Electrical Resistivity Tomography (ERT) method has been used to study two tectonically active areas of southern Apennine (Caggiano Faults and Ufita Basin). The main aim of this job was to study the structural setting of the investigated areas, i.e. the geometry of the basins at depth, the location of active faults at surface, and their geometrical characterization. The comparison between ERT and trench/drilling data allowed us to evaluate the efficacy of the ERT method in studying active faults and the structural setting of seismogenic areas. In the Timpa del Vento intermontane basin, high resolution ERT across the Caggiano Fault scarps, with different arrays, electrode spacing (from 1 to 10 m) and penetration depth (from about 5 to 40 m) was carried out. The obtained resistivity models allowed us to locate the fault planes along the hillslope and to gather information at depth, as later con- firmed by paleoseismological trenches excavated across the fault trace. In the Ufita River Valley a 3560-m-long ERT was carried out across the basin, joining 11 roll-along multi-channel acquisition system with an electrode spacing of 20mand reaching an investigation depth of about 170 m. The ERT allowed us to reconstruct the geometry and thickness of the Quaternary deposits filling the Ufita Valley. Our reconstruction of the depositional setting is in agreement with an interpretative geological section based on borehole data. [ABSTRACT FROM AUTHOR]

Abstract Background Information theory is an increasingly popular framework for studying how the brain encodes sensory information. Despite its widespread use for the analysis of spike trains of single neurons and of small neural populations, its application to the analysis of other types of neurophysiological signals (EEGs, LFPs, BOLD) has remained relatively limited so far. This is due to the limited-sampling bias which affects calculation of information, to the complexity of the techniques to eliminate the bias, and to the lack of publicly available fast routines for the information analysis of multi-dimensional responses. Results Here we introduce a new C- and Matlab-based information theoretic toolbox, specifically developed for neuroscience data. This toolbox implements a novel computationally-optimized algorithm for estimating many of the main information theoretic quantities and bias correction techniques used in neuroscience applications. We illustrate and test the toolbox in several ways. First, we verify that these algorithms provide accurate and unbiased estimates of the information carried by analog brain signals (i.e. LFPs, EEGs, or BOLD) even when using limited amounts of experimental data. This test is important since existing algorithms were so far tested primarily on spike trains. Second, we apply the toolbox to the analysis of EEGs recorded from a subject watching natural movies, and we characterize the electrodes locations, frequencies and signal features carrying the most visual information. Third, we explain how the toolbox can be used to break down the information carried by different features of the neural signal into distinct components reflecting different ways in which correlations between parts of the neural signal contribute to coding. We illustrate this breakdown by analyzing LFPs recorded from primary visual cortex during presentation of naturalistic movies. Conclusion The new toolbox presented here implements fast and data-robust computations of the most relevant quantities used in information theoretic analysis of neural data. The toolbox can be easily used within Matlab, the environment used by most neuroscience laboratories for the acquisition, preprocessing and plotting of neural data. It can therefore significantly enlarge the domain of application of information theory to neuroscience, and lead to new discoveries about the neural code.

Abstract Background The volume of data available on genetic variations has increased considerably with the recent development of high-density, single-nucleotide polymorphism (SNP) arrays. Several software programs have been developed to assist researchers in the analysis of this huge amount of data, but few can rely upon a whole genome variability visualisation system that could help data interpretation. Results We have developed GenotypeColour™ as a rapid user-friendly tool able to upload, visualise and compare the huge amounts of data produced by Affymetrix Human Mapping GeneChips without losing the overall view of the data. Some features of GenotypeColour™ include visualising the entire genome variability in a single screenshot for one or more samples, the simultaneous display of the genotype and Copy Number state for thousands of SNPs, and the comparison of large amounts of samples by producing "consensus" images displaying regions of complete or partial identity. The software is also useful for genotype analysis of trios and to show regions of potential uniparental disomy (UPD). All information can then be exported in a tabular format for analysis with dedicated software. At present, the software can handle data from 10 K, 100 K, 250 K, 5.0 and 6.0 Affymetrix chips. Conclusion We have created a software that offers a new way of displaying and comparing SNP and CNV genomic data. Background/Objectives : The International Organization for Standardization (ISO) 20776-2:2021, which replaces ISO 20776-2:2007, focuses solely on the performance of antimicrobial susceptibility testing (AST) assays, emphasizing the ISO 20776-1 broth microdilution method as the reference standard. Consequently, categorical agreement (CA) and associated errors should not be applied. We verified the Vitek Reveal AST assay according to both ISO 20776-2:2021 and ISO 20776-2:2007 criteria. Methods : Samples from 100 simulated and clinical Gram-negative (GN) positive blood cultures (PBCs) were tested at a large teaching hospital. The simulated GN-PBCs were obtained from a hospital collection of isolates selected to represent diverse antimicrobial resistance profiles. The Reveal assay results were compared with those from the reference assay, and the time to result (TTR) for the Reveal assay was calculated. Results : The essential agreement rates were 96.1% (816/849) for simulated and 98.8% (929/940) for clinical GN-PBC samples. The bias values were -3.1 for simulated and -11.0 for clinical samples. The CA rates were 97.7% (808/827) for simulated and 99.2% (924/931) for clinical samples. The mean TTR ± SD (hours) for resistant organisms was significantly lower (4.40 ± 1.15) than that for susceptible, increased exposure (5.52 ± 0.48) and susceptible (5.54 ± 0.49) organisms. Conclusions : Our findings reinforce the potential of the Reveal assay as a valuable tool and support its implementation in clinical microbiology laboratories.

