BACKGROUND: A variety of antibodies against the SARS-CoV-2 with different functional activities can lead to immune pathology and/or modulation of infection due to antibody-dependent enhancement. An important role in the development of ADE, as exemplified by Dengue fever, is played by low-affinity antibodies, which interacting with the virus, do not neutralize it, but on the contrary forms the virus-antibody complex to immune cells (monocytes/macrophages, B-cells). There are conflicting data on the antibody-dependent enhancement presence in COVID-19 coronavirus infection. Some studies indicate a relationship between the severity of infection in patients infected by SARS-СoV-2 and the level of antibodies to closely related coronaviruses. AIM: The aim of the study was to understand the possibility of interaction between antibodies to SARS-CoV-2 with closely related porcine coronavirus (alphacoronavirus) and their ability to modulate the development of infection in vitro. MATERIALS AND METHODS: In the study serums and leukocytes from patients, who are convalescent after COVID-19 infection in 2020, was used. Antibody titers (IgG/IgM) was checked by commercial kit (VektorBest, Russia), based on ELISA method. In the eukaryotic expression system Chinese hamster ovary cells, monoclonal antibody to SARS-CoV-2 spike protein and its Fab version without Fc domain were obtained. As a model, swine alphacoronavirus- transmissible gastroenteritis virus (TGEV) was used. Replication activity of TGEV was studied in porcine leukocyte cell culture. The ability of this virusto induce the interferon gamma synthesis in leukocytes from convalescents was assessed TigraTest SARS-CoV-2 Enzyme-Linked SPOT analysis (Russia). Virus titration and neutralization assay with sera and monoclonal antibodies were carried out in porcine kidney PK-15 cell line. RESULTS: Serum samples from 43 donors (59% female, 41% male) aged from 21 to 56 was studied for the presence of class M and G antibodies to SARS-CoV-2 by ELISA method. Class M antibodies were found in only one donor. The highest titers of class G antibodies (1:400 to 1:1600) were detected among donors 4050 years old. For four donors with the highest antibody titers, HLA-II type was determined. The DQA1 allele in 3 patients was found to have the *0501 variant, which, according to literature data, is associated with autoimmune thyroid diseases. For the DQB1 allele, two convalescents had exactly the same variants (*0602-8). Among HLA DRB1, all patients had different allele variants (*09, *11, *01,*03,*13,*15,*08,*13). Among the samples, studied by ELISPOT method, in 45,5% of samples a positive T-cell response was noted after stimulation of macrophages with the S peptides of the SARS-CoV-2 protein and in 22.7% of samples in response to the pool of N peptides of the SARS-CoV-2 protein. Interestingly, infection of macrophages from convalescents with the transmissible gastroenteritis virus caused interferon-gamma expression in 31,8% of cases. The monoclonal antibody to the Spike protein SARS-CoV-2 and its Fab variant were not able to neutralize the TGEV. Interestingly, serum samples from 16 donors convalescent after COVID-19 caused virus neutralization in PK-15 cell culture at dilutions of 1:4-1:8. CONCLUSIONS: We have shown that porcine alphacoronavirus induces (in 31.8% of cases) the synthesis of interferon gamma in macrophages of recovered afterCOVID-19 donors, which may indicate cross-recognition of the antigen of a closely related coronavirus. However, monoclonal antibodies against the Spike protein SARS-CoV-2 did not demonstrate to TGEV neutralization. In turn, the neutralization of the TGEV by sera from recovered COVID-19 donors suggests that not only Spike protein, but also other coronavirus antigens can play a significant role in antigenic imprinting and antibody-dependent enhancement of COVID-19.

Zika virus (ZIKV) is one of several examples of an unprecedented pandemic spread and against which there is currently no suitable vaccine or treatment. Here, we constructed and characterized recombinant baculovirus-derived ZIKV-like particles (Zika VLPs) to study ZIKV-antibody interactions. These VLPs, uniquely consisted of the full-length ZIKV capsid (C), pre-membrane (prM), and envelope (E) proteins with either: a) the viral nonstructural NS2B and NS3 protease unit under one or two different promoters or b) an alternative host-cell furin protease encoding cleavage sequence inserted between the C and prM genes, together with lobster tropomyosin leader and honeybee signal sequences with one promoter for increased extracellular secretion. All these Zika VLPs displayed typical virion morphology in transmission electron microscopic analysis when expressed in both insect (Sf9) and mammalian (HEK293T) cells and no uncleaved prM glycoprotein was detected, as are present on immature virions. The importance of glycosylation of the E glycoprotein was shown by the effects on both polyclonal and monoclonal antibody reactions after these N-linked carbohydrate residues were disrupted by oxidation or enzymatic cleavage. Importantly, the construct which contained the host-cell furin protease cleavage sequence together with a lobster tropomyosin leader and honeybee signal sequences under one promoter produced higher Zika VLP titers and protein concentrations and which can now be tested as a superior construct in multifunctional diagnostic (ELISA and neutralization/antibody-dependent enhancement) assays and immunogenic assessments possibly leading to vaccine trials.

