Concerns with distributional justice invariably arise in environmental governance, especially in the conservation and management of common-pool resources. These initiatives generate an array of costs and benefits that are typically heterogeneously distributed. Distribution of these impacts in a way that is considered fair by local stakeholders is not only a moral imperative, but instrumental to achieving social and ecological success given perceived unfairness fosters conflict and undermines cooperation. However, understandings of local stakeholders’ conceptions of distributional fairness are rare because research often assesses distributional outcomes based on tacit assumptions about what constitutes fairness (e.g. equality). We examine what local stakeholders consider distributional fairness with respect to monetary benefits arising from a collective payment for ecosystem services scheme in a co-managed marine protected area in Fiji. In six villages associated with the co-managed marine protected area, we elicited individuals’ fairness judgements of five distributional justice principles: equality, need, and three forms of proportionality based on customary rights, fisheries opportunity-costs, and involvement in co-management. We examine how fairness judgements are associated with socio-demographic characteristics indicative of key identities, thereby building on socially-aggregated approaches typical of the nascent literature on perceived fairness. We find the rights-based principle was considered the ‘most fair’ and the opportunity-costs principle the ‘least fair’. Our findings challenge prevailing understandings of distributional justice in conservation and commons management, which favour the principles of equality or opportunity-cost. We also find that education was significantly positively related to fairness judgements of all principles, whilst wealth was significantly related to the equality and the opportunity-based principles. These results provide insights into how fairness judgements could be influenced by key elements of current social change in the Global South (e.g. increasing formal education, market engagement and wealth accumulation). Overall, our results suggest that fair environmental governance requires explicit identification of distributional fairness conceptions of those most affected by such initiatives, especially in a context of increasing globalisation of conservation knowledge and practice.

Resilience of food systems is key to ensuring food security through crisis. The COVID-19 pandemic presents an unprecedented shock that reveals varying levels of resilience of increasingly interconnected food systems across the globe. We contribute to the ongoing debate about whether increased connectivity reduces or enhances resilience in the context of rural Pacific food systems, while examining how communities have adapted to the global shocks associated with the pandemic to ensure food security. We conducted 609 interviews across 199 coastal villages from May to October 2020 in Federated States of Micronesia, Fiji, Palau, Papua New Guinea, Solomon Islands, Tonga, and Tuvalu to understand community-level impacts and adaptations during the first 5-10 months of the COVID-19 crisis. We found that local food production practices and food sharing conferred resilience, and that imported foods could aid or inhibit resilience. Communities in countries more reliant on imports were almost twice as likely to report food insecurity compared to those least reliant. However, in places dealing with a concurrent cyclone, local food systems were impaired, and imported foods proved critical. Our findings suggest that policy in the Pacific should bolster sustainable local food production and practices. Pacific states should avoid becoming overly reliant on food imports, while having measures in place to support food security after disasters, supplementing locally produced and preserved foods with imported foods when necessary. Developing policies that promote resilient food systems can help prepare communities for future shocks, including those anticipated with climate change.

Locally managed marine areas (LMMAs) are often recommended as a strategy to achieve conservation and fisheries management, though few studies have evaluated their performance against these objectives. We assessed the effectiveness of eight periodically harvested closures (PHCs), the most common form of management within Fijian LMMAs, focusing on two outcomes: protection of resource units and biodiversity conservation. Of the eight PHCs, only one provided biodiversity benefits, whereas three were moderately successful in protecting resource units (targeted fish biomass). Protection of resource units was more likely when PHCs were harvested less frequently, less recently, and when total fish biomass in open areas was lower. Our findings further suggest that monitoring, enforcement, and clearly defined boundaries are critical, less frequent harvesting regimes are advised, and culturally appropriate management incentives are needed. Although PHCs have some potential to protect resource units, they are not recommended as a single strategy for broad-scale biodiversity conservation.

Worldwide, small-scale fisheries (SSF) are an important source of food and livelihoods for rural communities and contribute substantially to national economies. Women play crucial roles in these fisheries, yet their contributions are largely invisible, often ignored and unrecognized. We conducted household and focus group surveys to examine the role of indigenous Fijian (iTaukei) women in SSF, documenting fishing practices and contributions to household food security and income. Our results reinforced several traditional views, such as iTaukei women preferentially fishing closer to their villages; but also challenged other assumptions with women fishing a wider range of habitats (from inland rivers to the open ocean) and species than previously described, and many using a boat and fishing with men. In addition to gleaning for invertebrates and seaweed, women also caught over 100 species of fish. Women fished primarily for subsistence, emphasizing their significant contribution to household food security. Although almost half of the women sold part of their catch to supplement household incomes, they also engaged in other income earning livelihoods, and therefore were not solely dependent on fisheries. Of concern was the high targeting of nursery areas for fish and invertebrate species by women fishers, and species with low spawning potential ratios. Given the level of engagement in, and contributions to fisheries, the inclusion of iTaukei women fishers in fisheries planning and management is critical for ensuring the sustainability of SSF in Fiji. Furthermore, empowering women for full participation in fisheries and lifting them out of poverty requires a re-consideration of traditional gender norms in rural communities, which are already shifting and evolving.

