Milk fat plays an important role in the texture of yogurt. Thus, producing fat-free yogurt is a challenge for the food industry. We investigated and modeled the effect of yogurt composition and process parameters on the texture properties obtained by analyzing the texture profile analysis (TPA) and on the rheological properties. The experiment followed the Box-Behnken (DBB) design with three factors: centrifuge rotational speed (CRS) (3000, 4250 and 5500 rpm), casein (Cas) (3.4, 4.9 and 6.4% p/v) and transglutaminase enzyme (TgE) (0, 1 and 2 U/g of protein) concentrations. A complete second-order model better explained the effect of factors on texture properties. Desirability function was selected for the optimization of responses (firmness, cohesiveness and apparent viscosity placed at a maximum level and adhesiveness, gumminess, thixotropy and loss tangent (tan δ) were placed at a minimum level). The results suggest that the addition of TgE showed less effect on texture properties. The optimized values of each factor, Cas, TgE and CRS were, respectively, 3.4%, 1 U/g of protein and 5500 rpm with general desirability (D) of 0.6531. Under these conditions, it is possible to prepare a concentrated fat-free yogurt with a higher protein content due to the concentration, obtained after centrifugation, as well as obtaining desirable texture characteristics such as greater firmness and consistency, low gumminess and adhesiveness. La grasa de la leche desempeña un papel importante en la textura del yogur. Por ello, la producción de yogur sin grasa es un reto para la industria alimentaria. Investigamos y modelamos el efecto de la composición del yogur y de los parámetros del proceso sobre las propiedades de textura obtenidas mediante el análisis del perfil de textura (TPA) y reológia. El experimento siguió el diseño Box-Behnken (DBB) con tres factores: velocidad de rotación de la centrífuga (CRS) (3000, 4250 y 5500 rpm), caseína (Cas) (3.4, 4.9 y 6.4% p/v) y enzima transglutaminasa (TgE) (0, 1 y 2 U/g de proteína). Modelo completo de segundo orden explico el efecto de los factores sobre las propiedades de textura. Se seleccionó la función de deseabilidad para la optimización de las respuestas (la firmeza, la cohesividad y la viscosidad aparente se situaron en un nivel máximo y la adhesividad, la gomosidad, la tixotropía y la tangente de pérdida (tan δ) se situaron en un nivel mínimo). Los resultados sugieren que la adición de TgE mostró un menor efecto sobre las propiedades de textura. Los valores optimizados de cada factor, Cas, TgE y CRS fueron, respectivamente, 3.4%, 1 U/g de proteína y 5500 rpm con una deseabilidad general (D) de 0.6531. En estas condiciones, es posible preparar un yogur concentrado sin grasa con un mayor contenido en proteínas debido a la concentración, obtenida tras la centrifugación, así como obtener características de textura deseables como una mayor firmeza y consistencia, baja gomosidad y adhesividad. A gordura do leite desempenha um papel importante na textura do iogurte. Assim, a produção de iogurte sem gordura é um desafio para a indústria de alimentos. Nesse sentido, objetivou-se investigar e modelar o efeito da composição do iogurte e dos parâmetros do processo nas propriedades da textura obtidas por meio da análise do perfil da textura (TPA) e do estudo reológico. O experimento foi conduzido segundo Delineamento Box-Behnken (DBB) com três fatores: velocidade de rotação da centrífuga (CRS) (3000, 4250 e 5500 rpm), concentração de caseína (Cas) (3,4, 4,9 e 6,4% p/v) e de enzima transglutaminase (TgE) (0, 1 e 2 U/g de proteína). Os resultados sugerem que a adição de TgE teve menor efeito nas propriedades de textura. Os valores otimizados de cada fator Cas, TgE e CRS foram, respectivamente, 3,4%, 1 U/g de proteína e 5500 rpm com desejabilidade geral (D) de 0,6531. Nestas condições, é possível preparar um iogurte concentrado sem gordura com maior teor proteico devido à concentração obtida após centrifugação, bem como obter características de textura desejáveis, tais como maior firmeza e consistência e menor gomosidade e adesividade.

