- Abstract
Objective: To determine whether a multicomponent intervention based on physical activity with technological support and nutritional counselling prevents mobility disability in older adults with physical frailty and sarcopenia. Design: Evaluator blinded, randomised controlled trial. Setting: 16 clinical sites across 11 European countries, January 2016 to 31 October 2019. Participants: 1519 community dwelling men and women aged 70 years or older with physical frailty and sarcopenia, operationalised as the co-occurrence of low functional status, defined as a short physical performance battery (SPPB) score of 3 to 9, low appendicular lean mass, and ability to independently walk 400 m. 760 participants were randomised to a multicomponent intervention and 759 received education on healthy ageing (controls). Interventions: The multicomponent intervention comprised moderate intensity physical activity twice weekly at a centre and up to four times weekly at home. Actimetry data were used to tailor the intervention. Participants also received personalised nutritional counselling. Control participants received education on healthy ageing once a month. Interventions and follow-up lasted for up to 36 months. Main outcome measures: The primary outcome was mobility disability (inability to independently walk 400 m in <15 minutes). Persistent mobility disability (inability to walk 400 m on two consecutive occasions) and changes from baseline to 24 and 36 months in physical performance, muscle strength, and appendicular lean mass were analysed as pre-planned secondary outcomes. Primary comparisons were conducted in participants with baseline SPPB scores of 3–7 (n=1205). Those with SPPB scores of 8 or 9 (n=314) were analysed separately for exploratory purposes. Results: Mean age of the 1519 participants (1088 women) was 78.9 (standard deviation 5.8) years. The average follow-up was 26.4 (SD 9.5) months. Aim To describe the methods and feasibility of the nutritional intervention carried out within the SPRINTT Randomized cotrolled trial. We also illustrate how nutrition interventionists identified participants at risk of malnutrition and the lessons learnt from the nutrition intervention. Findings SPRINTT nutrition intervention was well-received by the majority of the participants. It was mainly carried out using tailored nutrition counselling, but also other means of delivering the intervention were successfully used. Compared with a standard nutrition prescription, an individualized protocol to diagnose malnutrition and follow-up by tailored nutrition counselling helped achieve nutritional targets more effectively in spite of diversity of population in nutritional habits and in some cases reluctance to accept changes. Message The SPRINTT nutrition intervention was feasible and allowed flexibility to the varying needs and cultural differences of this heterogeneous population of frail, older Europeans. It may serve as a model to educate and improve nutrition among community-dwelling older people at risk of mobility limitations. Supplementary Information The online version contains supplementary material available at 10.1007/s41999-020-00438-4., Background The “Sarcopenia and Physical Frailty in Older People: Multicomponent Treatment Strategies” (SPRINTT) project sponsored a multi-center randomized controlled trial (RCT) with the objective to determine the effect of physical activity and nutrition intervention for prevention of mobility disability in community-dwelling frail older Europeans. We describe here the design and feasibility of the SPRINTT nutrition intervention, including techniques used by nutrition interventionists to identify those at risk of malnutrition and to carry out the nutrition intervention. Methods SPRINTT RCT recruited older adults (≥ 70 years) from 11 European countries. Eligible participants (n = 1517) had functional limitations measured with Short Physical Performance Battery (SPPB score 3–9) and low muscle mass as determined by DXA scans, but were able to walk 400 m without assistance within 15 min. Participants were followed up for up to 3 years. The nutrition intervention was carried out mainly by individual nutrition counseling. Nutrition goals included achieving a daily protein intake of 1.0–1.2 g/kg body weight, energy intake of 25–30 kcal/kg of body weight/day, and serum vitamin D concentration ≥ 75 mmol/L. Survey on the method strategies and feasibility of the nutrition intervention was sent to all nutrition interventionists of the 16 SPRINTT study sites. Results Nutrition interventionists from all study sites responded to the survey. All responders found that the SPRINTT nutrition intervention was feasible for the target population, and it was well received by the majority. The identification of participants at nutritional risk was accomplished by combining information from interviews, questionnaires, clinical and laboratory data. Although the nutrition intervention was mainly carried out using individual nutritional counselling, other assisting methods were used as appropriate. Conclusion The SPRINTT nutrition intervention was feasible and able to adapt flexibly to varying needs of this heterogeneous population. The procedures adopted to identify older adults at risk of malnutrition and to design the appropriate intervention may serve as a model to deliver nutrition intervention for community-dwelling older people with mobility limitations. MAGIC Collaboration: et al., M 87 is one of the closest (z = 0.004 36) extragalactic sources emitting at very high energies (VHE, E > 100 GeV). The aim of this work is to locate the region of the VHE gamma-ray emission and to describe the observed broad-band spectral energy distribution (SED) during the low VHE gamma-ray state. The data from M 87 collected between 2012 and 2015 as part of a MAGIC monitoring programme are analysed and combined with multiwavelength data from Fermi-LAT, Chandra, HST, EVN, VLBA, and the Liverpool Telescope. The averaged VHE gamma-ray spectrum can be fitted from ∼100 GeV to ∼10 TeV with a simple power law with a photon index of (−2.41 ± 0.07), while the integral flux above 300 GeV is (1.44±0.13)×10−12cm−2s−1. During the campaign between 2012 and 2015, M 87 is generally found in a low-emission state at all observed wavelengths. The VHE gamma-ray flux from the present 2012–2015M 87 campaign is consistent with a constant flux with some hint of variability (∼3σ) on a daily time-scale in 2013. The low-state gamma-ray emission likely originates from the same region as the flare-state emission. Given the broad-band SED, both a leptonic synchrotron self-Compton and a hybrid photohadronic model reproduce the available data well, even if the latter is preferred. Tomato landraces, originated by adaptive responses to local habitats, are considered a valuable resource for many traits of agronomic interest, including fruit nutritional quality. Primary and secondary metabolites are essential determinants of fruit organoleptic quality, and some of them, such as carotenoids and phenolics, have been associated with beneficial proprieties for human health. Landraces' fruit taste and flavour are often preferred by consumers compared to the commercial varieties' ones. In an autumn-winter greenhouse hydroponic experiment, the response of three Southern-Italy tomato landraces (Ciettaicale, Linosa and Corleone) and one commercial cultivar (UC-82B) to different concentrations of sodium chloride (0 mM, 60 mM or 120 mM NaCl) were evaluated. At harvest, no losses in marketable yield were noticed in any of the tested genotypes. However, under salt stress, fresh fruit yield as well as fruit calcium concentration were higher affected in the commercial cultivar than in the landraces. Furthermore, UC-82B showed a trend of decreasing lycopene and total antioxidant capacity with increasing salt concentration, whereas no changes in these parameters were observed in the landraces under 60 mM NaCl. Landraces under 120 mM NaCl accumulated more fructose and glucose in the fruits, while salt did not affect hexoses levels in UC-82B. Ultra-performance liquid chromatography-tandem mass spectrometry analysis revealed differential accumulation of glycoalkaloids, phenolic acids, flavonoids and their derivatives in the fruits of all genotypes under stress. Overall, the investigated Italian landraces showed a different behaviour compared to the commercial variety UC-82B under moderate salinity stress, showing a tolerable compromise between yield and quality attributes. Our results point to the feasible use of tomato landraces as a target to select interesting genetic traits to improve fruit quality under stress conditions.

