Clavibacter michiganensis subsp. michiganensis (Smith) Davis et al. (Cmm) and Ralstonia solanacearum Yabuuchi et al. (Smith) (Rs) are important seed-borne bacterial pathogens of tomato (Solanum lycopersicum) listed as A2 pests in the EPPO (European and Mediterranean Plant Protection Organization) region. At present, there are few strategies to control these pathogens, and seed control with eco-compatible approaches is widely encouraged. In this work, the essential oils (EOs) of oregano (Origanum vulgare), garlic (Allium sativum), basil (Ocimum basilicum), cinnamon (Cinnamomum zeylanicum), clove buds (Syzygium aromaticum), thyme (Thymus vulgaris), and Trametes versicolor extract (Tve) were tested in vitro for their antimicrobial activity against Cmm and Rs (broth microdilution method). The tested EOs and the Tve extract caused a significant inhibition of bacterial growth, with very promising MBC (minimum bactericidal concentration) and MIC90 (minimum inhibitory concentration causing a 90% growth inhibition) values. Moreover, an in vivo germination test showed no major reduction in seed germination when the substances were applied as seed treatment. A rapid molecular screening method has been developed, through real-time PCR, for the specific quantification of Cmm in the presence of a vegetable matrix to test in vivo the antimicrobial efficacy of oregano and cinnamon oil on seed treatment without resorting to whole plant essays, which are time- and space-consuming.

The aim of this study was to test the hypothesis that duplication/hybridization of functional domains of naturally occurring pellicle peptides amplified the inhibitory effect of hydroxyapatite crystal growth, which is related to enamel remineralization and dental calculus formation. Histatin 3, statherin, their functional domains (RR14 and DR9), and engineered peptides (DR9-DR9 and DR9-RR14) were tested at seven different concentrations to evaluate the effect on hydroxyapatite crystal growth inhibition. The heterothallic ascomycete Fusarium fujikuroi (teleomorph: Gibberella fujikuroi) is the causal agent of bakanae of rice, a disease of increasing economic importance in the major rice-producing areas in the world and a serious threat for Italian rice cultivation. A few studies have characterized F. fujikuroi isolates in America and the Philippines but no data are available on the genetic structure of Italian pathogen populations. Microsatellite SSRs are useful tools to study the intraspecific diversity at population level. In this study, 19 polymorphic SSRs have been identified and applied to characterize the genetic variation of 334 isolates of F. fujikuroi coming from eight Italian rice-growing areas. A high degree of diversity at haplotype level has emerged: in the eight populations, 107 unique haplotypes were scored. Analysis of molecular variance (amova) showed that 98% of genetic variability occurred within F. fujikuroi Italian populations, as confirmed by the allelic Shannon index ranging from 0.56 to 1.06. The presence of a 1:1 ratio of mating type alleles in six out of eight of the Italian fungal populations suggests a potential for sexual reproduction in the field. However, the high fraction of clonality (43%), confirmed by neighbour-joining clustering analysis, and the high level of linkage disequilibrium observed, indicates that reproduction of F. fujikuroi is mostly clonal in Italy. All data suggest that the observed genetic variability was probably mediated by human activity and transmission by rice seeds.

Breeding for resistance is the most effective tool for controlling the corky root disease of tomato caused by Pyrenochaeta lycopersici. A comparative RNA-Seq-based transcriptomic analysis was conducted at 96 hpi (hours post infection) on two tomato cultivars: resistant Mogeor and its genetic background, and susceptible Moneymaker to investigate the differences in their transcriptomic response and identify the molecular bases of this plant-pathogen interaction. The number of differentially expressed genes (DEGs) identified was much higher in the susceptible than in the resistant genotype; however, the proportion of upregulated genes was higher in Mogeor (70.81%) than in Moneymaker (52.95%). Gene Ontology (GO) analysis enabled identification of 24 terms shared by the two cultivars that were consistent with responses to external stimulus, such as fungal infection. On the other hand, as many as 54 GO were enriched solely in Moneymaker, including terms related to defense response and cell wall metabolism. Our results could support the previous observations in other pathosystems, that susceptibility and resistance have overlapping signaling pathways and responses, suggesting that the P. lycopersici resistance gene pyl might be a recessive allele at a susceptibility locus, for which different candidate genes were identified based on the differences in induction or expression levels, observed between the resistant and susceptible genotype. MapMan analysis highlighted a complex hormone and transcription factors interplay where SA- and JA-induced pathways are modulated in a similar way in both genotypes and thus take part in a common response while the ethylene signaling pathways, induced mainly in susceptible Moneymaker, seem putatively contribute to its susceptibility.

