Keratin is an important byproduct of the animal industry, but almost all of it ends up in landfills due to a lack of efficient recycling methods. To make better use of keratin-based natural resources, the current extraction and processing strategies need to be improved or replaced by more sustainable and cost-effective processes. Here, we developed a simple and environmentally benign method to process extracted keratin, using HCl to induce the formation of a coacervate, a separate aqueous phase with a very high protein concentration. Remarkably, this pH-induced coacervation did not result in the denaturation of keratin, and we could even observe an increase in the amount of ordered secondary structures. The low-pH coacervates could be extruded and wet-spun into high-performance keratin fibers, without requiring heating or any organic solvents. The secondary structure of keratin was largely conserved in these regenerated fibers, which exhibited excellent mechanical performance. The process developed in this study represents a simple and environmentally friendly strategy to upcycle waste keratin into high-performance materials.

Keratin is one of the most abundant biopolymers, produced on a scale of millions of tons per year but often simply discarded as waste. Due to its abundance, biocompatibility, and excellent mechanical properties, there is an extremely high interest in developing protocols for the recycling of keratin and its conversion into protein-based materials. In this work, we describe a novel protocol for the conversion of keratin from wool into hybrid fibers. Our protocol uses a synthetic polyanion, which undergoes complex coacervation with keratin, leading to a viscous liquid phase that can be used directly as a dope for dry-spinning. The use of polyelectrolyte complexation allows us to use all of the extracted keratin, unlike previous works that were limited to the fraction with the highest molecular weight. The fibers prepared by this protocol show excellent mechanical properties, humidity responsiveness, and ion conductivity, which makes them promising candidates for applications as a strain sensor.

Because of their permanent charge, strong polyelectrolytes remain challenging to characterize, in particular, when they are combined with hydrophobic features. For this reason, they are typically prepared through a postmodification of a fully hydrophobic precursor. Unfortunately, these routes often result in an incomplete functionalization or otherwise require harsh reaction conditions, thus limiting their applicability. To overcome these problems, in this work a strategy is presented that facilitates the preparation of well-defined strong polyanions by starting from protected 3-sulfopropyl methacrylate monomers. Depending on the chemistry of the protecting group, the hydrophobic precursor could be quantitatively converted into a strong polyanion under nucleophilic, acidic, or basic conditions. As a proof of concept, orthogonally protected diblock copolymers were synthesized, selectively deprotected, and allowed to self-assemble in aqueous solution. Further conversion into a fully water-soluble polyanion was achieved by deprotecting the second block as well.

Encapsulation of proteins can have advantages for their protection, stability, and delivery purposes. One of the options to encapsulate proteins is to incorporate them in complex coacervate core micelles (C3Ms). This can easily be achieved by mixing aqueous solutions of the protein and an oppositely charged neutral-hydrophilic diblock copolymer. However, protein-containing C3Ms often suffer from salt-inducible disintegration due to the low charge density of proteins. The aim of this study is to improve the salt stability of protein-containing C3Ms by increasing the net charge of the protein by tagging it with a charged polypeptide. As a model protein, we used CotA laccase and generated variants with 10, 20, 30, and 40 glutamic acids attached at the C-terminus of CotA using genetic engineering. Micelles were obtained by mixing the five CotA variants with poly(N-methyl-2-vinyl-pyridinium)-block-poly(ethylene oxide) (PM2VP128-b-PEO477) at pH 10.8. Hydrodynamic radii of the micelles of approximately 31, 27, and 23 nm for native CotA, CotA-E20, and CotA-E40, respectively, were determined using dynamic light scattering (DLS) and fluorescence correlation spectroscopy (FCS). The encapsulation efficiency was not affected using enzymes with a polyglutamic acid tail but resulted in more micelles with a smaller number of enzyme molecules per micelle. Furthermore, it was shown that the addition of a polyglutamic acid tail to CotA indeed resulted in improved salt stability of enzyme-containing C3Ms. Interestingly, the polyglutamic acid CotA variants showed an enhanced enzyme activity. This study demonstrates that increasing the net charge of enzymes through genetic engineering is a promising strategy to improve the practical applicability of C3Ms as enzyme delivery systems.

3D printing of hydrogels usually relies on a combination of fine-tuned material chemistry and polymer chain architecture to obtain an ink with adequate yield-stress flow, shear-thinning, and self-healing behavior. Recent approaches in hydrogel ink design include introduction of reversible covalent or supramolecular bonds or jamming of microparticles into a granular hydrogel. However, the dimensional stability of such systems is typically afforded by a post-printing covalent cross-linking step that impedes further on-demand degradation of the scaffolds. Here, a jammed micro-gels ink made of thermosensitive poly(N-isopropylacrylamide) micro-gels incorporating terpyridine ligand linkers are proposed as a 3D printable ink that is cured post-printing by iron (II) cations for long-term stabilization. The uncross-linked micro-gels ink exhibits the rheological characteristics of granular materials, meaning yield-stress, shear-thinning and fast recovery. Upon curing, iron (II)-bis-terpyridine coordination complexes between neighboring micro-gels are formed. The supramolecular bonds are sufficiently strong and long-lived to maintain scaffold integrity during manual handling or immersion in liquid medium for over two months. The thermosensitivity of the micro-gels endows the printed construct with reversible and cyclable temperature-induced resolution enhancement, while the supramolecular cross-linking provides an asset of disintegration on-demand. The proposed micro-gel scaffolds are biocompatible, revealing the potential for biomedical applications and 4D bioprinting.

