A cervical rib is a rare condition in dogs and cats, but is widely described in humans. It is a congenital disorder involving the presence of additional ribs located mainly on one side or, less often, on both sides of the seventh cervical vertebra. Cervical ribs are mostly found accidentally during chest X-rays. Occasionally, however, the presence of a cervical rib can be symptomatic: neurological, cardiovascular, or respiratory symptoms are possible. An 8-year-old Shih-tzu female was presented with a progressive monoplegia of the left thoracic limb. Computed tomography revealed cervical ribs on both sides of the seventh cervical vertebra. The patient underwent surgery treatment and fully recovered.

Objective The aim of the study was to determine the effect of nesfatin-1 on bone properties in female rats in the conditions of developing osteopenia induced by ovariectomy (OVX). Material and methods The experiment was performed on 21 female Wistar rats assigned to 3 groups receiving intraperitoneally physiological saline (SHO, OVX-PhS) and nesfatin-1 in dose 2 μg/kg BW of (OVX-NES) once a day for 8 wks. At the end of the experiment, the rats were scanned using the DXA method to determine the body composition, tBMC, and tBMD. The isolated femora and tibia were tested with the DXA method for BMD and BMC, and with the pQCT method for separate analysis of the cortical and trabecular bone tissue. The bone strength parameters were also determined. The immunohistochemical method was used for determination of nesfatin-1 localization in growth cartilage. Bone metabolism markers (osteocalcin, bALP, and NTx) were identified using an ELISA kit. Results OVX exerts a negative effect on bone tissue. The nesfatin-1 administration influenced positively the DXA parameters of tibia. TvBMD and TbvBMD measured by pQCT in metaphysis of bones were significantly higher in the OVX-NES group than in OVX-PhS. No differences were found in the values of bone strength parameters between SHO and OVX-NES females. Extra- and intracellular immunohistochemical reaction for nesfatin-1 was observed in all zones of growth cartilage, with the strongest reaction detected in the calcifying zone. Nesfatin-1 administration caused a significant increase in the osteocalcin and bALP concentration in relation to the OVX-PhS animals. Conclusions The results of the experiment indicate that nesfatin-1 exerts a protective effect on bone tissue properties and can be used in the prevention of osteoporosis.

The investigations on the response of bone tissue under different loading conditions are important from clinical and engineering points of view. In this paper, the influence of nesfatin-1 administration on rat humerus mechanical properties was analyzed. The classical three-point bending and impact tests were carried out for three rat bone groups: control (SHO), the humerus of animals under the conditions of established osteopenia (OVX), and bones of rats receiving nesfatin-1 after ovariectomy (NES). The experiments proved that the bone strength parameters measured under various mechanical loading conditions increased after the nesfatin-1 administration. The OVX bones were most susceptible to deformation and had the smallest fracture toughness. The SEM images of humerus fracture surface in this group showed that ovariectomized rats had a much looser bone structure compared to the SHO and NES females. Loosening of the bone structure was also confirmed by the densitometric and qualitative EDS analysis, showing a decrease in the OVX bones’ mineral content. The samples of the NES group were characterized by the largest values of maximum force obtained under both quasi-static and impact conditions. The energies absorbed during the impact and the critical energy for fracture (from the three-point bending test) were similar for the SHO and NES groups. Statistically significant differences were observed between the mean Fi max values of all analyzed sample groups. The obtained results suggest that the impact test was more sensitive than the classical quasi-static three-point bending one. Hence, Fi max could be used as a parameter to predict bone fracture toughness.

The study was aimed to ascertain whether continuation or change in the offspring of the diet consumed by the parents modulates, in later life, the previously programmed bone metabolism. We used adult Wistar rats (16 males; 32 females), divided into groups that were fed either a standard (diet S) or a high-energy (diet F). After 90 days of obesity induction, the rats were submitted to obtain female offspring from parents S and F. The offspring stayed with their mothers until 21 days of age (weaning day). Our previous studies have proved the programming effects of parental obesity on the skeletal system of their offspring at the age of 21 days. Weaned female offspring were divided into groups: S/S-parents and offspring fed the S diet; S/F-parents fed the S diet and offspring fed the F diet; F/S-parents fed the diet F and offspring with the diet S; F/F-parents and offspring fed the F diet (F/F). After sacrifice, isolated femurs were assessed by peripheral quantitative computed tomography and by a three-point bending test. The bones were examined at 49 and 90 days of life. We found that nutritional programming has a significant influence on the development and metabolism of the skeletal system in females during growth and maturity. Moreover, the modification of nutrition alters the metabolism of bone tissue, and the osteotropic effects vary depending on the nature of the change, as well as the stage of development. Reducing the caloric content of the diet inhibits the mineralization and decreases the mechanical strength of the bones while increasing the caloric content of the diet has a beneficial osteotropic effect.

