Key points A fundamental question in sensory neuroscience revolves around how neurons represent complex visual stimuli. In mammalian primary visual cortex (V1), neurons decode intricate visual features to identify objects, with most being selective for edge orientation, but with half of those also developing invariance to edge position within their receptive fields. Position invariance allows cells to continue to code an edge even when it moves around. Combining feature selectivity and invariance is integral to successful object recognition. Considering the marsupial–eutherian divergence 160 million years ago, we explored whether feature selectivity and invariance was similar in marsupials and eutherians. We recovered the spatial filters and non‐linear processing characteristics of the receptive fields of neurons in wallaby V1 and compared them with previous results from cat cortex. We stimulated the neurons in V1 with white Gaussian noise and analysed responses using the non‐linear input model. Wallabies exhibit the same high percentage of orientation selective neurons as cats. However, in wallabies we observed a notably higher prevalence of neurons with three or more filters compared to cats. We show that having three or more filters substantially increases phase invariance in the V1s of both species, but that wallaby V1 accentuates this feature, suggesting that the species condenses more processing into the earliest cortical stage. These findings suggest that evolution has led to more than one solution to the problem of creating complex visual processing strategies. Previous studies have shown that the primary visual cortex (V1) in mammals is essential for processing complex visual stimuli, with neurons displaying selectivity for edge orientation and position. Fire offers both opportunities and risks for wildlife. Its impact will depend on the fire's scale, how it alters key resources and how animals move. Understanding how wildlife respond to fire is crucial as climate change is predicted to increase wildfire risk and will likely result in more frequent prescribed fire to reduce wildfire risk. Invasive predators and inappropriate fire regimes in south-eastern Australia threaten the long-nosed potoroo (Potorous tridactylus), a vulnerable marsupial often residing in areas frequently exposed to fire. The cumulative impacts of fire and predation may increase the threat to P. tridactylus after fire, as predators can be more effective in the immediate post-fire environment and P. tridactylus is often dependent on thick ground cover. We present a before-after control-impact experiment describing the influence of prescribed fire on P. tridactylus. We fitted GPS collars to 52 individuals at nine independent sites to test if exposure to prescribed burning reduced their survival or altered their movement behavior. Prescribed fire reduced P. tridactylus survival, yet range size and diffusion (movement) rate remained largely unaffected. With limited fire exposure, P. tridactylus tended to continue using burnt areas whereas activity became restricted to unburnt areas when larger proportions of their home range burnt. Site fidelity was very high - individuals rarely moved their home ranges after fire, regardless of fire exposure. Our results suggest recently burnt areas may be particularly dangerous for P. tridactylus: areas that can be attractive yet confer lower fitness outcomes. P. tridactylus may benefit from smaller fire scars, retention of structurally complex vegetation, and integrating invasive predator control with prescribed burning. [ABSTRACT FROM AUTHOR]

\nIntroduction: In many mammals, the testes descend from its abdominal position on the mesonephric kidney and are housed in the scrotum. It has been speculated that metatherians and eutherians might have acquired the scrotal testis independently because metatherians have the scrotum cranially to the phallus, while eutherians, such as humans and mice, possess it caudally. Rather, recent studies based on sequence comparisons of testis-descent-related genes indicate that the metatherian-eutherian common ancestor might already possess the descent mechanisms. To further elucidate the path of scrotal testis evolution, it is informative to compare the processes of the descent and scrotum development between metatherian and eutherian model animals. Methods: In this study, we histologically and molecularly compare these processes in gray short-tailed opossum (Monodelphis domestica), the most commonly used metatherian experimental model, and compare them with those in mouse. Results: Our observations indicate that, while transabdominal phase of the descent appears to be largely similar, scrotal phase differs due to their distinct scrotum positions. Our cell-labeling analyses and dynamic expression of Gsc1 reveal extensive cell/tissue rearrangements in murine scrotal development. In contrast, Gsc1 is not expressed in the developing genitalia and scrotal primordium of the opossum. Conclusion: Our results suggest recruitment of new regulatory pathways for the scrotum development and the scrotal phase of the testis descent during the evolution of eutherian mammals. Identifying and analysing isolated populations is critical for conservation. Isolation can make populations vulnerable to local extinction due to increased genetic drift and inbreeding, both of which should leave imprints of decreased genome‐wide heterozygosity. While decreases in heterozygosity among populations are frequently investigated, fewer studies have analysed how heterozygosity varies among individuals, including whether heterozygosity varies geographically along lines of discrete population structure or with continuous patterns analogous to isolation by distance. Here we explore geographical patterns of differentiation and individual heterozygosity in the threatened eastern barred bandicoot (Perameles gunnii) in Tasmania, Australia, using genomic data from 85 samples collected between 2008 and 2011. Our analyses identified two isolated demes undergoing significant genetic drift, and several areas of fine‐scale differentiation across Tasmania. We observed discrete genetic structures across geographical barriers and continuous patterns of isolation by distance, with little evidence of recent or historical migration. Using a recently developed analytical pipeline for estimating autosomal heterozygosity, we found individual heterozygosities varied within demes by up to a factor of two, and demes with low‐heterozygosity individuals also still contained those with high heterozygosity. Spatial interpolation of heterozygosity scores clarified these patterns and identified the isolated Tasman Peninsula as a location where low‐heterozygosity individuals were more common than elsewhere. Our results provide novel insights into the relationship between isolation‐driven genetic structure and local heterozygosity patterns. These may help improve translocation efforts, by identifying populations in need of assistance, and by providing an individualised metric for identifying source animals for translocation. The common opossum (Didelphis marsupialis) is a marsupial widely distributed in the Neotropics, where it lives in urbanized environments. The apparent scarcity and fragmentation of available habitat, and high rates of vehicle collisions, may represent barriers to dispersal. To assess the functional connectivity of this species and its potential use as a model in urban ecology, we evaluated the structure and genetic diversity of opossums in the metropolitan area of the Aburrá Valley (AMVA), Colombia, using sequences of the mitochondrial Cytochrome B (Cytb) gene and nine autosomal microsatellite loci. Although Cytb presented low levels of variation, microsatellite markers revealed high genetic diversity (He = 0.852 and Ho = 0.698). The geographic distribution of mitochondrial lineages and a spatial principal component analysis (sPCA) of microsatellites showed the absence of genetic structure and effective barriers to dispersal of opossums in the study area. This scenario suggests that hostile landscapes such as those in urban environments may be suitable enough to allow the effective connectivity of some species such as the common opossum and others with high dispersal ability in urbanized areas. Nonetheless, it is necessary to evaluate the impact of releases of rescued fauna made by the environmental authority on the population structure of urban opossums. [ABSTRACT FROM AUTHOR]

Sarcoptes scabiei mites and skin lesions consistent with severe sarcoptic mange were identified in a Tasmanian devil (Sarcophilus harrisii) and Bennett's wallaby (Notamacropus rufogriseus) from Tasmania, Australia. The devil and wallaby both had severe hyperkeratotic skin lesions. All stages of mite development were identified in the devil, suggesting parasite reproduction on the host. The devil was also affected by devil facial tumor disease and several other parasites. This expands the global host range of species susceptible to this panzootic mange disease. Southern greater gliders (Petauroides volans) are endangered Australian marsupials for which there is little health data currently available. Chlamydia pecorum is the only reported pathogen of greater gliders and infects a broad range of hosts, including other marsupials, ruminants, swine and birds. Conjunctival and cloacal swabs collected from thirty-two southern greater gliders across southeastern New South Wales, Australia were screened for Chlamydia spp. by 23S real-time PCR. None of the ninety-six swabs tested returned a positive result for Chlamydia DNA. We found no evidence of Chlamydia infection among the four geographically disparate populations sampled, suggesting that Chlamydia may not pose a widespread health risk to southern greater gliders in this region, based on the current data. However, various clinical signs of ophthalmic pathology were observed, warranting further investigation to determine their underlying aetiology. [ABSTRACT FROM AUTHOR]

