Magnetic Fe3O4 nanoparticles (MNPs) functionalized with (3-aminopropylo)trietoksysilan (APTES) or N-carboxymethylchitosan (CMC) were proposed as nanocarriers of methotrexate (MTX) to target ovarian cancer cell lines. The successful functionalization of the obtained nanostructures was confirmed by FT-IR spectroscopy. The nanoparticles were characterized by transmission electron spectroscopy (TEM) and dynamic light scattering (DLS) techniques. Their potential zeta, magnetization, and hyperthermic properties were also explored. MTX was conjugated with the nanocarriers by ionic bonds or by amide bonds. The drug release kinetics were examined at different pH and temperatures. The MTT assay showed no toxicity of the MNPs[APTES] and MNPs[CMC]. Finally, the cytotoxicity of the nanostructures with MTX attached towards the ovarian cancer cells was measured. The sensitivity and resistance to methotrexate was determined in simplistic 2D and spheroid 3D conditions. The cytotoxicity tests of the tested nanostructures showed similar values for inhibiting the proliferation of ovarian cancer cells as methotrexate in its free form. Conjugating MTX with nanoparticles allows the drug to be directed to the target site using an external magnetic field, reducing overall toxicity. Combining this approach with hyperthermia could enhance the therapeutic effect in vivo compared to free MTX, though further research on advanced 3D models is needed. [ABSTRACT FROM AUTHOR]

Textile industry effluents contain several hazardous substances, such as dye-containing effluents, which pose environmental and aesthetic challenges. Presently, the microbial-based remediation process is in use. This study investigated the application of ferrous–ferric oxide (Fe3O4) nanoparticles, a readily formulated nanoadsorbent, to remove scattered dye molecules from industrial effluents. The ferrous–ferric oxide nanoparticles were prepared using a chemical co-precipitation method. The nanoparticles had 26.93 emu g−1 magnetization, with sizes smaller than 20 nm, and possessed a highly purified cubic spinel crystallite structure. The catalytic activity of the iron oxide depended on the dose, photocatalytic enhancer, i.e., H2O2 level, pH of the reaction medium, and dye concentration. We optimized the Fenton-like reaction to work best using 1.0 g/L of ferrous–ferric oxide nanoparticles, 60 mM oxalic acid at pH 7.0, and 60 ppm of dye. Iron oxides act as photocatalysts, and oxalic acid generates electron–hole pairs. Consequently, higher amounts of super-radicals cause the rapid degradation of dye and pseudo-first-order reactions. Liquid chromatography–mass spectrometry (LC-MS) analysis revealed the ferrous–ferric oxide nanoparticles decolorized and destroyed Disperse Red 277 in 180 min under visible light. Hence, complete demineralization is observed using a photo-Fenton-like reaction within 3 h under visible light. These high-capacity, easy-to-separate next-generation adsorption systems are suggested to be suitable for industrial-scale use. Abstract Background New immunotherapies activate tumor-associated macrophages (TAMs) in the osteosarcoma microenvironment. Iron oxide nanoparticles (IONPs) are phagocytosed by TAMs and, therefore, enable TAM detection on T2*- and T2-weighted magnetic resonance images. We assessed the repeatability and reproducibility of T2*- and T2-mapping of osteosarcomas in a mouse model. Methods Fifteen BALB/c mice bearing-murine osteosarcomas underwent magnetic resonance imaging (MRI) on 3-T and 7-T scanners before and after intravenous IONP infusion, using T2*-weighted multi-gradient-echo, T2-weighted fast spin-echo, and T2-weighted multi-echo sequences. Each sequence was repeated twice. Tumor T2 and T2* relaxation times were measured twice by two independent investigators. Repeatability and reproducibility of measurements were assessed. Results We found excellent agreement between duplicate acquisitions for both T2* and T2 measurements at either magnetic field strength, by the same individual (repeatability), and between individuals (reproducibility). The repeatability concordance correlation coefficient (CCC) for T2* values were 0.99 (coefficients of variation (CoV) 4.43%) for reader 1 and 0.98 (CoV 5.82%) for reader 2. The reproducibility of T2* values between the two readers was 0.99 (CoV 3.32%) for the first acquisitions and 0.99 (CoV 6.30%) for the second acquisitions. Regarding T2 values, the repeatability of CCC was similar for both readers, 0.98 (CoV 3.64% for reader 1 and 4.45% for reader 2). The CCC of the reproducibility of T2 was 0.99 (CoV 3.1%) for the first acquisition and 0.98 (CoV 4.38%) for the second acquisition. Conclusions Our results demonstrated high repeatability and reproducibility of quantitative T2* and T2 mapping for monitoring the presence of TAMs in osteosarcomas. Relevance statement T2* and T2 measurements of osteosarcomas on IONP-enhanced MRI could allow identifying patients who may benefit from TAM-modulating immunotherapies and for monitoring treatment response. The technique described here could be also applied across a wide range of other solid tumors. Cancer immunotherapy has emerged as a groundbreaking field, offering promising and transformative tools for oncological research and treatment. However, it faces several limitations, including variations in cancer types, dependence on the tumor microenvironments (TMEs), immune cell exhaustion, and adverse reactions. Magnetic nanoparticles, particularly magnetite nanoparticles (MNPs), with established pharmacodynamics and pharmacokinetics for clinical use, hold great promise in this context and are now being explored for therapeutic aims. Numerous preclinical studies have illustrated their efficacy in enhancing immunotherapy through various strategies, such as modulating leukocyte functions, creating favorable TMEs for cytotoxic T lymphocytes, combining with monoclonal antibodies, and stimulating the immune response via magnetic hyperthermia (MHT) treatment (Front Immunol. 2021;12:701485. doi: 10.3389/fimmu.2021.701485). However, the current clinical trials of MNPs are mostly for diagnostic aims and as a tool for generating hyperthermia for tumor ablation. With concerns about the adverse effects of MNPs in the systems, clinical translation and clinical study of MNP-boosted immunotherapy remains limited. The lack of extensive clinical investigations poses a current barrier to patient application. Urgent efforts are needed to ascertain both the efficacy of MNP-enhanced immunotherapy and its safety profile in combination therapy. This article reviews the roles, potential, and challenges of using MNPs in advancing cancer immunotherapy. The application of MNPs in boosting immunotherapy, and its perspective role in research and development is also discussed.

