Lead (Pb) is a non-essential, highly toxic, and persistent element widely recognized as one of the most concerning pollutants. It is listed on the Priority List of Hazardous Substances. Widespread environmental contamination from Pb is a serious issue for human health and wildlife. In fish, Pb mainly accumulates in the liver, which is a key component for metal detoxification and excretion processes. In this study, we investigated, for the first time, the morphological and functional injuries induced in zebrafish (Danio rerio) liver by two very low and environmentally relevant concentrations of Pb (2.5 and 5 μg/L) after 48, 96, and 192 h of exposure. We observed significant histological alterations in all the exposed samples, and it was demonstrated that the extent of injuries increased with dose and exposure time. The most common modifications observed were congestion of blood vessels and sinusoids, cytoplasmic vacuolizations, parenchyma dyschromia, and macrophage proliferation. Pb administration also resulted in a significant increase in lipid content and the upregulation of key genes that are involved in metal detoxification (mtf1) and the defensive response against oxidative stress (sod1 and cat). We show that even very low doses of Pb can disrupt liver morphology and function.

We analyzed the body length, age structure, and age at sexual maturity of the invasive Asian common toad Duttaphrynus melanostictus from different sites in Toamasina, east Madagascar. We used skeletochronology as a proxy for age estimation, while gonads were histologically analyzed to determine the age of sexual maturity. The analysis of pooled age data from three sites investigated in 2016 showed that both sexes were larger, although not older, than those of native populations. For the individuals from Madagascar, the males were significantly smaller and younger (mean ± SD, SVL: 71.4 ± 1.6 mm; age: 1.8 ± 0.7 years) than the females (SVL: 78.42 ± 1.9 mm; age: 2.7 ± 1.3 years), when the data were pooled, but when the data were analyzed separately for each of the three sites, similar results were obtained only for one site. The oldest recorded male and female were 3 and 6 years old, respectively. Gonadal histology showed that the males and females reach sexual maturity after the first and second years of age, respectively. Further studies are needed to understand if the larger size and faster growth rates observed in the invasive population of D. melanostictus in Madagascar are a consequence of more favorable environmental conditions with respect to the native range (e.g., the availability of larger trophic niches, a lack of competitors, and lower predatory pressure), and we suggest to extend the monitoring of these life history traits to understand how they might influence the invasion.

We provide here the first karyotype description of eight Uroplatus species and a characterization of their chromosomal diversity. We performed a molecular taxonomic assessment of several Uroplatus samples using the mitochondrial 12S marker and a comparative cytogenetic analysis with standard karyotyping, silver staining (Ag-NOR) and sequential C-banding + Giemsa, +Chromomycin A3 (CMA3), +4′,6-diamidino-2-phenylindole (DAPI). We found chromosomal variability in terms of chromosome number (2n = 34–38), heterochromatin composition and number and localization of loci or Nucleolar Organizer Regions (NORs) (alternatively on the 2nd, 6th, 10th or 16th pair). Chromosome morphology is almost constant, with karyotypes composed of acrocentric chromosomes, gradually decreasing in length. C-banding evidenced a general low content of heterochromatin, mostly localized on pericentromeric and telomeric regions. Centromeric bands varied among the species studied, resulting in CMA3 positive and DAPI negative or positive to both fluorochromes. We also provide evidence of a first putative heteromorphic sex chromosome system in the genus. In fact, in U. alluaudi the 10th pair was highly heteromorphic, with a metacentric, largely heterochromatic W chromosome, which was much bigger than the Z. We propose an evolutionary scenario of chromosome reduction from 2n = 38 to 2n = 34, by means of translocations of microchromosomes on larger chromosomes (often involving the NOR-bearing microchromosomes). Adding our data to those available from the literature, we show that similar processes characterized the evolutionary radiation of a larger gecko clade. Finally, we hypothesize that sex chromosome diversification occurred independently in different genera.

