1. A hybrid discrete-continuum modelling approach for the interactions of the immune system with oncolytic viral infections

Abstract

Oncolytic virotherapy, utilizing genetically modified viruses to combat cancer and trigger anti-cancer immune responses, has garnered significant attention in recent years. In our previous work arXiv:2305.12386, we developed a stochastic agent-based model elucidating the spatial dynamics of infected and uninfected cells within solid tumours. Building upon this foundation, we present a novel stochastic agent-based model to describe the intricate interplay between the virus and the immune system; the agents' dynamics are coupled with a balance equation for the concentration of the chemoattractant that guides the movement of immune cells. We formally derive the continuum limit of the model and carry out a systematic quantitative comparison between this system of PDEs and the individual-based model in two spatial dimensions. Furthermore, we describe the traveling waves of the three populations, with the uninfected proliferative cells trying to escape from the infected cells while immune cells infiltrate the tumour. Simulations show a good agreement between agent-based approaches and numerical results for the continuum model. Some parameter ranges give rise to oscillations of cell number in both models, in line with the behaviour of the corresponding nonspatial model, which presents Hopf bifurcations. Nevertheless, in some situations the behaviours of the two models may differ significantly, suggesting that stochasticity plays a key role in the dynamics. Our results highlight that a too rapid immune response, before the infection is well-established, appears to decrease the efficacy of the therapy and thus some care is needed when oncolytic virotherapy is combined with immunotherapy. 2. Reducing N Fertilization without Yield Penalties in Maize with a Commercially Available Seed Dressing

Abstract

Introducing smart and sustainable tools for climate change adaptation and mitigation is a major need to support agriculture’s productivity potential. We assessed the effects of the processed gypsum seed dressing SOP® COCUS MAIZE+ (SCM), combined with a gradient of N fertilization rates (i.e., 0%, 70% equal to 160 kg N ha−1, and 100% equal to 230 kg N ha−1) in maize (Zea mays L.), on: (i) grain yield, (ii) root length density (RLD) and diameter class length (DCL), (iii) biodiversity of soil bacteria and fungi, and (iv) Greenhouse Gases (GHGs, i.e., N2O, CO2, and CH4) emission. Grain yield increased with SCM by 1 Mg ha−1 (+8%). The same occurred for overall RLD (+12%) and DCL of very fine, fine, and medium root classes. At anthesis, soil microbial biodiversity was not affected by treatments, suggesting earlier plant-rhizosphere interactions. Soil GHGs showed that (i) the main driver of N losses as N2O is the N-fertilization level, and (ii) decreasing N-fertilization in maize from 100% to 70% decreased N2O emissions by 509 mg N-N2O m−2 y−1. Since maize grain yield under SCM with 70% N-fertilization was similar to that under Control with 100% N-fertilization, we concluded that under our experimental conditions SCM may be used for reducing N input (−30%) and N2O emissions (−23%), while contemporarily maintaining maize yield. Hence, SCM can be considered an available tool to improve agriculture’s alignment to the United Nation Sustainable Development Goals (UN SDGs) and to comply with Europe’s Farm to Fork strategy for reducing N-fertilizer inputs.

Despite the key role they play in modern medicine, their cardio and neurotoxicity (together with their short half-life) stress the need for developing implantable devices for tailored local drug release, with the aim of counterbalancing their side effects and prolonging their pharmacological activity. This review discusses the evolution of the physical forms of local anesthetic delivery systems during the past decades. Depending on the use of different biocompatible materials (degradable polyesters, thermosensitive hydrogels, and liposomes and hydrogels from natural polymers) and manufacturing processes, these systems can be classified as films or micro- or nanostructured devices. We analyze and summarize the production techniques according to this classification, focusing on their relative advantages and disadvantages. The most relevant trend reported in this work highlights the effort of moving from microstructured to nanostructured systems, with the aim of reaching a scale comparable to the biological environment. Improved intracellular penetration compared to microstructured systems, indeed, provides specific drug absorption into the targeted tissue and can lead to an enhancement of its bioavailability and retention time. Nanostructured systems are realized by the modification of existing manufacturing processes (interfacial deposition and nanoprecipitation for degradable polyester particles and high- or low-temperature homogenization for liposomes) or development of novel strategies (electrospun matrices and nanogels). The high surface-to-volume ratio that characterizes nanostructured devices often leads to a burst drug release. This drawback needs to be addressed to fully exploit the advantage of the interaction between the target tissues and the drug: possible strategies could involve specific binding between the drug and the material chosen for the device, and a multiscale approach to reach a tailored, prolonged drug release. 4. Antibiotic-free PHA-based antibacterial materials for bone regeneration

