Background Despite disproportionate rates of mental ill-health compared with non-Indigenous populations, few programs have been tailored to the unique health, social, and cultural needs and preferences of young Aboriginal and Torres Strait Islander males. This paper describes the process of culturally adapting the US-based Young Black Men, Masculinities, and Mental Health (YBMen) Project to suit the needs, preferences, culture, and circumstances of Aboriginal and Torres Strait Islander males aged 16–25 years in the Northern Territory, Australia. YBMen is an evidence-based social media-based education and support program designed to promote mental health, expand understandings of gender and cultural identities, and enhance social support in college-aged Black men. Methods Our adaptation followed an Extended Stages of Cultural Adaptation model. First, we established a rationale for adaptation that included assessing the appropriateness of YBMen’s core components for the target population. We then investigated important and appropriate models to underpin the adapted program and conducted a non-linear, iterative process of gathering information from key sources, including young Aboriginal and Torres Strait Islander males, to inform program curriculum and delivery. Results To maintain program fidelity, we retained the core curriculum components of mental health, healthy masculinities, and social connection and kept the small cohort, private social media group delivery but developed two models: ‘online only’ (the original online delivery format) and ‘hybrid in-person/online’ (combining online delivery with weekly in-person group sessions). Adaptations made included using an overarching Aboriginal and Torres Strait Islander social and emotional wellbeing framework and socio-cultural strengths-based approach; inclusion of modules on health and wellbeing, positive Indigenous masculinities, and respectful relationships; use of Indigenous designs and colours; and prominent placement of images of Aboriginal and Torres Strait Islander male sportspeople, musicians, activists, and local role models. Conclusions This process resulted in a culturally responsive mental health, masculinities, and social support health promotion program for young Aboriginal and Torres Strait Islander males. Next steps will involve pilot testing to investigate the adapted program’s acceptability and feasibility and inform further refinement. Abstract In contrast to single‐phase materials the manufacturing process of composites creates multiscale interfaces among constituent materials and between laminar multilayers. Targeting the interfaces in these composite structures, this paper presents a perspective study on the impact safety of potential future composite rail vehicles. The aim is to conceptually explore the key role of interfacial discontinuities of composite material structures as they are a critical issue affecting the impact performance of future composite rail vehicles. Following a theoretical description, the issues are addressed in two parts. First, composite materials are characteristically analyzed from the perspective of their interfacial discontinuities within the materials and between the laminate multilayers to identify their influence. Second, the structural conditions required for crashworthiness are determined in relation to the dual requirements of global stability and local deformability for efficient energy absorption. The key findings are: (1) The interfacial discontinuities of the material phases and the designed structural assemblies need to be tailored for crashworthiness performance and (2) Global stability and locally deformability are the key dual requirements for the energy absorbing progressive deformations that are essential for application of composite for crashworthiness of rail vehicles. The research conceptually explores a key issue of the impact mechanics of composite structures from the perspective of impact safety of composite rail vehicles.

Abstract Quality control (QC) is a critical component of single-cell RNA-seq (scRNA-seq) processing pipelines. Current approaches to QC implicitly assume that datasets are comprised of one cell type, potentially resulting in biased exclusion of rare cell types. We introduce SampleQC, which robustly fits a Gaussian mixture model across multiple samples, improves sensitivity, and reduces bias compared to current approaches. We show via simulations that SampleQC is less susceptible to exclusion of rarer cell types. We also demonstrate SampleQC on a complex real dataset (867k cells over 172 samples). SampleQC is general, is implemented in R, and could be applied to other data types.

To meet the various demands for bus platforms tailored for the emerging new power sources, an adaptive design method is proposed for a bus chassis development using a design structure matrix. The structural feature parameters of the product modules are extracted and the corresponding design structure matrix is established to analyze structural correlations between modules. An adaptive design method process model is constructed following quantification of the design structure matrix. A digital matrix of the module interface is then built to characterize integration and interchangeability of functional modules. Reconfiguration of modules is conducted using the parent-child relationship premise so that the interface matrixes are well integrated. The adaptability of these products is subsequently improved by sharing dominant modules and replacement of local modules. Finally, the implementation of the proposed methodology is illustrated for a current bus chassis.

