Abstract Most embedding media for live and fixed samples were not designed for microscopy and have issues including long polymerization times, peak of toxicity toward the sample during the sol-gel transition, and irreversibility of this transition. Gels derived from biological sources are widely used in microscopy, but their precise composition is ill-defined and can vary between batches. Non-physiological temperatures and/or specific enzymatic solutions are often needed to revert the gel back to the sol state to allow sample recovery. Recovering the sample undamaged is important for multiple purposes, from the ability to release a living organism back into its environment and re-observe it at a later stage, to interrogating the sample once freed from the gel after imaging. We describe a supramolecular hydrogel that enables the observation of small living organisms using light microscopy, with simple sample recovery through vigorous pipetting with water. The organisms can be recovered alive and capable of further development into adulthood, which represents a significant advancement, as most other matrices require release conditions such as heating, the addition of chemicals, or mechanical disruption, which can damage or kill the embedded organisms. Furthermore, the gel is compatible with super-resolution multi-colour STED nanoscopy.

Peroxisomes are essential organelles involved in critical metabolic processes in animals such as fatty acid oxidation, ether phospholipid production and reactive oxygen species detoxification. We have generated transgenic Drosophila melanogaster models expressing fluorescent reporters for the selective autophagy of peroxisomes, a process known as pexophagy. We show that these reporters are colocalized with a peroxisomal marker and that they can reflect pexophagy induction by iron chelation and inhibition by depletion of the core autophagy protein Atg5. Using light sheet microscopy, we have been able to obtain a global overview of pexophagy levels across the entire organism at different stages of development. Tissue-specific control of pexophagy is exemplified by areas of peroxisome abundance but minimal pexophagy, observed in clusters of oenocytes surrounded by epithelial cells where pexophagy is much more evident. Enhancement of pexophagy was achieved by feeding flies with the iron chelator deferiprone, in line with past results using mammalian cells. Specific drivers were used to visualize pexophagy in neurons, and to demonstrate that specific depletion in the larval central nervous system of Hsc70-5, the Drosophila homologue of the chaperone HSPA9/mortalin, led to a substantial elevation in pexophagy.

Understanding how sea turtle species move through the environment and respond to environmental features is fundamental for sustainable ecosystem management and effective conservation. This study investigates the habitat suitability of the loggerhead sea turtle (Caretta caretta) in the Adriatic and Northern Ionian Seas (Central-Eastern Mediterranean) by developing and validating a multidisciplinary framework that leverages machine learning to investigate movement patterns collected by satellite tags Argos satellite tags. Satellite tracking data, enriched with sixteen environmental variables from the Copernicus Marine Service and EMODnet-bathymetry, were analyzed using Random Forest models, obtaining an accuracy of 80.9% when classifying presence versus pseudo-absence of loggerhead sea turtles. As main findings, sea bottom depth, surface chlorophyll (chl-a), and mixed layer depth (MLD) were identified as the most influential features in the habitat suitability of these specimens. Moreover, statistically significant differences, evaluated using t-test statistics, were found between coastal and pelagic locations, for the different seasons, in mixed layer depth, chl-a, 3D-clorophyll, salinity and phosphate. Although based on a limited sample of tagged animals, this study demonstrates that the distribution patterns of loggerhead sea turtles in Mediterranean coastal and pelagic areas are primarily influenced by sea water features linked to productivity and, consequently, to potential prey abundance. Additionally, this multidisciplinary framework presents a replicable approach that can be adapted for various species and regions.

Chlorophyll-a (Chl-a) and Total Suspended Matter (TSM) are key health indicators of the coastal ocean and seas. The former is linked to primary productivity, while the latter is associated with water quality; both are influenced by change in climate. Recent studies have highlighted a declining trend in Chl-a levels along the Mediterranean coastal region. River discharge plays an important role in regulating the coastal Chl-a concentration levels. The present research primarily focuses on understanding the significance of Tiber River −driven spatial dynamics of Chl-a and TSM along the central Tyrrhenian Sea coasts. The research also focuses on evaluating the applicability of Sentinel-2 and identifying a suitable method for estimating Chl-a and TSM from Sentinel-2. Neural networks and dark spectrum fitting techniques were applied using multiple algorithms to estimate the dynamic distribution of Chl-a and TSM driven by the Tiber River in the study area. Multiple statistical analyses were performed, and statistically significant relationships were observed. The Case-2 Regional Coast Colour Neural Network (C2RCC-Net) outperformed all other algorithms, with an R2 value of 0.903 for Chl-a and an R2 value of 0.966 for TSM. Furthermore, the present research also identified a positive pixel to pixel spatial correlation between Chl-a and TSM in all four seasons, highlighting the positive impact of Tiber River on maintaining Chl-a levels along the coasts of Tyrrhenian Sea. This stands in contrast with the negative trend seen in the Mediterranean scale.

Abstract Among marine ecosystems globally, those in the Mediterranean Sea, are facing many threats. New technologies are crucial for enhancing our understanding of marine habitats and ecosystems, which can be complex and resource-intensive to analyse using traditional techniques. We tested, for the first time, an integrated multi-platform approach for mapping the coastal benthic habitat in the Civitavecchia (northern Latium, Italy) coastal area. This approach includes the use of an Unmanned Surface Vehicle (USV), a Remote Operated Vehicle (ROV), and in situ measurements of ecosystem functionality. The echosounder data allowed us to reconstruct the distribution of bottom types, as well as the canopy height and coverage of the seagrass Posidonia oceanica. Our study further involved assessing the respiration (Rd) and net primary production (NCP) rates of P. oceanica and its associated community through in situ benthic chamber incubation. By combining these findings with the results of USV surveys, we were able to develop a preliminary spatial distribution model for P. oceanica primary production (PP-SDM). The P. oceanica PP-SDM was applied between the depths of 8 and 10 m in the studied area and the obtained results showed similarities with other sites in the Mediterranean Sea. Though in the early stages, our results highlight the significance of multi-platform observation data for a thorough exploration of marine ecosystems, emphasizing their utility in forecasting biogeochemical processes in the marine environment.