Abstract

The current world problem of antibacterial resistance to antibiotics has driven the need to develop new therapeutics as alternatives to antibiotic to prevent and fight infections. In the biomedical field, bone regeneration is one of the applications with the highest risk of infection. Therefore, alongside biological and functional properties, materials used for this application should be able to provide antibacterial features able to inhibit bacterial adhesion and growth. In such a scenario, this work focused on the development of novel antibiotic-free materials based on polyhydroxyalkanoates (PHAs) for bone regeneration following two strategies. For the first strategy, inherently antibacterial PHAs, thioester containing PHAs, were produced by bacterial fermentation. Such materials exhibited antibacterial activity by contact against S. aureus 6538P. Physical blending of thioester-PHA with P(3HHx-co-3HO-co-3HD) was investigated and a polymer ratio of 80:20 (P(3HHx-co-3HO-co-3HD): thioester-PHA) was identified as optimal to develop handleable and processable samples possessing antibacterial and in vitro biocompatibility properties. For the second strategy, novel 2D antibacterial composite materials were produced by combining P(3HB) and P(3HO-co-3HD-co-3HDD) with a novel antibacterial co-substituted hydroxyapatite, containing two therapeutic ions, selenium and strontium, conferring osteoinductive and antibacterial properties to the ceramic. The materials developed showed to be able to induce an inhibitory effect against both Gram positive and Gram negative bacteria. The possibility of developing 3D scaffolds using melt 3D printing of mcl-PHAs was investigated in this study for the first time. The process parameters were successfully optimized to produce well-defined and reproducible 3D P(3HO-co-3HD-co-3HDD) scaffolds. 3D scaffolds with dual porosity were also developed through the combination of 3D printing with salt leaching. Finally, the obtained 2D antibacterial films were used as starting materials for the development of 3D scaffolds for antibacterial synthetic bone substitutes using the optimized melt 3D printing process. Novel 3D composite scaffolds were produced using the 2D composite materials based on P(3HO-co-3HD-co-3HDD) and the antibacterial hydroxyapatite. Based on the antibacterial and cell compatibility studies, the composite material with 10 wt% of Se-Sr-HA as a filler was identified as the optimal candidate for the development of antibacterial scaffolds for bone regeneration. Novel inherently antibacterial scaffolds were produced by melt 3D printing of 20:80 blend films of thioester-PHA with P(3HHx-co-3HO-co-3HD). The 3D blend scaffolds showed an inhibitory effect against S. aureus 6538P by direct contact testing and were able to support the growth of MC3T3-E1 cells, showing potential for bone regeneration.

