Soil salinization is considered as one of the most important risks for agricultural soils. The objective of this experiment was to study the ecophysiological and the biochemical behaviour of two cultivars of Olea europaea L., 'Chemlali' and 'Koroneiki' under two salinity levels (100 and 200 mM NaCl) and the potential alleviation induced by gibberellic acid (GA3) foliar sprays. Salinity treatments significantly decreased photosynthetic assimilation rate and stomatal conductance compared to the control for both cultivars, but ‘Chemlali’ showed a higher resistance to increasing NaCl salinity compared to 'Koroneiki'. Leaf chlorophyll index also reduced gradually with increasing salinity concentration compared to the control. At the end of the experiment, a decrease in growth and dry matter accumulation was observed. Under high salinity stress, a significant decrease in root DW was recorded by 37% and 59% for ‘Chemlali’ and ‘Koroneiki’, respectively. High salinity stress decreased also shoot DW up to 51% for ‘Chemlali’. However, mannitol concentration increased under increasing salinity levels compared to control for ‘Chemlali’ cultivars. Interestingly, foliar application of GA3 alleviated the negative effects of salinity on ecophysiological parameters especially for ‘Koroneiki’. Indeed, GA3 improved photosynthetic assimilation up to 14% for ‘Chemlali’ and 36% for ‘Koroneiki’ compared to high salinity treatment. Both cultivars showed an increase in leaf chlorophyll index after applying GA3. Under high salinity combined with GA3, growth and dry weight were increased compared to salt stressed plants without GA3. The obtained results report that ‘Chemlali’ cultivar is more tolerant to salinity than ‘Koroneiki’ and suggest that GA3 plays an important role to reduce negative effects of NaCl salinity.

An efficient regeneration protocol was applied to regenerate shoots on salt stress-tolerant calli lines of aubergine (Solanum melongena). These NaCl-tolerant cell lines were obtained by two different methods. On the one hand, the developed callus tissue was transferred to a medium with a continuous salt content of 40, 80, 120, or 160 mM NaCl. On the other hand, the callus tissue was subjected to a stepwise increasing salinity to 160 mM NaCl every 30 days. With the second method, calli which could be selected were characterized by compact growth, a greenish color, and absence of necrotic zones. When grown on salt-free medium again, NaCl-tolerant calli showed a decline in relative growth rate and water content in comparison to the control line. This was more obvious in the 120 mM NaCl-tolerant callus. Lipid peroxidase activity increased in 40 and 80 mM NaCl-tolerant calli; yet did not increase further in 120 mM-tolerant callus. An increase in ascorbic acid content was observed in 80 and 120 mM NaCl-tolerant calli compared to the 40 mM NaCl-tolerant lines, in which ascorbic acid content was twice that of the control. All NaCl-tolerant lines showed significantly higher superoxide dismutase (SOD) (208–305–370 µmol min−1 mg−1 FW) and catalase (CAT) (136–211–238 µmol min−1 mg−1 FW) activities compared to control plants (231 and 126 µmol min−1 mg−1 FW). Plants were regenerated on the calli lines that could tolerate up to 120 mM NaCl. From the 32 plants tested in vitro, ten plants with a higher number of leaves and root length could be selected for further evaluation in the field. Their high salt tolerance was evident by their more elevated fresh and dry weight, their more increased relative water content, and a higher number and weight of fruits compared to the wild-type parental control. The presented work shows that somaclonal variation can be efficiently used to develop salt-tolerant mutants.

Hybridization in flowering plants depends, in the first place, on the delivery of pollen to a receptive stigma and the subsequent growth of pollen tubes through the style to the ovary, where the sperm nucleus of the pollen grain can ultimately fertilize the egg cell. However, reproductive failure is often observed in distant crosses and is caused by pre- and/or post-zygotic barriers. In this study, the reproductive pre-fertilization barriers of intertribal crosses between Anemone coronaria L. and Ranunculus asiaticus L., both belonging to the Ranunculaceae, were investigated. Despite the incongruity of intertribal crosses between A. coronaria and R. asiaticus having been of low intensity at the stigmatic level, interstylar obstructions of the pollen tube growth occurred, which confirmed the presence of pre-fertilization barriers. We show that these barriers could be partially bypassed by combining pollination with a stigma treatment. More specifically, a significantly higher ratio of the pollen tube length to the total style length and a better seed set were observed when the stigma was treated with the auxin 2,4-dichlorophenoxyacetic acid (2,4-D, 1 mg·mL−1) together with the cytokinin kinetin (KIN, 0.5 mg·mL−1) 24 h after pollination, irrespective of the cross direction. More specifically, the stigma treatments with any form of auxin (combined or not combined with cytokinin) resulted in a full seed set, assuming an apomictic fruit set, because no pollination was needed to obtain these seeds.

