Los métodos tradicionales para obtener líneas puras u homocigotas involucran varias generaciones de autofecundación con un requerimiento importante de tiempo. La obtención de plantas haploides y doble haploides, representa una alternativa que reduce a una generación, la producción de líneas con alelos idénticos en todos sus cromosomas. El objetivo de este trabajo fue implementar el protocolo de obtención de embriones androgénicos en poblaciones de chile mulato y ancho del programa de mejoramiento de chile en el Campo Experimental Bajío del Instituto Nacional de Investigaciones Forestales, Agrícolas y Pecuarias durante 2019 y 2020. Treinta genotipos de chile comprendidos en dos poblaciones. a) población criolla heterogénea de chiles mulato y ancho con característica de esterilidad masculina citoplasmática; y b) población de cruzas interespecíficas de chile ancho con chile habanero (C. annuum x C. chinense) resistentes a geminivirus (PepGMV) y Phytophthora sp., fueron sometidas a tratamiento de inducción androgénica. El tratamiento consistió en N 6 -furfuriladenina (cinetina) (0.01 mg L-1) y ácido 2,4-diclorofenoxiacético (2,4-D) (0.01 mg L-1) para la inducción de embriogénesis somática y posteriormente de cinetina (0.1 mg L-1) para la inducción de germinación de embriones. La duplicación cromosómica se realizó por medio de colchicina (0.5%) previo a su adaptación in vivo. La población conformada por genotipos derivados de cruzas interespecíficas obtuvo mayor eficiencia de plantas regeneradas doble haploides entre 4.29% y 14.67%, mientras que la población criolla generó embriones en menor proporción entre 1.02% y 5.26%. Estos resultados son los primeros reportados para obtención de plantas doble haploides de chiles mulato y ancho.

Phytophthora capsici, es el agente causal de la ‘marchitez del chile’ una de las enfermedades más importantes de este cultivo. Para establecer estrategias de control eficientes es necesario el conocimiento de la diversidad del patógeno, para esto, se realizó esta investigación cuyo objetivo fue determinar el grado de variabilidad de 29 aislados monozoospóricos obtenidos de chile (Capsicum annuum L.) de seis municipios del estado de Guanajuato y dos cepas de referencia de A1 y A2. Todos fueron caracterizados por la forma de crecimiento, tipo de compatibilidad, crecimiento a 35 °C, sensibilidad a mefenoxam y grado de virulencia. Los resultados obtenidos indican que los aislados, presentaron cinco formas de crecimiento: estrellada (62.5%), turuloso (15.62%), estolonífera (12.5%), filamentoso (6.25%) y petaloide (3.12%); su crecimiento a 35 °C fue menor a 30.7% respecto al control crecido a 27 °C, su tipo de compatibilidad fue 65.5% del tipo A1, se encontró sensibilidad intermedia al mefenoxam en 65.5% de los aislados, siendo el resto sensibles. El aislado más virulento fue el D3 y los avirulentos fueron D8, D11 y D12 todos originarios de Dolores Hidalgo. No se observa una relación directa entre la forma de crecimiento, el crecimiento a 35 °C, el tipo de compatibilidad, la resistencia a mefenoxam y el grado de virulencia.

Hydrogen peroxide is an important signal molecule in plant defense against biotic and abiotic stress. Pepper golden mosaic virus (PepGMV) is a whitefly-transmitted geminivirus causing significant yield and quality losses in pepper and other horticultural crops in Mexico. Several pesticides have been used trying to control whiteflies and thus, PepGMV disease in host crops. The present work examined the effect of exogenous application of hydrogen peroxide (H2O2) on inducing resistance to PepGMV infection in pepper plants. Experiments were carried out under greenhouse conditions aiming to evaluate phenotypical, biochemical and molecular features in these pepper plants. Hydrogen peroxide at 6, 14 and 18 mM induced tolerance to PepGMV either by absence of symptoms as well as by attenuating and/or delaying them. The protection observed was directly dependent on the concentration of H2O2 sprayed on plants. Moreover, PepGMV DNA levels were inversely proportional to the protection level. Enzymatic and gene expression profiles related with plant defense were induced in protected, in comparison to susceptible control plants. Interestingly, the levels of some phenolic compounds were also associated with plant protection. Taking together, these results suggested that exogenous foliar applications of H2O2 protect pepper plants against PepGMV infection inducing the plant host defense arsenal.

Chili pepper (Capsicum annuumL.) production is affected by wilt and root rot, the most devastating disease caused by the pathogen complex of oomycetePhytophthora capsiciLeon and the fungiFusarium oxysporumSchlecht andRhizoctonia solaniKühn, infecting roots, stems, leaves, and fruits. Fungicides are currently inefficient against this disease and have a high environmental impact. The use of elicitors is a sustainable alternative for inducing resistance to wilting and root rot. DNA fragments of an organism’s own origin (conspecific or self-DNA) have shown the ability to inhibit growth and activate defense mechanisms in some plant species. In this investigation, the effect of the fragmented DNA mixture ofPhytophthora capsiciL.,Fusarium oxysporumS., andRhizoctonia solaniK. on the protection against wilt and root rot ofCapsicum annuumL. plants was evaluated. Changes in plant performance, phenolics, and flavonoids contents, as well as gene expression involved in the production of defense metabolites after the fragmented and unfragmented DNA mixture in three concentrations (20, 60, and 100 μg mL–1) in chili peppers, were studied. The results obtained showed a decrease in plant height in 60 and 100 μg mL–1concentrations in absence of pathogens. Moreover, the treatment with fragmented DNA 100 μg mL–1showed significant increase in the content of phenolic compounds and total flavonoids as well as gene expression associated to plant defense in comparison with control plants. Interestingly, foliar application of DNA fragments of the pathogen complex to a concentration of 100 μg mL–1caused a 40% decrease in the mortality of infected plants with the pathogens at 30 days post-inoculation compared with control plants inoculated with the pathogen complex but not sprayed with DNA fragments. These results suggested a perspective for application of fragmented DNA of these pathogens at the agricultural level in crop protection strategies to cope with wilt and root rot inCapsicum.

