Abstract

Selenium (Se) is an essential trace element of fundamental importance for human health. Se is incorporated into selenoproteins (SPs) which are endowed with pleiotropic effects including antioxidant and anti-inflammatory effects and active production of thyroid hormones. It has also been suggested that Se plays a crucial role in the pathogenesis of various human diseases. The therapeutic effects of supplementation with Se have already been described in various thyroid diseases. However, there are still conflicting results regarding the optimal dose of Se to administer and the duration of treatment, efficacy, and safety. The highly beneficial effects of supplementation with Se have been observed in subjects with thyroid disease in the hyperthyroid phase. In line with these observations, clinical studies have shown that in patients with Basedow's disease (BD) and autoimmune thyroiditis (AT), treatment with a combination of antithyroid drugs and Se restores the euthyroid state faster than administration of antithyroid drugs alone. However, the efficacy of this therapeutic approach remains to be better evaluated.

Abstract

Citrus fruits are the main fruits of the Mediterranean diet and have been long recognized for their beneficial effects on human health. Observational studies have shown a significant association between dietary flavo-noid intake and reduced risk of cardiovascular and malignant diseases. The beneficial effects of citrus fruits on human health appear to be due to their high content in vitamins, minerals and fibers. In particular, the an-tioxidant and anti-inflammatory activities have been indicated as some of the mechanisms through which citrus fruits may thwarts the development of chronic degenerative diseases such as atherosclerosis and can-cer. This review would critically examine the results from numerous studies carried out in order assess the contribute of citrus flavonoids to the prevention of chronic pathological conditions including athero-sclerosis and cancer.

Abstract

Dark photons are hypothetical massive vector particles that could mix with ordinary photons. The simplest theoretical model is fully characterised by only two parameters: the mass of the dark photon m$_{\gamma^{\mathrm{D}}}$ and its mixing parameter with the photon, $\varepsilon$. The sensitivity of the SHiP detector is reviewed for dark photons in the mass range between 0.002 and 10 GeV. Different production mechanisms are simulated, with the dark photons decaying to pairs of visible fermions, including both leptons and quarks. Exclusion contours are presented and compared with those of past experiments. The SHiP detector is expected to have a unique sensitivity for m$_{\gamma^{\mathrm{D}}}$ ranging between 0.8 and 3.3$^{+0.2}_{-0.5}$ GeV, and $\varepsilon^2$ ranging between $10^{-11}$ and $10^{-17}$.

Abstract

We propose to build and operate a detector that, for the first time, will measure the process $pp\to\nu X$ at the LHC and search for feebly interacting particles (FIPs) in an unexplored domain. The TI18 tunnel has been identified as a suitable site to perform these measurements due to very low machine-induced background. The detector will be off-axis with respect to the ATLAS interaction point (IP1) and, given the pseudo-rapidity range accessible, the corresponding neutrinos will mostly come from charm decays: the proposed experiment will thus make the first test of the heavy flavour production in a pseudo-rapidity range that is not accessible by the current LHC detectors. In order to efficiently reconstruct neutrino interactions and identify their flavour, the detector will combine in the target region nuclear emulsion technology with scintillating fibre tracking layers and it will adopt a muon identification system based on scintillating bars that will also play the role of a hadronic calorimeter. The time of flight measurement will be achieved thanks to a dedicated timing detector. The detector will be a small-scale prototype of the scattering and neutrino detector (SND) of the SHiP experiment: the operation of this detector will provide an important test of the neutrino reconstruction in a high occupancy environment., Comment: Letter of Intent