The behaviour of mountain glaciers on decadal time scales is a useful indicator for assessing climate change. Although less monitored and studied than the ice sheet, local glaciers and ice caps along the coast of Greenland are substantial contributors to meltwater runoff and sea level rise. This study analyses the cumulative area, ice mass and Equilibrium Line Altitude (ELA) change that occurred on 4100 glaciers and ice caps in West Greenland from 1985 to approximately 2020, using remotely sensed data and including glaciers smaller than 1 km2 in the calculations. The glaciers involved in the study decreased in area by 1774 ± 229 km2 which corresponds to almost −15%. Their surface elevation decreased on average by 20.6 ± 3.9 m, corresponding to a rate of −0.5 ± 0.1 m w.e. a−1. The ELA shows a median regional rise of 150 m with marked local variability and higher median rise in the northern part of the study area. Strong regional gradients in ELA of individual glaciers are found, both towards the ice sheet and in areas where local orography affects precipitation. The observed high spatial variability of changes suggests that more monitoring on sub-regional level is needed.

BackgroundYoung people with chronic health conditions and disabilities rely on the healthcare system to maintain their best possible health. The appropriate delivery and utilization of healthcare services are key to improve their autonomy, self-efficacy and employment outcomes. The research question of our study is directed toward investigating if poor availability and accessibility of healthcare services in general, as identified by unmet needs in healthcare, are associated with dissatisfaction with healthcare.MethodsWithin a European multicenter observational study, 357 young adults with cerebral palsy aged 19–28 were included. We assessed special healthcare needs, utilization of healthcare services, and satisfaction with healthcare applying the short-form of the YHC-SUN-SF, environmental and social variables (EAEQ) as well as indicators for severity of condition and functionality (e.g., GMFCS) of these participants based on a self-, assisted self- or proxy-reports. We used correlation analyses to explore associations between satisfaction with healthcare and respective indicators related to availability and accessibility of healthcare services as well as severity of the condition. In addition, we included reference values for satisfaction with heath care from young adults with various chronic conditions assessed within population-based surveys from some of the European countries included in the study.ResultsWe identified several unmet healthcare needs, especially for widely used and established services (e.g., physical therapy). Satisfaction with healthcare (YHC-SUN-SF general and subscale scores) was moderate to high and almost consistently better for the sample of young adults with cerebral palsy as compared to reference values for young adults with various chronic conditions assessed within general population surveys). Correlation coefficients between satisfaction with healthcare and utilization of services and (unmet) healthcare needs were low, also with different indicators for severity of the condition or functionality.ConclusionYoung adults with cerebral palsy reports of unmet healthcare needs varied largely but showed substantial deficits in some aspects. This seems to have no impact on the satisfaction with healthcare those patients currently receive. We conclude that these are two different constructs and somewhat independent indicators to evaluate the quality of healthcare. Clinicians and other practitioners should consider this distinction when monitoring patient needs in their daily practice.

Abstract Background Cancer cachexia is associated with reduced body weight, appetite and quality of life (QOL) with no approved treatments. Growth hormone secretagogues like macimorelin have potential to mitigate these effects. Methods This pilot study assessed the safety and efficacy of macimorelin for 1 week. Efficacy was defined a priori as 1‐week change in body weight (≥0.8 kg), plasma insulin‐like growth factor (IGF)‐1 (≥50 ng/mL) or QOL (≥15%). Secondary outcomes included food intake, appetite, functional performance, energy expenditure and safety laboratory parameters. Patients with cancer cachexia were randomized to 0.5 or 1.0 mg/kg macimorelin or placebo; outcomes were assessed non‐parametrically. Results Participants receiving at least one of either macimorelin dose were combined (N = 10; 100% male; median age = 65.50 ± 2.12) and compared with placebo (N = 5; 80% male; median age = 68.00 ± 6.19). Efficacy criteria achieved: body weight (macimorelin N = 2; placebo N = 0; P = 0.92); IGF‐1 (macimorelin N = 0; placebo N = 0); QOL by Anderson Symptom Assessment Scale (macimorelin N = 4; placebo N = 1; P = 1.00) or Functional Assessment of Chronic Illness Therapy‐Fatigue (FACIT‐F; macimorelin N = 3; placebo N = 0; P = 0.50). No related serious or non‐serious adverse events were reported. In macimorelin recipients, change in FACIT‐F was directly associated with change in body weight (r = 0.92, P = 0.001), IGF‐1 (r = 0.80, P = 0.01), and caloric intake (r = 0.83, P = 0.005), and inversely associated with change in energy expenditure (r = −0.67, P = 0.05). Conclusions Daily oral macimorelin for 1 week was safe and numerically improved body weight and QOL in patients with cancer cachexia compared with placebo. Longer term administration should be evaluated for mitigation of cancer‐induced reductions in body weight, appetite and QOL in larger studies.

Coastal areas are extremely exposed to litter pollution. In this work, we assessed the abundance and distribution of microlitter in the water column and surface waters, as well as its summertime transport trajectories in the northern Latium coastal area (Tyrrhenian Sea, Italy). Microlitter items were classified according to their different shapes (fragments, films, and filaments), sizes, colours, and types by visual sorting and by optical microscopy; their composition was addressed via spectroscopic (Raman and FTIR) microanalysis. Microplastics (MPs) were abundant in all water samples; polypropylene, polyethylene, and poly-methyl methacrylate were identified. The results show a higher concentration of microlitter items in the water column (average concentration of 5.9 ± 1.38 items/m³) compared to surface waters (average concentration of 0.16 ± 0.03 items/m³). These abundances are similar to those found in the Mediterranean Sea and Oceanic waters. The sampling sites closer to the coast showed a higher abundance of particles, especially filaments, in the water column compared to the offshore areas. This feature can be explained based on the presence of land-based pollution sources, bathing areas, and the resuspension of microlitter items from the bottom. Analysis of the surficial trajectories pointed to a general northward transport direction of microlitter and the presence of possible enrichment spots along the coast.