La aplicación de elicitores induce una respuesta en el nivel de metabolitos secundarios y en la metilación del ADN de las plantas. Este trabajo tuvo como objetivo aplicar dos elicitores a plantas de estevia y evaluar el contenido de varios compuestos fenólicos y glucosidos de esteviol; así como, el nivel de metilación en plantas elicitada y no elicitada. Los fenólicos y glucosidos fueron determinados por HPLC y espectrofotométria mientras que la metilación por 5-mC DNA ELISA. Como resultado de la elicitación, se observó un ligero incremento de los ácidos clorogénico y ferulico. Los fenoles totales se incrementaron a los siete días después de la aplicación de los elicitores, sin mostrar ningún incremento a los 15 y 21 días. Por el contrario, los flavonoides y los taninos aumentaron significativamente a los 7, 15 y 21 días de la aplicación. El contenido del esteviosido y los rebaudiosidos A y C aumentaron 9.5, 5.3 y 3.3 veces más que el grupo control. A lo largo del experimento, el rebaudiosido C fue mayor en su concentración que la suma del esteviol y del rebaudiosido A. Se observó que las plantas elicitadas presentaron un nivel de metillación menor que las no elicitadas Por su alto contenido de rebaudiosido C, la estevia criollo, aquí utilizada, puede producir un edulcorante menos amargo que otros materiales. La elicitacion de estevia puede ser una estrategia para incrementar el rendimiento de edulcorantes.

Background : Attention to amaranth grains has increased in recent years due to the nutritional value of their seed proteins, which have high levels of the amino acid lysine. However, there is no detailed study describing the stages of seed development in Amaranthus hypochondriacus. Question : How are the developmental patterns of the female gametophyte and young seed in Amaranthus hypochondriacus ? Species studied : Amaranthus hypochondriacus L ’Revancha’ (Amaranthaceae). Study site and years of study : Plants were growth and collected from 2014 to 2016, in a greenhouse at Instituto Tecnologico de Celaya, Guanajuato, Mexico. Methods: Glomerules were collected before pollination and two weeks after anthesis. The ovules at different development stages were fixed and cleared and were analyzed by light microscopy. A clearing protocol was used to observe the developmental stages during female gametogenesis and embryogenesis. Results: We observed that the Amaranthus hypochondriacus ovule has a campylotropous form. The female gametophyte showed a Polygonum -type pattern of development. We were also able to identify all the stages from the megaspore mother cell to the cotyledon embryo stage. After meiosis, the micropylar megaspore differentiates into the functional megaspore. The embryo did not show symmetric divisions, although the final pattern is similar to that of in eudicotyledons. The suspensor showed additional longitudinal divisions, giving rise to a 2-rowed suspensor, while the endosperm showed a helobial development. Conclusions: These results will be used as baseline to identify morphological changes during seed development and to develop new strategies to improve seed quality or increase the yield.

El chile (Capsicum annuum L.) es un cultivo importante en México debido a su creciente demanda en los últimos años. Por lo que, se deben de buscar alternativas para aumentar su producción, una de ellas, es el uso de reguladores de crecimiento, de los cuales las giberelinas son importantes reguladores involucrados en el amarre y desarrollo de fruto, floración, elongación celular y expansión en plantas. Con el objetivo de evaluar el efecto de la aplicación de giberelinas (GA3) en el rendimiento de chile jalapeño producido en invernadero, se realizó la presente investigación en dos experimentos sucesivos. En el experimento 1 se evaluaron cinco dosis de giberelinas: 0 (testigo), 10, 30, 50 y 500 mg L-1 que se asperjaron en una y dos aplicaciones. La fertilización en este experimento fue con solución nutritiva (N, P, K y microelementos). En el experimento 2 se evaluaron las mismas dosis de GA3, también en una y dos aplicaciones, pero la fertilización se realizó con nitrato de calcio y nitrato de potasio solubles en agua. En el experimento 1 no se detectó un incremento en el rendimiento. En cambio, en el experimento 2, la dosis de 10 mg L-1 de GA3 en combinación con la fertilización con nitrato de calcio y nitrato de potasio incrementó el rendimiento de chile jalapeño. Los resultados muestran a las giberelinas como elementos clave combinados con baja fertilización para incrementar la producción de chile.

Una de las principales limitantes para la producción de chile son los hongos patógenos causantes de la enfermedad conocida como “marchitez del chile” o “secadera”. Esta enfermedad puede ser devastadora cuando las condiciones climáticas son favorables para el patógeno. A pesar de que se han intentado diferentes medios de control (químicos y culturales) ninguno ha tenido éxito. Una alternativa para su control es producir germoplasma resistente, sin embargo para poder establecer un programa de mejoramiento efectivo es necesario conocer la distribución y diversidad genética de los patógenos involucrados, particularmente de Rhizoctonia solani, que por su ubicuidad representa un peligro potencial en todas las zonas productoras. Por ello el objetivo fue caracterizar a R. solani en las zona Centro Norte de México y determinar su diversidad genética. Para cumplir con este objetivo se consideraron los estados de Chihuahua, Durango, Zacatecas, San Luis Potosí, Colima, Querétaro y Guanajuato donde en 2009 se colectaron plantas adultas de Chile con síntomas de marchitez, se aisló al hongo y se encontró una incidencia del 33%, encontrándose tanto en tallo como en raíz. Las células miceliales fueron multinucleadas, características de las cepas patogénicas. Las pruebas de anastomosis demostaron la presencia en México de los grupos GA4, GA-2.1, GA-IIB, GA-2IV, GA7, GA11, GA12 y GA13. La diversidad genética de este hongo fue muy alta, de tal manera que las relaciones demostradas por la construcción de dendrogramas no muestran tendencias homogéneas pues los principales grupos formados contienen elementos de todos los estados.