To use magnetic iron oxide nanoparticles (Fe3O4) for drug delivery, it is essential to modify the coated surface of nanoparticles with appropriate functional groups to decrease their toxicity. This study aims to explore how silica-coated magnetic iron oxide nanoparticles decorated with –NH2, –SH, and –OH affect the structure and function of lysozyme. Magnetite iron oxide nanoparticles were synthesized and coated with mesoporous silica (mSiO2@Fe3O4) and next decorated with functional groups of –NH2, –SH, and –OH then physically and structurally characterized using FTIR, VSM, XRD, DLS, zeta potential, FESEM, and TEM. Afterward, it assessed the effects of the synthesized nanoparticles on structural and conformational changes of hen egg white lysozyme (HEWL) as a model protein. The interaction between lysozyme and nanoparticles was evaluated through ATR-FTIR spectroscopy, intrinsic fluorescence, ANS and thioflavin T (ThT) fluorescence, and thermal aggregation assay. Our findings demonstrated that mSiO2@Fe3O4 altered the lysozyme's conformation and decreased its activity, whereas NH2-mSiO2@Fe3O4 increased lysozyme activity and prevented lysozyme aggregation. All the synthesized nanoparticles with the functional groups increased the protein stability during the thermal aggregation and inhibited lysozyme aggregation. [ABSTRACT FROM AUTHOR]

Single-walled carbon nanotubes (SWCNTs) loaded with magnetic iron oxide nanoparticles were prepared by the arc discharge method and air heat treatment. The nanocomposite was characterized by X-ray diffraction, scanning electron microscopy, Raman spectroscopy, vibrating sample magnetometry, etc. The results showed that the heat-treated nanocomposites (SWCNTs/FexOy) had iron oxide phases and superparamagnetic properties with a saturation magnetization of 33.32 emu/g. Compared with the non-heat-treated materials, SWCNTs/FexOy had a larger specific surface area and pore volume. Using SWCNTs/FexOy to remove the organic contaminant (bisphenol A, BPA), it was found that under the conditions of pH = 3 and adsorbent dosage of 0.2 g/L, the maximum adsorption capacity of the composite was 117 mg/g, and the adsorption could reach more than 90% in only 5 min when the BPA content was below 0.05 mmol/L. The fitting results of the Langmuir and D-R models are more consistent with the experimental data, indicating a relatively uniform distribution of the adsorption sites and that the adsorption process is more consistent with physical adsorption. The kinetic calculations showed that the SWCNTs/FexOy exhibits chemical effects on both the surface and the gap, and the adsorption process is controlled by the π-π bonds and the hydrophobicity of the SWCNTs/FexOy. Simple Summary: Image-guided stereotactic body radiation therapy (SBRT), utilizing biocompatible superparamagnetic iron oxide nanoparticles (SPION), like ferumoxytol, has emerged as a non-invasive, safe, and effective therapy for liver tumors. However, the radiomodulating properties of ferumoxytol on hepatic macrophages have never been directly investigated. We showed that ferumoxytol affected human monocytes increasing their resistance to radiation-induced cell death. These findings provide the basis for mechanism-based optimization of SPION-enhanced image-guided functional treatment planning platform for reducing hepatotoxicity in patients with advanced hepatic cirrhosis undergoing liver SBRT for liver cancer before liver transplant. Superparamagnetic iron oxide nanoparticles (SPION) have attracted great attention not only for therapeutic applications but also as an alternative magnetic resonance imaging (MRI) contrast agent that helps visualize liver tumors during MRI-guided stereotactic body radiotherapy (SBRT). SPION can provide functional imaging of liver parenchyma based upon its uptake by the hepatic resident macrophages or Kupffer cells with a relative enhancement of malignant tumors that lack Kupffer cells. However, the radiomodulating properties of SPION on liver macrophages are not known. Utilizing human monocytic THP-1 undifferentiated and differentiated cells, we characterized the effect of ferumoxytol (Feraheme®), a carbohydrate-coated ultrasmall SPION agent at clinically relevant concentration and therapeutically relevant doses of gamma radiation on cultured cells in vitro. We showed that ferumoxytol affected both monocytes and macrophages, increased the resistance of monocytes to radiation-induced cell death and inhibition of cell activity, and supported the anti-inflammatory phenotype of human macrophages under radiation. Its effect on human cells depended on the duration of SPION uptake and was radiation dose-dependent. The results of this pilot study support a strong mechanism-based optimization of SPION-enhanced MRI-guided liver SBRT for primary and metastatic liver tumors, especially in patients with liver cirrhosis awaiting a liver transplant. Retinoblastoma (RB) is one of the most important cancers in children with a higher rate of prevalence in developing countries. Despite different approaches to the treatment of RB, it seems necessary to discover a new approach to its treatment. Today, mitochondria are recognised as an important target in the treatment of cancer. Superparamagnetic iron oxide nanoparticles (SPIONs) have been studied by researchers due to their important biological effects. In this study, the effects of SPIONs on mitochondria isolated from Y79 retinoblastoma cells were investigated. The results showed that SPIONs were able to increase the reactive oxygen species (ROS) level and subsequently damage the mitochondrial membrane and release cytochrome c a as one of the important pro-apoptotic proteins of RB mitochondria. Furthermore, the results indicated a decrease in cell viability and an increase in caspase-3 activity in Y79 retinoblastoma cells. These events can lead to the killing of cancerous mitochondria. This study synthesizes magnetic iron oxide nanoparticles from agro-waste sweet pepper extract, exploring their potential as antioxidant additives and in food preservation. Iron (III) chloride hexahydrate is the precursor, with sweet pepper extract as both a reducing and capping agent at pH 7.5. Characterization techniques, including microscopy and spectroscopy, analyze the sweet pepper extract-magnetic iron oxide nanoparticles. Antioxidant capacities against 2,2-diphenyl-1-picrylhydrazyl are assessed, incorporating nanoparticles into banana-based bioplastic for grape preservation. Microscopy reveals cubic and quasi-spherical structures, and spectroscopy confirms functional groups, including Fe–O bonds. X-ray diffraction identifies cubic and monoclinic magnetite with a monoclinic hematite presence. Sweet pepper extract exhibits 100% inhibitory activity in 20 min, while sweet pepper extract-magnetic iron oxide nanoparticles show an IC50 of 128.1 µg/mL. Furthermore, these nanoparticles, stabilized with banana-based bioplastic, effectively preserve grapes, resulting in a 27.4% lower weight loss rate after 144 h compared to the control group (34.6%). This pioneering study encourages institutional research into the natural antioxidant properties of agro-waste sweet pepper combined with magnetic iron and other metal oxide nanoparticles, offering sustainable solutions for nanopackaging and food preservation. This work is aimed at studying the drug delivery applications of iron oxide (Fe3O4) nanoparticles with strontium (Sr) doping with varying molar ratios prepared by the co-precipitation route. The impact of increased strontium content on the particle size and magnetic properties was investigated. The impending of these nanoparticles for drug loading, drug release, and their respective cytotoxicity was also inspected. First, iron oxide nanoparticles were doped with various amounts of strontium, from 0.25, 0.50, and 0.75, to 1 mol using co-precipitation method. These synthesized nanoparticles were characterized by XRD, SEM, EDX, VSM, and FTIR for evaluating crystal structure, phase purity, morphology, composition, magnetic properties, and functional groups, respectively. Drug loading and drug release properties were determined using UV–vis spectroscopy, whereas MTT assay evaluated cytotoxicity. Colloidal stability was assessed using zeta potential in PBS solution. The findings confirmed the successful doping of iron oxide with strontium via XRD and EDX. SEM results confirmed spherical morphology for all and needle-like structure for 1 mol strontium doped sample. For VSM results, a single domain structure was established. It was also observed that the drug encapsulation efficiency increases with increased strontium content. Cytotoxicity results by MTT assay revealed increased cytotoxicity with increasing nanoparticle concentration, and ibuprofen-loaded nanoparticles showed higher cytotoxicity than un-loaded nanoparticles at the same concentration. Zeta potential results showed colloidal stability of iron oxide nanoparticles increased by the addition of strontium. This study provided predominantly comparison of the cytotoxicity of ibuprofen-loaded and non-loaded nanoparticles on Hep-2 cancer cells at similar concentrations for the first time for both Fe3O4 particles and Sr-doped Fe3O4 nanoparticles and enclosed the impact of increasing Sr doping content on Fe3O4 nanoparticles. Abstract Medical textiles, including surgical gowns, masks are used as obstacles to prevent the risk of infection for both doctor and patient. The widespread of bacteria and viruses, e.g. chronic hepatitis B, hepatitis C and currently Covid-19 viruses in the patient population is very common. In this work, multifunctional eco-friendly polyester fabrics have been produced by in situ impregnation of 2-((E)-(2-(2,4-dinitrophenyl)hydrazono)methyl)-4-((E)-phenyldiazenyl)phenol disperse dye onto magnetic iron oxide nanoparticles. The technique endowed polyester fabrics with a new color as well as magnetic and antibacterial functionalities. The colored magnetic nanoparticles showed high affinity toward fabrics. Besides, the unbound dye could be easily collected from wastewater by a magnet, significantly facilitate the wastewater treatment. The treated fabrics were analyzed by energy dispersive X-ray spectroscopy, scanning electron microscopy, X-ray diffraction and vibrating sample magnetometry. Colorimetric values, tensile properties and fastness of the composite fabrics were also measured. The tensile properties of the composite were increased after functionalization. The wettability features of the fabric were investigated and showed a significant improvement. Also, the toxicity of the resulted fabric was exhibited low toxicity against wi-38 cell line. These results indicate the potentiality of the suggested technique in producing multifunctional fabrics with various applications, especially as medical textiles.

