Abstract

The saw-scaled viper venom causes continued tissue damage that may cause death. Here the authors show that the venom attracts neutrophils to the bite site and induces neutrophil extracellular traps that capture venom components causing tissue damage, which can be prevented by enzymatic DNA degradation.

Abstract

Aberrant activation of nuclear factor kappa B (NF-κB) has been linked with the pathogenesis of several proinflammatory diseases including number of cancers and inflammatory bowel diseases. In the present work, we evaluated the anticancer activity of 1,2-oxazines derivatives against colorectal cancer cell lines and identified 2-((2-acetyl-6,6-dimethyl-4-phenyl-5,6-dihydro-2H-1,2-oxazin-3-yl)methyl)isoindoline-1,3-dione (API) as the lead anticancer agent among the tested compounds. The apoptosis inducing effect of API was demonstrated using flow cytometry analysis and measuring the caspase 3/7 activity in API treated cells. Based on the literature on inhibition of NF-κB by oxazines, we evaluated the effect of 1,2-oxazines against the ability of NF-κB binding to DNA, NF-κB-dependent luciferase expression and IκBα phosphorylation. We found that, API abrogate constitutive activation of NF-κB and inhibits IκBα phosphorylation in HCT116 cells. Our in silico analysis revealed the binding of oxazines to the hydrophobic cavity that present between the interface of p65 and IκBα. Given the relevance with aberrant activation of NF-κB in inflammation bowel disease (IBD), we evaluated the effect of API on dextran sulphate sodium-induced IBD mice model. The treatment of IBD induced mice with API decreased the myeloperoxidase activity in colonic extract, modulated the colon length and serum levels of pro- and anti-inflammatory cytokines such as TNF-α, IFN-γ, IL-6, IL-1β and IL-10. Furthermore, the histological analysis revealed the restoration of the distorted cryptic epithelial structure of colon in the API treated animals. In conclusion, we comprehensively validated the NF-κB inhibitory efficacy of API that targets NF-κB in in vitro colon cancer and an in vivo inflammatory bowel disease model.

Abstract

Thrombocytopenia in methotrexate (MTX)-treated cancer and rheumatoid arthritis (RA) patients connotes the interference of MTX with platelets. Hence, it seemed appealing to appraise the effect of MTX on platelets. Thereby, the mechanism of action of MTX on platelets was dissected. MTX (10 μM) induced activation of pro-apoptotic proteins Bid, Bax and Bad through JNK phosphorylation leading to ΔΨm dissipation, cytochrome c release and caspase activation, culminating in apoptosis. The use of specific inhibitor for JNK abrogates the MTX-induced activation of pro-apoptotic proteins and downstream events confirming JNK phosphorylation by MTX as a key event. We also demonstrate that platelet mitochondria as prime sources of ROS which plays a central role in MTX-induced apoptosis. Further, MTX induces oxidative stress by altering the levels of ROS and glutathione cycle. In parallel, the clinically approved thiol antioxidant N-acetylcysteine (NAC) and its derivative N-acetylcysteine amide (NACA) proficiently alleviate MTX-induced platelet apoptosis and oxidative damage. These findings underpin the dearth of research on interference of therapeutic drugs with platelets, despite their importance in human health and disease. Therefore, the use of antioxidants as supplementary therapy seems to be a safe bet in pathologies associated with altered platelet functions.

Abstract

Drugs such as necopidem, saripidem, alpidem, zolpidem, and olprinone contain nitrogen-containing bicyclic, condensed-imidazo[1,2-α]pyridines as bioactive scaffolds. In this work, we report a high-yield one pot synthesis of 1-(2-methyl-8-aryl-substitued-imidazo[1,2-α]pyridin-3-yl)ethan-1-onefor the first-time. Subsequently, we performed in silico mode-of-action analysis and predicted that the synthesized imidazopyridines targets Phospholipase A2 (PLA2). In vitro analysis confirmed the predicted target PLA2 for the novel imidazopyridine derivative1-(2-Methyl-8-naphthalen-1-yl-imidazo [1,2-α]pyridine-3-yl)-ethanone (compound 3f) showing significant inhibitory activity towards snake venom PLA2 with an IC50 value of 14.3 μM. Evidently, the molecular docking analysis suggested that imidazopyridine compound was able to bind to the active site of the PLA2 with strong affinity, whose affinity values are comparable to nimesulide. Furthermore, we estimated the potential for oral bioavailability by Lipinski's Rule of Five. Hence, it is concluded that the compound 3f could be a lead molecule against snake venom PLA2.