In this paper we describe our work aimed at designing a system able to measure the particulate matter (PM) concentrations via an optical particle counter (OPC) and simultaneously collect it via standard 2.5 cm filters for laboratory characterization. The Port of Civitavecchia (Italy), one of the most important maritime hubs of the Mediterranean Sea, was selected as a test site for an eight-months monitoring campaign. Comparison between the data provided by our device with those from the referenced and certified monitoring stations from the governmental Regional Agency for the Protection of the Environment (ARPA Lazio) allowed to define clear threshold values (PM10 = 25 μg/m3 and PM2.5 = 10 μg/m3). These threshold values need to be considered when correcting the OPC raw data with respect to the humidity (RH) conditions. The sample material was characterized through optical microscopy and SEM-EDS (scanning electron microscopy coupled with energy-dispersive X-ray fluorescence) and spectroscopy (FTIR, Raman), showing a variegate composition from Al-Fe-oxides to silicates, carbonates, and sulfates, to coal and amorphous carbon together with microplastics and textile fibers. As a final test, we analyzed the PM trends provided by our device during the COVID-19 lockdown, when stringent restrictions in the human activities caused well-known fluctuations in the atmospheric pollution. We again observed an evolution of the PM peaks in excellent agreement with the results yielded by the ARPA Lazio monitoring stations. This result provides a valuable confidence test for our devices highlighting the effectiveness of the presented strategy for airborne particulate-matter monitoring.

Remarkable advancements have been made in wave energy converters, notably the innovative WaveSAX technology—an oscillating water column system conceptually designed for coastal structures. Proven effective in existing installations, particularly at Civitavecchia Port, Italy, where the WaveSAX-1 prototype was tested in 2018 and the WaveSAX-2 was updated in 2021. The device’s power generation capacity was evaluated using a Simulating WAves Nearshore (SWAN) model that simulated 30 years of wave conditions. Validation with radar and Acoustic Doppler Profiler instruments showed excellent performance in wave height simulation. Results revealed higher wave power generation near the harbor breakwater, surpassing offshore levels, especially in central and northern sections. WaveSAX-2, featuring an improved Wells turbine, demonstrated a remarkable 37% increase in average efficiency compared to WaveSAX-1. If a linear WaveSAX array were installed, it could produce 4 GWh annually, satisfying 20% of the port’s energy needs and reducing around 2800 tCO2e/year. Doubling arrays and incorporating triangular modules could significantly enhance sustainability for port operations, offering a promising path toward self-sufficiency and environmental responsibility.

Abstract Background Effective inclusion in society for young people with disabilities is increasingly seen as generating opportunities for self-development, and improving well-being. However, significant barriers remain in the vast majority of activities meaningful for young adults. Research argues that various personal (disabilities, health) and environmental (access to the resources needed, accessible environment, discrimination, lack of personal economic independence) factors contribute to limited participation. However, previous studies conducted in young people with cerebral palsy (CP) mainly investigated the transition period to adulthood, and did not fully consider the whole range of impairment severity profiles or environmental barriers. In this study, we will use the follow-up of the SPARCLE cohort and a comparison group from the general population (1) to investigate the impact of the environment on participation and quality of life of young adults with CP, (2) to determine predictors of a successful young adulthood in educational, professional, health and social fields, (3) to compare quality of life and frequency of participation in social, work and recreational activities with the general population, (4) to document on participation and quality of life in those with severe disabilities. Methods The SPARCLE3 study has a combined longitudinal and cross-sectional design. Young adults with CP aged 22 to 27 years in 6 European regions previously enrolled in the SPARCLE cohort or newly recruited will be invited to self-complete a comprehensive set of questionnaires exploring participation (daily life and discretionary activities), health-related quality of life, body function, personal factors (health, personal resources), and contextual factors (availability of needed environmental items, family environment, services provision) during home visits supervised by trained researchers. Proxy-reports or adapted questionnaires will be used for those with the most severe impairments. The recruitment of a large group from the general population (online survey) will enable to identify life areas where the discrepancies between young people with CP and their able-bodied peers are the most significant. Discussion This study will help identify to what extent disabilities and barriers in environment negatively affect participation and quality of life, and how previous valued experiences during childhood or adolescence might modulate these effects.

The orchid genus Phloeophila is distributed from southern Mexico to Brazil and Bolivia, as well as Cuba. A taxonomic revision including the three Phloeophila species present in Ecuador is presented. Morphological characteristics, an identification key, maps of known localities and illustrations of the species are also included. In Ecuador, species of Phloeophila are only known from the Amazonian rainforests, growing from 890 to 1600 meters of altitude. Phloeophila condorana is described as a new species based on specimens collected in the Ecuadorian province of Zamora-Chinchipe and compared to Phloeophila nummularia . Phloeophila nummularia is reported for the first time in Peru. A lectotype for Pleurothallis echinantha is selected.

In permafrost regions, ground surface deformations induced by freezing and thawing threaten the integrity of the built environment. Mapping the frost susceptibility of the ground at a high spatial resolution is of practical importance for the construction and planning sectors. We processed Sentinel-1 Interferometric Synthetic Aperture Radar (InSAR) data from thawing seasons 2015 to 2019, acquired over the area of Ilulissat, West Greenland. We used a least-squares inversion scheme to retrieve the average seasonal displacement (S) and long-term deformation rate (R). We secondly investigated two different methods to extrapolate active layer thickness (ALT) measurements, based on their statistical relationship with remotely sensed surface characteristics. A generalized linear model (GLM) was first implemented, but the model was not able to fit the data and represent the ALT spatial variability over the entire study domain. ALT were alternatively averaged per vegetation class, using a land cover map derived by supervised classification of Sentinel-2 images. We finally estimated the active layer ice content and used it as a proxy to map the frost susceptibility of the ground at the community scale. Fine-grained sedimentary basins in Ilulissat were typically frost susceptible and subject to average seasonal downward displacements of 3 to 8 cm. Areas following a subsiding trend of up to 2.6 cm/yr were likely affected by permafrost degradation and melting of ground ice below the permafrost table. Our approach enabled us to identify frost-susceptible areas subject to severe seasonal deformations, to long-term subsidence induced by degrading permafrost, or to both. Used in combination with traditional site investigations, InSAR maps provide valuable information for risk management and community planning in the Arctic.