Adsorption of red blood cells (hemadsorption) on the surface of macrophages infected with African swine fever virus (ASFV) is a unique phenomenon allowing to determine virus infectious titer in hemadsorption unit (HAU) and differentiate virus strains phenotypically. In the meantime, hemadsorption of particular ASFV strain can by inhibited by homologous anti-ASFV serum containing antibody to the serogroup-specific virus protein (CD2v). Here, we describe a hemadsorption inhibition assay (HADIA) to phenotype ASFV strains to one of the known nine serogroups using blood-derived swine macrophages. The HADIA is a powerful method in the ASFV immunopathology and vaccine research since it provides additional antigenic and phenotypic characteristics of virus strains that can't be defined by other assays.

Introduction. African swine fever virus (ASF) is a large DNA virus that is the only member of the Asfarviridae family. The spread of the ASF virus in the territory of the Russian Federation, Eastern Europe and China indicates the ineffectiveness of existing methods of combating the disease and reinforces the urgent need to create effective vaccines. One of the most significant antigens required for the formation of immune protection against ASF is a serotype-specific CD2v protein. The purpose of the study. This study presents the results of immuno-informatics on the identification of B- and T-cell epitopes for the CD2v protein of the ASF virus using in silico prediction methods. Material and methods. The primary sequence of the CD2v protein of the ASFV virus strain Georgia 2007/1 (ID-FR682468) was analyzed in silico by programs BCPred, NetCTLpan, VaxiJen, PVS and Epitope Conservancy Analysis. Results. Using the BCPred and VaxiJen programs, 4 major B-cell immunogenic epitopes were identified. Analysis of the secretory region of ASF virus CD2v protein in NetCTLpan revealed 5 T-cell epitopes from the 32nd to the 197th position of amino acids that cross-link from the 1st to the 13th allele of the MHC-I of pig Discussion. This study presents the results in silico prediction to identify B- and T-cell epitopes of ASF virus CD2v protein. The soluble region of the CD2v protein can be included in the recombinant polyepitope vaccine against African swine fever. Conclusion. B- and T-cell epitopes in the secretory region of the CD2v protein (from 17 to 204 aa) of ASF virus were identified by in silico prediction. An analysis of the conservatism of the identified B- and T-cell epitopes allowed us to develop a map of the distribution of immune epitopes in the CD2v protein sequence.

Gene and cell therapies are widely recognized as future cancer therapeutics but poor controllability limits their clinical applications. Optogenetics, the use of light-controlled proteins to precisely spatiotemporally regulate the activity of genes and cells, opens up new possibilities for cancer treatment. Light of specific wavelength can activate the immune response, oncolytic activity and modulate cell signaling in tumor cells non-invasively, in dosed manner, with tissue confined action and without side effects of conventional therapies. Here, we review optogenetic approaches in cancer research, their clinical potential and challenges of incorporating optogenetics in cancer therapy. We critically discuss beneficial combinations of optogenetic technologies with therapeutic nanobodies, T-cell activation and CAR-T cell approaches, genome editors and oncolytic viruses. We consider viral vectors and nanoparticles for delivering optogenetic payloads and activating light to tumors. Finally, we highlight herein the prospects for integrating optogenetics into immunotherapy as a novel, fast, reversible and safe approach to cancer treatment.