Nanomedicine presents exciting new opportunities for the detection and treatment of cancer. Current cancer imaging methods and treatment approaches in clinics frequently fall short of entirely curing cancer and can have severe side effects. Theranostic nanoparticles, however, have the potential to revolutionize effective cancer treatment and early cancer detection. The objective of this study is to show how magnetic iron oxide nanoparticles and photoluminescent gold nanoclusters (MN‐AuNCs) may be combined effectively to produce bimodal imaging nanoparticles that possess magnetic and optical properties and can be used for both magnetic resonance imaging and optical biopsy. These findings demonstrate that MN‐AuNCs, when exposed to visible light, also have the capability to produce singlet oxygen, which is necessary for photodynamic therapy of cancer. In addition, it shows that they are non‐toxic, accumulate inside the cells, and cause cell death during exposure to visible light. The creation of these MN‐AuNCs offers a novel remedy for the current shortcomings in cancer diagnosis and treatment. Background: Discovering new cytotoxic compounds has received significant attention due to the rise in drug resistance and the adverse effects associated with chemotherapy drugs. In this study, the cytoplasmic extract of Lactobacillus casei was used to produce iron oxide nanoparticles (Fe2o3 NPs), and the cytotoxic effects of NPs were investigated on MCF-7 and human embryonic kidney 293 (HEK-293) cells. Methods: The cytoplasmic extract of L. casei was mixed with 10³M iron sulfate solution and incubated for 3 weeks at 37 °C and 5% CO2. The coprecipitation method was used to synthesize chemical Fe2o3 NPs. The synthesis of NPs was studied by electron microscopy and X-ray diffraction (XRD) analysis, and the cytotoxic effects were evaluated with dilutions (10, 100, and 1000 µg/mL) on MCF-7 and HEK cells. Results: X-ray diffraction analysis and scanning electron microscopy presented the mean of NPs synthesized by the green method to be about 15 nm and their shape to be spherical, as well as the average of chemically synthesized NPs to be about 20 nm with cubic structure. Chemical and green synthesized NPs only at a concentration of 1000 µg/mL were able to significantly reduce the survival rate of normal HEK-293 cells; chemically synthesized NPs decreased MCF-7 cell survival only at 1000 µg/mL and green synthesis at 100 µg/mL and 1000 µg/mL. Conclusion: Generating Fe2o3 NPs is biologically safe using the green synthesis method and the cytoplasmic extract of L. casei, which may be a suitable candidate for the treatment of cancer cells. Highly monodispersed and colloidal magnetite nanoparticles (MNPs) with controlled particle sizes and biocompatible polymer coatings have recently revealed a good ability for magnetic fluid hyperthermia (MFH) applications. Herein, we focused on probing the thermal abilities of polyacrylic acid (PAA)-decorated MNPs (PMNPs) under alternating magnetic field (AMF) with different frequencies and field amplitudes. The as-prepared PMNPs were fully characterized using various techniques including TEM, FTIR, DLS, TGA, and VSM. The results illustrated uniform colloidal well-dispersed ultra-small MNPs (core = 5 nm) with hydrodynamic sizes of (DH = 60 nm) and superparamagnetic behaviour. PAA-MNPs in water showed a good self-heating efficiencies, particularly when minimal concentration of MNPs (2.5 mg/mL) were employed. Heating profiles depicted that hyperthermia temperatures (42 °C) can be reached in relatively short times and could rise up to 53 °C. Heating abilities and SAR values as functions of frequency, field amplitude of AMF and concentration were systematically investigated. A remarkable increase of SAR with decreasing concentration and increasing frequency as well as amplitude was found. For instance, SAR was found to be 36 W/g and 6.75 for concentration of 2.5 and 10 mg/ml, respectively. The in vivo imaging of the drug distribution during cancer treatment plays an important role in helping physicians and pharmacists to monitor the progress of treatment and improve the effectiveness of therapy. Our goal in this work was to prepare a multifunctional nano drug delivery system that not only co‐loaded iron oxide nanoparticles (Fe3O4 NPs) and Doxorubicin (Dox) but also attached the near‐infrared fluorescent agent Cyanine 5.5 based on poly(lactide)‐tocopheryl polyethylene glycol 1000 succinate (PLA‐TPGS) copolymer. Cyanine 5.5 was attached to the surface Fe3O4 NPs via the NH−CO chemical bond, followed by co‐loading of Dox onto the PLA‐TPGS copolymer by the emulsion solvent evaporation method. The structural, morphological, dimensional, stability, and optical‐magnetic properties of the as‐synthesized nanosystem were fully characterized. From the drug‐released study, the prolonged release of Dox accelerated with increasing acidic conditions (at pH 5.0 and 6.5), while a slow release was observed at pH 7.4. Through the MTT assay, the nanosystem had cellular toxicity against Hep−G2 and HeLa cell lines with low IC50 values of 0.42 and 0.78 μg.mL−1 in terms of Dox concentration, respectively. At a magnetic field of 7 T, this nanosystem also exhibited a high r2/r1 ratio of 164.5 and a strong contrast magnetic resonance (MR) image in the liver. In vivo biodistribution studies revealed that the highest fluorescence intensity achived in the liver after 6 h, and the nanosystem was eliminated from the body after at least 24 h. Collectively, these results demonstrated that the PLA‐TPGS‐Fe3O4‐Cyanine 5.5‐Dox nanosystem is a good candidate for simultaneously increasing contrast for MR and near‐infrared fluorescence imaging. Background: Discovering new cytotoxic compounds has received significant attention due to the rise in drug resistance and the adverse effects associated with chemotherapy drugs. In this study, the cytoplasmic extract of Lactobacillus casei was used to produce iron oxide nanoparticles (Fe2o3 NPs), and the cytotoxic effects of NPs were investigated on MCF-7 and human embryonic kidney 293 (HEK-293) cells. Methods: The cytoplasmic extract of L. casei was mixed with 103M iron sulfate solution and incubated for 3 weeks at 37 °C and 5% CO2. The coprecipitation method was used to synthesize chemical Fe2o3 NPs. The synthesis of NPs was studied by electron microscopy and X-ray diffraction (XRD) analysis, and the cytotoxic effects were evaluated with dilutions (10, 100, and 1000 µg/mL) on MCF-7 and HEK cells. Results: X-ray diffraction analysis and scanning electron microscopy presented the mean of NPs synthesized by the green method to be about 15 nm and their shape to be spherical, as well as the average of chemically synthesized NPs to be about 20 nm with cubic structure. Chemical and green synthesized NPs only at a concentration of 1000 µg/mL were able to significantly reduce the survival rate of normal HEK-293 cells; chemically synthesized NPs decreased MCF-7 cell survival only at 1000 µg/mL and green synthesis at 100 µg/mL and 1000 µg/mL. Conclusion: Generating Fe2o3 NPs is biologically safe using the green synthesis method and the cytoplasmic extract of L. casei, which may be a suitable candidate for the treatment of cancer cells.