Introduction: While most people with cerebral palsy (CP) will have a life expectancy similar to that of the general population, international research has primarily focused on childhood and adolescence; and knowledge about the quality of life (QoL) of young adults with CP, its trajectories, and associated factors remains scarce.Methods: This longitudinal study included young adults with CP living in five European regions and who had previously participated in the SPARCLE cohort as children and/or adolescents. Their QoL in the psychological well-being and social relationships domains was estimated using age-appropriate validated instruments (KIDSCREEN-52 in childhood/adolescence and WHOQOL-Bref in young adulthood). We used generalized linear mixed-effect models with random intercept to estimate long-term trajectories of QoL in both domains and to investigate whether severity of impairment, pain, and seizure influenced these trajectories. We sought to identify potentially different trajectories of QoL from childhood to adulthood using a shape-based clustering method.Results: In total, 164 young adults with CP aged 22–27 years participated in the study. Psychological well-being linearly decreased by 0.78 points (scale 0–100) per year (95% confidence interval (CI) −0.99 to −0.56) from childhood to young adulthood, whereas QoL in the social relationships domain increased (β coefficient 1.24, 95% CI 0.92–1.55). Severity of impairment was associated with reduced QoL in all life periods of the study (childhood, adolescence, and young adulthood): motor impairment with social relationships, and more nuancedly intellectual disability with psychological well-being and social relationships. At all periods, frequent pain significantly reduced psychological well-being, and seizures were associated with lower QoL in the social relationships domain. In both domains, we identified a group of individuals with CP who presented a reverse trajectory compared with the general QoL trajectory.Conclusion: Identification of QoL trajectories and their associated factors yields improved knowledge about the experience of individuals with CP until young adulthood. Further studies are needed to better understand the determinants that have the greatest influence on the different shapes of long-term trajectories of QoL.

The ecological structure of Posidonia oceanica (L.) Delile meadows was evaluated on the northern coast of Lazio, Italy (central Tyrrhenian, Mediterranean sea). This is an infra-littoral zone with a wide range of anthropogenic activities and high geo-morphological variability, which reflects heterogeneity in shoot density, leaf morphology and biomass in fragmented patches. Genetic variability in populations corresponds to the formation of 3 sub-clusters, in the diverse impacted zones (north, centre and south), being correlated to the geographical distance between sites. AMOVA estimated a high genetic variation showing 43.05% individual differences within populations with a marked differentiation among the populations (56.9%) indicated by Fst value (0.57). These results revealed the role of the genetic structure of seagrasses for determining selectivity of fragmented habitat, in response to natural drivers. They showed that site-specific self-recruitment is related to biodiversity capacity and to the geo-morphological characteristic of the coast.

Biological invasions result in negative and unpredictable impacts on oceans worldwide. Non-indigenous macrophytes often synthesize secondary metabolites for defensive purposes and increased competition efficiency: this is the case of Caulerpa cylindracea, which has entered the Mediterranean Sea in 1990 and competed against local flora and fauna since. It was demonstrated that the white seabream Diplodus sargus (i) has included the algae into its diet, (ii) is subject to the peculiar Abnormally Tough Specimen (ATS) condition post-cooking, and (iii) suffers physiological and behavioral disturbances from caulerpin, one of the three major algal secondary metabolites. This paper confirms a feeding relationship between the fish and the algae, quantifies caulerpin accumulation in the liver, suggests a possible mollusk- and echinoderm-driven biomagnification, and highlights the fact that all ATS specimens were males. Multivariate analyses on a multi-biomarker panel reveals differential correlations to key cellular processes such as oxidative stress, metabolism, neurotoxicity, and lipid peroxidation as well as to condition indexes.

Background: While SARS-CoV-2 similarly infects men and women, COVID-19 outcome is less favorable in men. Variability in COVID-19 severity may be explained by differences in the host genome. Methods: We compared poly-amino acids variability from WES data in severely affected COVID-19 patients versus SARS-CoV-2 PCR-positive oligo-asymptomatic subjects. Findings: Shorter polyQ alleles (≤22) in the androgen receptor (AR) conferred protection against severe outcome in COVID-19 in the first tested cohort (both males and females) of 638 Italian subjects. The association between long polyQ alleles (≥23) and severe clinical outcome (p = 0.024) was also validated in an independent cohort of Spanish men

Nos anos 1930 e 1940, o poeta Carlos Drummond de Andrade viveu sua fase de maior engajamento político, aproximando-se do Partido Comunista e incorporando à sua poesia, antes intimista, temas coletivos e sociais. A partir de 1947, no entanto, esta postura começa a esmorecer, levando o escritor a uma isenção ideológica ou, antes disso, a uma clara aversão a seus antigos companheiros de jornada. Este artigo esmiúça o ponto mais tumultuado deste conflito, que foi a atuação de Drummond junto à Associação Brasileira de Escritores, expondo as oscilações do poeta na sua ação política e, simultaneamente, também em sua obra. A trajetória do autor testemunha os limites da comunhão de escritores em torno de causas coletivas, sobretudo nos períodos de maior tensão político-ideológica.

Purpose: Dose information from organ sub-regions has been shown to be more predictive of genitourinary toxicity than whole organ dose volume histogram information. This study aimed to identify anatomically-localized regions where 3D dose is associated with genitourinary toxicities in healthy tissues throughout the pelvic anatomy.Methods and Materials: Dose distributions for up to 656 patients of the Trans-Tasman Radiation Oncology Group 03.04 RADAR trial were deformably registered onto a single exemplar CT dataset. Voxel- based multiple comparison permutation dose difference testing, Cox regression modeling and LASSO feature selection were used to identify regions where 3D dose-increase was associated with late grade ≥ 2 genitourinary dysuria, incontinence and frequency, and late grade ≥ 1 haematuria. This was externally validated by registering dose distributions from the RT01 (up to n = 388) and CHHiP (up to n = 247) trials onto the same exemplar and repeating the voxel-based tests on each of these data sets. All three datasets were then combined, and the tests repeated.Results: Voxel-based Cox regression and multiple comparison permutation dose difference testing revealed regions where increased dose was correlated with genitourinary toxicity. Increased dose in the vicinity of the membranous and spongy urethra was associated with dysuria for all datasets. Haematuria was similarly correlated with increased dose at the membranous and spongy urethra, for the RADAR, CHHiP, and combined datasets. Some evidence was found for the association between incontinence and increased dose at the internal and external urethral sphincter for RADAR and the internal sphincter alone for the combined dataset. Incontinence was also strongly correlated with dose from posterior oblique beams. Patients with fields extending inferiorly and posteriorly to the CTV, adjacent to the membranous and spongy urethra, were found to experience increased frequency.Conclusions: Anatomically-localized dose-toxicity relationships were determined for late genitourinary symptoms in the urethra and urinary sphincters. Low-intermediate doses to the extraprostatic urethra were associated with risk of late dysuria and haematuria, while dose to the urinary sphincters was associated with incontinence.