The main criteria for current vaccines design are effectiveness, efficaciousness and safety. Increasing requirements for vaccine safety and purity push forward not only classical vaccine development, but also new generation vaccine technology, including sub-unit, recombinant, anti-idiotypic, DNA vaccines etc. This recombinant technology has already demonstrated its advantage, efficaciousness and safety in a large field of therapeutic and curative drug development for animal and human (S. Khan et al., 2016). In 2011, six novel drugs were created based on the new Fc-fusion protein technology. Most of the newly developed drugs affect receptor-ligand interactions, acting as antagonists by blocking direct receptor binding, i.e. Enbrel (etanercept; Amgen, USA), Zaltrap (aflibercept; Sanofi, France), Arcalyst (rilonacept; Regeneron, USA), or as agonists for direct stimulation of receptor function which augment immune response as Amevive (alefacept, Astellas, USA) does, or decrease immune response as Nplate (romiplostim; Amgen, USA) does. In this review, we pay attention to the most relevant results from the last few years for virus and bacterial vaccine designed based on Fc-fusion technology. The Fc-chimeras are hybrid sequences in which Fc-fragment of IgG (Fc-IgG) and targeted therapeutic protein are fused in an entire protein molecule (V. Pechtner et al., 2017). In this fusion, the hinge region of Fc-IgG is a flexible spacer between therapeutic protein and conservative part of IgG. It helps to minimize potential negative effect of two functional domains to each other. Therapeutic drugs based on Fc-fusion proteins are divided in three types, the receptor-Fc, peptide-Fc, and monomer-Fc. The Fc-fused proteins have tremendous therapeutic potential, since Fc domain in this molecules helps to specifically augment the pharmaco-dynamics values. Presence of Fc-domain in hybrid molecules prolongs half elimination of protein from plasma, which extends drug therapeutic activity and slows down kidney clearance for large molecules. Here, we summarize the most significant experimental data of Fc-fusion technology application against such pathogens as human immunodeficiency virus (D. Capon et al., 1989), Ebola virus (K. Konduru et al., 2011), Dengue virus (M.Y. Kim et al., 2018), influenza virus (L. Du et al., 2011), Mycobacterium tuberculosis (S. Soleimanpour et al., 2015), classical swine fever virus (Z. Liu et al., 2017). We also discuss the critical aspects of mechanism of action, drug design and Fc-fused protein production. Targeted activation of effector systems boosts protective potential of immuno-genic molecules and broadens its application. The interest of this review is focused on an application of Fc-fused proteins as potential vaccines against infectious human and animal diseases. We also briefly discuss the perspectives of Fc-fused antigens for novel effective medicine developments using African swine fever virus as an example.

Porcine epidemic diarrhea (PED) is a contagious viral disease in pigs, caused by the coronavirus porcine epidemic diarrhea virus (PEDV). PEDV infection results in significant mortality in piglets in unvaccinated herds. Like many others RNA viruses, PEDV has high evolutionary rate and is prone to genetic mutations. In this study, we analyzed the complete genome sequence of the recently sequenced isolate PEDV/Belgorod/dom/2008. A recombination event in S gene of PEDV/Belgorod/dom/2008 was detected. Pairwise identity analysis of the whole genome sequences revealed that PEDV/Belgorod/dom/2008 is an intermediate between PEDV and transmissible gastroenteritis virus (TGEV) strains. These results can be used for further analysis of the evolutionary variability, prevalence, and epidemiology of the porcine epidemic diarrhea virus.

The introduction of genotype II African swine fever (ASF) virus (ASFV) into the Caucasus in 2007 resulted in unprecedented disease propagation via slow geographical expansion through wild boar populations, short- and long-distance human-mediated translocations, and incursions into naive wild boar and domestic pig populations. The disease is now widespread in eastern and central Europe as well as in Asia, including China. The global dimension of the current epidemic shows that all countries need to be prepared for an introduction. In its natural habitat in Africa, ASFV is maintained within an ancient cycle between soft argasid ticks and the common warthog. Once introduced to the domestic pig population, direct and indirect virus transmission occurs with or without involvement of the tick vector in the pig-tick and domestic pig epidemiological cycles respectively. In the domestic pig cycle, human activities involving pigs or pig derived products are the dominating driver of virus transmission. ASF epidemiology in the presence of wild boar and northern European climates has proved to have specific characteristics, described in the wild boar-habitat epidemiological cycle. In this cycle wild boar carcasses and the resulting contamination of the environment play key roles in virus persistence. In both the wild boar-habitat and the domestic pig epidemiological cycle, fully implemented biosecurity is the key for stopping virus transmission and controlling the disease. Positive examples from the Czech Republic and Belgium show that control and eradication of ASF from the wild boar-habitat cycle can be achieved. Both these cases, as well as the example of Sardinia, where ASFV genotype I now seem very close to eradication after more than 40 years presence, further underline the importance of involving, engaging and understanding all stakeholders in the value chains from farm and forest to fork in order to accomplish ASF control and eradication.