Despite recent developments in controlled polymerization techniques, the straightforward synthesis of block copolymers that feature both strong anionic and charge-neutral segments remains a difficult endeavor. In particular, solubility issues may arise during the direct synthesis of strong amphiphiles and typical postpolymerization deprotection often requires harsh conditions. To overcome these challenges, we employed Cu(0)-mediated reversible deactivation radical polymerization (Cu(0)-RDRP) on a hydrophobic isobutoxy-protected 3-sulfopropyl acrylate. Cu(0)-RDRP enables the rapid synthesis of the polymer, reaching high conversions and low dispersities while using a single solvent system and low amounts of copper species. These macromolecules are straightforward to characterize and can subsequently be deprotected in a mild yet highly efficient fashion to expose their strongly charged nature. Furthermore, a protected sulfonate segment could be grown from a variety of charge-neutral macroinitiators to produce, after the use of the same deprotection chemistry, a library of amphiphilic, double-hydrophilic as well as thermoresponsive block copolymers (BCPs). The ability of these various BCPs to self-assemble in aqueous media was further studied by dynamic light scattering, ζ-potential measurements as well as atomic force and electron microscopy.

The application of complex coacervates in promising areas such as coatings and surgical glues requires a tight control of their viscous and elastic behaviour, and a keen understanding of the corresponding microscopic mechanisms. While the viscous, or dissipative, aspect is crucial at pre-setting times and in preventing detachment, elasticity at long waiting times and low strain rates is crucial to sustain a load-bearing joints. The independent tailoring of dissipative and elastic properties proves to be a major challenge that can not be addressed adequately by the complex coacervate motif by itself. We propose a versatile model of complex coacervates with customizable rheological fates by functionalization of polyelectrolytes with terpyridines, which provide transient crosslinks through complexation with metals. We show that the rheology of the hybrid complexes shows distinct footprints of both metal-ligand and coacervate dynamics, the former as a contribution very close to pure Maxwell viscoelasticity, the latter approaching a sticky Rouse fluid. Strikingly, when the contribution of metal-ligand bonds is dominant at long times, the relaxation of the overall complex is much slower than either the "native"coacervate relaxation time or the dissociation time of a comparable non-coacervate polyelectrolyte-metal-ligand complex. We recognize this slowing-down of transient bonds as a synergistic effect that has important implications for the use of complementary transient bonding in coacervate complexes.

Encapsulation of charged proteins into complex coacervate core micelles (C3Ms) can be accomplished by mixing them with oppositely charged diblock copolymers. However, these micelles tend to disintegrate at high ionic strength. Previous research showed that the addition of a homopolymer with the same charge sign as the protein improved the stability of protein-containing C3Ms. In this research, we used fluorescence correlation spectroscopy (FCS) and dynamic light scattering (DLS) to study how the addition of the homopolymer affects the encapsulation efficiency and salt stability of the micelles. We studied the encapsulation of laccase spore coat protein A (CotA), a multicopper oxidase, using a strong cationic-neutral diblock copolymer, poly(N-methyl-2-vinyl-pyridinium iodide)-block-poly(ethylene oxide) (PM2VP(128)-b-PEO477), and a negatively charged homopolymer, poly(4-styrenesulfonate) (PSS215). DLS indeed showed an improved stability of this three-component C3M system against the addition of salt compared to a two-component system. Remarkably, FCS showed that the release of CotA from a three-component C3M system occurred at a lower salt concentration and over a narrower concentration range than the dissociation of C3Ms. In conclusion, although the addition of the homopolymer to the system leads to micelles with a higher salt stability, CotA is excluded from the C3Ms already at lower ionic strengths because the homopolymer acts as a competitor of the enzyme for encapsulation.

Complex coacervate core micelles (C3Ms) are formed by mixing aqueous solutions of a charged (bio)macromolecule with an oppositely charged-neutral hydrophilic diblock copolymer. The stability of these structures is dependent on the ionic strength of the solution; above a critical ionic strength, the micelles will completely disintegrate. This instability at high ionic strengths is the main drawback for their application in, e.g., drug delivery systems or protein protection. In addition, the stability of C3Ms composed of weak polyelectrolytes is pH-dependent as well. The aim of this study is to assess the effectiveness of covalent crosslinking of the complex coacervate core to improve the stability of C3Ms. We studied the formation of C3Ms using a quaternized and amine-functionalized cationic-neutral diblock copolymer, poly(2-vinylpyridine)-block-poly(ethylene oxide) (QP2VP-b-PEO), and an anionic homopolymer, poly(acrylic acid) (PAA). Two different core-crosslinking strategies were employed that resulted in crosslinks between both types of polyelectrolyte chains in the core (i.e., between QP2VP and PAA) or in crosslinks between polyelectrolyte chains of the same type only (i.e., QP2VP). For these two strategies we used the crosslinkers 1-ethyl-3-(3′-dimethylaminopropyl)carbodiimide hydrochloride (EDC) and dimethyl-3,3′-dithiopropionimidate dihydrochloride (DTBP), respectively. EDC provides permanent crosslinks, while DTBP crosslinks can be broken by a reducing agent. Dynamic light scattering showed that both approaches significantly improved the stability of C3Ms against salt and pH changes. Furthermore, reduction of the disulphide bridges in the DTBP core-crosslinked micelles largely restored the original salt-stability profile. Therefore, this feature provides an excellent starting point for the application of C3Ms in controlled release formulations.