In the recent years, there has been a great necessity to increase crop productivity of staple crops in order to provide the food that is needed by a growing world population. Nowadays, not only crop yield is important, but also the ability to keep and, if possible, increase their bioactive compounds, the content of which is beneficial effects on human health, which have been widely reported. Nevertheless, various abiotic stresses affect crop productivity. Especially if the stress is prolonged, plant growth and productivity are severely diminished. In this sense, water scarcity constitutes a crucial constraint for agricultural productivity. Drought is one of the most serious abiotic stresses that restricts plant growth, development and productivity, and affects their physiological and biochemical mechanisms. However, there is evidence that a controlled water deficit (WD) may improve fruit quality through higher concentration of flavor compounds and phytochemicals. This review presents information about how a controlled WD may be used as an abiotic stress factor of crop phytochemicals, thereby improving health benefits for all consumers.

Plants require effective sensing and responses to environmental stimuli, for example in the case of biotic and abiotic stresses. Reactive oxygen species (ROS) are well-recognised key signal molecules for the stress response. In previous work in our group, several transgenic tobacco lines were generated constitutively expressing a germin-like gene from Capsicum chinense (CchGLP), producing higher endogen levels of hydrogen peroxide that correlated with tolerance to geminivirus infections and drought stress. Differential transcriptomic analysis between transgenic tobacco lines of low (L1) and high (L8) CchGLP gene expression showed key changes in differential profiles related to the crosstalk between basal metabolism, biochemical and stress-related pathways that might explain the observed tolerant phenotype. El cáncer bacteriano causado por Clavibacter michiganensis ssp. michiganensis (Cmm), es una enfermedad devastadora del jitomate alrededor del mundo. El objetivo del presente estudio fue describir un procedimiento rápido basado en PCR para la detección de Cmm patogénica en plantas de jitomate. El DNA fue aislado por el método de hervido y se realizó PCR para la amplificación del gen pat-1 (serina proteasa), empleando los oligonucleótidos CMM5F y CMM6R, cuyo tamaño de amplicón fue de 608 pb. El fragmento fue clonado, secuenciado y comparado con la base de datos del NCBI (Centro Nacional para la Información Biotecnológica), mostrando 100% de identidad con secuencias del gen pat-1 de Cmm. Adicionalmente, de la secuencia obtenida se diseñaron 2 pares de oligonucleótidos anidados para producir amplicones de 250 pb a una temperatura de alineamiento de 62 °C. Los resultados descritos en el presente trabajo permitirán la identificación rápida y precisa de Cmm por PCR en semilla y en tejido vegetal de jitomate, al reducir el tiempo de análisis de 24 a 4 h aproximadamente.

Resumen Introducción. Conocer la diversidad a nivel genético, químico y morfológico que existe entre individuos y poblaciones, es de gran utilidad en los programas de mejoramiento genético, ya que facilita la organización del material y la selección adecuada de genotipos superiores para el desarrollo de una población mejorada. Objetivo. El objetivo del presente trabajo fue caracterizar genética, química y agronómicamente líneas avanzadas de tomate de cáscara (Physalis ixocarpa Brot.), del programa de mejoramiento de hortalizas del Campo Experimental Bajío (CE-Bajío) del Instituto Nacional de Investigaciones Forestales, Agrícolas y Pecuarias (INIFAP), México. Materiales y métodos. Durante la temporada primavera-verano y otoño-invierno de 2017, se cuantificó la variabilidad genética con marcadores moleculares de ADN de tipo AFLP (Amplified Fragment Length Polymorphism); se determinaron los compuestos fenólicos y tomatidina en fruto, y en lo agronómico el porcentaje de germinación, número de frutos por planta, peso del fruto, diámetro ecuatorial y polar, y el rendimiento. Resultados. Se obtuvo un promedio general de similitud entre los genotipos de 0,86. De acuerdo con las relaciones genéticas, se detectaron las líneas 4 y 70 como posibles progenitores de híbridos y un patrón geográfico en los agrupamientos. La diversidad química indicó que el mayor contenido de flavonoides (51,1 mg EAG/100 g) se presentó en la L-86, de fenoles y antocianinas (396, 8 y 7,22 mg EAG/100 g, resp.) en la L-182, y de taninos (188,4 mg EAG/100 g) en la L-97, mientras que la tomatidina fue mayor en frutos verdes (2,23-3,81 mg EAG/100 g) que en morados. El intervalo de rendimiento de las líneas fue de 11,4 a 47,6 t/ha, donde el 20% de las líneas superaron la media de rendimiento nacional (40 t/ha). Se destacó la L-37 con 47,6 t/ha, que además produjo mayor número de frutos por planta, diámetro ecuatorial y polar, y una tasa de germinación de 93,3%. Conclusión. Las líneas 37, 25, 27 y 167, tienen potencial para ser validadas para uso comercial y como líneas parentales. Abstract Introduction. Knowledge about the genetic, chemical and morphological diversity that exists between individuals and populations is very useful in breeding programs, because it facilitates the organization of the material and the appropriate selection of superior genotypes for the development of an improved population. Objective. The aim of this research was to make a genetic, chemical and agronomic characterization in twenty advanced husk tomato lines (Physalis ixocarpa Brot.), belonging to the vegetable breeding program of the Campo Experimental Bajío (CE-Bajío), of the Instituto Nacional de Investigaciones Forestales, Agrícolas y Pecuarias (INIFAP), México. Materials and methods. During the spring-summer and autumn-winter 2017, the genetic variability was quantified with molecular markers of DNA, type AFLP (Amplified Fragment Length Polymorphisms), likewise, the phenolic and tomatidine compounds in the fruit were determined, and agronomic traits like germination percentage, number of fruits per plant, weight of the fruit, equatorial and polar diameter per fruit, and fruit yield were determined. A general average of similarity between the genotypes of 0.86 was obtained. Results. A general similitude mean between genotypes of 0.86 was obteined. According to the genetic relationships, a geographic pattern was identified and genotypes 4 and 70 were detected as possible progenitors of improved hybrids. Chemical diversity indicated that the fruits flavonoids content in L-86 was the highest (51.1mg EAG/100g), phenols (396.8 mg EAG/100 g) and anthocyanin’s (7.22 mg EAG/100g) for L-182 and tannins (188.4 mg EAG/100 g) for L-97, while tomatidine (2.23-3.81 mg EAG/100 g) was higher in green fruits than purple fruits. The agronomic results indicate that the lines fruit yield ranged from 11.4 to 47.6 t/ha, the 20% of the lines has a superior yield than the national mean yield (40 t/ha), The L-37 was noticeable, since it has the highest fruit yield with 47.6 t/ha, and has the highest number of fruits number per plant, equatorial and polar diameters, and germination rate (93.3%). Conclusion. Based on the results the lines 37, 25, 27 and 167 are positioned as lines with potential for commercial use and as parental lines.