Abstract Wildlife harbour a diverse range of microorganisms that affect their health and development. Marsupials are born immunologically naïve and physiologically underdeveloped, with primary development occurring inside a pouch. Secretion of immunological compounds and antimicrobial peptides in the epithelial lining of the female’s pouch, pouch young skin, and through the milk, are thought to boost the neonate’s immune system and potentially alter the pouch skin microbiome. Here, using 16S rRNA amplicon sequencing, we characterised the Tasmanian devil pouch skin microbiome from 25 lactating and 30 non-lactating wild females to describe and compare across these reproductive stages. We found that the lactating pouch skin microbiome had significantly lower amplicon sequence variant richness and diversity than non-lactating pouches, however there was no overall dissimilarity in community structure between lactating and non-lactating pouches. The top five phyla were found to be consistent between both reproductive stages, with over 85% of the microbiome being comprised of Firmicutes, Proteobacteria, Fusobacteriota, Actinobacteriota, and Bacteroidota. The most abundant taxa remained consistent across all taxonomic ranks between lactating and non-lactating pouch types. This suggests that any potential immunological compounds or antimicrobial peptide secretions did not significantly influence the main community members. Of the more than 16,000 total identified amplicon sequence variants, 25 were recognised as differentially abundant between lactating and non-lactating pouches. It is proposed that the secretion of antimicrobial peptides in the pouch act to modulate these microbial communities. This study identifies candidate bacterial clades on which to test the activity of Tasmanian devil antimicrobial peptides and their role in pouch young protection, which in turn may lead to future therapeutic development for human diseases.

X chromosome inactivation (XCI) is an epigenetic process that results in the transcriptional silencing of one X chromosome in the somatic cells of females. This phenomenon is common to both eutherian and marsupial mammals, but there are fundamental differences. In eutherians, the X chosen for silencing is random. DNA methylation on the eutherian inactive X is high at transcription start sites (TSSs) and their flanking regions, resulting in universally high DNA methylation. This contrasts XCI in marsupials where the paternally derived X is always silenced, and in which DNA methylation is low at TSSs and flanking regions. Here, we examined the DNA methylation status of the tammar wallaby X chromosome during spermatogenesis to determine the DNA methylation profile of the paternal X prior to and at fertilization. Whole genome enzymatic methylation sequencing was carried out on enriched flow-sorted populations of premeiotic, meiotic, and postmeiotic cells. We observed that the X displayed a pattern of DNA methylation from spermatogonia to mature sperm that reflected the inactive X in female somatic tissue. Therefore, the paternal X chromosome arrives at the egg with a DNA methylation profile that reflects the transcriptionally silent X in adult female somatic tissue. Leadbeater's possums (Gymnobelideus leadbeateri) are a critically endangered marsupial found in a restricted area of cold, wet forest in South‐Eastern Australia. The majority of Leadbeater's possums inhabit highland forest, with one outlying lowland population. In 2012, a breeding program was established for the lowland Leadbeater's possums when this genetically distinct population faced imminent extinction. Successful reproduction by highland Leadbeater's possums in the international zoo‐based population between 1970 and 2010 led to the widespread belief that the species bred readily in captivity. Lowland possums have not bred in the 2012–2021 contemporary captive conservation breeding program. This study reviewed the historic captive‐breeding data and found that of the 84% (162/194) that reached reproductive maturity; 37% of males (n = 30) and 39.5% of females (n = 32) bred, and this success was highly skewed towards a subset of highly fecund individuals (14% of females and 15% of males produced 75% and 80% of all offspring). Although lack of reproductive output in the captive lowland animals could be explained if age at mortality was lower than that of highlands possums, comparison of the longevity of highland and lowland animals had no significant difference. Conservation objectives that specify how captive breeding may support in situ recovery of wild populations are integral to the success of captive programs. A lack of reflective analysis of past husbandry records allowed misconceptions of success and approaches implemented in the management of the breeding program, reducing the benefits for the conservation of this high profile threatened species. This case study provides a lesson for the management of conservation breeding programs and illustrates the importance of well‐defined conservation objectives, integration of in situ and ex situ strategies, and the importance of objective, systematic and timely analysis of available evidence to inform management objectives and improve conservation outcomes in real time. Males of the endangered northern quoll (Dasyurus hallucatus) are facultatively semelparous, often dying following their first breeding season. This is often driven by high breeding costs: males travel and mate extensively in breeding season, resulting in reduced body condition. Annual post‐breeding survival rates for male northern quolls have been recorded between 0% and 12.5% across their range in northern Australia. We highlight an observed 50% survival rate of male northern quolls (3 of 6) living into their second year in a Pilbara mining landscape. High male survival here may be attributed to above average rainfall in the landscape and reduced breeding opportunities, due to low female abundance—likely driven by large amounts of unfavourable mining land. This likely led to lower hormonal stress associated with breeding for males, as well as increased resource availability. Given northern quoll population persistence is highly dependent on breeding success, high survival of male northern quolls may inadvertently stabilize populations if they can breed again in a second year, though this is yet to be observed in the Pilbara region. Nevertheless, semelparity of male northern quolls likely varies with environmental and demographic factors, demonstrating that northern quoll populations may be resilient to disturbance in an unpredictable semi‐arid environment. Global biodiversity loss continues unabated, and in Australia, the rate of recent mammal extinctions is among the worst in the world. Meanwhile, the diversity among and within many endemic mammal species remains undescribed. This information is crucial to delineate species boundaries and thus inform decision‐making for conservation. Sminthopsis virginiae (the red‐cheeked dunnart) is a small, dasyurid marsupial found in four disjunct populations around the northern coast of Australia and New Guinea. There are three currently recognized subspecies, each occupying a distinct geographic location. Sminthopsis v. virginiae occurs in Queensland, S. v. rufigenis is distributed across New Guinea and the Aru Islands, and S. v. nitela has populations in the Top End of the Northern Territory and the Kimberley region of Western Australia. Previous molecular work has suggested the current subspecies definitions are not aligned with DNA sequence data, though the sampling was limited. We undertook a comprehensive genetic and morphological review of S. virginiae to clarify relationships within the species. This included mitochondrial (CR, 12S, and cytb) and nuclear (omega‐globin, IRBP, and bfib7) loci, and morphometric analysis of skulls and whole wet‐preserved specimens held in museums. Maximum Likelihood and Bayesian phylogenetic analyses resolved samples into two distinct clades, demarcated by the Gulf of Carpentaria in Australia's north. Sminthopsis. v. nitela was consistently separated from S. v. virginiae and S. v. rufigenis, based on the overall body and skull size and craniodental features, while S. v. virginiae and S. v. rufigenis were more difficult to distinguish from each other. Thus, we redescribed S. virginiae, recognizing two species, S. nitela (raised from subspecies) and S. virginiae (now comprising the subspecies S. v. virginiae and S. v. rufigenis). This study highlights the importance of recognizing cryptic mammal fauna to help address the gap in our knowledge about diagnosing diversity during a time of conservation crisis. The major histocompatibility complex (MHC) plays a vital role in the vertebrate immune system due to its role in infection, disease and autoimmunity, or recognition of "self". The marsupial MHC class II genes show divergence from eutherian MHC class II genes and are a unique taxon of therian mammals that give birth to altricial and immunologically naive young providing an opportune study system for investigating evolution of the immune system. Additionally, the MHC in marsupials has been implicated in disease associations, including susceptibility to Chlamydia pecorum infection in koalas. Due to the complexity of the gene family, automated annotation is not possible so here we manually annotate 384 class II MHC genes in 29 marsupial species. We find losses of key components of the marsupial MHC repertoire in the Dasyuromorphia order and the Pseudochiridae family. We perform PGLS analysis to show the gene losses we find are true gene losses and not artifacts of unresolved genome assembly. We investigate the associations between the number of loci and life history traits, including lifespan and reproductive output in lineages of marsupials and hypothesize that gene loss may be linked to the energetic cost and tradeoffs associated with pregnancy and reproduction. We found support for litter size being a significant predictor of the number of DBA and DBB loci, indicating a tradeoff between the energetic requirements of immunity and reproduction. Additionally, we highlight the increased susceptibility of Dasyuridae species to neoplasia and a potential link to MHC gene loss. Finally, these annotations provide a valuable resource to the immunogenetics research community to move forward and further investigate diversity in MHC genes in marsupials. [ABSTRACT FROM AUTHOR]

