28. Diagnostic value of p53 immunohistochemical expression in high-grade serous ovarian carcinoma (HGSC): casuistry in our center in the last three years

High-grade serous carcinoma (HGSC) is the most common ovarian carcinoma (70%). Age over 60 years, a family history of breast/ovarian carcinoma, and infertility are risk factors. Etiology remains unknown. These tumours arise from tubal-type epithelium, usually in the fallopian fimbria and less commonly in the ovarian surface or within ovarian epithelial inclusion cysts. Nearly every tumour harbours a deleterious TP53 mutation [1].[ Methods: A review of all cases diagnosed in our centre of high-grade serous ovarian carcinoma in surgical specimens, between 1 January 2019 and 31 December 2021, was carried out. The immunohistochemical profile (WT-1, p16 and p53) was compared with the results obtained in the massive NGS sequencing panel, focusing on the study of p53. Results: In our centre, 32 cases of high-grade ovarian serous carcinoma were diagnosed in 2019, 2020 and 2021, of which 31 showed a mutates immunohistochemical staining pattern (65% with p53 overexpression and 32% with null pattern), and 1 a non-mutated pattern. Of these 31 patients, 19 (61%) underwent genetic sequencing (Oncomine). Of these 19 patients, 16 (84%) had a p53 somatic lineage mutation and 3 (16%) were non-mutated. Immunostaining was positive for p16 in 30 cases, with intense cytoplasmic and nuclear staining expression in 27 of them and patchy and diffuse in 3, WT1 was positive in all of them. Discussion and conclusions: In this case series we would like to share our centre´s case reports on this pathology, with special emphasis on the importance of p53 immunohistochemical staining as a surrogate marker for sequencing techniques. Its use allows patients to be classified into high or low grade serous carcinomas, important in biopsy for diagnostic and therapeutic orientation due to its high degree of sensitivity to chemotherapy treatment in those with mutation, making this marker one of the most relevant in ovarian tumour pathology. Abstract

AIM: To evaluates the effectiveness and safety of the first generation, NS3/4A protease inhibitors (PIs) in clinical practice against chronic C virus, especially in patients with advanced fibrosis. METHODS: Prospective study and non-experimental analysis of a multicentre cohort of 38 Spanish hospitals that includes patients with chronic hepatitis C genotype 1, treatment-nai¨ve (TN) or treatment-experienced (TE), who underwent triple therapy with the first generation NS3/4A protease inhibitors, boceprevir (BOC) and telaprevir (TVR), in combination with pegylated interferon and ribavirin. The patients were treatment in routine practice settings. Data on the study population and on adverse clinical and virologic effects were compiled during the treatment period and during follow up. RESULTS: One thousand and fifty seven patients were included, 405 (38%) were treated with BOC and 652 (62%) with TVR. Of this total, 30% (n = 319) were TN and the remaining were TE: 28% (n = 298) relapsers, 12% (n = 123) partial responders (PR), 25% (n = 260) null-responders (NR) and for 5% (n = 57) with prior response unknown. The rate of sustained virologic response (SVR) by intention-to-treatment (ITT) was greater in those treated with TVR (65%) than in those treated with BOC (52%) (P < 0.0001), whereas by modified intention-to-treatment (mITT) no were found significant differences. By degree of fibrosis, 56% of patients were F4 and the highest SVR rates were recorded in the non-F4 patients, both TN and TE. In the analysis by groups, the TN patients treated with TVR by ITT showed a higher SVR (P = 0.005). However, by mITT there were no significant differences between BOC and TVR. In the multivariate analysis by mITT, the significant SVR factors were relapsers, IL28B CC and non-F4; the type of treatment (BOC or TVR) was not significant. The lowest SVR values were presented by the F4-NR patients, treated with BOC (46%) or with TVR (45%). 28% of the patients interrupted the treatment, mainly by non-viral response (51%): this outcome was more frequent in the TE than in the TN patients (57% vs 40%, P = 0.01). With respect to severe haematological disorders, neutropaenia was more likely to affect the patients treated with BOC (33% vs 20%, P = 0.0001), and thrombocytopaenia and anaemia, the F4 patients (P = 0.000, P = 0.025, respectively). CONCLUSION: In a real clinical practice setting with a high proportion of patients with advanced fibrosis, effectiveness of first-generation PIs was high except for NR patients, with similar SVR rates being achieved by BOC and TVR.