Realistic predictions of climate change effects on natural resources are central to adaptation policies that try to reduce these impacts. However, most current forecasting approaches do not incorporate species-specific, process-based biological information, which limits their ability to inform actionable strategies. Mechanistic approaches, incorporating quantitative information on functional traits, can potentially predict species- and population-specific responses that result from the cumulative impacts of small-scale processes acting at the organismal level, and can be used to infer population-level dynamics and inform natural resources management. Here we present a proof-of-concept study using the European anchovy as a model species that shows how a trait-based, mechanistic species distribution model can be used to explore the vulnerability of marine species to environmental changes, producing quantitative outputs useful for informing fisheries management. We crossed scenarios of temperature and food to generate quantitative maps of selected mechanistic model outcomes (e.g., Maximum Length and Total Reproductive Output). These results highlight changing patterns of source and sink spawning areas as well as the incidence of reproductive failure. This study demonstrates that model predictions based on functional traits can reduce the degree of uncertainty when forecasting future trends of fish stocks. However, to be effective they must be based on high spatial- and temporal resolution environmental data. Such a sensitive and spatially explicit predictive approach may be used to inform more effective adaptive management strategies of resources in novel climatic conditions.

Abstract Background Little is known about the brain mechanisms underlying cancer‐associated weight loss (C‐WL) in humans despite this condition negatively affecting their quality of life and survival. We tested the hypothesis that patients with C‐WL have abnormal connectivity in homeostatic and hedonic brain pathways together with altered brain activity during food reward. Methods In 12 patients with cancer and 12 healthy controls, resting‐state functional connectivity (RSFC, resting brain activity observed through changes in blood flow in the brain which creates a blood oxygen level‐dependent signal that can be measured using functional magnetic resonance imaging) was used to compare three brain regions hypothesized to play a role in C‐WL: the hypothalamus (homeostatic), the nucleus accumbens (hedonic), and the habenula (an important regulator of reward). In addition, the brain reward response to juice was studied. Participants included 12 patients with histological diagnosis of incurable cancer (solid tumours), a European Cooperative Oncology Group performance status of 0–2, and a ≥5% involuntary body weight loss from pre‐illness over the previous 6 months and 12 non‐cancer controls matched for age, sex, and race. RSFC between the hypothalamus, nucleus accumbens, and habenula and brain striatum activity as measured by functional MRI during juice reward delivery events were the main outcome measures. Results After adjusting for BMI and compared with matched controls, patients with C‐WL were found to have reduced RSFC between the habenula and hypothalamus (P = 0.04) and between the habenula and nucleus accumbens (P = 0.014). Patients with C‐WL also had reduced juice reward responses in the striatum compared with controls. Conclusions In patients with C‐WL, reduced connectivity between both homeostatic and hedonic brain regions and the habenula and reduced juice reward were observed. Further research is needed to establish the relevance of the habenula and striatum in C‐WL.

Overall, the present pilot study provides detailed information on clinical management for Autism Spectrum Disorder (ASD) referral and diagnosis processes that are mandatory for child and adolescent mental health management. The analysis of ASD management, even if carried out on a selected sample of Child and Adolescent Mental Health (CAMH) units, represents a good approximation of how, in Italian outpatient settings, children and adolescents with ASD are recognised and eventually diagnosed. One of the aims of the study was to verify the adherence of Italian CAMH units to international recommendations for ASD referral and diagnosis and whether these processes can be traced using individual chart reports. Overall, the analysis evidenced that Italian CAMH units adopt an acceptable standard for ASD diagnosis, although the reporting of the ASD managing process in the individual chart is not always accurate. Furthermore, data collected suggest some improvements that CAMH units should implement to fill the gap with international recommendations, namely, establishing a multidisciplinary team for diagnosis, improving the assessment of physical and mental conditions by the use of standardised tools, implementing a specific assessment for challenging behaviours that could allow timely and specific planning of intervention.

La estimación de costos es un proceso vital en la planificación de la producción industrial, garantizando una mayor exactitud. Un producto en su ciclo de vida parte de la estimación de costos, logrando con ello establecer los elementos de diseño, viabilidad y márgenes de utilidad. El algoritmo metaheurístico poblacional Optimización de Enjambre de Partículas (PSO) desde su aparición ha demostrado ser eficiente en la solución de problemas de optimización, la variación de sus parámetros ha permitido mejorar su eficiencia. Entre los parámetros de configuración se encuentran los coeficientes de aceleración c1 y c2. El presente trabajo está centrado en realizar un estudio del ajuste paramétrico de c1 y c2 en el desempeño de PSO para resolver un problema de estimación de costos en la fabricación de tuberías. Dado que los diseñadores son propensos a cometer errores y en la búsqueda de modelos que representen al fenómeno estudiado, se ha demostrado que la modelación de problemas no lineales, basada en Redes Neuronales Artificiales (ANNs) provee aproximaciones con bajo margen de error. En consecuencia, en este artículo es mostrada la implementación de una Red Neuronal Artificial (ANN) tipo feedforward sigmoidal con aprendizaje PSO para lograr mejorar la exactitud en la estimación de costos. El estudio de caso utilizado para evaluar el modelo, corresponde a datos provenientes de la producción de tubería para la transferencia de fluidos en la industria minera con información relacionada al peso, tipo de soldadura, diámetro y el correspondiente costo. Los valores evaluados como coeficientes de aceleración estuvieron en el rango 0 a 10, los demás parámetros, en este caso factor inercial y el tamaño de enjambre se trabajaron con valores fijos. La función objetivo utilizada es el Error Cuadrático Medio (MSE), calculado entre los valores observados y los valores estimados por la ANN. A partir de los resultados se puede observar que valores muy pequeños de c1 y c2 obtienen baja exactitud en la estimación de costos de fabricación de tubería, en tanto que la mejor exactitud es lograda por medio de una ANN con aprendizaje PSO, cuyos coeficientes de aceleración son mayores o iguales a 0.5.

Posidonia oceanica (L.) Delile meadows are recognized to be one of the most productive ecosystems of the Mediterranean basin. Due to the impacts of human activities in coastal areas, seagrasses are experiencing a critical decline. In this context, the understanding of the dynamics of production and photosynthesis in response to the environmental factors is essential to address efficient conservation strategies that limit this trend and to assess the ecological status of marine ecosystems. Pulse Amplitude Modulated (PAM) fluorometry has been widely implemented to assess seagrass health and productivity. Here we analyzed the photosynthetic dynamics of P. oceanica according to its bathymetric distribution and daily light availability along a depth gradient to be used as baseline for monitoring purposes on the health status of the seagrass meadows in the Northern Tyrrhenian Sea. Moreover, to investigate the effects of the environmental factors on the health status of P. oceanica within the study area through a multidisciplinary approach, the models contained in the Civitavecchia Coastal Environmental Monitoring System were used. In this study, significant photo-physiological changes have been observed among the investigated meadows. Moreover, the integration of physiological and hydrodynamic information allowed the description of how P. oceanica modulates its photosynthetic capacity at different environmental conditions.

