Abstract: ITER envisages the use of two heating neutral beam injectors plus an optional one as part of the auxiliary heating and current drive system. The 16.5 MW expected neutral beam power per injector is several notches higher than worldwide existing facilities. A Neutral Beam Test Facility (NBTF) was established at Consorzio RFX, exploiting the synergy of two test beds, SPIDER and MITICA. SPIDER is dedicated to developing and characterizing large efficient negative ion sources at relevant parameters in ITER-like conditions: source and accelerator located in the same vacuum where the beam propagates, immunity to electromagnetic interferences of multiple radio-frequency (RF) antennas, avoidance of RF-induced discharges on the outside of the source. Three years of experiments on SPIDER have addressed to the necessary design modifications to enable full performances. The source is presently under a long shut-down phase to incorporate learnings from the experimental campaign. Parallelly, developments on MITICA, the full-scale prototype of the ITER NBI featuring a 1 MV accelerator and ion neutralization, are underway including manufacturing of in-vessel components, while power supplies and auxiliary plants are already under final testing and commissioning. Integration, commissioning and tests of the 1MV power supplies are essential for this first-of-kind system, unparalleled both in research and industry field. The integrated test to confirm 1MV output by combining invertor systems, DC generators and transmission lines extracted errors/accidents in some components. To realize a concrete system for ITER, solutions for the repair and the improvement of the system were developed. Hence, NBTF is emerging as a necessary facility, due to the large gap with existing injectors, effectively dedicated to identify issues and find solutions to enable successful ITER NBI operations in a time bound fashion.

Parallelly, developments on MITICA, the full-scale prototype of the ITER Neutral Beam Injector (NBI) featuring a 1 MV accelerator and ion neutralization, are underway including manufacturing of the beam source, accelerator and the beam line components, while power supplies and auxiliary plants, already installed, are under final testing and commissioning. Abstract: BACKGROUND: Among children, sex-specific differences in disease prevalence, age of onset, and susceptibility have been observed in health conditions including asthma, immune response, metabolic health, some pediatric and adult cancers, and psychiatric disorders. Epigenetic modifications such as DNA methylation may play a role in the sexual differences observed in diseases and other physiological traits. METHODS: We performed a meta-analysis of the association of sex and cord blood DNA methylation at over 450,000 CpG sites in 8438 newborns from 17 cohorts participating in the Pregnancy And Childhood Epigenetics (PACE) Consortium. We also examined associations of child sex with DNA methylation in older children ages 5.5-10 years from 8 cohorts (n = 4268). RESULTS: In newborn blood, sex was associated at Bonferroni level significance with differences in DNA methylation at 46,979 autosomal CpG sites (p < 1.3 × 10-7) after adjusting for white blood cell proportions and batch. Most of those sites had lower methylation levels in males than in females. Of the differentially methylated CpG sites identified in newborn blood, 68% (31,727) met look-up level significance (p < 1.1 × 10-6) in older children and had methylation differences in the same direction. CONCLUSIONS: This is a large-scale meta-analysis examining sex differences in DNA methylation in newborns and older children. Expanding upon previous studies, we replicated previous findings and identified additional autosomal sites with sex-specific differences in DNA methylation. Differentially methylated sites were enriched in genes involved in cancer, psychiatric disorders, and cardiovascular phenotypes.

Abstract: Heating neutral beam (HNB) injectors, necessary to achieve burning conditions and to control plasma instabilities in ITER, are characterized by such demanding parameters that a neutral beam test facility (NBTF) dedicated to their development and optimization is being realized in Padua (Italy) with direct contributions from the Italian government (through Consorzio RFX as the host entity) and the ITER international organization (with kind contributions from the ITER domestic agencies of Europe, Japan and India) and technical and scientific support from various European laboratories and universities. The NBTF hosts two experiments: SPIDER, devoted to ion source optimization for the required source performance, and MITICA, the full-size prototype of the ITER HNB, with an ion source identical to SPIDER. This paper gives an overview of the progress towards NBTF realization, with particular emphasis on issues discovered during this phase of activity and on solutions adopted to minimize the impact on the schedule and maintain the goals of the facilities. The realization of MITICA is well advanced; operation is expected to start in 2023 due to the long procurement time of the in-vessel mechanical components. The beam source power supplies, operating at 1 MV, are in an advanced phase of realization; all high-voltage components have been installed and the complex insulation test phase began in 2018. At the same time, construction and installation of SPIDER plant systems was successfully completed with their integration into the facility. The mechanical components of the SPIDER ion source were installed inside the vessel and connected to the plants. Integrated commissioning with the control, protection and safety systems ended positively and the first experimental phase has begun. The first results of the SPIDER experiment, with data from operational diagnostics, and the plans for the 1 MV insulation tests on the MITICA high-voltage components are presented.