Neospora caninum is a parasite relevant to the veterinary field. Innate and adaptive responses against N. caninum induce effector mechanisms that limit parasite replication, but little is known about their role in humoral response. Our work aimed to verify whether key molecules in the TLR2/MyD88-mediated response would impact the production of specific IgM and IgG antibodies in mice during immunization with soluble antigens of N. caninum. We observed that lack of IFN-gamma did not negatively affect the production of specific antibodies. However, mice genetically deficient in Toll-like receptor 2, Myeloid differentiation factor 88, Interleukin 12 and inducible nitric oxide synthase presented significant decrease in antibody levels against N. caninum antigens, which also reflected in the diversity of the antigen recognized by their serum. In that sense, we show here that molecules within this innate recognition pathway may present a direct impact in the induction of an antibody response against N. caninum.

Neospora caninum is a protozoan associated with abortions in ruminants and neuromuscular disease in dogs. Classically, the immune response against apicomplexan parasites is characterized by the production of proinflammatory cytokines, such as IL-12, IFN-γ and TNF. TNF is mainly produced during the acute phases of the infections and binds to TNF receptor 1 (CD120a, p55, TNFR1) activating a variety of cells, hence playing an important role in the induction of the inflammatory process against diverse pathogens. Thus, in this study, we aimed to evaluate the role of TNF in cellular and humoral immune responses during N. caninum infection. For this purpose, we used a mouse model of infection based on wildtype (WT) and genetically deficient C57BL/6 mice in TNFR1 (Tnfr1 -/-). We observed that Tnfr1 -/- mice presented higher mortality associated with inflammatory lesions and increased parasite burden in the brain after the infection with N. caninum tachyzoites. Moreover, Tnfr1 -/- mice showed a reduction in nitric oxide (NO) levels in vivo. We also observed that Tnfr1 -/- mice showed enhanced serum concentration of antigen-specific IgG2 subclass, while IgG1 production was significantly reduced compared to WT mice, suggesting that TNFR1 is required for regular IgG subclass production and antigen recognition. Based on our results, we conclude that the TNF-TNFR1 complex is crucial for mediating host resistance during the infection by N. caninum.