Methanolic extracts of Acaciella angustissima pods (MEA) were analyzed in order to determine some biological activities such as antioxidant, antimutagenic and antimicrobial properties by 1,1-diphenyl-2-picrylhydrazyl (DPPH), 2,2′-azinobis-3 ethylbenzothiazoline-6-sulfonic acid (ABTS), ferric reducing antioxidant power (FRAP), Kado microsuspension assay and in vitro antimicrobial assays, respectively. Moreover, a transcriptomic study of Timbe grown in greenhouse conditions was also performed using subtractive suppression hybridization methodology (SSH). MEA presented significant antioxidant activity by DPPH, ABTS and FRAP methods. The maximum concentration of MEA inhibited 80.33% and 77.39% of mutagenicity induced either by 4-nitro-O-phenylenediamine or sodium azide, respectively. MEA presented fungistatic activity against Fusarium oxysporum, Rhizoctonia solani and Phytophtora capsici. Moreover, a good in vivo protection against P. capsici was obtained in comparison to a commercial antifungal used toward this pathogen. Transcriptomic study displayed the expression of genes that have biological importance, such as protein TIC55, GATA transcription factor, maturase-like protein, and NAC domain protein. NAC domain protein might have some biotechnological or industrial application, because, NAC transcription factor plays an important role in responses to biotic and abiotic stress from some plants. Results suggest that Timbe might be useful in pharmaceutical, food and agro-industrial sectors.

Hydrogen peroxide (H2O2) is an important ROS molecule (Reactive oxygen species) that serves as a signal of oxidative stress and activation of signaling cascades as a result of the early response of the plant to biotic stress. This response can also be generated with the application of elicitors, stable molecules that induce the activation of transduction cascades and hormonal pathways, which trigger induced resistance to environmental stress. In this work, we evaluated the endogenous H2O2 production caused by salicylic acid (SA), chitosan (QN), and H2O2 elicitors in Capsicum annuum L. Hydrogen peroxide production after elicitation, catalase (CAT) and phenylalanine ammonia lyase (PAL) activities, as well as gene expression analysis of cat1, pal, and pathogenesis-related protein 1 (pr1) were determined. Our results displayed that 6.7 and 10 mM SA concentrations, and, 14 and 18 mM H2O2 concentrations, induced an endogenous H2O2 and gene expression. QN treatments induced the same responses in lesser proportion than the other two elicitors. Endogenous H2O2 production monitored during several days, showed results that could be an indicator for determining application opportunity uses in agriculture for maintaining plant alert systems against a stress.

Common bean (Phaseolus vulgaris L.) is the most important grain legume for direct human consumption; however, bean production is affected by several diseases such as Rhizoctonia root rot. Few bean cultivars have been identified that effectively resist the attack of this fungus. Herein, we used the P. vulgaris Pv-2094 landrace, which is less susceptible to Rhizoctonia root rot, for the construction of a suppressive subtractive hybridization cDNA library in order to isolate plant defense-related genes. Total RNAs obtained after 8 and 16 h from inoculated and non-inoculated roots with R. solani Kühn, were used as the source of the "tester" and the "driver" samples, respectively. A total of 136 unigenes were obtained and classified into 12 functional categories. Six unigenes were selected to analyze for differential expression by qRT-PCR, including a receptor-like kinase (PvRK20-1), an acid phosphatase associated to defense (PA), a pathogenesis related protein (PR1), an ethylene responsive factor (ERF), a polygalacturonase inhibitor protein (PGIP), and an alpha-dioxygenase (α-DOX). These genes were found to be differentially expressed in a time-dependent manner in bean roots during the interaction with R. solani. Data generated from this study will contribute to the understanding of the molecular mechanisms associated with plant defense against root rot in common bean.

Cascalote (Caesalpinia cacalaco) is a tree located in Pacific Mexican coast, and it is an excellent source of phenolics as gallic and tannic acids used in Mexican tannery industry, and with a potent antioxidant and antimutagenic activities. Based on the phenolics properties, these compounds present potential antimicrobial activity against a myriad of microorganisms. Thus, in this study it was evaluated the antimicrobial activity of cascalote phenolics against phytopathogenic fungus Colletotrichum lindemuthianum which causes anthracnose disease in common beans (Phaseolus vulgaris). The results indicate that cascalote phenolics have in vitro fungistatic activity against C. lindemuthianum races R-0 and R-1472. Moreover, these compounds provoked on the whole, in vitro inhibition of spores germination and cellulase and polygalacturonase activities in these fungi. In vivo assays with cascalote phenolics under greenhouse conditions using susceptible cv. PI 206272 of common bean resulted in a good protection against anthracnose severity especially as a preventive treatment. Several aspects of the results and implications on the potential use of cascalote phenolics as organic antifungal are discussed.