3D bioprinting is a powerful fabrication technique in biomedical engineering, which is currently limited by the number of available materials that meet all physicochemical and cytocompatibility requirements for biomaterial inks. Inspired by the key role of coacervations in the extrusion and spinning of many natural materials, hyaluronic acid-chitosan complex coacervates are proposed here as tunable biomaterial inks. Complex coacervates are obtained through an associative liquid-liquid phase separation driven by electrostatic attraction between oppositely charged macromolecules. They offer bioactive properties as well as facile modulation of their mechanical properties through mild physicochemical changes in the environment, rendering them attractive for 3D bioprinting. Fine-tuning the salt concentration, pH, and molecular weight of the constituent polymers results in biomaterial inks that are printable in air and water. The biomaterial ink, initially a viscoelastic fluid, transitions into a viscoelastic solid upon printing due to dehydration (for printing in air) or due to a change in pH and ionic composition (for printing in water). Consequently, scaffolds printed using the complex coacervate inks are stable without the need for post-printing processing. Cell culture scaffolds fabricated in this way are cytocompatible and show long-term topological stability. These results pave the way to a new class of easy-to-handle tunable biomaterials for biofabrication.

Ultrasound can be used to promote the physical interlocking of adhesives and tissues

The assembly of oppositely charged block copolymers, containing small thermoresponsive moieties, was investigated as a function of salt concentration and temperature. Aqueous solutions of poly-[N-isopropylacrylamide]-b-poly[dimethylaminoethyl methacrylate] (NIPAM44-b-DMAEMA216) and PNIPAM-b-poly[acrylic acid]-b-PNIPAM (NIPAM35-b-AA200-b-NIPAM35) were mixed in equal charge stoichiometry, and analysed by light scattering (LS), NMR spectroscopy and small angle X-ray scattering (SAXS). At room temperature, two different micelle morphologies were found at different salt concentrations. At NaCl concentrations below 0.75 M, complex coacervate core micelles (C3M) with a PNIPAM corona were formed as a result of interpolyelectrolyte complexation. At NaCl concentrations exceeding 0.75 M, the C3M micelles inverted into PNIPAM cored micelles (PCM), containing a water soluble polyelectrolyte corona. This behavior is ascribed to the salt concentration dependence of both the lower critical solution temperature (LCST) of PNIPAM, and the complex coacervation. Above 0.75 M NaCl, the PNIPAM blocks are insoluble in water at room temperature, while complexation between the polyelectrolytes is prevented because of charge screening by the salt. Upon increasing the temperature, both types of micelles display a cloud point temperature (Tcp), despite the small thermoresponsive blocks, and aggregate into hydrogels. These hydrogels consist of a complexed polyelectrolyte matrix with microphase separated PNIPAM domains. Controlling the morphology and aggregation of temperature sensitive polyelectrolytes can be an important tool for drug delivery systems, or the application and hardening of underwater glues., The assembly of oppositely charged block copolymers, containing small thermoresponsive moieties, was investigated as a function of salt concentration and temperature.

Probabilistic fasteners are known to provide strong attachment onto their respective surfaces. Examples are Velcro® and the "3M dual lock"system. However, these systems typically only function using specific counter surfaces and are often destructive to other surfaces such as fabrics. Moreover, the design parameters to optimize their functionality are not obvious. Here, we present a surface patterned with soft micrometric features inspired by the mushroom shape showing a nondestructive mechanical interlocking and thus attachment to fabrics. We provide a scalable experimental approach to prepare these surfaces and quantify the attachment strength with rheometric and video-based analysis. In these "probabilistic fasteners,"we find that higher feature densities result in higher attachment force; however, the individual feature strength is higher on a low feature density surface. We interpret our results via a load-sharing principle common in fiber bundle models. Our work provides new handles for tuning the mechanical attachment properties of soft patterned surfaces that can be used in various applications including soft robotics.

Adhesive proteins of marine organisms contain significant amounts of hydrophobic amino acids. Therefore, inter- and intramolecular hydrophobic interactions are expected to play an important role in both adhesion and cohesion. Here, we mimic the hydrophobicity of adhesive proteins by using temperature-responsive polyelectrolyte complexes (TERPOCs) with a high poly(N-isopropylacrylamide) (PNIPAM) content. Upon mixing aqueous solutions of PNIPAM-b-poly(acrylic acid)-b-PNIPAM and poly(2-(dimethylamino) ethyl methacrylate) (PDMAEMA), complexation between the oppositely charged polyelectrolytes occurs. At low temperatures, complex coacervate core micelles (C3Ms) with a PNIPAM corona are formed, and upon a temperature increase, the solution turns into a hydrogel by the formation of a network of hydrophobic PNIPAM domains. Consequently, an abrupt increase in viscosity is observed upon heating which facilitates injectability of the adhesive. The gelation temperature, Tgel, and (adhesive) strength of the TERPOC can be adjusted by altering the salt and polymer concentration, which changes the balance between the electrostatic and hydrophobic interactions. Despite the importance of hydrophobic groups in strong underwater adhesives, we conclude that TERPOCs with a high PNIPAM content (70 wt%) are unstable due to water release. Consequently, there is a limited amount of hydrophobic groups that can be inserted in this type of systems. Nevertheless, TERPOCs show promising and tunable properties for application as injectable underwater adhesives, for example in biomedical applications.

