The application of ecological theory to urban planning is becoming more important as land managers focus on increasing urban biodiversity as a way to improve human welfare. City authorities must decide not only what types of biodiversity-focused infrastructure should be prioritized, but also where new resources should be positioned and existing resources protected or enhanced. Careful spatial planning can contribute to the successful return and conservation of urban nature by maximizing the contribution of green infrastructure to landscape connectivity. By using ecological connectivity theory as a planning tool, governments can quantify the effect of different interventions on the ease with which wildlife can move across the landscape. Here we outline an approach to a) quantify ecological connectivity for different urban wildlife species and b) use this to test different urban planning scenarios using QGIS. We demonstrate four extensions to the work by Deslaurier et al. (2018) and Spanowicz & Jaeger (2019) which improve the application of this method as a planning tool for local government:A step-by-step method for calculating effective mesh size using the open-source software QGIS.Conversion of the effective mesh size value (meff) to a “probability of connectedness” (Pc, for easier interpretation by local government and comparisons between planning scenarios).Guidance for measuring species-specific connectivity, including how to decide what spatial information should be included and which types of species might be most responsive to connectivity planning.Advice for using the method to measure the outcome of different urban planning scenarios on ecological connectivity.

Membrane proteins (MPs) are the gatekeepers between different biological compartments separated by lipid bilayers. Being receptors, channels, transporters, or primary pumps, they fulfill a wide variety of cellular functions and their importance is reflected in the increasing number of drugs that target MPs. Functional studies of MPs within a native cellular context, however, is difficult due to the innate complexity of the densely packed membranes. Over the past decades, detergent-based extraction and purification of MPs and their reconstitution into lipid mimetic systems has been a very powerful tool to simplify the experimental system. In this review, we focus on proteoliposomes that have become an indispensable experimental system for enzymes with a vectorial function, including many of the here described energy transducing MPs. We first address long standing questions on the difficulty of successful reconstitution and controlled orientation of MPs into liposomes. A special emphasis is given on coreconstitution of several MPs into the same bilayer. Second, we discuss recent progress in the development of fluorescent dyes that offer sensitive detection with high temporal resolution. Finally, we briefly cover the use of giant unilamellar vesicles for the investigation of complex enzymatic cascades, a very promising experimental tool considering our increasing knowledge of the interplay of different cellular components.

It is increasingly recognised that urban trees can contribute to reducing stormwater runoff by intercepting and retaining a fraction of rainfall received. What is less studied is the translation of this to reduced pollutant loads being transferred to receiving streams, rivers, and water bodies. In this paper, we assess interception of two tree species (Eucalyptus microcorys and Ulmus procera) in an urban park. These data are used in simple water balance modelling to predict the environmental and economic benefit of reducing nitrogen loads to receiving waterways as a function of reduced runoff volume resulting from rainfall interception by urban trees on public land (21% of the catchment area). We use a highly urbanized catchment in Melbourne, Australia to demonstrate the impact of an urban forest dominated by deciduous trees, evergreen trees or a mixed tree canopy cover. We found that doubling the urban canopy cover in the catchment, while keeping the current mix ratio of deciduous and evergreen trees, could reduce annual runoff volume by 30 mm (92 MLyr−1). Using the prescribed values that developers must pay the local water authority for nitrogen treatment as a condition of new development, we calculate that this would deliver a nitrogen load removal benefit of AUD$ 200/tree. If only deciduous trees are planted, the annual runoff reduction would decrease to 24 mm (73 MLyr−1) and increases to 37 mm (112 MLyr−1) if only evergreen trees are planted. This study highlights both the additional benefits of public street trees and the differences in deciduous and evergreen trees which should be accounted for by policy makers.

The original Cerdà plan (Pla Cerdà) of 1855 for the extension of Barcelona is famous for its grid array of large blocks and wide streets to promote circulation. Each block was originally intended to have an area of open space in the center to provide for the needs of residents. Already by the 1920s, however, the center of the blocks had been filled with buildings. Barcelona currently suffers from a chronic lack of open space, excess noise, and air pollution that exceeds European Union law. This article aims to trace the agency of play as a dynamic process in urban planning to provide new sources of open space through Superblocks (aka Superillas and Supermanzanas). We begin with a brief history of their implementation in the city of Barcelona and the evolution of their justification from the 1990s onward. We then explain the capacity for urban transformation that Superblocks have and how they can modify the urban mobility model and the public space model. We then analyze the role of different forms of play that are used in the area of Poblenou to suggest how play can be used to increase the legitimacy of public participation. Finally, we observe the contingent role of play within urban socialization and in motivating and shaping the urban planning processes of implementing Superblocks.