Abstract: Background: Studies looking at associations between environmental chemicals and child behaviour usually consider only one exposure or family of exposures.Objective: This study explores associations between prenatal exposure to a wide range of environmental chemicals and child behaviour.Methods: We studied 703 mother-child pairs from five European cohorts recruited in 2003-2009 We assessed 47 exposure biomarkers from eight chemical exposure families in maternal blood or urine collected during pregnancy. We used the Strengths and Difficulties Questionnaire (SDQ) to evaluate child behaviour between three and seven years of age. We assessed associations of SDQ scores with exposures using an adjusted least absolute shrinkage and selection operator (LASSO) considering all exposures simultaneously and an adjusted exposome-wide association study (ExWAS) considering each exposure independently.Results: LASSO selected only copper (Cu) as associated with externalizing behaviour. In the ExWAS, bisphenol A [BPA, incidence rate ratio (IRR): 1.06, 95% confidence interval (95%CI): 1.01; 1.12] and mono-n-butyl phthalate (MnBP, IRR: 1.06, 95%CI: 1.00; 1.13) were associated with greater risk of externalizing behaviour problems. Cu (IRK: 0.90, 95%CI: 0.82; 0.98), perfluoroundecanoate (PFUnDA, IRR: 0.92, 95%CI: 0.84;0.99) and organochlorine compounds (OCs) were associated with lower risk of externalizing behaviour problems, however the associations with OCs were mainly seen among women with insufficient weight gain during pregnancy. Internalizing score worsen in association with exposure to diethyl thiophosphate (DETP, IRR: 1.11, 95%CI: 1.00;124) but the effect was driven by the smallest cohort. Internalizing score improved with increased concentration of perfluorooctane sulfonate (PFOS, IRR: 0.92, 95%CI: 0.85;1.00), however the association was driven by the two smallest cohorts with the lowest PFOS concentrations.Discussion: This study added evidence on deleterious effects of prenatal exposure to BPA and MnBP on child behaviour. Other associations should be interpreted cautiously since they were not consistent with previous studies or they have not been studied extensively. (C) 2020 The Authors. Published by Elsevier B.V.

Internalizing score worsen in association with exposure to diethyl thiophosphate (DETP, IRR: 1.11, 95%CI: 1.00;1.24) but the effect was driven by the smallest cohort. Abstract: Early life is an important window of opportunity to improve health across the full lifecycle. An accumulating body of evidence suggests that exposure to adverse stressors during early life leads to developmental adaptations, which subsequently affect disease risk in later life. Also, geographical, socio-economic, and ethnic differences are related to health inequalities from early life onwards. To address these important public health challenges, many European pregnancy and childhood cohorts have been established over the last 30 years. The enormous wealth of data of these cohorts has led to important new biological insights and important impact for health from early life onwards. The impact of these cohorts and their data could be further increased by combining data from different cohorts. Combining data will lead to the possibility of identifying smaller effect estimates, and the opportunity to better identify risk groups and risk factors leading to disease across the lifecycle across countries. Also, it enables research on better causal understanding and modelling of life course health trajectories. The EU Child Cohort Network, established by the Horizon2020-funded LifeCycle Project, brings together nineteen pregnancy and childhood cohorts, together including more than 250,000 children and their parents. A large set of variables has been harmonised and standardized across these cohorts. Abstract: The exposome represents the totality of life course environmental exposures (including lifestyle and other non-genetic factors), from the prenatal period onwards. This holistic concept of exposure provides a new framework to advance the understanding of complex and multifactorial diseases. Prospective pregnancy and birth cohort studies provide a unique opportunity for exposome research as they are able to capture, from prenatal life onwards, both the external (including lifestyle, chemical, social and wider community-level exposures) and the internal (including inflammation, metabolism, epigenetics, and gut microbiota) domains of the exposome. In this paper, we describe the steps required for applying an exposome approach, describe the main strengths and limitations of different statistical approaches and discuss their challenges, with the aim to provide guidance for methodological choices in the analysis of exposome data in birth cohort studies. An exposome approach implies selecting, pre-processing, describing and analyzing a large set of exposures. Several statistical methods are currently available to assess exposome-health associations, which differ in terms of research question that can be answered, of balance between sensitivity and false discovery proportion, and between computational complexity and simplicity (parsimony). Assessing the association between many exposures and health still raises many exposure assessment issues and statistical challenges. The exposome favors a holistic approach of environmental influences on health, which is likely to allow a more complete understanding of disease etiology.

Abstract: The ITER Heating Neutral Beam (HNB) injectors, one of the tools necessary both to achieve burning conditions and to control plasma instabilities, are characterized by such demanding parameters as to require the construction of a Neutral Beam Test Facility (NBTF) dedicated to their development and optimization. The NBTF was realized in Padua (Italy), with the direct contribution of the Italian government, through the Consorzio RFX as the host entity, IO, the in kind contributions of three Domestic Agencies (F4E, JADA, INDA) and the technical and scientific support of various European laboratories and universities. The NBTF hosts two experiments: SPIDER and MITICA. The former is devoted to the optimization of the HNB and DNB ion sources and to the achievement of the required source performances. MITICA is the full size prototype of the ITER HNB, with an ion source identical to the one used in SPIDER. This paper gives an overview of the progress of the NBTF realization with particular emphasis on issues discovered during this phase of activities and to the adopted solutions in order to minimize the impact on the schedule while maintaining the goals of the facilities. The realization of MITICA is well advanced; it is expected to enter into operation in 2023 due to the long procurement times of the in-vessel mechanical components. The power supply designed to operate at 1MV are in an advanced phase of realization; all the high voltage components have been installed and the complex insulation test phase has begun in 2018. The construction and installation of SPIDER plant systems was successfully completed with their integration into the facility. The mechanical components of the ion source were installed inside the vessel and connected to the plants. The integrated commissioning with control (CODAS), protection and safety systems ended positively and the first experimental phase began. Finally, the first results of the SPIDER experimentation and of the 1MV insulation tests on the MITICA high voltage components are presented.