While nature has optimized its antifouling strategies over millions of years, synthetic antifouling coatings have not yet reached technological maturity. For an antifouling coating to become technically feasible, it should fulfill many requirements: high effectiveness, long-term stability, durability, ecofriendliness, large-scale applicability, and more. It is therefore not surprising that the search for the perfect antifouling coating has been going on for decades. With the discovery of metal-based antifouling paints in the 1970s, fouling was thought to be a problem of the past, yet its untargeted toxicity led to serious ecological concern, and its use became prohibited. As a response, research shifted focus toward a biocompatible alternative: polymer-based antifouling coatings. This has resulted in numerous advanced and innovative antifouling strategies, including fouling-resistant, fouling-release, and fouling-degrading coatings. Here, these novel and exciting discoveries are highlighted while simultaneously assessing their antifouling performance and practical feasibility.

Fouling is a critical issue in membrane process operation as it greatly compromises the efficiency of the treatment processes. A promising approach to overcome this problem is the production of easy-to-clean membranes by incorporating stimuli-responsive pores. In this study, we fabricated thermo-responsive polystyrene-b-poly(N-isopropyl acrylamide) (PS-b-PNIPAM) block copolymer membranes using the self-assembly and non-solvent induced phase separation (SNIPS) method and systematically varied several membrane casting parameters, i.e. evaporation time, polymer concentration, solvent type and water content, to obtain nano- and isoporous membranes. Membranes with a disordered surface were obtained for PS selective solvents, whereas isoporous membranes were obtained when the block copolymers were dissolved in PNIPAM selective solvent mixtures. Using 1,4-dioxane/ tetrahydrofuran mixtures resulted in isoporous membranes for a large parameter space, indicating the robustness of structure formation in the PS-b-PNIPAM system. Permeability tests at various temperatures demonstrated fully reversible thermo-responsive behavior of the membranes.

A combined experimental and molecular dynamics (MD) study is performed to investigate the effect of polymer concentration on the zero shear rate viscosity η0 of a salt-free aqueous solution of poly(N,N-dimethylaminoethyl methacrylate) (PDMAEMA), a flexible thermoresponsive weak polyelectrolyte with a bulky 3-methyl-1,1-diphenylpentyl unit as the terminal group. The study is carried out at room temperature (T = 298 K) with relatively short PDMAEMA chains (each containing N = 20 monomers or repeat units) at a fixed degree of ionization (α+ = 100%). For the MD simulations, a thorough validation of several molecular mechanics force fields is first undertaken for assessing their capability to accurately reproduce the experimental observations and established theoretical laws. The generalized Amber force field in combination with the restrained electrostatic potential charge fitting method is eventually adopted. Three characteristic concentration regimes are considered: the dilute (from 5 to 10 wt %), the semidilute (from 10 to 20 wt %), and the concentrated (from 20 to 29 wt %); the latter two are characterized by polymer concentrations cp higher than the characteristic overlap concentration cp*. The structural behavior of the PDMAEMA chains in the solution is assessed by calculating the square root of their mean-square radius of gyration «Rg 2»0.5, the square root of the average square chain end-to-end distance «Ree 2»0.5, the ratio «Ree 2»/«Rg 2», and the persistence length Lp. It is observed that at low polymer concentrations, PDMAEMA chains adopt a stiffer and slightly extended conformation because of excluded-volume effects (a good solvent is considered in this study) and electrostatic repulsions within the polymer chains. As the polymer concentration increases above 20 wt %, the PDMAEMA chains adopt more flexible conformations, as the excluded-volume effects seize and the charge repulsion within the polymer chains subsides. The effect of total polymer concentration on PDMAEMA chain dynamics in the solution is assessed by calculating the orientational relaxation time τc of the chain, the center-of-mass diffusion coefficient D, and the zero shear rate viscosity η0; the latter is also measured experimentally here and found to be in excellent agreement with the MD predictions.