Abstract Background Despite widespread study of dendritic cell (DC)-based cancer immunotherapies, the in vivo postinjection fate of DC remains largely unknown. Due in part to a lack of quantifiable imaging modalities, this is troubling as the amount of DC migration to secondary lymphoid organs correlates with therapeutic efficacy. Magnetic particle imaging (MPI) has emerged as a suitable modality to quantify in vivo migration of superparamagnetic iron oxide (SPIO)-labeled DC. Herein, we describe a popliteal lymph node (pLN)-focused MPI scan to quantify DC in vivo migration accurately and consistently. Methods Adenovirus (Ad)-transduced SPIO+ (Ad SPIO+) and SPIO+ C57BL/6 bone marrow-derived DC were generated and assessed for viability and phenotype, then fluorescently labeled and injected into mouse hind footpads (n = 6). Two days later, in vivo DC migration was quantified using whole animal, pLN-focused, and ex vivo pLN MPI scans. Results No significant differences in viability, phenotype and in vivo pLN migration were noted for Ad SPIO+ and SPIO+ DC. Day 2 pLN-focused MPI quantified DC migration in all instances while whole animal MPI only quantified pLN migration in 75% of cases. Ex vivo MPI and fluorescence microscopy confirmed that pLN MPI signal was due to originally injected Ad SPIO+ and SPIO+ DC. Conclusion We overcame a reported limitation of MPI by using a pLN-focused MPI scan to quantify pLN-migrated Ad SPIO+ and SPIO+ DC in 100% of cases and detected as few as 1000 DC (4.4 ng Fe) in vivo. MPI is a suitable preclinical imaging modality to assess DC-based cancer immunotherapeutic efficacy. Relevance statement Tracking the in vivo fate of DC using noninvasive quantifiable magnetic particle imaging can potentially serve as a surrogate marker of therapeutic effectiveness. Textile industry effluents contain several hazardous substances, such as dye-containing effluents, which pose environmental and aesthetic challenges. Presently, the microbial-based remediation process is in use. This study investigated the application of ferrous–ferric oxide (Fe3O4) nanoparticles, a readily formulated nanoadsorbent, to remove scattered dye molecules from industrial effluents. The ferrous–ferric oxide nanoparticles were prepared using a chemical co-precipitation method. The nanoparticles had 26.93 emu g−1 magnetization, with sizes smaller than 20 nm, and possessed a highly purified cubic spinel crystallite structure. The catalytic activity of the iron oxide depended on the dose, photocatalytic enhancer, i.e., H2O2 level, pH of the reaction medium, and dye concentration. We optimized the Fenton-like reaction to work best using 1.0 g/L of ferrous–ferric oxide nanoparticles, 60 mM oxalic acid at pH 7.0, and 60 ppm of dye. Iron oxides act as photocatalysts, and oxalic acid generates electron–hole pairs. Consequently, higher amounts of super-radicals cause the rapid degradation of dye and pseudo-first-order reactions. Liquid chromatography–mass spectrometry (LC-MS) analysis revealed the ferrous–ferric oxide nanoparticles decolorized and destroyed Disperse Red 277 in 180 min under visible light. Hence, complete demineralization is observed using a photo-Fenton-like reaction within 3 h under visible light. These high-capacity, easy-to-separate next-generation adsorption systems are suggested to be suitable for industrial-scale use. Ferrous–ferric oxide nanoparticles with increased adsorption and magnetic properties could be utilized to clean environmental pollution.