Excess fat deposition and insulin resistance are considered the main risk factors for nonalcoholic fatty liver disease (NAFLD), and therefore, not surprisingly, the global prevalence of NAFLD increases in parallel with both obesity and type 2 diabetes. Although deterioration of bone homeostasis in patients with NAFLD is commonly observed, its etiology has not been fully elucidated yet. It was shown in several studies that bone tissue seems to be independently associated with NAFLD. A mechanistic perspective puts the liver at the center of mutual interdependencies obviously involving adipose tissue and muscles and also the bone matrix and bone cells, which are relatively novel. In this review, various pathophysiological mechanisms and possible mediating molecules that may interplay between NAFLD and bone tissue are described. Chronic inflammation, vitamin D3, growth hormone, insulin-like growth factor 1, osteopontin, fetuin-A, irisin, osteocalcin, and osteoprotegerin from osteoblasts have been proposed as mediators of mutual interactions among the skeleton, fatty tissue, and liver. Although to date there are still many issues that have not been elucidated, growing evidence suggests that screening and surveillance of bone mineral density in patients with NAFLD should be considered in future strategies and guidelines for NAFLD management.

The study was aimed at determining the osteotropic effects of diets containing snail meat as a sole protein source. In our experiment, we tested three different diets incorporating snail meat originating from Helix pomatia (HP), Cornu.aspesa maxima (CAM) and Cornu.aspersum aspersum (CAA) and compared these to a control diet (CON) in which casein was the source of protein. In all diets, the protein content amounted to 10%, as calculated on a dry weight basis. In the study, forty male Wistar rats with an initial body mass of 50 ± 2 g were randomly placed within the control and three experimental groups. After 28 days of experimental feeding, the rats were sacrificed, and the body mass, total skeletal density and body composition were recorded. Moreover, blood serum (osteocalcin, CTX) and isolated tibia (pQCT, DXA, 3D micro-CT, 3-point bending test) were stored for further analysis. The results reveal that a diet incorporating snail meat significantly decreased BMC (bone mineral content), as well as area of total skeleton and isolated tibia, and was without influence on BMD (bone mineral density). Furthermore, the 3D micro-CT analysis of trabecular compartment documented a reduced Tb.Th (trabecular thickness), as well as Tb.N (trabecular number), and an increased Tb.Sp (trabecular separation). Beyond the aforementioned, the snail-based diets had an influence upon the architectonical properties of the tibia-decreasing its resistance to mechanical loading. Finally, snail meat, when used as an alone source of protein, negatively influenced the metabolism of the bone tissue in growing animals-making bone smaller and weaker.

This study was undertaken to determine the osteotropic effect of different doses of lipoic acid (LA) on the mineralization of bone tissue in female Wistar rats with experimental osteopenia induced by bilateral ovariectomy. Fifty-six rats were randomly selected and submitted to either a sham operation (n = 8) or an ovariectomy (n = 48). The ovariectomized rats were randomly placed into two control groups, treated subcutaneously with either physiological saline or 17β-estradiol in the dose of 4 μg/kg body mass per day, and four experimental groups that received LA subcutaneously in the doses of 12.5, 25, 50, and 100 mg/kg body mass per day (n = 8 in each group). After 28 days of experimental treatment, the rats were sacrificed, and body mass, total skeletal density, and body composition were recorded. Blood serum and isolated femora were stored for further analysis. Our results revealed that the osteoprotective effect of LA was dose-dependent and was observed in rats treated with 50 and 100 mg/kg of LA. Moreover, the LA applied to the ovariectomized rats in the dose of 50 mg/kg not only stopped the bone resorption, but stimulated its formation.

