Plywood made of poplar are limited to indoor usages since poplar exhibits a rather low natural durability. Recently, wood heat treatments have been applied to improve properties such as decay susceptibility and dimensional stability. This study examines the potential of exposing poplar plywood to heat treatment to extend the potential of applications of this engineered wood product to outdoor end uses, and new markets accordingly. Plywood panels were glued with two different adhesive formulations based on the same melamine-urea-formaldehyde (MUF) resin to compare their respective ability to resist to the heat treatment. These different plywoods were thermally modified in saturated steam conditions at 215 °C for 2 hours following the ThermoWood® process, up to reach 14% in mass loss. The durability improvement brought by the heat treatment was assessed in order to evaluate any possible outdoor uses for such plywood. After all the conducted analyses, the potential to use heat treated poplar plywoods in humid interior and protected exterior service conditions was confirmed.

The kinetics of several poplar (Populus alba L.) wood properties during thermal modification conducted in conventional oven with air recirculation were analysed and modelled in this paper. A wide range of properties was assessed, such as: equilibrium moisture content, sorption diagram, shrinkage coefficients, specific shrinkage coefficients, mass loss, modulus of elasticity, strength and colour. The tests were executed at different temperatures ranging from 90 °C to 180 °C and with different durations. The time-temperature equivalency was checked and property modifications over time analysed through master curves in order to obtain a general model connecting together properties, treatment temperature and duration. Different activation energies arising from each property evolution with treatment temperature and duration are provided showing that every modification could occur with different kinetics.

To reduce the size of exit burrs induced by broaching, it is possible to use a low value martyred part clamped to the functional one on the tool exit side. Thence, this research work deals with an in-situ analysis of the burr formation at a bi-specimens interface in orthogonal cutting configuration. New mechanisms of burr formation at the interface formed by Inconel 718 and cast iron specimens are identified. During machining, most of the thermal sources are resulting from the conversion of mechanical energy during the chip shearing mechanism and the intense frictions between the tool, the chip, and the machined surface. Thermal gradients are high and localized, especially for low thermal diffusivity material like titanium alloys. Moreover, heat rates come close to few million degrees per second in the shearing zones. Even though several authors performed thermography techniques to determine the temperature distribution into the machined material, their results remain underestimated due to experimental limitations. In this research, a new temperature measurement technique has been developed based on a single wire thermocouple to measure at one exact location both the temperature and the heating rate during orthogonal cutting test. The tests are performed with uncut chip thicknesses of 0.020 mm and 0.100 mm and with a cutting speed of 120 m/min with two different cutting angles. Cryogenic assistance is used to improve machining performance and to enhance tool life under extreme contact conditions (pressure and temperature). This paper focuses on the influence of cryogenic cooling conditions using liquid nitrogen within the context of machining Ti6Al4 V with CrN-coated carbide cutting tools. Friction behavior of both CrN-coated and uncoated pins with and without cryogenic cooling were evaluated using a home-made laboratory tribometer. The adhesive and plastic friction coefficients were evaluated using a finite element model of the pin-Ti6Al4 V system through an inverse methodology. Finally, the contribution of the CrN coatings was evaluated under both cryogenic cooling and dry conditions. [ABSTRACT FROM AUTHOR]