International audience; Many marine organisms have developed adhesives that are able to bond under water, overcoming the challenges associated with wet adhesion. A key element in the processing of several natural underwater glues is complex coacervation, a liquid–liquid phase separation driven by complexation of oppositely charged macromolecules. Inspired by these examples, the development of a fully synthetic complex coacervate‐based adhesive is reported with an in situ setting mechanism, which can be triggered by a change in temperature and/or a change in ionic strength. The adhesive consists of a matrix of oppositely charged polyelectrolytes that are modified with thermoresponsive poly(N‐isopropylacrylamide) (PNIPAM) grafts. The adhesive, which initially starts out as a fluid complex coacervate with limited adhesion at room temperature and high ionic strength, transitions into a viscoelastic solid upon an increase in temperature and/or a decrease in the salt concentration of the environment. Consequently, the thermoresponsive chains self‐associate into hydrophobic domains and/or the polyelectrolyte matrix contracts, without inducing any macroscopic shrinking. The presence of PNIPAM favors energy dissipation by softening the material and by allowing crack blunting. The high work of adhesion, the gelation kinetics, and the easy tunability of the system make it a potential candidate for soft tissue adhesion in physiological environments.

International audience; Most commercially available soft tissue glues offer poor performance in the human body. Inspired by sea water organisms, we have developed an injectable adhesive whose setting mechanism is activated by a variation in environmental factors (e.g temperature and/or ionic strength). The material is prepared by mimicking a mechanism observed in many natural glues: complex coacervation, a liquid-liquid phase separation between oppositely charged polyelectrolytes. Complex coacervates are characterized by a high-water content, which inevitably weakens the glue. Here, we target the increase of the adhesive properties by systematically tuning the water content, a key parameter affecting the material performance. Among the several strategies here explored, the most effective one is the mechanical removal of water using an extruder, resulting in a work of adhesion increase of one order of magnitude compared to the original formulation.

Underwater adhesion represents a huge technological challenge as the presence of water compromises the performance of most commercially available adhesives. Inspired by natural organisms, we have designed an adhesive based on complex coacervation, a liquid&ndash, liquid phase separation phenomenon. A complex coacervate adhesive is formed by mixing oppositely charged polyelectrolytes bearing pendant thermoresponsive poly(N-isopropylacrylamide) (PNIPAM) chains. The material fully sets underwater due to a change in the environmental conditions, namely temperature and ionic strength. In this work, we incorporate silica nanoparticles forming a hybrid complex coacervate and investigate the resulting mechanical properties. An enhancement of the mechanical properties is observed below the PNIPAM lower critical solution temperature (LCST): this is due to the formation of PNIPAM&ndash, silica junctions, which, after setting, contribute to a moderate increase in the moduli and in the adhesive properties only when applying an ionic strength gradient. By contrast, when raising the temperature above the LCST, the mechanical properties are dominated by the association of PNIPAM chains and the nanofiller incorporation leads to an increased heterogeneity with the formation of fracture planes at the interface between areas of different concentrations of nanoparticles, promoting earlier failure of the network&mdash, an unexpected and noteworthy consequence of this hybrid system.

Block copolymers that accommodate both hydrophobic and ionic elements are interesting materials for numerous applications, such as stabilizing agents, lubricants and proton-exchange membranes. Frequently these copolymers are based on weak polyelectrolytes, but the pH-dependent charge density restricts their use to a limited pH window. Although strong polyelectrolytes do not suffer this problem, the most commonly employed post-modification approach limits the choice of the hydrophobic component, as harsh reaction conditions are usually involved. Moreover, this method often results in incomplete functionalization of the precursor copolymer. To avoid these difficulties a mild route was developed that is based on a hydrophobic protected poly(3-sulfopropyl methacrylate) intermediate that enables the preparation of well-defined strong anionic polyelectrolytes. The potential of this method was demonstrated by synthesizing hydrophobic/strong anionic diblock copolymers, and their self-assembly in aqueous solution was studied.

The synthesis of charged polymers often requires the polymerization of protected monomers, followed by a polymer-analogous reaction to the polyelectrolyte product. We present a mild, facile method to cleave tert-butyl groups from poly(tert-butyl acrylate) blocks that yields poly(acrylic acid) (pAA) blocks free of traces of the ester. The reaction utilizes a slight excess of HCl in hexafluoroisopropanol (HFIP) at room temperature and runs to completion within 4 h. We compare deprotection in HFIP with the common TFA/DCM method and show that the latter does not yield clean pAA. We show the effect of complete tert-butyl cleavage on a ABA triblock copolymer, where poly(N-isopropylacrylamide) (pNIPAM) is A and pAA is B, by means of viscosimetry, DLS, and SAXS on solutions above overlap. The pNIPAM blocks dehydrate, and their increased self-affinity above the lower critical solution temperature (LCST) results in network formation by the triblocks. This manifests itself as an increase in viscosity and a slowing down of the first-order correlation function in light scattering. However, this stickering effect manifests itself exclusively when the pAA block is tert-butyl-free. Additionally, SAXS shows that the conformational properties of tert-butyl-free pAA copolymers are markedly different from those with residual esters. Thus, we illustrate a surprising effect of hydrophobic impurities that act across blocks and assert the usefulness of HCl/HFIP in pAA synthesis.

