IntroductionSerotonin (5-HT) is critical for neurodevelopment and the serotonin transporter (SERT) modulates serotonin levels. Perturbed prenatal and postnatal dietary exposures affect the developing offspring predisposing to neurobehavioral disorders in the adult. We hypothesized that the postnatal brain 5-HT-SERT imbalance associated with gut dysbiosis forms the contributing gut-brain axis dependent mechanism responsible for such ultimate phenotypes.MethodsEmploying maternal diet restricted (IUGR, n=8) and high fat+high fructose (HFhf, n=6) dietary modifications, rodent brain serotonin was assessed temporally by ELISA and SERT by quantitative Western blot analysis. Simultaneously, colonic microbiome studies were performed.ResultsAt early postnatal (P) day 2 no changes in the IUGR, but a ~24% reduction in serotonin (p = 0.00005) in the HFhf group occurred, particularly in the males (p = 0.000007) revealing a male versus female difference (p = 0.006). No such changes in SERT concentrations emerged. At late P21 the IUGR group reared on HFhf (IUGR/HFhf, (n = 4) diet revealed increased serotonin by ~53% in males (p = 0.0001) and 36% in females (p = 0.023). While only females demonstrated a ~40% decrease in serotonin (p = 0.010), the males only trended lower without a significant change within the HFhf group (p = 0.146). SERT on the other hand was no different in HFhf or IUGR/RC, with only the female IUGR/HFhf revealing a 28% decrease (p = 0.036). In colonic microbiome studies, serotonin-producing Bacteriodes increased with decreased Lactobacillus at P2, while the serotonin-producing Streptococcus species increased in IUGR/HFhf at P21. Sex-specific changes emerged in association with brain serotonin or SERT in the case of Alistipase, Anaeroplasma, Blautia, Doria, Lactococcus, Proteus, and Roseburia genera.DiscussionWe conclude that an imbalanced 5-HT-SERT axis during postnatal brain development is sex-specific and induced by maternal dietary modifications related to postnatal gut dysbiosis. We speculate that these early changes albeit transient may permanently alter critical neural maturational processes affecting circuitry formation, thereby perturbing the neuropsychiatric equipoise.

The ever-growing need for improved in vitro models of human tissues to study both healthy and diseased states is advancing the use of techniques such as 3D Bioprinting. We here present our results on the development of a vascularized and perfusable 3D tumor mimic for studies of the early steps of Neuroblastoma metastatic spread. We used a multi-material and sacrificial bioprinting approach to fabricate vascularized 3D cell-laden structures and developed a customized perfusion system enabling maintenance of growth and viability of the constructs for up to 3 weeks. Cell phenotypes and densities in co-culture for both the bulk of the construct and the endothelialization of the vascular channels were optimized to better replicate in vivo conditions and ideally simulate tumor progression. We proved the formation of an endothelium layer lining the vascular channel after 14 days of perfused culture. Cells in the bulk of the construct reflected Neuroblastoma growth and its tendency to recruit endothelial cells contributing to neovascularization. We also collected preliminary evidence of Neuroblastoma cells migration towards the vascular compartment, recapitulating the first stages of metastatic dissemination.

Intra-Uterine Growth Restriction (IUGR) is a risk factor for many adult-onset chronic diseases, such as diabetes and obesity. These diseases are associated with intestinal microbiome perturbations (dysbiosis). The establishment of an intestinal microbiome begins in utero and continues postnatally (PN). Hypercaloric diet-induced dysbiosis is a major driver of childhood obesity. We hypothesized that different postnatal diets superimposed on IUGR will alter the postnatal intestinal microbiome. We compared four experimental rat groups: (1) Ad lib fed regular chow diet pre- and postnatally (CON), (2-3) IUGR induced by maternal caloric restriction prenatally followed postnatally (PN) by either (2) the control diet (IUGR-RC) or (3) High-Fat-high-fructose (IUGR-HFhf) diet, and lastly (4) HFhf ad lib pre- and postnatally (HFhf). Fecal samples were collected from dams and male and female rat offspring at postnatal day 2, 21, and adult day 180 for 16S rRNA gene sequencing. Maternal diet induced IUGR led to dysbiosis of the intestinal microbiome at PN21. Postnatal HFhf diet significantly reduced microbial diversity and worsened dysbiosis reflected by an increased Gammaproteobacteria/Clostridia ratio. Dysbiosis arising from a mismatch between IUGR and a postnatal HFhf diet may contribute to increased risk of the IUGR offspring for subsequent detrimental health problems.