Guiding urban planners on the cooling returns of different configurations of urban vegetation is important to protect urban dwellers from adverse heat impacts. To this end, we estimated statistical models that fused multi-temporal very fine spatial (20 cm) and vertical (1 mm) resolution imagery, that captures the complexity of urban vegetation, with remotely sensed temperature data to assess how urban vegetation configuration influences urban temperatures. Perth, Western Australia, was used as a case-study for this analysis. Panel regression models showed that within a location an increase in tree and shrub cover has a larger cooling effect than grass coverage. On average, holding all else equal, an approximate 1 km2 increase in shrub (tree) cover within a location reduces surface temperatures by 12 °C (5 °C). We included a range of robustness checks for the observed relationships between urban vegetation type and temperature. Geographically weighted regression models showed spatial variation in the cooling effect of different vegetation types; this indicates that i) unobserved factors moderate temperature-vegetation relationships across urban landscapes, and ii) that urban vegetation type and temperature relationships are complex. Machine learning models (Random Forests) were used to further explore complex and non-linear relationships between different urban vegetation configurations and temperature. The Random Forests showed that vegetation type explained 31.84% of the out-of-bag variance in summer surface temperatures, that increased cover of large vegetation within a location increases cooling, and that different configurations of urban vegetation structure can lead to cooling gains. The models in this study were trained with vegetation data capturing local detail, multiple time-periods, and entire city coverage. Thus, these models illustrate the potential to develop locally-detailed and spatially explicit tools to guide planning of vegetation configuration to optimise cooling at local- and city-scales.

This paper presents the outcomes from a joint research project that aims to develop a smartphone application/online platform to model the most thermally comfortable active transport route to a planned destination using heat information and tree shading (Shadeway). Here, we provide a summary of our systematic review of academic literature and applications from the Google Play and Apple App Store, to identify current knowledge about personal adaptation strategies when navigating travel in cities during high temperatures. The review identifies that there is a lack of attention regarding the use of smartphone applications to address urban thermal comfort for active transport by government and private industry. We then present the initial results of original research from three community focus groups and an online survey that elicited participants’ opinions about Shadeways in the City of Greater Bendigo (CoGB), Australia. The results clearly show the need for better management of Shadeways in CoGB. For example, 52.3% of the routes traveled by participants suffer from either no or poor levels of shading, and 53 of the shaded areas were located along routes that also experience heavy traffic, which can have an adverse effect on perceptions and actual safety. It is expected that this study will contribute to improve understanding of the methods used to identify adaptation strategies to increasingly extreme temperatures.

Multiplexed gene-signature-based phenotypic assays are increasingly used for the identification and profiling of small molecule-tool compounds and drugs. Here we introduce a method (provided as R-package) for the quantification of the dose-response potency of a gene-signature as EC50 and IC50 values. Two signaling pathways were used as models to validate our methods: beta-adrenergic agonistic activity on cAMP generation (dedicated dataset generated for this study) and EGFR inhibitory effect on cancer cell viability. In both cases, potencies derived from multi-gene expression data were highly correlated with orthogonal potencies derived from cAMP and cell growth readouts, and superior to potencies derived from single individual genes. Based on our results we propose gene-signature potencies as a novel valid alternative for the quantitative prioritization, optimization and development of novel drugs.

Cities tend to be built in areas of high biodiversity, and the accelerating pace of urbanization threatens the persistence of many species and ecological communities globally. However, urban environments also offer unique prospects for biological conservation, with multiple benefits for humans and other species. We present seven ecological principles to conserve and increase the biodiversity of cities, using metaphors to bridge the gap between the languages of built-environment and conservation professionals. We draw upon John Ruskin's famous essay on the seven lamps of architecture, but more generally on the thinking of built-environment pioneers such as Patrick Geddes (1854–1932) who proposed a synoptic view of the urban environment that included humans and non-humans alike. To explain each principle or ‘lamp’ of urban biodiversity, we use an understanding from the built-environment disciplines as a base and demonstrate through metaphor that planning for the more-than-human does not require a conceptual leap. We conclude our discussion with ten practical strategies for turning on these lamps in cities. Urban planners, architects, landscape architects, engineers and other built-environmental professionals have a key role to play in a paradigm shift to plan for the more-than-human, because of their direct influence on the evolving urban environment. This essay is intended to increase dialogue between ecologists and members of these professions, and thus increase the biodiversity of cities around the world.