The endemic Malagasy Scincinae lizards are amongst the most species-rich squamate groups on the island. They colonised all bioclimatic zones and display many ecomorphological adaptations to a fossorial (burrowing) lifestyle. Here we propose a new phylogenetic hypothesis for their diversification based on the largest taxon sampling so far compiled for this group. We estimated divergence times and investigated several aspects of their diversification (diversification rate, body size and fossorial lifestyle evolution, and biogeography). We found that diversification rate was constant throughout most of the evolutionary history of the group, but decreased over the last 6–4 million years and independently from body size and fossorial lifestyle evolution. Fossoriality has evolved from fully quadrupedal ancestors at least five times independently, which demonstrates that even complex morphological syndromes – in this case involving traits such as limb regression, body elongation, modification of cephalic scalation, depigmentation, and eyes and ear-opening regression – can evolve repeatedly and independently given enough time and eco-evolutionary advantages. Initial diversification of the group likely occurred in forests, and the divergence of sand-swimmer genera around 20 Ma appears linked to a period of aridification. Our results show that the large phenotypic variability of Malagasy Scincinae has not influenced diversification rate and that their rich species diversity results from a constant accumulation of lineages through time. Heavy metal contamination is recognized worldwide as a serious threat to human health and wildlife, and reducing their emissions is a priority of international and EU actions. Due to its persistence, high bioaccumulation tendency, and toxicity properties, lead (Pb) is one of the heavy metals of greatest concern. Even at low concentrations, lead induces various clinical and subclinical conditions in both humans and animals, and it has been included in the priority list of hazardous substances. In the present study, we used zebrafish's early stages as a model, given their well-acknowledged predictive value in the risk assessment of chemicals. This study was designed to investigate the morphological and morphometric alterations induced by Pb during zebrafish's early development and disclose the putative effects stage- and/or dose-dependent. We examined injuries induced by two environmentally relevant and extremely low concentrations of Pb (2.5 μg/L and 5 μg/L) during two exposure windows: early (between 1 and 7 dpf) and late (between 2 and 8 dpf). We clearly demonstrated that the incidence and severity of morphological abnormalities increased with increasing Pb dose and exposure time in both early and late-exposed groups. Furthermore, we revealed that malformation severity was significantly higher in the early exposed group than in the late exposure group at all exposure times and for both tested doses, thus highlighting the high sensitivity of zebrafish during the initial stages of development. The information presented in this paper emphasizes the effectiveness of morphological biomarkers in unveiling threatening situations and supports the role of zebrafish embryos and larvae in risk assessment and environmental monitoring.

We characterized the body size (as snout-vent length), age, sexual size dimorphism, and growth rate in a population of one of the larger riparian frog from Madagascar (Mantidactylusgrandidieri) from a rainforest patch close to Vevembe, SE Madagascar. We identified a significant female-biased sexual size dimorphism. Age was estimated using phalangeal skeletochronology and was significantly higher in females than in males. Modal age class turned out to be 4 years in both sexes but a large percentage of adult females (75%) fell in the 5–6 years-old classes, while no male exceeded 4 years. We here report M.grandidieri as a medium-long-lived anuran species. Von Bertalanffy’s model showed similar growth trajectories between the sexes although the growth coefficient in females (k = 0.335) was slightly but not significantly higher than in males (k = 0.329).

Lizards represent unique model organisms in the study of sex determination and sex chromosome evolution. Among tetrapods, they are characterized by an unparalleled diversity of sex determination systems, including temperature-dependent sex determination (TSD) and genetic sex determination (GSD) under either male or female heterogamety. Sex chromosome systems are also extremely variable in lizards. They include simple (XY and ZW) and multiple (X1X2Y and Z1Z2W) sex chromosome systems and encompass all the different hypothesized stages of diversification of heterogametic chromosomes, from homomorphic to heteromorphic and completely heterochromatic sex chromosomes. The co-occurrence of TSD, GSD and different sex chromosome systems also characterizes different lizard taxa, which represent ideal models to study the emergence and the evolutionary drivers of sex reversal and sex chromosome turnover. In this review, we present a synthesis of general genome and karyotype features of non-snakes squamates and discuss the main theories and evidences on the evolution and diversification of their different sex determination and sex chromosome systems. We here provide a systematic assessment of the available data on lizard sex chromosome systems and an overview of the main cytogenetic and molecular methods used for their identification, using a qualitative and quantitative approach.

