Dans cet article, nous présentons une étude micro-mécanique de l’effet du pourcentage de particules fines sur le comportement mécanique des matériaux granulaires à granulométrie lacunaire en utilisant la simulation numérique réalisée avec la Méthode des Éléments Discrets. Différents échantillons ayant un pourcentage de particules fines variant de 0% à 30% sont simulés. Cette étude montre que le pourcentage de particules fines influence significativement la résistance au cisaillement et la dilatance des échantillons granulaires. Lorsque le pourcentage de particules fines augmente, des particules fines viennent en contact avec des particules grossières and renforcent les dernières. Within inshore or fluvial environments, submerged fine matter mud banks are characterized by a high water content, a great spatial variability, and a strong deformability. The study of their instabilities induced by the variation of hydraulic stress requires a coupled modeling of sliding, erosion, and deposition mechanisms. In order to predict the impact of dam reservoir emptying on the stability of immersed upstream slopes, the method of approach to the problem proposed here combines theoretical developments, numerical modeling, site observations, mid measurements. First, the theoretically achieved sliding criterion is compared with unstable mud height measurements. For more accuracy in the representation of the natural events, the sliding criterion is then integrated within a numerical code which couples the computation of hydrodynamic conditions, the erosion, and deposition of mud and the banks sliding. Finally, the results of the combination of all these mechanisms are compared with the variations in the bathymetric profiles obtained on the experimental site. Une patiente née en 1957 est adressée pour élévation majeure de l’ACTH plasmatique (>2000ng/mL N<60ng/mL) demandé devant une pigmentation cutanée acquise. La patiente présente une forme extrêmement rare de porphyrie, une coproporphyrie héréditaire à l’état homozygote diagnostiquée à l’âge de 10 ans devant des douleurs abdominales et une hyponatrémie à 113mmol/L. Elle évolue sans suivi, ni crise aiguë depuis de nombreuses années avec une fatigue chronique.

La surverse et l’érosion interne apparaissent comme les deux principales causes de la rupture des digues et barrages en remblais. Quatre processus d’érosion interne peuvent être distingués et cette étude porte sur la suffusion qui mobilise la fraction fine du sol. En considérant uniquement la distribution granulométrique et éventuellement la densité du sol, de nombreux critères granulométriques ont été proposés dans la littérature. Toutefois la suffusion étant une interaction fluide-solide, il convient évidemment de prendre aussi en compte la sollicitation hydraulique. Par ailleurs, la filtration de certaines particules détachées lors de la suffusion peut induire un colmatage du milieu poreux et donc une variation de la pression interstitielle et du débit d’écoulement. En conséquence, la sollicitation hydraulique est modélisée par la puissance dissipée par le fluide. Les différentes campagnes de caractérisation expérimentale de la suffusion décrites dans la littérature, ont été menées sur des échantillons de tailles très diverses et différents historiques de chargement hydraulique qui peuvent être très différents des conditions de sollicitation in-situ. Enfin les essais de suffusion ont été très majoritairement réalisés sous écoulement vertical alors que sur site, la direction de l’écoulement peut être bien différente. Le premier objectif de l’étude menée est donc de s’assurer que la caractérisation de la sensibilité à la suffusion n’est pas significativement affectée par des effets d’échelles spatiale et temporelle. A cause de la diversité des sols qui constituent les ouvrages hydrauliques et leurs fondations et de l’importance du linéaire de ces ouvrages, la sensibilité à la suffusion à l’échelle d’un ouvrage est extrêmement variable. Le deuxième objectif de l’étude est donc d’estimer à partir de paramètres physiques aisément mesurables la sensibilité des sols à la suffusion. Enfin le troisième objectif est d’identifier les zones les plus sensibles d’un ouvrage en confrontant la sensibilité des sols avec la distribution spatiale de la puissance.