Abstract

ImportanceDelays in screening programs and the reluctance of patients to seek medical attention because of the outbreak of SARS-CoV-2 could be associated with the risk of more advanced colorectal cancers at diagnosis.ObjectiveTo evaluate whether the SARS-CoV-2 pandemic was associated with more advanced oncologic stage and change in clinical presentation for patients with colorectal cancer.Design, Setting, and ParticipantsThis retrospective, multicenter cohort study included all 17 938 adult patients who underwent surgery for colorectal cancer from March 1, 2020, to December 31, 2021 (pandemic period), and from January 1, 2018, to February 29, 2020 (prepandemic period), in 81 participating centers in Italy, including tertiary centers and community hospitals. Follow-up was 30 days from surgery.ExposuresAny type of surgical procedure for colorectal cancer, including explorative surgery, palliative procedures, and atypical or segmental resections.Main Outcomes and MeasuresThe primary outcome was advanced stage of colorectal cancer at diagnosis. Secondary outcomes were distant metastasis, T4 stage, aggressive biology (defined as cancer with at least 1 of the following characteristics: signet ring cells, mucinous tumor, budding, lymphovascular invasion, perineural invasion, and lymphangitis), stenotic lesion, emergency surgery, and palliative surgery. The independent association between the pandemic period and the outcomes was assessed using multivariate random-effects logistic regression, with hospital as the cluster variable.ResultsA total of 17 938 patients (10 007 men [55.8%]; mean [SD] age, 70.6 [12.2] years) underwent surgery for colorectal cancer: 7796 (43.5%) during the pandemic period and 10 142 (56.5%) during the prepandemic period. Logistic regression indicated that the pandemic period was significantly associated with an increased rate of advanced-stage colorectal cancer (odds ratio [OR], 1.07; 95% CI, 1.01-1.13; P = .03), aggressive biology (OR, 1.32; 95% CI, 1.15-1.53; P P = .03).Conclusions and RelevanceThis cohort study suggests a significant association between the SARS-CoV-2 pandemic and the risk of a more advanced oncologic stage at diagnosis among patients undergoing surgery for colorectal cancer and might indicate a potential reduction of survival for these patients.

Abstract

Schizophrenia (SCZ) is a mental illness characterized by aberrant synaptic plasticity and connectivity. A large bulk of evidence suggests genetic and functional links between postsynaptic abnormalities and SCZ. Here, we performed quantitative PCR and Western blotting analysis in the dorsolateral prefrontal cortex (DLPFC) and hippocampus of SCZ patients to investigate the mRNA and protein expression of three key spine shapers: the actin-binding protein cyclase-associated protein 2 (CAP2), the sheddase a disintegrin and metalloproteinase 10 (ADAM10), and the synapse-associated protein 97 (SAP97). Our analysis of the SCZ post-mortem brain indicated increased DLG1 mRNA in DLPFC and decreased CAP2 mRNA in the hippocampus of SCZ patients, compared to non-psychiatric control subjects, while the ADAM10 transcript was unaffected. Conversely, no differences in CAP2, SAP97, and ADAM10 protein levels were detected between SCZ and control individuals in both brain regions. To assess whether DLG1 and CAP2 transcript alterations were selective for SCZ, we also measured their expression in the superior frontal gyrus of patients affected by neurodegenerative disorders, like Parkinson’s and Alzheimer’s disease. Interestingly, also in Parkinson’s disease patients, we found a selective reduction of CAP2 mRNA levels relative to controls but unaltered protein levels. Taken together, we reported for the first time altered CAP2 expression in the brain of patients with psychiatric and neurological disorders, thus suggesting that aberrant expression of this gene may contribute to synaptic dysfunction in these neuropathologies.

Abstract

The transmission of constant envelope orthogonal frequency division multiplexing (CE-OFDM) signals is experimentally demonstrated in this paper to increase the reach of metro-regional optical links. A recirculating fiber loop was implemented for this aim. A reach extension of 100 km of standard single-mode fiber was achieved with the CE-OFDM-based system, considering 16 quadrature amplitude modulation (16-QAM) as subcarrier mapping and links without optical dispersion compensation. Link lengths as long as 700 km, with spectral efficiency of 1.89 b/s/Hz, were accomplished with performances below the forward error correction limit.

Abstract

Introduction: Video slot machines (VSMs), which are available on electronic gaming machines and online platforms, are widely recognized for their high potential to trigger harmful gambling behaviors. Yet, the psychological factors associated with VSM use among adolescents remain poorly investigated. Methods: We estimated adjusted odds ratios (OR) and corresponding 95% confidence intervals (CI) of current regular (at least monthly) VSM use according to self-efficacy beliefs regarding everyday problem-solving ability, along with current and expected personal fulfillment at the social, and educational level among 7,959 Italian high school students aged 15–17. Results: Current regular VSM use was reported by 108 students (1.4%). Notably, this behavior was more frequently reported by those with either particularly high or low self-ratings in problem-solving ability, current and expected social fulfillment, as well as by those with lower self-ratings in both current and expected educational/work fulfillment. Conclusions: These findings underscore that during adolescence, gambling behaviors are associated with diverse psychological profiles, each entailing distinct vulnerabilities. Effectively addressing adolescent gambling necessitates a comprehensive approach, with a strong emphasis on educational support. Given the increasing challenges in curbing underage exposure and access to the most harmful gambling formats, this study offers valuable insights for the design of health promotion and prevention strategies.