Single-walled carbon nanotubes (SWCNTs) loaded with magnetic iron oxide nanoparticles were prepared by the arc discharge method and air heat treatment. The nanocomposite was characterized by X-ray diffraction, scanning electron microscopy, Raman spectroscopy, vibrating sample magnetometry, etc. The results showed that the heat-treated nanocomposites (SWCNTs/FexOy) had iron oxide phases and superparamagnetic properties with a saturation magnetization of 33.32 emu/g. Compared with the non-heat-treated materials, SWCNTs/FexOy had a larger specific surface area and pore volume. Using SWCNTs/FexOy to remove the organic contaminant (bisphenol A, BPA), it was found that under the conditions of pH = 3 and adsorbent dosage of 0.2 g/L, the maximum adsorption capacity of the composite was 117 mg/g, and the adsorption could reach more than 90% in only 5 min when the BPA content was below 0.05 mmol/L. The fitting results of the Langmuir and D-R models are more consistent with the experimental data, indicating a relatively uniform distribution of the adsorption sites and that the adsorption process is more consistent with physical adsorption. The kinetic calculations showed that the SWCNTs/FexOy exhibits chemical effects on both the surface and the gap, and the adsorption process is controlled by the π-π bonds and the hydrophobicity of the SWCNTs/FexOy.

Abstract Nanomedicine presents exciting new opportunities for the detection and treatment of cancer. Current cancer imaging methods and treatment approaches in clinics frequently fall short of entirely curing cancer and can have severe side effects. Theranostic nanoparticles, however, have the potential to revolutionize effective cancer treatment and early cancer detection. The objective of this study is to show how magnetic iron oxide nanoparticles and photoluminescent gold nanoclusters (MN‐AuNCs) may be combined effectively to produce bimodal imaging nanoparticles that possess magnetic and optical properties and can be used for both magnetic resonance imaging and optical biopsy. These findings demonstrate that MN‐AuNCs, when exposed to visible light, also have the capability to produce singlet oxygen, which is necessary for photodynamic therapy of cancer. In addition, it shows that they are non‐toxic, accumulate inside the cells, and cause cell death during exposure to visible light. The creation of these MN‐AuNCs offers a novel remedy for the current shortcomings in cancer diagnosis and treatment. Since they have both therapeutic and imaging capabilities, MN‐AuNCs have the potential to improve patient outcomes while lowering the risk of negative side effects.

Background: Drug resistance in cancer cells is a major concern in chemotherapy. Cisplatin (CIS) is one of the most effective chemotherapeutics for ovarian cancer. Here, we investigated an experimental approach to increase CIS cytotoxicity and overcome cell resistance using nanoparticle-based combination treatments. Methods: Polyethylenimine (PEI)-based magnetic iron oxide nanocomplexes were used for drug delivery in genetically matched CIS-resistant (A2780/CP) and -sensitive (A2780) ovarian cancer cells in the presence of a 20 mT static magnetic field. Magnetic nanoparticles (MNPs) were synthesized and bonded to PEI cationic polymers to form binary complexes (PM). The binding of CIS to the PM binary complexes resulted in the formation of ternary complexes PM/C (PEI-- MNP/CIS) and PMC (PEI--MNP--CIS). Results: CIS cytotoxicity increased at different concentrations of CIS and PEI in all binary and ternary delivery systems over time. Additionally, CIS induced cell cycle arrest in the S and G2/M phases and reactive oxygen species production in both cell lines. Ternary complexes were more effective than binary complexes at promoting apoptosis in the treated cells. Conclusion: PEI-based magnetic nanocomplexes can be considered novel carriers for increasing CIS cytotoxicity and likely overcoming drug resistance of ovarian cancer cells. [ABSTRACT FROM AUTHOR]