The electron temperature and density pedestals tend to vary in their relative radial positions, as observed in DIII-D (Beurskens et al 2011 Phys. Plasmas 18 056120) and ASDEX Upgrade (Dunne et al 2017 Plasma Phys. Control. Fusion 59 14017). This so-called relative shift has an impact on the pedestal magnetohydrodynamic (MHD) stability and hence on the pedestal height (Osborne et al 2015 Nucl. Fusion 55 063018). The present work studies the effect of the relative shift on pedestal stability of JET ITER-like wall (JET-ILW) baseline low triangularity (δ) unseeded plasmas, and similar JET-C discharges. As shown in this paper, the increase of the pedestal relative shift is correlated with the reduction of the normalized pressure gradient, therefore playing a strong role in pedestal stability. Furthermore, JET-ILW tends to have a larger relative shift compared to JET carbon wall (JET-C), suggesting a possible role of the plasma facing materials in affecting the density profile location. Experimental results are then compared with stability analysis performed in terms of the peeling-ballooning model and with pedestal predictive model EUROPED (Saarelma et al 2017 Plasma Phys. Control. Fusion). Stability analysis is consistent with the experimental findings, showing an improvement of the pedestal stability, when the relative shift is reduced. This has been ascribed mainly to the increase of the edge bootstrap current, and to minor effects related to the increase of the pedestal pressure gradient and narrowing of the pedestal pressure width. The dependence of plasma transport and confinement on the main hydrogenic ion isotope mass is of fundamental importance for understanding turbulent transport and, therefore, for accurate extrapolations of confinement from present tokamak experiments, which typically use a single hydrogen isotope, to burning plasmas such as ITER, which will operate in deuterium-tritium mixtures. Knowledge of the dependence of plasma properties and edge transport barrier formation on main ion species is critical in view of the initial, low-activation phase of ITER operations in hydrogen or helium and of its implications on the subsequent operation in deuterium-tritium. The favourable scaling of global energy confinement time with isotope mass, which has been observed in many tokamak experiments, remains largely unexplained theoretically. Moreover, the mass scaling observed in experiments varies depending on the plasma edge conditions. In preparation for upcoming deuterium-tritium experiments in the JET tokamak with the ITER-like Be/W Wall (JET-ILW), a thorough experimental investigation of isotope effects in hydrogen, deuterium and tritium plasmas is being carried out, in order to provide stringent tests of plasma energy, particle and momentum transport models. Recent hydrogen and deuterium isotope experiments in JET-ILW on L-H power threshold, L-mode and H-mode confinement are reviewed and discussed in the context of past and more recent isotope experiments in tokamak plasmas, highlighting common elements as well as contrasting observations that have been reported. The experimental findings are discussed in the context of fundamental aspects of plasma transport models.

Erosion of plasma-facing materials and successive transport and redeposition of eroded material are crucial processes determining the lifetime of plasma-facing components and the trapped tritium inventory in redeposited material layers. Erosion and deposition in the JET divertor were studied during the second JET ITER-like wall campaign ILW-2 in 2013-2014 by using a poloidal row of specially prepared divertor marker tiles including the tungsten bulk tile 5. The marker tiles were analyzed using elastic backscattering with 3-4.5 MeV incident protons and nuclear reaction analysis using 0.8-4.5 MeV 3He ions before and after the campaign. The erosion/deposition pattern observed during ILW-2 is qualitatively comparable to the first campaign ILW-1 in 2011-2012: deposits consist mainly of beryllium with 5-20 at.% of carbon and oxygen and small amounts of Ni and W. The highest deposition with deposited layer thicknesses up to 30 μm per campaign is still observed on the upper and horizontal parts of the inner divertor. Outer divertor tiles 5, 6, 7 and 8 are net W erosion areas. The observed D inventory is roughly comparable to the inventory observed during ILW-1. The results obtained during ILW-2 therefore confirm the positive results observed in ILW-1 with respect to reduced material deposition and hydrogen isotopes retention in the divertor.

Post mortem analyses of JET ITER-Like-Wall tiles and passive diagnostics have been completed after each of the first two campaigns (ILW-1 and ILW-2). They show that the global fuel inventory is still dominated by co-deposition; hence plasma parameters and sputtering processes affecting material migration influence the distribution of retained fuel. In particular, differences between results from the two campaigns may be attributed to a greater proportion of pulses run with strike points in the divertor corners, and having about 300 discharges in hydrogen at the end of ILW-2. Recessed and remote areas can contribute to fuel retention due to the larger areas involved, e.g. recessed main chamber walls, gaps in castellated Be main chamber tiles and material migration to remote divertor areas. The fuel retention and material migration due to the bulk W Tile 5 during ILW-1 are presented. Overall these tiles account for only a small percentage of the global accountancy for ILW-1.