Introduction: The aim of this study was to investigate the influence of diet supplementation with sodium butyrate and Yucca schidigera extract (0.2% and 0.3%) on femur quality of growing pigs (n = 45). Material and Methods: At the age of 28, 35 and 56 d, five piglets from each group fed a different diet were euthanised and the femora were collected for further analyses. The bone characteristics were assessed based on weight, length, densitometric analysis of BMC and BMD, pQCT analysis (area, mineral content, volumetric density of trabecular and cortical part of metaphysis and diaphysis, respectively), ultimate strength, and geometrical parameters (cross-sectional area and second moment of inertia). Results: There were no significant differences in femur bone parameters among experimental groups on the 28th d of life. On the 35th d of life, piglets with 0.2% supplementation of sodium butyrate and Yucca schidigera extract had significantly lower values of weight and second moment of inertia, and significantly higher trabecular BMD and BMC compared to other experimental groups. In 56-day-old pigs, the higher values were observed in both experimental groups regarding BMC, ultimate strength, geometrical parameters, cortical BMC, diaphyseal total area, and endosteal circumference (P < 0.05). Significant differences between experimental groups were observed only in bone weight and cortical thickness. Conclusion: This study proved that simultaneous supplementation with sodium butyrate and Yucca schidigera extract positively influences bone quality in pigs in the post-weaning period. However, there were no differences in bone characteristics between the addition of 0.2% and 0.3% preparations.

Simple Summary Overweight and obesity can cause many diseases, and several studies indicate a close relationship between the obesity of parents and the health of their offspring. Our aim was to investigate whether there is a programming influence of parental obesity on the skeletal system in weaned female and male offspring rats. In undertaking this, analysis of bone material was carried out using isolated tibia, and densitometry (DXA), peripheral (pQCT) and micro (µCT) computed tomography were performed. Mechanical tests and blood serum biochemistry were also carried out. Our work showed a significant programming influence of parental obesity on neonatal skeletal development. The tibiae isolated from offspring originating from obese parents were characterized by more intense mineralization and higher fracture resistance. However, numerous studies demonstrate the destructive effect of obesity on the skeletal system. Our research and the available literature suggest the existence of a “fat threshold”, the exceeding of which changes of the osteotropic effect of adipose tissue to become unfavorable. Therefore, there is a need for further research to determine the time-dependent metabolic relationship between adipose tissue and bone in both animals and humans. Abstract Our study aimed to verify the hypothesis of the existence of a programming effect of parental obesity on the growth, development and mineralization of the skeletal system in female and male rat offspring on the day of weaning. The study began with the induction of obesity in female and male rats of the parental generation, using a high-energy diet (group F). Females and males of the control group received the standard diet (group S). After 90 days of dietary-induced obesity, the diet in group F was changed into the standard. Rats from groups F and S were mated to obtain offspring which stayed with their mothers until 21 days of age. Tibia was tested using dual-energy X-ray absorptiometry (DXA), peripheral quantitative computed tomography (pQCT), micro-computed tomography (µCT) and mechanical strength using the three-point bending test. Biochemical analysis of blood serum bone metabolism markers was performed. DXA analysis showed higher tibia bone mineral content (BMC) and area. pQCT measurements of cortical and trabecular tissue documented the increase of the volumetric bone mineral density and BMC of both bone compartments in offspring from the F group, while µCT of the trabecular tissue showed an increase in trabecular thickness and a decrease of its separation. Parental obesity, hence, exerts a programming influence on the development of the skeletal system of the offspring on the day of the weaning, which was reflected in the intensification of mineralization and increased bone strength.

In industrial poultry production, especially in broiler chickens, coccidiosis, caused by Eimeria protozoa parasites in the digestive tract, continues to be a serious health and economic problem. In economic terms, the losses are mainly caused by the costs of prevention and treatment, as well as the reduced weight gain and reduced condition of the birds. Currently, the prophylactic program of this disease unit is mainly based on the use of chemotherapeutics. Another way to control coccidiosis is active immunoprophylaxis, based on the vaccination of chickens for fattening, and passive immunoprophylaxis, based on vaccination of the parent flock. An alternative, effective way to reduce the negative effects of protozoan infestation by Eimeria may be the use of herbs and their extracts, pre- and probiotics and betaine.