Graphene-based nanomaterials (GBNMs), specifically graphene oxide (GO) and reduced graphene oxide (rGO), have shown great potential in cancer therapy owing to their physicochemical properties. As GO and rGO strongly absorb light in the near-infrared (NIR) region, they are useful in photothermal therapy (PTT) for cancer treatment. However, despite the structural similarities of GO and rGO, they exhibit different influences on anticancer treatment due to their different photothermal capacities. In this review, various characterization techniques used to compare the structural features of GO and rGO are first outlined. Then, a comprehensive summary and discussion of the applicability of GBNMs in the context of PTT for diverse cancer types are presented. This discussion includes the integration of PTT with secondary therapeutic strategies, with a particular focus on the photothermal capacity achieved through near-infrared irradiation parameters and the modifications implemented. Furthermore, a dedicated section is devoted to studies on hybrid magnetic-GBNMs. Finally, the challenges and prospects associated with the utilization of GBNM in PTT, with a primary emphasis on the potential for clinical translation, are addressed. Background and aim: The present study was conducted to evaluate the diagnostic accuracy of Superparamagnetic iron oxide nanoparticles in breast cancer.Material and methods: We examined all international databases, including PubMed, Scopus, Science Direct, ISI, Web of Knowledge, and Embase, searching until January 2023 based on keywords related to the objectives of the study. Based on the PRISMA 2020 checklist, the current study was conducted, and the Google Scholar search engine was also used to find related articles. A fixed-effect model and inverse-variance method were used to calculate the 95% confidence interval risk ratio. The meta-analysis was conducted using Stata/MP v. 17 software.Results: The abstracts of 477 articles were reviewed, and 51 articles were selected for full-text review, of which 15 articles were included in the meta-analysis. The risk ratio of patient detection rate between superparamagnetic iron oxide nanoparticles and the standard method was 1.02 (RR, 1.02 95% CI 0.52, 1.53; p>0.05). Superparamagnetic iron oxide nanoparticles (SPION) have attracted great attention not only for therapeutic applications but also as an alternative magnetic resonance imaging (MRI) contrast agent that helps visualize liver tumors during MRI-guided stereotactic body radiotherapy (SBRT). SPION can provide functional imaging of liver parenchyma based upon its uptake by the hepatic resident macrophages or Kupffer cells with a relative enhancement of malignant tumors that lack Kupffer cells. However, the radiomodulating properties of SPION on liver macrophages are not known. Utilizing human monocytic THP-1 undifferentiated and differentiated cells, we characterized the effect of ferumoxytol (Feraheme®), a carbohydrate-coated ultrasmall SPION agent at clinically relevant concentration and therapeutically relevant doses of gamma radiation on cultured cells in vitro. We showed that ferumoxytol affected both monocytes and macrophages, increased the resistance of monocytes to radiation-induced cell death and inhibition of cell activity, and supported the anti-inflammatory phenotype of human macrophages under radiation. Its effect on human cells depended on the duration of SPION uptake and was radiation dose-dependent. The results of this pilot study support a strong mechanism-based optimization of SPION-enhanced MRI-guided liver SBRT for primary and metastatic liver tumors, especially in patients with liver cirrhosis awaiting a liver transplant.

This study synthesizes magnetic iron oxide nanoparticles from agro-waste sweet pepper extract, exploring their potential as antioxidant additives and in food preservation. Iron (III) chloride hexahydrate is the precursor, with sweet pepper extract as both a reducing and capping agent at pH 7.5. Characterization techniques, including microscopy and spectroscopy, analyze the sweet pepper extract-magnetic iron oxide nanoparticles. Antioxidant capacities against 2,2-diphenyl-1-picrylhydrazyl are assessed, incorporating nanoparticles into banana-based bioplastic for grape preservation. Microscopy reveals cubic and quasi-spherical structures, and spectroscopy confirms functional groups, including Fe–O bonds. X-ray diffraction identifies cubic and monoclinic magnetite with a monoclinic hematite presence. Sweet pepper extract exhibits 100% inhibitory activity in 20 min, while sweet pepper extract-magnetic iron oxide nanoparticles show an IC50 of 128.1 µg/mL. Furthermore, these nanoparticles, stabilized with banana-based bioplastic, effectively preserve grapes, resulting in a 27.4% lower weight loss rate after 144 h compared to the control group (34.6%). This pioneering study encourages institutional research into the natural antioxidant properties of agro-waste sweet pepper combined with magnetic iron and other metal oxide nanoparticles, offering sustainable solutions for nanopackaging and food preservation. Current research focuses on refining experimental parameters and investigating diverse applications for sweet pepper extract-magnetic iron oxide nanoparticles in varied contexts.

In pursuit of a preventive therapeutic for maternal autoantibody-related (MAR) autism, we assessed the toxicity, biodistribution, and clearance of a MAR specific peptide-functionalized dextran iron oxide nanoparticle system in pregnant murine dams. We previously synthesized ~15 nm citrate-coated dextran iron oxide nanoparticles (DIONPs), surface-modified with polyethylene glycol and MAR peptides to produce systems for nanoparticle-based autoantibody reception and entrapments (SNAREs). First, we investigated their immunogenicity and MAR lactate dehydrogenase B antibody uptake in murine serum in vitro. To assess biodistribution and toxicity, as well as systemic effects, we performed in vivo clinical and post mortem pathological evaluations. We observed minimal production of inflammatory cytokines-interleukin 10 (IL-10) and IL-12 following in vitro exposure of macrophages to SNAREs. We established the maximum tolerated dose of SNAREs to be 150 mg/kg at which deposition of iron was evident in the liver and lungs by histology and magnetic resonance imaging but no concurrent evidence of liver toxicity or lung infarction was detected. Further, SNAREs exhibited slower clearance from the maternal blood in pregnant dams compared to DIONPs based on serum total iron concentration. These findings demonstrated that the SNAREs have a prolonged presence in the blood and are safe for use in pregnant mice as evidenced by no associated organ damage, failure, inflammation, and fetal mortality. Determination of the MTD dose sets the basis for future studies investigating the efficacy of our nanoparticle formulation in a MAR autism mouse model.