Impact-derived ejecta covers most of the lunar surface, originating from recent impacts through to the beginning of the geologic record. Despite how common ejecta is, accurate measurements of ejecta thickness are difficult to obtain, and existing estimates of ejecta thickness vary widely. This study uses excavation by meter-scale impacts on the fresh ejecta blankets of larger, kilometer-scale impacts to make point measurements of ejecta thickness. We estimate ejecta thickness at the rims of 73 lunar craters (0.1–4.8 km diameter) and create isopach maps of ejecta thickness for three craters. We derive an equation for ejecta thickness, $t=0.14\pm 0.062{R}^{(0.77\pm 0.080)}{\left(r/R\right)}^{(-B)}$ , where r is the horizontal distance from the center of the crater, R is the center-to-rim crater radius, and B describes the rate at which ejecta thickness decays with radial distance. Our average value for B (2.8 ± 0.1) is similar to previous work, though we observe that B can vary significantly within an ejecta blanket.

The genetic prehistory of central America has not been well explored. Here, the authors find evidence from ancient DNA from twenty individuals who lived in Belize 9,600 to 3,700 years ago of a migration from the south that coincided with the first evidence for forest clearing and the spread of maize horticulture.

Abstract Buried pipelines are an essential component of the urban infrastructure of modern cities. Traditional buried pipes are mainly made of metal materials. With the development of material science and technology in recent years, non-metallic pipes, such as plastic pipes, ceramic pipes, and concrete pipes, are increasingly taking the place of pipes made from metal in various pipeline networks such as water supply, drainage, heat, industry, oil, and gas. The location technologies for the location of the buried metal pipeline have become mature, but detection and location technologies for the non-metallic pipelines are still developing. In this paper, current trends and future perspectives of detection and location of buried non-metallic pipelines are summarized. Initially, this paper reviews and analyzes electromagnetic induction technologies, electromagnetic wave technologies, and other physics-based technologies. It then focuses on acoustic detection and location technologies, and finally introduces emerging technologies. Then the technical characteristics of each detection and location method have been compared, with their strengths and weaknesses identified. The current trends and future perspectives of each buried non-metallic pipeline detection and location technology have also been defined. Finally, some suggestions for the future development of buried non-metallic pipeline detection and location technologies are provided.

The structural feasibility of using a pultrusion of carbon-fibre-reinforced polymers (CFRP) for the lightweight design of a mast for overhead line railway electrification was investigated and simulated. Material characterisation was undertaken using three-point bending and finite element analysis to identify the orthotropic properties of the pultruded tubes designed for a composite mast for overhead electrification. An innovative design of the mast was proposed and verified using a simulation that compared the deflection and stress levels under wind and inertial load. From the simulation results, it was concluded that the proposed composite structure design complies with the mechanical performance requirements for its implementation and benefits the application with a weight reduction of more than 80% with respect to the current steel mast design.

Expensive tooling often constraints the use of composites in the design and development of automotive parts. While there is significant confidence and knowledge in sheet and bulk metals, composite processes are less understood in mass production environment. The processes used to produce composites and resulting properties are influenced by fiber length attrition, resin to fiber ratio, process waste etc. Tool designs are determined very early in the engineering process. It is cost prohibitive to build additional tools, in the event it becomes obvious a better processing method and material would be beneficial, the original decision is not easily changed. In the present work we recognize the bottleneck of tooling costs and provide an approach of multi-process tooling. The innovation of this work is the design and demonstration of a single tool for different processes namely injection, injection-compression and extrusion-compression. The materials used in this study were long and short fiber thermoplastics (LFTs and SFTs). The resulting structure-property relationships have been reported for the materials and processing methods with a battery tray (BT) tool.