A rapid rise of urban population is making cities denser. Consequently, the proportion of impervious surface cover has enlarged, increasing the amount and speed of run‐off reaching urban catchment areas, which may cause flash flooding. Trees play a key role to reduce run‐off in the city, as they intercept rainfall and store part of it on their leaves and branches, reducing the amount and speed of water running onto impervious surfaces. Storage capacity will depend on the rainfall event, the climate conditions and tree characteristics and canopy density. These canopy characteristics vary greatly among different species and their phenology. Furthermore, these canopy characteristics can vary greatly among individual trees of the same age, size, and species. This study tested how canopy density and leaf characteristics of three different tree species affect storage capacity under simulated rainfall conditions. Three species were selected (Ulmus procera, Platanus × acerifolia, and Corymbia maculata), each being of the same height and similar canopy dimensions. Storage capacity was measured using a mass balance approach during a 15‐min indoor, simulated rainfall event (2.5 mm/hr). Canopy metrics were estimated using a terrestrial laser scanner. Canopy surface area was measured through destructive harvest and leaf/twig/branch scanning. To investigate variations in the canopy leaf density, leaves were systematically removed to create four treatments: full, half, quarter, and woody. Canopy storage capacity was well correlated to plant surface area (m2), plant area index (m2/m2), and plant area density (m2/m3). All analyses indicated U. procera as the most efficient species for storing rainfall water within a canopy of equal volume or area index. Results reveal the complexity of evaluating interception of rainfall by tree canopies. While it is well recognised that increasing vegetation cover reduces the Urban Heat Island (UHI) effect in cities, less is understood about the spatial pattern of vegetation type required to maximise cooling benefits. This study examines how different urban vegetation spatial configuration and composition impact on the UHI phenomenon. We investigated this on a set of sites in Metropolitan Melbourne, Australia. Urban vegetation raster data at 20 cm resolution was used to define five height-based vegetation cover types (grass, shrub, small, medium and large trees); and to calculate eight landscape metrics: percentage of landscape (PLAND), mean patch area (AREA_MN), patch density (PD), edge density (ED), mean patch shape index (SHAPE_MN), mean Euclidean nearest-neighbour distance (ENN_MN), Shannon’s diversity index (SHDI) and Landscape shape index (LSI). These vegetation landscape metrics of vegetation types were established as independent variables and statistically analysed with the UHI intensity as the dependent variable using Classification and Regression Tree analysis (CRT). The CRT model was developed based on 6469 records and including depth of 5, 41 nodes and 21 terminal nodes. From the total 43 independent variables, 11 were identified as high impact factors on UHI intensity. Our findings revealed a consistent negative statistical relationship between UHI intensity and PLAND-landscape, PLAND-large tree and PLAND-medium tree across the study sites. The PLAND, ED and AREA_MN were the most prevalent metrics to explain UHI effect, which has also principally demonstrated the joint impact of the three metrics on UHI effect. This study presents a new framework of a fine-scale assessment and modelling for the impact of urban vegetation on UHI; and elaborates a practical approach of using CRT technique to design local-based UHI mitigating strategies taking advantage of different vegetation structure.

In what ways can we advance the community’s understanding of cities and enhance possibilities for creative responses to urban problems? Patrick Geddes’ communication methodologies remain relevant to answering these questions. Two constants across his long career were his propagandizing and pedagogical activities. The bedrock of both were exhibitions. Inspired by the succession of Great Exhibitions from the mid-nineteenth century, Geddes directed his thirst for acquiring and sharing knowledge into a rambling collection of planning materials gathered from all corners of the globe and organised into his own series of exhibitions which prompted viewers to relate the work of other places to their own. Drawing on assemblage thinking, this paper argues that Geddes' exhibitionary work prefigures McFarlane’s notion of “urban learning” across the requisite activities of translation, coordination and dwelling. We describe these changes and the interplay between Geddes' epistemology and his exhibition practices to reflect on how his approach to civic education retains pertinence to the present day.

