This research aimed to evaluate the adoption of Good Manufacturing Practices (GMP) for mussels and shrimp sold in street markets in the Grande Vitória Region, state of Espírito Santo, Brazil. The study included 11 street market, where 36 establishments (street stalls) were sampled. The assessment of sanitary and hygienic conditions was carried out using a checklist based on Resolução da Diretoria Colegiada (RDC) n° 216/2004 and RDC n° 275/2002. It was observed that 36 (100%) street stalls did not have exclusive handwashing sinks, a supply of potable water, equipment and utensils in proper condition, records of cleaning operations, and proper waste disposal. Only 18 (50%) establishments had handlers using aprons, and none of the handlers washed their hands or used gloves and caps. It is noteworthy that 27 (75%) establishments had flies and lacked protective barriers for products, and no establishment monitored the temperature of exposed products. No establishment had a Manual of Good Manufacturing Practices (GMP) and Standard Operating Procedures (SOP), although all had a sanitary permit. Overall, all establishments were classified as "poor," with a total average of 9.2% compliance and 90.8% non-compliance. It is concluded that the GMP adoption determined by current legislation are not being properly applied in the shellfish trade at street markets in the Grande Vitória Region. [ABSTRACT FROM AUTHOR]

Bovine cysticercosis (BCC) is an important disease in Brazil due to its detection in abattoirs, resulting in economic losses for cattle farmers, and a public health corcern. Besides its importance, the knowledge about BCC epidemiology and impact remains not fully described in several areas in this country, requiring more studies. Thus, this study focused on establishing BCC prevalence in the regions and municipalities of the state of Espírito Santo from 2017 to 2019, establishing the associated risk factors, and estimating the economic losses for cattle farmers. A set of 2,330 cases of BCC was detected in the 407,529 bovine inspected by Federal Inspection Service (0.57%; C.I. 95% 0.55 - 0.60%) with a higher detection of unviable cysticercus (66.14%). The beef producers lost at least US$153,000.20 due to BCC through this period. The highest risk for BCC infection occurred in Metropolitana (OR = 9.19), Litoral Sul (OR = 7.77), Caparó (OR = 7.44), and Central Sul (OR = 7.19) mesoregions, respectively. The BCC was detected in animals from 67 municipalities and its prevalence increased in areas with high human population density (OR = 1.58;; p = 0.01). In conclusion, BCC is an important disease for beef production chain in the Brazilian state of Espírito Santo and a cause of economic losses, mainly in some areas, and urgently requires the adoption of prophylactic strategies to reduce the ocurrence of this parasite. - Abstract
The genomes of 262 Bacillus cereus isolates were analyzed including 69 isolates sampled from equipment, raw milk and dairy products from Brazil. The population structure of isolates showed strains belonging to known phylogenetic groups II, III, IV, V and VI. Almost all the isolates obtained from dairy products belonged to group III. Investigation of specific alleles revealed high numbers of isolates carrying toxin-associated genes including cytK (53.62%), hblA (59.42%), hblC (44.93%), hblD (53.62%), nheA (84.06%), nheB (89.86%) and nheC (84.06%) with isolates belonging to groups IV and V having significant higher prevalence of hblACD and group IV of CytK genes. Strains from dairy products had significantly lower prevalence of CytK and hblACD genes compared to isolates from equipment and raw milk/bulk tanks. Genes related to sucrose metabolism were detected at higher frequency in isolates obtained from raw milk compared to strains from equipment and utensils. The population genomic analysis demonstrated the diversity of strains and variability of putative function among B. cereus group isolates in Brazilian dairy production, with large numbers of strains potentially able to cause foodborne illness. This detailed information will contribute to targeted interventions to reduce milk contamination and spoilage associated with B. cereus in Brazil.

