Emerging antimicrobial-resistant (AMR) Candida auris presents a formidable global health challenge, causing severe healthcare-associated infections (HAIs) with high mortality rates. Its ability to colonize surfaces and resist standard disinfectants undermines traditional hygiene practices, prompting an urgent need for new strategies. Ultraviolet C (UVC) light offers a promising approach with rapid and broad-spectrum germicidal efficacy. This review examines current literature on UVC LED technology in combating C. auris, highlighting its effectiveness, limitations, and applications in healthcare hygiene. UVC light has potent activity against C. auris, with up to 99.9999% inactivation depending on certain conditions such as microbial load, type of organism, surface, environmental, equipment, and UVC radiation factors. UVC LEDs can effectively combat C. auris, driving down healthcare costs and reducing attributable global mortality. Here, we explore implementation strategies for the targeted disinfection of high-risk areas and equipment, air handling units (AHUs), and water treatment systems. Challenges associated with UVC LED disinfection devices in healthcare settings, current performance limitations, and radiation safety are discussed. This will help in optimizing application protocols for effective disinfection and radiation safety. To further strengthen healthcare facility hygiene practices and curb the global spread of C. auris, recommendations for integrating UVC LED disinfection into infection control programs are shared. Here, we present the complete genome sequence of Streptomyces sp. strain CCM_MD2014 (phylum Actinobacteria), isolated from surface soil in Woods Hole, MA. Its single linear chromosome of 8,274,043 bp in length has a 72.13% G+C content and contains 6,948 coding sequences.

Here, we present the 3,443,800-bp complete genome sequence of Curtobacterium sp. strain MR_MD2014 (phylum Actinobacteria). This strain was isolated from soil in Woods Hole, MA, as part of the 2014 Microbial Diversity Summer Program at the Marine Biological Laboratory in Woods Hole, MA.

Ultraviolet-C (UV-C) germicidal light can effectively inactivate airborne pathogens and mitigate the transmission of infectious diseases. As the application of UV-C for disinfection gains popularity, practical estimation of UV irradiance is essential in determining the UV fluence (dose) and designing tubular UV lamp configurations for indoor air treatment. It is generally understood that the inverse square (~1/d²) law (i.e., irradiance is proportional to the inverse square of the distance) applies well to point light sources. However, there has been a recognition that the ~1/d² law does not work well for tubular light sources in the commonly defined near-field applications where the UV source is relatively close to the treated air. Therefore, practical near-field irradiation estimation is needed for designing portable air cleaners and heating, ventilation, and air conditioning (HVAC) ducts with built-in UV light bulbs. This research investigated UV-C light irradiance from tubular (L = 0.9 m) light bulbs at near distances inside an air cleaner prototype duct under three power output (1-, 4-, and 8-bulb) scenarios and conducted theoretical estimation based on a line-source irradiation model. Similarly sized visible fluorescent bulbs were used as a reference. The data were fitted on both ~1/d² and ~1/d correlation of irradiance with distance. Both measured and line source estimated data fit better (i.e., evaluated by R-square, standard errors, root mean squared errors) with the ~1/d than the ~1/d² relationship in the near distance. Although the differences between the measured and the modeled were observed, the pattern of light distribution generally follows an inverse relationship (~1/d) with distances (d) shorter than two tubular bulb lengths (d < 2L). The pattern applies to both UV and visible light tested in this study. It is recommended that the inverse (~1/d) correlation be used for near-distance estimation of light distribution, especially for disinfection purposes in air ducting for indoor air quality improvement and airborne disease mitigation. Background: Due to the increase in multidrug-resistant pathogens, it is important to investigate further antimicrobial options. In order not to have to work directly with pathogens, the investigation of possible surrogates is an important aspect. It is examined how suitable possible surrogate candidates for ESKAPE pathogens are for UVC applications. In addition, the inactivation sensitivities to 222 and 254 nm radiation are compared in relation. Methods: Non-pathogenic members (Enterococcus mundtii, Staphylococcus carnosus, Acinetobacter kookii, Pseudomonas fluorescens and Escherichia coli) of genera of ESKAPE strains were photoinactivated in PBS with irradiation wavelengths of 222 and 254 nm (no non-pathogenic Klebsiella was available). Log reduction doses were determined and compared to published photoinactivation results on ESKAPE pathogens. It was assumed that non-pathogenic bacteria could be designated as surrogates for one wavelength and one ESKAPE strain, if the doses were between the 25 and 75% quantiles of published log reduction dose of the corresponding pathogen. Results: For all non-pathogen relatives (except A. kookii), higher average log reduction doses were required for irradiation at 222 nm than at 254 nm. Comparison by boxplot revealed that five of eight determined log reduction doses of the possible surrogates were within the 25 and 75% quantiles of the data for ESKAPE pathogens. The measured log reduction dose for non-pathogenic E. coli was above the 75% quantile at 222 nm, and the log reduction dose for S. carnosus was below the 25% quantile at 254 nm. Conclusion: For more than half of the studied cases, the examined ESKAPE relatives in this study can be applied as surrogates for ESKAPE pathogens. Because of lack of data, no clear statement could be made for Enterococcus faecalis at 222 nm and Acinetobacter baumannii at both wavelengths. [ABSTRACT FROM AUTHOR]