The withdrawal of antibiotic growth stimulators as of 1 January 2006 in the European Union countries has forced the search for alternative solutions to improve the health and productivity of poultry. The poultry industry also faces the challenge of developing alternative feeding systems with the restriction or exclusion of coccidiostats. One of them is the use of probiotic strains as feed additives. This study aimed to determine the effect of the use in compound feed of a probiotic containing Bacillus licheniformis on rearing rates and postmortem performance of broiler chickens. The experiment was performed on 8012 unsexed broiler chickens of the Ross 308 line divided into four equal groups. The experimental factors were the addition of a probiotic containing Bacillus licheniformis bacteria and the addition of a coccidiostat. The introduction of probiotic bacteria into compound feed did not increase the body weight of birds, in particular, control weights, and at the end of rearing, but it decreased the feed conversion rate (FCR) and mortality. Addition of probiotic in compound feed did not have a significant effect on bird muscle. The mean weight of pectoral muscle between control (C+, C–) and study (C–BL, C+C–BL) groups did not show statistically significant differences; however, the highest mean weight of the evaluated parameter was visible in the C+ group (0. 665) and the lowest in the C–BL group (0. 623). Similarly to the mean weight of the carcass, also in the mean weight of thigh muscles, the lowest statistically significant values were observed in chickens belonging to the C+C–BL group (vs. C–; P

Colostrum (COL) is a valuable discharge of the mammary glands of mammal females, which contains many bioactive components that are indispensable for the normal development of the young organism. Lactoferrin (LF) is one of the colostrum components serving a wide range of functions in the organism, e.g. it is involved in iron metabolism and has anti-inflammatory or immune activity. The experiment was conducted on 36 female Wistar rats in conditions of established osteopenia (60 days after gonadectomy). Females from the experimental groups were administered lactoferrin at the doses of 20 and 40 mg/kg b.w. and colostrum at the doses of 0.5 and 1.0 ml/100g b.w. The results indicate that application of both COL and LF in conditions of established bone tissue atrophy induced by prolonged deficiency of sex steroid hormones has an osteoprotective effect on bone tissue. LF and COL inhibit a decrease in femoral BMC and BMD and improve bone strength parameters at the doses LF=20mg/kg b.w. and COL=5 ml/100 g b.w. However, high doses LF=40 mg/kg b.w. and COL=10 ml/100g b.w. may impede the improvement of bone strength. In femoral cancellous bone tissue, LF reduces the decline in BMC and vBMD, likewise COL, which additionally inhibits atrophy of the cancellous bone tissue. Administration of LF and COL reduced body mass gain and decreased the content of adipose tissue in the OVX rats. The presented results suggest a possibility of therapeutics use of lactoferrin and colostrum for improving the condition of bone tissue.

This study investigated the effect of alpha-ketoglutarate sodium salt (AKG) and ipriflavone (IP) treatment on the mineralization of the tibia in male rats during the development and after the establishment of osteopenia. One hundred and twenty eight male rats were randomly selected and submitted to either sham-operation (SHO) or orchidectomy (ORX), after which each group were then randomly divided between the two experiments. In Experiment-1, treatment with AKG or/and IP started after a 7-day recovery period, whereas in Experiment-2, the experimental protocol proceeded after a 60-day period of osteopenia establishment. AKG was then administered as an experimental drinking, at a concentration of 1.0 mol/l. As a control, a placebo solution was administered. IP at 50 mg/kg b.w., and physiological saline–PhS (as a control for IP) were applied daily via gavage. After 60 days of experimental treatment, in both experiments, the rats were sacrificed, their body weight recorded, while blood serum (Osteocalcin, CTX) and isolated tibia (weight, length, pQCT, DXA, 3-point bending test) were stored for further analysis. Our results show that during the development of osteopenia, AKG and IP when applied exclusively, counteracts osteopenia development, whereas their usage after the establishment of osteopenia, significantly limits the development of bone disorders. Furthermore, combined treatment of AKG and IP exceeded the effects of their sole usage. In addition, during the development of osteopenia, AKG and IP not only inhibited bone resorption, but markedly stimulated the formation of bone tissue. Finally, after the development of osteopenia, combined treatment with AKG and IP protected the bone tissue against orchidectomy-induced bone loss.