An organogenic regeneration protocol in common bean (Phaseolus vulgaris L.) using embryo-axes derived from mature seeds from commercial cultivars Flor de Junio Marcela (FJM) and Flor de Mayo Anita (FMA) is reported. Embryonic axes were excised from surface sterilized seeds and established in MS medium containing different benzylaminopurine (BAP) and adenine (A) concentrations. Efficient regeneration was achieved when 5 or 10 mg/l BAP was added to the medium regardless of the A concentration, inducing the formation of differentiated cells as bud clusters. These clusters mainly formed at the internodal segment of the embryo for FJM and at the apical segment for FMA embryonic axes. Regeneration efficiency varied considerably between the two cultivars where FJM showed higher bud cluster formation (90%) and full plant regeneration (83%), whereas FMA showed 18% bud cluster formation, and 50% full plant regeneration. The division and transfer of FMA and FJM bud clusters to induction media every 15 days resulted in the formation of three to five new bud clusters each transfer. A single 5 mm cluster formed up to 15 shoots from which 1–2 plants were regenerated. Genetic variation analysis by amplified fragment length polymorphism (AFLP) showed no variation between regenerated plants.

Although several reports have described the occurrence of the teleomorphic state of Glomerella lindemuthiana (anamorph, Colletotrichum lindemuthianum), there has been a lack of continuity in this research. To identify G. lindemuthiana isolates capable of developing the teleomorphic state, 19 Mexican isolates were analyzed. Three types of response were observed: (i) negative, where only mycelial growth with or without acervuli was observed; (ii) potential, where in addition to the above, spherical perithecia-like structures were observed; (iii) positive, where perithecia containing asci and ascospores were observed. All strains were self-sterile and only one combination of strains produced fertile perithecia. From this fertile combination 168 individual ascospore cultures were isolated, including five from a single ascus. Forty-four monoascospore cultures were characterized with AFLP, confirming that these individuals were progeny from a sexual cross between the original two G. lindemuthiana isolates and that sexual reproduction in G. lindemuthiana is heterothallic in nature. Analysis of the parental strains with degenerate PCR primers indicated that sequences homologous to the HMG box of the MAT1-2 idiomorph are present in both parental isolates. This supports previous observations in other Glomerella species where the standard ascomycete configuration of distinct idiomorphs at the MAT locus does not hold true. The significance of these results is discussed.

There is an increasing awareness and interest in the antioxidant behaviour and potential health benefits of phenolic acids. The identification of novel sources of phenolic acids has been also of scientific interest. Cascalote (Caesalpinia cacalaco) pods are known to be a good source of ‘tannins’, the name by which industry in Mexico recognizes phenolic extract. Phenolics were determined as gallic acid equivalents g−1. The antimutagenic activity against aflatoxin B1 and the antioxidant activity, using two different methods, of the extract were also evaluated. Gallic acid accounts for almost 90% of the phenolic extract of cascalote, the remaining 10% was tannic acid. Antimutagenic activity of cascalote phenolics was dose-dependent, showing an inhibition level of 64.42% at the highest dose assayed. Antioxidant and antiradical activities were also dose-dependent. The highest antioxidant activity showed by cascalote phenolics was 73.5%, higher than that of Trolox. The highest antiradical activity of cascalote phenolics was 75.3%, higher than that of BHT and Trolox. Cascalote pods are an outstanding source of gallic and tannic acids. The pathotypes of 17 isolates of Colletotrichum lindemuthianum from the central region of Mexico were characterized to determine the genetic relationship among isolates from this region and other regions of Mexico, and to evaluate the resistance present in the elite germ plasm collection of Phaseolus vulgaris at INIFAP. Eight pathotypes were identified, including pathotype 292, which is reported for the first time in Mexico. The lack of isolates infecting cultivar TU carrying the Co-5 resistance gene suggests that this cultivar is a useful source of resistance. Six pathotypes produced susceptible reactions on only differential cultivars of Middle American origin, one pathotype on a single cultivar of Andean origin, and one pathotype on cultivars of both Middle American and Andean origin. Comparison of amplified fragment length polymorphism (AFLP) genotypes of 21 isolates confirmed suggestions that populations of C. lindemuthianum are comprised of asexually reproducing clonal lineages. Analysis of five different pathotypes of C. lindemuthianum on 21 elite genotypes of P. vulgaris identified four genotypes from different races of P. vulgaris resistant to all five pathotypes. This information will allow breeders and farmers to select the resistant genotypes most suited to their needs.

En la región de el Bajío en el centro de México, el rendimiento del garbanzo (Cicer arietinum L.) es limitado por enfermedades de la raíz como la marchitez vascular causada Fusarium oxysporum f. sp. ciceris (FOC). El conocimiento de las razas existentes de este patógeno permitirá usar los cultivares resistentes apropiados. El objetivo fue identificar las razas de FOC existentes en El Bajío, estado de Guanajuato por medio de marcadores moleculares específicos. Se analizaron aislados provenientes de cinco municipios productores de garbanzo (Celaya, Salvatierra, Valle de Santiago, Huanímaro y Pénjamo). Se probaron 12 iniciadores, siete de secuencia arbitraria tipo RAPD (OP1-01, OPI-09, OPI-18, OPF-06, OPF-10, OPF-12 y OPF-16) asociados a las razas 0, 1A, 1B/C, 2, 3, 4, 5 y 6 de F. oxysporum f. sp. ciceris , y cinco de secuencia tipo SCAR (FocR0-M15, FocR1B/C-N5, FocR5-L10, FocR6-O2, FocR6-P18-FocR0-M15), procedentes de los fragmentos marcadores raciales tipo RAPD inicialmente identificados. Los marcadores señalaron la presencia de cuatro razas: 0, 1B/C, 4 y 6, de FOC; las razas de mayor frecuencia fueron la 0 y 1B/C, que inducen el síntoma de amarillamiento, y la raza 6 que induce el síntoma de marchitez. Con las razas identificadas en Guanajuato y la raza 5 aislada en el estado de Sinaloa, se realizó una prueba de patogenicidad sobre las líneas diferenciales: JG 62, Sanford, P 2245, CRIL 1-53, CRIL 1-94, CRIL 1-17, CRIL 1-36, y WR 315, por el método de inmersión de raíces. Los resultados de la inoculación verificaron la identificación de razas con los marcadores. Las diferenciales resistentes fueron CRIL 1-36, WR 315, y CRIL 1-17, las que pueden utilizarse como progenitores en el mejoramiento de la resistencia contra FOC en México.