Retroviruses are an ancient viral family that have globally coevolved with vertebrates and impacted their evolution. In Australia, a continent that has been geographically isolated for millions of years, little is known about retroviruses in wildlife, despite the devastating impacts of a retrovirus on endangered koala populations. We therefore sought to identify and characterize Australian retroviruses through reconstruction of endogenous retroviruses from marsupial genomes, in particular the Tasmanian devil due to its high cancer incidence. We screened 19 marsupial genomes and identified over 80,000 endogenous retrovirus fragments which we classified into eight retrovirus clades. The retroviruses were similar to either Betaretrovirus (5/8) or Gammaretrovirus (3/8) retroviruses, but formed distinct phylogenetic clades compared to extant retroviruses. One of the clades (MEBrv 3) lost an envelope but retained retrotranspositional activity, subsequently amplifying throughout all Dasyuridae genomes. Overall, we provide insights into Australian retrovirus evolution and identify a highly active endogenous retrovirus within Dasyuridae genomes. [ABSTRACT FROM AUTHOR]

Context: Fire regimes are changing with ongoing climate change, which is leading to an increase in fire frequency and severity. Australia's Black Summer wildfires burned >12 million hectares in 2019–2020, affecting numerous threatened animal species. One of the species predicted to be most impacted was the threatened southern greater glider, an arboreal, hollow-dependent folivore, endemic to eastern Australia's eucalypt forests. Aims: This study aimed to assess how the 2019–2020 wildfires affected greater glider abundance and the resources they depend on in Woomargama National Park, New South Wales, Australia. Methods: We categorised 32 sites into four fire severity treatments with eight sites for each treatment: unburned (continuous unburned vegetation); refuges (unburned patches within the fire's perimeter); low-moderate severity; and high severity. We carried out two spotlight surveys per site using the double-observer method, beginning 21 months after the fires. We also conducted vegetation assessments on the same transects. To analyse the data, we used Generalised Linear Models to compare habitat differences based on fire severity, and N-mixture models to model greater glider detectability and abundance in relation to habitat and fire severity. Key results: We found that fire severity depleted several habitat variables including canopy cover and the number of potentially hollow-bearing trees, a resource that greater gliders rely on. Greater glider abundance also decreased in all burn categories, with the greatest decline experienced in areas burned at high severity. We also found that greater glider abundance was much lower in fire refuges than unburned habitat outside of the fire zone. Conclusions: Greater glider declines following severe wildfire can be at least partly attributed to the level of vegetation loss and the associated loss of key habitat resources. The contribution of direct mortality to population declines remains unknown. Implications: Greater glider conservation will rely heavily on protecting expansive unburned areas of suitable habitat and maintaining hollow-bearing trees. Background: Although urban wildlife has increased during the last years, the effects of urbanization on wildlife are poorly understood. Common opossum (Didelphis marsupialis) is a frequent dweller of the Metropolitan Area of Valle del Aburrá (AMVA) in the Antioquia province (Colombia) where opossum-human conflicts result in large numbers of injured or dead opossums. Objective: To evaluate the effects of urban and rural habitats on the morphometry and reproductive output (litter size in marsupium) of opossums. Methods: A total of 131 opossums were captured: 94 in the AMVA, and 37 in rural areas of Antioquia. Opossums were weighed, measured, sexed, clinically inspected, and photographed. Results: Urban opossums were heavier and larger than rural individuals, regardless of sex, and no differences were observed in terms of reproductive output between both habitats. Additionally, urban opossums were more abundant than rural opossums, possibly explained by the fact that cities favor survival -through higher food availability-but not reproduction. Alternatively, bigger animals could also be explained by inadequate diets rich in carbohydrates which may also have deleterious health effects. We observed sexual size dimorphism in head and tail length but not in body length of opossums. Conclusion: We confirm that urban environments have a significant effect on body mass and size of opossums; however, it is unknown whether this is beneficial or detrimental for these animals. Dispersal is an important process that is widely studied across species, and it can be influenced by intrinsic and extrinsic factors. Intrinsic factors commonly assessed include the sex and age of individuals, while landscape features are frequently-tested extrinsic factors. Here, we investigated the effects of both sex and landscape composition and configuration on genetic distances among bare-nosed wombats (Vombatus ursinus)—one of the largest fossorial mammals in the world and subject to habitat fragmentation, threats from disease, and human persecution including culling as an agricultural pest. We analyzed a data set comprising 74 Tasmanian individuals (30 males and 44 females), genotyped for 9,064 single-nucleotide polymorphisms. We tested for sex-biased dispersal and the influence of landscape features on genetic distances including land use, water, vegetation, elevation, and topographic ruggedness. We detected significant female-biased dispersal, which may be related to females donating burrows to their offspring due to the energetic cost of excavation, given their large body sizes. Land use, waterbodies, and elevation appeared to be significant landscape predictors of genetic distance. Land use potentially reflects land clearing and persecution over the last 200 years. If our findings based on a limited sample size are valid, retention and restoration of nonanthropogenic landscapes in which wombats can move and burrow may be important for gene flow and maintenance of genetic diversity. The genus Gracilinanus ranges from savannas to dense forests in South America, yet its systematics have never been thoroughly investigated across its wide distributional range. We assessed Gracilinanus phylogenetic relationships, species boundaries, and geographical limits using mtDNA sequences. Our analysis confirmed the distinctiveness of the six recognized species (G. aceramarcae, G. agilis, G. emiliae, G. marica, G. microtarsus, and G. peruanus), with a mean p-distance for interspecific nucleotide sequence divergences ranging from 13–16.2% and robust phylogenetic support (BPP > 0.95; BS > 75%). Refined species delimitation approaches (GMYC, PTP, ASAP) revealed potential cryptic diversity, suggesting up to 20 candidate species. Three geographically structured and divergent lineages (4.1–4.8% sequence divergence) were identified within G. agilis, extending its Cerrado range. Within G. emiliae, we found divergence values ranging from 4.7–5.7% and the first known record for the northeastern Atlantic Forest. Three divergent clades were recovered within G. microtarsus (9.0–9.8% sequence divergence), including a new lineage for the northern Atlantic Forest. For G. peruanus, we found two divergent lineages (7.2%) and the first documented occurrence for Amazonian lowland forest. This comprehensive sampling revealed greater genetic diversity in Gracilinanus, extending its geographic limits. Here we propose nine putative new species, emphasizing a hidden diversity that warrants formal description and further increases the taxonomic diversity of this genus. Kenngoor (Phascogale calura) persist in < 1% of their original distribution, occupying highly fragmented remnant habitat in south-west Western Australia, with very little known of the genetic diversity of the remaining wild populations. Recently, the species has been translocated to managed reserves to improve its conservation. Understanding genetic structure and patterns of genetic diversity is crucial to inform conservation translocations for species recovery. This study aims to (1) assess genetic structure and genetic diversity across remaining wild locations, (2) assess long-term genetic outcomes of a mixed-source wild-to-wild translocation, and (3) estimate global effective population size. We genotyped 209 samples from 13 locations of fragmented remnant habitat using reduced representation sequencing. An isolation by distance model best explained genetic structure across the survey areas, with evidence of fine scale divergence of two northern locations. Allelic richness and autosomal heterozygosity measures indicated that diversity is spread uniformly across locations, and no locations showed signs of inbreeding or strong genetic drift. The mixed-source translocation has retained the diversity of the wider species ten years post-translocation. Overall, our results suggest that connectivity between survey areas has largely been maintained and that no location has substantially lower genetic diversity, despite the highly fragmented nature of remnant kenngoor habitat. Future translocations should aim to represent a mixture of genetically divergent locations to maintain the diversity present at the species level. Background: Koalas, an Australian arboreal marsupial, depend on eucalypt tree leaves for their diet. They selectively consume only a few of the hundreds of available eucalypt species. Since the koala gut microbiome is essential for the digestion and detoxification of eucalypts, their individual differences in the gut microbiome may lead to variations in their eucalypt selection and eucalypt metabolic capacity. However, research focusing on the relationship between the gut microbiome and differences in food preferences is very limited. We aimed to determine whether individual and regional differences exist in the gut microbiome of koalas as well as the mechanism by which these differences influence eucalypt selection. Methods: Foraging data were collected from six koalas and a total of 62 feces were collected from 15 koalas of two zoos in Japan. The mitochondrial phylogenetic analysis was conducted to estimate the mitochondrial maternal origin of each koala. In addition, the 16S-based gut microbiome of 15 koalas was analyzed to determine the composition and diversity of each koala's gut microbiome. We used these data to investigate the relationship among mitochondrial maternal origin, gut microbiome and eucalypt diet selection. Results and Discussion: This research revealed that diversity and composition of the gut microbiome and that eucalypt diet selection of koalas differs among regions. We also revealed that the gut microbiome alpha diversity was correlated with foraging