8. Salinity Stress Acclimation Strategies in Chlamydomonas sp. Revealed by Physiological, Morphological and Transcriptomic Approaches.

Abstract

Climate changes may include variations in salinity concentrations at sea by changing ocean dynamics. These variations may be especially challenging for marine photosynthetic organisms, affecting their growth and distribution. Chlamydomonas spp. are ubiquitous and are often found in extreme salinity conditions. For this reason, they are considered good model species to study salinity adaptation strategies. In the current study, we used an integrated approach to study the Chlamydomonas sp. CCMP225 response to salinities of 20‰ and 70‰, by combining physiological, morphological, and transcriptomic analyses, and comparing differentially expressed genes in the exponential and stationary growth phases under the two salinity conditions. The results showed that the strain is able to grow under all tested salinity conditions and maintains a surprisingly high photosynthetic efficiency even under high salinities. However, at the highest salinity condition, the cells lose their flagella. The transcriptomic analysis highlighted the up- or down-regulation of specific gene categories, helping to identify key genes responding to salinity stress. Overall, the findings may be of interest to the marine biology, ecology, and biotechnology communities, to better understand species adaptation mechanisms under possible global change scenarios and the potential activation of enzymes involved in the synthesis of bioactive molecules. Abstract

Saltworks are interesting sites for aquatic ecology, where marine phytoplankton plays a fundamental role in regulating ecosystem processes and sustaining large-scale biodiversity. This paper presents successional, structural, and dynamical traits of a phytoplankton metacommunity in a disused saltwork, where the industrial hydraulic management has been changed, leading to a different overlapping of natural and anthropic induced scales. We have considered the total phytoplankton biomass, size classes, and chemotaxonomical composition in monitoring the metacommunity dynamics in a series of single ponds spanning from marine to hypersaline ones between July 2008 and October 2009. Results indicate a large accumulation of biomass during summer to early autumn in hypersaline ponds, with the prevalence of small-size cells (<20 μm) in the entire system. Despite severe salinity fluctuations as a consequence of rainy events and evaporating processes, chlorophytes represented the dominant group of hypersaline ponds, while diatoms were more abundant where the influence of seawater inflow was higher. Despite the absence of ordinary maintenance, the phytoplankton metacommunity dynamic and patterns observed suggest that the existing saline gradient ensures qualitative and quantitative aspects of the primary production usually found in active saltworks. This makes the Saline di Tarquinia an important site not only for the preservation of Mediterranean biodiversity and hypersaline biotopes but also for ecological studies dealing with the trait-based responses of the phytoplankton community to large environmental fluctuations. However, the number of studies reporting the simultaneous diversity of the phytoplankton and bacterioplankton in Antarctic waters are limited. Here, we report data on the bacterial diversity in relation to phytoplankton community structure in the surface waters of the Ross Sea during the Austral summer 2017. Our results show partially overlapping bacterioplankton communities between the stations located in the Terra Nova Bay (TNB) coastal waters and the Ross Sea Open Waters (RSOWs), with a dominance of members belonging to the bacterial phyla Bacteroidetes and Proteobacteria. In the TNB coastal area, microbial communities were characterized by a higher abundance of sequences related to heterotrophic bacterial genera such as Polaribacter spp., together with higher phytoplankton biomass and higher relative abundance of diatoms. On the contrary, the phytoplankton biomass in the RSOW were lower, with relatively higher contribution of haptophytes and a higher abundance of sequences related to oligotrophic and mixothrophic bacterial groups like the Oligotrophic Marine Gammaproteobacteria (OMG) group and SAR11. We show that the rate of diversity change between the two locations is influenced by both abiotic (salinity and the nitrogen to phosphorus ratio) and biotic (phytoplankton community structure) factors. 29. Phytoplankton biomass and main functional groups of the Sele river mouth (Southern Italian Tyrrhenian coast) in a severe drought period

Abstract

The phytoplankton community structure of the terminal stretches of the Sele river, one of the most important rivers of the southern Tyrrhenian coast of Italy, was investigated. Main functional groups and total biomass were determined from samples collected in proximity of drainage channels and under a period of severe drought. The overall sampling area was characterized by a strong vertical stratification driven by intrusion of seawater, with large hypoxic or anoxic areas near the bottom. High concentrations of Chl-a up to 95.73 μg/l were observed in the surface water layer (0.1-1.0 m depth) characterized by oxygen oversaturated waters. Cryptophytes and diatoms were the dominant groups, but high variability of minor groups was observed among stations. Abstract