Macrophomina phaseolina infects a wide range of plant species. In the case of Phaseolus vulgaris (the common bean), the fungus causes charcoal rot which can lead to severe losses especially under arid conditions. In order to combat the disease, it is necessary to have information on the population structure of the pathogen in terms of pathogenesis on different species which may be grown simultaneously or successively within a region. Previous reports have demonstrated the heterogeneity of the pathogen both in terms of morphological characteristics and molecular genotype; however no systematic attempt has been made to compare pathogenesis of different isolates in relation to genotype, species of origin or geographical location. With this aim 84 isolates of M. phaseolina from different plant species in different geographical regions of Mexico were analysed by inoculation on a set of P. vulgaris cultivars differing in reaction to the fungus and by determination of their amplified fragment length polymorphism (AFLP) genotype. Forty three distinct pathotypes were identified, only two of which were found in more than one species. All 84 isolates had distinct AFLP genotypes and cluster analysis showed a tendency of the isolates to form groups related to geographical origin or species of origin.

La pudrición blanca, una de las enfermedades más preocupantes para los productores de ajo (Allium sativum), es causada por el hongo fitopatógeno Sclerotium cepivorum Berk. Con el fin de desarrollar procedimientos de biología molecular para el hongo, en este trabajo se describe un método de transformación empleando fusión de protoplastos en presencia de polietilenglicol 3350 (PEG). Se empleó el plásmido pAN7-1 que confiere resistencia a higromicina B, el cual contiene el gen higromicina fosfotransferasa B (hph) de Escherichia coli bajo el promotor gliceraldehído fosfato deshidrogenasa (gpdA) y el terminador antranilato sintetasa (trpC) del hongo filamentoso Aspergillus nidulans. Las transformantes recuperadas en el medio de cultivo de regeneración papa dextrosa agar-sorbitol (PDA-S) en presencia de 200 μgmL-1 de higromicina B se analizaron amplificando un fragmento del gen bacteriano hph mediante PCR con oligonucleótidos específicos: Hig 1, Sentido 5 ́ GAC CTG CTG AGG TCC CTC 3 ́ y Hig 2, antisentido 5 ́ GCC CTC GGA CGA GTG CTG 3 ́. Se obtuvieron frecuencias de 12-18 transformantes de S. cepivorum por μg de ADN vector. Los resultados descritos en el presente trabajo contribuirán en la caracterización molecular de genes involucrados en el fenómeno de patogénesis de S. cepivorum sobre el ajo.

Chili pepper blight observed on pepper farms from north Aguascalientes was monitored for the presence ofPhytophthora capsiciduring 2008–2010. Initially, ELISA tests were directed to plant samples from greenhouses and rustic nurseries, showing an 86% of positive samples. Later, samples of wilted plants from the farms during the first survey were tested with ELISA. The subsequent survey on soil samples included mycelia isolation and PCR amplification of a 560 bp fragment of ITS-specific DNA sequence ofP. capsici. Data was analyzed according to four geographical areas defined by coordinates to ease the dispersal assessment. In general, one-third of the samples from surveyed fields containedP. capsici, inferring that this may be the pathogen responsible of the observed wilt. Nevertheless, only five sites from a total of 92 were consistently negative toP. capsici. The presence of this pathogen was detected through ELISA and confirmed through PCR. The other two-thirds of the negative samples may be attributable toFusariumandRhizoctonia, both isolated instead ofPhytophthorain these areas. Due to these striking results, this information would be of interest for local plant protection committees and farmers to avoid further dispersal of pathogens to new lands.

A germin-like gene (CchGLP) cloned from geminivirus-resistant pepper (Capsicum chinense Jacq. Line BG-3821) was characterized and the enzymatic activity of the expressed protein analyzed. The predicted protein consists of 203 amino acids, similar to other germin-like proteins. A highly conserved cupin domain and typical germin boxes, one of them containing three histidines and one glutamate, are also present in CchGLP. A signal peptide was predicted in the first 18 N-terminal amino acids, as well as one putative N-glycosylation site from residues 44–47. CchGLP was expressed in E. coli and the recombinant protein displayed manganese superoxide dismutase (Mn-SOD) activity. Molecular analysis showed that CchGLP is present in one copy in the C. chinense Jacq. genome and was induced in plants by ethylene (Et) and salicylic acid (SA) but not jasmonic acid (JA) applications in the absence of pathogens. Meanwhile, incompatible interactions with either Pepper golden mosaic virus (PepGMV) or Pepper huasteco yellow vein virus (PHYVV) caused local and systemic CchGLP induction in these geminivirus-resistant plants, but not in a susceptible accession. Compatible interactions with PHYVV, PepGMV and oomycete Phytophthora capsici did not induce CchGLP expression. Thus, these results indicate that CchGLP encodes a Mn-SOD, which is induced in the C. chinense geminivirus-resistant line BG-3821, likely using SA and Et signaling pathways during incompatible interactions with geminiviruses PepGMV and PHYVV.

