A dysregulated immune response toward self-antigens characterizes autoimmune and autoinflammatory (AIF) disorders. Autoantibodies or autoreactive T cells contribute to autoimmune diseases, while autoinflammation results from a hyper-functional innate immune system. Aside from their differences, many studies suggest that monocytes and macrophages (Mo/Ma) significantly contribute to the development of both types of disease. Mo/Ma are innate immune cells that promote an immune-modulatory, pro-inflammatory, or repair response depending on the microenvironment. However, understanding the contribution of these cells to different immune disorders has been difficult due to their high functional and phenotypic plasticity. Several factors can influence the function of Mo/Ma under the landscape of autoimmune/autoinflammatory diseases, such as genetic predisposition, epigenetic changes, or infections. For instance, some vaccines and microorganisms can induce epigenetic changes in Mo/Ma, modifying their functional responses. This phenomenon is known as trained immunity. Trained immunity can be mediated by Mo/Ma and NK cells independently of T and B cell function. It is defined as the altered innate immune response to the same or different microorganisms during a second encounter. The improvement in cell function is related to epigenetic and metabolic changes that modify gene expression. Although the benefits of immune training have been highlighted in a vaccination context, the effects of this type of immune response on autoimmunity and chronic inflammation still remain controversial. Induction of trained immunity reprograms cellular metabolism in hematopoietic stem cells (HSCs), transmitting a memory-like phenotype to the cells. Thus, trained Mo/Ma derived from HSCs typically present a metabolic shift toward glycolysis, which leads to the modification of the chromatin architecture. During trained immunity, the epigenetic changes facilitate the specific gene expression after secondary challenge with other stimuli. Consequently, the enhanced pro-inflammatory response could contribute to developing or maintaining autoimmune/autoinflammatory diseases. However, the prediction of the outcome is not simple, and other studies propose that trained immunity can induce a beneficial response both in AIF and autoimmune conditions by inducing anti-inflammatory responses. This article describes the metabolic and epigenetic mechanisms involved in trained immunity that affect Mo/Ma, contraposing the controversial evidence on how it may impact autoimmune/autoinflammation conditions.

3. Glycated Albumin Triggers an Inflammatory Response in the Human Airway Epithelium and Causes an Increase in Ciliary Beat Frequency

The role of inflammation in airway epithelial cells and its regulation are important in several respiratory diseases. When disease is present, the barrier between the pulmonary circulation and the airway epithelium is damaged, allowing serum proteins to enter the airways. We identified that human glycated albumin (GA) is a molecule in human serum that triggers an inflammatory response in human airway epithelial cultures. We observed that single-donor human serum induced IL-8 secretion from primary human airway epithelial cells and from a cystic fibrosis airway cell line (CF1-16) in a dose-dependent manner. IL-8 secretion from airway epithelial cells was time dependent and rapidly increased in the first 4 h of incubation. Stimulation with GA promoted epithelial cells to secrete IL-8, and this increase was blocked by the anti-GA antibody. The IL-8 secretion induced by serum GA was 10–50-fold more potent than TNFα or LPS stimulation. GA also has a functional effect on airway epithelial cells in vitro, increasing ciliary beat frequency. Our results demonstrate that the serum molecule GA is pro-inflammatory and triggers host defense responses including increases in IL-8 secretion and ciliary beat frequency in the human airway epithelium. Although the binding site of GA has not yet been described, it is possible that GA could bind to the receptor for advanced glycated end products (RAGE), known to be expressed in the airway epithelium; however, further experiments are needed to identify the mechanism involved. We highlight a possible role for GA in airway inflammation.

Heme oxygenase (HO) is the primary antioxidant enzyme involved in heme group degradation. A variety of stimuli triggers the expression of the inducible HO-1 isoform, which is modulated by its substrate and cellular stressors. A major anti-inflammatory role has been assigned to the HO-1 activity. Therefore, in recent years HO-1 induction has been employed as an approach to treating several disorders displaying some immune alterations components, such as exacerbated inflammation or self-reactivity. Many natural compounds have shown to be effective inductors of HO-1 without cytotoxic effects; among them, most are chemicals present in plants used as food, flavoring, and medicine. Here we discuss some naturally derived compounds involved in HO-1 induction, their impact in the immune response modulation, and the beneficial effect in diverse autoimmune disorders. We conclude that the use of some compounds from natural sources able to induce HO-1 is an attractive lifestyle toward promoting human health. This review opens a new outlook on the investigation of naturally derived HO-1 inducers, mainly concerning autoimmunity.