We undertook the first study systematically evaluating the risk of Anisakis-sensitization in Croatian fish-processing workers and potential genetic susceptibility to anisakiasis. Anti-Anisakis IgE seroprevalence and risk factors for 600 employees of Croatian fish processing facilities and 466 blood donor controls, were assessed by indirect ELISA targeted with: recombinant Ani s 1 and Ani s 7 allergens, an Anisakis crude extract, the commercial ImmunoCAP kit, and questionnaires. Genetic susceptibility to anisakiasis was evaluated by genotypisation of human leukocytes alleles (HLA). Anti-Anisakis seropositive and a fraction of negative subjects were also assessed by ELISA and Western Blot (WB) for IgG seroprevalence to Trichinella spp. Overall, the observed anti-Anisakis seroprevalence inferred by indirect ELISA was significantly higher in fish processing workers (1.8%, 95% CI 0.9–3.3%) compared to the controls (0%, 0–0.8%). Seven out of 11 Ani s 1 and Ani s 7-positives and none of selected 65 negative sera, tested positive on whole-Anisakis extract (ImmunoCAP), whereas Anisakis crude extract ELISA detected 3.9% (2.4–6.0%) seropositives in fish processing workers, three (14%) of which showed IgE reactivity to milk proteins. The highest risk associated with Anisakis-sensitization among workers was fishing in the free time, rather than any of attributes related to the occupational exposure. Although no association was observed between anti-Anisakis seropositivity and wearing gloves or protective goggles, the majority of workers (92%) wore protective gloves, minimizing the risk for Anisakis sensitization via skin contact. Six HLA alleles within DRB1 gene were significantly associated with seropositivity under dominant, allelic or recessive models. All sera confirmed negative for anti-Trichinella spp. IgG. The study exhaustively covered almost all marine fish processing workers in Croatia, reflecting real-time Anisakis sensitization status within the industry, already under the influence of wide array of allergens., Author summary Anisakiasis is a human disease caused by ingestion of live Anisakis spp. larvae by raw seafood. Fish processing and aquaculture workers (approximately 60 million people world-wide) are at occupational risk of becoming sensitized to Anisakis. This is the first study systematically evaluating the risk of Anisakis-sensitization in Croatian fish-processing workers and potential genetic susceptibility to anisakiasis in the analysed population. Observed seroprevalence was significantly higher in fish processing workers (1.8%) compared to the controls (0%). Surprisingly, the highest risk associated with Anisakis-sensitization among workers was fishing in their free time, rather than any of attributes related to the occupational exposure. Additionally, seroprevalence to another important food-borne helminth, Trichinella spp. assessed in all Anisakis-positive subjects showed to be negative. Almost all marine fish processing workers in Croatia were part of this research, reflecting real-time Anisakis-sensitization status within the industry. This sets a baseline for the future tracking of sensitization in the sector already under the influence of wide array of allergens.

Trichinellosis is a zoonotic disease due to the ingestion of raw or undercooked meat from animals infected with the larvae of nematodes belonging to the genus Trichinella. In January–February 2015, an outbreak of trichinellosis occurred in Genoa, Northern Italy. The epidemiological link was traced back to a dinner served at an agritourism farm on 31 December 2014, where a majority of the 52 guests had consumed the ‘beef’ steak tartare. The source of infection was not traced; however, it was noted that the amount of beef purchased officially for providing at the dinner did not correspond with that served, suggesting that meat of a different origin had been added to the beef to prepare the steak tartare. Clinical and laboratory data of 30 individuals out of the 52 (57.7%), of which four were hospitalized, were consistent with that of the case definition of trichinellosis. Western blot patterns of the sera from patients with confirmed trichinellosis were similar to the diagnostic pattern identified for the reference sera of Trichinella pseudospiralis but different from those of the control sera tested for patients infected with Trichinella spiralis and Trichinella britovi. Identification of T. pseudospiralis as the aetiological agent responsible for the outbreak of trichinellosis using an indirect tool represents an advancement in the epidemiological investigation of this zoonotic disease.

As individuals increasingly live in cities, methods to study their everyday movements and the data that can be collected becomes important and valuable. Eye-tracking informatics are known to connect to a range of feelings, health conditions, mental states and actions. But because vision is the result of constant eye-movements, teasing out what is important from what is noise is complex and data intensive. Furthermore, a significant challenge is controlling for what people look at compared to what is presented to them. The following presents a methodology for combining and analyzing eye-tracking on a video of a natural and complex scene with a machine learning technique for analyzing the content of the video. In the protocol we focus on analyzing data from filmed videos, how a video can be best used to record participants' eye-tracking data, and importantly how the content of the video can be analyzed and combined with the eye-tracking data. We present a brief summary of the results and a discussion of the potential of the method for further studies in complex environments.

