Cancer drug resistance poses a significant obstacle to successful chemotherapy, primarily driven by the activity of ATP‐binding cassette (ABC) transporters, which actively efflux chemotherapeutic agents from cancer cells, reducing their intracellular concentrations and therapeutic efficacy. Recent studies have highlighted the pivotal role of long noncoding RNAs (lncRNAs) in regulating this resistance, positioning them as crucial modulators of ABC transporter function. lncRNAs, once considered transcriptional noise, are now recognized for their complex regulatory capabilities at various cellular levels, including chromatin modification, transcription, and post‐transcriptional processing. This review synthesizes current research demonstrating how lncRNAs influence cancer drug resistance by modulating the expression and activity of ABC transporters. lncRNAs can act as molecular sponges, sequestering microRNAs that would otherwise downregulate ABC transporter genes. Additionally, they can alter the epigenetic landscape of these genes, affecting their transcriptional activity. Mechanistic insights reveal that lncRNAs contribute to the activity of ABC transporters, thereby altering the efflux of chemotherapeutic drugs and promoting drug resistance. Understanding these interactions provides a new perspective on the molecular basis of chemoresistance, emphasizing the regulatory network of lncRNAs and ABC transporters. This knowledge not only deepens our understanding of the biological mechanisms underlying drug resistance but also suggests novel therapeutic strategies. Doxorubicin (DOX) is an effective chemotherapeutic agent used for the treatment of various types of cancer. However, its poor solubility, undesirable side effects, and short half-life have remained a challenge. We used a formulation based on graphene oxide as an anticancer drug delivery system for DOX in MCF-7 breast cancer cells, to address these issues. In vitro release studies confirmed that the synthesized formulation has an improved release profile in acidic conditions (similar to the tumor microenvironment). Further in vitro studies, including MTT, uptake, and apoptosis assays were performed. The toxic effects of the nanocarrier on the kidney, heart and liver of healthy rats were also evaluated. We observed that the DOX-loaded carrier improved the cytotoxic effect of DOX on the breast cell line compared to free DOX. In summary, our results introduce the DOX-loaded carrier as a potential platform for in vitro targeting of cancer cells and suggest further studies are necessary to investigate its in vivo anti-cancer potential.

This study aimed to investigate the underlying mechanisms in anti-tumorigenesis effects of exercise through evaluation of inflammation and apoptosis. Twenty-four Wistar rats were divided into control, exercise, 1,2-dimethylhydrazine (DMH), and DMH + exercise. After a week, rats in the DMH group were given DMH twice a week for 2 weeks. Animals in the exercise groups performed exercise on a treadmill 5 days/week for 8 weeks. After 8 weeks of training, levels of COX-2, PCNA, Bax, Bcl-2, and procaspase-3/cleaved caspase-3 were assessed. Histological changes, number of aberrant crypt foci (ACF), and serum levels of TNF-[alpha] and IL-6 were also analyzed. ACF number was significantly decreased following the exercise program. Protein levels of COX-2 and PCNA and serum levels of IL-6 and TNF-[alpha] were significantly elevated in the rats receiving DMH and downregulated after performing the exercise program (P < 0.05). Exercise upregulated apoptosis, which was evident from the increased Bax/Bcl2 ratio, and enhanced the expression levels of activated caspase-3 as compared to the DMH group. The colonic architecture was improved in DMH + exercise. Exercise can effectively attenuate DMH-induced increase of inflammatory markers. Exercise induces apoptosis at the downstream of the inflammatory response. Therefore, exercise may play a role as a moderator of inflammation to exert protective effects against colon cancer. Cette etude portait sur les modes d'action sous-jacents des effets anti-tumorigenes de l'exercice physique par l'intermediaire de l'inflammation et de l'apoptose. Nous avons reparti 24 rats Wistar dans les groupes suivants: temoin, exercice physique, 1,2-dimethylhydrazine (DMH) et DMH + exercice physique. Apres une semaine, nous avons administre aux rats DMH du DMH 2 fois par semaine pendant 2 semaines. Les animaux des groupes exercice physique ont couru sur un tapis roulant 5 jours par semaine pendant 8 semaines. Apres 8 semaines d'entrainement, nous avons evalue les taux de COX-2, de PCNA, de Bax, de Bcl-2 et de caspase 3 scindee par la procaspase 3. Nous avons aussi evalue les variations histologiques, le nombre de foyers de cryptes aberrantes, ainsi que les taux seriques de TNF-[alpha] et d'IL-6. Le nombre de foyers de cryptes aberrantes etait nettement moins eleve a la suite du programme d'exercice physique. Les taux de COX-2 et de PCNA en proteines et les taux seriques d'IL-6 et de TNF-[alpha] etaient nettement plus eleves chez les rats recevant du DMH, avec une regulation a la baisse apres le programme d'exercice physique (P < 0,05). L'exercice physique entrainait une regulation a la hausse de l'apoptose, ce dont temoignait une augmentation du ratio Bax/Bcl2 avec une augmentation des taux d'expression de la caspase 3 activee par rapport au groupe DMH. Nous avons observe une amelioration de l'architecture du colon dans le groupe DMH + exercice physique. L'exercice physique peut attenuer efficacement la hausse des marqueurs de l'inflammation engendree par le DMH. L'exercice physique engendre de l'apoptose en aval de la reaction inflammatoire. Par consequent, l'exercice physique pourrait jouer un role en tant que moderateur de l'inflammation pour exercer des