The heme oxygenase (HO) system involves three isoforms of this enzyme, HO-1, HO-2, and HO-3. The three of them display the same catalytic activity, oxidating the heme group to produce biliverdin, ferrous iron, and carbon monoxide (CO). HO-1 is the isoform most widely studied in proinflammatory diseases because treatments that overexpress this enzyme promote the generation of anti-inflammatory products. However, neonatal jaundice (hyperbilirubinemia) derived from HO overexpression led to the development of inhibitors, such as those based on metaloproto- and meso-porphyrins inhibitors with competitive activity. Further, non-competitive inhibitors have also been identified, such as synthetic and natural imidazole-dioxolane-based, small synthetic molecules, inhibitors of the enzyme regulation pathway, and genetic engineering using iRNA or CRISPR cas9. Despite most of the applications of the HO inhibitors being related to metabolic diseases, the beneficial effects of these molecules in immune-mediated diseases have also emerged. Different medical implications, including cancer, Alzheimer´s disease, and infections, are discussed in this article and as to how the selective inhibition of HO isoforms may contribute to the treatment of these ailments.

6. In vivo expression of ß-galactosidase by rat oviduct exposed to naked DNA or messenger RNA

Intra-oviductal administration of RNA obtained from oviducts of estradiol-treated rats resulted in accelerated egg transport (Ríos et al., 1997). It is probable that estradiol-induced messenger RNA (mRNA) entered oviductal cells and was translated into the proteins involved in accelerated egg transport. In order to test this interpretation we deposited in vivo 50 µg of pure ß-galactosidase (ß-gal) mRNA, 50 µg of pure DNA from the reporter gene ß-gal under SV40 promoter or the vehicle (control oviducts) into the oviductal lumen of rats. Twenty four hours later the ß-gal activity was assayed in oviductal tissue homogenates using o-nitrophenyl-ß-D-galactopyranoside as a substrate. The administration of ß-gal mRNA and pSVBgal plasmid increased ß-gal activity by 71% and 142%, respectively, over the control oviducts. These results indicate that naked DNA and mRNA coding for ß-gal can enter oviductal cells and be translated into an active enzyme. They are consistent with the interpretation that embryo transport acceleration caused by the injection of estradiol-induced RNA in the oviduct involves translation of the injected mRNA

Estradiol accelerates oviductal embryo transport in the rat through changes of genomic expression in oviductal cells. However, the genes involved are unknown. We used a differential display by reverse transcription-polymerase chain reaction to detect estradiol (E2)-dependent genes in the rat oviduct. Rats on day 2 of pregnancy were untreated or treated with 10 µg of E2 and the oviducts were extracted at 30, 180 and 360 min later and used to isolate RNA. Products of reverse transcriptase-PCR, made with pairs of arbitrary and oligo-deoxythymidine primers, were separated on denaturing polyacrylamide gels and candidate bands were excised and reamplified. Truly positive cDNA fragments determined by a single strand conformation polymorphism assay were cloned and sequenced. A ribonuclease protection assay confirmed that clone 25 is up-regulated by E2 in the oviduct at 30, 180 and 360 min. This clone exhibited no homology with known genes and in situ hybridization showed it is only expressed in the epithelial cells of the isthmic segment. Clone 25 is likely to represent a new gene, which is up-regulated by E2 in the epithelium of the isthmic segment of the rat oviduct. Its time frame of response is compatible with a mediator of the effect of E2 on oviductal embryo transport

O câncer de mama tem maior incidência em mulheres no Brasil. A mastectomia é uma das possibilidades de tratamento, porém com repercussões na saúde física e psíquica da mulher. Foi objetivo desta pesquisa descrever os principais impactos da mastectomia em mulheres acolhidas na Associação de Apoio à Pessoa com Câncer (AAPC) na Bahia. Trata-se de um estudo qualitativo, descritivo, realizado com dez mulheres entre 32 e 63 anos, mastectomizadas e que frequentavam a associação. Foi realizada entrevista semisestruturada individual, e os dados foram analisados por meio da técnica de análise de conteúdo de Bardin. Após análise, foram extraídas três categorias: (1) sentimentos diante do diagnóstico e tratamento da neoplasia; (2) luto simbólico e impactos na autoestima das mulheres mastectomizadas; e (3) estratégias de enfrentamento diante da mastectomia. Os sentimentos identificados foram tristeza, medo, desespero, aceitação, fé e esperança. Houve vivência do luto simbólico e impacto na autoestima devido à mastectomia e perda de cabelo. Já as estratégias de enfrentamento do adoecimento foram o fortalecimento de redes de apoio, a espiritualidade, a significação do processo via trabalho voluntário e o apoio da equipe de saúde. Conclui-se que são muitos os impactos psicológicos do adoecimento e do tratamento, sendo o acompanhamento psicológico um cuidado potencial.