Abstract

The canonical biplot method (CB) is used to determine the discriminatory power of volatile chemical compounds in cheese. These volatile compounds were used as variables in order to differentiate among 6 groups or populations of cheeses (combinations of two seasons (winter and summer) with 3 types of cheese (cow, sheep and goat’s milk). We analyzed a total of 17 volatile compounds by means of gas chromatography coupled with mass detection. The compounds included aldehydes and methyl-aldehydes, alcohols (primary, secondary and branched chain), ketones, methyl-ketones and esters in winter (WC) and summer (SC) cow’s cheeses, winter (WSh) and summer (SSh) sheep’s cheeses and in winter (WG) and summer (SG) goat’s cheeses. The CB method allows differences to be found as a function of the elaboration of the cheeses, the seasonality of the milk, and the separation of the six groups of cheeses, characterizing the specific volatile chemical compounds responsible for such differences., Papel del método biplot canónico en el estudio de compuestos volátiles en quesos de composición variable. El método biplot canónico (CB) se utiliza para determinar el poder discriminatorio de compuestos químicos volátiles en queso. Los compuestos volátiles se utilizan como variables con el fin de diferenciar entre los 6 grupos o poblaciones de quesos (combinaciones de dos temporadas (invierno y verano) con 3 tipos de queso (vaca, oveja y cabra). Se analizan un total de 17 compuestos volátiles por medio de cromatografía de gases acoplada con detección de masas. Los compuestos incluyen aldehídos y metil-aldehídos, alcoholes (primarios de cadena, secundaria y ramificada), cetonas, metil-cetonas y ésteres. Los seis grupos de quesos son, quesos de vaca de invierno (WC) y verano (SC); quesos de oveja de invierno (WSh) y verano (SSh) y quesos de cabra de invierno (WG) y verano (SG). 45. NIR spectroscopy to identify and quantify imazapyr in soil

Abstract

The analysis of herbicide residues applied to the soil is commonly conducted by using chromatographic techniques which use organic solvents, are costly and time consuming. Having a simple and inexpensive discriminant method to determine samples with and without herbicide residues would be highly beneficial to lower costs of analysis and save time. The general objective of the present work was to develop such methodology. In particular, the presence of imazapyr was quantified using a near-infrared spectroscopy (NIRS) and a chemometrics approach. The experimental soil for a qualitative experiment of discrimination was Phaeozem with different agricultural managements and the following treatments: dry soil doped with imazapyr, rehydrated soil also doped with the herbicide, and control soil without the chemical. A discriminant algorithm was developed to allow the identification of soils containing imazapyr and those which did not contain the herbicide. The qualitative approach was based on a discriminant partial least squares (DPLS). An accurate identification of 100 percent of the soils containing imazapyr and 98 percent of those which did not contain the pesticide was achieved. Quantification of imazapyr was performed in five soil types (Andosol, Vertisol, Acrisol, Cambisol and Phaeozem) with different concentrations of the herbicide with an equation generated by modified partial least squares (MPLS) regression. As a reference for the above analytical method, a high performance liquid chromatography (HPLC) analysis was performed. The resulting equation had a RSQ of 0.81 and a prediction capacity (RPD) of 2.4. The results showed that with NIR spectroscopy it was possible to discriminate soils that contained imazapyr from those that did not contain the herbicide. This finding allows for a reduction in the number of samples to be evaluated to determine their concentration. Moreover, with this technique it is also possible to estimate the amount of herbicide in the soil with similar results to those accomplished using the reference method (HPLC), but faster and at lower cost

Abstract

We investigated the composition in vitamins A and E of cheeses made from the milks of ewes, goats and cows. A total of 84 cheeses of known composition were prepared and controlled to determine the influence of different factors, e.g. the variable proportions of cow’s, ewe’s, and goat’s milks, seasonality (winter/summer), and evolution during the course of ripening. The variable proportions of milk from the different species did not vary in either the amount of vitamin A or that of vitamin E in the cheeses. Seasonality and ripening were seen to have a significant effect on the concentration of vitamin A

Abstract

The study deals with the effects of the origin of milk (cow, ewe, goat, at different proportions), seasonality, and ripen- ing time on the contents of conjugated linoleic acid (CLA) in 224 samples of cheese. The sum of the cis 9, trans 11 and trans 10, cis 12 isomers was determined by GC-FID, after the extraction and methylation of the fatty acids of the samples, observing that the mean amount of CLA was 2.22, 2.72, and 3.54 mg/g of cheese, depending on the proportions of cow’s, goat’s, or ewe’s milks, respectively. The contents in cow’s, ewe’s, and goat’s milk, together with the ripening time and seasonality, were seen to have significant effects ( P < 0.05) on the concentration of CLA. The Pearson correlation re- vealed an inverse correlation between the content of CLA and the % of cow’s milk ( r = –0.269, P < 0.01) and seasonality ( r = –0.290, P < 0.01), and a direct correlation between CLA content and the % of ewe’s milk ( r = 0.312, P < 0.01) and the month of ripening ( r = 0.188, P < 0.01)