The knowledge of demographic traits such as longevity, growth rates and age at sexual maturity is crucial for understanding the structure of a population in its natural environment and implementing appropriate strategies for its management and conservation. Based on counts of growth layer groups in sections of decalcified teeth using the paraffin technique, we estimated the age and growth of 25 individuals of striped dolphin (Stenella coeruleoalba) found dead stranded along the coast of Campania and Calabria (south Italy, central-western Mediterranean) from 2013 to 2018. Seven individuals, with TL of 100–110 cm, were calves under 1 year old. The oldest male and female individuals were 19 and 14 years old, respectively. Growth curve estimated using the Gompertz growth model (GGM) showed that in S. coeruleoalba male growth trajectories are partly in accordance with those reported in other studies on the same species from different Mediterranean areas. The high frequency (28%) of calves strongly suggests that females of this species use the marine area all around the south-western Italian coasts to give birth to their offspring. Furthermore, a comparison with the estimated age of striped dolphins from other Mediterranean marine areas shows that the longevity of the individuals examined in this study is much lower. Our study provides information toward understanding the demographic traits of S. coeruleoalba from Mediterranean Sea. The results reported here can be useful for future research aimed at understanding population structure, mortality patterns and the effects of anthropogenic activity on the survival of this species in this marine area.

Simple Summary The Lycian salamanders of the genus Lyciasalamandra are characterized by a debated taxonomy and phylogenetic relationships. They have been the subject of various molecular and phylogenetic analyses, but their chromosomal diversity is completely unknown. We here present a comparative cytogenetic analysis on five out of the seven described species and seven subspecies of Lyciasalamandra, providing the first karyological assessment on the genus and comparing them to closely related representatives of the genus Salamandra. We analyzed the occurrence and distribution of different conserved (chromosome number and morphology) and highly variable karyological features. We found an impressive diversity in the configuration of nucleolus organizing regions (NORs), which alternatively occur either as heteromorphic or homomorphic loci on distinct regions of different chromosome pairs. We highlight that the observed peculiar taxon-specific pattern of chromosome markers supports the taxonomic validity of the different studied evolutionary lineages and is consistent with a scenario of synchronous evolution in the Lycian salamanders. Abstract We performed the first cytogenetic analysis on five out of the seven species of the genus Lyciasalamandra, including seven subspecies, and representatives of its sister genus Salamandra. All the studied species have a similar karyotype of 2n = 24, mostly composed of biarmed elements. C-bands were observed on all chromosomes, at centromeric, telomeric and interstitial position. We found a peculiar taxon-specific NOR configuration, including either heteromorphic and homomorphic NORs on distinct regions of different chromosomes. Lyciasalamandra a. antalyana and L. helverseni showed two homomorphic NORs (pairs 8 and 2, respectively), while heteromorphic NORs were found in L. billae (pairs 6, 12), L. flavimembris (pairs 2, 12), L. l. luschani (pairs 2, 12), L. l. basoglui (pairs 6, 12), L. l. finikensis (pairs 2, 6) and S. lanzai (pairs 8, 10). Homomorphic NORs with an additional supernumerary site were shown by S. s. salamandra (pairs 2, 8) and S. s. gigliolii (pairs 2, 10). This unexpected highly variable NOR configuration is probably derived from multiple independent NOR translocations and paracentric inversions and correlated to lineage divergence in Lyciasalamandra. These results support the taxonomic validity of the studied taxa and are consistent with a hypothesized scenario of synchronous evolution in the genus.

Except for polyploidy, parthenogenesis and hybridogenesis, the role of chromosome rearrangements in speciation is still highly debated. This paper highlights an unusual karyotype diversification and a putative case of chromosomal speciation in the endemic Malagasy tomato frogs. The tomato frogs include Dyscophus insularis as well as the sister species D. antongilii and D. guineti, which are characterized by a disjunct distribution and a low genetic divergence, probably due to a recent hybridization. Our results show that the three Dyscophus species present 2n = 26 biarmed chromosomes, differing in chromatin pattern. Interestingly, both sexes of D. guineti show a heteromorphic chromosome pair 11, with one element larger (11a) than the other (11b) due to the presence of a large terminal tract on the short arm with an euchromatic band enclosed between two heterochromatic bands. This fixed condition reminds that of some newts of the genus Triturus, but in Dyscophus it probably characterizes the beginning of a cladogenetic process. We hypothesize that in D. guineti the raising of the heteromorphic 11th chromosome pair followed the hybridization with D. antongilii, originating a post-zygotic barrier and representing a case of sympatric or parapatric chromosomal speciation. This speciation mechanism may also have had important biogeographic consequences, shaping the current disjunct distribution of D. antongilii and D. guineti, so far not sufficiently explained.