6. Novel benzoxazine-based aglycones block glucose uptake in vivo by inhibiting glycosidases.

Abstract

Glycoside hydrolases catalyze the selective hydrolysis of glycosidic bonds in oligosaccharides, polysaccharides, and their conjugates. β-glucosidases occur in all domains of living organisms and constitute a major group among glycoside hydrolases. On the other hand, the benzoxazinoids occur in living systems and act as stable β-glucosides, such as 2-(2,4-dihydroxy-7-methoxy-2H-1,4-benzoxazin-3(4H)-one)-β-D-gluco-pyranose, which hydrolyse to an aglycone DIMBOA. Here, we synthesized the library of novel 1,3-benzoxazine scaffold based aglycones by using 2-aminobenzyl alcohols and aldehydes from one-pot reaction in a chloroacetic acid catalytic system via aerobic oxidative synthesis. Among the synthesized benzoxazines, 4-(7-chloro-2,4-dihydro-1H-benzo[d][1,3]oxazin-2-yl)phenol (compound 7) exhibit significant inhibition towards glucosidase compared to acarbose, with a IC50 value of 11.5 µM. Based upon results generated by in silico target prediction algorithms (Naïve Bayesian classifier), these aglycones potentially target the additional sodium/glucose cotransporter 1 (where a log likelihood score of 2.70 was observed). Furthermore, the in vitro glucosidase activity was correlated with the in silico docking results, with a high docking score for the aglycones towards the substrate binding site of glycosidase. Evidently, the in vitro and in vivo experiments clearly suggest an anti-hyperglycemic effect via glucose uptake inhibition by 4-(7-chloro-2,4-dihydro-1H-benzo[d][1,3]oxazin-2-yl)phenol in the starved rat model. These synthetic aglycones could constitute a novel pharmacological approach for the treatment, or re-enforcement of existing treatments, of type 2 diabetes and associated secondary complications.

7. Novel apigenin based small molecule that targets snake venom metalloproteases.

Abstract

The classical antivenom therapy has appreciably reduced snakebite mortality rate and thus is the only savior drug available. Unfortunately, it considerably fails to shield the viper bite complications like hemorrhage, local tissue degradation and necrosis responsible for severe morbidity. Moreover, the therapy is also tagged with limitations including anaphylaxis, serum sickness and poor availability. Over the last decade, snake venom metalloproteases (SVMPs) are reported to be the primary component responsible for hemorrhage and tissue degradation at bitten site. Thus, antivenom inability to offset viper venom-induced local toxicity has been a basis for an insistent search for SVMP inhibitors. Here we report the inhibitory effect of compound 5d, an apigenin based molecule against SVMPs both in silico and in vivo. Several apigenin analogues are synthesized using multicomponent Ugi reactions. Among them, compound 5d effectively abrogated Echis carinatus (EC) venom-induced local hemorrhage, tissue necrosis and myotoxicity in a dose dependant fashion. The histopathological study further conferred effective inhibition of basement membrane degradation, and accumulation of inflammatory leucocytes at the site of EC venom inoculation. The compound also protected EC venom-induced fibrin and fibrinogen degradation. The molecular docking of compound 5d and bothropasin demonstrated the direct interaction of hydroxyl group of compound with Glu146 present in hydrophobic pocket of active site and does not chelate Zn2+. Hence, it is concluded that compound 5d could be a potent agent in viper bite management.

Abstract

Thrombocytopenia is a serious issue connected with the pathogenesis of several human diseases including chronic inflammation, arthritis, Alzheimer's disease, cardiovascular diseases (CVDs) and other oxidative stress-associated pathologies. The indiscriminate use of antibiotics and other biological drugs are reported to result in thrombocytopenia, which is often neglected during the treatment regime. In addition, augmented oxidative stress induced by drugs and pathological conditions has also been shown to induce thrombocytopenia, which seems to be the most obvious consequence of elevated rate of platelet apoptosis. Thus, blocking oxidative stress-induced platelet apoptosis would be of prime importance in order to negotiate thrombocytopenia and associated human pathologies. The current study presents the synthesis and platelet protective nature of novel ibuprofen derivatives. The potent anti-oxidant ibuprofen derivative 4f was selected for the study and the platelet protective efficacy and platelet aggregation inhibitory property has been demonstrated. The compound 4f dose dependently mitigates the oxidative stress-induced platelet apoptosis in both platelet rich plasma and washed platelets. The platelet protective nature of compound 4f was determined by assessing various apoptotic markers such as ROS generation, cytosolic Ca2+ levels, PS externalization, cytochrome C translocation, Caspase activation, mitochondrial membrane depolarization, cytotoxicity, LDH leakage and tyrosine phosphorylation of cytosolic proteins. Furthermore, compound 4f dose dependently ameliorated agonist induced platelet aggregation. Therefore, compound 4f can be estimated as a potential candidate in the treatment regime of pathological disorders associated with platelet activation and apoptosis. In addition, compound 4f can be used as an auxiliary therapeutic agent in pathologies associated with thrombocytopenia.