Abstract: The application of metabolic phenotyping in clinical and epidemiological studies is limited by a poor understanding of inter-individual, intra-individual and temporal variability in metabolic phenotypes. Using 1H NMR spectroscopy we characterised short-term variability in urinary metabolites measured from 20 children aged 8–9 years old. Daily spot morning, night-time and pooled (50:50 morning and night-time) urine samples across six days (18 samples per child) were analysed, and 44 metabolites quantified. Intraclass correlation coefficients (ICC) and mixed effect models were applied to assess the reproducibility and biological variance of metabolic phenotypes. Excellent analytical reproducibility and precision was demonstrated for the 1H NMR spectroscopic platform (median CV 7.2%). Pooled samples captured the best inter-individual variability with an ICC of 0.40 (median). Trimethylamine, N-acetyl neuraminic acid, 3-hydroxyisobutyrate, 3-hydroxybutyrate/3-aminoisobutyrate, tyrosine, valine and 3-hydroxyisovalerate exhibited the highest stability with over 50% of variance specific to the child. The pooled sample was shown to capture the most inter-individual variance in the metabolic phenotype, which is of importance for molecular epidemiology study design. A substantial proportion of the variation in the urinary metabolome of children is specific to the individual, underlining the potential of such data to inform clinical and exposome studies conducted early in life.

Abstract: The ITER Neutral Beam Test Facility (NBTF), called PRIMA (Padova Research on ITER Megavolt Accelerator), is hosted in Padova, Italy and includes two experiments: MITICA, the full-scale prototype of the ITER heating neutral beam injector, and SPIDER, the full-size radio frequency negative-ions source. The NBTF realization and the exploitation of SPIDER and MITICA have been recognized as necessary to make the future operation of the ITER heating neutral beam injectors efficient and reliable, fundamental to the achievement of thermonuclear-relevant plasma parameters in ITER. This paper reports on design and R&D carried out to construct PRIMA, SPIDER and MITICA, and highlights the huge progress made in just a few years, from the signature of the agreement for the NBTF realization in 2011, up to now - when the buildings and relevant infrastructures have been completed, SPIDER is entering the integrated commissioning phase and the procurements of several MITICA components are at a well advanced stage

Abstract: SPIDER is one of two projects of the ITER Neutral Beam Test Facility under construction in Padova, Italy, at the Consorzio RFX premises. It will have a 100 keV beam source with a full-size prototype of the radiofrequency ion source for the ITER neutral beam injector (NBI) and also, similar to the ITER diagnostic neutral beam, it is designed to operate with a pulse length of up to 3600 s, featuring an ITER-like magnetic filter field configuration (for high extraction of negative ions) and caesium oven (for high production of negative ions) layout as well as a wide set of diagnostics. These features will allow a reproduction of the ion source operation in ITER, which cannot be done in any other existing test facility. SPIDER realization is well advanced and the first operation is expected at the beginning of 2018, with the mission of achieving the ITER heating and diagnostic NBI ion source requirements and of improving its performance in terms of reliability and availability. Abstract: Substantial progresses have been achieved in the realization of the ITER Neutral Beam Test Facility (NBTF) hosted in Padova, Italy; the buildings, completed by the end of 2015, are being progressively filled with new systems and components. The realization of SPIDER, the ITER full-size negative ion source, is well advanced and important progress is also recorded for MITICA, the full-scale prototype of the ITER HNB injector. The paper gives an overview of the achieved results, highlighting the main challenges faced.

Abstract: Summary Among the drugs commonly used in the treatment of memory disorders of the elderly, vincamine and hydergine® have been shown to moderately increase the firing rate of noradrenergic locus coeruleus (LC) neurons. Since changes in electrical activity of noradrenergic neurons are generally reflected in corresponding alterations of the turnover of this transmitter, the effects of these drugs on the accumulation of 3,4-dihyroxyphenylacetic acid (DOPAC) and dopamine (DA) in the presence and absence of the dopamine-?-hydroxylase inhibitor, FLA 63, were studied in the LC as well as in two of its projection areas, the hippocampus and the cerebellum. Characterization of this procedure with the a2-adrenoceptor antagonist, idazoxan, the corresponding agonist, clonidine, the a1-adrenoceptor antagonist prazosine, and haloperidol, suggested that- DOPAC changes are more suitable than those of DA or DOPAC/DA ratios in reflecting changes in noradrenaline (NA) turnover,- inhibiting DBH is advantageous if NA turnover is to be measured in projection areas, but not in LC, and- haloperidol and prazosine, in principle, did not affect NA turnover.