© 2017 Technical University of Denmark. We demonstrate the measurement of a 2D MeV-range ion velocity distribution function by velocity-space tomography at JET. Deuterium ions were accelerated into the MeV-range by third harmonic ion cyclotron resonance heating. We made measurements with three neutron emission spectrometers and a high-resolution γ-ray spectrometer detecting the γ-rays released in two reactions. The tomographic inversion based on these five spectra is in excellent agreement with numerical simulations with the ASCOT-RFOF and the SPOT-RFOF codes. The length of the measured fast-ion tail corroborates the prediction that very few particles are accelerated above 2 MeV due to the weak wave-particle interaction at higher energies.

The risk assessment of internal erosion made by Erinoh is reported in three steps: 1. laboratory tests: main mechanisms of internal erosion are evaluated in lab tests on intact or compacted samples, 2. methods of detection on site : the flaws or defects on site (leaks, cavities, other anomalies) are localized and sometimes monitored by accurate methods of detection, 3. engineering methods : the risk assessment is finally driven by functional analysis, behaviour assessment and engineering rules based on filter criteria and erosion thresholds. Every step is described in a handbook. This report presents some outputs of the Erinoh Project and invites the practitioners to read the 3 handbooks for applications.; Ce rapport traite de l'érosion interne. En France, chaque année un ouvrage hydraulique est victime d'une rupture par érosion interne. La réduction de ce risque de rupture par érosion interne fut l'enjeu d'un projet national de recherche appelé Erinoh. - Abstract
Suffusion, one of the main internal erosion processes, selectively erodes the fine particles which move through the voids formed by the coarser particles. In the literature, the mechanical consequences of suffusion on soil were investigated by several researchers. However, no general conclusion can be drawn because the already published results appear to be opposite. The objective of this study is to investigate these consequences. A large triaxial erodimeter was developed in order to perform suffusion tests and triaxial tests on coarse soils which are often encountered in French dikes. For the suffusion characterisation, the proposed interpretative method is based on the energy expended by the seepage flow and the cumulative loss of dry mass. The results of triaxial tests on non-eroded and eroded specimens show the significant effect of an achieved suffusion process on the mechanical strength of soils.

International audience; Ensuring the safety of hydraulic embankment structures, including embankment dams, levees and dykes, is a major concern all over the world. Still today, about one to two large dams fail every year in the world and hundreds, probably thousands, of dykes and levees fail every year in the world. Embankment hydraulic structures can be subject to three potential failure modes: instability, internal erosion and external erosion. The physical parameters driving instability are the pore pressures inside the embankment and its foundation. This failure mode covers both general slope sliding and instabilities due to seismic loadings. Statistics show that 6% of the failure of large embankment dams are due to that process. Internal erosion can be initiated in the embankment and/or in its foundation by four several types of processes: crack erosion, contact erosion, regressive erosion and suffusion. Crack erosion and regressive erosion can lead to one of the most sudden and dangerous failure mode: piping. Statistics show that 46% of the failures of large embankment dams are due to that process. External erosion is due to the detachment of the embankment material by the overtopping flow. Erosion of the upstream toe of dykes and levees can also lead to failures. Statistics show that 48% of the failures of large embankment dams are due to these processes of external erosion. Study of real cases of embankment dam failures and researches performed in the last two decades on hydraulic embankments erosion both show that breach parameters (such as the breach peak outflow, the final breach length or the breach opening duration) can not be correctly predicted only by geometrical parameters of the embankment and the upstream reservoir. Taking into account the behaviour of the embankment and foundation material under the flow stress, that is called the material erodibility, is now considered essential for safety assessments of that type of structures.Different experimental devices, called erodimeters, have been developed for sediment transport or embankment issues. Their use is always based on an assumption of an erosion law, which is the relation between the rate of erosion (i.e. quantity of eroded material per time unit per surface unit) and parameters characterizing the flow and the soil material.Two erodimeters have been selected for the assessment of the erodibility of hydraulic embankment structures, based on the physical processes they are representing and their practical characteristics. These two erodimeters are the Jet Erosion Test and the Hole Erosion Test. After presenting the physical principles of these two tests and the modelling upon which they are based, their engineering application is illustrated as well as the research needs that remain.