- Abstract
A pandemia gerada com a disseminação do vírus Sars-Cov-2 e o consequente isolamento social reconfiguraram o modo de ensino tradicionalmente presencial para o modelo remoto. O objetivo deste estudo é avaliar as implicações desse novo modelo na formação dos futuros médicos a partir de uma revisão integrativa. Assim, a pesquisa tem caráter qualitativo e foi realizada por meio da busca de artigos indexados nas bases de dados Publisher Medline (PubMed) e Scientific Eletronic Library Online (SciELO), utilizando os seguintes descritores: “COVID-19”; “Pandemia COVID-19”; “Infecção pelo SARS-CoV-2”; “Medicina”; “Medicine”; “Educação à distância”; “Education. Distance”; “Estudantes de Medicina”; “Students, Medical”. Foram encontrados 133 artigos (n=133) que, após a aplicação dos critérios de inclusão e exclusão, foram reduzidos para 80 artigos (n=80) os quais, após análise por leitura integral, foram reduzidos para 54 artigos (n=54). Ainda não ficou claro o impacto que esse novo modelo de ensino causará a longo prazo na formação médica, no entanto, é imprescindível que o ensino remoto e o currículo das escolas médicas sejam aperfeiçoados devido a continuidade da pandemia da COVID-19 ou outros eventos que afetem a interação social e, consequentemente, o ensino médico presencial.

In Brazil, the pandemic impacted the ways of life of families, through housing and neighbourhood designs. This research identified built environment design factors that impacted life during the pandemic. A Systematic Literature Review was carried out to evaluate the impacts of the pandemic on the built environment and identify recommendations and actions to reduce those impacts. As a result, we recommend changes to the built environment and especially housing. Improvements are pointed out to ensure the well-being of dwellers, in present and future scenarios.

This study aims to analyze the epidemiological profiles of Visceral Leishmaniasis (VL) and Visceral Leishmaniasis-HIV (VL-HIV) co-infection in the state of Piauí, from 2007 to 2019. The research method used was the descriptive epidemiological study, of the quantitative type, regarding the epidemiological profile of cases of visceral leishmaniasis and LV-HIV co-infection in Piauí from 2007 to 2019, notified and confirmed in the SINAN (Information System for Notifiable Diseases). Of the 2,797 cases of visceral leishmaniasis notified, the “white/ignored” cases (n=486) marked in subfield 34 of the SINAN LV notification form were discarded, totaling a sample of 2,311 cases, of which 2015 (87.2%) had only LV and 296 (12.8%) LV-HIV co-infection. In Piauí, VL is considered a serious public health problem and it was found that patients with visceral leishmaniasis with and without HIV co-infection were mostly men and from urban areas, aged 1 to 4 years for VL and 20 to 39 years for LV-HIV, with the main method of laboratory confirmation in both groups, with a good percentage of cure (80%), a high lethality (10.13%) and recurrence (10%). Este estudio tiene como objetivo analizar los perfiles epidemiológicos de la coinfección por Leishmaniasis Visceral (LV) y Leishmaniasis Visceral-VIH (LV-VIH) en el estado de Piauí, de 2007 a 2019. El método de investigación utilizado fue el estudio epidemiológico descriptivo, de la tipo cuantitativo, referido al perfil epidemiológico de los casos de leishmaniasis visceral y coinfección VI-VIH en Piauí de 2007 a 2019, notificados y confirmados en el SINAN (Sistema de Información de Enfermedades Notificables). De los 2.797 casos de leishmaniasis visceral notificados, se descartaron los casos “blancos / ignorados” (n = 486) marcados en el subcampo 34 del formulario de notificación SINAN LV, totalizando una muestra de 2.311 casos, de los cuales 2015 (87,2%) Coinfección LV y 296 (12,8%) LV-VIH. En Piauí, la LV se considera un problema de salud pública grave y se encontró que los pacientes con leishmaniasis visceral con y sin coinfección por VIH eran en su mayoría hombres y de áreas urbanas, de 1 a 4 años para LV y de 20 a 39 años para LV- VIH, con el principal método de confirmación de laboratorio en ambos grupos, con un buen porcentaje de curación (80%), alta letalidad (10,13%) y recurrencia (10%). O presente estudo tem como objetivo analisar os perfis epidemiológicos da Leishmaniose Visceral (LV) e da coinfecção Leishmaniose Visceral-HIV (LV-HIV) no estado Piauí, no período de 2007 a 2019. O método de pesquisa utilizado foi o estudo epidemiológico descritivo, do tipo quantitativo, referente ao perfil epidemiológico dos casos de leishmaniose visceral e da coinfecção LV-HIV no Piauí do período de 2007 a 2019, notificados e confirmados no SINAN (Sistema de Informações de Agravos de Notificação). No Piauí, a LV é considerada um grave problema de saúde pública e foi constatado que os pacientes com leishmaniose visceral com e sem coinfecção HIV eram, em sua maioria, homens e procedentes da zona urbana, com idade de 1 a 4 anos para a LV e 20 a 39 anos para LV-HIV, com o principal método de confirmação laboratorial em ambos os grupos, tendo um bom percentual de cura (80%), uma alta letalidade (10,13%) e recidiva (10%).

