11. IgG Autoantibodies Induced by T. cruzi During Pregnancy: Correlation with Gravidity Complications and Early Outcome Assessment of the Newborns

Abstract

Objective The aim of the present research was to evaluate the correlation of vertically transmitted IgG antibodies induced by T. cruzi and newborn early outcome assessment, mainly birth weight and gestational age. Methods We performed a cross-sectional study with 183 pregnant women (64 with asymptomatic Chagas disease) and their newborns. Both were subjected to complete clinical examination. Peripheral parasitemia was assessed in mother and neonates by parasite detection through microscopic examination of the buffycoat from mother's peripheral and cord blood. Antibodies induced by T. cruzi, such as anti-FRA, anti-B13, anti-p2[beta] and anti-T. cruzi were assessed by immunoassay. Birth weight, general condition evaluation by APGAR Score and gestational age by Capurro Score, were determined in newborns. Results The rate of stillbirth background and pregnancy-induced hypertension were higher in patients with Chagas disease (p = 0.01 and p = 0.02, respectively). Parasitemia was detectable in 17 mothers and 4 newborns. The newborns of mothers with detectable parasitemia presented decreased gestational age (p = 0.006) and body weight (p = 0.04). Mostly all the mothers with Chagas disease and all their newborns have positive values of antibodies induced by T. cruzi; however, only anti-p2[beta] showed to be related to the presence of complication during pregnancy (OR 2.35, p = 0.036), and to low birth weight (OR 1.55, p = 0.02). Conclusions Low birth weight and decreased postnatal estimation of maturity were related to detectable parasitemia in the mother. Abstract

This study aimed to determine the clonal relationship among 137 Streptococcus uberis isolates from bovine milk with subclinical or clinical mastitis in Argentina and to assess the prevalence and conservation of pauA and sua genes. This information is critical for the rational design of a vaccine for the prevention of bovine mastitis caused by S. uberis. The isolates were typed by random amplified polymorphic DNA (RAPD) analysis and by pulsed-field gel electrophoresis (PFGE). The 137 isolates exhibited 61 different PFGE types and 25 distinct RAPD profiles. Simpson's diversity index was calculated both for PFGE (0.983) and for RAPD (0.941), showing a high discriminatory power in both techniques. The analysis of the relationship between pairs of isolates showed 92.6 % concordance between both techniques indicating that any given pair of isolates distinguished by one method tended to be distinguished by the other. The prevalence of the sua and pauA genes was 97.8 % (134/137) and 94.9 % (130/137), respectively. Nucleotide and amino acid sequences of the sua and pauA genes from 20 S. uberis selected isolates, based on their PFGE and RAPD types and geographical origin, showed an identity between 95 % and 100 % with respect to all reference sequences registered in GenBank. These results demonstrate that, in spite of S. uberis clonal diversity, the sua and pauA genes are prevalent and highly conserved, showing their importance to be included in future vaccine studies to prevent S. uberis bovine mastitis. Este estudio pretendió determinar la relación clonal entre 137 aislamientos de S. uberis obtenidos de leche de bovinos con mastitis clínica o subclínica en la Argentina, como así también la prevalencia y la conservación de los genes sua y PauA entre dichos aislamientos. Esta información es crítica para el diseño racional de una vacuna que prevenga la mastitis bovina por S. uberis. Los aislamientos se tipificaron molecularmente por amplificación al azar del ADN polimórfico (RAPD) y mediante electroforesis de campos pulsados (PFGE). Los 137 aislamientos mostraron 61 pulsotipos mediante PFGE y 25 tipos de RAPD diferentes. Los índices de Simpson calculados fueron 0,983 por PFGE y 0,941 por RAPD; esto evidencia el elevado poder discriminatorio de ambas técnicas. El análisis de la relación entre pares de aislamientos mostró un 92,6 % de concordancia entre ambas técnicas, lo que indica que cualquier par de aislamientos que fue distinguido por un método tendió a ser distinguido por el otro. La prevalencia de los genes sua y puaA fue del 97,8 % (134/137) y 94,9 % (130/137), respectivamente. Las secuencias de nucleótidos y de aminoácidos codificados por los genes sua y pauA de los 20 aislamientos de S. uberis seleccionados sobre la base de su tipo de PFGE y RAPD y origen geográfico tuvieron un porcentaje de identidad de entre 95 % y 100 % con respecto a todas las secuencias de referencia registradas en GenBank. Estos resultados demuestran que, a pesar de la diversidad clonal de S. uberis, los genes sua y pauA son prevalentes y están altamente conservados y deberían ser incluidos en futuros estudios de vacunas para prevenir mastitis bovina causada por S. uberis. Abstract

Monodisperse polymer particles with carboxyl and acetal functionalities were synthesized through a two-step emulsion polymerization process. In the first step, latex particles were synthesized by batch emulsion polymerization of styrene (St); and in the second step, the functional monomers (methacrylic acid or acrolein diethyl acetal) were copolymerized with St onto the previously formed polystyrene particles. The synthesized “core-shell” latexes were used as support for their sensitization (by covalent coupling) with two antigenic recombinant proteins of Trypanosoma cruzi (RP1 and CP1). Polymer latexes and latex-protein complexes were characterized by measuring their colloidal stability, average particle size, shell thickness and protein thickness through conductimetric titration, dynamic light scattering, turbidimetry and scanning electron microscopy. 23. Survey of potential factors involved in the low frequency of CP5 and CP8 expression in Staphylococcus aureusisolates from mastitis of dairy cattle from Argentina, Chile, and Uruguay

