Le CBL (Club des Bactéries Lactiques) est une manifestation scientifique qui réunit chercheurs, enseignants-chercheurs et industriels R&D, pour échanger sur les avancées scientifiques et techniques réalisées dans le domaine des bactéries lactiques. Les thèmes abordés sont le reflet des larges potentiels de ces bactéries et de leurs applications : fermentation alimentaire, santé humaine et animale, probiotique, environnement…; International audience; En fabrication fromagère, les bactéries lactiques (BL) et propioniques sont des actrices clés de la production de métabolites conférant les qualités nutritionnelles et organoleptiques aux fromages. Pourtant, la contribution de chaque espèce à la qualité finale du fromage n’est pas totalement élucidée. L’objectif était de déterminer quelles espèces contribuent à acidifier et produire des composés d’arôme, par quelles voies métaboliques, selon quelle temporalité et aussi d’investiguer les interactions métaboliques bactériennes contribuant au fonctionnement de l’écosystème fromager.Nous avons séquencé et annoté : Lactococcus lactis subsp. lactis biovar diacetylactis CIRM-BIA1206 (LL), Lactiplantibacillus plantarum CIRM-BIA465 (LP), Propionibacterium freudenreichii CIRM-BIA122 (PF). Nous avons reconstruit les voies métaboliques et élaboré un modèle métabolique de communauté. Un fromage à pâte pressée non cuite sans croûte a été réalisé avec ces bactéries et analysé pendant la fabrication et l’affinage (4 réplicats). Des dénombrements bactériens, des dosages de sucres, d’acides organiques et de composés d’arôme [1] ont été réalisés ainsi qu’un séquençage des ARN bactériens. L’analyse des gènes différentiellement exprimés a permis de montrer à quel moment de la fabrication étaient induits les gènes impliqués dans le catabolisme du lactose et du citrate et dans les voies de synthèse de différents composés d’arômes : acides lactique, acétique, propionique, isovalérique (arôme ‘vieux fromage’), diacétyle (arôme beurré). Le catabolisme du lactose était opéré chez LL via la voie du tagatose pendant l’acidification puis via la voie de Leloir et uniquement par cette dernière voie chez LP. La production de diacétyle était effectuée par LL dès le début de la fabrication puis plus modérément par LP. Les synthèses d’acides propionique et isovalérique ont été attribuées à PF et les métabolismes correspondants induits pendant la phase d’affinage. Enfin, les voies de fermentation mixtes ayant été induites pendant l’affinage, l’acide acétique a été vraisemblablement produit par les trois espèces à cette étape. L’implémentation du modèle métabolique communautaire dans l’outil Smetana [2] a révélé les bases moléculaires du commensalisme précédemment évoqué entre BL et PF [3–5]. Les BL produisent de l’acide lactique et potentiellement du glycérol, de la sérine et de la phénylalanine au profit de PF. Ces interactions identifiées in silico restent à valider in vitro.L’ensemble de ces résultats font de la métatranscriptomique associée aux modèles métaboliques, des outils de choix pour mieux comprendre et maîtriser les métabolismes et les interactions régissant le fonctionnement des écosystèmes fromagers.

The formation of cheese flavor mainly results from the production of volatile compounds by microorganisms. We investigated how fine-tuning cheese-making process parameters changed the cheese volatilome in a semi-hard cheese inoculated with

Backgrounds and Aims. Inflammatory Bowel Diseases (IBD), including Ulcerative Colitis (UC), coincide with alterations in the gut microbiota. Consumption of immunomodulatory strains of probiotic bacteria may induce or prolong remission in UC patients. Fermented foods, including cheeses, constitute major vectors for bacteria consumption. New evidences revealed anti-inflammatory effects in selected strains of Propionibacterium freudenreichii. We thus hypothesized that consumption of a functional cheese, fermented by such a strain, may exert a positive effect on IBD. Methods. We investigated the impact of cheese fermented by P. freudenreichii on gut inflammation. We developed an experimental single-strain cheese solely fermented by a selected immunomodulatory strain of P. freudenreichii, CIRM-BIA 129. We moreover produced, in industrial conditions, an Emmental cheese using the same strain, in combination with Lactobacillus delbrueckii CNRZ327 and Streptococcus thermophilus LMD-9, as starters. Consumption of both cheeses was investigated with respect to prevention of Dextran Sodium Sulphate (DSS)-induced colitis in mice. Results. Consumption of the single-strain experimental cheese, or of the industrial Emmental, both fermented by P. freudenreichii CIRM-BIA 129, reduced severity of subsequent DSS-induced colitis, weight loss, disease activity index and histological score. Both treatments, in a preventive way, reduced small bowel Immunoglobulin A (IgA) secretion, restored occludin gene expression and prevented induction of Tumor Necrosis Factor &alpha, (TNF&alpha, ), Interferon &gamma, (IFN&gamma, ) and Interleukin-17 (IL-17). Conclusions. A combination of immunomodulatory strains of starter bacteria can be used to manufacture an anti-inflammatory cheese, as revealed in an animal model of colitis. This opens new perspectives for personalised nutrition in the context of IBD.