Studies investigating autistic community research priorities indicate a mismatch between what autism research focuses on and what autistic people want to see researched. Furthermore, there has not been a research priority-setting exercise specifically with autistic people in Scotland, where there are unique cultural, political and social contexts. Using a community-based participatory design, we aimed to identify the research priorities of autistic adults living in Scotland. Autistic and non-autistic researchers designed and conducted a survey where 225 autistic adults rated and ranked research topics in order of importance and provided qualitative feedback on issues and questions important to them. The top five research priorities were mental health/well-being, identification and diagnosis of autistic people, support services, knowledge and attitudes towards autistic people and issues impacting autistic women. There were differences in priorities according to different intersections of identity, and qualitative responses indicated a desire for research to focus on support and understanding. The bottom three priorities concerned genetics, treatments and interventions and causes. These findings emphasise the need to address the gap between what autism research focuses on and the everyday lives of autistic people.

Ground penetrating radar (GPR) is a well-known useful tool for subsurface exploration. GPR data can be recorded at a relatively high speed in a continuous way with hyperbolas being artifacts and evidence of disturbances in the soil. Automatic and accurate detection and interpretation of hyperbolas in GPR remains an open challenge. Recently deep learning techniques have achieved remarkable success in image recognition tasks and this has potential for interpretation of GPR data. However, training reliable deep learning models requires massive labeled data, which is challenging. To address the challenges, this work proposes a Generative Adversarial Nets (GANs)-based deep learning framework, which generates new training data to address the scarcity of GPR data, automatically learns features and detects subsurface objects (via hyperbola) through an end-to-end solution. We have evaluated our proposed approach using real GPR B-scan images from rail infrastructure monitoring applications and compared this with the state-of-the-art methods for object detection (i.e. Faster-RCNN, Cascade R-CNN, SSD and YOLO V2). The proposed approach outperforms the existing methods with high accuracy of 97% being the mean Average Precision (mAP). Moreover, the proposed approach also demonstrates the good generalizability through cross-validation on independent datasets.

There has been much debate around the role of metrics in scholarly communication, with particular focus on the misapplication of journal metrics, such as the impact factor in the assessment of research and researchers. Various initiatives have advocated for a change in this culture, including the Declaration on Research Assessment (DORA), which invites stakeholders throughout the scholarly communication ecosystem to sign up and show their support for practices designed to address the misuse of metrics. This case study provides an overview of the process undertaken by a large academic publisher (Taylor & Francis Group) in signing up to DORA and implementing some of its key practices in the hope that it will provide some guidance to others considering becoming a signatory. Our experience suggests that research, consultation and flexibility are crucial components of the process. Additionally, approaching signing with a project mindset versus a ‘sign and forget’ mentality can help organizations to understand the practical implications of signing, to anticipate and mitigate potential obstacles and to support cultural change.

Railway Tunnel SubSurface Inspection (RTSSI) is essential for targeted structural maintenance. ‘Effective’ detection, localisation and characterisation of fully concealed features (i.e., assets, defects) is the primary challenge faced by RTSSI engineers, particularly in historic masonry tunnels. Clear conveyance and communication of gathered information to end-users poses the less frequently considered secondary challenge. The purpose of this review is to establish the current state of the art in RTSSI data acquisition and information conveyance schemes, in turn formalising exactly what constitutes an ‘effective’ RTSSI visualisation framework. From this knowledge gaps, trends in leading RTSSI research and opportunities for future development are explored. Literary analysis of over 300 resources (identified using the 360-degree search method) informs data acquisition system operation principles, common strengths and limitations, alongside leading studies and commercial tools. Similar rigor is adopted to appraise leading information conveyance schemes. This provides a comprehensive whilst critical review of present research and future development opportunities within the field. This review highlights common shortcomings shared by multiple methods for RTSSI, which are used to formulate robust criteria for a contextually ‘effective’ visualisation framework. Although no current process is deemed fully effective; a feasible hybridised framework capable of meeting all stipulated criteria is proposed based on identified future research avenues. Scope for novel analysis of helical point cloud subsurface datasets obtained by a new rotating ground penetrating radar antenna is of notable interest.