Abstract

Background: In experimental models of Parkinson's disease (PD), different degrees of degeneration to the nigrostriatal pathway produce distinct profiles of synaptic alterations that depend on progressive changes in N-methyl-D-aspartate receptors (NMDAR)-mediated functions. Repetitive transcranial magnetic stimulation (rTMS) induces modifications in glutamatergic and dopaminergic systems, suggesting that it may have an impact on glutamatergic synapses modulated by dopamine neurotransmission. However, no studies have so far explored the mechanisms of rTMS effects at early stages of PD. Objectives: We tested the hypothesis that in vivo application of rTMS with intermittent theta-burst stimulation (iTBS) pattern alleviates corticostriatal dysfunctions by modulating NMDAR-dependent plasticity in a rat model of early parkinsonism. Methods: Dorsolateral striatal spiny projection neurons (SPNs) activity was studied through ex vivo whole-cell patch-clamp recordings in corticostriatal slices obtained from 6-hydroxydopamine-lesioned rats, subjected to a single session (acute) of iTBS and tested for forelimb akinesia with the stepping test. Immunohistochemical analyses were performed to analyze morphological correlates of plasticity in SPNs. Results: Acute iTBS ameliorated limb akinesia and rescued corticostriatal long-term potentiation (LTP) in SPNs of partially lesioned rats. This effect was abolished by applying a selective inhibitor of GluN2B-subunit-containing NMDAR, suggesting that iTBS treatment could be associated with an enhanced activation of specific NMDAR subunits, which are major regulators of structural plasticity during synapse development. Morphological analyses of SPNs revealed that iTBS treatment reverted dendritic spine loss inducing a prevalence of thin-elongated spines in the biocytin-filled SPNs. Abstract

Feed additives have received increasing attention as a viable means to reduce enteric emissions from ruminants, which contribute to total anthropogenic methane (CH4) emissions. The aim of this study was to investigate the efficacy of the commercial feed additive SOP STAR COW (SOP) to reduce enteric emissions from dairy cows and to assess potential impacts on milk production. Twenty cows were blocked by parity and days in milk and randomly assigned to one of two treatment groups (n = 10): supplemented with 8 g/day SOP STAR COW, and an unsupplemented control group. Enteric emissions were measured in individual head chambers over a 12-hour period, every 14 days for six weeks. SOP-treated cows over time showed a reduction in CH4 of 20.4% from day 14 to day 42 (P = 0.014), while protein % of the milk was increased (+4.9% from day 0 to day 14 (P = 0.036) and +6.5% from day 0 to day 42 (P = 0.002)). However, kg of milk protein remained similar within the SOP-treated cows over the trial period. The control and SOP-treated cows showed similar results for kg of milk fat and kg of milk protein produced per day. No differences in enteric emissions or milk parameters were detected between the control and SOP-treated cows on respective test days.

Abstract

Background: Lumbar articular fusion with the facet wedge (FW) technique is gaining increasing interest among surgeons for the treatment of vertebral instability due to its limited invasiveness and ease of use. Studies on cadavers have reported biomechanical properties similar to pedicle screws. Yet, the evidence supporting their use is still limited and moreover focused only on spinal degenerative disease. Methods: 96 cases of lumbar articular fusion with the FW techniques performed at 3 different centers between 2014 and 2022 were retrospectively analyzed based on the specific surgical indications: 1) degenerative spondylolisthesis/unstable lumbar stenosis; 2) synovial cysts; 3) adjacent segment disease (ASD). Medical records were reviewed to identify rates of complications and measures of functional outcome (ODI, low back pain VAS and modified Macnab scale) were collected both at baseline and at the follow-up visits. Wilcoxon signed-rank test was adopted to test for significant functional improvements. Results: Significative clinical improvements were observed from baseline to follow-up regarding ODI and VAS scores. Overall rate of moderate and severe complications (according to Landriel–Ibañez scale) was 7.9%. Only 3.4% of patients with degenerative disease developed ASD requiring reoperations. Only one case of radicular deficit and one of device mobilization were reported. 2/4 cases of synovial cysts treated with unilateral fusions developed contralateral complications. 9 out of 16 (56.25%) patients who underwent long-term postoperative CT scans presented adequate degree of articular fusion. Conclusion: FW technique is easy, safe, and effective. Its low rate of complications justifies its use for cases of mild lumbar instability.