The data presented in this paper stand as supplementary information of the associated article “Karyological characterization of the common chameleon (Chamaeleo chamaeleon) provides insights on the evolution and diversification of sex chromosomes in Chamaeleonidae” [1] . This work provides (i) raw experimental data on the karyology of the common chameleon Chamaeleo chamaeleon and (ii) the results of bioinformatic analysis on sex-specific and repeated DNA sequences found in the same species. The karyological information here presented includes traditional staining method (Giemsa staining) and sequential C-banding + fluorochromes performed on Tunisian samples of the species. The sequence data include the alignments of the isolated DNA sequences with homologous sequences found in squamate Short Read Archives (SRAs) and the results of searches in public nucleic acid databases.

Information on demographic and life-history traits of endangered vertebrate species, such as sea turtles, is crucial for planning management and conservation actions. We applied skeletochronology of phalanges to estimate the age of loggerhead turtles, Caretta caretta, found dead stranded along the beaches of Campania (western Mediterranean) from 2013 to 2017. To obtain maturity data, we examined gonads from a subsample of 7 males and 11 females. Overall, curved carapace length (CCL) ranged from 5.6 to 90.8 cm, but for most turtles (89%) it was 50–79.9 cm. Predominance of stranded females (62%) was recorded. Ten out of the eighteen histologically examined gonads allowed estimating maturity. Based on the lines of arrested growth counting, the estimated age of the examined specimens ranged from 0 (hatchling) to 26 years. The modal age was 14 years for males and 17 years for females. The smallest male with spermatogenetic activity had a CCL of 65 cm and was estimated to be 16 years old. The smallest female with follicular development stage, characterising the transition towards adulthood, had a CCL of 69.5 cm and was estimated to be 20 years old. Anthropogenic factors were responsible for 36% of the mortality of individuals, followed by parasitic/infective pathologies (20%).

Sexual differentiation (SD) during development results in anatomical, metabolic, and physiological differences that involve not only the gonads, but also a variety of other biological structures, such as the brain, determining differences in morphology, behavior, and response in the breeding season. In many reptiles, whose sex is determined by egg incubation temperature, such as the leopard gecko, Eublepharis macularius, embryos incubated at different temperatures clearly differ in the volume of brain nuclei that modulate behavior. Based on the premise that "the developmental decision of gender does not flow through a single gene", we performed an analysis on E. macularius using three approaches to gain insights into the genes that may be involved in brain SD during the thermosensitive period. Using quantitative RT-PCR, we studied the expression of genes known to be involved in gonadal SD such as WNT4, SOX9, DMRT1, Erα, Erβ, GnRH, P450 aromatase, PRL, and PRL-R. Then, further genes putatively involved in sex dimorphic brain differentiation were sought by differential display (DDRT-PCR) and PCR array. Our findings indicate that embryo exposure to different sex determining temperatures induces differential expression of several genes that are involved not only in gonadal differentiation (PRL-R, Wnt4, Erα, Erβ, p450 aromatase, and DMRT1), but also in neural differentiation (TN-R, Adora2A, and ASCL1) and metabolic pathways (GP1, RPS15, and NADH12). These data suggest that the brains of SDT reptiles might be dimorphic at birth, thus behavioral experiences in postnatal development would act on a structure already committed to male or female.