Multi-scale social-ecological systems (SES) approaches to conservation and commons management are needed to address the complex challenges of the Anthropocene. Although SES approaches to monitoring and evaluation are advocated in global science and policy arenas, real-world applications remain scarce. Here, we describe the first operationalization and implementation of Ostrom’s influential SES framework for monitoring practice across multiple countries. Designed to inform management aimed at sustaining coral reefs and the people that depend on them, we developed our SES monitoring framework through a transdisciplinary process involving academics and practitioners with expertise in social and ecological sciences. We describe the SES monitroing framework, including how it operationalizes key insights from the SES and program evaluation literatures, and demonstrate how insights from its implementation in more than 85 communities in four countries (Fiji, Indonesia, Kenya and Madagascar) are informing decision-making at multiple levels. Responding to repeated calls for guidance on applying SES approaches to monitoring and management practice, we outline the key steps of the transdisciplinary development of the framework and lessons learnt. Therefore, our work contributes to bridging the gap between SES science and commons management practice through not only providing an SES monitoring framework that can be readily applied to coral reefs and other commons, but also through demonstrating how to operationalize SES approaches for real-world monitoring and management practice.

Purpose – The purpose of this paper is to underpin a scoping study commissioned by community leaders to assess the potential for creating a “health café” in the centre of Boston, in eastern England, UK, to facilitate healthier lifestyles. Design/methodology/approach – A mixed methods and framework analytic approach was adopted, using documentary, focus group, interview and survey data. The paper drew on social marketing principles to enhance the community relevance of findings. Findings – Community stakeholders and public were generally supportive of a “health café” facility in the town centre. Accessibility and a welcoming environment were seen as key factors. A wide range of health-related services in addition to providing healthy foods were proposed. Key issues identified were: a wider role of the facility as a community “health hub”; appropriate marketing approaches; food provision issues and sustainability. All groups contacted saw the word “health” as off-putting. Research limitations/implications – As with many commissioned scoping studies, the timetable for delivery was very short, just three months, significantly influencing the choices of methodological approaches taken up. This made it important to provide a multi-disciplinary multi-methods design to enhance triangulation and a research team with extensive community research experience including previous research in this region. It was also important to specify and locate any knowledge claims from the findings. Practical implications – The research helped engage community stakeholders to tap a diversity of views which could be adopted by community leaders into their ongoing health strategies and development plans for a “health hub” for Boston. Originality/value – The paper provides important information for those embarking on community health education projects and particularly in how to tailor health research methods to real-world timescales and stakeholder perspectives. Insights are also provided into community attitudes, understandings and behaviours towards healthy living in a part of the UK with a well-documented history of poor health.

Introduction: Food allergy is a recognized health problem, but little has been reported on its cost for health services. The EuroPrevall project was a European study investigating the patterns, prevalence and socio-economic cost of food allergy. Aims: To investigate the health service cost for food-allergic Europeans and the relationship between severity and cost of illness. Methods: Participants recruited through EuroPrevall studies in a case-control study in four countries, and cases only in five countries, completed a validated economics questionnaire. Individuals with possible food allergy were identified by clinical history, and those with food-specific immuno-globulin E were defined as having probable allergy. Data on resource use were used to estimate total health care costs of illness. Mean costs were compared in the case-control cohorts. Regression analysis was conducted on cases from all 9 countries to assess impact of country, severity and age group. Results: Food-allergic individuals had higher health care costs than controls. The mean annual cost of health care was international dollars (I$)2016 for food-allergic adults and I$1089 for controls, a difference of I$927 (95% confidence interval I$324-I$1530). A similar result was found for adults in each country, and for children, and was not sensitive to baseline demographic differences. Cost was significantly related to severity of illness in cases in nine countries. Conclusions: Food allergy is associated with higher health care costs. In this paper we examine the gendered nature of international and internal migration that originates in the central valleys of Oaxaca Mexico. Our goals are to define migration patterns and outcomes for Oaxacan women from the central valleys region and note the differences that mark migrant men and women. We use ethnographic data from anthropological research in 12 of Oaxacas central valley communities to argue that local concepts of what defines correct behaviour (for both men and women) are critical to the outcomes and the differences that exist in the practices of migrant men and women. We have determined that the human SEPW1 (selenoprotein W) gene maps to chromosome 19q13.3, spans approximately 6.3 kb and comprises six exons, in contrast to the previously published five exons. The gene lacks canonical TATA and CAAT boxes, but has numerous Sp1 consensus binding sites upstream of multiple transcription start sites. SEPW1 is expressed in all of the 22 tissues assayed, and shows highest expression in skeletal muscle and heart. Additionally, we have also identified a retroprocessed SEPW1 pseudogene, SEPW1P, which maps to chromosome 1p34-35.