The impact of salinity on the physiological and biochemical parameters of tolerant ('Bonica') and susceptible ('Black Beauty') eggplant varieties (

Rose is the most economically important ornamental plant. However, cold stress seriously affects the survival and regrowth of garden roses in northern regions. Cold acclimation was studied using two genotypes (Rosa wichurana and R. hybrida ‘Yesterday’) selected from a rose breeding program. During the winter season (November to April), the cold hardiness of stems, soluble sugar content, and expression of dehydrins and the related key genes in the soluble sugar metabolism were analyzed. ‘Yesterday’ is more cold-hardy and acclimated faster, reaching its maximum cold hardiness in December. R. wichurana is relatively less cold-hardy, only reaching its maximum cold hardiness in January after prolonged exposure to freezing temperatures. Dehydrin transcripts accumulated significantly during November–January in both genotypes. Soluble sugars are highly involved in cold acclimation, with sucrose and oligosaccharides significantly correlated with cold hardiness. Sucrose occupied the highest proportion of total soluble sugars in both genotypes. During November–January, downregulation of RhSUS was found in both genotypes, while upregulation of RhSPS was observed in ‘Yesterday’ and upregulation of RhINV2 was found in R. wichurana. Oligosaccharides accumulated from November to February and decreased to a significantly low level in April. RhRS6 had a significant upregulation in December in R. wichurana. This study provides insight into the cold acclimation mechanism of roses by combining transcription patterns with metabolite quantification.

Vertical farming using light-emitting diode offers potential for the early production phase (few weeks) of young ornamental plants. However, once transferred to the greenhouse, the photosynthetic acclimation of these young plants might depend on this initial light regime. To obtain insight about this acclimatization, Chrysanthemum (sun species) and Spathiphyllum (shade species) were preconditioned in growth chambers for 4 weeks under four light qualities: blue (B), red (R), red/blue (RB, 60% R) and white (W) at 100 mu mol m(-2) s(-1). Monochromatic light (R and B) limited leaf development of both species, which resulted in a lower leaf mass per area when compared to multispectral light (RB for Chrysanthemum, RB and W for Spathiphyllum). R-developed leaves had a lower photosynthetic efficiency in both species. After the light quality pretreatment, plants were transferred to the greenhouse with high-intensity natural light conditions. On the first day of transfer, R and B preconditioned leaves of both species had an inhibited photosynthesis. After 1 week in natural light condition, rapid light curve parameters of Chrysanthemum leaves that developed under B acclimated to sunlight had a similar level than RB-developed leaves unlike R-leaves. Spathiphyllum leaves showed a decrease in maximum electron transport rate and this was most pronounced for the R pretreatment. After 1 month, R-preconditioned Chrysanthemum had the lowest dry mass, while no effects on the dry weight of Spathiphyllum with respect to the pretreatments were observed. Light quality during preconditioning affected the leaf ability to acclimate to natural high light intensities in greenhouse environment.

Male flower shortage is a major reason of insufficient pollination in zucchini (Cucurbita pepo L.), leading to pointy fruits that rot quickly in the post-harvest phase. Harvesting pollen grains early in the season, when male flowers are abundant, followed by storage and subsequent manual pollination, could be a solution, provided that high pollen viability is maintained. Therefore, insight in parameters affecting pollen viability during anthesis and under storage are needed to advice growers on the most successful pollination strategy. Pollen viability was determined using Fluorescein Diacetate (FDA) and digital image analysis performed with Image J. Under practice conditions, viability started to diminish three hours after anthesis. At flower closing, only 60% of pollen grains remained viable and the next morning no more than 10%. In order to assess the impact of storage on pollen viability, pollen grains were subjected to different relative humidities and storage times. Stored at 20°C and 80% RH or lower for four hours, at least 90% of the pollen turned out to be non-viable. At 20°C and 90%, RH pollen viability remained above 50% for eight hours. When female flowers were pollinated with stored pollen (five days or longer), none of the fruits set sufficiently to be marketable, while fresh harvested pollen resulted in 90% marketable fruits. Therefore, pollen should be harvested and applied on the stigma within three hours after anthesis in order to ensure good fruit setting. Pollen could not be stored longer than eight hours without severe loss of viability or pollinating capacity.