- Abstract
Low-protein, low-phosphorus diets (LPD) are prescribed to patients with chronic renal failure (CRF) to slow down the rate of progression of CRF and to control uraemic symptoms. A satisfactory adherence of patients to the prescribed diet is needed to meet these two goals. We studied the compliance of CRF patients to a LPD providing daily (per kg body weight) 0.3 g protein, 3–5 mg phosphorus, 35 kcal, and supplemented with essential amino-acids and ketoanalogues. Forty CRF patients were studied for 23.3±10.8 months (range 12–45). Compliance to LPD was evaluated by dietary inquiry and protein intake estimated from urinary urea excretion. According to compliance to LPD, patients were retrospectively assigned to the compliant (=27) or the non-compliant (=13) group. GFR measured by the urinary clearance of [Cr]-EDTA was identical in the two groups at the start of the study: compliant patients 15.7±5.3 ml/mn, non-compliant patients 15.4±5.9 ml/mn. The decrease of GFR was − 0.08±0.22 ml/min per month in compliant patients versus −0.31±0.37 ml/min per month in non-compliant patients (<0.02). These results were not demonstrated if the progression of CRF was assessed by the linear regressions over time of creatinine clearance or the reciprocal of creatinine. Serum bicarbonate, serum phosphorus and PTH levels were corrected by LPD in compliant patients (< 0.005 for all parameters) but not in non-compliant patients. These results suggest that evaluation of compliance is necessary to assess the response of CRF patients to LPD, whether the aim is to slow the progression of CRF or to control its metabolic consequences. A beneficial effect of compliance to LPD was demonstrated upon these two goals. [ABSTRACT FROM PUBLISHER]

International audience; In this paper, we present a coupled Discrete Element (DE) and Lattice Boltzmann (LB) method to model fluid-solid interactions. This method is applied to study and model a two-dimensional piping erosion through a set-up inspired by the Hole Erosion Test (HET). In this work, we mainly focus on grain detachments under hydraulic loading. Simulation results show that the erosion law classically used in such experiments, can be retrieved with our model. It illustrates that such coupled systems can be investigated with the LB-DE approach developed in this work. This approach is a promising tool which is well designed to perform such investigations at the grain scale

- Abstract
Rationale: Daily oral synthetic levothyroxine (LT4) is the main treatment for hypothyroidism, which, in most cases, allows the regression of symptoms and the normalization of the thyroid function. However, rarely, despite a high dose of oral LT4, hypothyroidism persists and is called refractory hypothyroidism. Intravenous or intramuscular treatment is then often necessary. We report the case of a patient with refractory hypothyroidism successfully treated with subcutaneous LT4., Interventions and Outcomes: After 4 weeks of weekly intravenous injections of 200 µg LT4 in complement to the oral treatment, thyroid balance was improved (TSH: 21.8 mIU/L). We tested the replacement of intravenous with subcutaneous injections of LT4 and gradually increased injection frequency from 1 to 3 injections per week (600 µg/week). Simultaneously, oral treatment was gradually tapered off, and within a few months, thyroid function tests were normalized. Two years later, hormone levels remained normal without symptoms of hypothyroidism. The only side effect was a local reaction in the first few weeks of injections, which spontaneously resolved., Lessons: In this case of unexplained oral LT4 malabsorption, subcutaneous injection allowed a self-administrated physiological dose of LT4 3 times weekly. We report the case of an alcoholic patient with severe pancytopenia with low plasma folate level but normal erythrocyte folates and cobalamin levels. The bone marrow smear concluded to a pancytopenia due to folates and/or cobalamin deficiency. Severe pancytopenia due to acute plasma folate deficiency can be observed despite normal erythrocyte folates level which reflects the organism's folates store.