Genes involved in human male sex determination and spermatogenesis are likely to be located on the Y chromosome. In an effort to identify Y-linked, testis-expressed genes, a cDNA selection library was generated by selecting testis cDNA with Y-cosmid clones. Resultant clones containing repetitive or vector material were eliminated, and 79 of the remaining clones were sequenced. Nineteen cDNAs showed homology with the TTY2 gene, and indicated that TTY2 is part of a large gene family. Screening of a panel of Y-linked cosmids revealed that the TTY2 gene family includes at least 26 members organized in 14 subfamilies. Further investigation revealed that TTY2 genes are arranged in tandemly arrayed clusters on both arms of the Y chromosome, and each gene comprises a series of tandemly arranged repeats. RT-PCR studies for two of these genes revealed that they are expressed in adult and fetal testis, as well as in the adult kidney. None of the genes investigated in detail contain an open reading frame. We conclude that the TTY2 gene family is composed of multiple copies, some of which may function as noncoding RNA transcripts and some may be pseudogenes.[The sequence data described in this paper for TTY2L12A and TTY2L2A have been submitted to the EMBL data library, under accession nos.AJ297963 and AJ297964.]

Locus control regions (LCRs) are gene regulatory elements in mammals that can overcome the highly repressive effects normally associated with heterochromatic transgene locations (for example the centromere) in mice1,2,3. Deletion of essential LCR sequences renders the cognate gene susceptible to this form of repression, so a proportion of the cells from transgenic mice that would normally express the transgene are silenced—a phenomenon known as position effect variegation4,5,6 (PEV). We show here that PEV can also occur when the transgene is non-centromeric and that the extent of variegation can be developmentally regulated. Furthermore, by overexpressing a mammalian homologue (M31) of Drosophila melanogaster heterochromatin protein 1 (HP1; refs 7,8) in transgenic mouse lines that exhibit PEV, it is possible to modify the proportion of cells that silence the transgene in a dose-dependent manner. Thus, we show M31 overexpression to have two contrasting effects which are dependent on chromosomal context: (i) it enhanced PEV in those lines with centromeric or pericentromeric transgene locations; and (ii) it suppressed PEV when the transgene was non-centromeric. Our results indicate that components or modifiers of heterochromatin may have a chromosomal-context-dependent role in gene silencing and activation decisions in mammals.

Doyne honeycomb retinal dystrophy (DHRD) is a late-onset autosomal dominant disorder that causes degeneration of the retina and can lead to blindness. We have previously assigned DHRD to a 5-cM region of chromosome 2p16 between marker loci D2S2739 and D2S378. Using sequence-tagged sites (STSs), expressed sequence tags (ESTs) and polymorphic markers within the DHRD region, we have identified 18 yeast artificial chromosomes (YACs) encompassing the DHRD locus, spanning approximately 3 Mb. The YAC contig was constructed by STS content mapping of these YACs and incorporates 13 STSs, including four genes and six polymorphic marker loci. We also report the genetic mapping of two families with a dominant drusen phenotype to the DHRD locus, and genetic refinement of the disease locus to a critical interval flanked by microsatellite marker loci D2S2352 and D2S2251, a distance of approximately 700 kb. These studies exclude a number of candidate genes and provide a resource for construction of a transcriptional map of the region, as a prerequisite to identification of the DHRD disease-causing gene and genes for other diseases mapping in the region, such as Malattia leventinese and Carney complex.

Tbx6 is a member of the T-box family of proteins, which share a region of homology corresponding to the DNA-binding domain of the transcription factor T. Previous expression studies and knockout experiments in mice indicate that Tbx6 is important for specification of paraxial mesoderm structures. We have isolated and characterized the human orthologue, TBX6. Sequence comparisons show that overall the nucleotide homology between human and mouse TBX6/Tbx6 is 84%; within the T-box there is 89% nucleotide homology and 96% amino acid identity. TBX6 maps to chromosome 16 p11.2, a region syntenic with mouse chromosome 7, at 61 cM, the map position of mouse Tbx6. RT-PCR studies of RNA distribution indicate that this gene is expressed not only during gastrulation but has a second phase of expression in some adult tissues including testis. DNA/protein-binding studies demonstrate that Tbx6 binds to the same target DNA as T protein and can form a dimeric complex with DNA. We could find no evidence that Tbx6 forms a heterodimer with T.