The introgression of rol-genes of rhizogenic Agrobacterium into the plant genome induces changes in plant phenotype and physiology. However, only limited experience with this technique is available for woody ornamentals. To induce new variation within the Escallonia genus, several species were co-cultivated with rhizogenic Agrobacterium strains. Co-cultivation of three rhizogenic Agrobacterium strains (Arqua1, LMG 63 and MAFF210266) with four Escallonia species (E. illinita, E. myrtoidea, E. rosea, and E. rubra), resulted in hairy roots production with a varying efficiency. Co-cultivation of E. rubra with MAFF210266, and E. myrtoidea with LMG63 did not yield hairy roots, while co-cultivation of E. rubra leaves with LMG63 was most successful for hairy root production (up to 80.6%). In addition, the efficiency of hairy root induction depended on the explant type (leaves or nodal sections). The presence of inserted rol-genes and aux-genes in hairy roots was molecularly confirmed using qPCR. Few shoots regenerated from hairy roots, but regeneration needs to be optimized for efficient implementation of rol-genes introgression in Escallonia breeding. This research provides a protocol for the production of hairy roots with rol-genes inserted after co-cultivation of several species of Escallonia with rhizogenic Agrobacterium strains.

Light stimulates the production of anthocyanins in leaves. Assimilation light is needed in the production of red colored foliage plants during the dark winter period in order to obtain a commercially attractive product. In this study we tested whether this can be achieved by growing those plants for a limited period at the end of the production cycle in a growth chamber under specific LED (Light Emitting Diodes) light spectra. The experiment was conducted on three species: Hypoestes phyllostachya ‘decor pink’ and ‘decor red’ or polka dot plant, Guzmania lingulata ‘Theresa’ with yellow and red bracts and Cryptanthus carnosus ‘Tricolor’, also known as earth star. Different combinations of blue, red and far red light were applied and control treatments were given with and without HPS (High Pressure Sodium) lamps in the greenhouse. In all three species a minimal amount of blue light is compulsory. When only red LEDs are used in Hypoestes, leaves become curly and plants are not sufficiently compact. Without the addition of blue light the Guzmania bracts turn entirely yellow and Cryptanthus leaves are much more pale. In all cases, the use of LED was an improvement for the anthocyanin content when compared to plants that were grown without supplementary light. We demonstrated that for all three species a limited growing period under specific light spectra at the end of the production cycle can be a good way to produce attractive colored foliage plants when natural light is limited.

Crassulacean acid metabolism (CAM) is a specialized photosynthetic pathway present in a variety of genera including many epiphytic orchids. CAM is under circadian control and can be subdivided into four discrete phases during a diel cycle. Inherent to this specific mode of metabolism, carbohydrate availability is a limiting factor for nocturnal CO2 uptake and biomass production. To evaluate the effects of light quality on the photosynthetic performance and diel changes in carbohydrates during the CAM cycle. Phalaenopsis plants were grown under four different light qualities (red, blue, red + blue and full spectrum white light) at a fluence of 100 µmol m-2 s-1 and a photoperiod of 12 h for 8 weeks. In contrast to monochromatic blue light, plants grown under monochromatic red light showed already a significant decline of the quantum efficiency (ΦPSII) after 5 days and of the maximum quantum yield (Fv/Fm) after 10 days under this treatment. This was also reflected in a compromised chlorophyll and carotenoid content and total diel CO2 uptake under red light in comparison with monochromatic blue and full spectrum white light. In particular, CO2 uptake during nocturnal phase I was affected under red illumination resulting in a reduced amount of vacuolar malate. In addition, red light caused the rate of decarboxylation of malate during the day to be consistently lower and malic acid breakdown persisted until 4 h after dusk. Because the intrinsic activity of PEPC was not affected, the restricted availability of storage carbohydrates such as starch was likely to cause these adverse effects under red light. Garden roses have a high ornamental and economic value. They are usually produced in one climatic zone, then sold to another climatic zone. Cold stress can severely inhibit their regrowth and ornamental quality. Knowledge of cold hardiness of specific genotypes is therefore necessary before commercialization to colder regions. 17 roses with diverse genetic backgrounds and belonging to different USDA (United States Department of Agriculture) plant hardiness zones were chosen for a three-year cold hardiness screening based on the index of injury method. Stems were sampled on 3rd February 2015, 20th January 2016 and 23rd January 2017 after a freezing period of six successive days. Cold hardiness levels were determined by controlled freezing to −20 °C. Genotypes with the highest and lowest levels of cold hardiness were confirmed during these three years. Soluble carbohydrates were analyzed. The results showed that both sucrose and oligosaccharides contributed to cold hardiness in the studied roses. When genotypes with known cold hardiness are included in screening programs, this approach can lead to rapid screening of new rose genotypes.

