Significance: Hypoxic-ischemic events due to intrapartum complications represent the second cause of neonatal mortality and initiate an acute brain disorder known as hypoxic-ischemic encephalopathy (HIE). In HIE, the brain undergoes primary and secondary energy failure phases separated by a latent phase in which partial neuronal recovery is observed. A hypoxic-ischemic event leads to oxygen restriction causing ATP depletion, neuronal oxidative stress, and cell death. Mitochondrial dysfunction and enhanced oxidant formation in brain cells are characteristic phenomena associated with energy failure. Recent Advances: Mitochondrial sources of oxidants in neurons include complex I of the mitochondrial respiratory chain, as a key contributor to O2•- production via succinate by a reverse electron transport mechanism. The reaction of O2•- with nitric oxide (•NO) yields peroxynitrite, a mitochondrial and cellular toxin. Quantitation of the redox state of cytochrome c oxidase, through broadband near-infrared spectroscopy, represents a promising monitoring approach to evaluate mitochondrial dysfunction in vivo in humans, in conjunction with the determination of cerebral oxygenation and their correlation with the severity of brain injury. Critical Issues: The energetic failure being a key phenomenon in HIE connected with the severity of the encephalopathy, measurement of mitochondrial dysfunction in vivo provides an approach to assess evolution, prognosis, and adequate therapies. Restoration of mitochondrial redox homeostasis constitutes a key therapeutic goal. Future Directions: While hypothermia is the only currently accepted therapy in clinical management to preserve mitochondrial function, other mitochondria-targeted and/or redox-based treatments are likely to synergize to ensure further efficacy.