The Mediterranean region is a hot spot for climate change, and cities of this area will be exposed to both increasing temperatures and decreasing precipitations. Green Infrastructures (GIs) can lower urban temperatures through evapotranspiration with an adequate soil moisture content. Grey water reuse can both guarantee the right soil moisture content and reduce freshwater exploitation. In order to test the effectiveness of soil moisture on reducing air temperature, two modelling simulations ran with the microclimate CFD-based model ENVI-met 4.0. The chosen day was a registered heat wave (7 July 2019) in Lecce, a city of south Italy, which was selected as case study for the Mediterranean area. The results demonstrated the effectiveness of soil moisture on evapotranspiration in reducing air temperature. From a circular economy perspective, the supply of grey water for urban GIs represents a strategic adaptation strategy to the expected effects of climate change on the Mediterranean basin.

The use of biomass can be a strategic way to realize a carbon-neutral energy plan, ensuring a fuel feedstock. Residual biomass arising from pruning is demonstrated to be an important energy resource in terms of quantity and quality. In the Salento peninsula, Apulia Region, in the south of Italy, a dramatic outbreak of Xylella fastidiosa has decimated olive trees since 2013, gaining a considerable amount of wood biomass. This paper, starting from the need to find a way to optimize the use of this available stock, reviews the main technologies on the utilization of olive wood for energy purposes. In particular, processes and products are here described, and an energy analysis compares lower heating value (LHV), higher heating value (HHV), mass yield, process operating conditions, and energy generated and spent by the process in order to find the most effective technology in order to optimize the energy use of olive biomass. The conclusions show the advantages and disadvantages of each technology. Pyrolysis performs well, showing the best results for both char HHV and syngas yield under different operating conditions. Gasification seems to be the most appropriate among conversion technologies to optimize olive tree pruning for energy purposes, as it can be used to produce both electrical and thermal energy. In terms of economic valorization, char is the most promising material representing a value-added product, the quality and versatility of which ranges from fuel to soil improvers and additives for the construction of supercapacitors. Conversely, its disadvantages are mainly represented by high ash content, which can slightly decrease the boiler efficiency. Finally, the amount of alkali metals can produce several problems, such as fouling, slagging, corrosion, etc., posing a challenge for combustion control and pollutant minimization.

Thyroid autoimmunity is extremely common in the adult population and can affect pregnancy outcomes. Signs in the newborn can range from absent to severe, making the diagnosis easy to miss. We present an interesting case of neonatal Graves disease associated with intrauterine growth restriction, premature delivery, and liver failure with severely high ferritin, thought to be secondary to hemochromatosis. Treatment of the underlying hyperthyroidism caused a rapid resolution of the elevated ferritin and liver failure. Abstract Objectives Assessment of the adaptive immune response against severe acute respiratory syndrome coronavirus 2 (SARS‐CoV‐2) is crucial for studying long‐term immunity and vaccine strategies. We quantified IFNγ‐secreting T cells reactive against the main viral SARS‐CoV‐2 antigens using a standardised enzyme‐linked immunospot assay (ELISpot). Methods Overlapping peptide pools built from the sequences of M, N and S viral proteins and a mix (MNS) were used as antigens. Using IFNγ T‐CoV‐Spot assay, we assessed T‐cell and antibody responses in mild, moderate and severe SARS‐CoV‐2 patients and in control samples collected before the outbreak. Results Specific T cells were assessed in 60 consecutive patients (mild, n = 26; moderate, n = 10; and severe patients, n = 24) during their follow‐up (median time from symptom onset [interquartile range]: 36 days [28;53]). T cells against M, N and S peptide pools were detected in n = 60 (100%), n = 56 (93.3%), n = 55 patients (91.7%), respectively. Using the MNS mix, IFNγ T‐CoV‐Spot assay showed a specificity of 96.7% (95% CI, 88.5–99.6%) and a specificity of 90.3% (75.2–98.0%). The frequency of reactive T cells observed with M, S and MNS mix pools correlated with severity and with levels of anti‐S1 and anti‐RBD serum antibodies. Conclusion IFNγ T‐CoV‐Spot assay is a reliable method to explore specific T cells in large cohorts of patients. This test may become a useful tool to assess the long‐lived memory T‐cell response after vaccination. Our study demonstrates that SARS‐CoV‐2 patients developing a severe disease achieve a higher adaptive immune response.