Rocket cultivation is increasing to supply the expanding ready-to-eat market because of its unique taste, but crops are often over fertilized to avoid nitrogen deficiencies. This leads to nitrate accumulation in leaves, and the products of their degradation (nitrites and nitrosamines) have been related to several health problems. Nitrate concentrations in rocket and other leafy vegetables are subject to limits by the EU legislation, yet rocket holds a great nutritional value. Degradation products of glucosinolates (isothiocyanates) have been consistently linked with benefits to human health. We investigated the influence of nitrogen application (1 and 8 mM), species [Eruca sativa (L.) Cav. and Diplotaxis tenuifolia (L.) DC.] and light spectrum (full spectrum, red, blue and red + blue) on the nitrate concentration, nitrate reductase activity and glucosinolate content of rocket grown in a soil-less system. Red light decreased the nitrate concentration with respect to the blue spectrum (4,270 vs. 7,100 mg.kg(-1) of fresh weight, respectively), but such reduction was influenced by the species and the nitrogen level (significantly higher in D. tenuifolia and with the higher concentration of N). The nitrate reductase activity increased under red light in D. tenuifolia, with the lower N concentration. Rocket is known to contain several health-promoting compounds mainly antioxidants and glucosinolates, as secondary metabolites that act as part of plant defense mechanisms. The total content of glucosinolates was mainly affected by the species (D. tenuifolia showed the highest concentrations). Our results will help growers to tailor light spectra with the aim of reducing nitrate concentration and to remain within EU legislative limits, without any detrimental influence on other qualitative parameters in rocket.

In this study, the effects of both leaf age and light quality on leaf resistance against Botrytis cinerea were investigated. Strawberry plants were grown in a growth chamber equipped with white, blue, red or red + blue light-emitting diodes (LEDs) at a photosynthetic photon flux (PPF) of 100 μmol m−2 s−1. The effect of leaf age and light quality on leaf morphology, resistance to B. cinerea, hydrogen peroxide levels, metabolites and pigments was studied. Leaf number increased under red light, while leaf petiole length was reduced by red + blue light. Leaf resistance to B. cinerea dramatically increased with leaf age from 1- to 4-week-old leaves but decreased again in 5-week-old leaves. Red light significantly improved leaf resistance, while white and blue-light treated leaves were the most susceptible to B. cinerea at all leaf ages. Hydrogen peroxide levels positively correlated with disease severity and were influenced by both leaf age and light quality. They were lowest in 4-week-old leaves and in red light-grown leaves, irrespective of leaf age. Chlorophyll and carotenoids levels negatively correlated with disease severity and increased with leaf age but were lowest in blue light-grown leaves. Total phenolics and flavonoid levels were very high in the very susceptible 1-week-old leaves and considerably lower in the older leaves. Red light stimulated total phenolics in 1–4-week-old leaves. Proline levels were strongly stimulated by blue light, especially in 1 and 5-week-old leaves. Overall, low hydrogen peroxide levels and high chlorophyll and carotenoids levels appear to be the best indicators for leaf resistance to B. cinerea in strawberry leaves. Moreover, leaf age should be taken into account when assessing the effect of light quality on disease resistance.

