The biophysical and biomechanical properties of the extracellular matrix (ECM) are crucial in the processes of cell differentiation and proliferation. However, it is unclear to what extent tumor cells are influenced by biomechanical and biophysical changes of the surrounding microenvironment and how this response varies between different tumor forms, and over the course of tumor progression. The entire ensemble of genes encoding the ECM associated proteins is called matrisome. In cancer, the ECM evolves to become highly dysregulated, rigid, and fibrotic, serving both pro-tumorigenic and antitumorigenic roles. Tumor desmoplasia is characterized by a dramatic increase of α-smooth muscle actin expressing fibroblast and the deposition of hard ECM containing collagen, fibronectin, proteoglycans, and hyaluronic acid and is common in many solid tumors. In this review, we described the role of inflammation and inflammatory cytokines, in desmoplastic matrix remodeling, tumor state transition driven by microenvironment forces and the signaling pathways in mechanotransduction as potential targeted therapies, focusing on the impact of qualitative and quantitative variations of the ECM on the regulation of tumor development, hypothesizing the presence of matrisome drivers, acting alongside the cell-intrinsic oncogenic drivers, in some stages of neoplastic progression and in some tumor contexts, such as pancreatic carcinoma, breast cancer, lung cancer and mesothelioma. Cystinosis is a rare lysosomal storage disorder caused by autosomal recessive mutations in the CTNS gene that encodes for the cystine transporter cystinosin, which is expressed on the lysosomal membrane mediating the efflux of cystine. Cysteamine bitartrate is a cystine-depleting aminothiol agent approved for the treatment of cystinosis in children and adults. In this study, we developed and validated a liquid chromatography–tandem mass spectrometry (LC-MS/MS) method for the determination of cysteamine levels in plasma samples. This LC-MS/MS method was validated according to the European Medicines Agency (EMA)'s guidelines for bioanalytical method validation. An ultra-performance liquid chromatograph (UPLC) coupled with a 6470 mass spectrometry system was used for cysteamine determination. Our validated method was applied to plasma samples from n = 8 cystinosis patients (median, interquartile range (IQR) = 20.5, 8.5–26.0 years). The samples were collected before cysteamine oral administration (pre-dose) and 1 h after (post-dose). Our bioanalytical method fulfilled the regulatory guidelines for method validation. The cysteamine plasma levels in pre-dose samples were 2.57 and 1.50–3.31 μM (median and IQR, respectively), whereas the post-dose samples reported a cysteamine median concentration of 28.00 μM (IQR: 17.60–36.61). Our method allows the rapid determination of cysteamine plasma levels. This method was successfully used in cystinosis patients and, therefore, could be a useful tool for the evaluation of therapy adherence and for future pharmacokinetic (PK) studies involving a higher number of subjects. Summary: Little is known of the course of COVID‐19 and the antibody response to infection or vaccination in patients with hairy‐cell leukaemia (HCL). Among a total of 58 HCL cases we studied in these regards, 37 unvaccinated patients, mostly enjoying a relatively long period free from anti‐leukaemic treatment, developed COVID‐19 between March 2020 and December 2021 with a usually favourable outcome (fatality rate: 5/37, 14%); however, active leukaemia, older age and more comorbidities were associated with a worse course. Postinfection (n = 11 cases) and postvaccination (n = 28) seroconversion consistently developed, except after recent anti‐CD20 or venetoclax therapy, correlating with perivaccine B‐cell count. Vaccination appeared to protect from severe COVID‐19 in 11 patients with breakthrough infection. [ABSTRACT FROM AUTHOR]

The Acque Albule Basin in Tivoli (Rome, Central Italy) represents one of the largest exposures of travertine deposits in the world. A detailed study of the Upper Pleistocene Testina travertine, constituting the poorly lithified top part of the Lapis Tiburtinus travertines, is presented. Fieldwork performed in an area of 10 km2 and petrographic analysis allow the recognition of six geobodies with different geometries, degree of diagenetic alteration and architectural elements, composed of seven lithotypes. The Testina travertine appears in general as a poorly lithified travertine, even if it shows a high degree of cementation. It is mainly characterised by blocky, drusy or bladed microsparite and sparite calcite cements, related to thermal and meteoric phreatic to vadose water circulation, affecting in particular those travertine deposits located in the northern part of the study area. The Testina, neglected until now in the several studies performed in the Acque Albule Basin, shows a progradational trend from the north to the south and is characterised by environments, evidencing deposition on a gentle slope, in an alluvial plain and in a shallow lake, with gradual transitions between low‐energy environments and high‐energy environments. Furthermore, based on correlation between the different geobodies in the overall study area, the Testina travertine depositional system was influenced by tectonic activity in the northern part, by an increasing topographic gradient and water discharge flow in the central part and by flooding events related to the Aniene River in the southern part. This study aims to differentiate such influencing factors facilitating the analysis of other travertine depositional systems and also to aid in the interpretation of subsurface analogues, as in the case of South Atlantic Pre‐Salt reservoirs. Barium zirconate (BaZrO3), when substituted with trivalent acceptor ions to replace Zr4+, is a proton conducting material of interest for several electrochemical applications. The local coordination environments, and vibrational dynamics, of the protons are known to critically influence the material's proton conducting properties, however, the nature of the static and dynamic structure around the protons and, especially, how it is affected by the dopant atoms for high doping concentrations, remains to be elucidated. Here we report results from X-ray powder diffraction, infrared (IR) spectroscopy, inelastic neutron scattering (INS) and ab initio molecular dynamics (AIMD) simulations on a hydrated sample of BaZrO3 substituted with 50% In3+. The investigation of the momentum-transfer (Q) dependence of the INS spectrum is used to aid the analysis of the spectra and the assignment of the spectral components to fundamental O–H bend and O–H stretch modes and higher-order transitions. The AIMD simulations show that the INS spectrum is constituted of the overlapping spectra of protons in several different local structural environments, whereas the local proton environments for specific protons are found to vary with time as a result of thermally activated vibrations of the perovskite lattice. It is argued that, converse to more weakly doped systems, such as 20% Y-doped BaZrO3, the dopant–proton association effect does not hinder the diffusion of protons due to the presence of percolation paths of dopant atoms throughout the perovskite lattice.