Essa doença possui grande importância econômica pois causa abortamentos em bovinos. Atualmente não há tratamento e nem vacinas e, por isso, o estudo do parasito e das suas relações com o meio ambiente é de suma importância. Sabe-se que aves são reservatórios de N. caninum, porém o papel da neosporose nestes hospedeiros ainda não está claro. O presente trabalho busca caracterizar o genótipo de uma nova cepa de N. caninum, descrita como Nc-UDI4, isolada do cérebro de uma galinha da região de Uberlândia/MG. Buscou-se ainda compreender o comportamento biológico desta cepa comparando-a com outras duas cepas de N. caninum, Nc-1 e Nc-Liverpool (Nc-Liv). Análises sorológicas mostraram que este novo isolado induz anticorpos específicos anti- N. caninum em camundongos e, através da reação de cadeia em polimerase (PCR), identificou-se que Nc-UDI4 possui o gene Nc5 específico de N. caninum. Além disso, Nc-UDI4 teve uma taxa de alinhamento de 99,8% com o genoma de referência Nc-Liv, conforme as análises de sequenciamento de nova geração (NGS). Foram identificados polimorfismos de nucleotídeo único (SNPs), inserções e deleções (InDels) entre Nc-UDI4 e Nc-Liv. Os principais genes que tiveram mutações foram os responsáveis pela adesão, invasão e sobrevivência do parasito e, conforme mostrado no ensaio de placa, Nc-UDI4 conseguiu invadir e se replicar menos que Nc-Liv. Foram feitas análises de microssatélites que dividiram os isolados de N. caninum em 2 clusters principais, sendo o genótipo de Nc-UDI4 mais próximo de cepas Argentinas e de Nc-1. Nc-UDI4 demonstrou ser menos virulento que Nc-Liv, pois induziu menor mortalidade de camundongos e perfil de citocinas diferente. Portanto, camundongos C57BL/6 infectados com Nc-UDI4 e Nc-1 conseguem ser mais resistentes a infecção por equilibrarem a resposta imune pró e anti-inflamatória, enquanto Nc-Liv modula essa resposta favorecendo a infecção do parasito no hospedeiro. Neospora caninum is a protozoan that belongs to the Apicomplexan phylum, has worldwide distribution and causes neosporosis. This disease induces abortions in cattle and has a significant economic impact. There are no treatment nor vaccines against the parasite so, it is important to study the parasite and its relationship with the environment. It is known that birds are reservoirs for N. caninum, but the actual relevance of neosporosis in birds it is not clear yet. In the present work we aimed to characterize the genotype of a new strain of N. caninum isolated from the brain of a chicken in the city of Uberlandia/MG, Brazil, and compare its biological features with Nc-1 and Nc-Liv. Serological analysis showed that this new strain induces anti-N. caninum antibodies in mice and, using polymerase chain-reaction (PCR), we amplified the Nc5 sequence in its genomic DNA. Furthermore, Nc-UD4 genome aligned 99,8% to the reference genome, Nc-Liv, according to new generation sequencing (NGS) analysis. We identify single nucleotide polymorphisms (SNPs) and insertions/deletions (InDels) between Nc-UDI4 and Nc-Liv. Genes related to adhesion, invasion and survival of the parasite were altered and, as we showed in plaque assay, Nc-UDI4 had lower invasion/replication rates compared to Nc-Liv. Microsatellite analysis shared N. caninum in two clusters and revealed that the genotype of Nc-UDI4 is closely related to the Argentinian strains and Nc-1. Nc-UDI4 and Nc-1 are less virulent than Nc-Liv, as it induced low mortality of C57BL/6 mice and also a different cytokine profile. In that sense, mice infected with Nc-UDI4 and Nc-1 are more resistant to the infection, allied to a more favourable balance of pro- and anti-inflammatory immune responses against N. caninum, while Nc-Liv modulated this response in its favor, contributing to a worst outcome to its host. Coordenação de Aperfeiçoamento de Pessoal de Nível Superior O transporte de pessoas e de mercadorias contribui na deterioração do meio ambiente nos centros urbanos, associado à geração de alguma forma de poluição, seja atmosférica, sonora, do solo e da água ou intrusão visual. Os veículos pesados, que utilizam óleo diesel como combustível, são os principais responsáveis pela emissão de óxidos de nitrogênio (NOx) e material particulado (MP), contribuindo para a participação do setor de transportes na poluição atmosférica. Além disso, há a emissão de Gases do Efeito Estufa (GEE) e o seu principal componente, o dióxido de carbono (CO2). Na maioria das grandes cidades, o transporte coletivo por ônibus é muitas vezes considerado uma alternativa menos poluente comparada aos veículos particulares, tendo em vista o potencial de reduzir, por passageiro, as emissões de GEE e poluentes atmosféricos. A área de estudo foi a cidade de Uberlândia e os objetos de estudo foram as linhas troncais do Sistema Integrado de Transporte (SIT). Foram quantificadas as emissões de NOx, MP e CO2 por meio da abordagem bottom-up em que foi utilizado o trajeto de cada linha de ônibus e também o consumo de combustível obtido por meio de simulação no software TSIS. O software possui algumas limitações quanto aos resultados e não informa no relatório a emissão de poluentes por ônibus, além de não se poder alterar especificações quanto ao combustível utilizado na frota. Com os resultados obtidos nos cálculos de emissões de poluentes e GEE pela abordagem bottom-up observou-se que, ao utilizar o consumo de combustível obtido em simulação, a emissão resultou maior que ao utilizar a distância. Conclui-se, também, que levando em conta os combustíveis e as distâncias percorridas há uma redução nas emissões ao se utilizar o etanol quando comparadas ao óleo diesel. The transport of people and goods contributes to the deterioration of the environment in urban areas because of the generation of pollution, such as, air, noise, soil, water or visual degradation. The heavy vehicles that use diesel as fuel are mainly responsible for the emission of nitrogen oxides (NOx) and particulate matter (PM), contributing to participation of the transport sector in air pollution. In addition, there is emission of Greenhouse Gas (GHG) whose main component is carbon dioxide (CO2). In most major cities, public transportation is often considered as a less polluting alternative compared to the private vehicle, in view of the potential to reduce, per passenger, the emissions of GHG and air pollutants. The study area was the city of Uberlândia and the objects of study were the trunk lines of the Sistema Integrado de Transporte (SIT). The emissions of NOx, PM and CO2 were estimated through the bottom-up approach which used the route of each bus line and also fuel consumption obtained through simulation from the TSIS software. The software has some result limitations, there are no report about the emission of pollutants by bus, and it is not able to change specifications for the fuel used by the fleet. The results obtained through calculations of pollutants and GHG emission by the bottom-up approach show that the emission is higher when using fuel comsuption obtained in simulation than using distance. For the results considering fuel and distance there was a reduction in emissions comparing ethanol and diesel. Dissertação (Mestrado)