2-Oxoglutaric acid (2-Ox), a precursor to hydroxyproline – the most abundant amino acid in bone collagen, exerts protective effects on bone development during different stages of organism development; however, little is known about the action of 2-Ox on cartilage. The aim of the present study was to elucidate the influence of dietary 2-Ox supplementation on the growth plate, articular cartilage and bone of growing rats. A total of twelve male Sprague–Dawley rats were used in the study. Half of the rats received 2-oxoglutarate at a dose of 0·75 g/kg body weight per d in their drinking-water. Body and organ weights were measured. Histomorphometric analyses of the cartilage and bone tissue of the femora and tibiae were conducted, as well as bone densitometry and peripheral quantitative computed tomography (pQCT). The study aimed at the determination of dynamic relationship between mineralisation processes and antioxidative/oxidative status during the development of osteopenia. One hundred and two healthy female Wistar rats at the age of 2 months and initial body weight of 200 g were used in the experiment. The rats were divided into control (CON, n=6), sham operated (SHO, n=48), and ovariectomised (OVX, n=48) groups. Animals from SHO (n=6) and OVX (n=6) groups were sacrificed every week during 8 weeks of the experiment in order to detect dynamic changes in examined parameters. The samples were collected weekly from day 7 to day 56. The femora were examined with the use of DXA (bone mineral density) and pQCT (area, mineral content, volumetric density of trabecular and cortical part of distal femora). The pQCT scans were performed 5 mm from distal end of the tibia. The determination of activity of glutathione peroxidase (GSH-Px) and superoxide dismutase (SOD) in haemolysates of erythrocytes were performed spectrophotometrically. Obtained data showed wave like changes in both enzyme activities and bone parameters and indicated the importance of the 2nd-3rd and 5th-6th week after surgery as a key moment for bone metabolism and activity of enzymatic antioxidative defence during the development of osteopenia induced by bilateral ovariectomy. The obtained results proved that alterations in activity of GSH-Px and SOD, and pQCT ahead the changes registered by DXA by 7 d.

The purpose of the study was to determine the effect of alpha-ketoglutarate sodium salt (AKG) treatment on the mineralization of the tibia in female rats during the development of osteopenia (Experiment-1) and in the condition of established osteopenia (Experiment-2). Thirty-two female rats were ovariectomized (OVX) to induce osteopenia and osteoporosis and another 32 female rats were sham-operated (SHO) and then randomly divided between the two experiments. In Experiment-1, the treatment with AKG started after a 7-day period of convalescence, whereas in Experiment-2 the rats were subjected to a 60-day period of osteopenia fixation, after which the actual experimental protocol commenced. AKG was administered in the experimental solution for drinking at a concentration of 1.0 mol/l and a placebo (PLC) was used as a control solution. After 60 days of experimental treatment the rats in both experiements were sacrificed, the body weight recorded, and blood serum and isolated tibia were stored for further analysis. The bones were analyzed using tomography and densitometry, and for estimation of mechanical properties the 3-point bending test was used. Serum concentrations of osteocalcin and collagen type I crosslinked C-telopeptide were measured. The anabolic effects of AKG on bone during osteopenia development in Experiment-1 not only stopped the degradation of bone tissue, but also stimulated its mineralization. The usage of AKG in animals with established osteopenia (Experiment-2) was not able to prevent bone atrophy, but markedly reduced its intensity. The stimulation of tibia mineralization after AKG treatment has been also argued in healthy SHO animals. The results obtained prove the effectiveness of AKG usage in the prophylaxis and therapy of osteopenia and osteoporosis, induced by bilateral gonadectomy. Additionally, the results clearly prove that treatment with AKG improves the mineralization of bone tissue in healthy animals.