The From'Innov cheesemaking process was recently suggested to increase process control and flexibility while reducing the use of energy and fresh water. It focuses on building cheese texture and aroma separately, in a shorter time than any other cheese technology, by enriching a liquid precheese with aromatic matrices fermented by traditional ripening microorganisms. This study assesses the economic benefits, and the use of energy and fresh water of the From'Innov process through predictive analysis. A singleday production of industrial bloomy soft cheese (Camembert-type) was simulated in three distinct virtual plants (100,000 kg of milk per day) using different technological approaches: the From'Innov, the Maubois-Mocquot-Vassal (MMV, another liquid pre-cheese process) or the traditional processes. With its higher margin and its lower specific energy and fresh water consumption, the From'Innov process appeared to be the most efficient.

Milk is often subjected to technological treatments which have impacts on the structure of milk constituents and the characteristics of rennet curds. In this paper, the influence of the dairy fat structure on the biochemical and textural characteristics of curds coagulated by an extract of Calotropis procera leaves was studied. Standardized milks were reconstituted with the same contents in protein (35 g·kg− 1) and fat (35 g·kg− 1) but with different structures of fat i.e. homogenized anhydrous milk fat (HAMF), homogenized cream (HC) and non-homogenized cream (NHC). As expected, the size distributions of fat globules in the different milks were different. After their coagulations by the plant extract, the physico-chemical characteristics of the curds and respective wheys were determined. No difference was observed in the coagulation time between the three milks but the whey removed more quickly from HAMF and HC curds than NHC-curd. The biochemical analyses of curds revealed a lower content in dry matter and fat in the NHC-curd compared to HAMF- and HC-curds. Otherwise, the NHC-whey exhibited the highest amount of fat. Observations by confocal microscopy showed that the fat globules were homogenously distributed and well trapped in the protein networks of HAMF- and HC-curds. In the NHC-curd, the fat globules were located in whey pockets, with less connectivity with the protein network. The textural analysis showed that the NHC-curd was more elastic, soft and adhesive than HAMF- and HC-curds. Homogenization significantly reduced the loss of fat during cheese manufacturing and conferred specific textural characteristics to the curds coagulated by an extract of Calotropis procera.

The heat-stable protease Ser2 is secreted by the species Serratia liquefaciens, a psychrotrophic bacteria frequently found in raw milk. To understand the physicochemical modifications of casein micelles induced by Ser2 and to confirm its implication in UHT milk destabilization, the enzyme was purified and added to microfiltered raw milk before UHT treatment. UHT milk destabilization was investigated during 90 days of storage. A visual destabilization appeared after 8 days of storage with the presence of sediment. Zeta potential increase and formation of aggregates were observed during the storage. Using tandem mass spectrometry, numerous released peptides from the four caseins were identified at the end of storage. Caseins were hydrolyzed in the preferential order β- > αs1- > κ- > αs2. No specific peptidic hydrolysed bond was detected. The present study confirmed that the presence of the protease Ser2 in raw milk can be one of the main causes of UHT milk destabilization.

International audience; In cheese production, lactic acid bacteria (LAB) and propionic bacteria are key players in the production of metabolites that confer nutritional and organoleptic qualities. However, the contribution of each species to the final quality of cheeses is not fully elucidated. The objective was to determine which species contribute to acidify and produce flavour compounds, by which metabolic pathways, according to which temporality and also to investigate the bacterial metabolic interactions contributing to the functioning of the cheese ecosystem.We sequenced and annotated: Lactococcus lactis subsp. lactis biovar diacetylactis CIRM-BIA1206 (LL), Lactiplantibacillus plantarum CIRM-BIA465 (LP), Propionibacterium freudenreichii CIRM-BIA122 (PF). We reconstructed the metabolic pathways and developed a community metabolic model. Four semi-hard cheeses were made with LL, LP and PF and were analyzed throughout manufacturing. Bacteria, sugars, organic acids and flavour compounds were quantified and RNA sequenced.The analysis of differentially expressed genes showed at which moment of the manufacturing process the genes involved in the catabolism of lactose and citrate and in the synthesis pathways of different flavour compounds: lactic, acetic, propionic, isovaleric (old cheese flavour), acetoin (cheesy flavour) acids were induced. Lactose catabolism was induced in LP and PF (Leloir pathway) during acidification and then in LL (tagatose pathway) during ripening. Acetoin production was induced in LL, LP and PF from the beginning of the manufacturing process to the beginning of ripening. The synthesis of propionic and isovaleric acids were attributed to PF and the corresponding metabolisms were induced from the beginning of the manufacturing then stably expressed during the first month of the ripening. Since the mixed fermentation pathways were induced during ripening, acetic acid was likely produced by LL, LP and PF at this stage. Implementation of the metabolic community model in the Smetana tool revealed the molecular basis of the previously discussed commensalism between LAB and PF. LAB produce lactic acid and potentially ribose, succinate, glycerol, serine and phenylalanine for the benefit of PF. These interactions, identified in silico, remain to be validated in vitro.All these results make metatranscriptomics associated with metabolic models, tools of choice to better understand and control the metabolisms and interactions governing the functioning of cheese ecosystems.