Most studies of urban forest management look at vegetation on public land. Yet, to meet ambitious urban forest targets, cities must attempt to maintain or increase trees and canopy cover on private urban land too. In this study, we review and evaluate international approaches to protecting and retaining trees on private urban land. Our study combines a systematic academic literature review, two empirical social science studies on the views of urban forest professionals, and a global case study review of innovative regulations and incentives aimed at protecting and retaining trees on private urban land. Case studies were evaluated for the extent they exceeded minimum standards or went beyond ?business-as-usual?. We found that the most innovative mechanisms combine many regulations, instead of relying on a single regulation, and use financial incentives to retain or plant trees in newly developed or re-developed sites, as well as private residences. We did not find any cases where appropriate monitoring was in place to determine the efficacy and efficiency of these mechanisms. We also found no single simple solution that could effectively and efficiently protect and retain trees on private land. Only by combining policies, planning schemes, local laws, and financial incentives with community engagement and stewardship will cities protect and retain trees on private land. Useful and innovative ways to protecting and retaining trees on private land involves providing solutions at multiple governments levels, embedding trees in existing strategic policy and management solutions, incentivising positive behavior, creating regulations that require payment up front, and engaging the broader community in private tree stewardship.

Hot weather not only impacts upon human physical comfort and health, but also impacts the way that people access and experience active travel options such as walking and cycling. By evaluating the street thermal environment of a city alongside an assessment of those communities that are the most vulnerable to the effects of heat, we can prioritise areas in which heat mitigation interventions are most needed. In this paper, we propose a new approach for policy makers to determine where to delegate limited resources for heat mitigation with most effective outcomes for the communities. We use eye-level street panorama images and community profiles to provide a bottom-up, human-centred perspective of the city scale assessment, highlighting the situation of urban tree shade provision throughout the streets in comparison with environmental and social-economic status. The approach leverages multiple sources of spatial data including satellite thermal images, Google street view (GSV) images, land use and demographic census data. A deep learning model was developed to automate the classification of streetscape types and percentages at the street- and eye-view level. The methodology is metrics based and scalable which provides a data driven assessment of heat-related vulnerability. The findings of this study first contribute to sustainable development by developing a method to identify geographical areas or neighbourhoods that require heat mitigation; and enforce policies improving tree shade on routes, as a heat adaptation strategy, which will lead to increasing active travel and produce significant health benefits for residents. The approach can be also used to guide post COVID-19 city planning and design.

• Zoonotic nematodes of the genus Trichinella are known to occur in countries bordering the Mediterranean Sea. For example, Trichinella spiralis was reported in pigs and humans at the turn of the Second World War in Sicily and Trichinella britovi in pigs, red foxes and dogs from Corsica and Sardinia since 2004. Following the discovery of T. britovi in Corsica and Sardinia, a question arose as to whether this species was also present in Sicily which is located only 3.14 km from continental Italy. To address this query, we investigated the presence of anti-Trichinella antibodies in the serum of Nebrodi black pigs, a breed that is bred in the wild in the Nebrodi Regional Park, a protected area of the island rich in flora and fauna. Blood samples were collected from 112 Nebrodi black pigs from five farms. Sera were tested by ELISA and ELISA positive sera were confirmed by Western blot (Wb) using excretory/secretory antigens. Eighteen (16.1%) serum samples belonging to 17 fattening pigs and 1 boar tested positive by Wb. Positive sera tested by Wb using crude worm extract antigens (CWE), displayed a banding pattern similar to the CWE-Wb pattern of T. spiralis and T. britovi reference pig sera but different to that of T. pseudospiralis reference pig sera. No larvae were detected in muscles of serologically positive pigs by artificial digestion. The presence of anti-Trichinella antibodies in the absence of larvae in the muscles, suggests that the pigs were infected with T. britovi and not T. spiralis whose larvae survive in the muscles for at least two years. These results suggest that T. britovi is circulating in Nebrodi Regional Park in Sicily. ELISA testing may constitute a suitable tool for large-scale screening of Trichinella spp. infection in free-ranging pigs, when ELISA-positive sera are confirmed by Wb. Free-ranging animals can act as sentinels for the presence of zoonotic nematodes of the genus Trichinella in wildlife.

Households’ links with local Government provide important support for disaster resilience and recovery on the Bangladeshi coast. Few previous studies of disaster resilience and recovery have explored how linking social networks—and in particular local government—contribute. Using household surveys, focus groups, and key informant interviews, we examine strengths and weaknesses of local government’s contribution, using two cyclone-affected coastal villages as case studies. The findings show that local government provides important support, for example relief distribution, livelihood assistance, and reconstruction of major community services. However, patronage relationships (notably favouring political supporters) and bribery play a substantial role in how those responsibilities are discharged. The equity and efficiency of these contributions to recovery are markedly diminished by corruption. Reducing corruption in UP’s contributions to disaster recovery could significantly improve resilience; however, general reform of governance in Bangladesh would needed to bring this about.