Chameleons display high karyological diversity in chromosome number (from 2n = 20 to 62), morphology, heterochromatin distribution and location of specific chromosomal markers, making them unique study models in evolutionary cytogenetics. However, most available cytogenetic data are limited to the description of the chromosome number and morphology. Concerning sex chromosomes, our knowledge is limited to ZZ/ZW and Z1Z1Z2Z2/Z1Z2W systems in the genus Furcifer and the isolation of sex-linked, male-specific, sequences in Chamaeleo calyptratus, but the putative XY chromosomes have still to be identified in Chamaeleo and the conservation of male heterogamety in the genus needs confirmation from other species. In this study we performed a molecular and a cytogenetic analysis on C. chamaeleon, using standard, banding methods and molecular cytogenetics to provide a throughout karyological characterization of the species and to identify and locate the putative XY chromosomes. We confirm that the chromosome formula of the species is 2n = 24, with 12 metacentric macrochromosomes, 12 microchromosomes and NORs on the second chromosome pair. Heterochromatin was detected as weak C-bands on centromeric regions, differently from what was previously reported for C. calyptratus. Fluorescence in situ hybridization (FISH) showed the occurrence of interspersed telomeric signals on most macrochromosomes, suggesting that ancient chromosome fusions may have led to a reduction of the chromosome number. Using a combination of molecular and FISH analyses, we proved that male specific Restriction site-Associated DNA sequences (RADseq) isolated in C. calyptratus are conserved in C. chamaeleon and located the putative XY chromosomes on the second chromosome pair. We also identified different transposable elements in the focal taxa, which are highly interspersed on most chromosome pairs.

The common lizard (Zootoca vivipara) displays characteristic cytogenetic, reproductive, molecular, and biogeographic variability. This species comprises oviparous and viviparous populations with disjunct distribution and sex chromosome polymorphisms, from simple ZZ/ZW to complex Z1Z1Z2Z2/Z1Z2W systems with different morphologies of the W chromosome. In this study, we used the primers SINE A and SINE B and a newly designed primer pair to (1) obtain information on the presence and distribution of transposable elements (TEs) in 8 squamate families and (2) assess the chromosomal location of SINE Squam elements in Z. vivipara. PCR amplification with SINE A and SINE B produced single or multiple products in different Z. vivipara populations, subsequently used to design the SINE-Zv primers. Using the newly designed SINE-Zv primers, we identified 2 sequences of about 700 and 300 bp (SINE-Zv 700 and SINE-Zv 300) in all the investigated populations of Z. vivipara. Fluorescence in situ hybridizations showed a preferential localization of SINE-Zv sequences in the peritelomeric regions of almost all chromosomes, with the exception of the W. Both sequences contained a distinct segment of SINE Squam2. SINE-Zv 700 appeared to be restricted to Z. vivipara, while SINE-Zv 300 contained a partial Gypsy sequence that is highly conserved among Squamata and showed high identity values (72-93%) with several transcripts from different species. Using the same primers, we also highlighted the presence of another highly conserved Gypsy-like fragment in snakes which displayed significant similarity with the stomatin-like protein 2 of colubrids. Our results suggest that SINEs and the Gypsy-like elements are widely distributed among squamates and may have played an active role in their genomic evolution and differentiation.

Background: Endemic fluorosis induced by high concentrations of fluoride in groundwater and soils is a major health problem in several countries, particularly in volcanic areas.Aim: To evaluate the occurrence of dental fluorosis resulting from exposure to high levels of environmental fluoride in 79 AD Herculaneum and close Vesuvius towns.Subjects and methods: The occurrence of dental fluorosis from teeth of the Herculaneum victims of the 79 AD eruption and some individuals from Pompeii (14-37 AD) and Nocera Inferiore (Salerno, IV sec. AD) was detected by means of Particle Induced Gamma-ray Emission technique (PIGE).Results: A clinical and analytical scenario of dental fluorosis resulted from the extreme high fluorine tooth content detected in teeth from Herculaneum and the Vesuvius area inhabitants. The adoption of PIGE technique has proved to be particularly effective in showing moderate as well as milder forms of dental fluorosis, otherwise not clearly detectable by clinical and histological analysis.Conclusions: Morphological, histological and elemental analysis of teeth of the 79 AD Herculaneum population show that in this area fluorosis occurred since Roman times.