Contains fulltext : 171021.pdf (Publisher’s version ) (Open Access) This study describes the design, production, and testing of functionalized variants of a recombinant protein-based polymer that forms nanofibrillar hydrogels with self-healing properties. With a view to bone tissue engineering applications, we equipped these variants with N-terminal extensions containing either (1) integrin-binding (RGD) or (2) less commonly studied proteoglycan-binding (KRSR) cell-adhesive motifs. The polymers were efficiently produced as secreted proteins using the yeast Pichia pastoris and were essentially monodisperse. The pH-responsive protein-based polymers are soluble at low pH and self-assemble into supramolecular fibrils and hydrogels at physiological pH. By mixing functionalized and nonfunctionalized proteins in different ratios, and adjusting pH, hydrogel scaffolds with the same protein concentration but varying content of the two types of cell-adhesive motifs were readily obtained. The scaffolds were used for the two-dimensional culture of MG-63 osteoblastic cells. RGD domains had a slightly stronger effect than KRSR domains on adhesion, activity, and spreading. However, scaffolds featuring both functional domains revealed a clear synergistic effect on cell metabolic activity and spreading, and provided the highest final degree of cell confluency. The mixed functionalized hydrogels presented here thus allowed to tailor the osteoblastic cell response, offering prospects for their further development as scaffolds for bone regeneration. Traditional paints consist of hydrophobic polymers dissolved in hydrocarbons; they are appreciated for their rheological properties and the smooth and glossy films they form upon drying and crosslinking, but are now largely banned because of the hazards associated with the solvents. In terms of health, water borne paints based on colloidal resin dispersions are an improvement but these systems lack the rheological and film forming properties of traditional paints. We present here a bio-inspired alternative that combines the best of both worlds: a water soluble polymer that can crosslink by mild oxidation to a fully water resistant and adhesive coating. Using free radical polymerization, we copolymerized two water soluble monomers, namely borax-protected dopamine acrylamide (DAA-p) and 2-aminoethylmethacrylamide (AEMA) in various proportions. We determined the reactivity ratio of these monomers using an in situ1H NMR monitoring method and found values of 0.0 and 0.46 for DAA-p and AEMA, respectively. From this we conclude that in the polymers DAA-p pairs do not occur, while DAA-AEMA diads are relatively frequent. After removing the protective borax by hydrolysis, we obtain polymers which are soluble in water at low pH (pH 2) but which undergo rapid oxidative crosslinking when the catechol groups of DAA react with amines from AEMA. This leads to a water resistant, mildly hydrophobic film with a water/air contact angle of about 40 degrees which adheres well to glass substrates.

Nature shows excellent control over the mechanics of fibrous hydrogels by assembling protein fibers into bundles of well-defined dimensions. Yet, obtaining artificial materials displaying controlled bundling remains a challenge. Here, we developed genetically engineered protein-based polymers functionalized with heparin-binding KRSR domains and show controlled bundling using heparin as a binder. The protein polymer forms fibers upon increasing the pH to physiological values and at higher concentrations fibrous gels. We show that addition of heparin to the protein polymer with incorporated KRSR domains, induces bundling, which results in faster gel formation and stiffer gels. The interactions are expected to be primarily electrostatic and fiber bundling has an optimum when the positive charges of KRSR are approximately in balance with the negative charges of the heparin. Our study suggests that, generally, a straightforward method to control the properties of fibrous gels is to prepare a fiber former with specific binding domains and then simply adding an appropriate amount of binder.

We used the self-consistent field (SCF) formalism of Scheutjens and Fleer (SF-SCF) to complement existing theoretical investigations on the phase behavior of block copolymer melts. This method employs the freely jointed chain (FJC) model for finite chain length and systematic differences exist compared to the classical SCF predictions. We focus on the critical and hexagonal (HEX) to lamellar (LAM) phase transition region at intermediate and strong segregations. Chain length (N) dependence of the critical point ( χ c r ) was found to be χ c r N = 10.495 ( 1 + 4 / N ) . The characteristic spacing (D) of LAM was found as D = 4 / 3 N at the critical conditions. We present SF-SCF predictions for the phases single gyroid (SG), double gyroid (DG) and hexagonally perforated lamellar (HPL), in the region where HEX and LAM compete. At χ N = 30 , N = 300 ; we found SG and HPL were metastable with respect to LAM or HEX, DG was stable in a narrow region of the asymmetry ratio. In contrast to the latest predictions, at strong segregation χ N = 120 , DG was found to be metastable. From the structural evolution of HPL, we speculate that this may be an intermediate phase that allows the system to go through various connectivity regimes between minority and majority blocks.

Nature has developed protein‐based adhesives whose underwater performance has attracted much research attention over the last few decades. The adhesive proteins are rich in catechols combined with amphiphilic and ionic features. This combination of features constitutes a supramolecular toolbox, to provide stimuli‐responsive processing of the adhesive, to secure strong adhesion to a variety of surfaces, and to control the cohesive properties of the material. Here, the versatile interactions used in adhesives secreted by sandcastle worms and mussels are explored. These biological principles are then put in a broader perspective, and synthetic adhesive systems that are based on different types of supramolecular interactions are summarized. The variety and combinations of interactions that can be used in the design of new adhesive systems are highlighted.

In this article, catechol-functionalized polymers are synthesized by free radical polymerization of dopamine methacrylamide (DMA) and 2-methoxyethyl methacrylate (MEA) at 60 °C in DMF. By varying the DMA content in the polymer, it is found that during free radical polymerization, the catechol groups in DMA react with the propagating radicals, resulting in the formation of a crosslinked structure. We systematically study the effect of DMA content and crosslinking on the adhesion properties of the polymers. Under both dry and wet conditions, maximum adhesion is obtained for a polymer composed of 5 mol% DMA. This polymer exhibits an optimum balance between the catechol content to strengthen the interface, compliance to ensure good contact formation and cohesive strength to resist separation. An increase in the crosslinking degree of the polymer resulted in reduced dry adhesion.