Abstract

This study validated the utilization of triacylglycerides (TAGs) by Pseudomonas mendocina CH50, a wild type strain, resulting in the production of novel mcl-PHAs with unique physical properties. A PHA yield of 58% dcw was obtained using 20g/L of coconut oil. Chemical and structural characterisation confirmed that the mcl-PHA produced was a terpolymer comprising of three different repeating monomer units, 3-hydroxyoctanoate, 3-hydroxydecanoate and 3-hydroxydodecanoate or P(3HO-3HD-3HDD). Bearing in mind the potential of P(3HO-3HD-3HDD) in biomedical research, especially in neural tissue engineering, in vitro biocompatibility studies were carried out using NG108-15 (neuronal) cells. Cell viability data confirmed that P(3HO-3HD-3HDD) supported the attachment and proliferation of NG108-15 and was therefore, confirmed to be biocompatible in nature and suitable for neural regeneration.

Abstract

This study demonstrated the utilisation of unprocessed biodiesel waste as a carbon feedstock for Pseudomonas mendocina CH50, for the production of PHAs. A PHA yield of 39.5% CDM was obtained using 5% (v/v) biodiesel waste substrate. Chemical analysis confirmed that the polymer produced was poly(3-hydroxyhexanoate-co-3-hydroxyoctanoate-co-3- hydroxydecanoate-co-3-hydroxydodecanoate) or P(3HHx-3HO-3HD-3HDD). P(3HHx-3HO- 3HD-3HDD) was further characterised and evaluated for its use as a tissue engineering scaffold (TES). This study demonstrated that P(3HHx-3HO-3HD-3HDD) was biocompatible with the C2C12 (myoblast) cell line. In fact, the % cell proliferation of C2C12 on the P(3HHx-3HO-3HD-3HDD) scaffold was 72% higher than the standard tissue culture plastic confirming that this novel PHA was indeed a promising new material for soft tissue engineering.

Abstract

Recent studies support the therapeutic utility of repetitive transcranial magnetic stimulation in Parkinson's disease (PD), whose progression is correlated with loss of corticostriatal long-term potentiation and long-term depression. Glial cell activation is also a feature of PD that is gaining increasing attention in the field because astrocytes play a role in chronic neuroinflammatory responses but are also able to manage dopamine (DA) levels. METHODS: Intermittent theta-burst stimulation protocol was applied to study the effect of therapeutic neuromodulation on striatal DA levels measured by means of in vivo microdialysis in 6-hydroxydopamine-hemilesioned rats. Effects on corticostriatal synaptic plasticity were studied through in vitro intracellular and whole-cell patch clamp recordings while stepping test and CatWalk were used to test motor behavior. Immunohistochemical analyses were performed to analyze morphological changes in neurons and glial cells. RESULTS: Acute theta-burst stimulation induced an increase in striatal DA levels in hemiparkinsonian rats, 80 minutes post-treatment, correlated with full recovery of plasticity and amelioration of motor performances. With the same timing, immediate early gene activation was restricted to striatal spiny neurons. Intense astrocytic and microglial responses were also significantly reduced 80 minutes following theta-burst stimulation. CONCLUSION: Taken together, these results provide a first glimpse on physiological adaptations that occur in the parkinsonian striatum following intermittent theta-burst stimulation and may help to disclose the real potential of this technique in treating PD and preventing DA replacement therapy-associated disturbances. © 2017 International Parkinson and Movement Disorder Society.