Abstract

Background: Nowadays, thanks to the introduction of the VI-RADS scoring system, mpMRI has shown promising results in pre-TURBT assessment of muscular invasiveness of BCa, even if its application in everyday practice is still limited. This might be due to a lack in the literature about the learning curve of radiologists and about the characteristics of the exam. With the aim to reduce scan time and patient discomfort while maintaining diagnostic accuracy, bpMRI has been introduced as a possible alternative to mpMRI in this group of patients. This study reports a single-center experience using mpMRI and the VI-RADS scoring system to differentiate NMIBC from MIBC. The primary aim of the study is to assess diagnostic accuracy of mpMRI using the VI-RADS scoring system. The secondary aim is to evaluate the learning curve of an experienced mpMRI radiologist. Additionally, we perform a retrospective assessment of the same group of patients evaluating only DWIs and T2-weighted images, as they underwent bpMRI, to compare the performance of mpMRI and bpMRI. Materials and Methods: From 11/2021 to 11/2023, patients with suspected newly diagnosed BCa were enrolled in this prospective study. All patients underwent mpMRI prior to TURBT in a highly specialized radiology center for MRI. According to VI-RADS, a cutoff of ≥3 was assumed to define MIBC. Histological TURBT reports were compared with preoperative VI-RADS scores to assess the accuracy of mpMRI in discriminating between NMIBC and MIBC. Furthermore, to assess the learning curve of the reading radiologist we analyzed the rate of patients correctly classified as MIBC at MRI. Finally, we evaluated the performance of a hypothetic biparametric MRI in classifying our cohort according to VI-RADS score and compared it with mpMRI performance by using DeLong's test. Data analysis was performed using Jamovi software v.2.3 and R software v.4.2.1. Results: A total of 133 patients were enrolled. mpMRI showed sensitivity and specificity of 86% (95% confidence interval [CI]: 64–97) and 95% (95% CI: 89–98), respectively. The learning curve analysis of the reading radiologist showed that the rate of patients correctly classified as MIBC rapidly increases reaching its plateau after 40 cases. The hypothetic bpMRI showed a sensitivity of 76% (95% CI: 53–92) and a specificity of 93% (95% CI: 86–97), with no significant difference with mpMRI performance (p = 0.10). Conclusions: Our study confirms the effectiveness of MRI, particularly with the VI-RADS scoring system, in differentiating NMIBC from MIBC. The learning curve analysis underscores the importance of radiologist training in optimizing diagnostic accuracy. Future research should focus on enhancing the sensitivity of bpMRI and further validating these findings in larger and multicentric studies. [ABSTRACT FROM AUTHOR]

Abstract

A new general purpose fixed target facility is proposed at the CERN SPS accelerator which is aimed at exploring the domain of hidden particles and make measurements with tau neutrinos. Hidden particles are predicted by a large number of models beyond the Standard Model. The high intensity of the SPS 400~GeV beam allows probing a wide variety of models containing light long-lived exotic particles with masses below ${\cal O}$(10)~GeV/c$^2$, including very weakly interacting low-energy SUSY states. The experimental programme of the proposed facility is capable of being extended in the future, e.g. to include direct searches for Dark Matter and Lepton Flavour Violation., Comment: Technical Proposal

Abstract

Introduction: In this study, we employed a combined electromagnetic recording technique, i.e., electroencephalogram (EEG)/event-related potentials (ERPs) plus standardized weighted low-resolution electromagnetic tomography (swLORETA), to investigate the neural mechanism subserving the orthographic processing of symbols in language and music. While much is known about word processing, the current literature remains inconclusive regarding music reading, as its mechanisms appear to be left lateralized in some cases (as suggested by music-alexia clinical case reports) and either right-sided or bilateral in others, depending on the study and the methodology used. Methods: In this study, 90 right-handed participants with varying musical abilities and sexes performed an attentional selection task that involved the recognition of target letters and musical notes, while their EEG signals were recorded from 128 sites. Results: The occipito/temporal N170 component of ERPs (170–210 ms) was found strictly left-sided during letter selection and bilateral (with a right-hemispheric tendency) during note selection. Source reconstruction data indicated the preponderant engagement of the right posterior fusiform gyrus (BA19) for processing musical notes. Also involved were other brain regions belonging to the word reading circuit, including the left-sided visual word form area (VWFA) and frontal eye-fields (FEFs). Discussion: This finding provides an explanation for the infrequent appearance of musical alexia cases (previously observed only in patients with left hemispheric lesions). It also suggests how musical literacy could be a rehabilitative and preventive factor for dyslexia, by promoting neuroplasticity and bilaterality in the reading areas.

This mini-review discusses the existing evidence on various forms of humour and humour-like behaviour in non-human animals, combining ontogenetic and phylogenetic perspectives. The first section describes humour-like behaviours, from the simplest to the most complex form (from laughing, tickling, joking, and chasing to ToM humour). In the second section, we propose the SPeCies (Social, Physiological, and Cognitive) Perspective, which frames the various types of humour based on Social motivation, Physiological state, and Cognitive skills. Finally, in the third section, we discuss future directions for further development.