Objetivo: Analisar o perfil epidemiológico das pessoas diagnosticadas com HIV/AIDS. Métodos: Trata-se de um estudo quantitativo feito por meio do DATASUS. Foram avaliados individualmente sexo, idade, escolaridade, região e a raça/cor de pessoas diagnosticadas com HIV/AIDS no período de 2015 a 2019. Resultados: O sexo masculino é o mais atingido pela doença com 69%, a faixa etária é entre 25 a 49 anos (67% do total de casos). A escolaridade 36% das pessoas que foram diagnosticadas com HIV/AIDS ensino fundamental incompleto, 56% não concluíram o Ensino médio, 26% tinham o ensino médio completo, 6% Ensino Superior Incompleto e 11% concluíram o Ensino Superior. A região Sudeste foi a que mais teve casos com 39% do total e a raça/cor dessas pessoas, foi uma informação ignorada em 76.295 dos registros. Sendo assim, utilizou-se como 100% dos casos apenas 94.218, sendo a maioria eram de etnia parda (46%). Conclusão: Os resultados deste trabalho reforçam a necessidade de uma maior implementação de políticas públicas de disseminação de saberes sobre a prevenção da infecção pelo hiv. Assim, prevenindo também a AIDS, visto que a população mais atingida é a de baixa escolaridade e jovens-adultos homens.