diversity in koalas. These individual and regional differences would result from vertical (maternal) transmission of the gut microbiome and represent an intraspecific variation in koala foraging strategies. Further, we demonstrated that certain gut bacteria were strongly correlated with both mitochondrial maternal origin and eucalypt foraging patterns. Bacteria found to be associated with mitochondrial maternal origin included bacteria involved in fiber digestion and degradation of secondary metabolites, such as the families Rikenellaceae and Synergistaceae. These bacteria may cause differences in metabolic capacity between individual and regional koalas and influence their eucalypt selection. Conclusion: We showed that the characteristics (composition and diversity) of the gut microbiome and eucalypt diet selection of koalas differ by individuals and regional origins as we expected. In addition, some gut bacteria that could influence eucalypt foraging of koalas showed the relationships with both mitochondrial maternal origin and eucalypt foraging pattern. These differences in the gut microbiome between regional origins may make a difference in eucalypt selection. Given the importance of the gut microbiome to koalas foraging on eucalypts and their strong symbiotic relationship, future studies should focus on the symbiotic relationship and coevolution between koalas and the gut microbiome to understand individual and regional differences in eucalypt diet selection by koalas. Introduced predators have been implicated in the extinction or range reduction of many Australian species, potentially due to native wildlife exhibiting prey naivety. We used traps with predator faeces present, monitored by camera traps, to determine if Australian mammals responded to native and introduced predator faeces. The presence of either faeces did not influence the trap or camera captures of any species. Some Australian mammals may exhibit prey naivety and not react to introduced predators. There are no detailed descriptions of deaths in species of marsupials in the scientific literature, nor of their responses to dead conspecifics. A camera trap captured the death of a southern hairy-nosed wombat (Lasiorhinus latifrons) and the responses of other wombats to its body. The emaciated wombat lay down and died over 2–3 h near a burrow. In the following months, its body was inspected 22 times by at least four wombats. It is unclear whether the wombats purposefully visited the dead wombat. Inspecting the body was likely to be related to encountering the dead wombat and being stimulated to investigate it. Wombats are interested in their dead. We found that Southern hairy-nosed wombats of a warren often inspected the body of a wombat that died there. Nutrition is essential for not only survival but also successful growth and reproduction. Dietary demands are increased in a diseased state due to the increased energy and nutritional requirements associated with immune response, inflammation and convalescence. The herbivorous bare-nosed wombat (Vombatus ursinus) is notably susceptible to sarcoptic mange, a disease caused by the mite, Sarcoptes scabiei , which causes debilitating pruritic skin disease and leads to secondary bacterial infections and increased wombat morbidity and mortality, as well as regionally variable population declines. It is unknown why wombats are so susceptible to sarcoptic mange and if nutrition may play a role in disease expression, particularly relating to seasonality. The objective of this study was to quantify the differences in the nutritive value, over four seasons, of plants (mostly grasses and sedges) that are available as food items for bare-nosed wombats. We collected plants over four seasons from five wombat habitats that were known to have wombats affected by sarcoptic mange. We found seasonal and site differences for macro and micronutrients in the plants analysed. Monitoring the diet quality of wombats in the wild is useful for managing their populations and understanding population dynamics in relation to food resource quality. Nutritional value of plants available as food for bare-nosed wombats was assessed at sites within NSW. Significant seasonal and site differences were determined. Wombats are affected by sarcoptic mange, which affects their metabolic rate and nutrient needs; therefore, supporting nutrition and health of wombats is important for conservation of this species. The Virginia opossum (Didelphis virginiana) has a rapidly expanding distribution in North America, but many aspects of its ecology remain relatively understudied, particularly in rural areas of its core range. We collected GPS telemetry data from 93 opossums in a rural, non-agricultural landscape in South Carolina, USA (2018–2019) to examine factors influencing space use and resource selection. Estimated male home ranges (99% utilization distributions) were on average 50% larger than those of females (mean home range 115.9 ± 103.7 ha vs 76.7 ± 75.0 ha). The home range size decreased on average by 20% with each 20% increase in deciduous land cover but was not affected by season or other landscape factors. Core area sizes (65% utilization distributions) were not influenced by sex (mean core area size 29.1 ± 23.7 ha and 22.4 ha ± 13.8 for males and females, respectively) or season, but the core area size decreased by 14% with each 400 m increase in distance from a permanent water source. Resource selection by opossums primarily occurred at the landscape level. Both males and females generally selected for wetlands while avoiding pine forests and developed/open areas, likely the result of differences in resource availability and predation risk between habitats. Opossums also tended to select for linear features such as unpaved roads and edge habitat, which may facilitate movement across the landscape. The home ranges we documented are among the largest recorded for opossums in the USA, likely the result of the relatively low resource abundance throughout our study area due to comparatively minimal anthropogenic influence. Simple Summary: This study focuses on the spontaneous lesions observed in a captive colony of geriatric Julia Creek dunnarts, an endangered carnivorous marsupial. Despite their conservation status, no prior research has focused on the conditions affecting captive individuals attaining senescence. We examined one wild and thirty-five captive-born, mostly elderly dunnarts that failed to reproduce over several breeding periods. From these, ten dunnarts had normal findings. Among females, the most common issue was cystic glandular hyperplasia (eight cases); cutaneous lesions were infrequent (two cases). Males showed testicular degeneration, aspermatogenesis, or atrophy (three cases). Cutaneous lesions compatible with epitheliotropic T cell lymphomas were observed in both sexes (five cases) and thus, an underlying oncogenic viral etiology is suspected. This is the first study that documents spontaneous diseases in aging Julia Creek dunnarts, shedding light on geriatric conditions within a conservation context. Julia Creek dunnarts are an endangered species of carnivorous marsupials and the focus of multiple conservation strategies involving significant resources such as captive breeding programs. Despite the relevance for conservation, no study to date has focused on evaluating geriatric diseases in dunnarts. This study describes the pathology findings in a group of one wild and thirty-five captive-born, mostly geriatric Julia Creek dunnarts that failed to produce offspring over multiple breeding periods. A total of 20 females and 16 males were submitted for a postmortem examination, with ages ranging from 9 to 42 and 12 to 42 months for females and males, respectively. Of these, 10 had unremarkable findings. The most common condition in females was cystic glandular hyperplasia (n = 8), typical of hormonal dysregulation profiles in senescence, particularly hyperestrogenism. Rarely, cutaneous disease represented by unidentified dermal round cell infiltrates was observed in females (n = 2). Primary reproductive hormonal dysregulation was also suspected in males diagnosed with testicular degeneration, aspermatogenesis and/or atrophy (n = 3). Cutaneous round cell infiltrates, possibly compatible with epitheliotropic lymphomas, were seen in males (n = 3), and 2/3 affected males also had concurrent testicular degeneration or atrophy, indicating male sex could be a predictor for lymphoid neoplasia in aged dunnarts, especially in individuals with concurrent testosterone-luteinizing hormone dysregulation as it occurs in gonadectomized animals. The role of an underlying viral etiology is also explored. This study is the first to describe major spontaneous diseases in endangered aged Julia Creek dunnarts, providing an important understanding of senescence and geriatric diseases within a conservation context. Abstract Global biodiversity loss continues unabated, and in Australia, the rate of recent mammal extinctions is among the worst in the world. Meanwhile, the diversity among and within many endemic mammal species remains undescribed. This information is crucial to delineate species boundaries and thus inform decision‐making for conservation. Sminthopsis virginiae (the red‐cheeked dunnart) is a small, dasyurid marsupial found in four disjunct populations around the northern coast of Australia and New Guinea. There are three currently recognized subspecies, each occupying a distinct geographic location. Sminthopsis v. virginiae occurs in Queensland, S. v. rufigenis is distributed across New Guinea and the Aru Islands, and S. v. nitela has populations in the Top End of the Northern Territory and the Kimberley region of Western Australia. Previous molecular work has suggested the current subspecies definitions are not aligned with DNA sequence data, though the sampling was limited. We undertook a comprehensive genetic and morphological review of S. virginiae to clarify relationships within the species. This included mitochondrial (CR, 12S, and cytb) and nuclear (omega‐globin, IRBP, and bfib7) loci, and morphometric analysis of skulls and whole wet‐preserved specimens held in museums. Maximum Likelihood and Bayesian phylogenetic analyses resolved samples into two distinct clades, demarcated by the Gulf of Carpentaria in Australia's north. Sminthopsis. v. nitela was consistently separated from S. v. virginiae and S. v. rufigenis, based on the overall body and skull size and craniodental features, while S. v. virginiae and S. v. rufigenis were more difficult to distinguish from each other. Thus, we redescribed S. virginiae, recognizing two species, S. nitela (raised from subspecies) and S. virginiae (now comprising the subspecies S. v. virginiae and S. v. rufigenis). This study highlights the importance of recognizing cryptic mammal fauna to help address the gap in our knowledge about diagnosing diversity during a time of conservation crisis.