Abstract Background Current pre-operative Sentinel Lymph Node (SLN) mapping using dual tracing is associated with drawbacks (radiation exposure, logistic challenges). Superparamagnetic iron oxide (SPIO) is a non-inferior alternative for SLN mapping in breast cancer patients. Limited research has been performed on SPIO use and pre-operative MRI in melanoma patients to identify SLNs. Methods Healthy participants underwent MRI-scanning pre- and post SPIO-injection during 20 min. Workflow protocols varied in dosage, massage duration, route of administration and injection sites. The first lymph node showing a susceptibility artefact caused by SPIO accumulation was considered as SLN. Results Artefacts were identified in 5/6 participants. Two participants received a 0.5 ml subcutaneous injection and 30-s massage, of which one showed an artefact after one hour. Four participants received a 1.0 ml intracutaneous injection and two-minute massage, leading to artefacts in all participants. All SLNs were observed within five minutes, except after lower limb injection (30 min). Conclusion SPIO and pre-operative MRI-scanning seems to be a promising alternative for SLN visualization in melanoma patients. An intracutaneous injection of 1.0 ml SPIO tracer, followed by a two-minute massage seems to be the most effective technique, simplifying the pre-operative pathway. Introduction: Due to the side effects of drugs, the development of nanoscale drug delivery systems has led to a significant improvement in medicinal therapies due to drug pharmacokinetics changes, decreased toxicity, and increased half-life of the drug. This study aimed to synthesize tamoxifen (TMX)-loaded L-lysine coated magnetic iron oxide nanoparticles as a nano-carrier to investigate its cytotoxic effects and anti-cancer properties against MCF-7 cancer cells. Methods: Magnetic Fe3O4 nanoparticles were synthesized and coated with L-lysine (F-Lys NPs). Then, TMX was loaded onto these NPs. The characteristics of synthesized nanoparticles (F-Lys-TMX NPs) were evaluated by X-ray diffraction (XRD), Fourier-transform infrared spectroscopy (FTIR), scanning electron microscopy (SEM), transmission electron microscopy (TEM), dynamic light scattering (DLS), differential scanning calorimetry (DSC), vibrating sample magnetometer (VSM), and thermogravimetric analysis (TGA). The drug release was analyzed at pH 5.8 and pH 7.4. The MCF-7 cells were exposed to F-Lys-TMX NPs, F-Lys NPs, and TMX for 24, 48, and 72 hours. To evaluate the cytotoxic potential of designed nanoparticles, MTT and apoptosis assays, real-time PCR, and cell cycle analysis was carried out. Results: The F-Lys-TMX NPs had spherical morphology with a size ranging from 9 to 30 nm. By increasing the nanoparticles concentration and treatment time, more cell proliferation inhibition and apoptosis induction were observed in F-Lys-TMX NPs-treated cells compared to the TMX. The expression levels of ERBB2, cyclin D1, and cyclin E genes were down-regulated and expression levels of the caspase-3 and caspase-9 genes were up-regulated. Studies on the drug release revealed a slow and controlled pH-dependent release of the nanoparticles. Cell cycle analysis indicated that F-Lys-TMX NPs could arrest the cells at the G0/G1 phase. Conclusion: The findings suggest that F-Lys-TMX NPs are more effective and have the potential for cell proliferation inhibition and apoptosis induction compared to the TMX. Hence, F-Lys-TMX NPs can be considered as an anti-cancer agent against MCF-7 breast cancer cells.

In clinical applications for cancer treatment, chemotherapy coupled with thermotherapy is highly considered. The development of multifunctional nanocomposite materials is an appealing strategy for use in various applications including biomedical applications. We present the preparation of dopamine-modified mesoporous silica material, in which magnetic iron oxide nanoparticles (FeNP) were grown onto the outer surface via the complexation of iron (Fe(III) and Fe(II)) ions with the dopamine groups modified on the silica hybrid and subsequent chemical reduction approaches. The prepared magnetic iron oxide incorporated with mesoporous silica hybrid composite nanoparticles (FeNP@MSHC NPs) had a large surface area (346 m2/g), pore size (3.2 nm), and pore volume (0.048 cm3/g). The formation of FeNP on the outer surface of the FeNP@MSHC NPs results in superparamagnetic characteristics. Furthermore, the prepared FeNP@MSHC NPs have a high drug (Dox) loading capacity (~62%) as well as pH- and temperature-responsive drug release efficiency. In addition, the MTT assay result shows the biocompatibility of the prepared FeNP@MSHC NPs. As a result, the FeNP@MSHC NPs could be utilized in cancer treatment for pH and temperature-sensitive delivery of chemotherapeutic agents to the target sites. [ABSTRACT FROM AUTHOR]