Abstract Camera traps are increasingly used to monitor wildlife populations and management activities. Failing to detect target occurrence and/or behaviour inhibits the robustness of wildlife surveys. Based on user‐testing, it is reasonable to expect some equipment to malfunction but other sources of failure, such as those caused by theft and vandalism, are largely unquantified. Between May 2016 and October 2017, we undertook an international survey of professional practitioners who use camera traps for wildlife research and management projects to quantify theft and vandalism, and to document the subsequent effects on project outcomes. We also sought to record the methods used by practitioners to avoid theft and vandalism and whether or not practitioners believed those actions were effective. Most (59%) of the 407 respondents were wildlife researchers and university academics. The survey results revealed that camera trap theft and vandalism is a global issue that not only adds to costs via equipment loss (approx. USD $1.48 million from n = 309 respondents between 2010 and 2015) and theft prevention (c. USD $800 000 spent by respondents between 2010 and 2015) but also influences survey design. Vandalism and theft are clearly a global problem, with responses suggesting that they occur across a diverse array of geographic locations, at varying proximity to human settlements, in multiple habitat types and across device placements. Methods to deter human interference included using camouflaging (73%), security devices such as chains (63%) and boxes (43%), use of decoy camera traps, shortening deployment periods, setting the camera relatively high or low to the ground, or moving away from human traffic. Despite this, the responses suggest that attempts to mitigate losses are often not effective. In review of our findings, we make recommendations for the future of camera trapping that requires implementation and testing.

This article provides a critical review of the evidence on ‘thinking and working politically’ (TWP) in development. Scholars and practitioners have increasingly recognised that development is a fundamentally political process, and there are concerted efforts underway to develop more politically-informed and adaptive ways of thinking and working in providing development assistance. However, while there are interesting and engaging case studies in the emerging, largely practitioner-based literature, these do not yet constitute a strong evidence base that shows these efforts can be clearly linked to more effective aid programming. Much of the evidence used so far to support these approaches is anecdotal, does not meet high standards for a robust body of evidence, is not comparative and draws on a small number of self-selected, relatively well-known success stories written primarily by programme insiders. The article discusses the factors identified in the TWP literature that are said to enable politically-informed programmes to increase aid effectiveness. It then looks at the state of the evidence on TWP in three areas: political context, sector, and organisation. The aim is to show where research efforts have been targeted so far and to provide guidance on where the field might focus next. In the final section, the article outlines some ways of testing the core assumptions of the TWP agenda more thoroughly, to provide a clearer sense of the contribution it can make to aid effectiveness.

Modular design for a bus chassis is a practical solution to address different bus types and their differing powertrains. Physics interfaces are used to connect interacting subsystems in bus chassis modules. There are few studies focusing on the criteria for assessing interface connectivity, with significantly non-standard characteristics, between bus chassis modules. This study proposes criteria for assessing the interface connectivity in bus chassis modules in accordance with the digital expression of the module interface. Criteria for judging the compatibility of the connection between bus chassis modules are proposed and modular reconfiguration methods are provided, based on the criteria. Reconfiguration of the process matrix is constructed to indicate changes in position, shape, and size after the component is modified, based on the connectivity between modules. Finally, the proposed method is applied to the reconfiguration and replacement of the front suspension frame module of a bus chassis. The results demonstrated and proved the effectiveness of this method.

A forward head and rounded shoulder posture is a poor posture that is widely seen in everyday life. It is known that sitting in such a poor posture with long hours will bring health issues such as muscle pain. However, it is not known whether sitting in this poor posture for a short period of time will affect human activities. This paper investigates the effects of a short-duration poor posture before some typical physical activities such as push-ups. The experiments are set up as follows. Fourteen male subjects are asked to do push-ups until fatigue with two surface electromyography (sEMG) at the upper limb. Two days later, they are asked to sit in this poor posture for 15 min with eight sEMG sensors located at given back muscles. Then they do the push-ups after the short-duration poor posture. The observations from the median frequency of sEMG signals at the upper limb indicate that the short-duration poor posture does affect the fatigue procedure of push-ups. A significant decreasing trend of the performance of push-ups is obtained after sitting in this poor posture. Such effects indicate that some parts of the back muscles indeed get fatigued with only 15 min sitting in this poor posture. By further investigating the time-frequency components of sEMG of back muscles, it is observed that the low and middle frequencies of sEMG signals from the infraspinatus muscle of the dominant side are demonstrated to be more prone to fatigue with the poor posture. Although this study focuses only on push-ups, similar experiments can be arranged for other physical exercises as well. This study provides new insights into the effect of a short-duration poor posture before physical activities. These insights can be used to guide athletes to pay attention to postures before physical activities to improve performance and reduce the risk of injury.