The possession of fur or hair is a defining characteristic of mammals and can occur in a variety of colours and patterns. While genetic determinants of coat colour are well described in eutherian ‘placental’ mammals, the other major mammalian infraclass, marsupials, is grossly understudied. The fur of the common brushtail possum (Trichosurus vulpecula), an iconic native mammal found throughout Australia and introduced into Aotearoa New Zealand, possesses two main colour morphs: grey and black. To identify genetic variants associated with coat colour, we performed a genome-wide association study (GWAS) with genotype by sequencing (GBS) data. Single nucleotide variants (SNVs) on chromosome 3, close to the agouti signalling protein (ASIP) gene that controls the temporal and spatial distribution of pigments in eutherian mammals, were identified. Fine-mapping identified a C>T variant at chr3:100483705 that results in a ASIP:p.Arg115Cys missense substitution, and animals homozygous for this variant have black fur. In addition to uncovering the first genetic determinant of coat colour in a natural marsupial population, comparative analysis of ASIP in divergent marsupial species identified the dasyurids as having accelerated evolution, reflecting their well described diversity of coat colour and pattern.

Abstract Background Efficient movement and energy expenditure are vital for animal survival. Human disturbance can alter animal movement due to changes in resource availability and threats. Some animals can exploit anthropogenic disturbances for more efficient movement, while others face restricted or inefficient movement due to fragmentation of high-resource habitats, and risks associated with disturbed habitats. Mining, a major anthropogenic disturbance, removes natural habitats, introduces new landscape features, and alters resource distribution in the landscape. This study investigates the effect of mining on the movement of an endangered mesopredator, the northern quoll (Dasyurus hallucatus). Using GPS collars and accelerometers, we investigate their habitat selection and energy expenditure in an active mining landscape, to determine the effects of this disturbance on northern quolls. Methods We fit northern quolls with GPS collars and accelerometers during breeding and non-breeding season at an active mine site in the Pilbara region of Western Australia. We investigated broad-scale movement by calculating the movement ranges of quolls using utilisation distributions at the 95% isopleth, and compared habitat types and environmental characteristics within observed movement ranges to the available landscape. We investigated fine-scale movement by quolls with integrated step selection functions, assessing the relative selection strength for each habitat covariate. Finally, we used piecewise structural equation modelling to analyse the influence of each habitat covariate on northern quoll energy expenditure. Results At the broad scale, northern quolls predominantly used rugged, rocky habitats, and used mining habitats in proportion to their availability. However, at the fine scale, habitat use varied between breeding and non-breeding seasons. During the breeding season, quolls notably avoided mining habitats, whereas in the non-breeding season, they frequented mining habitats equally to rocky and riparian habitats, albeit at a higher energetic cost. Conclusion Mining impacts northern quolls by fragmenting favoured rocky habitats, increasing energy expenditure, and potentially impacting breeding dispersal. While mining habitats might offer limited resource opportunities in the non-breeding season, conservation efforts during active mining, including the creation of movement corridors and progressive habitat restoration would likely be useful. However, prioritising the preservation of natural rocky and riparian habitats in mining landscapes is vital for northern quoll conservation.