- Abstract
KiSS1 is a putative metastasis suppressor gene in melanoma and breast cancer-encoding kisspeptins, which are also described as neuroendocrine regulators of the gonadotropic axis. Negative as well as positive regulation of KiSS1 gene expression by estradiol (E(2)) has been reported in the hypothalamus. Estrogen receptor alpha (ERalpha level is recognized as a marker of breast cancer, raising the question of whether expression of KiSS1 and its G-protein-coupled receptor (GPR54) is down- or upregulated by estrogens in breast cancer cells. KiSS1 was found to be expressed in MDA-MB-231, MCF7, and T47D cell lines, but not in ZR75-1, L56Br, and MDA-MB-435 cells. KiSS1 mRNA levels decreased significantly in ERalpha-negative MDA-MB-231 cells expressing recombinant ERalpha. In contrast, tamoxifen (TAM) treatment of ERalpha-positive MCF7 and T47D cells increased KiSS1 and GPR54 levels. The clinical relevance of this negative regulation of KiSS1 and GPR54 by E(2) was then studied in postmenopausal breast cancers. KiSS1 mRNA increased with the grade of the breast tumors. ERalpha-positive invasive primary tumors expressed sevenfold lower KiSS1 levels than ERalpha-negative tumors. Among ERalpha-positive breast tumors from postmenopausal women treated with TAM, high KiSS1 combined with high GPR54 mRNA tumoral levels was unexpectedly associated with shorter relapse-free survival (RFS) relative to tumors expressing low tumoral mRNA levels of both genes. The contradictory observation of putative metastasis inhibitor role of kisspeptins and RFS to TAM treatment suggests that evaluation of KiSS1 and its receptor tumoral mRNA levels could be new interesting markers of the tumoral resistance to anti-estrogen treatment.

- Abstract
BRCA1 is a central component of the DNA damage response mechanism and defects in BRCA1 confer sensitivity to a broad range of DNA damaging agents. BRCA1 is required for homologous recombination and DNA damage-induced S and G(2)/M phase arrest. We show here that BRCA1 is required for ATM- and ATR-dependent phosphorylation of p53, c-Jun, Nbs1 and Chk2 following exposure to ionizing or ultraviolet radiation, respectively, and is also required for ATM phosphorylation of CtIP. In contrast, DNA damage-induced phosphorylation of the histone variant H2AX is independent of BRCA1. We also show that the presence of BRCA1 is dispensable for DNA damage-induced phosphorylation of Rad9, Hus1 and Rad17, and for the relocalization of Rad9 and Hus1. We propose that BRCA1 facilitates the ability of ATM and ATR to phosphorylate downstream substrates that directly influence cell cycle checkpoint arrest and apoptosis, but that BRCA1 is dispensable for the phosphorylation of DNA-associated ATM and ATR substrates.

- Abstract
BRCA1 plays an important role in mechanisms of response to double-strand breaks, participating in genome surveillance, DNA repair, and cell cycle checkpoint arrests. Here, we identify a constitutive BRCA1-c-Abl complex and provide evidence for a direct interaction between the PXXP motif in the C terminus of BRCA1 and the SH3 domain of c-Abl. Following exposure to ionizing radiation (IR), the BRCA1-c-Abl complex is disrupted in an ATM-dependent manner, which correlates temporally with ATM-dependent phosphorylation of BRCA1 and ATM-dependent enhancement of the tyrosine kinase activity of c-Abl. The BRCA1-c-Abl interaction is affected by radiation-induced modification to both BRCA1 and c-Abl. We show that the C terminus of BRCA1 is phosphorylated by c-Abl in vitro. In vivo, BRCA1 is phosphorylated at tyrosine residues in an ATM-dependent, radiation-dependent manner. Tyrosine phosphorylation of BRCA1, however, is not required for the disruption of the BRCA1-c-Abl complex. BRCA1-mutated cells exhibit constitutively high c-Abl kinase activity that is not further increased on exposure to IR. We suggest a model in which BRCA1 acts in concert with ATM to regulate c-Abl tyrosine kinase activity.