Bacterial pathogens in coastal aquatic ecosystems pose a potential public health hazard for bathing water use. The European Bathing Water Directive (2006/7/EC) currently relies on the culturability of fecal pollution bacterial indicators such as Escherichia coli, without considering dormant or quiescent (Viable But Not Culturable, VBNC) cells, whose possible resuscitation after bathers ingestion cannot be excluded. Standard methods are also time-consuming and therefore hardly meet early warning needs of marine monitoring. To solve this issue, a new index, the Bathing Water Quality Index (BWQI), has here been developed, allowing to identify the most favorable coastal zones for recreational use. The index was calculated by combining numerical simulations of living and dormant E. coli abundances and their residence times. To specifically set up the model with the different physiological states of the whole E. coli population, an ad hoc experiment based on the fluorescent antibody method was performed. The BWQI application to Santa Marinella bathing area highlights a potential risk for human health in the zone most frequented by bathers. This study provides a predictive tool to support preventive decisions of the competent authorities and to properly protect bathers’ health, stressing the need for improved methods for environmental monitoring.

Svalbard archipelago is experiencing the effects of climate changes (i.e., glaciers’ thickness reduction and glacier front retreat), but how ice melting affects water biogeochemistry is still unknown. Microbial communities often act as environmental sentinels, modulating their distribution and activity in response to environmental variability. To assess microbial response to climate warming, within the ARctic: present Climatic change and pAst extreme events (ARCA) project, a survey was carried out along a transect in Konsfjorden from off-shore stations towards the Kronebreen glacier. Total bacterial abundance and the fraction of actively respiring cells (labelled by cyanotetrazolium chloride, CTC), cultivable heterotrophic bacterial abundance, and extracellular enzymatic activities (leucine aminopeptidase (LAP), beta-glucosidase (GLU), and alkaline phosphatase (AP)) were measured. In addition, water temperature, salinity, dissolved oxygen, turbidity, total suspended matter (TSM), particulate and chromophoric dissolved organic matter (CDOM), chlorophyll-a (Chl-a), and inorganic compounds were determined, in order to evaluate whether variations in microbial abundance and metabolism were related with changes in environmental variables. Colder waters at surface (3.5–5 m) depths and increased turbidity, TSM, and inorganic compounds found at some hydrological stations close to the glacier were signals of ice melting. CDOM absorption slope values (275–295 nm) varied from 0.0077 to 0.0109 nm−1, and total bacterial cell count and cultivable heterotrophic bacterial abundance were in the order of 106 cells/mL and 103 colony forming units/mL, respectively. Enzymatic rates p < 0.05); and LAP with Chl-a, Particulate Organic Carbon (POC) and Particulate Organic Nitrogen (PON) (0.97, 0.780, 0.734, p < 0.01), suggesting that fresh material from ice melting stimulated the metabolism of the cultivable fraction.

The Arctic region is greatly affected by climate change, with evident alterations in both physical and biological processes: temperatures are changing at a rate that is twice the global average and phytoplankton productivity is directly affected by ice melting. Continuous monitoring of this ecosystem is fundamental to gain greater understanding of the impact of changes on the natural environment, but the Global Ocean Observing System only provides partial coverage in these extreme areas, which are particularly difficult to reach. Technological progress in oceanographic measurement capabilities is indispensable for the implementation of marine observatories, especially in these remote regions. In recent years, autonomous systems and cost-effective technologies have proved to be valuable for increasing spatial and temporal coverage of data. This is the case with the innovative ArLoC (Arctic Low-Cost) probe, which was designed and developed for easy integration into various types of platforms, enabling continuous measurement of temperature, pressure and fluorescence of chlorophyll a. This work reports on the results of two scientific campaigns carried out in Kongsfjorden (Svalbard Islands) in 2018 in the framework of the UVASS (Unmanned Vehicles for Autonomous Sensing and Sampling) research project. The ArLoC probe was integrated onboard the PROTEUS (Portable RObotic TEchnology for Unmanned Surveys) unmanned semi-submersible vehicle and this allowed us to collect important data in the stretches of sea near tidewater glacier fronts. The acquired data showed several significant effects of glacier melting such as: high temperature and salinity gradients, which cause considerable variations in water mass stratification, and an increase in turbidity and the chlorophyll a concentration, which directly affects primary productivity and the trophic chain. During the surveys, ArLoC proved to be an easy-to-integrate, very reliable instrument, which permitted high spatial resolution investigation of ecological processes during glacier melting as never studied before.

Dissolved organic matter present in natural aquatic environments is a heterogeneous mixture of allochthonous and autochthonous materials. In coastal areas vulnerable to sewage waste, its biologically active component, the chromophoric dissolved organic matter (CDOM), is expected to change its composition and distribution in relation to anthropogenic activities, suggesting the possible use of CDOM as a proxy of fecal contamination. This study aimed at testing such hypothesis by investigating and relating the optical properties of CDOM with Escherichia coli abundance, physiological state, and enzymatic activities in a bathing area of the Northern Tyrrhenian Sea (Latium, Italy) affected by urban wastewaters. The parallel factor analysis (PARAFAC) applied to the excitation–emission matrices (EEMs) of CDOM allowed us to distinguish three main components: C1 (λEx/λEm = 342 nm/435 nm), C2 (λEx/λEm = 281–373 nm/460 nm), and C3 (λEx/λEm = 286 nm/360 nm). C1 and C2 corresponded to humic acids of terrestrial origin, while C3 to tryptophan, whose fluorescence peak was detected close to sewage sites, strongly related to active E. coli cells. The comparison between spectral and microbiological methods is suggested as a suitable approach to monitor bathing water quality for the implementation of coastal observing system capability.