Esse parasito possui uma grande importância na medicina veterinária, pois está relacionado à doença neuromuscular em cães e ao abortamento em bovinos, gerando perdas afetivas e econômicas. A resposta imune de padrão Th1 é uma das principais responsáveis pelo controle da infecção, mediando a ativação de vias celulares culminando na produção de moléculas efetoras que reduzem a replicação do parasito. Esse trabalho teve como objetivo, avaliar o papel de alguns destes fatores na produção e função de anticorpos específicos contra os antígenos de N. caninum. Para avaliar a produção e função dos anticorpos produzidos, camundongos WT e geneticamente deficientes para TLR2, MyD88, IFN-, IL-12, MHCII e iNOS foram imunizados com antígeno solúvel do parasito (NLA). Amostras de soros foram obtidos em cinética durante a imunização e quantificados por ELISA indireto. O perfil de reconhecimento antigênico destas amostras foi avaliado por immunoblotting e as funções biológicas desses anticorpos, analisadas por meio de ensaios de lise mediada pelo complemento e opsonização dos parasitos. Os resultados obtidos mediante estas análises demonstraram que a ausência de alguns dos fatores cruciais para a indução de uma resposta Th1 (TLR2, IFN- e IL-12) não alteraram a produção de anticorpos IgM e IgG, camundongos MyD88-/-, MHCII-/- e iNOS-/- apresentaram uma redução significativa desses níveis de anticorpos e seu reconhecimento antigênico. Tal redução na produção desses anticorpos nestes animais geneticamente deficientes encontrou-se associado a redução de suas funções clássicas, como lise mediada pelo complemento e opsonização. Neste sentido, demonstramos que MyD88, MHCII e iNOS, fatores relacionados a resposta imune celular de padrão Th1, são cruciais para o desenvolvimento de uma resposta imune humoral adequada contra N. caninum. Th1-based immune responses against N. caninum is one of the major responsible to control infection, inducing the release of effector molecules that limits parasite replication. In that context, this study aimed to define the role of key Th1-related molecules in the synthesis and functions of antibodies during exposure to N. caninum antigens. To evaluate the production and function of these antibodies, WT and genetically deficient mice for TLR2, MyD88, IFN-, IL-12, MHCII and iNOS were immunized with soluble antigens of N. caninum (NLA). Kinetics of serum samples during immunization were measured by indirect ELISA. Profile of antigen recognition were evaluated by immunoblotting and biological functions assays analyzed by complement-mediated lysis and opsonization. We observed that in the absence of crucial factors in induction of Th1 response (TLR2, IFN- e IL-12) specific production of IgM and IgG antibodies was not impaired. Mice MyD88-/-, MHCII-/- and iNOS-/- presented significant decrease in the levels of these antibodies and in antigenic recognition. That reduction in these groups directly reflected in compromised lysis and opsonization properties of the hyperimmune sera. So, we demonstrate that factors related to Th1 immune response as MyD88, MHCII and iNOS, are crucial to induce humoral immune responses against N. caninum. Dissertação (Mestrado)

Activities of phospholipases (PLAs) have been linked to pathogenesis in various microorganisms, and implicated in cell invasion and so the interest in these enzymes as potential targets that could contribute to the control of parasite survival and proliferation. Chicken eggs immunized with BnSP-7, a Lys49 phospholipase A2 (PLA2) homologue from Bothrops pauloensis snake venom, represent an excellent source of polyclonal antibodies with potential inhibitory activity on parasite PLAs. Herein, we report the production, characterization and anti-parasitic effect of IgY antibodies from egg yolks of hens immunized with BnSP-7. Produced antibodies presented increasing avidity and affinity for antigenic toxin epitopes throughout immunization, attaining a plateau after 4weeks. Pooled egg yolks-purified anti-BnSP-7 IgY antibodies were able to specifically recognize different PLA2s from Bothrops pauloensis and Bothrops jararacussu venom. Antibodies also neutralized BnSP-7 cytotoxic activity in C2C12 cells. Also, the antibodies recognized targets in Leishmania (Leishmania) amazonensis and Toxoplasma gondii extracts by ELISA and immunofluorescence assays. Anti-BnSP-7 IgY antibodies were cytotoxic to T. gondii tachyzoite and L. (L.) amazonensis promastigotes, and were able to decrease proliferation of both parasites treated before infection. These data suggest that the anti-BnSP-7 IgY is an important tool for discovering new parasite targets and blocking parasitic effects.

Objetivo: a hiperplasia condilar unilateral é uma condição rara do desenvolvimento ósseo causada pela hiperatividade do centro de crescimento que resulta em um crescimento excessivo e autolimitado do côndilo mandibular. Caracteriza-se e pelo alongamento progressivo do processo condilar levando a características como assimetria facial, alongamento do terço inferior da face, má oclusão e ocasionalmente disfunção temporomandibular. Diante das formas de tratamento, o preparo ortodôntico associado à cirurgia ortognática e a condilectomia apresentam resultados satisfatórios. Relato de Caso: caso clínico de um paciente do sexo masculino, 28 anos de idade, portador de hiperplasia condilar unilateral com assimetria facial severa, protrusão mandibular e desvio do mento. O tratamento foi realizado no Hospital Federal dos Servidores do Estado através de cirurgia ortognática maxilomandibular com mentoplastia associada à condilectomia alta, planejada previamente utilizando modelos estereolitográficos. A condilectomia resultou na remoção do centro responsável pela hiperatividade condilar permitindo um segundo tempo cirúrgico para correção da deformidade esquelética com melhora estético-funcional do paciente. Conclusão: o mesmo se encontra em controle pós-operatório de seis anos, sem recidiva e com um padrão de oclusão satisfatório.