Id proteins antagonise the functional properties of DNA-binding, basic helix-loop-helix transcription factors. Id proteins inhibited cell differentiation in various model systems, both in vitro and in vivo. They are transcriptionally and post-transcriptionally regulated during cell cycle progression and promote cell proliferation. In order to establish the molecular and functional properties of Id4, we analysed structure, chromosomal localisation and expression of the murine Id4 gene. Sequence analysis indicated that the Id4 gene consists of three exons. Multiple transcription start sites map about 300 bp upstream of the ATG translational start codon within a 30-bp region of the Id4 promoter, which lacks a classic TATA box. Expression of the Id4 gene results in four major transcripts, most likely generated by differential use of polyadenylation sites. Abundance of the four transcripts varies across tissues, suggesting tissue-specific regulation of polyadenylation and/or post-transcriptional regulation of Id4 expression. However, the Id4 gene seems to be expressed as a single protein. Id4 expression is switched on during embryogenesis between day 7.5 and 9.5 of gestation and is most abundant in adult brain, kidney and testis. Id4 maps to chromosome 13 of the mouse.

Abstract —Voltage-activated Ca 2+ channels exist as multigene families that share common structural features. Different Ca 2+ channels are distinguished by their electrophysiology and pharmacology and can be classified as either low or high voltage–activated channels. Six α1 subunit genes cloned previously code for high voltage–activated Ca 2+ channels; therefore, we have used a database search strategy to identify new Ca 2+ channel genes, possibly including low voltage–activated (T-type) channels. A novel expressed sequence–tagged cDNA clone of α1G was used to screen a cDNA library, and in the present study, we report the cloning of α1H (or Ca v T.2), a low voltage–activated Ca 2+ channel from human heart. Northern blots of human mRNA detected more α1H expression in peripheral tissues, such as kidney and heart, than in brain. We mapped the gene, CACNA1H , to human chromosome 16p13.3 and mouse chromosome 17. Expression of α1H in HEK-293 cells resulted in Ca 2+ channel currents displaying voltage dependence, kinetics, and unitary conductance characteristic of native T-type Ca 2+ channels. The α1H channel is sensitive to mibefradil, a nondihydropyridine Ca 2+ channel blocker, with an IC 50 of 1.4 μmol/L, consistent with the reported potency of mibefradil for T-type Ca 2+ channels. Together with α1G, a rat brain T-type Ca 2+ channel also cloned in our laboratory, these genes define a unique family of Ca 2+ channels.

Testicular germ cell tumors are unusual because they can be cured in over 80% of patients with combination chemotherapy. In order to identify chromosomes carrying genes controlling drug sensitivity, fusions were made between a mouse embryonal carcinoma (EC) cell line, F9, and a human bladder cancer cell line, MGH-UI. In contrast to some previous reports, interspecies hybrids of mouse EC cells with human cells were easy to produce. Six independent hybrids were cloned and grown for 10 further passages and karyotyped. Surprisingly, all the independent hybrids retained approximately 80% of the 40 mouse chromosomes and approximately 80% of the 83 human chromosomes. Despite the positive selection for mouse chromosomes and the absence of selection for human chromosomes, it appears that some of both sets of chromosomes are essential for these hybrids.

The molecular diversity of voltage-activated calcium channels was established by studies showing that channels could be distinguished by their voltage-dependence, deactivation and single-channel conductance. Low-voltage-activated channels are called 'T' type because their currents are both transient (owing to fast inactivation) and tiny (owing to small conductance). T-type channels are thought to be involved in pacemaker activity, low-threshold calcium spikes, neuronal oscillations and resonance, and rebound burst firing. Here we report the identification of a neuronal T-type channel. Our cloning strategy began with an analysis of Genbank sequences defined as sharing homology with calcium channels. We sequenced an expressed sequence tag (EST), then used it to clone a full-length complementary DNA from rat brain. Northern blot analysis indicated that this gene is expressed predominantly in brain, in particular the amygdala, cerebellum and thalamus. We mapped the human gene to chromosome 17q22, and the mouse gene to chromosome 11. Functional expression of the channel was measured in Xenopus oocytes. Based on the channel's distinctive voltage dependence, slow deactivation kinetics, and 7.5-pS single-channel conductance, we conclude that this channel is a low-voltage-activated T-type calcium channel.