In the present study we used the first rock glacier inventory for the entire French Alps to model spatial permafrost distribution in the region. Climatic and topographic data evaluated at the rock glacier locations were used as predictor variables in a Generalized Linear Model. Model performances are strong, suggesting that, in agreement with several previous studies, this methodology is able to model accurately rock glacier distribution. A methodology to estimate model uncertainties is proposed, revealing that the subjectivity in the interpretation of rock glacier activity and contours may substantially bias the model. The model highlights a North-South trend in the regional pattern of permafrost distribution which is attributed to the climatic influences of the Atlantic and Mediterranean climates. Further analysis suggest that lower amounts of precipitation in the early winter and a thinner snow cover, as typically found in the Mediterranean area, could contribute to the existence of permafrost at higher temperatures compared to the Northern Alps. A comparison with the Alpine Permafrost Index Map (APIM) shows no major differences with our model, highlighting the very good predictive power of the APIM despite its tendency to slightly overestimate permafrost extension with respect to our database. The use of rock glaciers as indicators of permafrost existence despite their time response to climate change is discussed and an interpretation key is proposed in order to ensure the proper use of the model for research as well as for operational purposes.

The development of low-cost instrumentation plays a key role in marine environmental studies and represents one of the most innovative aspects of current oceanographic research. These kinds of devices can be used for several applications, ranging from vertical profilers to stand-alone systems, and can be installed on different platforms (buoys, Voluntary Observing Ships, underwater vehicles, etc.). The availability of low-cost technologies enables the realization of extended observatory networks for the study of marine physical and biological processes through an integrated approach merging in situ observations, forecasting models and remotely sensed data. We present new low-cost sensors and probes developed to measure marine temperature, conductivity, chlorophyll a and Chromophoric Dissolved Organic Matter fluorescence, focusing on sensing strategies, general architecture, laboratory trials, in situ tests and comparison with standard instruments. Furthermore, we report the expendable (New T-FLaP), vertical profiler (T-FLaPpro) and stand-alone (Spectra) applications of these technological developments that were tested during several oceanographic surveys in the Mediterranean Sea.

The spatial distribution of sea bed covers and seagrass in coastal waters is of key importance in monitoring and managing Mediterranean shallow water environments often subject to both increasing anthropogenic impacts and climate change effects. In this context we present a methodology for effective monitoring and mapping of Posidonia oceanica (PO) meadows in turbid waters using remote sensing techniques tested by means of LAI (Leaf Area Index) point sea truth measurements. Preliminary results using Daedalus airborne sensor are reported referring to the PO meadows at Civitavecchia site (central Tyrrhenian sea) where vessel traffic due to presence of important harbors and huge power plant represent strong impact factors. This coastal area, 100 km far from Rome (Central Italy), is characterized also by significant hydrodynamic variations and other anthropogenic factors that affect the health of seagrass meadows with frequent turbidity and suspended sediments in the water column. During 2011–2012 years point measurements of several parameters related to PO meadows phenology were acquired on various stations distributed along 20 km of coast between the Civitavecchia and S. Marinella sites. The Daedalus airborne sensor multispectral data were preprocessed with the support of satellite (MERIS) derived water quality parameters to obtain here improved thematic maps of the local PO distribution. Their thematic accuracy was then evaluated as agreement (R2) with the point sea truth measurements and regressive modeling using an on purpose developd method.

The Arctic region is known to be severely affected by climate change, with evident alterations in both physical and biological processes. Monitoring the Arctic Ocean ecosystem is key to understanding the impact of natural and human-induced change on the environment. Large data sets are required to monitor the Arctic marine ecosystem and validate high-resolution satellite observations (e.g., Sentinel), which are necessary to feed climatic and biogeochemical forecasting models. However, the Global Observing System needs to complete its geographic coverage, particularly for the harsh, extreme environment of the Arctic Region. In this scenario, autonomous systems are proving to be valuable tools for increasing the resolution of existing data. To this end, a low-cost, miniaturized and flexible probe, ArLoC (Arctic Low-Cost probe), was designed, built and installed on an innovative unmanned marine vehicle, the PROTEUS (Portable RObotic TEchnology for Unmanned Surveys), during a preliminary scientific campaign in the Svalbard Archipelago within the UVASS project. This study outlines the instrumentation used and its design features, its preliminary integration on PROTEUS and its test results.

The Gulf of Gaeta, in the western margin of central Italy, is characterized by a coastal morphology that creates a natural sheltered area in which fine sediment settles. The new port regulatory plan provides for dock expansions and dredging works that could alter the suspended particulate matter (SPM) concentration. The present study investigates the dynamics of the Gulf of Gaeta with a focus on the dynamic processes that affect the fine particle concentration. The study was conducted through a multidisciplinary approach that involves remote sensing acquisitions (satellite imagery and X-band radar), measurements in situ (water sampling, wave buoy, weather station, turbidity station, CTD profiles), and numerical modelling (SWAN and Delft3D FLOW). The X-band radar system supports the analysis of the dynamic processes of the SPM concentration providing a large dataset useful for the hydrodynamic model’s validation. The analysis reveals a strong influence of nearby rivers in modulating the SPM at the regional scale. Short-term high and low fluctuations in SPM concentration within the gulf are triggered by the local effect of the main physical forces. In particular, the direction of events and bottom sediment resuspension play a key role in modulating the SPM concentration while micro-tidal regime does not appear to influence turbidity in the study area. This approach represents an important tool in improving the long-term coastal management strategy from the perspective of sustainable human activities in marine coastal ecosystems.

The range of biological outcomes generated by many signalling proteins in development and homeostasis is increased by their interactions with glycosaminoglycans, particularly heparan sulfate (HS). This interaction controls the localization and movement of these signalling proteins, but whether such control depends on the specificity of the interactions is not known. We used five fibroblast growth factors with an N-terminal HaloTag (Halo-FGFs) for fluorescent labelling, with well-characterized and distinct HS-binding properties, and measured their binding and diffusion in pericellular matrix of fixed rat mammary 27 fibroblasts. Halo-FGF1, Halo-FGF2 and Halo-FGF6 bound to HS, whereas Halo-FGF10 also interacted with chondroitin sulfate/dermatan sulfate, and FGF20 did not bind detectably. The distribution of bound FGFs in the pericellular matrix was not homogeneous, and for FGF10 exhibited striking clusters. Fluorescence recovery after photobleaching showed that FGF2 and FGF6 diffused faster, whereas FGF1 diffused more slowly, and FGF10 was immobile. The results demonstrate that the specificity of the interactions of proteins with glycosaminoglycans controls their binding and diffusion. Moreover, cells regulate the spatial distribution of different protein-binding sites in glycosaminoglycans independently of each other, implying that the extracellular matrix has long-range structure.