Abstract

Considerable evidence suggests that musical education induces structural and functional neuroplasticity in the brain. This study aimed to explore the potential impact of such changes on word-reading proficiency. We investigated whether musical training promotes the development of uncharted orthographic regions in the right hemisphere leading to better reading abilities. A total of 60 healthy, right-handed culturally matched professional musicians and controls took part in this research. They were categorised as normo-typical readers based on their reading speed (syl/sec) and subdivided into two groups of relatively good and poor readers. High density EEG/ERPs were recorded while participants engaged in a note or letter detection task. Musicians were more fluent in word, non-word and text reading tests, and faster in detecting both notes and words. They also exhibited greater N170 and P300 responses, and target-non target differences for words than controls. Similarly, good readers showed larger N170 and P300 responses than poor readers. Increased reading skills were associated to a bilateral activation of the occipito/temporal cortex, during music and word reading. Source reconstruction also showed a reduced activation of the left fusiform gyrus, and of areas devoted to attentional/ocular shifting in poor vs. good readers, and in controls vs. musicians. Data suggest that music literacy acquired early in time can shape reading circuits by promoting the specialization of a right-sided reading area, whose activity was here associated with enhanced reading proficiency. In conclusion, music literacy induces measurable neuroplastic changes in the left and right OT cortex responsible for improved word reading ability.

Abstract

Background: Mucosal Melanomas (MM) are highly aggressive neoplasms arising from mucosal melanocytes. Current treatments offer a limited survival benefit for patients with advanced MM; moreover, the lack of pre-clinical cellular systems has significantly limited the understanding of their immunobiology. Methods: Five novel cell lines were obtained from patient-derived biopsies of MM arising in the sino-nasal mucosa and designated as SN-MM1-5. The morphology, ultrastructure and melanocytic identity of SN-MM cell lines were validated by transmission electron microscopy and immunohistochemistry. Moreover, in vivo tumorigenicity of SN-MM1-5 was tested by subcutaneous injection in NOD/SCID mice. Molecular characterization of SN-MM cell lines was performed by a mass-spectrometry proteomic approach, and their sensitivity to PI3K chemical inhibitor LY294002 was validated by Akt activation, measured by pAkt(Ser473) and pAkt(Thr308) in immunoblots, and MTS assay. Results: This study reports the validation and functional characterization of five newly generated SN-MM cell lines. Compared to the normal counterpart, the proteomic profile of SN-MM is consistent with transformed melanocytes showing a heterogeneous degree of melanocytic differentiation and activation of cancer-related pathways. All SN-MM cell lines resulted tumorigenic in vivo and display recurrent structural variants according to aCGH analysis. Of relevance, the microscopic analysis of the corresponding xenotransplants allowed the identification of clusters of MITF-/CDH1-/CDH2 + /ZEB1 + /CD271 + cells, supporting the existence of melanoma-initiating cells also in MM, as confirmed in clinical samples. In vitro, SN-MM cell lines were sensitive to cisplatin, but not to temozolomide. Moreover, the proteomic analysis of SN-MM cell lines revealed that RICTOR, a subunit of mTORC2 complex, is the most significantly activated upstream regulator, suggesting a relevant role for the PI3K-Akt-mTOR pathway in these neoplasms. 43. Characteristics of Candida isolates from patients with diabetes mellitus

Abstract

The present research has investigated the molecular characterization of oral yeasts in healthy individuals from different dental clinical settings in the United Kingdom, and patients from the UK and Italy who were affected by diabetes mellitus (DM) to determine the impact of DM upon candidal infection of the mouth. In the present study of patients with DM from the UK and Italy a lower than expected incidence of oral candidal infections (7/249, 2.8%) was observed. The presence of oral yeasts and genotypic diversity of C. albicans was also not influenced by DM. There was no significant difference between the presence of oral yeasts in Italian and UK DM patients. However, more (p=0.04) C. dubliniensis isolates were found in non-DM individuals. Higher levels of Candida adhesion to fibronectin-coated paramagnetic beads were observed in isolates from DM patients. Isolates from patients with low oral yeast loads adhered significantly (p=0.0T) more than those from patients with high oral yeast loads. In general, there was no difference in proteolytic activity of isolates from DM or control patients. Candida isolates from UK DM patients had significantly greater in vitro resistance to azole antifungal agents (miconazole p<0.0001 fluconazole p=0.02 ketoconazole p=0.01) than those from Italian DM patients. In addition, C. albicans isolates from all examined patients were more susceptible to fluconazole (p=0.0008) and miconazole (p=0.01) than non-C. albicans strains. PCR fingerprinting and subsequent phylogenetic analysis revealed that C. albicans isolates from UK DM patients were the most diverse (p<0.0001) in comparison to those from Italian DM patients or from healthy subjects. Finally, it was observed that the activity of a rat IgM monoclonal anti-idiotypic antibody (mAbKlO) and a synthetic decapeptide (KP) had a significant dose- dependant fungicidal activity upon a wide spectrum of C. albicans and non-C albicans isolates from patients with and without DM.