Hymenoptera venoms constitute an interesting source of natural toxins that may lead to the development of novel therapeutic agents. The present study investigated the enzymatic and biological characteristics of the crude venom of the ant Odontomachus bauri. Its crude venom presents several protein bands, with higher staining for six proteins with gelatinolytic activity (17, 20, 26, 29, 43 and 48 kDa). The crude venom showed high proteolytic activity on azocasein at optimal pH 8.0 and 37 °C. In the presence of protease inhibitors as aprotinin, leupeptin and EDTA, the azocaseinolytic activity was reduced by 45%, 29% and 9%, respectively, suggesting that the enzymes present in the crude venom belong to the three classes of proteases, with the serine proteases in greater intensity. The crude venom degraded the fibrinogen α-chain faster than the β-chain, while the fibrinogen γ-chain remained unchanged. In biological assays, O. bauri venom showed hemolytic and coagulant activity in vitro, and defibrinating activity in vivo. In addition, the venom showed antimicrobial activity against Staphylococcus aureus and Escherichia coli as well as antiparasitic activity on Toxoplasma gondii infection in vitro. In that sense, this study sheds perspectives for pharmacological applications of O. bauri venom enzymes.

Introdução: A hiperplasia condilar é uma condição rara do desenvolvimento ósseo causada pela hiperatividade do côndilo mandibular que resulta em seu crescimento excessivo e autolimitado. Caracteriza-se pelo alongamento progressivo do processo condilar levando a assimetria facial, alongamento do terço inferior da face, má oclusão e ocasionalmente disfunção temporomandibular. Diante das formas de tratamento, o preparo ortodôntico associado à cirurgia ortognática e a condilectomia apresentam resultados satisfatórios. Objetivo: O objetivo deste trabalho é apresentar o caso clínico de um paciente do sexo masculino, 28 anos de idade, portador de hiperplasia condilar unilateral com assimetria facial severa, protrusão mandibular e desvio do mento. Relato de caso: O tratamento foi realizado no Hospital Federal dos Servidores do Estado/RJ, através de cirurgia ortognáticamaxilomandibular com mentoplastia associada à condilectomia alta, planejada previamente utilizando modelos estereolitográficos. A correção da deformidade esquelética, através da cirurgia ortognática, efetuou expressiva melhora estético-funcional do paciente. Conclusão: O mesmo se encontra em controle pós-operatório de seis anos, sem recidiva e com um padrão de oclusão satisfatório.

Objetivo: Identificar na literatura o uso da fonoforese para alívio da dor e inflamação em distúrbios osteomioarticulares, qualificando o nível de evidência e grau de recomendação dos estudos. Metodologia: Trata-se de um estudo de revisão de literatura, onde foram selecionados no Portal de Periódicos, da CAPES, os descritores com maior amplitude de pesquisa clínica em fonoforese: Osteoartrite de joelho, Lombalgia, Síndrome de impacto do ombro e Síndrome do Túnel do Carpo. Após selecionados estudos, foram coletados os dados de amostra, intervenção e os resultados obtidos para discussão neste trabalho. O nível de evidência de cada trabalho, bem como o grau de recomendação por seus resultados foram qualificados de acordo com as determinantes do OCEBM (Centre for Evidence-Based Medicine). Resultados: Todos os trabalhos selecionados tinham nível de evidência 1b, e grau de recomendação superior a B. Considerações finais: Das evidências encontradas, todas tinham um nível de evidência submáximo, e a maioria com o mais alto grau de recomendação, demonstrando a eficácia do recurso da fonoforese, em relação a outras formas de terapias comuns.