The repeat acquisition of high-resolution snow depth measurements has important research and civil applications in the Arctic. Currently the surveying methods for capturing the high spatial and temporal variability of the snowpack are expensive, in particular for small areal extents. An alternative methodology based on Unmanned Aerial Systems (UASs) and digital photogrammetry was tested over varying surveying conditions in the Arctic employing two diverse and low-cost UAS-camera combinations (500 and 1700 USD, respectively). Six areas, two in Svalbard and four in Greenland, were mapped covering from 1386 to 38,410 m2. The sites presented diverse snow surface types, underlying topography and light conditions in order to test the method under potentially limiting conditions. The resulting snow depth maps achieved spatial resolutions between 0.06 and 0.09 m. The average difference between UAS-estimated and measured snow depth, checked with conventional snow probing, ranged from 0.015 to 0.16 m. The impact of image pre-processing was explored, improving point cloud density and accuracy for different image qualities and snow/light conditions. Our UAS photogrammetry results are expected to be scalable to larger areal extents. While further validation is needed, with the inclusion of extra validation points, the study showcases the potential of this cost-effective methodology for high-resolution monitoring of snow dynamics in the Arctic and beyond.

Objective . Noninsulinoma pancreatogenous hypoglycemia syndrome (NIPHS) is a disorder of endogenous hyperinsulinemia that is clinically distinguishable from insulinoma, with a greater preponderance after Roux-en-Y gastric bypass (RYBG). Hyperinsulinemic hypoglycemia can predispose to attenuation of counterregulatory hormone responses to hypoglycemia, and consequent suppression of the hypothalamic–pituitary–adrenal (HPA) axis. This case series describes 3 individuals who were diagnosed with adrenal insufficiency (AI) after undergoing RYGB, complicated by NIPHS. Methods . A retrospective chart review was performed for each individual. Chart review applied particular attention to the onset of hyperinsulinemic hypoglycemia following bariatric surgery and the dynamic testing leading to the diagnoses of NIPHS and AI. Results . In each case, reactive hypoglycemia ensued within months to years after RYGB. Cosyntropin stimulation testing confirmed the diagnosis of AI. Hydrocortisone therapy reduced the frequency and severity of hypoglycemia and was continued until successful medical and/or surgical management of hyperinsulinism occurred. Follow-up testing of the HPA axis demonstrated resolution of AI. In all cases, hydrocortisone therapy was finally discontinued without incident. Conclusion . We speculate that transient AI is a potential complication in patients who experience recurrent hyperinsulinemic hypoglycemia after RYGB. The putative mechanism for this observation may be attenuation of the HPA axis after prolonged exposure to severe, recurrent hypoglycemia. We conclude that biochemical screening for AI should be considered in individuals who develop post-RYGB hyperinsulinemic hypoglycemia. If AI is diagnosed, supportive treatment should be maintained until hyperinsulinemic hypoglycemia has been managed effectively.

Quantum dots (QDs) are small nanocrystals widely used for labelling cells in order to enable cell tracking in complex environments in vitro, ex vivo and in vivo. They present many advantages over traditional fluorescent markers as they are resistant to photobleaching and have narrow emission spectra. Although QDs have been used effectively in cell tracking applications, their suitability has been questioned by reports showing they can affect stem cell behaviour and can be transferred to neighbouring cells. Using a variety of cellular and molecular biology techniques, we have investigated the effect of QDs on the proliferation and differentiation potential of two stem cell types: mouse embryonic stem cells and tissue-specific stem cells derived from mouse kidney. We have also tested if QDs released from living or dead cells can be taken up by neighbouring cells, and we have determined if QDs affect the degree of cell-cell fusion; this information is critical in order to assess the suitability of QDs for stem cell tracking. We show here that QDs have no effect on the viability, proliferation or differentiation potential of the two stem cell types. Furthermore, we show that the extent of transfer of QDs to neighbouring cells is 85%), they are rapidly depleted from both stem cell populations. Taken together, our results suggest that QDs are effective cell labelling probes that are suitable for short-term stem cell tracking.

Androgen insensitivity syndrome (AIS) is a rare disease associated with inactivating mutations of AR that disrupt male sexual differentiation, and cause a spectrum of phenotypic abnormalities having as a common denominator loss of reproductive viability. No established treatment exists for these conditions, however there are sporadic reports of patients (or recapitulated mutations in cell lines) that respond to administration of supraphysiologic doses (or pulses) of testosterone or synthetic ligands. Here, we utilize a novel high content analysis (HCA) approach to study AR function at the single cell level in genital skin fibroblasts (GSF). We discuss in detail findings in GSF from three historical patients with AIS, which include identification of novel mechanisms of AR malfunction, and the potential ability to utilize HCA for personalized treatment of patients affected by this condition.

Understanding how androgen receptor (AR) function is modulated by exposure to steroids, growth factors or small molecules can have important mechanistic implications for AR-related disease therapies (e.g., prostate cancer, androgen insensitivity syndrome, AIS), and in the analysis of environmental endocrine disruptors.We report the development of a high throughput (HT) image-based assay that quantifies AR subcellular and subnuclear distribution, and transcriptional reporter gene activity on a cell-by-cell basis. Furthermore, simultaneous analysis of DNA content allowed determination of cell cycle position and permitted the analysis of cell cycle dependent changes in AR function in unsynchronized cell populations. Assay quality for EC50 coefficients of variation were 5-24%, with Z' values reaching 0.91. This was achieved by the selective analysis of cells expressing physiological levels of AR, important because minor over-expression resulted in elevated nuclear speckling and decreased transcriptional reporter gene activity. A small screen of AR-binding ligands, including known agonists, antagonists, and endocrine disruptors, demonstrated that nuclear translocation and nuclear "speckling" were linked with transcriptional output, and specific ligands were noted to differentially affect measurements for wild type versus mutant AR, suggesting differing mechanisms of action. HT imaging of patient-derived AIS mutations demonstrated a proof-of-principle personalized medicine approach to rapidly identify ligands capable of restoring multiple AR functions.HT imaging-based multiplex screening will provide a rapid, systems-level analysis of compounds/RNAi that may differentially affect wild type AR or clinically relevant AR mutations.