The incorporation of stimuli-responsive pores in nanoporous membranes is a promising approach to facilitate the cleaning process of the membranes. Here we present fully reversible thermo-responsive nanoporous membranes fabricated by self-assembly and non-solvent induced phase separation (SNIPS) of polystyrene-poly(N-isopropylacrylamide) (PS-PNIPAM) block copolymers. A variety of PS-PNIPAM block copolymers were synthesized by reversible addition-fragmentation chain transfer (RAFT) polymerization and the reaction conditions were optimized. The target copolymers featured: (1) a thermo-responsive PNIPAM block, (2) a majority PS fraction, and (3) a well-defined high molecular weight, which are requirements for successful fabrication of free-standing responsive membranes using SNIPS. The resulting membranes exhibited a worm-like cylindrical morphology with interconnected nanopores. The thermo-responsive character of the membranes was studied by measuring the permeability of the membranes as a function of temperature. The permeability was found to increase by almost 400% upon going from room temperature to 50 °C and this thermo-responsive character was fully reversible.

International audience; In this work, we report the systematic investigation of a multiresponsive complex coacervate-based underwater adhesive, obtained by combining polyelectrolyte domains and thermoresponsive poly(N-isopropylacrylamide) (PNIPAM) units. This material exhibits a transition from liquid to solid but, differently from most reactive glues, is completely held together by non-covalent interactions, i.e., electrostatic and hydrophobic. Because the solidification results in a kinetically trapped morphology, the final mechanical properties strongly depend on the preparation conditions and on the surrounding environment. A systematic study is performed to assess the effect of ionic strength and of PNIPAM content on the thermal, rheological and adhesive properties. This study enables the optimization of polymer composition and environmental conditions for this underwater adhesive system. The best performance with a work of adhesion of 6.5 J/m 2 was found for the complex coacervates prepared at high ionic strength (0.75 M NaCl) and at an optimal PNIPAM content around 30% mol/mol. The high ionic strength enables injectability, while the hydrated PNIPAM domains provide additional dissipation, without softening the material so much that it becomes too weak to resist detaching stress.

Catechols play an important role in many natural systems. They are known to readily interact with both organic (e.g., amino acids) and inorganic (e.g., metal ions, metal oxides) compounds, thereby providing a powerful system for protein curing. Catechol crosslinked protein networks, such as sclerotized cuticle and byssal threads of the mussel, have been shown to exhibit excellent mechanical properties. A lot of effort has been devoted to mimicking the natural proteins using synthetic catechol-functionalized polymers. Despite the success in developing catechol-functionalized materials, the crosslinking chemistry of catechols is still a subject of debate. To develop materials with controlled and superior properties, a clear understanding of the crosslinking mechanism of catechols is of vital importance. This review describes the crosslinking pathways of catechol and derivatives in both natural and synthetic systems. We discuss existing pathways of catechol crosslinking and parameters that affect the catechol chemistry in detail. This overview will point towards a rational direction for further investigation of the complicated catechol chemistry.

In this study, we assess the utility of a normal force (pull-test) approach to measuring adhesion in organic solar cells and organic light-emitting diodes. This approach is a simple and practical method of monitoring the impact of systematic changes in materials, processing conditions, or environmental exposure on interfacial strength and electrode delamination. The ease of measurement enables a statistical description with numerous samples, variant geometry, and minimal preparation. After examining over 70 samples, using the Weibull modulus and the characteristic breaking strength as metrics, we were able to successfully differentiate the adhesion values between 8-tris(hydroxyquinoline aluminum) (Alq3) and poly(3-hexyl-thiophene) and [6,6]-phenyl C61-butyric acid methyl ester (P3HT:PCBM) interfaces with Al and between two annealing times for the bulk heterojunction polymer blends. Additionally, the Weibull modulus, a relative measure of the range of flaw sizes at the fracture plane, can be correlated with the roughness of the organic surface. Finite element modeling of the delamination process suggests that the out-of-plane elastic modulus for Alq3 is lower than the reported in-plane elastic values. We suggest a statistical treatment of a large volume of tests be part of the standard protocol for investigating adhesion to accommodate the unavoidable variability in morphology and interfacial structure found in most organic devices.

Catechol chemistry is used as a crosslinking tool abundantly in both natural organisms (e.g. mussels, sandcastle worms) and synthetic systems to achieve the desired mechanical properties. Despite this abundance and success, the crosslinking chemistry is still poorly understood. In this study, to simplify the system, yet to capture the essential chemistry, model compounds 4-methyl catechol and propylamine are used. The reaction of 4-methyl catechol (2 mM) with propylamine (6 mM) is carried out in the presence of NaIO4 (2 mM) in 10 mM Na2CO3 aqueous solution. A variety of spectroscopic/spectrometric and chromatographic methods such as 1H NMR, LC-MS, and UV-VIS are used to track the reaction and identify the products/intermediates. It is found that the crosslinking chemistry of a catechol and an amine is both fast and complicated. Within five minutes, more than 60 products are formed. These products encompass 19 different masses ranging from molecular weight of 179 to 704. By combining time-dependent data, it is inferred that the dominant reaction pathways: the majority is formed via aryloxyl-phenol coupling and Michael-type addition, whereas a small fraction of products is formed via Schiff base reactions.