Os peptídeos da estaterina (DR9) e da histatina 3 (RR14), que ocorrem naturalmente na película in vivo, amplificam o efeito inibitório do crescimento de cristais de hidroxiapatita, função relacionada à remineralizarão do esmalte e formação de cálculos dentários. A hipótese da duplicação/hibridação de domínios funcionais dos peptídeos DR9 da estaterina e RR14 da histatina 3 foi testada. Para isto, os peptídeos peptidomiméticos (DR9-DR9, DR9-RR14), além deles individualmente e suas proteínas intactas (DR9, RR14, estaterina e histatina 3) foram estudados em sete concentrações diferentes para avaliar o efeito da inibição do crescimento de cristais de hidroxiapatita. Foi utilizado um ensaio colorimétrico de microplaca para quantificar o crescimento de cristais de hidroxiapatita. As experiências foram feitas em triplicata e a concentração inibitória (IC50) foi estabelecida para cada grupo. A IC50 foi calculada para todos os peptídeos e proteínas testados. A histatina 3 e o RR14 não atingiram o valor de IC50. O DR9- RR14 atingiu o valor de IC50 a 3,80 M. Como esperado, DR9 e DR9-DR9 demonstraram um efeito inibitório significativo na atividade de crescimento de cristais, atingindo o valor de IC50 a 2,82 M e 1,07 M, respectivamente. Background Bakanae disease, caused by seed-borne Fusarium species, mainly F. fujikuroi, is a rice disease whose importance is considerably increasing in several rice growing countries, leading to incremental production losses. Results A germplasm collection of japonica rice was screened for F. fujikuroi resistance, allowing the identification of accessions with high-to-moderate levels of resistance to bakanae. A GWAS approach uncovered two genomic regions highly associated with the observed phenotypic variation for response to bakanae infection on the short arm of chromosome 1 (named as qBK1_628091) and on the long arm of chromosome 4 (named as qBK4_31750955). High levels of phenotypic resistance to bakanae were associated to the cumulated presence of the resistant alleles at the two resistance loci, suggesting that they can provide useful levels of disease protection in resistance breeding. A fine comparison with the genomic positions of qBK1_628091 and qBK4_31750955 with respect to the QTLs for bakanae resistance reported in the literature suggests that the resistant loci here described represent new genomic regions associated to F. fujikuroi resistance. A search for candidate genes with a putative role in bakanae resistance was conducted considering all the annotated genes and F. fujikuroi-related DEGs included in the two genomic regions highlighting several gene functions that could be involved in resistance, thus paving the way to the functional characterization of the resistance loci. Conclusions New effective sources for bakanae resistance were identified on rice chromosomes 1 and 4 and tools for resistance breeding are provided. Background Pyrenochaeta lycopersici is a soil-dwelling ascomycete pathogen that causes corky root rot disease in tomato (Solanum lycopersicum) and other Solanaceous crops, reducing fruit yields by up to 75%. Fungal pathogens that infect roots receive less attention than those infecting the aerial parts of crops despite their significant impact on plant growth and fruit production. Results We assembled a 54.9Mb P. lycopersici draft genome sequence based on Illumina short reads, and annotated approximately 17,000 genes. The P. lycopersici genome is closely related to hemibiotrophs and necrotrophs, in agreement with the phenotypic characteristics of the fungus and its lifestyle. Several gene families related to host–pathogen interactions are strongly represented, including those responsible for nutrient absorption, the detoxification of fungicides and plant cell wall degradation, the latter confirming that much of the genome is devoted to the pathogenic activity of the fungus. We did not find a MAT gene, which is consistent with the classification of P. lycopersici as an imperfect fungus, but we observed a significant expansion of the gene families associated with heterokaryon incompatibility (HI). Conclusions The P. lycopersici draft genome sequence provided insight into the molecular and genetic basis of the fungal lifestyle, characterizing previously unknown pathogenic behaviors and defining strategies that allow this asexual fungus to increase genetic diversity and to acquire new pathogenic traits.

Many fungal plant pathogens secrete an array of cell wall degrading enzymes mainly involved in the pathogenesis. In this work, a cDNA clone encoding an extracellular endo-1,4-β-glucanase (named PlEGL1) from the causal agent of the Corky Root Rot of tomato, Pyrenochaeta lycopersici, was isolated and characterized, in order to understand its putative role in the pathogenesis and its mechanism of action. Multiple alignment of the deduced amino acidic sequence shows a high homology with other endoglucanases from different phytopathogenic fungi and detects a well-defined conserved domain of the Glycosyl Hydrolase family 61 (GH61). In vitro, Plegl1 gene transcription is correlated to a cellulolytic activity of the fungus, regulated, in its turn, by the presence of sugar and/or cellulose in the culture medium. In the infected plants, expression level of Plegl1 is positively correlated to the development of the disease. PlEGL1 was heterologously expressed in Escherichia coli and the recombinant protein was purified and tested for its cellulolytic ability, showing a very weak activity, in agreement with all the endoglucanases belonging to GH61 family. The finding in this paper will provide the basis for further determination of biochemical properties of the PlEGL1 protein and its possible involvement in the host–pathogen interaction.