The objective of this study was to evaluate the effect of dietary conjugated linoleic acid (CLA) and false flax (Camelina sativa) seed oil (CS) on bone quality in broiler chickens. Experiment was carried out on 96 chickens randomly divided into four groups: control group fed diet with sunflower oil (SO) and three experimental groups fed diet with addition of CLA (2.86% starter, 4.32% grower) and diet with addition of CLA (1.43% starter, 2.16% grower) and SO or CS (1.43% starter, 2.16% grower). On the 35th d of life eight birds per treatment were slaughtered and the femur, humerus, and tibia were isolated for further analysis. Using DXA method, bone mineral content (BMC) and bone mineral density (BMD) were measured. Furthermore, weight and length of bones were assessed. The weight, BMD, and BMC in chickens from CS+CLA group were significantly higher when compared with CLA group. The significant differences were noted for BMC between control and CS+CLA birds. No marked differences of bone parameters were observed between control group and CLA, and SO+CLA groups. In conclusion, the results indicate that false flax oil and CLA can replace sunflower oil in chickens feeding, and demonstrate the effectiveness of false flax oil on the enhancement of bone properties in broiler chickens.

Bioactive substances including taurine, l -carnitine, polyunsaturated fatty acids, flavonoids and antioxidants were shown to influence the perinatal development of a majority of mammalian systems, nervous, gastrointestinal, immune, etc. A blend of bioactive substances and plants was composed to supplement sow feedstuff, and our study aimed to examine the bone parameters in their offspring. Control sows were fed with a standard diet alone ( n = 6). The sows from the supplemented group ( n = 6) received the supplementation from the gestation day 80 until the end of lactation. From each sow two piglets were sacrificed, one at 7 d and the second at 28 d of life. The femur and humerus were isolated, bone mineral density and mineral content were determined using dual energy X-ray absorptiometry. Bone maximum strength and elastic force were determined by a three-point bending test. No differences in the body weight were found. At day 7, the bones of piglets from the supplemented group were characterized by higher weight, physical parameters and bone mineral content as compared to control, no difference in length and bone mineral density was observed. At day 28, no differences in humerus parameters were found as well as in femur mineral density and mineral content. However, in relation to the body weight, the weight and physical parameters of femur were higher in the supplemented group as compared to control. In conclusion, the supplementation of sow diet with bioactive substances influenced the development of bones in their offspring.

The study was conducted using 120 commercial broiler chicks (Ross 308) randomly allocated to two experimental groups. The experimental diets, differing only in protein source, either solvent-extracted soya bean meal (SBM) or traditional (non-genetically modified) full-fat soya bean seeds (FFS), were prepared using practical corn-based formulation designed to meet nutritional requirements of broilers. Performance parameters were monitored weekly. Also, the subjects were evaluated daily for overt changes in skeletal anatomy and gait physiology. Randomly selected chickens from each group (seven males and seven females) were euthanized at 2, 3, 4 and 6 weeks of age, and bone specimens were collected for further study. The structural quality of the connective tissue is genetically determined and is influenced by hormonal and nutritional modification. An effect of a 2-Ox-rich diet on bone mineralization and structure and expression of non-collagenous protein in articular and growth cartilages of maternal dexamethasone-treated 9-month-old boars was considered in this study. Sows were treated i.m. with dexamethasone at the dose of 0.03 mg kg(-1) body weight every second day during the last 45 days of pregnancy. After the birth, the boars were divided into two groups: administered and not supplemented with 2-Ox for 9 months (0.4 g/kg body weight/day). Dexamethasone given during the prenatal time inhibited the growth and negatively influenced the mechanics, geometry and histomorphometrical parameters of long bones and cartilage irrespective of the diet. Moreover, maternal dexamethasone treatment resulted in expression of osteocalcin in the articular cartilage, and the diet rich in 2-Ox limited the OC expression. This study demonstrated that changes observed in adult boars initiated by dexamethasone treatment in the prenatal period were persistent and long-term use of alimentary 2-Ox supplementation can counteract only some of the destructive changes evoked by prenatal dexamethasone excess.