P2X3 is one of seven cloned ATP-gated non-selective cation channels. We have isolated a full-length mouse P2X3 gene from a phage lambda-129/Sv genomic library. The gene consists of 12 exons spanning a locus of approximately 40 kb. No significant similarities have been found between the genomic organisation of the mouse P2X3 gene and genes encoding other ion channels. The encoded mouse P2X3 protein consists of 397 amino acids and shows 99% identity with rat P2X3. Using RNase protection and primer extension assays, multiple transcription initiation sites have been mapped in the mouse P2X3 promoter to a region 162-168 bp upstream of the translation initiation codon. The P2X3 gene has been mapped to mouse chromosome 2p by fluorescence in situ hybridisation. The RAG locus-associated gene T160 is located 1.8 kb upstream of the transcription start site of mouse P2X3 gene. The promoter region of the mouse P2X3 gene lacks a conventional TATA and CCAAT consensus sites, and initiator elements. P2X3 is the first member of the P2X gene family to be completely characterised.

Small-diameter sensory neurons associated with unmyelinated axons express a tetrodotoxin-insensitive (TTXi) voltage-gated sodium channel (VGSC) that may play an important role in the transmission of nociceptive information to the spinal cord. A TTXi VGSC, named SNS, that accounts for the tetrodotoxin-resistant sodium current described in sensory neurons has been cloned from rat dorsal root ganglia. Using recombinant λ phage clones encoding a mouse 129/SV genomic library, we have determined the detailed structure of the mouse SNS gene (Scn10a), including the location of exon–intron boundaries and the nucleotide sequence of the exons. The gene consists of 27 exons spanning approximately 90 kb on chromosome 9. Mouse SNS shows 95.3% overall amino acid identity to rat SNS and 98.5% identity throughout the putative transmembrane segments and the intracellular loop linking domains 3 and 4. The sizes of the exons and the exon–intron junction positions of the mouse SNS and the human skeletal muscle VGSC genes are remarkably conserved. These results provide the basis for an evolutionary comparison of sodium channels, the construction and analysis of a mouse SNS null mutant as a direct approach to understanding the biological function of SNS, and the identification of regulatory elements that are responsible for the tissue- and cell-specific expression of SNS.

Phosphoglucomutase 1 (PGM1) deficiency is a stable characteristic of the erythroleukaemic cell line, K562, whereas the activity of the isozymes of the other two PGM loci (PGM2 and PGM3) is slightly elevated. In this study the molecular basis of PGM1 deficiency was investigated by a combined approach utilising protein electrophoresis, immunodetection, cytogenetic techniques, and DNA and RNA analysis. Isoelectric focusing and activity staining confirmed that K562 has no detectable PGM1 activity. Immunoblot analysis of extracts, separated by isoelectric focusing, starch gel and SDS gel electrophoresis, using monospecific anti-PGM1 antibodies showed that K562 contained no detectable immunoreactive material. Karyotype analysis revealed the presence of two intact chromosomes 1 and a derivative chromosome 1, der(1)t(1;11), each of which carried a copy of the PGM1 gene as demonstrated by fluorescence in situ hybridization using a PGM1 cosmid as probe. Southern blot analysis using a PGM1 cDNA clone as probe suggested that the PGM1 genes had not been subject to any gross structural rearrangements. We were also able to determine that K562 is type PGM1 2+1+ by restriction endonuclease analysis of genomic DNA. Very low levels of PGM1 mRNA which appeared to be full length transcripts were detected in K562 using a reverse transcriptase PCR technique. We conclude that the most likely cause of PGM1 enzyme deficiency in K562 is abnormal regulation of transcription.

Cervical tumors nearly all have complex karyotypes and more precise cytogenetic information is required to establish whether specific rearrangements occur, and if they are related to the type of HPV infection found. The karyotypes of five recently established cervical cancer cell lines, three from squamous cell carcinomas (two HPV 16 +ve and one HPV 18 +ve), one from an adenocarcinoma (HPV -ve), and one from an adenosquamous carcinoma (HPV 16 +ve), have been analysed using fluorescence in situ hybridization (FISH), with 23 chromosome specific paints, YACs and cosmids as probes, in addition to conventional G banding, in order to identify markers and clarify the breakpoints. Chromosomes 1 and 3 were rearranged in all cell lines. Breakpoints in the squamous lines were all in 3q. but in different regions. Small metacentrics involving chromosome 5 were a del(5q) in one line, and a t(X;5) in another, rather than i(5p). The region 6q21 was involved in three cases and chromosome 9 was rearranged in four. An i(8q) was found in three squamous carcinoma cell lines. Structural changes of 11q were found only in two cases, but a marker 11 representing amplification in the 11q14-22 region was duplicated in the adenosquamous line.

