High dose methotrexate (HDMTX) is an essential agent in chemotherapeutic regimens used in various hematological malignancies in Egyptian adults. The research for the impact of gene polymorphism on HDMTX induced toxicities and delayed elimination is an important ongoing objective in many studies, variable and conflicting results produced in the past years to clarify that impact. This study aimed to investigate the role of ABCB1 3435 C T rs1045642 and MTHFR 677 C T rs1801133 polymorphisms on HDMTX induced toxicity outcomes and delayed elimination in Egyptian adult patients with hematological malignancies. A prospective, observational cohort study was conducted on a total of 62 Egyptian adult patients with hematological malignancies age ≥ 18-years-old. All demographic, medical, and laboratory data were continuously collected from the patients' medical files in an up-to-date follow-up in selected clinics during the period from April 2018 to March 2020. Venous blood samples were collected for the purpose of genotyping, DNA extraction, and measurement of MTX levels. All the relevant data were statistically analyzed. The studied patients' median age was 25 years old with a range of (18-62) years. Forty-six patients were males with about 74%, and 16 were females with about 26%. Eighty-nine percent of the patients diagnosed with acute lymphoblastic leukemia 'ALL', 5% of the patients had B cell non-hodgkin lymphoma 'B-NHL' and 3% diagnosed with primary central nervous system lymphoma 'PCNSL' and Burkitt's lymphoma 'BL' Hematological, hepatic, renal and gastrointestinal toxicities observed post-HDMTX were recorded with the hematological toxicities toping on all the others, also patients with delayed elimination at 72 hours post the HDMTX dose were determined. Statistical analysis revealed a significant association between ABCB1 3435 C T rs1045642 and HDMTX delayed elimination with about 10 times higher risk among the minor allele 'T' carriers (

Mosquitoes can act as vectors for many disease-causing viruses and parasites through carrying these organisms from person to person. The objective of this study was to evaluate the susceptibility of Aedes aegypti larvae to some insecticides in Jeddah Governorate. Mosquito larvae were collected from all the available breeding sites in Jeddah municipalities using standard larval collection kits. The diagnostic doses (LC50 and LC90) were calculated from the mortality and concentration data. The larvicides Pirimiphos methyl, Fenitrothion, Bifenthrin and Cypermethrin were tested against the larvae of Ae. aegypti mosquito. The results showed positive correlation between the concentrations under investigation and mortality% of the treated larvae. Cypermethrin gave strong effect against Ae. aegypti larvae (LD50 0.0015 ml/L) followed by Pirimiphos methyl (LD50 0.0020 ml/L), Bifenthrin (LD50 0.039 ml/L), and at last rank Fenitrothion (0.041 ml/L). The same trend was obtained in LD90s. Ae. aegypti in Jeddah was susceptible to Bifenthrin, Cypermethrin, Pirimiphos methyl and Fenitrothion. Further studies should be run to conduct an intensive and appropriate vector control program in Jeddah Governorate.

Background: Lead contamination has turned into a major concern as it serves no useful purpose in the human body. Its presence in the body can lead to toxic effects on different organs. Aim: The purpose of the present study is to examine the efficacy of selenium in mitigating leadinduced oxidative stress and kidney injury in male albino Wistar rats. Further to study the underlying molecular mechanism. Methods: We hypothesized that selenium protect against lead nephrotoxicity by testing various parameters of kidney function in male rats treated for 28 days with o.5 mg/kg selenium versus control male rats that received a vehicle. Results: The results showed an increase of serum urea, creatinine, uric acid and urinary albumin in rats intoxicated with lead compared to control group. The increase of these parameters would indicate renal toxicity which is confirmed by histopathological changes. Lead intoxicated rats had significant increment in apoptosis as well as phosphorylated PERK, eIF2α and apoptotic proteins as CHOP, caspase 3 and 8 in the kidneys, which were attenuated by selenium treatment. Conclusion: This study clearly demonstrated that activation of PERK- eIF2α -CHOP signaling pathway was involved in lead induced nephrotoxicity in rats and selenium inhibited lead nephrotoxicity partly through suppression of PERK- eIF2α CHOP pathway.

AIM: This study aims to evaluate the accuracy and reliability of Kinect motion sensing input device’s three-dimensional (3D) models by comparing it with direct anthropometry and digital 2D photogrammetry. MATERIALS AND METHODS: Six profiles and four frontal parameters were directly measured on the faces of 80 participants. The same measurements were repeated using two-dimensional (2D) photogrammetry and (3D) images obtained from Kinect device. Another observer made the same measurements for 30% of the images obtained with 3D technique, and interobserver reproducibility was evaluated for 3D images. Intraobserver reproducibility was evaluated. Statistical analysis was conducted using the paired samples t-test, interclass correlation coefficient, and Bland-Altman limits of agreement. RESULTS: The highest mean difference was 0.0084 mm between direct measurement and photogrammetry, 0.027 mm between direct measurement and 3D Kinect’s models, and 0.018 mm between photogrammetry and 3D Kinect’s. The lowest agreement value was 0.016 in the all parameter between the photogrammetry and 3D Kinect’s methods. Agreement between the two observers varied from 0.999 Sn-Me to 1 with the rest of linear measurements. CONCLUSION: Measurements done using 3D Images obtained from Kinect device indicate that it may be an accurate and reliable imaging method for use in orthodontics. It also provides an easy low-cost 3D imaging technique that has become increasingly popular in clinical settings, offering advantages for surgical planning and outcome evaluation.