Their mandibles were isolated and investigated by densitometric (DXA), tomographic (pQCT) and morphometric methods. Moreover, the mechanical parameters (ultimate strength and Young’s modulus) of the mandibles were measured. The results revealed that snail meat as the sole source of protein significantly decreased the bone mineral density (BMD) and content (BMC) of the mandibles. Moreover, the tomographic analysis demonstrated that each type of snail-based diet had a negative influence on the bone cortical and trabecular compartments (which was especially noticeable in the decreasing pQCT parameters). The investigation of mechanical resistance of the mandibles also revealed lower values of the ultimate strength and Young’s modulus in the snail-based diet groups, compared with the casein group. .

A novel elastic hydroxyapatite-based composite of high surgical handiness has been developed. Its potential application in orthopedics as a filler of bone defects has been studied. The biomaterial was composed of carbonated hydroxyapatite (CHAP) granules and polysaccharide polymer (β-1,3-glucan). Cylinders of 4 mm in diameter and 6 mm in length were implanted into bone cavities created in the proximal metaphysis of tibiae of 24 New Zealand white rabbits. 18 sham-operated animals were used as controls. After 1, 3 or 6 months, the rabbits were euthanized, the bones were harvested and subjected to analysis. Radiological images and histological sections revealed integration of implants with bone tissue with no signs of graft rejection. Peripheral quantitative computed tomography (pQCT) indicated the stimulating effect of the biomaterial on bone formation and mineralization. Densitometry (DXA) analysis suggested that biomineralization of bones was preceded by bioresorption and gradual disappearance of porous ceramic granules. The findings suggest that the CHAP–glucan composite material enables regeneration of bone tissue and could serve as a bone defect filler.

The heterotopic pancreas, which is usually described as an untypical presence of pancreatic tissue without any anatomic or vascular continuity with the pancreas, is relatively rare. Clinical manifestations may include bleeding, inflammation, pain and obstruction; however, in most cases it remains silent and is diagnosed during autopsy. Here, we report a case of ectopic pancreatic lesion located in the gastric cardia. The patient was a 73-year-old woman who had a history (over four months) of chronic epigastric pain accompanied by heartburn. Esophagogastroduodenoscopy revealed inflammatory changes throughout the stomach and lower esophagus, as well as a flat polypoid mass with benign features located in the gastric cardia, approx. 10 mm below the “Z” line, measuring approx. 7 mm in diameter. Endoscopic biopsy forceps were used to remove the lesion. Histological examination of the lesion revealed the presence of heterotopic pancreatic tissue in the gastric mucosa. On the basis of the presented case, we suggest that pancreatic ectopia should be a part of differential diagnosis, not only when dealing with submucosal gastric lesions, but also with those that are small, flat and/or untypically located.

Objective: Proton-pump inhibitors, such as omeprazole, are widely used in the prevention and treatment of gastroesophageal diseases. However, an association between proton-pump inhibitors and the increased risk of bone fractures has been observed, especially in patients treated for extended periods. Conversely, 2-oxoglutarate, a precursor of hydroxyproline, the most abundant amino acid in bone collagen, counteracts the bone loss. The aim of the present study was to elucidate the influence of omeprazole on bone and investigate whether dietary 2-oxoglutarate supplementation could prevent the effects of omeprazole. Methods: Eighteen male Sprague-Dawley rats were used. Rats received omeprazole in the diet and 2-oxoglutarate in the drinking water. Body and organ weights and serum concentrations of cholecystokinin and gastrin were measured. The femurs, tibias, and calvarias were collected. Histomorphometric analysis of bone and cartilage tissues was conducted. Bone densitometric and peripheral quantitative computed tomographic analyses of the femur and tibia were performed. Results: Omeprazole decreased the femur and tibia weights, the mechanical properties of the femur, the volumetric bone density and content, the trabecular and cortical bone mineral content, the total, trabecular, and cortical bone areas, the mean cortical thickness, and the periosteal circumference of the femur. Omeprazole had a minor effect on the examined bone morphology and exerted negligible effects on the cartilage. 2-Oxoglutarate lowered the gastrin concentration. Conclusions: Omeprazole treatment exerts its effects mostly on bone mineralization and cancellous bone, adversely affecting bone properties. This adverse effect of omeprazole was not markedly abolished by 2-oxoglutaric acid, which acted as an anti-hypergastrinemic agent. (C) 2013 Elsevier Inc. All rights reserved. (Less)