Uno de los objetivos principales de la ecología fluvial es sintetizar la información ecológica para poder predecir cómo los taxa responden a los cambios ambientales antropogénicos o naturales. Estos ecosistemas fluviales se encuentran dentro de los que contienen la mayor biodiversidad planetaria, pero también forman parte de los ambientes con mayores riesgos y peligros de extinción debido al aumento en su uso para el desarrollo socioeconómico moderno. La provincia de Buenos Aires presenta la mayor concentración demográfica e industrial del país que junto con la producción agroganadera y el uso más intenso de agroquímicos, ha puesto en jaque a numerosos arroyos pampeanos. Tanto los cambios espaciales como temporales de los hábitats ocurren a diferentes escalas y otorgan a los organismos un mosaico cambiante de condiciones bióticas y abióticas, las que juegan un papel importante en su organización. Las interacciones de los organismos con las propiedades fluctuantes del entorno se llevan a cabo a través de sus características morfológicas, fisiológicas, de comportamiento o uso del hábitat, las que formarán una estrategia determinada de la cual dependerá la prevalencia o no de los mismos en ese hábitat. Debido a su diversidad de formas y hábitos, los ensambles de macroinvertebrados bentónicos son buenos candidatos para evaluar la calidad de los ecosistemas acuáticos ya que tienen el potencial de responder de muchas maneras al estrés ambiental. Las características mencionadas anteriormente forman parte de los rasgos funcionales, es decir, atributos del organismo referente a su biología, fisiología y ecología (rasgos biológicos y ecológicos), que repercuten en el crecimiento, la reproducción y la supervivencia a nivel individual. Un rasgo puede mostrar diferentes expresiones o atributos, y estos pueden ser expresados en categorías o rangos. La teoría del River Habitat Templet (RHT) predice que las fuentes de hábitats de los arroyos determinan la composición y diversidad de las comunidades bióticas en una red de variabilidad espaciotemporal. Entonces, desde ese concepto se podría potencialmente hacer predicciones cuantitativas de la representación de los grupos funcionales de organismos. Por grupos funcionales, se entiende a una combinación específica de atributos que define un tipo funcional (p.ej., grupos de taxa que comparten adaptaciones con una función específica). La identificación y evaluación de estos tipos funcionales es importante para describir los mecanismos bajo los cuales la comunidad responde a diferentes factores ambientales. El objetivo general del presente trabajo de tesis es proponer, por primera vez para la Ecorregión Pampa, un modelo gráfico que sintetice la variabilidad encontrada de acuerdo con la teoría del RHT para rasgos biológicos y ecológicos de macroinvertebrados, evaluando su uso como una herramienta potencial de biomonitoreo en los ambientes fluviales pampeanos. De forma complementaria se aplicaron índices biológicos tradicionales y de diversidad funcional. Para su cumplimiento se seleccionaron los usos del suelo periurbano, industrial, agroganadero y de reserva pertenecientes a dos complejos de la Ecorregión Pampa. En el complejo de la pampa ondulada se muestrearon los arroyos El Destino, Juan Blanco, El Pescado, Cajaravilla, Pérez, Del Gato y Chubichaminí; en el complejo de las sierras bonaerenses los arroyos Tandileofú, San Gabriel, La Cascada y su arroyo (del Sistema Tandilia), Atravesado, Sauce Chico, Ventana, Napostá Grande y río Sauce Grande (del Sistema Ventania). Se tomaron muestras del hábitat templet físico- químico y biológico (es decir, macroinvertebrados) de estos sistemas fluviales entre los años 2015 y 2017 durante dos estaciones climáticas contrastantes. Para estructurar la información biológica se documentaron las características relacionadas con el ciclo de vida (p.ej. "tamaño máximo"), la resistencia o resiliencia (p.ej. “formas de resistencia”), rasgos de sus historias de vida (p.ej. "locomoción"), rasgos fisiológicos generales (p.ej. "respiración") y aspectos del comportamiento de la reproducción o nutrición de los taxa (p.ej. "reproducción", “alimento”, 'hábitos alimentarios'). Para la información ecológica, se registraron las variables que reflejaban diferentes características espaciales del hábitat utilizado por los organismos (p.ej. “altitud absoluta”), y se codificaron las preferencias por los microhábitats (p.ej. “tipo de sustrato”, “velocidad de la corriente”, “temperatura”), y también el "estado trófico" del agua en el que cada taxón vive. Tanto los rasgos biológicos como ecológicos fueron sometidos a un proceso de “fuzzyficación” que consiste en colocar valores entre 0 y 3 para "traducir" el conocimiento disponible sobre los rasgos de los organismos a valores numéricos. Se indica con valor 0: cuando no hay afinidad de un taxón a una categoría dada o no hay información documentada, 1: ante una afinidad débil a esa categoría de rasgo o se menciona en la literatura, 2: cuando se distingue una afinidad sustancial a esa categoría de rasgos y 3: en los casos que se advierte una afinidad alta a esa categoría de rasgos. Se coloca “0” ante la falta de información de un determinado taxón para asegurar que los análisis multivariados promediarán con los demás taxa ese perfil para la variable correspondiente. Se realizó un Análisis de Correspondencia Difuso (FCA) para analizar de manera preliminar la tabla de rasgos biológicos y la de ecológicos de los taxa. Se utilizó el análisis RLQ, una técnica de ordenación, que vincula tres tablas: la ambiental (R) con la tabla de rasgos (Q) a través de la tabla de abundancia (L) para investigar la relación entre las características del hábitat y los rasgos. El método de la cuarta esquina fue utilizado para evaluar la significancia estadística de la asociación entre las categorías de rasgos individuales y los gradientes ambientales sobre las bases de las puntuaciones de RLQ. Luego se combinaron los métodos para probar la hipótesis nula donde al menos una tabla (R o Q) no esté vinculada a L contra la hipótesis alternativa de que tanto los rasgos como el entorno, influyan en la distribución de especies (es decir, los enlaces LQ y RL sean significativos). La posición de los 19 grupos taxonómicos (generalmente órdenes) en los vectores del análisis FCA de los rasgos biológicos indicó la separación en seis grupos según las diferencias o similitudes en su biología indicando una relativa homogeneidad en los rasgos biológicos para un determinado grupo. En tanto, el análisis de rasgos ecológicos demostró la separación según su “utilización” del hábitat a diferentes escalas espaciales donde las variables se encuentran formando gradientes entre taxa reófilos y taxa que tienen mayor afinidad por los hábitats potámicos. En los análisis de RLQ para rasgos biológicos y ecológicos se obtuvo un eje ambiental principal que explicó la distribución de las especies y sus características distintivas. Este eje incluyó variables de presión antropogénica (p.ej.: fosfatos y amonio relacionados al uso agroganadero y urbano) y variabilidad natural (p.ej.: altitud, temperatura y materia orgánica, relacionados con el clima y la geología) que están fuertemente interrelacionadas en el área de estudio, con un claro componente espacial. Los rasgos pueden mejorar la comprensión mecánica de las relaciones causa-efecto mediante la integración de la teoría ecológica (en este caso, el concepto RHT) en el biomonitoreo, por lo tanto, indican los factores estresantes responsables del deterioro biológico. En esta tesis se halló que diferentes rasgos biológicos y ecológicos pueden indicar diferentes intensidades de impacto humano a través de un claro cambio en las combinaciones de rasgos. Los rasgos biológicos asociados a los usos periurbanos en la pampa ondulada fueron: formas de resistencia como huevos-gémulas o estatoblastos, muy tolerantes a la contaminación, tamaño mayor a 20mm; organismos con modalidades de rasgos ecológicos que incluían tolerancias a: DBO5 superiores a 10mg/l, oxígeno disuelto inferior a 4 mg/l y pH entre 6.5 y 7. En los usos agroganaderos del complejo mencionado: tamaños máximos entre 10-15 mm, excavadores epibentónicos, muy tolerantes a la contaminación y ovovivíparos; en cuanto a los ecológicos, los organismos para sobrevivir en estos ambientes deben tener las mayores tolerancias a los sólidos en suspensión, a los ambientes eutrofizados y asociados a la vegetación flotante. Los potenciales rasgos funcionales que caracterizan los sitios de las sierras bonaerenses donde se practica agricultura y ganadería son: alimentación de detritos gruesos y respiración tegumentaria junto con organismos que toleran aguas más frías y de velocidades moderadas. En estos ambientes los efectos combinados de la geología, la altitud y el clima complejizaron la selección de rasgos correspondientes a los impactos ambientales antrópicos de las serranías. Dentro de este marco teórico, la variación climática natural se propuso como un eje de perturbación (impulsado por los procesos hidrológicos intensos típicos de los sistemas lóticos serranos), mientras que la presión antropogénica (principalmente agricultura intensiva) y los sedimentos finos, una consecuencia natural de la geología potenciada por el uso del suelo, fue propuesto como un eje de adversidad. En base a las variaciones de los rasgos bio-ecológicos a lo largo de los ejes de este templet, los organismos se dispusieron en función del concepto de selección r, K y A. Particular situations increase susceptibility to infection in neonates. The enhanced susceptibility to certain infections increases the risk of developing particular diseases that can progress to become morbidly severe. For example, during the current pandemic caused by the SARS-CoV-2 virus, epidemiological studies have established that pregnant women with COVID-19 disease are more likely to be hospitalized. However, the risk for intensive care unit admission and mechanical ventilation is not increased compared with nonpregnant women. Although much remains unknown with this particular infection, the elevated risk of progression during pregnancy towards more severe manifestations of COVID-19 disease is not associated with an increased risk of death. In addition, the epidemiological data available in neonates suggest that their risk of acquiring COVID-19 is low compared with infants (