Recent research has shown that the entire southern rim of Amazonia was inhabited by earth-building societies involving landscape engineering, landscape domestication and likely low-density urbanism during the Late Holocene. However, the scale, timing, and intensity of human settlement in this region remain unknown due to the dearth of archaeological work and the logistical difficulties associated with research in tropical forest environments. A case in point are the newly discovered Mound Villages (AD ~1000–1650) in the SE portion of Acre State, Brazil. Much of recent pioneering work on this new archaeological tradition has mainly focused on the excavation of single mounds within sites with little concern for the architectural layout and regional settlement patterns, thus preventing us from understanding how these societies were organised at the regional level. To address these shortcomings, we carried out the first Lidar survey with a RIEGL VUX-1 UAV Lidar sensor integrated into an MD 500 helicopter. Our novel results documented distinctive architectural features of Circular Mound Villages such as the presence of ranked, paired, cardinally oriented, sunken roads interconnecting villages, the occurrence of a diversity of mound shapes within sites, as well as the exposure the superimposition of villages. Site size distribution analysis showed no apparent signs of settlement hierarchy. At the same time, it revealed that some small groups of villages positioned along streams exhibit regular distances of 2.5–3 km and 5–6 km between sites. Our data show that after the cessation of Geoglyph construction (~AD 950), this region of SW Amazonia was not abandoned, but occupied by a flourishing regional system of Mound Villages. The results continue to call into question traditional views that portray interfluvial areas and the western sector of Amazonia as sparsely inhabited. A brief discussion of our findings in the context with pre-Columbian settlement patterns across other regions of Amazonia is conducted.

Summary: Background: The unequal distribution of income is a fundamental determinant of health inequalities. Decision making around economic policies could be enhanced by showing their potential health effects. We used scenario modelling to assess the effects of 12 income-based policies on years of life lost (YLL) and inequalities in YLL in Scotland for the 2017–21 period. Methods: In this modelling study, we used EUROMOD version H1.0+, a tax-benefit microsimulation model, to estimate the effects of hypothetical fiscal policies on household income for Scottish households in the 2014/15 Family Resources Survey (n=2871). The effects were modelled excluding housing costs. Income change from baseline was estimated for each quintile of the 2016 Scottish Index of Multiple Deprivation (SIMD) after weighting to account for differential non-response to the Family Resources Survey, and incomes were equivalised according to the Organisation for Economic Co-operation and Development's modified equivalence scale. A regression analysis of cross-sectional data was used to estimate the relationship between income change and all-cause mortality, followed up by a sensitivity analysis to account for uncertainties around the assumptions on effect size. Informing Interventions to reduce health Inequalities (Triple I), a health inequalities scenario modelling tool, was used to estimate policy effects on YLL and government spending after five years of theoretical implementation. The Triple I model used population estimates for 2016 stratified by sex, 5-year age group, and SIMD quintile, which were obtained from the National Records of Scotland. Preliminary estimates of relative policy costs were calculated from the EUROMOD-derived combined effects of each policy on tax bills, National Insurance contributions, and benefits receipts for Scottish households. Findings: Taxation-based policies did not substantially affect household incomes, whereas benefits-based policies had large effects across the quintiles. The best policy for improving health and narrowing health inequalities was a 50% increase to means-tested benefits (approximately 105 177 [4·7%] YLL fewer than the baseline of 2·2 million, and a 7·9% reduction in relative index of inequality). Effects on YLL and health inequalities were inversely correlated in response to changes in taxation policy. Citizen's Basic Income (CBI) schemes also substantially narrowed inequalities (3·7% relative index of inequality for basic scheme, 5·9% for CBI with additional payments for individuals with disability), and modestly reduced YLL (0·7% for the basic scheme and 1·4% with additional payments). The estimated government spending associated with a policy was proportional to its effect on YLL, but less closely related to its effect on inequalities in YLL. Interpretation: Policies that affect incomes could potentially have marked effects on health and health inequalities in Scotland. Our projections suggest that the most effective policies for reducing health inequalities appeared to be those that disproportionately increased incomes in the most deprived areas. Although modelling was subject to various assumptions, the approach can be useful to inform decisions around addressing the upstream determinants of health inequalities. Funding: None.