Tese de Doutoramento em História da Arte da Antiguidade Com o estudo dos mamíferos representados nos mosaicos romanos da Península Ibérica, foi possível estabelecer seriações homogéneas, por tipo ou espécie. Foi também possível entender qual o papel desempenhado pelos animais na Antiguidade, até porque assumiram neste contexto um lugar que ultrapassou o simples registo figurado. Constatou-se a existência de diferentes níveis de representatividade, cuja iconologia ultrapassou o mero aparato social, bem como registos específicos de índole diversa. De facto, os mamíferos desempenharam um importante papel nas sociedades Antiga e Tardo-Antiga, adquirindo um estatuto próprio que conduziu à sua representação, sob diversas formas, nos mosaicos. Os mamíferos assumiram, pois, um lugar destacado na sociedade, cultura, mentalidade e economia, justificando a sua presença nos mosaicos que pavimentavam espaços públicos, domus e uillæ. Afigurou-se-nos, por outro lado, importante efectuar uma leitura formal dessas representações e perceber se, no lato período considerado, sofreram ou não modificações substantivas no modo de as construir. Ou seja, se a introdução da policromia e a evolução do gosto e, consequentemente, do desenho contribuíram para a obtenção de níveis representativos distintos e, ainda, se existem, por exemplo no debuxo dos animais, elementos constantes e que, aprioristicamente permitam realizar uma identificação ou associações inequívocas, assim como as mudanças operadas na aparência e significado dessas representações, isto já em plena Antiguidade tardia.

A figura de Orlando Ribeiro e a sua influência na Geografia de Portugal, na segunda metade do século XX, são incontestáveis. Neste sentido, dirigindo o nosso estudo para uma área geográfica de singular valor paisagístico – o Alto Douro – tentamos compreender de que forma Orlando Ribeiro a construiu na sua obra como unidade regional. Na primeira parte deste trabalho dirigimos a nossa atenção para a figura do autor, o seu percurso académico e científico, de forma a analisar as influências teóricas e metodológicas dominantes. A segunda parte consistiu no estudo da evolução da divisão geográfica de Portugal, estudando-se as diferentes propostas e a posição de Orlando Ribeiro relativamente às mesmas. Destacamos por um lado os outros autores, particularmente, os geógrafos, e as suas obras, e por outro, os discípulos e teses orientadas por Orlando Ribeiro sobre a região do Alto Douro. Na terceira parte recolhemos através do estudo analítico das fontes disponíveis: obra científica editada, cadernos de campo e fotografias, os elementos físicos e humanos que no seu todo formam a unidade de paisagem do Alto Douro. A diversidade de elementos temáticos existentes levou a que neste ponto, seguíssemos a subdivisão proposta pelo próprio Orlando Ribeiro no seu mais conhecido livro Portugal o Mediterrâneo e o Atlântico. A quarta parte pretendeu destacar os autores e obras de referência, no Alto Douro de Orlando Ribeiro, assim como a herança entre os discípulos que, nos seus estudos académicos, escolheram esta região como objecto de estudo. Este estudo pretende constituir uma abordagem à Geografia praticada por Orlando Ribeiro, tendo por base a procura dos elementos fundamentais da leitura de paisagem que o mesmo traçou em relação ao Alto Douro. Parasitism by the endophagous braconid Aphidius ervi (Hymenoptera, Braconidae) has a negative impact on the reproductive activity of its host, Acyrthosiphon pisitin (Homoptera, Aphididae). The host castration is induced by the parasitoid venom and is reproduced by the injection of chromatographic fractions highly enriched with two proteins, of 18 (p18) and 36 kDa (p36) in size, respectively. Here we demonstrate that these bioactive proteins trigger apoptosis of the cells in the germaria and ovariole sheath of the host aphid. Both p18 and p36 were internally sequenced and the gathered information was matched against the deduced amino acid sequence of the putative proteins encoded by cDNA clones, randomly selected from a cDNA library, which was raised using mRNA extracted from A. ervi venom glands. The identified cDNA clones contained an insert corresponding to the RNA product of an interrupted gene, made of six exons and five introns, which was found to be transcribed at higher levels in adult females of A. ervi than in mates. This gene codes for a putative protein composed of 541 amino acids, with a calculated molecular mass of 56.9 kDa, which contained the amino acid sequences experimentally determined for both p18 and p36. This putative protein showed a significant level of sequence identity with gamma-glutamyl transpeptidases (gamma-GT), and it was named Ae-gamma-GT. The gamma-GTs are enzymes which play a key role in the metabolism of glutathione (GSH) and, as observed in most organisms, they are membrane-bound heterodimers formed by a large and a small subunit, which originate by post-translational processing of a single-chain precursor. The expression in insect cells of Ae-gamma-GT confirmed the occurrence of the expected post-translational processing, and demonstrated that, unlike other gamma-GTs, this protein is secreted in the extracellular environment. A measurable gamma-GT activity was detected in the venom of A. ervi and in the chromatographic fractions containing Ae-gamma-GT. Thus, we suggest that this venom protein may induce apoptosis in the host ovarioles by generating an alteration of the GSH metabolism and a consequent oxidative stress. (C) 2007 Elsevier Ltd. This cross-sectional study aimed at evaluating the oral health conditions of building construction workers from a city in the mid-west region of São Paulo, Brazil. This study involved 219 male subjects, aged 17 to 72. The definition of a random sample utilized the functional number of each worker as a criterion to the raffle, which took into account all 450 subjects registered in the Working Accidents Prevention Program. The examination of oral health conditions by DMFT index and need of treatment were carried out according to WHO criteria (1997).