3-Hydroxyglutaric acid (3HGA) accumulates in the inherited neurometabolic disorder known as glutaryl-CoA dehydrogenase deficiency. The disease is clinically characterized by severe neurological symptoms, frontotemporal atrophy and striatum degeneration. Because of the pathophysiology of the brain damage in glutaryl-CoA dehydrogenase deficiency is not completed clear, we investigated the in vitro effect of 3HGA (0.01-5.0mM) on critical enzyme activities of energy metabolism, including the respiratory chain complexes I-V, creatine kinase isoforms and Na(+),K(+)-ATPase in cerebral cortex and striatum from 30-day-old rats. Complex II activity was also studied in rat C6-glioma cells exposed to 3HGA. The effect of 3HGA was further investigated on the rate of oxygen consumption in mitochondria from rat cerebrum. We observed that 1.0mM 3HGA significantly inhibited complex II in cerebral cortex and C6 cells but not the other activities of the respiratory chain complexes. Creatine kinase isoforms and Na(+),K(+)-ATPase were also not affected by the acid. Furthermore, no inhibition of complex II activity occurred when mitochondrial preparations from cerebral cortex or striatum homogenates were used. In addition, 3HGA significantly lowered the respiratory control ratio in the presence of glutamate/malate and succinate under stressful conditions or when mitochondria were permeabilized with digitonin. Since 3HGA stimulated oxygen consumption in state IV and compromised ATP formation, it can be presumed that this organic acid might act as an endogenous uncoupler of mitochondria respiration. Finally, we observed that 3HGA changed C6 cell morphology from a round flat to a spindle-differentiated shape, but did not alter cell viability neither induced apoptosis. The data provide evidence that 3HGA provokes a moderate impairment of brain energy metabolism and do not support the view that 3HGA-induced energy failure would solely explain the characteristic brain degeneration observed in glutaryl-CoA dehydrogenase deficiency patients.

The matrix protein gene was cloned and sequenced for several recent isolates of avian paramyxovirus type 1 (APMV-1). Specifically, isolates from pigeons and doves, members of the Columbidae family were examined. APMV-1 is the causative agent of Newcastle disease and the virus is associated with disease among a diverse number of avian species. Newcastle disease virus (NDV) isolates from pigeons have also been classified as pigeon paramyxovirus type 1 (PPMV-1). Matrix protein gene sequences for PPMV-1 isolates clustered together as a group relative to isolates from other species phylogenetically. However, there were also isolates from pigeons or doves that grouped with APMV-1 isolates from other species. This indicates that PPMV-1 may be circulating among Columbidae members as a distinct lineage, but that these avian species may also harbor other NDV strains as well. Of particular interest was a dove isolate from Europe that had an aberrant fusion protein cleavage site and was an outlying member phylogenetically between the two major groups of APMV-1 isolates.