Previous studies of Pre-Columbian earthworks in the Amazon basin have left a gap in the Upper Tapajós Basin (UTB). Here, the authors detect 104 Pre-Columbian earthworks in the UTB, suggesting continuous occupation across southern Amazonia and higher population densities than previously estimated.

Abstract Background Accurate data to measure population cigarette consumption are vital for surveillance and for evaluating the impact of tobacco control policies. This study uses cigarette retail sales data to provide novel insights into trends and patterns in cigarette consumption in Great Britain. Methods Cigarette sales estimates derived from electronic sales from most large grocery stores and a weighted representative sample of smaller convenience stores were obtained from Nielsen. Data on the number of cigarette sticks sold per calendar month and per week were obtained for Scotland and England/Wales (combined) for the period January 2008 to December 2015. Cigarette consumption per adult smoker per month was calculated using survey-based smoking prevalence estimates and mid-year population estimates. Results Population cigarette consumption in Great Britain declined between 2008 and 2013. Cigarette sales have since stabilised at around 400 cigarettes per adult smoker per month. Cigarettes sold in 14- to 19-packs have substituted a sharp decline in 20-packs and now account for over half of all cigarettes sold in Great Britain. Cigarette consumption has been consistently higher in Scotland than England/Wales. This is due to higher sales of 20-packs in Scotland between 2008 and 2013, which has been substituted by higher sales of 14- to 19-packs in recent years. Conclusions Cigarette retail sales data provide unique insights into levels and patterns of cigarette consumption and should be used for monitoring and evaluating tobacco control policy in Great Britain.

The role of plants in early human migrations across the globe has received little attention compared to big game hunting. Tropical forests in particular have been seen as a barrier for Late Pleistocene human dispersals due to perceived difficulties in obtaining sufficient subsistence resources. Archaeobotanical data from the Cerro Azul rock outcrop in the Colombian Amazon details Late Pleistocene plant exploitation providing insight into early human subsistence in the tropical forest. The dominance of palm taxa in the assemblage, dating from 12.5 ka BP, allows us to speculate on processes of ecological knowledge transfer and the identification of edible resources in a novel environment. Following the hypothesis of Martin Jones from his 2009 work, “Moving North: archaeobotanical evidence for plant diet in Middle and Upper Paleolithic Europe”, we contend that the instantly recognizable and economically useful palm family (Arecaceae) provided a “gateway” to the unknown resources of the Amazon forest.

Salamanders are unique among extant tetrapods for their ability to completely regenerate their limbs. Here, Nogueira and colleagues show that lungfishes, the sister clade of tetrapods, regenerate their fins using analogous gene regulatory changes and morphological steps.

From the perspective of Central and South America, the peopling of the New World was a complex process lasting thousands of years and involving multiple waves of Pleistocene and early Holocene period immigrants entering into the neotropics. These Paleoindian colonists initially brought with them technologies developed for adaptation to environments and resources found in North America. As the ice age ended across the New World people adapted more generalized stone tools to exploit changing environments and resources. In the neotropics these changes would have been pronounced as patchy forests and grasslands gave way to broadleaf tropical forests. We document a late Pleistocene/early Holocene stone tool tradition from Belize, located in southern Mesoamerica. This represents the first endogenous Paleoindian stone tool technocomplex recovered from well dated stratigraphic contexts for Mesoamerica. Previously designated Lowe, these artifacts share multiple features with contemporary North and South American Paleoindian tool types. Once hafted, these bifaces appear to have served multiple functions for cutting, hooking, thrusting, or throwing. The tools were developed at a time of technological regionalization reflecting the diverse demands of a period of pronounced environmental change and population movement. Combined stratigraphic, technological, and population paleogenetic data suggests that there were strong ties between lowland neotropic regions at the onset of the Holocene.