Abstract

BackgroundTo date, there are no large studies delineating the clinical correlates of “pure” essential tremor (ET) according to its new definition.MethodsFrom the ITAlian tremor Network (TITAN) database, we extracted data from patients with a diagnosis of “pure” ET and excluded those with other tremor classifications, including ET-plus, focal, and task-specific tremor, which were formerly considered parts of the ET spectrum.ResultsOut of 653 subjects recruited in the TITAN study by January 2022, the data of 208 (31.8%) “pure” ET patients (86M/122F) were analyzed. The distribution of age at onset was found to be bimodal. The proportion of familial cases by the age-at-onset class of 20 years showed significant differences, with sporadic cases representing the large majority of the class with an age at onset above 60 years. Patients with a positive family history of tremor had a younger onset and were more likely to have leg involvement than sporadic patients despite a similar disease duration. Early-onset and late-onset cases were different in terms of tremor distribution at onset and tremor severity, likely as a function of longer disease duration, yet without differences in terms of quality of life, which suggests a relatively benign progression. Treatment patterns and outcomes revealed that up to 40% of the sample was unsatisfied with the current pharmacological options.DiscussionThe findings reported in the study provide new insights, especially with regard to a possible inversed sex distribution, and to the genetic backgrounds of “pure” ET, given that familial cases were evenly distributed across age-at-onset classes of 20 years. Deep clinical profiling of “pure” ET, for instance, according to age at onset, might increase the clinical value of this syndrome in identifying pathogenetic hypotheses and therapeutic strategies.

IntroductionIn the study of therapeutic strategies for the treatment of cancer, eco-evolutionary dynamics are of particular interest, since characteristics of the tumor population, interaction with the environment and effects of the treatment, influence the geometric and epigenetic characterization of the tumor with direct consequences on the efficacy of the therapy and possible relapses. In particular, when considering radiotherapy, oxygen concentration plays a central role both in determining the effectiveness of the treatment and the selective pressure due to hypoxia.MethodsWe propose a mathematical model, settled in the framework of epigenetically structured population dynamics and formulated in terms of systems of coupled non-linear integro-differential equations that aims to catch these phenomena and to provide a predictive tool for the tumor mass evolution and therapeutic effects.ResultsThe outcomes of the simulations show how the model is able to explain the impact of environmental selection and therapies on the evolution of the mass, motivating observed dynamics such as relapses and therapeutic failures.DiscussionThis novel modeling framework, together with the experimental results obtained so far, offers a first hint for the development of therapies which can be adapted to overcome problems of resistance and relapses. Further studies, based on a quantification of medical data, could include the development of a mathematical tool for medical support in optimizing therapeutic protocols.

En este artículo se estudia el tratamiento del mito de Cleopatra en cinco piezas del siglo XVII, cuatro españolas y una italiana. Los elementos considerados para el análisis son la proverbial belleza de la reina egipcia, por un lado, y su suicido, por el otro. Las formas de plasmarlos proporcionan las pautas para entender el valor dado a la figura histórica y para corroborar el proceso de «redención» sobre los escenarios. De las obras, quizás sea “El mayor monstruo del mundo” de Calderón la menos relevante, porque la historia de Cleopatra no es el núcleo dramático principal. “Los áspides de Cleopatra” de Rojas Zorrilla, “Marco Antonio y Cleopatra” (obra conservada en una suelta que la atribuye a Calderón) y “Los tres señores del mundo” de Belmonte Bermúdez, en cambio, dramatizan la historia haciendo diferente hincapié en la belleza de la reina pero coincidiendo en enaltecer su sacrificio final como ejemplo extremo de amor, un triunfo en toda regla. El punto álgido del rescate teatral de la reina egipcia se realiza en “La Cleopatra” de Giovanni Delfino, cuya belleza se transforma en atributo moral y cuya nobleza se expresa plenamente en la manera de afrontar la muerte. This paper analyses the treatment of Cleopatra’s myth in five plays —four Spanish and one Italian— of the seventeenth century. The elements used for my analysis are the proverbial beauty of the Egyptian Queen and her suicide. The ways of expressing them provide guidelines to understand the significance of this historic figure and to verify her redemption, a process then in progress on the stage. They are Calderón’s “El mayor monstruo del mundo”, “Marco Antonio y Cleopatra” (a play printed in one suelta attributed to Calderón), Rojas Zorrilla’s “Los áspides de Cleopatra”, Belmonte Bermúdez’s “Los tres señores del mundo” and Giovanni Delfino’s “La Cleopatra”. Of the five, perhaps Calderón’s is the least relevant, because in it (unlike the other four) Cleopatra’s story is not the central dramatic point. All others have dramatized it with special emphasis on the beauty of the queen but also highlighting her final sacrifice as an extreme example of love —a clear triumph. The apex in this process of redemption the Egyptian queen is Delfino’s La Cleopatra, where her beauty becomes a moral attribute and her nobility becomes evident in the manner in which she faces death.