Botrytis cinerea, a fungal pathogen that causes gray mold, displays a high degree of phenotypic diversity. Light emitting diodes (LEDs) with specific light spectrum are increasingly used as lighting resource for plant greenhouse production. The chosen light spectrum can also have an effect on the pathogens in this production system. In this study, we investigated the phenological diversity in 15 B. cinerea isolates upon different light treatments. Daylight, darkness, and LED lights with different wavelengths (white, blue, red, blue+red) were chosen as treatments. The 15 Botrytis isolates differed in their mycelial growth rate, conidia production, and sclerotia formation. Light quality had a limited effect on growth rate. All isolates sporulated under daylight treatment, red light resulted in lower sporulation, while white, blue, and blue+red light inhibited sclerotia formation in all isolates, and sporulation in most, but not all isolates. Pathogenicity of the Botrytis isolates was studied on 2-week-old strawberry (Fragaria × ananassa ‘Elsanta’) leaves grown under white, blue, and red LED lights. The isolates differed in virulence on strawberry leaves, and this was positively correlated to oxalic acid production by B. cinerea in vitro. Red LED light improved leaf basal resistance to all the tested Botrytis isolates. Blue light pretreatment resulted in decreased leaf resistance to some isolates. Furthermore, we used image analysis to quantify the virulence of the different Botrytis isolates based on changes in photosynthetic performance of the strawberry leaves: chlorophyll fluorescence (Fv/Fm), chlorophyll index (ChlIdx) and anthocyanin content (modified anthocyanin reflection index, mAriIdx). Fv/Fm showed a strong negative correlation with disease severity and can be an indicator for the early detection of gray mold on strawberry leaves.

Light plays an essential role in the outcome of plant-pathogen interactions. In this study, the effect of pre- and post-inoculation light quality on Botrytis cinerea disease development in strawberry leaves was investigated. Circadian variations in leaf susceptibility to Botrytis cinerea were tested by inoculating at 5 h, 12 h, or 16 h after the start of photoperiod. The effects of 4 h and/or 12 h of red or blue light combined with white light in a 16 h photoperiod on leaf susceptibility and metabolites were studied. Light presence significantly decreased leaf susceptibility to Botrytis irrespective of light quality. Leaf susceptibility to B. cinerea showed significant circadian variations. Susceptibility was highest in leaves inoculated 12 h after the start of photoperiod followed by incubation in the dark. When disease development took place under the light regime, the shorter the light period after inoculation the stronger the disease symptoms. Twelve hours of red light radiation in a 16 h photoperiod increased, while 12 h blue light radiation decreased leaf resistance, resulting in alterations in the circadian variation of leaf susceptibility. These alterations correlated with changes in leaf metabolites, such as hydrogen peroxide, total phenolics and hexoses. Overall, we conclude that twelve hours of red light followed by white light in a 16 h photoperiod efficiently increased leaf resistance to B. cinerea.

The effect of light quality at two light intensities on leaf anatomy, photosynthetic efficiency and pigmentation were investigated in Chrysanthemum. Four light qualities were applied at two light intensities of 40 μmol m−2 s−1 and 100 μmol m−2 s−1 and with a photoperiod of 14 h using light-emitting diodes, which were 100% red (R), 100% blue (B), 75% red with 25% blue (RB) and white (W), respectively. Leaf anatomy responses to a lower light intensity were observed, we saw a decrease in leaf thickness for blue, red + blue and multispectral white light but not for the red wavelength. At higher light intensity we also observed a favorable effect of blue light on the anatomical development of the leaves. Both light intensity and quality affected the stomatal development. Low light decreased the stomatal index and stomatal density but increased in the stomatal area for red + blue and multispectral white light. Light intensity affected the pigment accumulation but no quality effects were present. For the lowest light level an enhanced pigment concentration was observed in Chrysanthemum this as well for Chl a, Chl b and total carotenoids. Light quality influenced the photosynthetic efficiency as observed by chlorophyll fluorescence. Monochromatic red resulted in an inhibition of Photosystem II, this at both light intensities, resulting in a decline in maximum quantum yield (Fv/Fm) and quantum efficiency (ΦPSII). Light intensity significantly influenced biomass accumulation, higher light intensity increased plant dry weight. At a light intensity of 100 μmol m−2 s−1, blue light positively influenced the biomass compared to monochromatic red.

