Here, we compare face-selective and word-selective responses in a large group of patients (N = 37) implanted with intracerebral electrodes in the ventral occipito-temporal cortex (VOTC). Both face-selective (i.e., significantly different responses to faces vs. non-face visual objects) and word-selective (i.e., significantly different responses to words vs. pseudofonts) neural activity is isolated with frequency-tagging. Critically, this sensitive approach allows to objectively quantify category-selective neural responses and disentangle them from general visual responses. About 70% of significant electrode contacts show either face-selectivity or word-selectivity only, with the expected right and left hemispheric dominance, respectively. Spatial dissociations are also found within core regions of face and word processing, with a medio-lateral dissociation in the fusiform gyrus (FG) and surrounding sulci, respectively. In the 30% of overlapping face- and word-selective contacts across the VOTC or in the FG and surrounding sulci, between-category-selective amplitudes (faces vs. words) show no-to-weak correlations, despite strong correlations in both the within-category-selective amplitudes (face-face, word-word) and the general visual responses to words and faces. Overall, these observations support the view that category-selective circuitry for faces and written words is largely dissociated in the human adult VOTC.

Abstract

Both mild and severe epilepsies are influenced by variants in the same genes, yet an explanation for the resulting phenotypic variation is unknown. As part of the ongoing Epi25 Collaboration, we performed a whole-exome sequencing analysis of 13,487 epilepsy-affected individuals and 15,678 control individuals. While prior Epi25 studies focused on gene-based collapsing analyses, we asked how the pattern of variation within genes differs by epilepsy type. Specifically, we compared the genetic architectures of severe developmental and epileptic encephalopathies (DEEs) and two generally less severe epilepsies, genetic generalized epilepsy and non-acquired focal epilepsy (NAFE). Our gene-based rare variant collapsing analysis used geographic ancestry-based clustering that included broader ancestries than previously possible and revealed novel associations. Using the missense intolerance ratio (MTR), we found that variants in DEE-affected individuals are in significantly more intolerant genic sub-regions than those in NAFE-affected individuals. Only previously reported pathogenic variants absent in available genomic datasets showed a significant burden in epilepsy-affected individuals compared with control individuals, and the ultra-rare pathogenic variants associated with DEE were located in more intolerant genic sub-regions than variants associated with non-DEE epilepsies. MTR filtering improved the yield of ultra-rare pathogenic variants in affected individuals compared with control individuals. Finally, analysis of variants in genes without a disease association revealed a significant burden of loss-of-function variants in the genes most intolerant to such variation, indicating additional epilepsy-risk genes yet to be discovered. Taken together, our study suggests that genic and sub-genic intolerance are critical characteristics for interpreting the effects of variation in genes that influence epilepsy.

Abstract

Cette étude porte sur les changements de localisation des productions agricoles au sein de l'Union européenne. Ces phénomènes sont analysés grâce à la construction de deux modèles d'économie géographique. Le premier présente la répartition de deux activités agricoles sur un territoire. Il est centré sur la représentation des marchés agricoles et inspiré des travaux de Krugman (1991). Il permet d'identifier certains déterminants (coûts de transport, politiques publiques...) des phénomènes de spécialisation ou de diversification de la production agricole sur un territoire. Le second modèle présente la répartition de deux activités dans deux espaces productifs. Il montre comment, lors d'une intégration, les politiques économiques de nature réglementaire (différenciation territoriale des produits) peuvent infléchir les phénomènes de concentration géographique des activités agricoles. Afin de valider les résultats issus des deux modèles, l'étude de la localisation des biens agricoles est réalisée pour la période 1983/1995, selon une typologie (30 bassins de production) établie sur la base du Réseau d'information comptable agricole (RICA). Elle permet de mettre en évidence que les phénomènes de spécialisation et de concentration des productions sont propres à chaque produit, en fonction de leurs caractéristiques et de leur appartenance, ou non, à une Organisation commune de marché. La mise en place d'indicateurs de compétitivité des bassins et les tests économétriques qui en découlent permettent de présenter l'influence des différents déterminants sur l'évolution de la localisation géographique des biens agricoles. Alors que la concentration des produits soutenus est freinée par les politiques publiques, les produits "agro-industriels" non soutenus vont se concentrer dans les bassins disposant d'un accès au marché communautaire (coûts de transport).