Due to their particular phylogenetic position and biological characteristics, squamate reptiles and, in particular, snakes are becoming an increasingly important model for fields such as evolutionary biology, molecular ecology and adaptation. Recently, during a study to analyze the evolutionary history of European whip snakes, we found a LINE1 (L1)-like sequence (GenBank accession no. LM644476), herein called TRL1L, and while there are data on the abundance of L1 in snakes, their genomic and chromosome localization is still largely unexplored. We therefore performed a study to obtain information on TRL1L abundance, distribution and conservation in snake species, belonging to the Colubridae, Lamprophiidae and Viperidae families, using quantitative dot-blot and fluorescence in situ hybridization (FISH). TRL1L showed a high identity with homologous segments of L1s of lizards the Anolis carolinensis and Lacerta agilis and the zebrafish Danio rerio. The discovered sequences are truncated L1 elements which occur with a low copy number, about 0.1% of the genome of the species studied. This evidence suggests that L1 retroposons have a similar landscape in lizard and snake genomes, probably because similar processes limited L1 distribution in their genomes. TRL1L showed a non-random chromosome distribution pattern. It was scarcely located on autosomes and on the euchromatic W chromosome of Cerastes vipera, while mostly found on the heterochromatic W chromosome of Hierophis carbonarius and Elaphe quatuorlineata. Our data support the hypothesis that a ‘purifying selection’ against the accumulation of L1 elements takes place in recombining regions and highlight the possible role of these elements in the differentiation processes of the snake heterochromatic W chromosome. Interestingly, the preferential distribution of TRL1L on the heterochromatic W chromosomes of the studied snakes appears to be similar to that observed in mammals for L1 accumulation on differentiated Y chromosomes. This finding suggests that a convergent process may have taken place in the differentiation of vertebrate heterochromatic sex chromosomes

We used a multidisciplinary approach to infer the taxonomy and historical biogeography of Hierophis viridiflavus and H. gemonensis, performing molecular analyses of mitochondrial (16S, Cyt-b, ND4) and nuclear markers (PRLR), a landmark-based morphometric study and a cytogenetic analysis. Our data distinguished three main groups in the studied species, corresponding to H. gemonensis and to two monophyletic clades (E and W) within H. viridiflavus. Clades E and W display a significant genetic (about 4% for Cyt-b and ND4) and morphological divergence and a different morphology of the W sex chromosome (submetacentric in clade E and telocentric in clade W). Taking into account the existing divergence, these clades appear to represent independent phylogenetic units, deserving elevation to species status. Specific names should be H. viridiflavus (Lacepede, 1789) and H. carbonarius (Bonaparte 1833) for clades W and E, respectively. The phylogeography of the studied species is only partially concordant with a general pattern of ‘southern richness and northern purity’ of genetic diversity, whereas H. gemonensis exhibits high genetic diversity at low latitudes (especially in the Peloponnese), H. carbonarius shows a number of different haplotypes both at low (along the southern Italian Apennines and in Sicily) and high latitudes in Italy. Furthermore, a relaxed clock model hypothesizes the differentiation between H. gemonensis and H. viridiflavus sensu lato at about 7 Mya, in the Messinian. Subsequently, the speciation involving H. viridiflavus sensu stricto and H. carbonarius took place in the Quaternary, probably as a result of Pleistocene climatic oscillations. Furthermore, our results are consistent with the existence of several ‘refugia within refugia’ in Italy and in the Balkans and depict the major cladogenesis as allopatric events, mainly driven by paleoclimatic and geographical factors.

In this work, we performed a biogeographic analysis with Bayesian binary MCMC (BBM) statistical dispersal-vicariance analysis (S-DIVA) and species distribution models (SDM) on three phylogenetically closely related Mediterranean whipsnakes (Hierophis gemonensis, H. carbonarius, H. viridiflavus), to investigate the pathways of their geographical diversification and locate putative refugial areas in the last glacial maximum (LGM). Our analysis suggests that the diversification processes between the studied species overall followed an east-west route, from eastern Greece to the Iberian Peninsula and continental France, highlighting a significant role of dispersal and vicariance processes at both inter- and intraspecific levels. In particular, the main lineage-splitting events between H. gemonensis, H. carbonarius and H. viridiflavus coincide with two events of vicariance, involving respectively eastern Greece and eastern Italy, and eastern Italy and western Italy, Iberian Peninsula and continental France. SDM analyses highlight the occurrence of multiple putative glacial refugia in the Balkans, Italy and southern France, which represent well the occurrence of distinct "refugia within refugia" in the main Mediterranean peninsulas. Furthermore, our results suggest how these refugia may have played an important role during the Quaternary climatic oscillations in shaping the current haplotype distribution of European whipsnakes. © 2018 Elsevier GmbH.