Abstract Background Killer cell immunoglobulin-like receptors (KIRs) are a family of inhibitory and activatory receptors that are expressed by most natural killer (NK) cells. The KIR gene family is polymorphic: genomic diversity is achieved through differences in gene content and allelic polymorphism. The number of KIR loci has been reported to vary among individuals, resulting in different KIR haplotypes. In this study we report the genotypic structure of KIRs in 217 unrelated healthy Italian individuals from 22 immunogenetics laboratories, located in the northern, central and southern regions of Italy. Methods Two hundred and seventeen DNA samples were studied by a low resolution PCR-SSP kit designed to identify all KIR genes. Results All 17 KIR genes were observed in the population with different frequencies than other Caucasian and non-Caucasian populations; framework genes KIR3DL3, KIR3DP1, KIR2DL4 and KIR3DL2 were present in all individuals. Sixty-five different profiles were found in this Italian population study. Haplotype A remains the most prevalent and genotype 1, with a frequency of 28.5%, is the most commonly observed in the Italian population. Conclusion The Italian Caucasian population shows polymorphism of the KIR gene family like other Caucasian and non-Caucasian populations. Although 64 genotypes have been observed, genotype 1 remains the most frequent as already observed in other populations. Such knowledge of the KIR gene distribution in populations is very useful in the study of associations with diseases and in selection of donors for haploidentical bone marrow transplantation.