Abstract

Staphylococcus aureusproduces capsular polysaccharides (CPs) both in vivo and under defined culture conditions being serotypes 5 and 8 the most prevalent. S. aureusisolates that fail to produce CP5 or CP8 are defined as non-typeable (NT). Loss of capsule expression, however, may lead to S. aureuspersistence in a chronically infected host. The prevalence of NT strains of S. aureusisolated from bovine mastitis varies according to the geographic origin of the strain. The aims of this work were to detect phenotypically and genotypically the capsular profile of 144 S. aureusisolated from bovine mastitis in Argentina, Chile, and Uruguay and explore the factors that are considered to be associated with capsule expression as presence of IS257, IScap, and agrtyping of non-related collection. The detection of the IS257, IScap, capgenes, and agrtyping was performed using PCR. The detection and quantification of capsular polysaccharide production were performed by ELISA assays. We found that 96% of the S. aureusisolates investigated carried cap5(8)genes but over 75% of strains do not express capsule in the three countries studied. However, only 6 isolates from Argentina carried the IScapelement that totally suppressed the expression of the capsule, suggesting that other factors could influence on CP expression. Moreover, the agrI/NT association was statistically significant suggesting that this profile is a phenomenon observed not only in other parts of the world but also in our region.

Abstract

A successful Staphylococcus aureus vaccine should elicit a long-term antibody response that prevents establishment of the infection. The aim of the present study was to evaluate the functional role of antibodies raised against different S. aureus CP5 vaccines in invasion to bovine mammary epithelial cells (MAC-T) and phagocytosis by bovine milk macrophages in vitro. Sera and whey from cows immunized with a whole-cell S. aureus CP5 vaccine adjuvanted with Al(OH)3 or with ISCOM Matrix, significantly reduced internalization of S. aureus in MAC-T cells without significant differences between both groups. The effect of antibodies generated by a S. aureus whole-cell and a lysate vaccine formulated with ISCOM Matrix was also evaluated. Sera and whey from both immunized groups significantly reduced S. aureus internalization in MAC-T cells without significant differences between both groups. Whey antibodies against whole-cell and lysate vaccines were also able to inhibit internalization in MAC-T cells of a heterologous S. aureus strain. In addition, sera from animals vaccinated with S. aureus lysate or bacterin promoted milk macrophage phagocytosis. These results provide an insight into the potential mechanisms by which these vaccines can afford protection to the mammary gland against S. aureus intramammary infection. [ABSTRACT FROM AUTHOR]

Abstract

Tesis para la obtención del grado académico de Doctora en Ciencias Biológicas, de la Universidad Nacional del Litoral, en 2012 Staphylococcus aureus es el patógeno más frecuentemente aislado de casos de mastitis bovina en Argentina y el mundo. Las particulares características de las infecciones intramamarias por S. aureus, han llevado a la búsqueda de alternativas para coadyuvar al control de esta enfermedad, siendo una de ellas la inmunización. El objetivo general de este trabajo fue desarrollar una vacuna prototípica contra mastitis bovinas causadas por S. aureus. Se compararon las respuestas inmunes generadas en bovinos por bacterinas de S. aureus productoras de polisacárido capsular 5 (CP5), formuladas con Al(OH3), o Iscom Matrix. La formulación con Iscom Matrix potenció la producción de anticuerpos específicos en sangre y leche, con elevada capacidad opsónica. Posteriormente, se compararon las respuestas inmunes generadas en bovinos por una bacterina y un lisado de S. aureus CP5 formulados con Iscom Matrix. La formulación con lisado bacteriano mostró un mejor desempeño en cuanto a la producción de anticuerpos específicos y funcionales, y la expresión de citoquinas con potencial importancia en el control de infecciones por S. aureus. Finalmente, se obtuvo un inmunógeno prototípico compuesto por lisado de S. aureus CP5, suplementado con los antígenos recombinantes proteína de unión a fibronectina A (FnBP-A), clumpling factor A (ClfA) y β-toxina, formulado con Iscom Matrix. La inmunización de vaquillonas con el inmunógeno multicomponente potenció la respuesta de anticuerpos en sangre y leche dirigidos hacia los antígenos recombinantes. Los mismos resultaron eficientes en la opsonización del microorganismo y en la neutralización de la actividad de la β-toxina nativa de S. aureus in vitro. Staphylococcus aureus is the most frequently isolated pathogen from bovine mastitis in Argentina and worldwide. The peculiar pathogenic characteristics of S. aureus lead to the development of alternatives measures, like immunization, to control this disease. The aim of this study was to develop a prototype vaccine against S. aureus bovine mastitis. Immune responses induced in heifers by S. aureus bacterins from a capsular polisacharide 5 (CP5) producing strain, formulated either with Al(OH)3 or Iscom Matrix™, were compared. Formulation of experimental immunogen with Iscom MatrixTM strengthened humoral in blood and milk, with an augmented opsonic capacity. In addition, immune responses generated in heifers by either a S. aureus CP5 bacterin or lysate, formulated with Iscom MatrixTM were evaluated. The bacterial lysate formulation showed a better performance regarding production of specific and functional antibodies and induction of cytokines involved in early immune responses to the immunogen. Finally, a prototype immunogen composed by S. aureus CP5 lysate, supplemented with recombinant antigens fibronectin binding protein A (FnBP-A), clumping factor A (ClfA) and β-toxin, adjuvanted with Iscom MatrixTM, was obtained. Heifers’ immunization with the multicomponent immunogen strengthened antibody responses against recombinant antigens in blood and milk.