Abstract

Smoke from human-induced fires such as prescribed fires can occasionally cause significant reduction in visibility on highways in the southern United States. Visibility reduction to less than 3 m has been termed “superfog” and environmental conditions that lead to its formation have been proposed previously. Accurate characterization and prediction of precursor conditions for superfog is needed to prevent dangerous low visibility situations when planning prescribed fires. It is hypothesized that extremely hygroscopic cloud condensation nuclei from the smoldering phase of a fire can produce a large number of droplets smaller in size than in naturally occurring fog. This large number of small droplets can produce superfog conditions with relatively low liquid water content. A thermodynamics-based model for fog formation was developed. Laboratory generated superfog measured by a Phase Doppler Particle Analyzer determined that mean droplet radius was 1.5 μm and the size distribution could be modeled with a lognormal distribution. Experiments in an environmentally-conditioned wind tunnel using longleaf pine (Pinus palustris Mill.) needle fuel beds provided visibility, heat flux, temperature, humidity, and particle data for model validation. Numerical modeling was used to approximate the growth of a superfog boundary-layer with liquid water content values of 2 g m−3 or greater. The model successfully predicted previous superfog events.

Abstract

Background: Asthma and atopic dermatitis (AD) are chronic allergic conditions, along with allergic rhinitis and food allergy and cause high morbidity and mortality both in children and adults. This study aims to evaluate the global, regional, national, and temporal trends of the burden of asthma and AD from 1990 to 2019 and analyze their associations with geographic, demographic, social, and clinical factors. Methods: Using data from the Global Burden of Diseases (GBD), Injuries, and Risk Factors Study 2019, we assessed the age-standardized prevalence, incidence, mortality, and disability-adjusted life years (DALYs) of both asthma and AD from 1990 to 2019, stratified by geographic region, age, sex, and socio-demographic index (SDI). DALYs were calculated as the sum of years lived with disability and years of life lost to premature mortality. Additionally, the disease burden of asthma attributable to high body mass index, occupational asthmagens, and smoking was described. Results: In 2019, there were a total of 262 million [95% uncertainty interval (UI): 224–309 million] cases of asthma and 171 million [95% UI: 165–178 million] total cases of AD globally; age-standardized prevalence rates were 3416 [95% UI: 2899–4066] and 2277 [95% UI: 2192–2369] per 100,000 population for asthma and AD, respectively, a 24.1% [95% UI: −27.2 to −20.8] decrease for asthma and a 4.3% [95% UI: 3.8–4.8] decrease for AD compared to baseline in 1990. Both asthma and AD had similar trends according to age, with age-specific prevalence rates peaking at age 5–9 years and rising again in adulthood. The prevalence and incidence of asthma and AD were both higher for individuals with higher SDI; however, mortality and DALYs rates of individuals with asthma had a reverse trend, with higher mortality and DALYs rates in those in the lower SDI quintiles. Abstract

We are developing a model of vacuum arcs. This model assumes that arcs develop as a result of mechanical failure of the surface due to Coulomb explosions, followed by ionization of fragments by field emission and the development of a small, dense plasma that interacts with the surface primarily through self sputtering and terminates as a unipolar arc capable of producing breakdown sites with high enhancement factors. We have attempted to produce a self consistent picture of triggering, arc evolution and surface damage. We are modeling these mechanisms using Molecular Dynamics (mechanical failure, Coulomb explosions, self sputtering), Particle-In-Cell (PIC) codes (plasma evolution), mesoscale surface thermodynamics (surface evolution), and finite element electrostatic modeling (field enhancements). Abstract

Hydrogen remains an attractive alternative fuel to petroleum and a number of investigators claim that adding hydrogen to the air intake manifold of a diesel engine will reduce criteria emissions and diesel fuel consumption. Such claims are appealing when trying to simultaneously reduce petroleum consumption, greenhouse gases and criteria pollutants. The goal of this research was to measure the change in criteria emissions (CO, NOx, and PM 2.5) and greenhouse gases such as carbon dioxide (CO2), using standard test methods for a wide range of hydrogen addition rates. A two-stroke Detroit Diesel Corporation 12V-71TI marine diesel engine was mounted on an engine dynamometer and tested at three out of the four loads specified in the ISO 8178-4 E3 emission test cycle and at idle. The engine operated on CARB ultra-low sulfur #2 diesel with hydrogen added at flow rates of 0, 22 and 220 SLPM. As compared with the base case without hydrogen, measurements showed that hydrogen injection at 22 and 220 SLPM had negligible influence on the overall carbon dioxide specific emission, EFCO2. However, in examining data at each load the data revealed that at idle EFCO2 was reduced by 21% at 22 SLPM (6.9% of the added fuel energy was from hydrogen) and 37.3% at 220 SLPM (103.1% of the added fuel energy was from hydrogen). At all other loads, the influence of added hydrogen was insignificant. Specific emissions for nitrogen oxides, EFNOx, and fine particulate matters, EFPM 2.5, showed a trade-off relationship at idle. At idle, EFNO x was reduced by 28% and 41% with increasing hydrogen flow rates, whilst EFPM2.5 increased by 41% and 86% respectively. For other engine loads, EFNOx and EFPM2.5 did not change significantly with varying hydrogen flow rates. One of the main reasons for the greater impact of hydrogen at idle is that the contribution of hydrogen to the total fuel energy is much higher at idle as compared to the other loads. The final examination in this paper was the system energy balance when hydrogen is produced by an on-board electrolysis unit. An analysis at 75% engine load showed that hydrogen production increased the overall equivalent fuel consumption by 2.6% at 22 SLPM and 17.7% at 220 SLPM. © 2014, Hydrogen Energy Publications, LLC. Abstract