Eutirucallin is a lectin isolated from the latex of Euphorbia tirucalli, a plant known for its medical properties. The present study explores various characteristics of Eutirucallin including stability, cytotoxicity against tumor cells, antimicrobial and antiparasitic activities. Eutirucallin was stable from 2 to 40 days at 4°C, maintained hemagglutinating activity within a restricted range, and showed optimal activity at pH 7.0–8.0. Eutirucallin presented antiproliferative activity for HeLa, PC3, MDA-MB-231, and MCF-7 tumor cells but was not cytotoxic for non-tumorigenic cells such as macrophages and fibroblasts. Eutirucallin inhibited the Ehrlich ascites carcinoma in vivo and it was also observed that Eutirucallin inhibited 62.5% of Escherichia coli growth. Also, Eutirucallin showed to be effective when tested directly against Toxoplasma gondii infection in vitro. Therefore, this study sheds perspectives for pharmacological applications of Eutirucallin.

Films of silk fibroin (SF) and sodium alginate (SA) blends were prepared by solution casting technique. The miscibility of SF and SA in those blends was evaluated and scanning electron microscopy (SEM) revealed that SF/SA 25/75 wt.% blends underwent microscopic phase separation, resulting in globular structures composed mainly of SF. X-ray diffraction indicated the amorphous nature of these blends, even after a treatment with ethanol that turned them insoluble in water. Thermal analyses of blends showed the peaks of degradation of pristine SF and SA shifted to intermediate temperatures. Water vapor permeability, swelling capacity and tensile strength of SF films could be enhanced by blending with SA. Cell viability remained between 90 and 100%, as indicated by in vitro cytotoxicity test. The SF/SA blend with self-assembled SF globules can be used to modulate structural and mechanical properties of the final material and may be used in designing high performance wound dressing.

Silk fibroin (SF) films have been largely studied as biomaterials due to their biocompatibility and biodegradability. Casting a SF aqueous solution at room temperature is a common technique to produce SF films at relative low cost and processing time; however, their brittleness and solubility in water make them unsuitable for certain biomedical applications. In this study, the incorporation of additives, ethanol and glycerin, are presented as an alternative to both improve mechanical properties of SF films and decrease their solubility in water. SF films with additives were further characterized using scanning electron microscopy, X-ray diffraction, Fourier transformed infrared spectroscopy with attenuated total reflection, analysis of water solubility, mechanical test of traction, and in vitro cytotoxicity experiments. The results show that SF films containing additives are stable in water due to the effect of glycerin and ethanol, and do not require post-treatments. Furthermore, great improvements on elongation of the films were achieved, mainly in the presence of both additives. Silk fibroin (SF) is a protein fiber spun by Bombyx mori silkworm. SF fibers are about 10-25 μm wide in diameter and a single cocoon may provide over 1000 m of SF fibers. SF can present several conformations regarding protein secondary structure which ultimately define the structural properties of SF-based materials. For this reason, a rigorous control on its processing conditions shall be performed. It is known that SF has excellent properties to be used in biomaterials field, controlled release and scaffolds for tissue engineering. In addition, SF can be processed in several forms, such as films, fibers, hydrogels or microparticles. This work seeks to provide an overview on SF processing conditions, regarding the preparation of SF membranes (dense and porous), hydrogels and biocomposites, focusing on biomaterials application.