En México la enfermedad de raíz más importante del cultivo de chile es la marchitez, esta se controla principalmente con fumigantes y fungicidas que contribuyen a seleccionar aislados resistentes, y provocan daños al ambiente y a la salud. Una opción inocua con el ambiente podría ser el cultivo de variedades resistentes, sin embargo, hay pocas variedades con resistencia a esta enfermedad. El objetivo del presente trabajo fue aislar los patógenos asociados a la marchitez del chile, en la región centro y norte de México e identificar genotipos de chile resistentes. Durante 2006 y 2007, se colectaron plantas de chile con síntomas de marchitez en 118 lotes de los estados de Chihuahua, Colima, Durango, Guanajuato, Querétaro, San Luis Potosí y Zacatecas, a partir de las cuales se aislaron los patógenos y se obtuvieron cultivos puros. Estos se inocularon individualmente o en mezcla para seleccionar germoplasma resistente en 44 accesiones de chile del banco de germoplasma del INIFAP y 141 colectas procedentes de Durango, Guanajuato, Michoacán, San Luis Potosí y Zacatecas. Fusarium spp. fue aislado con una frecuencia de 42.6%, Rhizoctonia solani 37%, y Phytophthora capsici 3.9%. Se identificaron 26 colectas con al menos un individuo resistente a Fusarium spp., y seis a R. solani. Sólo las accesiones BG102 y BG107 del banco de germoplasma fueron resistentes a P. capsici y a la mezcla de los tres patógenos. Estos materiales tienen potencial para usarse en programas de mejoramiento genético del chile.

"El hongo Colletotrichum gloeosporioides, es el agente causal de antracnosis y representa la principal limitante para la cosecha de frutos con calidad comercial de aguacate Hass (Persea americana). La diversidad genética poblacional de este hongo se ha estudiado en diversas partes del mundo, pero en México se desconoce su estructura poblacional y particularmente los componentes de su diversidad. Se determinó la diversidad genética, patogénica y morfológica de 21 cepas monoconidiales de Colletotrichum gloeosporioides de Michoacán, México. La diversidad genética se estimó por medio del polimorfismo de ADN amplificado al azar y por la determinación de los perfiles de isoenzimas de cada cepa monoconidiales. La diversidad patogénica se midió mediante la capacidad invasiva de pulpa de aguacate y por la tasa de crecimiento in vitro. La diversidad morfológica in vitro se determinó por la comparación de caracteres del micelio de cada cepa monoconidiales. Los resultados mostraron alta diversidad de Colletotrichum gloeosporioides con un índice de variabilidad genética de 0.3031 y los estimadores más precisos fue el polimorfismo de ADN amplificado al azar (índice de variabilidad genética igual a 0.344) y perfiles de isoenzimas (índice de variabilidad genética igual a 0.35). Las características típicas fueron micelio blanco, consistencia algodonosa, colonia blanca, crecimiento de micelio al azar e hifas cortas en el borde de crecimiento. No se encontró relación entre la patogenicidad medida como capacidad invasiva de pulpa y la velocidad de crecimiento in vitro. Los análisis de agrupamiento establecieron que las relaciones entre cepas se dan principalmente con base en la localidad de origen y de manera secundaria, en el síntoma producido en el fruto."

La roya del tallo (Puccinia graminis Persoon f. sp. avenae) disminuye la productividad de avena en México. Las variedades que actualmente se siembran son susceptibles a esta enfermedad. El objetivo de este estudio fue buscar e identificar marcadores moleculares ligados a genes de resistencia a la roya del tallo en cuatro cultivares de avena. En primavera-verano de 2004 se evaluó en campo, en siete localidades, la reacción a la roya del tallo, y en 2005 en invernadero, la reacción a ocho aislados de roya del tallo colectados en diversas localidades del territorio nacional, de las variedades Chihuahua, Obsidiana, Cevamex y Karma. Se utilizaron 30 iniciadores con regiones NBSLRR en el genoma de la avena y 12 ligados a genes de resistencia a roya en gramíneas. La identificación de los marcadores se realizó con base en el concepto de genes análogos de resistencia (RGAs) y el mapa comparativo de la especie. Los resultados de la evaluación mostraron la susceptibilidad de la variedad Chihuahua; Obsidiana y Cevamex fueron moderadamente resistentes y Karma resistente; esta última mostró el mayor número (17) de marcadores polimórficos ligados a genes de resistencia. Los iniciadores: PIC11-K2/PIC11-WEL, Kinase2 D-E/ KQCFA 3-4, PLOOP1-4/WMA1-4, Kinase2 D-E/WMA1- 4, NBS B, F2/R2 y Hv3Lrk, pueden ser indicadores de los genes que confieren resistencia a la roya del tallo.

Quebracho extracts are used in tannery due to their high concentration of phenolics. The Mexican tannery industry uses around 450 kg/m 3 of which, 150 kg/m 3 remains in wastewaters and are discharged in drain pipe systems or rivers. The quebracho phenolics recovered from tannery wastewater (QPTW) was characterized by HPLC. The antimutagenic and antioxidant activities as well as the microbiological quality were evaluated. Total phenolic content of QPTW was 621 mg catechin equivalent/g sample. Gallic and protocatechuic acids were the major components characterized by HPLC. QPTW showed an inhibition range on aflatoxin B 1 mutagenicity from 16 to 60% and was dose-dependent. Antioxidant activity (defined as β-carotene bleaching) of QPTW (64.4%) at a dose of 12.3 mg/mL was similar to that of BHT (68.7%) at a dose of 0.33 mg/mL, but lower than Trolox (90.8% at a dose of 2.5 mg/mL); meanwhile antiradical activity (measured as reduction of DPPH) (60.8%) was higher than that of BHT (50.8%) and Trolox (34.2%). Quebracho residues were demonstrated to be an outstanding source of phenolic acids and for research and industrial uses.

Ornithine decarboxylase from respiratory fungal pathogen, Coccidioides immitis, cloned in the pETCiODC plasmid under control of T7lac promoter, was produced in E. coli BL21(DE3), BL21(DE3)pLysS, BLR(DE3) and EWH319 transformant strains. E. coli BL21(DE3)pLysS-pETCiODC expressed the highest specific activity of ODC, suggesting that this strain could be successfully used for protein structure and drug testing studies.