Salinity tolerance levels and physiological changes were evaluated in four eggplant cultivars ( Solanum melongena L.). Seeds and seedlings were exposed to increasing salinity levels. In vitro germination kinetics and young seedling growth divided the cultivars in a sensitive and a tolerant group. ‘Adriatica’ and ‘Black Beauty’ supported only moderate salt stress up to 40 mM NaCl while the tolerance level of ‘Bonica’ and ‘Galine’ was up to 80 mM NaCl. Effects of salinity levels (0, 20, 40, 80 and 160 mM of NaCl) were also tested in a greenhouse experiment. Increasing NaCl concentration increased strongly the levels of proline, malondialdehyde and soluble carbohydrates in the leaves of ‘Adriatica’ and ‘Black Beauty’. In contrast, ‘Bonica’ and ‘Galine’ showed a decrease in soluble carbohydrates and a significant increase in starch under saline stress. The midday leaf water potential (ψ l ) and leaf osmotic potential (ψ π ) were significantly affected in sensitive cultivars and remained quite stable in tolerant cultivars under salt stress. Leaf Na + and Cl − content was higher in sensitive than in tolerant cultivars. The leaf K + , Ca 2+ and Mg 2+ contents were reduced under salt stress in sensitive cultivars. Under increasing salinity ‘Bonica’ and ‘Galine’ combine low leaf Na + accumulation with a high plant tolerance index (PTI) thus maintaining a normal level of growth, while ‘Adriatica’ and ‘Black Beauty’ accumulate significantly higher concentrations of leaf Na + and fail to maintain a normal level of growth from 80 mM NaCl on.

There is a need for plant growing media that can support a beneficial microbial root environment to ensure that optimal plant growth properties can be achieved. We investigated the effect of five rhizosphere bacterial community inocula (BCI S1&ndash, 5) that were collected at three open field organic farms and two soilless farms on the performance of lettuce (Lactuca sativa L.). The lettuce plants were grown in ten different plant growing media (M1&ndash, 10) composed of 60% v/v peat (black peat or white peat), 20% v/v other organics (coir pith or wood fiber), 10% v/v composted materials (composted bark or green waste compost) and 10% v/v inorganic materials (perlite or sand), and one commercial plant growing medium inside a plant factory with artificial lighting. Fractional factorial design of experiments analysis revealed that the bacterial community inoculum, plant growing medium composition, and their interaction determine plant performance. The impact of bacterial amendments on the plant phenotype relied on the bacterial source. For example, S3 treatment significantly increased lettuce shoot fresh weight (+57%), lettuce head area (+29%), root fresh weight (+53%), and NO3-content (+53%), while S1 treatment significantly increased lettuce shoot dry weight (+15%), total phenolic content (+65%), and decreased NO3-content (&minus, 67%). However, the effectiveness of S3 and S1 treatment depended on plant growing medium composition. Principal component analysis revealed that shoot fresh weight, lettuce head area, root fresh weight, and shoot dry weight were the dominant parameters contributing to the variation in the interactions. The dominant treatments were S3-M8, S1-M7, S2-M4, the commercial plant growing medium, S1-M2, and S3-M10. Proper selection of plant growing medium composition is critical for the efficacy of bacterial amendments and achieving optimal plant performance inside a plant factory with artificial lighting.

Drought is a major threat in agriculture and horticulture, including commercial strawberry production. Here, we compare hormonal regulation of a first-line drought stress response, namely stomatal closure, in two Fragaria x ananassa cultivars, known to differ in their drought stress phenotype. We show that the observed difference in xylem abscisic acid accumulation cannot explain the different stomatal responses under osmotic stress. Foliar abscisic acid accumulation cannot fully account for the stomatal behavior in one of both cultivars either. An indirect effect of abscisic acid on stomatal conductance via an impact on leaf hydraulic conductance, possibly mediated via aquaporins, as is recently proposed in literature, was not observed here. Next, we show that these two cultivars respond differently to jasmonic acid and one of its precursors. This difference in sensitivity of the jasmonates pathway between both cultivars may partly explain the different stomatal response. This study contributes to the understanding of the regulation of an important drought stress response in an economically important crop prone to water deficit stress.