The world's largest extant carnivorous marsupial, the Tasmanian devil, is challenged by Devil Facial Tumor Disease (DFTD), a fatal, clonally transmitted cancer. In two decades, DFTD has spread across 95% of the species distributional range. A previous study has shown that factors such as season, geographic location, and infection with DFTD can impact the expression of immune genes in Tasmanian devils. To date, no study has investigated within-individual immune gene expression changes prior to and throughout the course of DFTD infection. To explore possible changes in immune response, we investigated four locations across Tasmania that differed in DFTD exposure history, ranging between 2 and >30 years. Our study demonstrated considerable complexity in the immune responses to DFTD. The same factors (sex, age, season, location and DFTD infection) affected immune gene expression both across and within devils, although seasonal and location specific variations were diminished in DFTD affected devils. We also found that expression of both adaptive and innate immune genes starts to alter early in DFTD infection and continues to change as DFTD progresses. A novel finding was that the lower expression of immune genes MHC-II, NKG2D and CD8 may predict susceptibility to earlier DFTD infection. A case study of a single devil with regressed tumor showed opposite/contrasting immune gene expression patterns compared to the general trends observed across devils with DFTD infection. Our study highlights the complexity of DFTD's interactions with the host immune system and the need for long-term studies to fully understand how DFTD alters the evolutionary trajectory of devil immunity. Unusual colorations are rare in Didelphis, but records are important to understand genetic aspects of the species. Albino individuals have a total absence of pigmentation throughout the body (skin and hair) and have eyes without traces of pigmentation, but with a red or pink color. We draw attention to the first two confirmed records of albinism in white-eared opossum (Didelphis albiventris) and their karyotype. This is the first record of albinism in D. albiventris in Northeastern Brazil. Introduction: During early development in most male mammals the testes move from a position near the kidneys through the abdomen to eventually reside in the scrotum. The transabdominal phase of this migration is driven by insulin-like peptide 3 (INSL3) which stimulates growth of the gubernaculum, a key ligament connecting the testes with the abdominal wall. While all marsupials, except the marsupial mole (Notoryctes typhlops), have a scrotum and fully descended testes, it is unclear if INSL3 drives this process in marsupials especially given that marsupials have a different mechanism of scrotum determination and position relative to the phallus compared to eutherian mammals. Methods: To understand if INSL3 plays a role in marsupial testicular descent we have sequenced and curated the INSL3 gene and its receptor (RXFP2) in a range of marsupials representing every order. Furthermore, we looked at single cell RNAseq and qPCR analysis of INSL3 in the fat-tailed dunnart testis (Sminthopsis crassicaudata) to understand the location and timing of expression during development. Results: These data show a strong phylogenetic similarity between marsupial and eutherian orthologues, but not with monotreme INSL3s which were more similar to the ancestral RLN3 gene. We have also shown the genomic location of INSL3, and surrounding genes is conserved in a range of marsupials and eutherians. Single cell RNA-seq and qPCR data show that INSL3 mRNA is expressed specifically in Leydig cells and expressed at higher levels during the testicular descent phase in developing marsupials. Discussion: Together, these data argue strongly for a therian origin of INSL3 mediated testicular descent in mammals and suggests that a coordinated movement of the testes to the abdominal wall may have preceded externalization in marsupials and therian mammals. The white‐eared opossum (Didelphis albiventris) is a Neotropical marsupial that occurs in the Brazilian territory. The larynx is an important organ of vocalization in mammals, although, other laryngeal functions are more fundamental for survival of mammals than phonation. The anatomical knowledge of respiratory structures is pivotal for a better understanding of the species. Thus, this study aimed to examine the larynx of the white‐eared opossum by gross anatomy, computed tomography and histological description. For this, 10 adult (six females and four males) white‐eared opossums (D. albiventris) were used. The white‐eared opossum larynx was formed by epiglottis, thyroid, cricoids and arytenoid cartilages and a corniculate process. There is a similarity between the larynx of this marsupial and those reported in other wild mammals, regarding the number of cartilages and their location. Histologically, the epiglottis consisted of elastic cartilage and thyroid, cricoid and arytenoid cartilages were composed of hyaline cartilage. The epiglottis protruded rostrally into the nasal part of the pharynx, above the soft palate, and this is probably a marsupial characteristic as the fact that the thyroid and cricoid cartilages were ventrally fused. The hyoid apparatus was similar to that of other animals, with the same bony constitution, but with a greatly reduced stylohyoid bone. Histologically, the larynx was similar to those of other species such as koala, armadillo, crab‐eating foxes and giant anteaters. The knowledge of the larynx morphology is important for the anatomical features of the species and clinical and surgical procedures, such as endotracheal intubation. [ABSTRACT FROM AUTHOR]

Small non-flying mammals represent 44% of Brazilian mammal species and have a wide range of habits and life-history strategies. This wide diversity requires different sampling methods in survey studies. We compared the efficiency of pitfall and live-traps in different vertical positions in relation to the alpha and beta diversity of small mammals in three forest fragments with different levels of conservation and in a continuous area in the southwestern Amazon, Acre state. Captures were carried out using a combination of pitfall traps and live-traps on the ground, understorey, and canopy. Taxonomic identification was performed by morphological and molecular analyses. Alpha diversity was evaluated using Hill numbers (q = 0 and q = 1). The turnover between different types of traps and different vertical strata (beta diversity) was analysed using permutation analysis of variance. Species richness between areas ranged from 6 to 21. The highest species richness was observed in ground traps, and the lowest species richness was observed in the canopy. Live-traps on the ground recorded a greater diversity in two areas. Pitfall traps recorded the greatest number of unique species in three areas. The different types of traps and the different vertical positions acted in a complementary way in the small mammal samplings. The turnover in relation to trap type and stratum indicated the formation of two significantly different groups: ground traps and aboveground traps. However, the use of canopy traps did not contribute significantly to an increase in the estimated species richness and diversity in three of the four localities. [ABSTRACT FROM AUTHOR]