We performed a phylogenetic analysis using nuclear (RAG-1, RAG-2) and mitochondrial (16S) markers, a statistical Bayesian reconstruction of ancestral distribution areas and a karyological analysis on most Malagasy species of the gekkonid genus Lygodactylus. The phylogenetic analysis largely confirms major basal branching pattern of previous molecular studies, but highlights significant differences concerning both the relationships between different species groups as well as those within groups. The biogeographic analysis supports a Malagasy origin of Lygodactylus, an oversea dispersal to continental Africa and a return to Madagascar. The L. madagascariensis group (also including a new candidate species identified herein) is the most basal clade in Lygodactylus, and the sister group of a clade with all the remaining species. The second most basal clade is the L. verticillatus group, placed as the sister group of a clade comprising African and Malagasy species. The sister lineage of the L. verticillatus group originated the African radiation through an oversea dispersal out of Madagascar. Eventually, the sister lineage of the L. capensis group originated secondary dispersals from Africa to Madagascar. In Madagascar, lineage diversification in different species groups mainly occurred from southern to northern and eastern regions. Dispersal, vicariance and paleoclimatic refugia probably played a relevant role in the evolutionary history of closely related taxa and in speciation mechanisms. The cytogenetic analysis evidenced a high karyotypic variability in Lygodactylus (from 2n = 34 to 2n = 40), which is at least partly consistent with the phylogenetic relationships and the composition of the various species group. Chromosome evolution occurred independently in different lineages, mainly through a reduction in the chromosome number and starting from a putative primitive karyotype of 2n = 40 with all telocentric elements.

n thermally damaged archaeological bones the quantity, quality, and amplifiability of DNA are very much reliant on both the extent of heating and the environmental conditions of the burial context. In this study we tested the possibility of extracting and amplifying ancient DNA from human bone remains of Herculaneum victims of the 79 AD eruption of Vesuvius using a combination of histochemical and molecular methods. Long bone samples with variable degrees of chromatic and morphological alterations consistent with exposure to temperatures of about 300 °C were taken from four specimens. Using histochemical stains, bone cryostat sections from three individuals revealed DNA within osteocyte lacunae but only for one sample the DNA was suitable for PCR amplification obtained, namely from reactions with the primer pair for X and Y amelogenin (AMEL) loci. The relative sequence differed from the homologous trait of AMELX deposited in GenBank for six bases, probably due to degradation processes following death. Our data are indicative that archaeological bones exposed to high temperatures of about 300 °C should be considered for DNA analysis, given the favorable conditions of corpse burial and skeleton preservation, such as those that occurred for the 79 AD victims in Herculaneum and Pompeii.

In this first cytogenetic survey on the lamprophiid snake subfamily Pseudoxyrhophiinae, we studied the karyology of ten snake species belonging to seven genera from Madagascar (Compsophis, Leioheterodon, Liophidium, Lycodryas, Madagascarophis, Phisalixella and Thamnosophis) using standard and banding methods. Our results show a wide range of different karyotypes ranging from 2n = 34 to 2n = 46 elements (FN from 40 to 48), with nucleolus organizer regions (NORs) on one (plesiomorphic) or two (derived/apomorphic) microchromosome pairs, and W chromosome at early or advanced states of diversification from the Z chromosome. The observed W chromosome variations further support the most accepted hypothesis that W differentiation from the Z chromosome occurred by progressive steps. We also propose an evolutionary scenario for the observed high karyotype diversity in this group of snakes, suggesting that it is derived from a putative primitive pseudoxyrhophiine karyotype with 2n = 46, similar to that of Leioheterodon geayi, via a series of centric fusions and inversions among macrochromosomes and translocations of micro- either to micro- or to macrochromosomes. This primitive Pseudoxyrhophiinae karyotype might have derived from a putative Lamprophiidae ancestor with 2n = 48, by means of a translocation of a micro- to a macrochromosome. In turn, the karyotype of this lamprophiid common ancestor may have derived from the assumed primitive snake karyotype (2n = 36 chromosomes, with 16 biarmed macro- and 20 microchromosomes) by a series of centric fissions and one inversion. © 2014 The Linnean Society of London, Biological Journal of the Linnean Society, 2014, 112, 450–460.