Cofilin is a widely-distributed, intracellular, actin binding protein which is involved in the translocation of actin-cofilin complex from cytoplasm to nucleus. We have cloned a non-muscle-type cofilin (CFL1) from a human promyelocytic cDNA library and mapped this to human chromosome 11 by PCR amplification of 3' untranslated sequence in a panel of rodent-human somatic cell hybrids, and to the interval 11q12-q13.2 in a chromosome 11 somatic cell hybrid mapping panel. Confirmation of regional localisation to 11q13 has been obtained by fluorescent in situ hybridisation of genomic cosmid clones, by demonstration of the presence of both SEA (the human homologue of avian retrovirus proviral tyrosine kinase, 11q13) and CFL1 in some of these clones and by close linkage of CFL1 to SEA in a panel of high-dose irradiation hybrids. We have identified human muscle-type cofilin sequences by comparison of human expressed sequence tags with M-type cofilins of other species and we have mapped the human M-type cofilin, CFL2, to chromosome 14.

Summary A panel of 22 hybrids was tested for the presence of the gene coding for human cysteine dioxygenase (CDO) by using human specific oligonucleotide primers in the polymerase chain reaction. Detection of human CDO completely correlated with the presence of human chromosome 5. A human total genome cosmid library was screened with a PCR product from the coding region of human CDO cDNA and the two positive clones identified were used in fluorescent in situ hybridisation (FISH) analysis on metaphase chromosome spreads. Fluorescent signals were seen on chromosome 5q22–23. Interspecific backcross mapping in the mouse indicated that Cdo, the mouse homologue of CDO, is situated in the central region of mouse chromosome 18 which shares a region of homology with human chromosome 5.

A detailed karyotype analysis using fluorescence in situ hybridization (FISH), with 24 chromosome-specific paint probes has been carried out on newly established cell lines from two testicular tumors, an i(12p)-positive teratoma, and an i(12p)-negative combined seminoma/teratoma. This has been correlated with loss of heterozygosity (LOH) and allelic imbalance, using DNA RFLP analysis to clarify the genetic changes and to identify any common regions of deletion or rearrangement. With G-banding alone, a total of 11 breakpoints were recognized. After FISH, the position of seven required revision, and 21 new ones were identified. The chromosomes involved most frequently in both tumors were numbers 1, 12, and 18. Breakpoints in 11q and 16q were also seen in both, and seven or more copies of 12p per cell were found in all clones. LOH was found for 18q in both tumors, and overall was much more frequent in underrepresented regions (one or two copies). On the whole, there was good agreement between the cytogenetic and DNA RFLP data; loci showing allelic imbalance generally had an odd number of copies of the chromosome region in which they were known to be located. Combined data on the chromosome 1 translocations in both tumors suggested that rearrangements were more complicated than cytogenetics alone had predicted.

To access a wide a variety of expressed sequence from human chromosome 21 we have placed this chromosome into undifferentiated P19 mouse embryonic carcinoma cells. Cell lines resulting from these experiments have a range of morphologies and a wide variety of karyotypes. We have studied the retinoic acid response of five cell lines, compared to P19 cells, by observing three markers of retinoic acid induced P19 differentiation--cell morphology, RAR alpha and Wnt1 transcription. We see an 'early' retinoic acid response effect, however this response breaks down by the time the 'late' gene. Wnt1 would be transcribed in P19 cells. A highly responsive cell line will be useful for cloning expressed sequences from human chromosome 21 which are produced by early genes in retinoic acid inducible pathways, such as those involved in neurogenesis.

HT-29 cells resistant to 10−6M methotrexate (HT29-MTX) secrete mucins with gastric immunoreactivity (Lesuffleur, T., Barbat, A., Dussaulx, E., and Zweibaum, A.(1990) Cancer Res. 50, 6334-6343). A 3310-base pair mucin cDNA clone (L31) was isolated from an HT29-MTX expression library using a polyclonal serum specific for normal gastric mucosa. It shows a high level of identity (98.6%) to clone NP3a isolated from a nasal polyp cDNA library (Meerzaman, D., Charles, P., Daskal, E., Polymeropoulos, M. H., Martin, B. M., and Rose, M. C.(1994) J. Biol. Chem. 269, 12932-12939). However, as a result of changes in reading frame, the 1042-amino acid deduced peptide contains four regions of a low similarity to the NP3a peptide. The amino acid sequence shows 36.3% similarity to part of the carboxyl-terminal sequence of MUC2 including the so-called D4 domain and 21.3% to the pro von Willebrand factor. A short amino acid sequence is similar to cysteine-rich sequences repeated in tracheobronchial, gastric, and colonic mucin cDNAs. The gene corresponding to L31 is located in the mucin gene cluster on chromosome 11p15.5. The patterns of mRNA expression were indistinguishable from those revealed with the JER58 probe (MUC5AC). Southern blot analysis indicates that the L31 and JER 58 sequences are within 20 kilobase pairs of each other. Together, these results suggest that L31 clone is the 3′ end of MUC5AC.