In this work, the adhesion of biomimetic polydimethylsiloxane (PDMS) pillar arrays with mushroom-shaped tips was studied on nano- and micro-rough surfaces and compared to unpatterned controls. The adhesion strength on nano-rough surfaces invariably decreased with increasing roughness, but pillar arrays retained higher adhesion strengths than unpatterned controls in all cases. The results were analyzed with a model that focuses on the effect on adhesion of depressions in a rough surface. The model fits the data very well, suggesting that the pull-off strength for patterned PDMS is controlled by the deepest dimple-like feature on the rough surface. The lower pull-off strength for unpatterned PDMS may be explained by the initiation of the pull-off process at the edge of the probe, where significant stress concentrates. With micro-rough surfaces, pillar arrays showed maximum adhesion with a certain intermediate roughness, while unpatterned controls did not show any measurable adhesion. This effect can be explained by the inability of micropatterned surfaces to conform to very fine and very large surface asperities.

Progress in adhesion technology over the last few decades has led to widespread replacement of mechanical fasteners with adhesive bonds. Despite the advances, it remains challenging to produce materials that are sticky on demand. In this feature article we highlight recent efforts to develop reversibly switchable adhesives, that exhibit the ability to trigger adhesion in response to an environmental change – for example, pH, solvent, temperature, mechanics, and electro/magnetic field. We review two initially separate design principles to induce switchable adhesion: (1) chemical functionality and (2) topography. Combining both approaches may lead to novel hierarchical adhesives with interesting property profiles.

Batteries, fuel cells and solar cells, among many other high-current-density devices, could benefit from the precise meso- to macroscopic structure control afforded by the silica sol-gel process. The porous materials made by silica sol-gel chemistry are typically insulators, however, which has restricted their application. Here we present a simple, yet highly versatile silica sol-gel process built around a multifunctional sol-gel precursor that is derived from the following: amino acids, hydroxy acids or peptides; a silicon alkoxide; and a metal acetate. This approach allows a wide range of biological functionalities and metals--including noble metals--to be combined into a library of sol-gel materials with a high degree of control over composition and structure. We demonstrate that the sol-gel process based on these precursors is compatible with block-copolymer self-assembly, colloidal crystal templating and the Stöber process. As a result of the exceptionally high metal content, these materials can be thermally processed to make porous nanocomposites with metallic percolation networks that have an electrical conductivity of over 1,000 S cm(-1). This improves the electrical conductivity of porous silica sol-gel nanocomposites by three orders of magnitude over existing approaches, opening applications to high-current-density devices.

Mesoporous and highly crystalline TiO2 (anatase)/carbon composites with large (>5?nm) and uniform pores were synthesized using PI-b-PEO block copolymers as structure directing agents. Pore sizes could be tuned by utilizing block copolymers with different molecular weights. The resulting mesoporous TiO2/carbon was successfully used as an anode material for Li ion batteries. Without addition of conducting aid (Super P), the electrode showed high capacity during the first insertion/desertion cycle due to carbon wiring inside the walls of mesoporous TiO2/carbon. The electrode further showed stable cycle performance up to 50 cycles and the specific charge capacity at 30?C was 38?mA h (g of TiO2)-1, which indicates CCM-TiO2/carbon can be used as a material for high rate use.

"To learn from nature" is a highly desirable goal of modern materials research. However, mimicking nature alone is not sufficient: a multidisciplinary approach based on biology, physics, chemistry, materials science and theoretical mechanics is necessary to understand the principles which have been successful in evolution and to apply them to technical systems. This allows the development of new adhesive surfaces, where a large industrial market becomes apparent - from household articles up to medical implants.

„Von der Natur lernen“ ist ein viel zitiertes Ziel moderner Materialforschung. Doch Nachahmen allein reicht nicht: Ein multidisziplinarer Ansatz, bestehend aus Biologie, Physik, Chemie, Materialwissenschaft und theoretischer Mechanik ist notwendig, um die in der Evolution erfolgreichen Prinzipien zu verstehen und auf technische Systeme zu ubertragen. So konnten neue Haftoberflachen entwickelt werden, fur die sich bereits ein groser industrieller Einsatzbedarf abzeichnet — vom Haushaltsartikel bis zum medizinischen Implantat.

Nature has developed elegant and economical strategies to produce materials that are well-adapted to their purposes. As biology evolved to remarkable and complex designs, synthetic mimics are evolving toward new levels of complexity achieving larger combinations of properties within one material. The field of bioinspiration encompasses a wide range of advanced materials ranging from biooptics to energy materials, to biomaterials. In this paper an overview is given of selected recent developments in the field of bioinspired material design focusing on gecko-inspired adhesives, mussel-inspired coatings, and spider silk-inspired biomaterials.