Hepatocellular carcinoma (HCC) is a major cause of cancer death in Egypt. There is still a risk for HCC development even after eradicating hepatitis C virus (HCV) by direct-acting antivirals (DAAs). Chitinase-3-like-protein-1 (CHI3L1), a biomarker for predicting many diseases, plays an essential role in inflammation, angiogenesis, and antiapoptosis. Tolloid-like protein 1 (TLL1) may be involved in hepatic fibrogenesis and carcinogenesis. This study aimed to determine the role and combined effect of CHI3L1 (rs880633), TLL1 (rs1503298), and an intergenic (rs597533) polymorphisms on the risk of developing HCC in Egyptian patients after achieving sustained virological response (SVR) by DAAs. Blood samples were collected from 68 HCC patients, 77 non-HCC subjects, and 80 healthy controls. The DNA was extracted and analyzed for rs880633, rs1503298, and rs597533 using Genotyping TaqMan™ assay. The result of the present study showed a significant difference in genotypes and alleles frequencies in both (rs880633) and (rs597533) in HCC group as compared to healthy control and also as compared to the non-HCC group. However, regarding to (rs1503298) genotypes and alleles between the HCC and non-HCC groups, there were no significant differences. Combined polymorphism in more than one gene simultaneously showed a higher risk to HCC after SVR than an individual locus. Both allelic and genotypic variations of the CHI3L1 gene (rs880633) and an intergenic (rs597533) seemed to be significant predictors confirming a great risk for HCC susceptibility in Egyptian patients achieved SVR. Patients with a polymorphism in more than one gene showed an increased risk to HCC after SVR rather than individual locus.

Novel series of pyrazoline carbothioamides, acetyl pyrazoles, pyridine-3-carbonitriles, pyridine-2-carbonitriles, and nicotinonitriles were synthesized. The structures of the newly synthesized compounds were established based on their spectral data, elemental analyses and alternative synthetic routes whenever possible. Also, the newly synthesized compounds were screened for their in vitro anticancer activity against T-47D (breast cancer human cell line) and UACC-257 (melanoma human cell line) by MTT assay (a colorimetric assay for assessing cell metabolic activity). Experimental results demonstrated that compounds 3a, 4a, and 8c showed excellent in vitro activity against T-47D cell line, while compounds 3d, 6d, and 5b exhibited excellent activity against UACC-257 cell line. Molecular docking studies were performed on compound 4a, as it showed the most promising activity against T-47D cell line, using Molecular Operation Environment (MOE 2008.10) software. Also, drug-likeness, pharmacokinetics, and toxicity assessment studies were carried out.

Background Pyrazolines show different biological activities. In recent years, interest in the chemistry of hydrazonoyl halides has been renewed. 1,3,4-Thiadiazoles are one of the most common heterocyclic pharmacophores with a wide range of biological activities. Results Ethyl 2-(5-(furan-2-yl)-3-(p-tolyl)-4,5-dihydro-1H-pyrazol-1-yl)-4-methyl-thiazole-5-carboxylate, 2-(5-(furan-2-yl)-3-(p-tolyl)-4,5-dihydro-1H-pyrazol-1-yl)thiazol-4(5H)-one, and 1-(2-(5-(furan-2-yl)-3-(p-tolyl)-4,5-dihydro-1H-pyrazol-1-yl)-4-methylthiazol-5-yl)ethan-1-one were synthesized from the reaction of 5-(furan-2-yl)-3-(p-tolyl)-4,5-dihydro-1H-pyrazole-1-carbothioamide with different halogenated compounds. Thiazole, 1,3,4-thiadiazole and pyrano[2,3-d]thiazole derivatives were also synthesized. The structures of the newly synthesized compounds were elucidated based on elemental analysis, spectral data, and alternative synthetic routes whenever possible. Additionally, the newly synthesized compounds were screened for antimicrobial activity against various microorganisms. Conclusions A new series of novel functionalized 1,3,4-thiadiazoles, 1,3-thiazoles, and pyrazoline-containing moieties were synthesized using hydrazonoyl halides as precursors and evaluated for their in vitro antibacterial, and antifungal activities. The antimicrobial results of the examined compounds revealed promising results and some derivatives have activities similar to the references used.