- Abstract
The loss of BRCA1 function appears as an essential step in breast and ovarian epithelial cells oncogenesis and is consistent with the concept that BRCA1 acts as a tumor suppressor gene. However, the mechanism underlying this activity is not understood. In 1996, a retroviral vector was used for BRCA1 delivery to demonstrate that the transfer of BRCA1 inhibits breast and ovarian cancer cell growth. Since this early observation, the tumor growth inhibitory activity of BRCA1 in vivo has not been further documented. Here we re-address this issue and report experiments designed to evaluate the potential of adenovirus-mediated BRCA1 delivery to suppress the growth of cells with various status of endogenous BRCA1 in comparison with p53 and p21. Delivery of wild-type BRCA1 by an adenovirus vector in breast and ovarian tumor cells, decreases in vitro proliferation and tumorigenicity. Similarly, in vivo administration of BRCA1 provokes tumor growth retardation or regression comparable to that obtained with p53 or p21. The antitumor effect of BRCA1 is not observed upon transfer of a mutant lacking the 542 C-terminal residues. The p53- or p21-mediated antiproliferative activities are likely to bear on their capacity to induce apoptosis and/or interfere with cell cycle checkpoint. By contrast, the data presented here show that neither of these mechanisms can account for the BRCA1-mediated antitumor activity and suggest the activation of an alternative route.

- Abstract
The breast and ovarian cancer susceptibility genes, BRCA1 and BRCA2, are likely to participate in DNA lesion processing. Oxidative lesions, such as 8-oxoguanine, occur in DNA after endogenous or exogenous oxidative stress. We show that deficiency for either BRCA1 or BRCA2 in human cancer cells leads to a block of the RNA polymerase II transcription machinery at the 8-oxoguanine site and impairs the transcription-coupled repair of the lesion, leading to a high mutation rate. Expression of wild-type BRCA1 from a recombinant adenovirus fully complements the repair defect in BRCA1-deficient cells. These results represent the first demonstration of the essential contribution of BRCA1 and BRCA2 gene products in the repair of the 8-oxoguanine oxidative damage specifically located on the transcribed strand in human cells. This suggests that cells from individuals predisposed to breast and/or ovarian cancer may undergo a high rate of mutations because of the deficiency of this damage repair pathway after oxidative stress.

There is now evidence to suggest that BRCA1 and BRCA2 are involved in the response of cells to DNA damage and cell cycle checkpoint control. This report examines the death pathways of human cells with various BRCA1 and BRCA2 genotypes after exposure to gamma-rays. A lack of functional BRCA1 and BRCA2 led to defective repair of DNA double-strand breaks in irradiated cells. This impairment resulted in a relaxation of cell cycle checkpoints, production of micronuclei, and a loss of proliferative capacity. Heterozygous BRCA1 and BRCA2 mutations also led to enhanced radiosensitivity, with an impaired proliferative capacity after irradiation. The existence of a phenotype related to radiosensitivity in BRCA1+/- and BRCA2+/- cells raises the question of the response of heterozygous women to radiation.

- Abstract
1. Since oxygen free radicals are directly involved in a variety of pathologies such as atherosclerosis, diabetes mellitus, inflammation and/or when a deficit of defences of the organism against radicals occurs, we developed a suitable and simple method to determine both the erythrocyte sensitivity to an oxidative stress and plasma antioxidant protective capacity. 2. This test is based on the introduction at 37 degrees C of a radical initiator, 2,2'-azobis (2-amidinopropane) dihydrochloride (AAPH), within an erythrocyte suspension leading to a membrane alteration and ultimately to haemolysis. The latter can be quantified by determining the lacticodeshydrogenase activity released in the medium. The erythrocyte sensitivity to haemolysis and the volume of plasma inhibiting 50% of the haemolysis were determined. 3. Intra-assay CVs were 1.9% for erythrocyte sensitivity to oxidative stress and 3.4% for inhibitory 50% plasma volume. Inter-assay CVs for both erythrocyte sensitivity and inhibitory 50% plasma volume were 4%. 4. The reliability of this method was assessed and applied to test the protective effect of vitamin E, a well known antioxidant agent, in six healthy volunteers. Two weeks after daily administration of 500 mg of vitamin E, the mean plasma vitamin E concentration increased by 41% from 10.7 +/- 2.0 mg l-1 before treatment (P < 0.05). As the vitamin E concentration increased, the mean inhibitory 50% plasma volume and the percentage of haemolysed erythrocytes decreased respectively by 29% from 3.35 +/- 0.5 microliter (P < 0.05) and 18% from 71.5 +/- 3.8% (P < 0.05). No significative variation of these parameters was observed in six adult men without vitamin E supplementation. 5. Thus, this global and simple test permits an antioxidant status evaluation of a patient. It can be applied to various pathologies and allows the potency of new antioxidant molecules to be evaluated.