El presente artículo se centra en la conceptualización de los territorios hidrosociales, en torno a la inserción de un megaproyecto turístico en Yucatán, México. Para su análisis se utilizó una estrategia metodológica cualitativa aplicada a un caso de estudio que abarca tres poblados de la península: Yalcobá, X’tut y Sisbichén, afectados actualmente por la construcción del megaproyecto turístico “Xibalbá”. El objetivo es identificar las racionalidades que conciben, definen y problematizan el territorio hídrico, desde las lógicas del estado-empresario, y revelar su inserción al territorio a través de mecanismos específicos. Concluye que el megaproyecto Xibalbá es un mecanismo de poder, caracterizado por lógicas que conciben el agua como recurso económico desde la industria cultural, que inserta racionalidades políticas a través de particularidades físicas, sociales, simbólicas y culturales del sitio acompañado por los siguientes mecanismos: La legislación, el discurso, las políticas públicas y la tecnología.

Introdução: O estado de alerta provocado pela doença do novo Coronavírus (COVID-19) pode impactar negativamente na saúde dos indivíduos. Os profissionais da saúde estão expostos à contaminação pelo vírus, em especial os Cirurgiões-Dentistas e Médicos, que atuam em ambiente hospitalar, devido a sua forma de atuação frequente próxima ao paciente. Sabe-se que a cefaleia pode estar relacionada aos equipamentos de proteção individuais (EPIS) utilizados no dia a dia desses profissionais. Objetivo: O presente estudo busca avaliar a associação da cefaleia e o uso de equipamentos de proteção individual em Cirurgiões-Dentistas e Médicos no Rio Grande do Norte. Metodologia: Foi realizado um estudo do tipo observacional, analítico e transversal com cirurgiões dentistas e médicos convidados a participar da pesquisa via mensagens eletrônicas através do Google formulários acerca de sinais e sintomas, através do questionário para cefaleia (Questionário de dores de cabeça associadas a equipamentos de proteção individual - Adaptado). Para análise estatística os dados foram reunidos em um banco de dados criado no programa Statistical Package for the Social Science (SPSS) 22.0. Foram realizados testes Qui quadrado de Pearson e Fischer para o cruzamento das variáveis, com um intervalo de confiança de 95%. Resultados: A amostra do estudo foi composta por 181 profissionais, sendo 23,2% médicos (n=42) e 76,8% cirurgiões dentistas (n=139), 34,8% (n=63) do gênero masculino e 65,2% (n=118) do gênero feminino. 84% (n=152) dos profissionais atuavam em ambiente ambulatorial. 43,6% dos profissionais (n=79) relataram uma resposta positiva à cefaleia. Um aumento do uso dos EPIS (87,3% n=158) (p=0,183), uso de máscaras faciais com óculos ou viseiras (96,7% n=175), (p=0,234) ou máscaras N95 ou PFF-2 (88,4% n=160) (p=0,062) não foram relacionados à cefaleia. El Perú, dentro de su territorio presenta más de 12000 lagunas, la gran mayoría ubicada en los Andes, se conoce poco de los macroinvertebrados acuáticos que en ellas habitan pese a que son muy importante en el funcionamiento de dichos ecosistemas. Se estudió los macroinvertebrados acuáticos de la zona litoral de la laguna Condorccocha (con un espejo de agua de 5,6 has), ubicada en el departamento de Ayacucho, a una altitud de 3600 msnm, de abril del 2017 hasta febrero del 2018 con muestreos mensuales, se tomaron muestras de cuatro zonas (distribuidas homogéneamente), utilizando una red tipo D Net con una luz de malla de 0,5 mm, modificada para colectar muestras desde la orilla, mediante arrastres desde el interior de la laguna. Se halló 16 géneros, pertenecientes a 13 familias, 8 órdenes y 5 clases (Insecta, Bivalvia, Gastropoda, Malacostraca y Turbellaria); siendo la Insecta la más diversa con once géneros. Los géneros persistentes durante los meses de muestreo fueron Hyalella, Limnaea, Notonecta, Aeshna, Cricotopus, Pseudochironomus, Ectemnostega, Telebasis y Aeshna; los géneros más abundantes fueron Hyalella (35,4%), Notonecta (16,4%) y Lymnaea (15,3%), mientras que el resto presenta abundancias menores a 10%. Se observa que las abundancias de los géneros hallados son semejantes en las cuatro zonas de muestreo, mientras que existen varias de ellas que muestran diferentes abundancias según los meses (Tropisternus, Ectemnostega, Pseudochironomus, Rheothanytarsus, Hyalella). El análisis de cluster (Paired group, Sorensen), se observa que todas las zonas y los meses son semejantes por encima del 80%. En el análisis de componentes principales, muestra que los géneros Tropisternus, Ectemnostega y Cricotopus se hallan asociadas a la zona IV, mientras que el Ectemnostega y Cricotopus al mes de junio., Resumen fue el mismo presentado en AQUATROP.