In order to improve the understanding of plant water relations under drought stress, the water use behavior of two Fragaria x ananassa Duch. cultivars, contrasting in their drought stress phenotype, is identified. Under drought, stomatal closure is gradual in Figaro. Based on this, we associate Figaro with conservative water use behavior. Contrarily, drought stress causes a sudden and steep decrease in stomatal conductance in Flair, leading to the identification of Flair as a prodigal water use behavior cultivar. Responses to progressive drought on the one hand and an osmotic shock on the other hand are compared between these two cultivars. Tonoplast intrinsic protein mRNA levels are shown to be upregulated under progressive drought in the roots of Figaro only. Otherwise, aquaporin expression upon drought or osmotic stress is similar between both cultivars, i.e. plasma membrane intrinsic proteins are downregulated under progressive drought in leaves and under short term osmotic shock in roots. In response to osmotic shock, root hydraulic conductivity did not change significantly and stomatal closure is equal in both cultivars. De novo abscisic acid biosynthesis is upregulated in the roots of both cultivars under progressive drought.

Pistachio (P. vera L, Anacardiaceae) is cultivated in regions where soil water deficits and salinity conditions are higher than usual. Despite adult pistachio trees having been documented as being drought tolerant; there is only a limited understanding of the physiological mechanisms pistachio cultivars use to survive drought. We therefore, carried out a greenhouse experiment to evaluate the effects of three osmotic drought stress treatments including; control coridithins (-0.1 MPa), moderate (-0.75 MPa) and severe drought (-1.5 MPa) stress, using PEG 6000 for 14days with a subsequent two weeks recovery period. Carbohydrate contents, relative water content, water use efficiency, stomatal characteristics, and nitrogen and carbon isotope composition were evaluated in three Iranian pistachio cultivars, i.e. Akbari, Kaleghochi and Ohadi. Results revealed that the drought stress treatments induced osmotic adjustment by carbohydrate accumulation. Both drought stress treatments increased soluble carbohydrate and starch contents of the leaves. Relative water content was only affected by drought stress in Kaleghochi. Stomata] density and morphology varied with pistachio cultivars but was hardly affected by the stress treatments. Drought stress significantly increased the overall mean of water use efficiency (intrinsic and instantaneous WUE). There were no significant differences between the leaf carbon isotope compositions of all pistachio cultivars under stress. This indicates that this relation may not be used to determine pistachio cultivars with appropriate WUE via leaf carbon isotope composition within the time frame of the experiment. SUMMARYThe term ‘stay-green’ (SG) is used in studies with varieties showing delayed senescence in the field. However, delayed appearance of visual symptoms of leaf senescence does not guarantee a longer duration of photosynthesis. The question arises whether the variation in photosynthetic capacity between silage maize varieties currently on the Belgian market is large enough to define different SG types. Furthermore, physiological traits were investigated as proxies for photosynthesis, including chlorophyll concentration, nitrogen (N) concentration, SPAD readings and greenness score. Finally, the functionality of the SG trait was determined by studying dry matter (DM) and N partitioning in the plants. To address these questions, eight silage maize varieties were monitored at two sites in Belgium over 2 years (2013–2014). Two plant types were found: hereafter called ‘normal’ and ‘SG’. The SG varieties had higher values for photosynthetic capacity and they coincided with higher values for the proxies. Because a higher photosynthetic capacity did not provoke higher assimilate accumulation in the leaves, the SG trait was characterized as a cosmetic one. The SG trait influenced N dynamics in the plant: lower N translocation from the leaves to the ear resulted in lower ear N concentration and lower ear DM yield. No differences in whole-crop N concentration and whole-crop DM yield were found. As the SG trait mainly provokes shifts in partition of DM and N between vegetative and generative tissues, the energy source also shifts from starch (provided by the ear) to cell wall material (provided by the stover).