A panel of 63 radiation-reduced hybrids has been derived from a mouse cell line containing a neo-marked human Chromosome (Chr) 6, primarily to provide a resource for higher resolution localization of new markers. Hybrids were generated with radiation doses of 40-400 Gy, selected in G418, and were shown by PCR to contain the neo gene. PCR was also used to score the retention of 15 loci that map from 6q13 to q25.2 of the current consensus map, plus six other loci assigned to 6q26-q27. An average retention frequency of 27.8% was observed, with the highest frequencies at D6S313 and D6S280 (63.5%) located near the centromere at 6q13, and at D6S283 (68.5%) at 6q16.3-q21, presumably close to the neo integration site. Lowest frequencies (4.8%) were observed for telomeric markers. All markers segregated independently except D6S297 and D6S193. Agreement and some improvement to the current consensus map of 6q was made by mapping 12 loci by the non-parametric statistical method of Falk. In addition, deletion mapping with informative hybrids allowed the ordering of six loci from 6q26 to q27 and permitted some integration of maps of this region.

Fiji has demonstrated a strong commitment to cetacean conservation via national, regional and international plans and agreements. To provide baseline information in support of these efforts, this paper provides an updated listing of cetacean species found in Fijian waters and identifies locations where cetaceans have been noted on a consistent basis. Information for this review was sourced from peer-reviewed publications, field reports, historical whaling records, national consultations, anecdotal and opportunistic sources, as well as a national database held by the Fiji Government’s Department of Fisheries. Reliable and recent records were confirmed for 10 cetacean species in Fijian waters. In addition, less reliable records and regional species’ information provides support for the occurrence of at least 14 additional species or groups of similar-looking species that could not be identified more specifically. Thirteen hotspot areas within the Fiji Economic Exclusive Zone were preliminarily identified as being particularly important for cetaceans, including numerous sites within the Vatu-i-Ra and Lomaiviti passages and surrounding waters. Issues with the available data include uneven coverage, inherent biases within available sources, and difficulties with species identification and verification in some cases. Nevertheless, it is hoped that this review will provide a reference point from which to move forward with cetacean management and conservation efforts in Fiji.

The kinetics and mutagen specificity of reversion of an HGPRT(-)TG(R) line of Chinese hamster cells have been examined in detail by measuring the frequency of HAT(R) colonies. Alkylating agents which produce relatively high levels of O-atom reaction were effective in inducing reversion. MMS, DMS and u.v. were less efficient, and aflatoxin B1, acridine orange and N-acetoxy-AAF were completely ineffective. For agents which were effective, the relationship between HAT(R) colony frequency and dose of mutagen was linear at early expression times (6 h). HAT(R) colony frequency fell subsequently at all doses and the rate and extent of the fall was inversely related to dose. These observations suggest repair of a pro-mutagenic DNA lesion. Other TG(R) mutants isolated from the same wild-type cell line under different selective conditions were also tested for revertibility after exposure to the same mutagens. The majority did not revert, this suggests that they carry deletions within the structural gene for HGPRT. The infrequent revertible lines all arose spontaneously and our evidence suggests that they carry nonsense mutations.

A spontaneous TG(R) HAT(s) V79 mutant had an HGPRT specific activity of less than 5% that of the wild type. Only two spontaneous revertants were recovered from more than 10(8) mutant cells screened. However, HAT resistant clones were recovered, after EMS mutagenesis at a frequency of 4.5 x 10(-6) and at a lower frequency from u.v. irradiated mutant cells. Of more than 400 clones tested, over 90% had regained a HAT(R) TG(s) HGPRT(+) revertant phenotype. Nine of these revertants that were further characterised had HGPRT specific activities within the wild type range and showed wild type electrophoretic mobility. These data suggest that the original HGPRT(-) phenotype was of a mutational rather than an adaptational origin.

This study estimates the effects of food hypersensitivity on individuals' perceived welfare and well-being compared to non-food hypersensitive individuals. Study respondents were recruited in the Netherlands, Poland, Spain and UK. The difference in welfare between food hypersensitive respondents and those asymptomatic to foods was estimated using a subjective welfare approach, including income evaluation. Well-being was measured using the Cantril Ladder-of-Life Scale, and health status using the Self-Perceived Health Scale. The difference in well-being, welfare and health status between participant groups was explained further using a number of background variables. No significant within-country differences in welfare between food hypersensitive respondents and respondents asymptomatic to foods were found. In terms of well-being, adult food hypersensitive respondents and their spouses reported significantly less happiness than respondents and their spouses asymptomatic to foods in the Netherlands and Poland. In Spain, the spouses of the food hypersensitive respondents were significantly less happy than respondents aymptomatic to foods. The well-being of children did not significantly differ between groups. The degree of severity of food hypersensitivity was negatively related to overall health status. In Poland, food hypersensitive respondents reported worse health status compared to asymptomatic respondents. In Spain, the converse was true. Food hypersensitive respondents were generally less happy with their life as a whole than respondents asymptomatic to foods, presumably because they experienced more negative effects, which were not related to perceived health status.