Face-selective contacts are three times more numerous than house-selective contacts and show a larger amplitude, with a right hemisphere advantage for faces. Most importantly, these category-selective contacts are spatially dissociated along the lateral-to-medial VOTC axis, respectively, consistent with neuroimaging evidence. At the minority of "overlap" contacts responding selectively to both faces and houses, response amplitude to the two categories is not correlated, suggesting a contribution of distinct populations of neurons responding selectively to each category. The medio-lateral dissociation also extends into the underexplored anterior temporal lobe (ATL). In this region, a relatively high number of intracerebral recording contacts show category-exclusive responses (i.e., without any response to baseline visual objects) to faces but rarely to houses, in line with the proposed role of this region in processing people-related semantic information. Altogether, these observations shed novel insight on the neural basis of human visual recognition and strengthen the validity of the frequency-tagging approach coupled with intracerebral recordings in epileptic patients to understand human brain function.

Item does not contain fulltext, The extent to which faces and written words share neural circuitry in the human brain is actively debated. We provide an original contribution to this debate by comparing face-selective and word-selective responses in a large group of patients (N=37) implanted with intracerebral electrodes in the ventral occipito-temporal cortex (VOTC). Both face-selective (i.e., significantly different responses to faces vs. nonface objects) and word-selective (i.e., significantly different responses to words vs. pseudofonts) neural activity is isolated through frequency-tagging (Jonas et al., 2016; Lochy et al., 2018, respectively). Critically, this approach allows disentangling category-selective neural responses from general visual responses. Overall, we find that 69.26% of significant contacts show either face- or word-selectivity, with the expected right and left hemispheric dominance, respectively (Fig.1A,B). Moreover, the center of mass for word-contacts is more lateral than for face-contacts, with no differences in postero-anterior axis (Fig.2A). Spatial dissociations are also found within core regions of face and word processing, with a medio-lateral dissociation in the fusiform gyrus (FG) and surrounding sulci (FG+sulci;Fig.2B), while a postero-anterior dissociation is found in the inferior occipital gyrus (IOG;Fig.2C). Despite their spatial dissociations in the FG+sulci and IOG, most overlap in category-selective responses is found in these regions (Fig.1C). Critically, in the overlap-contacts, across the whole brain or specifically in the FG+sulci, between-category (word-face) selective-amplitudes showed no-to-weak correlations, despite strong correlations for within-category (face-face, word-word) selective-amplitudes (Fig.3A), and a strong correlation in non-selective general-amplitudes to words-faces. Item does not contain fulltext, BACKGROUND: Results of clinical trials are often criticized by low inclusion rate and potential sampling bias in patient recruitment. The aim of this validation registry is to evaluate how far an all-comers design in the context of clinical research can ensure the representation of the true all-comers population. METHODS: This validation registry is a prospective international multicentre registry, conducted at 10 out of the total 21 centers, participating in TARGET-AC (registered under NCT02520180). During a predefined four-week period data were recorded prospectively on all PCIs performed in the participating centers, whether or not patients were enrolled in TARGET-AC. Data were collected on patient demographics, angiographic lesion- and procedural characteristics. For patients who were not enrolled in the study, operators were asked to declare the reason for not enrolling the patient, using a single-choice questionnaire. RESULTS: A total of 131 patients were enrolled in the TARGET-AC study during the investigated period (ER group), standing as 20% (range 4% and 54%) of all eligible cases per protocol. In the ER group more patients presented with stable angina (61% vs. 43%, respectively; P < .001). Whereas ST-elevation infarction was less common (5% vs. 26%, respectively; P < .001), there was no difference in non-ST elevation acute coronary syndrome (32% vs. 27%, respectively; P = .248). Risk factors and comorbidities did not show any difference between the ER and the non-enrolled (NER) groups, except for greater rate of significant valvular disease in the NER group (12% vs 19%, respectively; P = .037). The NER group presented more thrombotic stenoses than the ER group (20% vs 12%, respectively; P = .040).