The aim of the study was to determine the effect of alpha-ketoglutarate on the blood lipid profile using a rat animal model with experimentally induced hypercholesterolaemia.The female and male (30/30) Wistar rats had ad libitum access to a diet containing cholesterol (1 %) and lard (10 %) throughout the entire experimental period (120 days). On day 60 of the study, both the females and the males were divided into three groups, the first receiving a mixture of drinking water adjusted to pH 4.6 using HCl (control), the other two (experimental groups) receiving a solution containing 0.01 M and 0.1 M alpha-ketoglutarate (AKG) (pH adjusted to 4.6). Blood samples were taken on days 0, 30, 60 and 120.The concentrations of total cholesterol, triglycerides, HDL and LDL, respectively, in the blood serum were estimated spectrophotometrically. During the entire experimental period the total cholesterol, triglycerides and LDL levels of the control rats increased, whereas that of HDL decreased. The serum concentrations of total cholesterol, LDL and triglycerides in both the experimental groups receiving AKG decreased (days 60 to 120) (p0.05), while the HDL concentration tended to increase. The body gain in all groups receiving AKG was significantly lower than in the control group.These observations clearly prove that oral treatment with AKG can decrease the risk of hypercholesterolaemia developing and can lower the body weight. The relative concentrations of the plasma LDL and HDL changed to a more favourable ratio promoting good health.

Summary The lack of protective action of oestrogens which appears during menopausal period may be the reason of serious metabolic disturbances including oxidative stress. The hypothesis was stated that ovariectomy may induce the variations of antioxidant/oxidant status which can be detected in rat liver. A total of 102 healthy Wistar female rats were included in the experiment and divided into control (CON; n = 6), sham-operated (SHO; n = 48) and ovariectomized (OVX; n = 48) groups. Animals from SHO (n = 6) and OVX (n = 6) groups were killed every week during 8 weeks of experiment to detect dynamic changes in examined parameters. Anti-oxidative enzyme activities [glutathione peroxidase (GSH-Px); superoxide dismutase (SOD)] as well as total antioxidant capacity (TAC) and the intensity of lipid peroxidation, measured by the concentration of N,N,diethyl-p-phenylene diamine (DEPPD) radical, were determined in liver homogenates by the use of spectrophotometric methods. Wave-like patterns of examined parameters within 8 weeks of experiment were detected. GSH-Px activity tended to be higher in OVX animals and was significantly lower at 8th week when compared with 1st week of experiment. SOD activity was higher in SHO animals and showed significant differences between 3rd, 4th, 7th and 1st week. TAC values were significantly higher in OVX when compared with SHO groups in 2nd, 4th, 5th week and significantly lower in 3rd, 6th and 7th week of the experiment. The concentration of DEPPD radicals tended to increase in OVX group. In conclusion, ovariectomy which leads to oestrogen insufficiency is reflected as well in variation of anti-oxidative/oxidative parameters in rat liver homogenates.

Obesity is a multivariate syndrome which can negatively affect whole body functioning. It is most common in highly developed countries, and in recent years a progressive increase in obesity occurrence is noticeable. The aim of the study was to assess serum lipid profile and metabolic syndrome occurrence among obese rural women from Lublin Region in Eastern Poland. The study was conducted in the Institute of Agricultural Medicine in Lublin (IAM). All subjects had a negative history of diseases and treatment that could affect serum lipid profile or glucose measurements. The inclusion criterion for the study group was overweight and obesity, defined as a body mass index above 25 (BMI25) and living in a rural area. 44 women participated in the study. There were no women fulfilling the criteria or who had a history of incorrect fasting glucose (IFG) or incorrect glucose tolerance (IGT). In contrast, the prevalence of arterial hypertension (or treatment) was high - 53%. 22.7% women had normal serum TC values. The proportion of those with hyper-LDL-C was 38.6% and with hyper-TG - 18.2%. 20.5% of studied women had incorrect serum HDL-C levels, and in 15.9% hypo- HDL-C was accompanied by high serum TC levels. Analysis of correlation showed that serum TC was positively correlated with both LDL- and HDL-C. 55% of the studied obese or overweight women had at least 2 additional components of the metabolic syndrome.