Dermatophytosis is one of the most frequent anthropozoonoses on clinic of company animals, affecting about 4 to 15% of dogs and more than 20% of cats worldwide. It is also known that around 30% of infections in humans are of zoonotic cause, with transmission occuring through direct contact with infected animals, or indirectly, by the exposure to spores in the environment. Thus, the objective of the present manuscript was to perform an updated literature review about the zoonotic and public health aspects of dermatophytosis in small animals, due its highly contagious aspect and the increasing importance for public and animal health. This study focused on to verify the physical-chemical quality and the occurrence of fraud in heat treated milk by Ultra High Temperature (UHT) process produced in the state of São Paulo, Brazil and soldon the São Paulo Region Midwest. 63 samples of UHT milk were evaluated, 7 samples of different batches of 9 different brands through the physicochemical official analytical methods defined by the Ministry of Agriculture, Livestock and Supply. The parameters evaluated were: density at 15 ° C, acidity degree Dornic, fat, and total fat dry extract, freezing point and searches for fraudulent addition of chloride, starch addition, addition of acid reducers, addition of hydrogen peroxide, addition of formaldehyde, addition of chlorine and hypochlorite. Irregularities were observed only in relation to the percentage of fat, nonfat dry extract and the values of cryoscopic index in 31.74%, 34.92%, 22.22% of the samples, respectively. The surveyed fraudulent substances were not detected in this study.. It is concluded that non-compliances of the physicochemical parameters of UHT milk samples produced in the State of São Paulo were detected, demonstrating a need for greater surveillance to trade integrity products to consumers. This study focused on counting Staphylococcus spp. in curd cheeses "requeijão" and "especialidade láctea type requeijão" sold in Brazil, assessing the presence of mecA gene in obtained isolates and establishing antimicrobial resistance profile of the mecA gene positive isolates. To this, a set of 200 samples of these dairy products were evaluated. Low counts of Staphylococcus spp. were observed in these dairy products. All the isolates were determined as coagulase-negative strains using coagulase test and PCR. However, two isolates (3.70%) were carriers of mecA gene and they can be considered as risk for public health. These isolates presented resistance to penicillin, oxacillin and erythromycin. In conclusion, low counts of Staphylococcus were detected in curd cheese "requeijão" and "especialidade láctea type requeijão" sold in Brazil. However, coagulase-negative methicillin-resistant Staphylococcus spp. was detected in these dairy products. This is the first report of the detection of methicillin-resistant Staphylococcus spp. in heat-treated dairy products in Brazil. Results served as a warning to public sanitary authorities to control multidrug-resistant strains in veterinary and human medicine. [ABSTRACT FROM AUTHOR]

The UHT milk and milk beverage proteolysis during storage is the major factor that limits the shelf-life of these products. The GMP content has assumed importance because it can indirectly indicate the integrity of κ-casein in milk. So, this study focused on evaluates the κ-casein proteolysis in UHT milk and milk beverage during the shelf-life. 60 samples obtained of five different brands of each product were evaluated by spectrophotometric assay for quantifying acid sialic. For UHT milk was observed the values ranging from 3.83 to 15.27 μg acid sialic/ml at the begin of the shelf-life, from 5.95 to 17.16 mg sialic acid/ml at the middle of the period (60 days after the production) and 6.54 to 21.11 mg sialic acid/ml at the end of shelf-life (120 days). For milk beverages were observed ranges between 11.12 to 24.25, 15.74 to 24.73 μg of sialic acid/ml and from 17.19 to 26.49 mg sialic acid/ml respectively for the beginning, 60 and 120 days of the shelf-life. The proteolysis rate in UHT milk and milk beverage increased during the shelflife and is detectable after 60 days of the shelf-life. This study was focused on the determination of cysticercosis prevalence in swine and cattle carcasses in 56 slaughterhouses under supervision from Inspection Service of São Paulo State (SISP) between the years 2008 and 2013. Amongst the 1,305,723 pigs slaughtered, cysticercosis was present in only 125 (0.01%) of them in which, 116 (92.80%) were calcified. Nonetheless, cysticercosis was present in 24,278 (1.69%) of the 1,434,445 bovine carcasses evaluated; 19,826 (81.67%) were calcified. An inverse relationship (P <0.05) between time and the disease occurrence variable was observed. Sanitary preventive practices improvements, such as the Good Agricultural Practices, are required during the cattle rearing in order to decrease the cysticercosis occurrence to low prevalence levels as observed in swine rearing. Bovine mastitis is the most impacting disease of dairy industry, and it is characterized by a complexity of causal agents, which have revealed a geographical variation among regions and countries. The mastitis-related pathogens have been traditionally classified as contagious or environmental, based on habits of the microorganisms and transmission routes. Besides, the severity of mammary infections has been associated with the virulence of the pathogens, and immune and nutritional status of the hosts. Considering this scenario, we investigated the etiological pattern, clinical severity scores, and days in milk (DIM) data in 4,273 clinical cases of bovine mastitis among ten large-dairy farms located in Brazil. Streptococcus dysgalactiae (283/4,273 = 6.6%), Escherichia coli (190/4,273 = 4.4%), Prototheca