El chile (Capsicum annuum) es uno de los cultivos con mayor tradición en mesoamérica, ya que constituye un elemento básico dentro de la dieta de la población. Sin embargo, los rendimientos en México son bajos, debido en gran parte al ataque por agentes fitopatógenos, siendo Phytophthora capsici uno de los más importantes, ocasionando pérdidas entre 60-100% por la enfermedad conocida como marchitez del chile, junto con un complejo fúngico en el que recurrentemente se aíslan Rhizoctonia solani y Fusarium spp. Con la finalidad de detectar la presencia de P. capsici y prevenir daños por marchitez, se desarrolló una estrategia basada en PCR para el diagnóstico de esta enfermedad. Se emplearon iniciadores específicos para el gen que codifica para la cutinasa de P. capsici, obteniéndose amplificaciones específicas tanto en extracciones de DNA a partir de micelio, así como de plantas infectadas. Asímismo, se lograron determinar condiciones de amplificación específicas para diferenciar la presencia de P. capsici en plantas de chile en las que además se presentaron R. solani y Fusarium spp.

Trypsin inhibitors (TI), tannins, and lectins appear to have a role in preventing chronic diseases in humans. The genetic variability of these traits in common bean needs to be ascertained in order to increase levels through breeding. The variability of TI, tannin, and lectins was determined in five bean cultivars grown at five locations in Mexico. TI and tannins contents in colored beans that belong to the Jalisco race were higher (11.1-11.9 trypsin units inhibited (TUI)/mg and 29.0-38.1 mg catechin equivalent (CE)/g, respectively) than cultivars of the Durango race (7.9-8.3 TUI/mg and 16.8-19.9 CE/mg, respectively). Bayo Victoria, a Durango race cultivar, had three times more lectins than levels reported for soybean. Cultivar influenced TI and tannins contents (p < 0.001), whereas site affected lectins (p < 0.001). An increase in levels of TI and tannins could be enhanced through breeding.

Colletotrichum gloeosporioides presenta una alta variabilidad morfológica, patogénica y genética, lo cual ha sido difícil de evaluar usando los métodos tradicionales. En los últimos años, las técnicas moleculares han facilitado la separación y caracterización genética. En este trabajo se evaluó la diversidad genética de 21cepas monospóricas de C. gloeosporioides provenientes de frutos de papaya de Veracruz, Chiapas y Campeche, México. Se utilizó la técnica molecular RAPD para caracterizar la variabilidad genética del hongo, para lo cual se extrajo el DNA de cada una de las cepas. Se usaron 18 oligonucleótidos decámeros de secuencias aleatorias, los cuales se hicieron reaccionar con el DNA de cada una de las cepas para determinar la variabilidad. Se obtuvieron en total 134 bandas polimórficas; el OPX-16 presentó el menor número de bandas que fue de 4, en contraste, el OPX-04 y el OPM-20 que presentaron 12, siendo el mayor número de bandas. Se obtuvo un dendrograma en el cual se observó la formación de dos grupos principales de cepas (A y B) ampliamente separados, con un valor de 100% de asociación entre los agrupamientos. En el grupo A se asociaron las cepas Ca2, Ca3 y Ca5, las cuales genéticamente presentan características completamente diferentes al resto de las cepas agrupadas en B. En general, se observó una tendencia de agrupamientos de acuerdo a su lugar de procedencia.

El frijol tipo Flor de Junio, cuyo color de grano es de fondo crema con franjas rosas, se produce y consume en la región Centro de México. Se describe el proceso de obtención y características de Dalia, una nueva variedad de frijol tipo Flor de Junio de raza Jalisco, derivada de la cruza de Flor de Junio Silvia por Flor de Mayo Anita realizada en el Campo Experimental Bajío en 2003. El proceso de selección se basó inicialmente en la sanidad y el ciclo de la planta, así como en las características comerciales del grano en siembras bajo condiciones alternas de riego y temporal. En su fase final se realizó selección asistida por marcadores moleculares (SAM) para seleccionar la progenie que tuviera simultáneamente los marcadores ROC11 y SW13, ligados a los genes I y bc3, cuya piramidación confieren resistencia a todos los patotipos de los virus BCMV y BCMNV. Dalia es de hábito de crecimiento indeterminado tipo 3, con guías de tamaño medio y ciclo de cultivo intermedio de 105 días de siembra a la madurez fisiológica, y resistente a BCMV y BCMNV. En un ensayo nacional de rendimiento y adaptación, Dalia mostró un rendimiento similar a Flor de Junio Marcela, la variedad dominante en el mercado, pero en contraste con ésta, una nula interacción genotipo por ambiente, lo que indicó amplia adaptación y estabilidad de rendimiento. Dalia se recomienda para producción bajo riego y temporal en las regiones de El Bajío y el Altiplano del centro de México.

Malacomeles denticulata es un fruto nativo de México al que recientemente se le ha encontrado características funcionales para proponerlo como una opción frutal. Esta investigación tuvo como objetivo elucidar la variabilidad de doce poblaciones de M. denticulata mediante marcadores Inter Secuencias Simples Repetidas o Intermicrosatélites (ISSR). Todos los ISSR presentaron altos valores en el Contenido de Información Polimórfica (PIC, por sus siglas en inglés) y el índice de diferenciación poblacional de Nei (GST), así como altos porcentajes de polimorfismo. Se crearon tres grupos de variabilidad, donde siete poblaciones de Guanajuato y la población del Tepozán, Querétaro, conformaron un grupo; mientras que las poblaciones de Agua Zarca (Guanajuato), La Joya (Querétaro) y Santa Catarina del Monte (México) conformaron otro. Finalmente, la población de San Miguel Tlaixpan (México) quedó separada como un grupo atípico. De acuerdo con el Análisis Molecular de Varianza (Amova), la variabilidad intrapoblacional representa el 47% y la interpoblacional el 53% de la variabilidad total, lo que concuerda con la variabilidad en caracteres de semilla, pero difiere a lo reportado para marcadores SSR.