The ‘short’ neutral section is a feature of alternating current (AC) railway overhead line electrification that is often unreliable and a source of train delays. However hardly any dynamic analysis of its behaviour has been undertaken. This paper briefly describes the work undertaken investigating the possibility of modelling the behaviour using a novel approach. The potential for thus improving the performance of short neutral sections is evaluated, with particular reference to the UK situation. The analysis fundamentally used dynamic simulation of the pantograph and overhead contact line (OCL) interface, implemented using a proprietary finite element analysis tool. The neutral section model was constructed using physical characteristics and laboratory tests data, and was included in a validated pantograph/OCL simulation model. Simulation output of the neutral section behaviour has been validated satisfactorily against real line test data. Using this method the sensitivity of the neutral section performance in relation to particular parameters of its construction was examined. A limited number of parameter adjustments were studied, seeking potential improvements. One such improvement identified involved the additional inclusion of a lever arm at the trailing end of the neutral section. A novel application of pantograph/OCL dynamic simulation to modelling neutral section behaviour has been shown to be useful in assessing the modification of neutral section parameters.

Anthropogenic climate change driven by increased carbon emissions is leading to more severe fire seasons and increasing the frequency of mega-fires in the Amazon. This has the potential to convert Amazon forests from net carbon sinks to net carbon sources. Although modern human influence over the Earth is substantial, debate remains over when humans began to dominate Earth's natural systems. To date, little is known about the history of human land use in key regions like the Amazon. Here, we examine the history of human occupation from a ~8,500 year-old sediment core record from Lake Caranã (LC) in the eastern Amazon. The onset of pre-Columbian activity at LC (~4,500 cal yr B.P.) is associated with the beginning of fire management and crop cultivation, later followed by the formation of Amazonian Dark Earth soils (ADEs) ~2,000 cal yr B.P. Selective forest enrichment of edible plants and low-severity fire activity altered the composition and structure of forests growing on ADEs (ADE forests) making them more drought susceptible and fire-prone. Following European colonization (1661 A.D.), the Amazon rubber boom (mid-1800s to 1920 A.D.) is associated with record-low fire activity despite drier regional climate, indicating fire exclusion. The formation of FLONA Reserve in 1974 A.D. is accompanied by the relocation of traditional populations and a fire suppression policy. Despite suppression efforts, biomass burning and fire severity in the past decade is higher than any other period in the record. This is attributed to combined climate and human factors which create optimal conditions for mega-fires in ADE forests and threatens to transform the Amazon from a net carbon sink to a net carbon source. To help mitigate the occurrence of mega-fires, a fire management policy reducing fire-use and careful fire management for farming may help to reduce fuel loads and the occurrence of mega-fires in fire-prone ADE forests. As both natural and anthropogenic pressures are projected to increase in the Amazon, this study provides valuable insights into the legacy of past human land use on modern ADE forest composition, structure, and flammability that can inform ecological benchmarks and future management efforts in the eastern Amazon.

Hydroxychloroquine (HCQ) retinopathy can result in permanent vision loss. In early stages of HCQ retinopathy, patients are usually asymptomatic with preservation of visual acuity. We aspire that our review, in conjunction with the American Academy of Ophthalmology screening guidelines, shall shed light on effective screening measures utilizing multimodal imaging techniques to detect early signs of HCQ retinopathy before advanced changes manifest clinically.