Context. The distribution of the threatened greater bilby (Macrotis lagotis) has significantly contracted since the introduction of feral cats and foxes. To counteract these threats, bilbies have been reintroduced to multiple feral predator exclosures and offshore islands across Australia. Aims. The aims of this study were to monitor the reintroduction of bilbies to the feral predator exclosure at Currawinya National Park and assess three hypotheses: (1) captiveborn founders would establish stable home ranges and utilise habitats similar to those of wildborn bilbies in the exclosure; (2) founders would maintain adequate body condition and weight, enabling the survival of >50% of founders at 12 months post-release; and (3) the population would rapidly increase in the absence of feral predators, due to the high reproductive potential of bilbies. Methods. We used VHF/GPS telemetry to compare home range size and habitat use of 12 founders and 11 wild-born bilbies. Founders were monitored intensively to assess reproductive success, weight, body condition and survival. Pouch activity was monitored to examine reproductive output. Spatially explicit capture–recapture modelling was used to estimate population density/size. Key results. The population rapidly increased to >450 bilbies after 3 years, and founder survivorship was high (70% at 12 months post-release). Male founder home ranges (248.46 ha ± 97.22) were comparable in size to wild-born males (216.74 ha ± 54.19), but female founder home ranges (51.23 ha ± 12.22) were significantly larger than wild-born females (20.80 ha ± 2.94) (P = 0.04). The doubling of the population between 2021 and 2022 may have resulted in the contraction of female home ranges. Reproductive output decreased significantly in 2022, indicating that reproduction may be density-dependent at high densities. There was a significant relationship between track counts and population estimates, indicating that track counts are a reliable method for estimating population size inside the exclosure. Conclusions. This reintroduction has been highly successful to date, demonstrating that bilbies are an adaptable species capable of rapid population growth in the absence of feral predators. Bilbies are a model species for reintroductions to feral predator exclosures due to high survival rates, reproductive output, dietary flexibility and the ability to utilise a broad range of environments. Worldwide predation by feral cats has caused severe declines in many animal species. Prior to extinction, threatening processes typically alter species behaviour, habitat use, survival and reproductive rates. By monitoring these parameters, we may be able to determine if feral cats are a significant threatening process to a species prior to extinction. We aimed to test whether feral cats are negatively affecting the population size, morphometrics and movement patterns of mulgaras (Dasycercus blythi) on the Matuwa Indigenous Protected Area, in the arid rangelands of Western Australia. We hypothesised that mulgaras, which inhabit a cat-free environment, would have a larger home range and denser population, and be in better condition than those that inhabited an adjacent area with a suppressed density of feral cats. We trapped (n = 64), and radio-tracked (n = 19) mulgara inside and outside an I 100 ha cat-free fenced enclosure on five occasions in 2012. Treatment (i.e. inside or outside the cat-free enclosure) was a significant variable for explaining variation in mulgara density. There was no significant difference in morphometrics or the home ranges of mulgara between the sexes or treatments. Head length, pes length and weight, as well as tail width and length, were significantly different between sexes and over time, suggesting that seasonal variation influences mulgara populations more than low levels of cat predation. During a study of interactions among koalas, we observed interactions between adult males and dependent joeys. Most were passive in nature, suggesting that interactions with adult males are not a major threat to joey survival. However, one male-joey interaction resulted in a joey falling approximately 7 m to the ground, an incident that may have resulted in the death of the joey had we not intervened. We suggest that there is potential for infant death to occur under the generalised aggression hypothesis for koalas, although further observations are needed. [ABSTRACT FROM AUTHOR]

Australia's mammals are facing devastating effects from introduced predators such as cats and foxes. It is hypothesised that this may be because Australian mammals do not recognise some predators and are naïve to the threat they pose. We tested how wild-caught northern brown bandicoots (Isoodon macrourus) reacted to a variety of predators and their cues in captivity. These cues were live predators, taxidermied predators and predator faeces. Bandicoots spent less time feeding and took longer to begin feeding in the presence of a live dog. However, they did not display a significant difference in behaviours in the presence of any other predator cue when compared with the baseline. This suggests that bandicoots may perceive live dogs as a threat, and that the use of taxidermied predators and predator faeces may not elicit avoidance behaviours in bandicoots. Descrito como um “animal monstruoso” pelo cronista Pietro Martire d’Anghiera, esse quadrúpede despertou particular atenção por possuir uma bolsa no ventre onde carregava seus filhotes. Baseado nesse relato, Martin Waldseemüller incluiria uma imagem fantasiosa desse mamífero em sua célebre “Carta Marina” de 1516. Apesar de existirem figuras e notícias mais exatas já na primeira metade do século XVI, Martire d’Anghiera e Waldseemüller definiriam boa parte das narrativas e ilustrações posteriores, sendo reproduzidos ou adaptados em numerosas ocasiões. Sob o nome de “simivulpa” (i.e. “macaco-vulpino”), essa “besta prodigiosa” foi considerada um símbolo do continente americano ao longo de várias décadas, uma relação bem marcada no campo da cartografia. Background Koalas, an Australian arboreal marsupial, depend on eucalypt tree leaves for their diet. They selectively consume only a few of the hundreds of available eucalypt species. Since the koala gut microbiome is essential for the digestion and detoxification of eucalypts, their individual differences in the gut microbiome may lead to variations in their eucalypt selection and eucalypt metabolic capacity. However, research focusing on the relationship between the gut microbiome and differences in food preferences is very limited. We aimed to determine whether individual and regional differences exist in the gut microbiome of koalas as well as the mechanism by which these differences influence eucalypt selection. Methods Foraging data were collected from six koalas and a total of 62 feces were collected from 15 koalas of two zoos in Japan. The mitochondrial phylogenetic analysis was conducted to estimate the mitochondrial maternal origin of each koala. In addition, the 16S-based gut microbiome of 15 koalas was analyzed to determine the composition and diversity of each koala’s gut microbiome. We used these data to investigate the relationship among mitochondrial maternal origin, gut microbiome and eucalypt diet selection. Results and Discussion This research revealed that diversity and composition of the gut microbiome and that eucalypt diet selection of koalas differs among regions. We also revealed that the gut microbiome alpha diversity was correlated with foraging diversity in koalas. These individual and regional differences would result from vertical (maternal) transmission of the gut microbiome and represent an intraspecific variation in koala foraging strategies. Further, we demonstrated that certain gut bacteria were strongly correlated with both mitochondrial maternal origin and eucalypt foraging patterns. Bacteria found to be associated with mitochondrial maternal origin included bacteria involved in fiber digestion and degradation of secondary metabolites, such as the families Rikenellaceae and Synergistaceae. These bacteria may cause differences in metabolic capacity between individual and regional koalas and influence their eucalypt selection. Conclusion We showed that the characteristics (composition and diversity) of the gut microbiome and eucalypt diet selection of koalas differ by individuals and regional origins as we expected. In addition, some gut bacteria that could influence eucalypt foraging of koalas showed the relationships with both mitochondrial maternal origin and eucalypt foraging pattern. These differences in the gut microbiome between regional origins may make a difference in eucalypt selection. Given the importance of the gut microbiome to koalas foraging on eucalypts and their strong symbiotic relationship, future studies should focus on the symbiotic relationship and coevolution between koalas and the gut microbiome to understand individual and regional differences in eucalypt diet selection by koalas.

We report observations of Glironia venusta Thomas, 1912, Bushy-tailed Opossum, from coastal French Guiana, expanding the distribution of this species 300 km north of its previously known range.

The world’s largest extant carnivorous marsupial, the Tasmanian devil, is challenged by Devil Facial Tumor Disease (DFTD), a fatal, clonally transmitted cancer. In two decades, DFTD has spread across 95% of the species distributional range. A previous study has shown that factors such as season, geographic location, and infection with DFTD can impact the expression of immune genes in Tasmanian devils. To date, no study has investigated within-individual immune gene expression changes prior to and throughout the course of DFTD infection. To explore possible changes in immune response, we investigated four locations across Tasmania that differed in DFTD exposure history, ranging between 2 and >30 years. Our study demonstrated considerable complexity in the immune responses to DFTD. The same factors (sex, age, season, location and DFTD infection) affected immune gene expression both across and within devils, although seasonal and location specific variations were diminished in DFTD affected devils. We also found that expression of both adaptive and innate immune genes starts to alter early in DFTD infection and continues to change as DFTD progresses. A novel finding was that the lower expression of immune genes MHC-II, NKG2D and CD8 may predict susceptibility to earlier DFTD infection. A case study of a single devil with regressed tumor showed opposite/contrasting immune gene expression patterns compared to the general trends observed across devils with DFTD infection. Our study highlights the complexity of DFTD’s interactions with the host immune system and the need for long-term studies to fully understand how DFTD alters the evolutionary trajectory of devil immunity.