Vegetables and fruits are an important part of a healthy food diet, however, the eco-sustainability of the production of these can still be significantly improved. European farmers and consumers spend an estimated Euro15.5 billion per year on inorganic fertilizers and the production of N-fertilizers results in a high carbon footprint. We investigated if fertilizer type and medium constituents determine microbial nitrogen conversions in organic growing media and can be used as a next step towards a more sustainable horticulture. We demonstrated that growing media constituents showed differences in urea hydrolysis, ammonia and nitrite oxidation and in carbon dioxide respiration rate. Interestingly, mixing of the growing media constituents resulted in a stimulation of the function of the microorganisms. The use of organic fertilizer resulted in an increase in amoA gene copy number by factor 100 compared to inorganic fertilizers. Our results support our hypothesis that the activity of the functional microbial community with respect to nitrogen turnover in an organic growing medium can be improved by selecting and mixing the appropriate growing media components with each other. These findings contribute to the understanding of the functional microbial community in growing media and its potential role towards a more responsible horticulture.

Aim of this study was to investigate the effect of water shortage in wild jujube plants, Ziziphus lotus (L.) Lam, and how it is related to its ecological success. All leaf anatomical traits were significantly affected following soil drying. Stressed plants displayed more negative stem water potential (psi(w)) and osmotic potential values of ca. -3.5 and -4.5 MPa, respectively, after 30 d. The relative water content declined although it still maintained high values (>= 75%). The net photosynthetic rate, stomatal conductance, and transpiration rate were significantly inhibited as psi(w) decreased. However, the intrinsic water-use efficiency increased as water deficit was intensified; the difference became significant only after 30 d. As a consequence, the effective quantum yield of PSII photochemistry and the photochemical quenching coefficient significantly decreased although the maximal quantum yield of PSII photochemistry ratio remained statisticaly unchanged. Plants could maintain their water status sufficiently by increasing proline 1.7-fold and sucrose 1.8-fold in their leaves, respectively. The largest accumulation of both solutes may avoid photodamages at cellular level and play a critical role in maintaining osmotic adjustment.

Synthetic fertilizer production is associated with a high environmental footprint, as compounds typically dissolve rapidly leaching emissions to the atmosphere or surface waters. We tested two recovered nutrients with slower release patterns, as promising alternatives for synthetic fertilizers: struvite and a commercially available organic fertilizer. Using these fertilizers as nitrogen source, we conducted a rhizotron experiment to test their effect on plant performance and nutrient recovery in juvenile tomato plants. Plant performance was significantly improved when organic fertilizer was provided, promoting higher shoot biomass. Since the microbial community influences plant nitrogen availability, we characterized the root-associated microbial community structure and functionality. Analyses revealed distinct root microbial community structure when different fertilizers were supplied. However, plant presence significantly increased the similarity of the microbial community over time, regardless of fertilization. Additionally, the presence of the plant significantly reduced the potential ammonia oxidation rates, implying a possible role of the rhizosheath microbiome or nitrification inhibition by the plant. Our results indicate that nitrifying community members are impacted by the type of fertilizer used, while tomato plants influenced the potential ammonia-oxidizing activity of nitrogen-related rhizospheric microbial communities. These novel insights on interactions between recovered fertilizers, plant and associated microbes can contribute to develop sustainable crop production systems. To obtain crossing products, an efficient pollination and subsequent fertilization is essential. This efficient pollination is achieved by pollen germination and tube growth. Here, these pollen characteristics of 2 genetically differentiating cultivars of Anemone coronaria L. were investigated in vitro. For the essential components boron, calcium and an osmoticant, only calcium showed to be crucial for pollen germination. Boron concentrations influenced the pollen tube length with a concentration of 100 mg l−1 H3BO3 resulting in the longest pollen tubes. For the osmoticant sucrose, a concentration of 100 g l−1 caused a significant positive effect on both pollen germination and pollen tube length for the 2 cultivars. The cultivars reacted similarly with respect to the investigated compounds. Next to this, the pollen development was correlated with 8 different flower stages in A. coronaria. As a final point, the germination of an optimized pollen germination medium was compared with in vivo pollen germination in cross-pollinations within the same cultivar (identified by aniline blue staining). For ‘Mistral Wine’, pollen germination percentage was lower in vitro than in vivo, while ‘Wicabri Blue’ pollen showed no significant difference in germination rates in vivo and in vitro. To achieve fertilization a following requisite is that the stigma is receptive. To study this, the most receptive female flower stage of the 8 different flower stages for A. coronaria was characterized by aniline blue staining. A. coronaria clearly showed protogyny.