Background: The emergence of resistance to antimicrobials by pathogens has reached crisis levels, calling for identification of alternative means to combat diseases. Objective: To determine antimicrobial activity of crude methanolic extract of Aloe secundiflora Engl. from Lake Victoria region of Kenya. Materials and Methods: Extract was tested against four strains of mycobacteria (Mycobacterium tuberculosis, M. kansasii, M. fortuitum and M. smegmatis), Salmonella typhi, Staphylococcus aureus, Pseudomonas aeruginosa, Escherichia coli, Klebsiella pneumoniae and a fungus Candida albicans. activity of the extract was determined using BACTEC™ MGIT™ 960 system. General antibacterial and antifungal activity was determined using standard procedures: zones of inhibition, Minimum Inhibitory Concentrations (MICs) and Minimum Bactericidal/Fungicidal Concentrations (MBCs/MFCs). Results: The extract was potent against M. fortuitum, M. smegmatis and M. kansasii where it completely inhibited growth (Zero growth units (GUs)) in all the extract concentrations used. It gave strong antimycobacterial activity (157 GUs) against M. tuberculosis. It showed strong antimicrobial activity (P⩽0.05), giving inhibition zones ⩾9.00 mm against most microorganisms, such as P. aeruginosa (MIC 9.375 mg mL-1 and MBC of 18.75 mg mL-1), E. coli (both MIC and MBC of 18.75 mg mL-1), S. aureus and S. typhi (both with MIC and MBC of 37.5 mg mL-1). Preliminary phytochemistry revealed presence of terpenoids, flavonoids and tannins. Conclusion: The data suggests that Aloe secundiflora could be a rich source of antimicrobial agents. The result gives scientific backing to its use by the local people of Lake Victoria region of Kenya, in the management of conditions associated with the tested microorganisms. [ABSTRACT FROM AUTHOR]

Purpose: To determine the potential benefits of nine medicinal plants used by the Samburu community for the treatment of tuberculosis. Methods: The extract was tested against four strains of Mycobacteria namely; Mycobacterium tuberculosis (Mtb), M. Kansasii (Mk), M. fortuitum (Mf), and M. smegmatis (Ms) using BACTEC MGIT 960 system. The crude extracts were also analyzed for the presence of phytochemical constituents. Results: Both the extracts of Scadoxus multiflorus and Acacia nilotica showed strong antimycobacterial activity against the four tuberculosis-causing strains. Eurphobia scarlatina was the most active against both the slow (Mtb and Mk) and the fast (Mf and Ms) growers with Zero GUs at 0.5mg/ml. Phytochemical screening indicated presence or absence of tannins, saponins and flavonoids, terpenoids, cardiac glycosides and alkaloids in the extracts. Conclusion: The data suggest that some of the methanol extracts could be a rich source of antituberculosis agents. The results further show that there is some merit in the use of some of the plants studied in alternative medical practice. The purpose of the study was to investigate the antifungal, antibacterial and antimycobacterial properties of methanol extract of Entada abysinnica steudel ex. A. Rich (Fabaceae) leaves used by herbalists from the Lake Victoria region, Kenya. The extract was tested against four strains of mycobacteria (Mycobacterium tuberculosis, Mycobacterium kansasii, Mycobacterium fortuitum, and Mycobacterium smegmatis) using BACTEC Mycobacteria Growth Indicator Tube (MGIT) 960 system and the proportional method. Standard procedures were used to determine the zones of inhibition, minimum inhibitory concentrations (MICs) and minimum bactericidal/fungicidal concentrations (MBCs/MFCs) for Candida albicans, Salmonella typhi, Staphylococcus aureus, Pseudomonas aeruginosa, Escherichia coli, and Klebsiella pneumoniae. The extract showed activity against some mycobacteria strains, especially M. tuberculosis. It also showed strong antimicrobial activity (zones of inhibition were between 9.00 and 14.10 mm) against C. albicans, Sa. typhi, and St. aureus. The extract gave a better zone of inhibition against C. albicans than fluconazole whose zone of inhibition was 13.00 mm. The MICs and MBCs for C. albicans and Sa. typhi were good. The crude extracts were also analyzed for the presence of phytochemicals. Phytochemical screening indicated that the extract most abundantly contained tannins, saponins, and flavonoids. The data suggest that the methanolic leaves extract of E. abysinnica could be a rich source of antimicrobial agents, especially antifungals. The results further show that there is some merit in the use of the plant in alternative medical practices. Antibiotic resistance poses a grave global public health threat, exacerbated by widespread and often inappropriate antibiotic usage. Vigilant surveillance of antibiotic utilization and emergence of antimicrobial resistance (AMR) is essential. Of particular concern in the era of AMR is the persistent issue of chronic wound infections. To address this, we conducted a comprehensive evaluation of wound isolates from chronic wounds at Jaramogi Oginga Odinga Teaching and Referral Hospital (JOOTRH) in Kenya, to identify relevant bacteria and assess their drug resistance patterns.Wound samples were collected and processed using standard microbiological methods. Bacterial isolates were identified and assessed for their susceptibility to a panel of antibiotics using the Kirby-Bauer disk diffusion method. A total of 103 bacterial isolates were obtained from the wound samples, with a higher prevalence in male patients (59%). Staphylococcus aureus (20.7 %) emerged as the most predominant pathogen, followed by Klebsiella spp. (14.8 %), Pseudomonas aeruginosa spp. (14.8 %) and Escherichia coli (4.4 %) in wound samples. High levels of antibiotic resistance were observed among the isolates, with the highest resistance rates reported for cotrimoxazole (48.1 %), clindamycin (25.9 %) and erythromycin (25.9 %). Furthermore, among the isolates, 75 % produced haemolysin and protease, while 50 % produced lipase and phospholipase, factors that enhance virulence and survival. The findings of this study highlight the alarmingly high prevalence of antibiotic resistance among bacterial pathogens isolated from chronic wounds in Kenya. This poses a major challenge to the effective management of chronic wound infections.