Drought stress limits the growth and yield of crops, affecting the source-sink relationships. A complex and dynamic signalling network, in which hormones, reactive oxygen species (ROS) and sugars are mainly involved, is activated by plants to cope with water shortage. Tolerant plants have developed several efficient adaptive mechanisms to establish new cell metabolism homeostasis avoiding and/or reducing permanent impairments triggered by drought. The Southern Italy landrace of tomato (Solanum lycopersicum L.) "Ciettaicale" and the well-known tomato cultivar "Moneymaker" were compared evaluating their biometrical and metabolic responses to 20 dayswater deficits (high and moderate) under controlled growth chamber conditions. In order to investigate the drought induced adaptive mechanisms of the two tomato genotypes, we evaluated in vivo chlorophyll a fluorescence, gas exchanges and leaf water potential (LYw), together with the analyses of non-structural carbohydrates in source and sink organs and with the hormonal and antioxidative responses. A decline in actual photochemical efficiency of photosystem II in the light (FPSII) associated with an enhancement of non-photochemical fluorescence quenching (NPQ) were more pronounced in Moneymaker than Ciettaicale stressed plants, despite the maintenance of high potential efficiency of PSII photochemistry (Fv/Fm) in any case. Gas exchanges revealed higher water use efficiency (WUE) in Ciettaicale comparing to Moneymaker thanks to more efficient CO2 assimilation capacity. Higher sucrose levels observed in Moneymaker leaf and root tissues comparing to Ciettaicale ones indicated a potential role of these compounds as osmoprotectant or a reduced export demand from sink organs. Similarly, the elevated levels of hydrogen peroxide (H2O2), lipid peroxidation and abscisic acid (ABA) in Moneymaker leaf and root tissues supported the hypothesis of a drought induced alteration of source-sink relationships. In addition, Ciettaicale showed higher root metabolic activity, as supported also by elevated indole-3-acetic acid (IAA) levels. Changes in the antioxidant pool, including enzymes and compounds like ascorbate peroxidase, ascorbate and phenols, and compatible osmolites, such as proline, played a key role to counteract redox and osmotic pressure in both genotypes. Overall, while Moneymaker showed a survival strategy leading to the accumulation of metabolites to maintain more negative LYw but at same time showing the disruption of source-sink balance, Ciettaicale increased WUE and maintained carbon translocation from source to sink supporting the investment in root metabolism and growth to escape from an environmental osmotic pressure.

We describe a new, state-of-the-art, Earth System Regional Climate Model (RegCM-ES), which includes the coupling between the atmosphere, ocean, and land surface, as well as a hydrological and ocean biogeochemistry model, with the capability of using a variety of physical parameterizations. The regional coupled model has been implemented and tested over some of the COordinated Regional climate Downscaling Experiment (CORDEX) domains and more regional settings featuring climatically important coupled phenomena. Regional coupled ocean-atmosphere models can be especially useful tools to provide information on the mechanisms of air-sea interactions and feedbacks occurring at fine spatial and temporal scales. RegCM-ES shows a good representation of precipitation and SST fields over the domains tested, as well as realistic simulations of coupled air-sea processes and interactions. The RegCM-ES model, which can be easily implemented over any regional domain of interest, is open source, making it suitable for usage by the broad scientific community.

A dwarf mutant, dwarf2 (dw2), was isolated from sunflower (Helianthus annuus). The most obvious alterations of dw2 plants were the lack of stem growth, reduced size of leaves, petioles and flower organs, retarded flower development. Pollen and ovules were produced but the filaments failed to extrude the anthers from the corolla. The dw2 phenotype was mainly because of reduced cell size. In dw2 leaves, the dark-green color was not so much due to higher pigment content, but was correlated with a changed leaf morphology. The mutant responded to the application of bioactive gibberellins (GAs). The levels of ent-7a-hydroxykaurenoic acid, GA19, GA20 and GA1 in dw2 seedlings were severely decreased relative to those in its wild type (WT). ent-Kaurenoic acid was actively converted to ent-7a-hydroxykaurenoic acid in WT plants but quite poorly in dw2 plants. All together these data suggested that the dw2 mutation severely reduced the flux through the biosynthetic pathway leading to active GAs by hampering the conversion of ent-kaurenoic acid to GA12. Two ent-kaurenoic acid oxidase (KAO) genes were identified. HaKAO1 was expressed everywhere in sunflower organs, while HaKAO2 was mainly expressed in roots. We demonstrated that a DNA deletion in HaKAO1 of dw2 generated aberrant mRNA-splicing, causing a premature stop codon in the amino acid sequence. In dw2 calli, Agrobacterium-mediated transfer of WT HaKAO1 cDNA restored the WT endogenous levels of GAs. In segregating BC1 progeny, the deletion co-segregated with the dwarf phenotype. The deletion was generated near to a breakpoint of a more complex chromosome rearrangement.

A new version of the RegCM regional climate modeling system, RegCM4, has been recently developed and made available for public use. Compared to previous versions, RegCM4 includes new land surface, planetary boundary layer, and air–sea flux schemes, a mixed convection and tropical band configuration, modifications to the pre-existing radiative transfer and boundary layer schemes, and a full upgrade of the model code towards improved flexibility, portability, and user friendliness. The model can be interactively coupled to a 1D lake model, a simplified aerosol scheme (including organic carbon, black carbon, SO4, dust, and sea spray), and a gas phase chemistry module (CBM-Z). After a general description of the model, a series of test experiments are presented over 4 domains prescribed under the CORDEX framework (Africa, South America, East Asia, and Europe) to provide illustrative examples of the model behavior and sensitivities under different climatic regimes. These experiments indicate that, overall, RegCM4 shows an improved performance in several respects compared to previous versions, although further testing by the user community is needed to fully explore its sensitivities and range of applications.