Amyloid self-assembly of α-synuclein (αSyn) is linked to the pathogenesis of Parkinson's disease (PD). Type 2 diabetes (T2D) has recently emerged as a risk factor for PD. Cross-interactions between their amyloidogenic proteins may act as molecular links. In fact, fibrils of islet amyloid polypeptide (IAPP) (T2D) can cross-seed αSyn amyloidogenesis and αSyn and IAPP colocalize in PD brains. Inhibition of both self- and IAPP-cross-seeded αSyn amyloidogenesis could thus interfere with PD pathogenesis. Here we show that macrocyclic peptides, designed to mimic IAPP self-/cross-interaction sites and previously found to inhibit amyloidogenesis of IAPP and/or Alzheimer's disease (AD) amyloid-β peptide Aβ40(42), are nanomolar inhibitors of both self- and IAPP-cross-seeded amyloid self-assembly of αSyn. Anti-amyloid function is mediated by nanomolar affinity interactions with αSyn via three αSyn regions which are identified as key sites of both αSyn self-assembly and its cross-interactions with IAPP. We also show that the peptides block Aβ42-mediated cross-seeding of αSyn as well. Based on their broad spectrum anti-amyloid function and additional drug-like features, these peptides are leads for multifunctional anti-amyloid drugs in PD, T2D, AD, and their comorbidities, while the identified αSyn key segments are valuable targets for novel, multi-site targeting amyloid inhibitors in PD and related synucleinopathies., (© 2025 The Author(s). Therapies for wound healing using the secretome and extracellular vesicles (EVs) of mesenchymal stem/stromal cells have been shown to be successful in preclinical studies. This study aimed to characterise the protein content of the secretome from stem cells from human exfoliated deciduous teeth (SHED) and analyse the in vitro effects of SHED-conditioned medium (SHED-CM) and SHED extracellular vesicles (SHED-EVs) on keratinocytes. EVs were isolated and characterised. The keratinocyte viability and migration of cells treated with SHED-EVs and conditioned medium (CM) were evaluated. An HaCaT apoptosis model induced by H 2 O 2 in vitro was performed with H 2 O 2 followed by 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide (MTT) and live/dead assays. Finally, the expression of vascular endothelial growth factor (VEGF) in keratinocytes treated with secretome and EVs was evaluated by immunofluorescence staining and confirmed with RT-qPCR. SHED-EVs revealed a cup-shaped morphology with expression of the classical markers for exosomes CD9 and CD63, and a diameter of 181 ± 87 nm. The internalisation of EVs by HaCaT cells was confirmed by fluorescence microscopy. Proteomic analysis identified that SHED-CM is enriched with proteins related to stress response and development, including cytokines (CXCL8, IL-6, CSF1, CCL2) and growth factors (IGF2, MYDGF, PDGF). The results also indicated that 50% CM and 0.4-0.6 μg/mL EVs were similarly efficient for improving keratinocyte viability, migration, and attenuation of H 2 O 2 -induced cytotoxicity. Additionally, expression of VEGF on keratinocytes increased when treated with SHED secretome and EVs. Furthermore, VEGF gene expression in keratinocytes increased significantly when treated with SHED secretome and EVs. In the thermography process, accurately determining emissivity is crucial to obtain precise temperature measurements as it enables the conversion of radiometric values to absolute temperatures. However, assessing emissivity is not a straightforward task as it depends on various other parameters. Traditional methods for measuring emissivity often involve costly materials and cannot be carried out simultaneously with infrared image acquisition. This article presents a method for obtaining pixel-wise emissivity using data from a multispectral infrared camera. Consequently, this method allows for direct emissivity measurement during infrared camera acquisition without the need for additional materials or experiments.

Background: Since the beginning of the pandemic, clinicians and researchers have been searching for alternative tests to improve the screening and diagnosis of the SARS-CoV-2 infection. Currently, the gold standard for virus identification is the nasopharyngeal (NP) swab. Saliva samples, however, offer clear, practical, and logistical advantages but due to a lack of collection, transport, and storage solutions, high-throughput saliva-based laboratory tests are difficult to scale up as a screening or diagnostic tool. With this study, we aimed to validate an intralaboratory molecular detection method for SARS-CoV-2 on saliva samples collected in a new storage saline solution, comparing the results to NP swabs to determine the difference in sensitivity between the two tests., Methods: In this study, 156 patients (cases) and 1005 asymptomatic subjects (controls) were enrolled and tested simultaneously for the detection of the SARS-CoV-2 viral genome by RT-PCR on both NP swab and saliva samples. Saliva samples were collected in a preservative and inhibiting saline solution (Biofarma Srl). Internal method validation was performed to standardize the entire workflow for saliva samples., Results: The identification of SARS-CoV-2 conducted on saliva samples showed a clinical sensitivity of 95.1% and specificity of 97.8% compared to NP swabs. The positive predictive value (PPV) was 81% while the negative predictive value (NPV) was 99.5%. Test concordance was 97.6% (Cohen's Kappa = 0.86; 95% CI 0.81-0.91). The LoD of the test was 5 viral copies for both samples., Conclusions: RT-PCR assays conducted on a stored saliva sample achieved similar performance to those on NP swabs, and this may provide a very effective tool for population screening and diagnosis. Background: Severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) infection has rapidly spread worldwide from the beginning of 2020. Quantitative reverse transcription-PCR (RT-qPCR) is, to this day, the preferred methodology for viral RNA detection, even if not without problems. To overcome some of the limitations still existing for the detection and quantification of nucleic acids in various applications, the use of one-step reverse transcription-droplet digital PCR (RT-ddPCR) has been established. The purpose of this study was, then, to evaluate the efficacy of ddPCR for the detection of SARS-CoV-2 RNA in nasopharyngeal swabs, optimizing the detection of low-viral load-burdened samples., Methods: The RT-ddPCR workflow was validated for sensitivity, specificity, linearity, reproducibility, and precision using samples from 90 COVID-19-infected patients referred to the Department of Laboratory Medicine of the University Hospital of Udine (Italy)., Results: The present study shows that RT-ddPCR allows the detection of as low as 10.3 copies of a SARS-COV-2 E-gene per sample with a higher level of accuracy and precision, especially at low concentration., Conclusion: During the postpeak phase of the SARS-CoV-2 pandemic, it is essential to rely on a highly robust molecular biology method to identify infected subjects, whether they have symptoms or not, in order to prepare appropriate containment measures., Competing Interests: The authors declare no conflicts of interest., (Copyright © 2021 Catia Mio et al.)

Severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) infection has rapidly spread worldwide from the beginning of 2020. The presence of viral RNA in samples by nucleic acid (NA) molecular analysis is the only method available to diagnose COVID-19 disease and to assess patients' viral load. Since the demand for laboratory reagents has increased, there has been a worldwide shortage of RNA extraction kits. We, therefore, developed a fast and cost-effective viral genome isolation method that, combined with quantitative RT-PCR assay, detects SARS-CoV-2 RNA in patient samples. The method relies on the addition of Proteinase K followed by a controlled heat-shock incubation and, then, E gene evaluation by RT-qPCR. It was validated for sensitivity, specificity, linearity, reproducibility, and precision. It detects as low as 10 viral copies/sample, is rapid, and has been characterized in 60 COVID-19-infected patients. Compared to automated extraction methods, our pretreatment guarantees the same positivity rate with the advantage of shortening the time of the analysis and reducing its cost. This is a rapid workflow meant to aid the healthcare system in the rapid identification of infected patients, such as during a pathogen-related outbreak. The set-up of machining parameters for non-ferric materials such as wood and wood-based materials is not yet defined on a scientific basis. In this paper, a new rapid experimental method to assess the specific cutting coefficients when routing isotropic and orthotropic wood-based materials is presented. The method consists of routing, with different depths of cut, a given material previously machined to a round shape after having it fixed on a dynamometric platform able to measure the cutting forces. The execution of subsequent cuts using different depths of cut allows the calculation of the specific cutting coefficients. With the measurement being done during real routing operations, a method to remove machine vibrations was also developed. The specific cutting coefficients were computed for the whole set of grain orientations for orthotropic materials and as an average for isotropic ones. The aim of this paper is to present and validate the whole method by machining selected materials such as Polytetrafluoroethylene-PTFE (isotropic), Medium Density Fiberboard-MDF (isotropic), beech Laminate Veneer Lumber-LVL (orthotropic) and poplar LVL (orthotropic). The method and the proposed analysis have been shown to work very effectively and could be used for optimization and comparison between materials and processes.

Metallic mercury may be self-injected for suicidal or self-harm purposes or sometimes for superstitious or other inadvisable reasons. Local tissue or systemic consequences such as mercurialism can frequently occur in cases of subcutaneous or deep injection, while death due to pulmonary embolism and cardiac, brain, hepatic or renal toxicity may occur in cases of high dosage intravenous administration. The aim of this review is to focus on the diagnostic difficulties facing coroners and forensic pathologists when the courts require confirmation that evidence of self-injection of metallic mercury is the result of suicide or self-harming. Forensic examination performed on the corpses of victims who died in or out of hospital or on surviving injured or intoxicated victims showing signs of mercurialism, demands the careful evaluation of the death scene, of all related circumstances and of the clinical and autopsy data. Rabbit meat breeding may be heavily affected by enterotoxaemia due to Clostridium spiroforme. Data on its antimicrobial susceptibility are insufficient, presumably because of difficulties in cultivating and identifying the pathogen. Our aim is therefore to provide this information to veterinary practitioners by focusing on a panel of therapeutics used in intensive rabbit units. Lincomycin was also checked in order to investigate the origin of resistance to macrolides. Minimal inhibitory concentrations (MICs) were determined with the agar dilution method according to the CLSI M11-A7 protocol (2007). MIC(50) and MIC(90) were, respectively, 64 and 64microg/ml for tiamulin, 32 and 32microg/ml for norfloxacin, 0.063 and 0.125microg/ml for amoxicillin, and 8 and 16microg/ml for doxycycline. MIC(50) and MIC(90) were 256microg/ml for sulphadimethoxine, spiramycin and lincomycin. Our results have shown that intrinsic or acquired antimicrobial resistances are diffuse in the C. spiroforme population and suggest focusing on prevention rather than on treatment of clostridial overgrowth, by reducing risk factors and using antimicrobials prudently.