Silk hydrogels are interesting materials to be used as matrix in controlled drug delivery devices. However, methods to accelerate fibroin gelation and allow the drug incorporation during the hydrogel preparation are needed in literature. In this article we report the preparation of silk fibroin hydrogels with addition of several contents of ethanol, used to accelerate fibroin gelation kinetics, and we also evaluate the potential of these hydrogels to be used as matrices for drug delivery. Chemical and conformational properties did not change despite the amount of ethanol incorporated in the hydrogel. Hydrogels containing diclofenac sodium dissolved in ethanol showed a faster initial release of the drug than hydrogels with the drug dissolved in water but equilibrium was reached later. This indicates a more sustained drug delivery from hydrogels in which the model drug was dissolved in ethanol. Objetivou-se avaliar o perfil metabólico energético, proteico e enzimático de vacas mestiças leiteiras com baixo escore de condição corporal (ECC) no periparto. Foram colhidas amostras sanguíneas uma semana antes do parto, no dia do parto, e aos sete, 14, 21, 28 e 43 dias pós-parto (DPP) de 36 animais, com média de ECC de 2,6±0,5, com eutocia e pós-parto fisiológico e sem tratamentos nesta fase. Analisaram-se as concentrações séricas de proteínas totais, albumina e globulinas para o perfil protéico; AST, ALT, GGT e fosfatase alcalina para o perfil enzimático; ácidos graxos não-esterificados (NEFA), β-hidroxibutirato (BHBA), triglicerídeos, colesterol e lipoproteínas (VLDL, HDL e LDL) para o perfil energético. As vacas apresentaram no pré-parto hipoproteinemia, hipoalbuminemia, hipocolesterolemia e aumento das enzimas GGT e AST. No dia do parto houve lipólise e hipoglobulinemia. Concluiu-se que vacas mestiças leiteiras com baixo ECC apresentam balanço energético negativo, hipoproteinemia com hipoalbuminemia e lesão hepática no periparto, com restabelecimento aos 30 DPP, mas não recuperam sua condição corporal até o final do puerpério. The aim of this study was to evaluate the metabolic profile of protein, energy and enzyme in crossbred dairy cows with low body condition score (BCS) in the peripartum period. Blood samples were collected from 36 animals with 2.6±0.5 BCS, eutocia, physiological postpartum and without any treatment, on following days: one week before calving, calving and 7, 14, 21, 28 and 43 days in milk (DIM). It was evaluated serum total protein, albumin and globulins for protein profile; AST, ALT, GGT and alkaline phosphatase for mineral profile; nonesterified fatty acids (NEFA), β-hydroxybutyrate (BHBA), triglycerides, cholesterol and lipoproteins (VLDL, HDL and LDL) for energy profile. Crossbred dairy cows had hypoproteinemia, hypoalbuminemia, hypocholesterolemia and increased that both enzymes AST and GGT at the precalving. There were lipolysis and hypoglobulinemia at parturition. It was concluded that crossbred dairy cows with low BCS have negative energy balance, hypoproteinemia with hypoalbuminemia and hepatic injury in the peripartum. This condition is restored at 30 DIM, but there is no recovery of the body condition by the end of puerperium.

In dairy cattle, uterine infections are not life threatening and often unavoidable; however, they reduce fertility and increase the production costs of properties. The aim of this study was to evaluate the incidence of subclinical endometritis from 32 to 70 days in milk (DIM) and its effects on the reproductive performance of crossbred dairy cows. Lactating cows (Holstein/Gir; n = 172), with no history of retained placenta, without clinical signs of uterine infection were used. The body condition score (BCS) was evaluated on a scale from 1 to 5. Ultrasound examination was performed to evaluate uterine lining and ovarian activity, while vaginal mucus was analyzed by gloved hand. The diagnosis of subclinical endometritis was performed by endometrial cytobrush technique. The samples were collected, stained, and examined microscopically; positive cases for subclinical endometritis were considered with the presence of ≥5 % of neutrophils. Later, the cows were submitted to conventional artificial insemination or timed artificial insemination. The incidence of subclinical endometritis in the herd was 26 %, and this was not affected by the season of calving, presence of corpus luteum, DIM, and parity. Cows with a BCS ≤2.50 had a higher incidence of subclinical endometritis. The conception rate to first insemination and pregnancy rate at 150 days postpartum were not influenced by the presence of subclinical endometritis in crossbred dairy cows.