Abstract

International audience; A disintegrin and metalloproteinase 10 (ADAM10) is the major α-secretase that catalyzes the amyloid precursor protein (APP) ectodomain shedding in the brain and prevents amyloid formation. Its activity depends on correct intracellular trafficking and on synaptic membrane insertion. Here, we describe that in hippocampal neurons the synapse-associated protein-97 (SAP97), an excitatory synapse scaffolding element, governs ADAM10 trafficking from dendritic Golgi outposts to synaptic membranes. This process is mediated by a previously uncharacterized protein kinase C phosphosite in SAP97 SRC homology 3 domain that modulates SAP97 association with ADAM10. Such mechanism is essential for ADAM10 trafficking from the Golgi outposts to the synapse, but does not affect ADAM10 transport from the endoplasmic reticulum. Notably, this process is altered in Alzheimer's disease brains. These results help in understanding the mechanism responsible for the modulation of ADAM10 intracellular path, and can constitute an innovative therapeutic strategy to finely tune ADAM10 shedding activity towards APP.

Abstract

ABSTRACT BACKGROUND: Although an association has been made between coronavirus disease 2019 (COVID-19) and microvascular disease, data on vascular complications (other than venous thromboembolism) are sparse. OBJECTIVE: To investigate the vascular complications in severely ill patients hospitalized with COVID-19 and their association with all-cause mortality. DESIGN AND SETTING: This cohort study was conducted at the Universidade Federal de São Paulo, Brazil. METHODS: All 305 consecutive patients diagnosed with COVID-19 and hospitalized in the intensive care unit (ICU) of a tertiary university hospital from April 2 to July 17, 2021, were included and followed up for 30 days. RESULTS: Of these, 193 (63.3%) were male, and the mean age was 59.9 years (standard deviation = 14.34). The mortality rate was 56.3% (172 patients), and 72 (23.6%) patients developed at least one vascular complication during the follow-up period. Vascular complications were more prevalent in the non-survivors (28.5%) than in the survivors (17.3%) group and included disseminated intravascular coagulation (DIC, 10.8%), deep vein thrombosis (8.2%), acrocyanosis (7.5%), and necrosis of the extremities (2%). DIC (adjusted odds ratio (aOR) 2.30, 95% confidence interval (CI) 1.01–5.24, P = 0.046) and acrocyanosis (aOR 5.21, 95% CI 1.48–18.27, P = 0.009) were significantly more prevalent in the non-survivors than in the survivors group. CONCLUSION: Vascular complications in critically ill COVID-19 patients are common (23.6%) and can be closely related to the mortality rate (56.3%) until 30 days after ICU admission. Abstract