The cell response of human HepG2 cells exposed to hypothermia with rewarming was analyzed. Ultrastructural findings in hypothermic stressed cells showed swollen mitochondria, dispersed chromatin, vacuoles and ring-shape nucleolar reorganization. These changes were coupled with significative differences in the induction of Hsp60, inducible Hsp70 and monomeric Hsfl in all treated samples, but not in Hsc 70 expression. Cellular response to hypothermia could be associated with the synergistic induction of Hsp expression.

The aim of the study was to determine the colonization of mutans streptococci (MS) in dental biofilm and saliva of children from a high-risk population with low socioeconomic status from San José, Costa Rica.A total of 68 healthy babies from 8 to 20 months of age (mean age=13 months), with caries-free status and cariogenic feeding habits (eg, nursing bottle- or breast-feeding on demand) were examined. Children divided in two groups (13 months and 14-20 months). Samples from dental biofilm and saliva from each subject were processed and plated in blood agar (BA) and Mitis-Salivarius-Bacitracin agar (MSB) to determine MS percentage. Statistical analysis included the analysis of variance test, and stratification by age of MS levels.Detectable MS levels in dental biofilm and saliva were found in 75% and 72% of children, respectively. Counts higher than 20000 colony forming units (CFU) per ml were found in 4% of saliva and in 35% of biofilm samples. The two groups showed significant differences in MS levels for dental biofilm (P=.036) and saliva (P=.009). Children older than 17 months presented higher MS levels (P.05). Analysis by MS levels (0.1%; 0.1-1.0%;1.0% of MS) The two groups showed an insignificant association with increasing mean age. MS density was associated with mean number of erupted primary molars.Mutans streptococci colonization of dental biofilm and saliva from high caries risk infants is associated with age and dental development.

This study examined Hsp70 accumulation and the subcellular characteristics of liver and lung when exposed to ethanol (EtOH), with and without l -carnitine protection. Female Sprague-Dawley rats, 150–200 g body weight, were randomized into four groups: Control (CON), Alcohol (ALC), l -carnitine (CAR) and Alcohol- l -carnitine (ALC-CAR). EtOH was administered per os at a dose of 4 g/kg body weight (1 ml) daily for 4 weeks. Before alcohol intake, an oral dose of 500 mg/kg body weight of l -carnitine was also administered to the ALC-CAR group. The liver and lung samples were subjected to Hsp70 Western blot and ultrastructural analysis. The Hsp70 accumulation was higher in the liver than in the lung samples. Hepatic Hsp70 accumulation was similar for all groups in contrast to lung, where the Hsp70 accumulation depends on the group studied. The ultrastructural results showed lung but not liver alterations, evidencing a stressful condition and subsequent cellular injury for lung tissue but not for liver. The ALC-CAR group showed less lung damage than the non-protected group and resembles the general appearance of the CON and CAR groups. EtOH intoxication induced differential cellular response in liver and lung in a dose and tissue dependent manner. l -carnitine seems to reduce lung EtOH-induced subcellular damage. The promotion of heat shock or stress proteins might represent one of the mechanisms involved that need to be further investigated.

Background: The ECLAMC (Estudio Colaborativo Latinoamericano de Malformaciones Congenitas) is an epidemiological surveillance program for congenital defects that operates in Chile since 1969. Aim: To communicate the frequency of disabling congenital defects in Chile in the period 1982-1997. Material and methods: A review of the ECLAMC registry, choosing 12 congenital defects: amelia, limb amputations, limb reductions, arthrogryposis, hip luxation and subluxation, spina bifida, hydrocephaly, microcephaly, cephalocele, talipes equinovarus, Down syndrome and multiple abnormalities. Results: In the study period, 283,403 births occurred and 7,917 newborns were malformed (7,654 born alive and 263 stillbirths). The congenital defects prevalence rates appeared higher in Chile than in other Latin American countries, specially among stillbirths. Among the studied maternity hospitals, the Clinical Hospital of the University of Chile, showed the higher prevalence of congenital defects. Rancagua and the Navy Hospital in Valparaiso have a high frequency of Down syndrome. Global rates in Chile and in the rest of ECLAMC for specific defects, do not have significant differences, except for hip subluxation, that has a lower incidence in Chile. Conclusions: The ECLAMC allows to have a good knowledge of the prevalence of congenital malformations in Latin America (Rev Med Chile 2001; 129: 67-74).