Accurate identification and rapid genotyping of Candida parapsilosis , a significant opportunistic pathogen in healthcare settings, is crucial for managing outbreaks, timely intervention, and effective infection control measures. This study includes 24 clinical samples and 2 positive environmental surveillance swabs collected during a fluconazole-resistant Candida parapsilosis outbreak at the Tuscany Rehabilitation Clinic (Clinica di Riabilitazione Toscana, CRT), located in the province of Arezzo, Italy. Fourier-transform infrared (FTIR) spectroscopy, genetic sequencing of the ERG11 gene, and short tandem repeat (STR) analysis was applied to track the fluconazole-resistant C. parapsilosis outbreak at the CRT facility. FTIR analysis clustered the isolates into two major groups, correlating with resistance-associated ERG11 mutations (Y132F and R398I), azole resistance levels, and year of isolation. The combined use of FTIR spectroscopy and STR typing provided a comprehensive approach to identify and track fluconazole-resistant C. parapsilosis isolates, which identified specific clusters of genetically similar isolates. By comparison with feasible molecular techniques, we conclude that FTIR spectroscopy applied in real time can inform targeted infection control strategies and aid in the effective management of nosocomial infections.

In 2019, Candida auris became the first fungal pathogen included in the list of the urgent antimicrobial threats by the Centers for Disease Control (CDC). Short tandem repeat (STR) analysis and whole-genome sequencing (WGS) are considered the gold standard, and can be complemented by other molecular methods, for the genomic surveillance and clade classification of this multidrug-resistant yeast. However, these methods can be expensive and require time and expertise that are not always available. The long turnaround time is especially not compatible with the speed needed to manage clonal transmission in healthcare settings. Fourier transform infrared (FTIR) spectroscopy, a biochemical fingerprint approach, has been applied in this study to a set of 74 C. auris isolates belonging to the five clades of C. auris (I-V) in combination with an artificial neural network (ANN) algorithm to create and validate "CLADE-FINDER", a tool for C. auris clade determination. The CLADE-FINDER classifier allowed us to discriminate the four primary C. auris clades (I-IV) with a correct classification for 96% of the samples in the validation set. This newly developed genotyping scheme can be reasonably applied for the effective epidemiological monitoring and management of C. auris cases in real time.

The identification of microorganisms at the species level has always constituted a diagnostic challenge for clinical microbiology laboratories. The aim of the present study has been the evaluation in a real-time assay of the performance of Autobio in comparison with the Bruker mass spectrometry system for the identification of bacteria and yeasts. A total of 535 bacteria and yeast were tested in parallel with the two systems by direct smear or fast formic acid extraction for bacteria and yeasts, respectively. Discordant results were verified by 16S, ITS rRNA or specific gene sequencing. Beyond giving comparable results for bacteria with respect to the MBT smart system, Autof MS2600 mass spectrometer provided excellent accuracy for the identification of yeast species of clinical interest.

Inflammatory bowel disease (IBD) includes ulcerative colitis (UC) and Crohn's disease (CD). These are autoimmune diseases of the gastrointestinal tract with a chronic relapsing and remitting course. Due to complex interactions between multiple factors in the etiology of IBD, the discovery of new predictors of disease course and response to therapy, and the development of effective therapies is a significant challenge. The dysregulation of microRNAs (miRNAs), a class of conserved endogenous, small non-coding RNA molecules with a length of 18-25 nucleotides, that regulate gene expression by an RNA interference process, is implicated in the complex pathogenetic context of IBD. Both tissue-derived, circulating, and fecal microRNAs have been explored as promising biomarkers in the diagnosis and the prognosis of disease severity of IBD. In this review, we summarize the expressed miRNA profile in blood, mucosal tissue, and stool and highlight the role of miRNAs as biomarkers with potential diagnostic and therapeutic applications in ulcerative colitis and Crohn's disease. Moreover, we discuss the new perspectives in developing a new screening model for the detection of colorectal cancer (CRC) based on fecal miRNAs.

Aim: The post-operative pain (POP) is accompanied by autonomic, psychological and behavioural responses which can result in a significant increase in morbidity and mortality. The purpose of this study was to compare the efficacy and safety of intravenous bolus administration of morphine versus subcutaneous administration., Material of Study: Fifty subjects scheduled for elective abdominal surgery were randomized into a group S (n = 26) and group E (n = 24). The first group was administered morphine (0.35 mg/kg) subcutaneously immediately after induction of anesthesia, while the second group the same drug at a dose of 0.25 mg/kg was administered intravenously 45' before the end of the intervention. All patients received intraoperative remifentanil (0.25-0.50 microg/kg/min), and at the end of surgery paracetameolo 1 g and ondansetron 4 mg. Upon awakening, the DPO has been reviewed by the NRS (numerical scale of pain) at time zero (TO), after 30' (t1), 1h (T2) and 24h (T3). Furthermore, it was annotated request for rescue doses of morphine and the collateral effects., Results: The two study groups did not show significant differences related to the anthropometric characteristics, duration of surgery, type of surgery, ASA class. No difference between the two groups was highlighted during the evaluation of POP through NRS scale and quantity of "rescue" doses requested. In group E there was a single case of respiratory depression., Conclusions: Our study suggests that whether intravenous or subcutaneous administration of morphine, both simple and economical analgesic techniques, they are able to guarantee the same quality control of POP in patients undergoing abdominal surgery.