IntroductionFunctional Motor Disorders (FMDs) represent nosological entities with no clear phenotypic characterization, especially in patients with multiple (combined FMDs) motor manifestations. A data-driven approach using cluster analysis of clinical data has been proposed as an analytic method to obtain non-hierarchical unbiased classifications. The study aimed to identify clinical subtypes of combined FMDs using a data-driven approach to overcome possible limits related to “a priori” classifications and clinical overlapping.MethodsData were obtained by the Italian Registry of Functional Motor Disorders. Patients identified with multiple or “combined” FMDs by standardized clinical assessments were selected to be analyzed. Non-hierarchical cluster analysis was performed based on FMDs phenomenology. Multivariate analysis was then performed after adjustment for principal confounding variables.ResultsFrom a study population of n = 410 subjects with FMDs, we selected n = 188 subjects [women: 133 (70.7%); age: 47.9 ± 14.4 years; disease duration: 6.4 ± 7.7 years] presenting combined FMDs to be analyzed. Based on motor phenotype, two independent clusters were identified: Cluster C1 (n = 82; 43.6%) and Cluster C2 (n = 106; 56.4%). Cluster C1 was characterized by functional tremor plus parkinsonism as the main clinical phenotype. Cluster C2 mainly included subjects with functional weakness. Cluster C1 included older subjects suffering from anxiety who were more treated with botulinum toxin and antiepileptics. Cluster C2 included younger subjects referring to different associated symptoms, such as pain, headache, and visual disturbances, who were more treated with antidepressants.ConclusionUsing a data-driven approach of clinical data from the Italian registry, we differentiated clinical subtypes among combined FMDs to be validated by prospective studies.

Abstract

This study was designed to monitor the biochemical profiles of serum and follicular fluid (FF) of postpartum dairy cows during the summer (n = 30) and winter (n = 30). Blood and FF (follicles ≥ 9 mm) were obtained from Girolando cows at 30, 45, 60, 75 and 90 days postpartum. The samples were collected and analysed to determine glucose, total cholesterol (TC), triglyceride (TG), urea, sodium (Na), potassium (K) and calcium (Ca) levels. Throughout the study, the following clinical variables were measured: rectal temperature (RT), respiratory rate (RR) and body condition score (BCS). In addition, the temperature humidity index (THI) was calculated for each season. Abstract

The integral membrane protein Thlaspi goesingense metal tolerance protein 1 (TgMTP1) has been suggested to play an important role in Zn hyperaccumulation in T. goesingense. Here, we show that the TgMTP1 protein is accumulated to high levels at the vacuolar membrane in shoot tissue of T. goesingense. TgMTP1 is likely to act in the transport of Zn into the vacuole, enhancing both Zn accumulation and tolerance. By specifically expressing TgMTP1 in Arabidopsis thaliana shoots, we show that TgMTP1, localized at the vacuolar membrane, can drive the enhanced shoot accumulation of Zn by initiating a systemic Zn deficiency response. The systematic response includes increased expression of Zn transporters (ZIP3, ZIP4, ZIP5 and ZIP9) in both shoot and root tissue. Furthermore, shoot-specific accumulation of TgMTP1 at the vacuolar membrane also leads to increased resistance to Zn in A. thaliana, probably through enhanced Zn compartmentalization in the vacuole. Such evidence leads to the conclusion that the high levels of TgMTP1 at the vacuolar membrane in shoot tissue of the Zn hyperaccumulator T. goesingense play a role in both Zn tolerance and enhanced Zn uptake and accumulation, via the activation of a systemic Zn deficiency response.

Abstract

Introduction Congenital heart disease (CHD) comprises the anatomic malformations that jeopardise the structure and function of the heart. It can be extremely complex and serious, corresponding to 30% of all deaths in the first month of life. The surgical approach for adequate treatment requires postoperative mechanical ventilation. The most critical decision related to the postoperative management of patients submitted to cardiac surgery is the right time for extubation, especially because not only abrupt or inadequate discontinuation of ventilatory support can lead to clinical decline and necessity of reintubation but also extended time of mechanical ventilation, which can lead to complications, such as pneumonia, atelectasis, diaphragm hypertrophy, and increasing morbidity and mortality.Methods and analysis This systematic review plans to include individual parallel, cross-over and cluster randomised controlled trials regarding any breathing trial test to predict extubation success in children submitted to cardiac surgery due to CHD. Studies with paediatric patients submitted to cardiac surgery for congenital cardiopathy repair, attended at a critical care unit, and under mechanical ventilatory support will be included. The main outcomes analysed will be success of extubation, reduction of pulmonary complications and time reduction of mechanical ventilation.Ethics and dissemination We will not treat patients directly; therefore, ethics committee approval was not necessary because it is not a primary study. We expect that this study may improve healthcare and medical assistance, helping healthcare professionals with routine daily decisions regarding the correct time for extubation.PROSPERO registration number CRD42021223999.