As the annual expanse of Arctic summer ice‐cover steadily decreases, concomitant biogeochemical and ecological changes in this region are likely to occur. Because the Central Arctic Ocean is often nutrient and light limited, it is essential to understand how environmental changes will affect productivity, phytoplankton species composition, and ensuing changes in biogeochemistry in the region. During the transition from late summer to early autumn, water column sampling of various biogeochemical parameters was conducted along an ice‐floe drift station near the North Pole. Our results show that as the upper water column stratification weakened during the late summer–early autumn transition, nutrient concentrations, particulate dimethylsulfoniopropionate (DMSPp) levels, photosynthetic efficiency, and biological productivity, as estimated by ΔO2/Ar ratios, all decreased. Chemotaxonomic (CHEMTAX) analysis of phytoplankton pigments revealed a taxonomically diverse picoautotrophic community, with chlorophyll (Chl) c3‐containing flagellates and the prasinophyte, Pyramimonas spp., as the most abundant groups, comprising ~ 30% and 20% of the total Chl a (TChl a) biomass, respectively. In contrast to previous studies, the picoprasinophyte, Micromonas spp., represented only 5% to 10% of the TChl a biomass. Of the nine taxonomic groups identified, DMSPp was most closely associated with Pyramimonas spp., a Chl b‐containing species not usually considered a high DMSP producer. As the extent and duration of open, ice‐free waters in the Central Arctic Ocean progressively increases, we suggest that enhanced light transmission could potentially expand the ecological niche of Pyramimonas spp. in the region.

Abstract

We analyse primary production processes during austral summer 1996 and 2001 in different environmental conditions such as ice-free waters and extensive ice-covered areas. Spatio-temporal distribution of phytoplankton biomass and functional groups along with photosynthetic parameters are presented. Production vs irradiance (PvsE) experiments were performed using 14C incubations at several stations and three or four different depths to define the eco-physiology of phytoplankton communities. The results of the oceanographic campaign conducted in ice-free waters of the Ross Sea (summer 1996) emphasize that these ecosystems are characterized by high nutrient low chlorophyll (HNLC) conditions due to limiting factors (eg. Fe). Conversely, the results of the oceanographic cruise in extensively packice-covered areas (summer 2001) indicate that the average phytoplankton biomass (estimated from Chla) was about three times the values recorded in ice-free conditions, but the primary production was relatively lower. In fact, in situ primary production and PvsE experiments over few days show that high primary production values occurred in most of the area, but only within the first five meters of the water column and the melting pack ice. Notwithstanding some high values of phytoplankton biomass during the 2001 campaign, water column stability, similar irradiance levels along the water column, photosynthetic capacity was suppressed in deeper layers, indicating a low carrying capacity of the pelagic ecosystem due to iron limitation, as shown by low values of the photo-chemical efficiency of photosystem II (PSII), variable fluorescence and maximal fluorescence ratio (Fv/Fm). In contrast with a very high variability in phytoplankton biomass at several temporal and spatial scales, photosynthetic parameters (PB max , α, Ek) varied within narrow ranges. Relevant changes in phytoplankton abundance and species composition are reported in this study although the environmental factors that drive these changes in primary production processes and prevalence of principal functional groups of phytoplankton communities compared to the past (1996 and 2001) are still unknown. The effect of these changes on the carrying capacity of Ross Sea ecosystems, carbon export and the potentially new asset of the food web will need to be determined.

47. Thermo resistant antioxidants from photoautotrophic microorganisms: screening and characterization.

Abstract

The demand for natural antioxidants to be used in food industry is increasing, as synthetic antioxidants are toxic and have high production costs. Specifically, food processing and preservation require antioxidants resistant to thermal sterilization processes. In this study, twenty-five strains among microalgae and cyanobacteria were screened as antioxidants producers. The species Enallax sp., Synechococcus bigranulatus and Galdieria sulphuraria showed the highest content of chlorophyll a and total carotenoids. In vitro stability and antioxidant activity of the ethanolic extracts were performed. The results revealed that pigments present in the extracts, obtained from the previously mentioned species, were stable at room temperature and exhibited in vitro free radical scavenging potential with IC50 values of 0.099 ± 0.001, 0.048 ± 0.001 and 0.13 ± 0.02 mg mL-1, respectively. Biocompatibility assay showed that the extracts were not toxic on immortalized cell lines. The antioxidant activity was also tested on a cell-based model by measuring intracellular ROS levels after sodium arsenite treatment. Noteworthy, extracts were able to exert the same protective effect, before and after the pasteurization process. Results clearly indicate the feasibility of obtaining biologically active and thermostable antioxidants from microalgae. Green solvents can be used to obtain thermo-resistant antioxidants from cyanobacteria and microalgae which can be used in the food industry.