Introduction: During early development in most male mammals the testes move from a position near the kidneys through the abdomen to eventually reside in the scrotum. The transabdominal phase of this migration is driven by insulin-like peptide 3 (INSL3) which stimulates growth of the gubernaculum, a key ligament connecting the testes with the abdominal wall. While all marsupials, except the marsupial mole (Notoryctes typhlops), have a scrotum and fully descended testes, it is unclear if INSL3 drives this process in marsupials especially given that marsupials have a different mechanism of scrotum determination and position relative to the phallus compared to eutherian mammals.Methods: To understand if INSL3 plays a role in marsupial testicular descent we have sequenced and curated the INSL3 gene and its receptor (RXFP2) in a range of marsupials representing every order. Furthermore, we looked at single cell RNA-seq and qPCR analysis of INSL3 in the fat-tailed dunnart testis (Sminthopsis crassicaudata) to understand the location and timing of expression during development.Results: These data show a strong phylogenetic similarity between marsupial and eutherian orthologues, but not with monotreme INSL3s which were more similar to the ancestral RLN3 gene. We have also shown the genomic location of INSL3, and surrounding genes is conserved in a range of marsupials and eutherians. Single cell RNA-seq and qPCR data show that INSL3 mRNA is expressed specifically in Leydig cells and expressed at higher levels during the testicular descent phase in developing marsupials.Discussion: Together, these data argue strongly for a therian origin of INSL3 mediated testicular descent in mammals and suggests that a coordinated movement of the testes to the abdominal wall may have preceded externalization in marsupials and therian mammals.

The stripe-faced dunnart (Sminthopsis macroura) is one of the most widespread dasyurids in Australia, occurring mostly in semiarid and arid habitats. It is not known to inhabit coastal regions of southern Australia, and no records have previously been recorded from latitudes greater than 28.5°S in Western Australia. Following the capture of an individual south of the known species range provisionally identified based on external morphology as S. macroura , we used DNA and craniodental morphology to corroborate the specimen's identification, providing a record of the species at Eucla, Western Australia. This represents a large range extension for S. macroura of 630 km from the nearest confirmed records of the species in Western Australia and South Australia. A suspected stripe-faced dunnart (Sminthopsis macroura) was captured at a coastal dune site near Eucla, Western Australia. The identity of the specimen was corroborated through DNA sequencing and morphological examination. This represents a range extension into novel habitat for the species in southern Western Australia. Recent didelphid marsupials resemble the assumed mammalian ancestor and are suitable to inform on the evolution of the mammalian lung. This study uses X‐ray computed tomography (μCT) to three‐dimensionally reconstruct the bronchial tree of the marsupial Gray short‐tailed opossum (Monodelphis domestica) in order to reveal the timeline of morphogenesis during the postnatal period. The development of the bronchial tree was examined in 37 animals from embryonic day 13, during the postnatal period (neonate to 57 days) and in adults. The first appearance and the branching of lobar, segmental and sub‐segmental bronchioles in the lungs were documented. Based on the reconstructions, the generation of end‐branching airways, the median and maximum generation and the number of branches were calculated for each pulmonary lobe. At birth, the lung of M. domestica has a primitive appearance since it consists of a simple system of branching airways that end in a number of terminal air spaces, lobar bronchioles, and first segmental bronchioles are present. During the postnatal period, the volumes of the lung and bronchial tree steadily increase and development, differentiation, and expansion of the bronchial tree takes place. By 14 days, the fundamental bronchial tree consisting of lobar, segmental, and sub‐segmental bronchioles has been established. A mature bronchial tree, including respiratory bronchioles and alveolar ducts is present by day 35. The asymmetry of the right (predominately four lobes) and the left lung (predominately two lobes), as present in M. domestica, can be considered as plesiomorphic for Mammalia. In marsupials, the process of branching morphogenesis, which takes place intrauterine in the placental fetus, is shifted to the postnatal period, but follows similar patterns as described in placentals. Lung maturation in general and the branching morphogenesis in particular seems to be highly conservative within mammalian evolution. [ABSTRACT FROM AUTHOR]

Simple Summary: The gut microbiome can provide valuable information on the ecology and health of wildlife. We profiled the gut microbiota of the southern greater glider, an endangered Australian marsupial that was heavily disturbed by the 2019–2020 Australian bushfire season. Geographic location was found to be a significant driver of gut microbial community diversity and structure. In addition, the wildfires were shown to shape some aspects of the gut microbiome. By establishing baseline microbiome data for the southern greater glider, we lay the foundation for the future monitoring of populations at risk of compromised health, as well as inform future conservation management decisions for this endangered species. Studying the gut microbiome can provide valuable insights into animal health and inform the conservation management of threatened wildlife. Gut microbiota play important roles in regulating mammalian host physiology, including digestion, energy metabolism and immunity. Dysbiosis can impair such physiological processes and compromise host health, so it is essential that the gut microbiome be considered in conservation planning. The southern greater glider (Petauroides volans) is an endangered arboreal marsupial that faced widespread habitat fragmentation and population declines following the 2019–2020 Australian bushfire season. This study details baseline data on the gut microbiome of this species. The V3–V4 region of the 16S rRNA gene was amplified from scats collected from individuals inhabiting burnt and unburnt sites across southeastern Australia and sequenced to determine bacterial community composition. Southern greater glider gut microbiomes were characterised by high relative abundances of Firmicutes and Bacteroidota, which is consistent with that reported for other marsupial herbivores. Significant differences in gut microbial diversity and community structure were detected among individuals from different geographic locations. Certain microbiota and functional orthologues were also found to be significantly differentially abundant between locations. The role of wildfire in shaping southern greater glider gut microbiomes was shown, with some significant differences in the diversity and abundance of microbiota detected between burnt and unburnt sites. Overall, this study details the first data on greater glider (Petauroides) gut microbiomes, laying the foundation for future studies to further explore relationships between microbial community structure, environmental stressors and host health. To maximize energy savings, entry into torpor should involve a fast reduction of metabolic rate and body temperature (T b); that is, animals should thermoconform. However, animals often defend against the decrease in T b via a temporary increase in thermoregulatory heat production, slowing the cooling process. We investigated how thermoregulating or thermoconforming during torpor entry affects temporal and thermoenergetic aspects in relation to body mass and age in juvenile and adult fat-tailed dunnarts (Sminthopsis crassicaudata ; Marsupialia: Dasyuridae). During torpor entry, juvenile thermoconformers cooled twice as fast as and used less energy during cooling than juvenile thermoregulators. While both juvenile and adult thermoconformers had a lower minimum T b, a lower torpor metabolic rate, and longer torpor bouts than thermoregulators, these differences were more pronounced in the juveniles. Rewarming from torpor took approximately twice as long for juvenile thermoconformers, and the costs of rewarming were greater. To determine the difference in average daily metabolic rate between thermoconformers and thermoregulators independent of body mass, we compared juveniles of a similar size (∼13 g) and similarly sized adults (∼17 g). The average daily metabolic rate was 7% (juveniles) and 17% (adults) less in thermoconformers than in thermoregulators, even though thermoconformers were active for longer. Our data suggest that thermoconforming during torpor entry provides an energetic advantage for both juvenile and adult dunnarts and may aid growth for juveniles. While thermoregulation during torpor entry is more costly, it still saves energy, and the higher T b permits greater alertness and mobility and reduces the energetic cost of endogenous rewarming.