A recent expansion of mass-housing programmes has occurred in emerging economies. The analysis of research on programmes raises questions about what type of research is produced and what its impact is on housing. The Brazilian ���My House, My Life��� (Minha Casa, Minha Vida ��� MCMV) programme demonstrates that more of the same type of housing is produced and that the focus of most research repeats the same mistakes. Three million homes have been built and the research community has examined the programme���s social, economic and environmental impacts. A total of 2477 scientific studies on MCMV are analyzed. Few studies were found to assess living conditions from a user perspective at the residential unit scale. Although improvements have occurred on some social issues, the siting of housing on the urban periphery is problematic for urban mobility, social segregation and aesthetic monotony. Also, the design model does not respond to the diverse needs of inhabitants. Opportunities are identified for actions and essential missing research on mass housing. Retrofit strategies are urgent and social cost studies should induce change to the design model. Evidence-based research is needed to support policies and design processes for affordable and quality housing solutions that value users, their needs and aspirations.

Extracellular ATP, in association with [Ca2+]i regulation, is required to maintain basal ciliary beat frequency.Increasing extracellular ATP levels increases ciliary beating in airway epithelial cells, maintaining a sustained response by inducing the release of additional ATP.Extracellular ATP levels in the millimolar range, previously associated with pathophysiological conditions of the airway epithelium, produce a transient arrest of ciliary activity.The regulation of ciliary beat frequency is dependent on ATP release by hemichannels (connexin/pannexin) and P2X receptor activation, the blockage of which may even stop ciliary movement.The force exerted by cilia, measured by atomic force microscopy, is reduced following extracellular ATP hydrolysis. This result complements the current understanding of the ciliary beating regulatory mechanism, with special relevance to inflammatory diseases of the airway epithelium that affect mucociliary clearance.

In order to have a tool to empirically test the ideas derived from a theoretical model, we extended a protocol for evaluation of episodic-like memory in rats, based on the triad "what, where, context" for definition of memories. As with the computational model, our intention was for the animal being tested to store a specific number of object-place-context configurations as different memories, which would then be retrievable from cues. The aim of this work was to evaluate the influence of the number of configurations to be memorized on the performance of the task. Sixty-five Wistar male rats were evaluated. In accordance with previous work, for two configurations, the recognition index was indicative of recognition of the element mismatching the original memory (mean = 0.28; SEM = 0.12). The recognition index for three configurations was lower (mean = 0.15; SEM = 0.10), evidencing less recall with increasing requirements. The results also showed a trend toward recognition of novelty for the first and the last memory when evaluating three configurations (a "U" shape in the exploratory preference's curve), showing the primacy and recency effects typical of memory both in humans and animals. Nonetheless, the data presented a high inter-subject variability which makes the test non-robust for small groups. However, if used before and after a treatment for a same subject, we suggest that the protocol presented in this work can be a useful behavioral test for the evaluation of episodic-like memory in rats in terms of a variable task demand.