The development of autonomous vehicles has been the subject of considerable research and development in recent years. The project aims to develop a 2-wheeled autonomous car using a Robotic Operating System (ROS) with lidar and camera sensors as inputs. The car's control system will be designed to make decisions based on the information provided by these sensors, allowing it to navigate and avoid obstacles in real-time. The project will begin by selecting appropriate hardware components such as motors, batteries and sensors and integrating them into a functional prototype. Lidar and camera sensors will be used to detect and localize the position of the car, detect obstacles and determine their distance from the vehicle. The control system will be developed using ROS, an open source software framework for robot development, and will include algorithms for navigation, obstacle avoidance and decision making. The car will be programmed to follow a predetermined route. Finally, the autonomous car's performance will be evaluated through various tests, including its ability to follow a specific path and avoid obstacles. The results of these tests will be analyzed, and possible improvements will be identified. The outcome of this project is expected to provide a proof-of-concept for the development of autonomous vehicles using ROS with lidar and camera sensors as input. Abstract

The currently existing label-based techniques for the detection of circulating tumor cells (CTCs) target natural surface proteins of cells and are therefore applicable to only limited cancer cell types. We report optomicrofluidic detection of cancer cells in the pool of peripheral blood mononuclear cells (PBMCs) by exploiting the difference in their cell metabolism. We employ metabolic glycoengineering as a click chemistry tool for tagging cells that yields several fold-higher fluorescence signals from cancer cells compared to that from PBMCs. The effects of concentrations of the tagging compounds and cell incubation time on the fluorescence signal intensity are studied. The tagged cells were encapsulated in droplets ensuring that cells enter the detection region two-dimensionally focused in single-file and optically detected with a high detection efficiency and low coefficient of variation of the signals. The metabolic tagging approach showed a significantly higher tagging efficiency and average fluorescence signal compared to the well-established and widely adopted anti-EpCAM-FITC-based tagging. We demonstrated the detection of three different cancer cell lines – EpCAM-negative cervical cancer cell, HeLa, weakly EpCAM positive, and triple-negative breast cancer cell, MDA-MB-231, and strongly EpCAM positive breast cancer cell, MCF7, highlighting that the proposed technique is independent of naturally occurring cell surface proteins and widely applicable. The metabolically tagged and optically detected cells were successfully recultured, proving the compatibility of the proposed technique with downstream assays. The proposed technique is then utilised for the detection of CTCs in metastatic cancer patients' blood. The current work provides a new strategy for detecting cancer cells in the blood that can find potential applications in both fundamental research and clinical studies involving CTCs as well as in single-cell sequencing. Abstract

This study focuses on the effect of using silane-treated groundnut shell powder (GSP) as a filler material in varying weight proportions (1 %, 3 %, 5 %, and 7 % wt.) in the fabrication of ramie fibre-reinforced epoxy composites. This study also deals with the mechanical, thermal, and hydrophobic properties of ramie fibre-epoxy composites. A biological waste filler made from groundnut shell (Arachis hypogaea L.), which incorporates cellulose, hemi-cellulose, and lignin, is surface-treated with silane (3-aminopropyltriethoxysilane) using the wet solution technique. Ramie fibre-epoxy composites were created using hand layup and ambient temperature curing. The highest tensile strength of a composite made with 5 wt. % GSP particles in an epoxy matrix is 171 MPa; the maximum flexural strength is 228 MPa, the Izod impact toughness is 6.7 J; and the micro-hardness is 91 Shore-D. Although the thermal stability rises as the filler loading increases, nanocomposites also show a nearly similar tendency toward thermal stability at higher loadings. The silane-treated GSP contributed to an improvement in wear resistance of the composite specimens ERG1, ERG2, ERG3, and ERG4 compared to the untreated ones. The composite specimens (ERG4) with more filler showed greater water absorption. After 45 days of immersion, the ERG4 specimens show a 17 % moisture absorption (the untreated specimen) and a 15% moisture absorption (the treated specimen).