spp. (112/4,273 = 2.6%), and Streptococcus uberis (95/4,273 = 2.2%) were the predominant pathogens isolated. Among 4,273 clinical cases, clinical severity score was available in 43.8% (1,871/4,273) animals. From these, 69.8% (1,306/1,871), 27.3% (510/1,871), and 2.9% (55/1,871) were scored as mild, moderate, and severe, respectively. Pathogens were observed mainly in samples obtained during the first 100 days in milk, and clinical severity scored mainly as mild. Our results contributed to the etiological identification, clinical severity scoring, and milking aspects of mastitic cows in dairy farms with a history of clinical mammary infections. Rabies is a viral encephalitis that affects mammals, including humans. Rapid and effective laboratory diagnosis of the rabies virus is critical for public health. This study evaluated the operational, technical, and financial viability of the RT-qPCR in replacement of inoculation in mice for diagnosing rabies in the laboratory routine. A total of 316 samples of mammalian brains were analyzed, 121 positives and 195 negatives, previously diagnosed by direct immunofluorescence (DIF) and mouse inoculation test (MIT). The samples were submitted to the duplex TaqMan RT-qPCR technique. The accuracy, sensitivity, and specificity of RT-qPCR were analyzed. We analyzed the costs for performing the RT-qPCR technique and compared it with the cost of MIT. The results showed 99.37% accuracy, 99.17% sensitivity, and 99.49% specificity by RT-qPCR when related to DIF and MIT results, which proved to be a robust and repeatable technique. The minimum time for a positive diagnosis was reduced in RT-qPCR (1 day) if compared to MIT (9.64 days), with a 17.7% reduction in the cost of the molecular technique. The present study demonstrated that the molecular biology technique is an efficient tool to diagnose rabies in the laboratory routine, being able to replace MIT. [ABSTRACT FROM AUTHOR]

Bovine fasciolosis is a neglected parasitic zoonosis that causes economic losses to beef production chains due to liver condemnation at abattoirs. Despite the characterization of Bovine fasciolosis in several areas of Brazil, an update on its prevalence in the states of Rio de Janeiro and Espirito Santo is lacking. Thus, fasciolosis prevalence from 2018 to 2021 is presented here according to cattle origin (municipalities of both states). In addition, the associated economic impact on an abbatoir was analyzed through liver condemnation of bovines from both states. The prevalence of fasciolosis in the state of Espírito Santo (10.4%) was significantly higher than Rio de Janeiro (3.6%) (P < 0.01), and resulted in an economic loss of US$12,678.60 for the abattoir. The following municipalities located in the state of Espírito Santo presented prevalences above the mean (10.3%) established in the study: Vargem Alta, Anchieta, Cachoeiro do Itapemirim, Castelo, Atilio Vivacqua, Apiacá and Ibitirama. Thus, majority of these municipalities are considered high-risk areas. Although, there is an aparrent reduction of fasciolosis prevalence, it continues to be a cause of economic losses in abattoirs especially in the state of Espírito Santo. With the objective to evaluate the hygienic and sanitary conditions of grated parmesan cheese acquired from the retail business, total 120 cheese samples were acquired: 60 of which were obtained from four different brands of cheese that were grated in factories, and the other 60 samples were obtained from another four brands of cheese that are normally acquired by retailers (supermarkets) in blocks and grated at the moment of sale. The population of heterotrophic mesophilic microorganisms ranged from 1.2×10³ to 1.1×107 colony-forming units (CFU)·g-1. All samples analyzed contained Staphylococcus spp. with populations varying from 1.2×10³ to 8.7×106CFU·g-1, from which 60.0% were classified as coagulase-positive Staphylococcus and 52.5% of the samples possessed populations above the permissible limit set by legislation. Staphylococcus aureus was detected in 57.5% of the samples. Population of molds and yeasts varied from <10 to 1.8×106CFU·g-1. Salmonella spp. was not isolated in this research. A difference was observed between the parmesan cheese grated in factories and that grated in supermarkets, where the former presented better microbiological quality than the latter. Thus, procedures must be proposed to minimize the presence of pathogenic agents reported in grated parmesan cheeses evaluated in the present study because of the public health risk associated with food bacterial contamination. The present study focused on isolation Bacillus cereus during the UHT milk production and shelf life, to assess the enterotoxigenic production capacity of isolates and to evaluate the use of the RAPD-PCR technique to verify whether Bacillus cereus isolated at different phases of UHT milk processing belongs to the same strain. For this, six groups of milk samples composed of raw, pasteurized and UHT milk were collected from a processing plant. The results revealed that bacteria belonging to the Bacillus cereus group were isolated from 51.6%, 81.6% and from 13.8% of raw, pasteurized and UHT milk samples, respectively. About 50.0% of isolates from raw milk, 19.2% isolates from pasteurized milk and 70.7% isolates from UHT milk were capable of producing enterotoxins. It was confirmed the genetic similarity among Bacillus cereus isolates from raw, pasteurized and UHT milk, therefore demonstrating that the microorganism is able to withstand UHT treatment. These results should serve as a warning to health authorities, given that 13.8% of samples were not in accordance with standards established by the Department of Health for containing a potentially pathogen agent, therefore indicating that contamination of milk by sporulating bacteria should be avoided.