The removal of pollutants, such as heavy metals, aromatic compounds, dyes, pesticides and pharmaceuticals, from water is still an open challenge. Many methods have been developed and exploited for the purification of water from contaminants, including photocatalytic degradation, biological treatment, adsorption and chemical precipitation. Absorption-based techniques are still considered among the most efficient and commonly used approaches thanks to their operational simplicity. In recent years, polydopamine-coated magnetic nanoparticles have emerged for the uptake of heavy metals in water treatment, since they combine specific affinity towards pollutants and magnetic separation capacity. In this context, this work focuses on the synthesis of polydopamine (PDA)-coated Super Paramagnetic Iron Oxide Nanoparticles (PDA@SPIONs) as adsorbents for Cu2+ ions, designed to serve as functional nanostructures for the removal of Cu2+ from water by applying a magnetic field. The synthetic parameters, including the amount of SPIONs and PDA, were thoroughly investigated to define their effects on the nanostructure features and properties. Subsequently, the ability of the magnetic nanostructures to bind metal ions was assessed on Cu2+-containing solutions. A systematic investigation of the prepared functional nanostructures was carried out by means of complementary spectroscopic, morphological and magnetic techniques. Inductively coupled plasma atomic emission spectroscopy (ICP-AES) measurements were performed in order to estimate the Cu2+ binding ability. Introduction: Here, we assess the therapeutic effects of photobiomodulation (PBM) and curcumin (CUR)-loaded superparamagnetic iron oxide nanoparticles (SPIONs), alone or together, on the maturation step of a type 1 diabetes (DM1) rat wound model. Methods: Full-thickness wounds were inflicted in 36 rats with diabetes mellitus (DM) induced by the administration of streptozotocin (STZ). The rats were randomly allocated to four groups. Group one was untreated (control); group two received CUR; group 3 received PBM (890 nm, 80 Hz, 0.2 J/cm2); group 4 received a combination of PBM plus CUR. On days 0, 4, 7, and 15, we measured microbial flora, wound closure fraction, tensile strength, and stereological analysis. Results: All treatment groups showed a substantial escalation in the wound closure rate, a substantial reduction in the count of methicillin-resistant Staphylococcus aureus (MRSA), a substantial improvement in wound strength, a substantially improvement in stereological parameters compared to the control group, however, the PBM + CUR group was superior to the other treatment groups (all, P ≤ 0.05). Conclusion: All treatment groups showed significantly improved wound healing in the DM1 rat model. In the present study, we explored whether magnetic iron oxide nanoparticles (MNPs-Fe3O4) can be used to alleviate the toxicity of 3-nitrophenol (3-NP) to rice (Oryza sativa L.) seedlings grown under hydroponic conditions. The results showed that 3-NP from 7 to 560 μM decreased the growth, photochemical activity of the photosystem II (PS II), and chlorophyll content of the seedlings in a concentration-dependent manner. In the presence of 3-NP, 2,000 mg L−1 MNPs-Fe3O4 were added to the growth medium as the absorbents of 3-NP and then were separated with a magnet. The emergence of MNPs-Fe3O4 effectively alleviated the negative effects of 3-NP on rice seedlings. In addition, the long-term presence of MNPs-Fe3O4 (from 100 to 2,000 mg L−1) in the growth medium enhanced the growth, production of reactive oxygen species (ROS), activities of antioxidant enzymes, photochemical activity of PS II, and chlorophyll content of the rice seedlings. These results suggest that MNPs-Fe3O4 could be used as potential additives to relieve the physiological toxicity of 3-NP to rice seedlings.

Establishing a rapid in vitro evaluation system for drug screening is essential for the development of new drugs. To reproduce tissues/organs with functions closer to living organisms, in vitro three-dimensional (3D) culture evaluation using microfabrication technology has been reported in recent years. Culture on patterned substrates with controlled hydrophilic and hydrophobic regions (Cell-ableTM) can create 3D liver models (miniature livers) with liver-specific Disse luminal structures and functions. MRI contrast agents are widely used as safe and minimally invasive diagnostic methods. We focused on anionic polysaccharide magnetic iron oxide nanoparticles (Resovist®) and synthesized the four types of nanoparticle derivatives with different properties. Cationic nanoparticles (TMADM) can be used to label target cells in a short time and have been successfully visualized in vivo. In this study, we examined the morphology of various nanoparticles. The morphology of various nanoparticles showed relatively smooth-edged spherical shapes. As 3D liver models, we prepared primary hepatocyte–endothelial cell heterospheroids. The toxicity, CYP3A, and albumin secretory capacity were evaluated in the heterospheroids labeled with various nanoparticles. As the culture period progressed, the heterospheroids labeled with anionic and cationic nanoparticles showed lower liver function than non-labeled heterospheroids. In the future, there is a need to improve the method of creation of artificial 3D liver or to design a low-invasive MRI contrast agent to label the artificial 3D liver. [ABSTRACT FROM AUTHOR]

Background: Current pre-operative Sentinel Lymph Node (SLN) mapping using dual tracing is associated with drawbacks (radiation exposure, logistic challenges). Superparamagnetic iron oxide (SPIO) is a non-inferior alternative for SLN mapping in breast cancer patients. Limited research has been performed on SPIO use and pre-operative MRI in melanoma patients to identify SLNs.  METHODS: Healthy participants underwent MRI-scanning pre- and post SPIO-injection during 20 min. Workflow protocols varied in dosage, massage duration, route of administration and injection sites. The first lymph node showing a susceptibility artefact caused by SPIO accumulation was considered as SLN.Results: Artefacts were identified in 5/6 participants. Two participants received a 0.5 ml subcutaneous injection and 30-s massage, of which one showed an artefact after one hour. Four participants received a 1.0 ml intracutaneous injection and two-minute massage, leading to artefacts in all participants. All SLNs were observed within five minutes, except after lower limb injection (30 min).Conclusion: SPIO and pre-operative MRI-scanning seems to be a promising alternative for SLN visualization in melanoma patients. An intracutaneous injection of 1.0 ml SPIO tracer, followed by a two-minute massage seems to be the most effective technique, simplifying the pre-operative pathway. Graphene-based nanomaterials (GBNMs), specifically graphene oxide (GO) and reduced graphene oxide (rGO), have shown great potential in cancer therapy owing to their physicochemical properties. As GO and rGO strongly absorb light in the near-infrared (NIR) region, they are useful in photothermal therapy (PTT) for cancer treatment. However, despite the structural similarities of GO and rGO, they exhibit different influences on anticancer treatment due to their different photothermal capacities. In this review, various characterization techniques used to compare the structural features of GO and rGO are first outlined. Then, a comprehensive summary and discussion of the applicability of GBNMs in the context of PTT for diverse cancer types are presented. This discussion includes the integration of PTT with secondary therapeutic strategies, with a particular focus on the photothermal capacity achieved through near-infrared irradiation parameters and the modifications implemented. Furthermore, a dedicated section is devoted to studies on hybrid magnetic-GBNMs. Finally, the challenges and prospects associated with the utilization of GBNM in PTT, with a primary emphasis on the potential for clinical translation, are addressed.