Summary: Background: The Omicron variant has challenged the control of the COVID-19 pandemic due to its immuno-evasive properties. The administration of a booster dose of a SARS-CoV-2 vaccine showed positive effects in the immunogenicity against SARS-CoV-2, effect that is even enhanced after the administration of a second booster. Methods: During a phase-3 clinical trial, we evaluated the effect of a second booster of CoronaVac®, an inactivated vaccine administered 6 months after the first booster, in the neutralization of SARS-CoV-2 (n = 87). In parallel, cellular immunity (n = 45) was analyzed in stimulated peripheral mononuclear cells by flow cytometry and ELISPOT. The acrosome reaction (AR) is the exocytosis of the acrosomal vesicle in response to different physiological and non-physiological stimuli. Particularly in mammals, the AR is needed for sperm to fuse with the oocyte plasma membrane, and it occurs only in capacitated sperm. Previous evidence in the literature indicates that extracellular ATP induces the AR in capacitated human and bovine spermatozoa, but its receptor has not yet been identified. The aim of this work was to define a putative ATP receptor in rat spermatozoa using pharmacological and biochemical approaches. We found that ATP induced the AR only in capacitated rat spermatozoa, which was inhibited in the presence of two general inhibitors of ATP receptors (P2 receptors), Suramin, and oxidized ATP (oATP), and one inhibitor of P2X receptor (pyridoxalphosphate-6-azophenyl-2',4'-disulfonic acid [PPADS]). In addition, the AR induced by ATP in capacitated rat spermatozoa was inhibited by brilliant blue-G (BB-G) and 17-β-oestradiol, two blockers of P2X7 receptors. Moreover, the ATP analog 2'(3')-O-(4-benzoylbenzoyl) ATP (BzATP) was almost 500 times more potent than ATP to induce the AR, which agrees with the pharmacology of a P2X7 receptor. Here, we show the presence of P2X7 receptor by Western blot and its localization in the tail and acrosome by indirect immunofluorescence. Finally, we quantify the presence of ATP in the rat oviduct during the estrous cycle. We found that the ATP concentration within the lumen of the oviduct is similar to those required to induce acrosome reaction, which agree with its role during in vivo fertilization. Therefore, our results strongly suggest that ATP induces the AR in capacitated rat spermatozoa through a P2X7 receptor, which may be functional during in vivo fertilization.

ABSTRACT Multiple vaccines against severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) have been evaluated in clinical trials. However, trials addressing the immune response in the pediatric population are scarce. The inactivated vaccine CoronaVac has been shown to be safe and immunogenic in a phase 1/2 clinical trial in a pediatric cohort in China. Here, we report interim safety and immunogenicity results of a phase 3 clinical trial for CoronaVac in healthy children and adolescents in Chile. Participants 3 to 17 years old received two doses of CoronaVac in a 4-week interval until 31 December 2021. Local and systemic adverse reactions were registered for volunteers who received one or two doses of CoronaVac. Whole-blood samples were collected from a subgroup of 148 participants for humoral and cellular immunity analyses. The main adverse reaction reported after the first and second doses was pain at the injection site. Four weeks after the second dose, an increase in neutralizing antibody titer was observed in subjects relative to their baseline visit. Similar results were found for activation of specific CD4+ T cells. Neutralizing antibodies were identified against the Delta and Omicron variants. However, these titers were lower than those for the D614G strain. Importantly, comparable CD4+ T cell responses were detected against these variants of concern. Therefore, CoronaVac is safe and immunogenic in subjects 3 to 17 years old, inducing neutralizing antibody secretion and activating CD4+ T cells against SARS-CoV-2 and its variants. IMPORTANCE This work evaluated the immune response induced by two doses of CoronaVac separated by 4 weeks in healthy children and adolescents in Chile. To date, few studies have described the effects of CoronaVac in the pediatric population. Therefore, it is essential to generate knowledge regarding the protection of vaccines in this population. Along these lines, we reported the anti-S humoral response and cellular immune response to several SARS-CoV-2 proteins that have been published and recently studied. Here, we show that a vaccination schedule consisting of two doses separated by 4 weeks induces the secretion of neutralizing antibodies against SARS-CoV-2. Furthermore, CoronaVac induces the activation of CD4+ T cells upon stimulation with peptides from the proteome of SARS-CoV-2. These results indicate that, even though the neutralizing antibody response induced by vaccination decreases against the Delta and Omicron variants, the cellular response against these variants is comparable to the response against the ancestral strain D614G, even being significantly higher against Omicron.