A long held view about the occupation of southern proto-Jê pit house villages of the southern Brazilian highlands is that these sites represent cycles of long-term abandonment and reoccupation. However, this assumption is based on an insufficient number of radiocarbon dates for individual pit houses. To address this problem, we conducted a programme of comprehensive AMS radiocarbon dating and Bayesian modelling at the deeply stratified oversized pit House 1, Baggio I site (Cal. A.D. 1395-1650), Campo Belo do Sul, Santa Catarina state, Brazil. The stratigraphy of House 1 revealed an unparalleled sequence of twelve well preserved floors evidencing a major change in occupation dynamics including five completely burnt collapsed roofs. The results of the radiocarbon dating allowed us to understand for the first time the occupation dynamics of an oversized pit house in the southern Brazilian highlands. The Bayesian model demonstrates that House 1 was occupied for over two centuries with no evidence of major periods of abandonment, calling into question previous models of long-term abandonment. In addition, the House 1 sequence allowed us to tie transformations in ceramic style and lithic technology to an absolute chronology. Finally, we can provide new evidence that the emergence of oversized domestic structures is a relatively recent phenomenon among the southern proto-Jê. As monumental pit houses start to be built, small pit houses continue to be inhabited, evidencing emerging disparities in domestic architecture after AD 1000. Our research shows the importance of programmes of intensive dating of individual structures to understand occupation dynamics and site permanence, and challenges long held assumptions that the southern Brazilian highlands were home to marginal cultures in the context of lowland South America.

Sterilising immunity is a desired outcome for vaccination against human immunodeficiency virus (HIV) and has been observed in the macaque model using inactivated simian immunodeficiency virus (SIV). This protection was attributed to antibodies specific for cell proteins including human leucocyte antigens (HLA) class I and II incorporated into virions during vaccine and challenge virus preparation. We show here, using HLA bead arrays, that vaccinated macaques protected from virus challenge had higher serum antibody reactivity compared with non-protected animals. Moreover, reactivity was shown to be directed against HLA framework determinants. Previous studies failed to correlate serum antibody mediated virus neutralisation with protection and were confounded by cytotoxic effects. Using a virus entry assay based on TZM-bl cells we now report that, in the presence of complement, serum antibody titres that neutralise virus infectivity were higher in protected animals. We propose that complement-augmented virus neutralisation is a key factor in inducing sterilising immunity and may be difficult to achieve with HIV/SIV Env-based vaccines. Understanding how to overcome the apparent block of inactivated SIV vaccines to elicit anti-envelope protein antibodies that effectively engage the complement system could enable novel anti-HIV antibody vaccines that induce potent, virolytic serological response to be developed.

The efficacy of capecitabine as adjuvant therapy in colon cancer is well demonstrated and its lower toxicity rates when compared with 5-FU make it an increasingly more favourable option for patients. This case highlights the awareness of a potentially severe side effect related to the use of capecitabine, yet through the early identification of symptoms patients can be managed conservatively.

BackgroundLive attenuated simian immunodeficiency virus (SIV) vaccines represent the most effective means of vaccinating macaques against pathogenic SIV challenge. However, thus far, protection has been demonstrated to be more effective against homologous than heterologous strains. Immune correlates of vaccine-induced protection have also been difficult to identify, particularly those measurable in the peripheral circulation.Methodology/principal findingsHere we describe potent protection in 6 out of 8 Mauritian-derived cynomolgus macaques (MCM) against heterologous virus challenge with the pathogenic, uncloned SIVsmE660 viral stock following vaccination with live attenuated SIVmac251/C8. MCM provided a characterised host genetic background with limited Major Histocompatibility Complex (MHC) and TRIM5α allelic diversity. Early protection, observed as soon as 3 weeks post-vaccination, was comparable to that of 20 weeks vaccination. Recrudescence of vaccine virus was most pronounced in breakthrough cases where simultaneous identification of vaccine and challenge viruses by virus-specific PCR was indicative of active co-infection. Persistence of the vaccine virus in a range of lymphoid tissues was typified by a consistent level of SIV RNA positive cells in protected vaccinates. However, no association between MHC class I/II haplotype or TRIM5α polymorphism and study outcome was identified.Conclusion/significanceThis SIV vaccine study, conducted in MHC-characterised MCM, demonstrated potent protection against the pathogenic, heterologous SIVsmE660 challenge stock after only 3 weeks vaccination. This level of protection against this viral stock by intravenous challenge has not been hitherto observed. The mechanism(s) of protection by vaccination with live attenuated SIV must account for the heterologous and early protection data described in this study, including those which relate to the innate immune system.