effets protecteurs contre le cancer du colon. A significant increase in the worldwide incidence and prevalence of type 2 diabetic mellitus (T2DM) has elevated the need for studies on novel and effective therapeutic strategies. Sirtuin 1 (SIRT1) is an NAD + dependent protein deacetylase with a critical function in the regulation of glucose/lipid metabolism, insulin resistance, inflammation, oxidative stress, and mitochondrial function. SIRT1 is also involved in the regulation of insulin secretion from pancreatic β-cells and protecting these cells from inflammation and oxidative stress-mediated tissue damages. In this regard, major SIRT1 activators have been demonstrated to exert a beneficial impact in reversing T2DM-related complications including cardiomyopathy, nephropathy, retinopathy, and neuropathy, hence treating T2DM. Therefore, an accumulating number of recent studies have investigated the efficacy of targeting SIRT1 as a therapeutic strategy in T2DM. In this review we aimed to discuss the current understanding of the physiological and biological roles of SIRT1, then its implication in the pathogenesis of T2DM, and the therapeutic potential of SIRT1 in combating T2DM. Introduction: Serum amyloid-A (SAA) and protein carbonyl group are rigorously related with cardiovascular diseases (CVDs) as a sensitive marker of an acute inflammatory state and as an important index of oxidative stress, respectively. Moreover, diet is one of the main factors that canmodify cardiovascular risks. Therefore, this study aimed to investigate the effects of Ramadanfasting on SAA and protein carbonyl group levels in patients with CVDs.Methods: Twenty-one patients (21 male; mean age 52±9 years old) with CVDs (coronaryartery disease, cerebrovascular, or peripheral arterial diseases) were participated in this study.Biochemical parameters were measured in patients 2 days before and 2 days after Ramadanfasting. SAA levels were assessed using enzyme-linked immunosorbent assay and Cayman’sprotein carbonyl colorimetric assay was provided for measuring protein carbonyl groups.Results: According to the findings of the study, post-Ramadan levels of inflammatory biomarker,SAA was decreased significantly in patients with CVDs in comparison with the baseline beforefastingvalues (16.84±8.20 vs. 24.40±6.72 μg/ml, P = 0.021). In addition, Ramadan fastingsignificantly reduced the levels of protein carbonyl group in patients as compared with those ofbaseline values (33.08±15.31 vs. 43.65±16.88 nmol/ml, P = 0.039).Conclusion: Ramadan fasting has impressive effects on modulating CVDs by decreasinginflammation and oxidative stress markers. However, to get a clear conclusion with more results,further investigation is warranted.

Respiratory viral infections are common respiratory diseases. Influenza viruses, RSV and SARS-COV2 have the potential to cause severe respiratory infections. Numerous studies have shown that unregulated immune response to these viruses can cause excessive inflammation and tissue damage. Therefore, regulating the antiviral immune response in the respiratory tract is of importance. In this regard, recent years studies have emphasized the importance of vitamin D in respiratory viral infections. Although, the most well-known role of vitamin D is to regulate the metabolism of phosphorus and calcium, it has been shown that this vitamin has other important functions. One of these functions is immune regulation. Vitamin D can regulate the antiviral immune response in the respiratory tract in order to provide an effective defense against respiratory viral infections and prevention from excessive inflammatory response and tissue damage. In addition, this vitamin has preventive effects against respiratory viral infections. Some studies during the COVID-19 pandemic have shown that vitamin D deficiency may be associated with a higher risk of mortality and sever disease in patients with COVID-19. Since, more attention has recently been focused on vitamin D. In this article, after a brief overview of the antiviral immune response in the respiratory system, we will review the role of vitamin D in regulating the antiviral immune response comprehensively. The third most common malignancy has been identified as Colorectal cancer (CRC) that conducive to death in most cases. Chemoresistance is a common obstacle to CRC treatment. Circulating exosomal microRNAs (miRNAs) have been shown to reverse chemo‐resistance and are promising biomarkers for CRC. The capacity of engineered exosomes to cross biological barriers and deliver functional miRNAs could be used to achieve these proposes. The object of this review is the investigation of the role of exosomal miRNA in the chemo‐resistance, diagnosis, and prognosis of CRC. Using Preferred Reporting Items for Systematic Reviews and Meta‐Analyses guidelines, electronic databases, PubMed, EMBASE, Web of Science, Scopus were searched from January 1990 to November 2020. Ultimately, eight articles included five in vitro (16 cell lines) and three in vivo examinations. Three studies demonstrated that increasing or decreasing mRNA expression was associated with increasing and decreasing cell proliferation in vitro. The presence of miRNA in two studies increased the sensitivity of the drug and exhibited a considerable growth inhibitory effect on cancer cell proliferation. The apoptotic rate was significantly increased in four studies by increased mRNA expression and reduced mrna expression. Tumor volume of xenograft models in three studies suppressed by antitumor miRNA activity. In contrast, anti‐miRNA activity in one study decreased the tumor volume. Exosomal miRNAs can be regulators of chemo‐resistance and predict adverse outcomes in CRC patients. In sum, exosomes containing miRNAs can be a promising biomarker for the prognosis and diagnosis of CRC.