The cadherins are a large family of cell adhesion molecules involved in calcium-dependent recognition and adhesion events. We have used YAC analysis to determine the structure of the human N-cadherin gene. An 850-kb YAC was isolated and the entire N-cadherin gene mapped to a 250-kb region spanning three putative CpG islands. A PCR and cosmid subcloning strategy was used to define the boundaries for the 16 exons that compose the gene. These were shown to be not only highly conserved between mouse and human N-cadherin genes, but also similar to other cadherins. The first and second introns of the gene are large, a property conserved between the mouse and human genes. In situ hybridization with YAC DNA refined the map position of N-cadherin to human chromosome 18q11.2.

A panel of microcell hybrids containing fragments of chromosome 2 was analyzed for the presence of human DPP4, the gene that codes for dipeptidyl peptidase IV (or CD26), by specific PCR amplification of a fragment of the 3' untranslated region of the gene. This analysis placed DPP4 between LCT and GAD in bands q21 to q31. The localization was confirmed by in situ hybridization using two genomic probes that each revealed a hybridization signal in band q24. We also use the recent identification of the ADA binding protein as DPPIV to propose that the gene ADCP2 should be renamed DPP4.

Pancreatitis-associated protein (PAP) is a secretory pancreatic protein present in small amounts in normal pancreas and overexpressed during the acute phase of pancreatitis. In this paper, the authors describe the cloning, characterization, and chromosomal mapping of the human PAP gene. The gene spans 2748 bp and contains six exons interrupted by five introns. The gene has a typical promoter containing the sequences TATAAA and CCAAT 28 and 52 bp upstream of the cap site, respectively. They found striking similarities in genomic organization as well as in the promoter sequences between the human and rat PAP genes. The human PAP gene was mapped to chromosome 2p12 using rodent-human hybrid cells and in situ chromosomal hybridization. This localization coincides with that of the reg/lithostathine gene, which encodes a pancreatic secretory protein structurally related to PAP, suggesting that both genes derived from the same ancestral gene by duplication. 35 refs., 4 figs., 1 tab.

The gene LCT which codes for the intestinal disaccharidase lactase-phlorizin hydrolase has previously been mapped, using somatic cell hybrids and linkage analysis, using the CEPH families, to chromosome 2. We describe here the regional localization of LCT to chromosome 2q21 by polymerase chain reaction (PCR) analysis of somatic cell hybrids and in situ hybridization. LCT is closely linked to D2S44, with a lod score of 30.6 at theta = 0.10.

High-dose irradiation hybrids containing fragments of chromosome 11 have been generated, with a view to isolating new region-specific markers. Forty-seven lines were scored for 34 markers: average retention was 6%. Fourteen lines contain markers from 11q14 to 11q23. One of these, Jo12, has 11q markers extending from tyrosinase (q14-q21) to PBGD (q23.3) plus one marker (TYRL, p11.2) from 11p. In situ hybridization using Alu PCR products from Jo12 as probe confirmed that the human DNA is derived from two regions, one in proximal 11p and a second, larger region in 11q23. Plasmid libraries of Alu PCR products from this and three other hybrids have been made. Six of eight recombinants identified as having single-copy inserts were mapped back to the regions of 11q22-q23 detected in the originating hybrid; only one mapped to a region not originally detected, and one was of hamster origin. These six clones provide new markers in 11q22-q23 that can be used directly for polymorphism studies. This series of hybrids is therefore a valuable resource for the rapid generation of markers from specific, defined regions of chromosome 11.

Summary The pancreatic stone protein (lithostathine) secreted by the exocrine pancreas is an inhibitor of CaCO3 crystal growth. This protein, which is also present in endocrine pancreas, has also been called the regeneration protein (reg). Here we report the mapping of the REG gene to chromosome 2 using the polymerase chain reaction for the specific amplification of human reg sequences in rodentlhuman somatic cell hybrid DNA. A regional assignment has been made by in situ hybridization to metaphase chromosomes using two different fluorescently labelled genomic probes corresponding to the REG gene and a related gene REGL. Both probes hybridized to chromosome 2p12 suggesting the tandem organization of these genes.

The gene MUC3 which codes for a mucin expressed in intestine (Gum et al. 1990) has previously been mapped, using somatic cell hybrids, to chromosome 7. We describe here the regional localization of MUC3 to chromosome 7q22 by in situ hybridization. Preliminary linkage analysis using CEPH (Centre d'Etude du Polymorphisme Humain) families supports this assignment and places MUC3 in the same linkage group as COL1A2 and CFTR.