Lepidosaurs are frequently described as having highly kinetic skulls, and different forms of cranial kinesis have been described as being characteristic of their radiation. The model of amphikinesis proposed by Frazzetta, J Morphol 1962; 111:287–319, which was long considered a synapomorphy of the large suborder Sauria, is now much debated given its uncertain distribution among the various lizard taxa and the lack of data about its morphological correlates. In this article, we analyze the anatomical correlates of different forms of cranial kinesis, with particular regard to the putative saurian amphikinesis, describing the possible diverse skull movements of several species of European gekkotans (Hemidactylus turcicus, Mediodactylus kotschyi, and Tarentola mauritanica) and lacertids (Lacerta agilis, L. bilineata, Podarcis muralis, P. siculus, and Teira dugesii). Using serial and whole-mount histology, we found clear differences between gekkotans and lacertids in the structure of several cranial joints underlining the existence of two degrees of intracranial mobility. The lacertid species possess the anatomical features for streptostyly (quadrate joints) and metakinesis (parietal-supraoccipital and parabasisphenoid-pterygoid joints) and lack the anatomical correlates for mesokinesis (mobility of frontal-parietal and palatine-pterygoid joints) and amphikinesis (coupled mesokinesis, metakinesis, and streptostyly). In contrast, geckos present all the anatomical correlates for amphikinesis as described by the traditional quadratic crank model. Finally, we present a comprehensive summary of the different forms of squamate cranial kinesis, advancing two alternative hypotheses about the evolutionary origin of amphikinesis. A karyological analysis on six Italian populations the slow worm (Anguis veronensis Pollini, 1818) was performed and their genetic differentiation at the mitochondrial 16S rRNA gene fragment from a Spanish sample has been assessed. The Italian populations were karyologically uniform, all showing 2n= 44 elements, of which 20 were macrochromosomes and 24 microchromosomes. Comparison with literature data on Central European populations showed a difference on the morphology of the 10th chromosome pair: submetacentric in Italian populations and telocentric in the Central European ones. Our analysis showed the presence of a fragile site on chromosomes of this pair, suggesting its propensity for structural rearrangements. Analysis of the 16S rRNA gene fragment showed uniformity among Italian populations (uncorrected genetic distance of 0.4%), and their genetic distinctness from the Spanish individual (uncorrected genetic distance of 4.2%). Our results confirm the existence of two different Anguis fragilis Linnaeus, 1758 lineages, each one characterized by a different cytotype.

Sexual differentiation (SD) during development results in anatomical, metabolic, and physiological differences that involve not only the gonads, but also a variety of other biological structures, such as the brain, determining differences in morphology, behavior, and response in the breeding season. In many reptiles, whose sex is determined by egg incubation temperature, such as the leopard gecko, Eublepharis macularius, embryos incubated at different temperatures clearly differ in the volume of brain nuclei that modulate behavior. Based on the premise that "the developmental decision of gender does not flow through a single gene", we performed an analysis on E. macularius using three approaches to gain insights into the genes that may be involved in brain SD during the thermosensitive period. Using quantitative RT-PCR, we studied the expression of genes known to be involved in gonadal SD such as WNT4, SOX9, DMRT1, Erα, Erβ, GnRH, P450 aromatase, PRL, and PRL-R. Then, further genes putatively involved in sex dimorphic brain differentiation were sought by differential display (DDRT-PCR) and PCR array. Our findings indicate that embryo exposure to different sex determining temperatures induces differential expression of several genes that are involved not only in gonadal differentiation (PRL-R, Wnt4, Erα, Erβ, p450 aromatase, and DMRT1), but also in neural differentiation (TN-R, Adora2A, and ASCL1) and metabolic pathways (GP1, RPS15, and NADH12). These data suggest that the brains of SDT reptiles might be dimorphic at birth, thus behavioral experiences in postnatal development would act on a structure already committed to male or female.
