8. The curious case of tryptophan in pregnancy

Abstract

Tryptophan-derived indoles L-Tryptophan is an uncommon amino acid in a protein. It is encoded by triplet UGG, the odd person out in the trio of translation-termination codons (UAA, UGA, and [...], For patients and caregivers to be fully informed about how living organ donation or prior kidney injury affects future health, we need to better understand the role of kidney reserve in physiological adaptation, especially during pregnancy. Importantly, epidemiological studies reason that live kidney donors are at increased risk for developing preeclampsia, a hypertensive disorder of pregnancy with serious implications for maternal and fetal health. Despite the import of this finding, the mechanistic basis for this increased risk is not understood. In this issue of the JCI, Dupont, Berg, and co-authors provide strong evidence that impaired placental perfusion, placental ischemia, increased soluble fms-like tyrosine kinase 1 (sFLT1), and a maternal preeclampsia-like phenotype are associated with an inability to upregulate the L-tryptophan-derived L-kynurenine pathway during pregnancy in mice with blunted renal reserve. These surprising revelations underscore the curious quiddity of L-tryptophan.

10. Theory and experiment of entanglement in a quasi-phase-matched two-crystal source

Abstract

We report new results regarding a source of polarization entangled photon-pairs created by the process of spontaneous parametric downconversion in two orthogonally oriented, periodically poled, bulk KTiOPO4 crystals (PPKTP). The source emits light colinearly at the non-degenerate wavelengths of 810 nm and 1550 nm, and is optimized for single-mode optical fiber collection and long-distance quantum communication. The configuration favors long crystals, which promote a high photon-pair production rate at a narrow bandwidth, together with a high pair-probability in fibers. The quality of entanglement is limited by chromatic dispersion, which we analyze by determining the output state. We find that such a decoherence effect is strongly material dependent, providing for long crystals an upper bound on the visibility of the coincidence fringes of 41% for KTiOPO4, and zero for LiNbO3. The best obtained raw visibility, when canceling decoherence with an extra piece of crystal, was 91 \pm 0.2%, including background counts. We confirm by a violation of the CHSH-inequality (S = 2.679 \pm 0.004 at 55 s^{-1/2} standard deviations) and by complete quantum state tomography that the fibers carry high-quality entangled pairs at a maximum rate of 55 x 10^3 s^{-1}THz^{-1}mW^{-1}., Comment: 12 pages, 10 figures, REVTeX4

Abstract

Measurements of spin relaxation processes were made in the technologically interesting InGaAs/InP quantum well system. The first non-linear pump and probe measurements were performed in a single 80Å lattice-matched InGaAs/InP quantum well layer. To make these measurements a synchronously-pumped parametric oscillator was constructed, using periodically-poled lithium niobate as its non-linear element. Investigations of dominant carrier species and their associated spin relaxation were made as a function of temperature. A model was constructed for comparison to the experimental data showing the dominance of excitons at low temperature, with un-bound electron-hole effects dominating at temperatures above 100K.

Abstract

Although mesenchymal stromal (stem) cell (MSC) administration attenuates sepsis-induced lung injury in pre-clinical models, the mechanism(s) of action and host immune system contributions to its therapeutic effects remain elusive. We show that treatment with MSCs decreased expression of host-derived microRNA (miR)-193b-5p and increased expression of its target gene, the tight junctional protein occludin (Ocln), in lungs from septic mice. Mutating the Ocln 3' untranslated region miR-193b-5p binding sequence impaired binding to Ocln mRNA. Inhibition of miR-193b-5p in human primary pulmonary microvascular endothelial cells prevents tumour necrosis factor (TNF)-induced decrease in Ocln gene and protein expression and loss of barrier function. MSC-conditioned media mitigated TNF-induced miR-193b-5p upregulation and Ocln downregulation in vitro When administered in vivo, MSC-conditioned media recapitulated the effects of MSC administration on pulmonary miR-193b-5p and Ocln expression. MiR-193b-deficient mice were resistant to pulmonary inflammation and injury induced by lipopolysaccharide (LPS) instillation. Silencing of Ocln in miR-193b-deficient mice partially recovered the susceptibility to LPS-induced lung injury. In vivo inhibition of miR-193b-5p protected mice from endotoxin-induced lung injury. Finally, the clinical significance of these results was supported by the finding of increased miR-193b-5p expression levels in lung autopsy samples from acute respiratory distress syndrome patients who died with diffuse alveolar damage.

Para caracterizar a clínica da leishmaniose tegumentar em crianças de 0 a 5 anos de idade, foram avaliadas, retrospectivamente, 4.464 fichas clínicas do Centro de Saúde de Corte de Pedra, Presidente Tancredo Neves, Bahia, Brasil, área endêmica de leishmaniose tegumentar americana, entre maio de 1987 e dezembro de 1995. Foram registrados neste período 4.275 casos novos de leishmaniose, dos quais, 491 (11,5%) correspondiam a crianças de 0 a 5 anos. A razão entre gênero masculino e feminino nas crianças foi 1,1:1.