Rubber tree plantations may improve the soil’s physical and chemical properties, and they may sequester atmospheric carbon in the biomass or the soil. However, the potential role of these plantations in sequestering carbon in the soil and plant biomass has not been fully evaluated. This study evaluated rubber tree plantations at Paranapoema, which is located in the northwestern region of the Paraná state of Brazil, to measure the biomass in plantations of different ages and to determine the organic carbon content and d13C in the soils. Biomass accumulation was evaluated using the destructive method in plantations of different ages. The total carbon stock in the top 60 cm of the soil was 63.4 Mg C ha-1 for the pasture adjacent to the plantations and 66.8 and 79.3 Mg C ha-1 for the 4- and 15-year-old rubber tree plantations, respectively. These values are equivalent to an annual increase in soil carbon stocks of 0.85 and 1.06 Mg ha-1, respectively, and they do not include the accumulation of carbon as tree woody biomass. The soil d13C indicated a relatively fast conversion from the previous C4-C (pasture; Brachiaria-Urochloa brizantha) to C3-C (rubber tree). The results from this study suggest that rubber tree plantations have untapped potential to sequester carbon over a relatively short time period.

Utilizando técnica de aquisição automática de dados atmosféricos e de teor de água do solo, este trabalho quantificou o consumo diário de água em cultura do trigo em Latossolo Vermelho do município de Ponta Grossa, Estado do Paraná, durante o período de agosto a dezembro de 2003, procurando dar ênfase à contribuição das chuvas e dos fluxos ascendentes de água das camadas mais profundas do solo nesse consumo. Os resultados mostraram que no período monitorado: (a) a lâmina média diária de água evapotranspirada pela cultura do trigo foi de 6,75 mm, com o fluxo ascendente de água no perfil de solo contribuindo com 62 % desse total; (b) as taxas de evapotranspiração estimadas pelo método de Penman e pela equação do balanço hídrico (pedológico) se transladaram no tempo com simetria aproximadamente igual, mas com defasagem aproximada de sete dias, como se o solo respondesse às variações impostas pela atmosfera cerca de uma semana depois; (c) as chuvas tiveram efeito importante no armazenamento de água no solo, contribuindo para elevação das taxas evapotranspirativas; e (d) pelo fato de o potencial mátrico médio na zona das raízes ter-se apresentado próximo do limite crítico para a cultura, concluiu-se que a irrigação poderia produzir impactos potencialmente positivos para a cultura, por disponibilizar mais água no solo e garantir níveis evapotranspirativos mais altos, como é agronomicamente desejável.The daily water consumption of a wheat culture was quantified on an Oxisoil using atmospheric and soil data measured automatically on an experimental farm in Ponta Grossa, Paraná, Brazil. The measurement period was August through December 2003. The rain contribution to soil moisture and the vertical upward movement of water within the soil were particularly emphasized. Our results show that in the evaluated period (a) wheat evapotranspiration amounted to 6.75 mm a day, to which upward water flux contributed with 62 %; (b) soil water-budget evaporation lags behind Penman evapotranspiration estimates by about 7 days, as if the soil responded to atmospheric variations with a weekly delay; (c) as expected, rain significantly affected soil storage and usually raised evapotranspiration rates, and (d) given the fact that the mean matric potential in the root zone was near the critical value for wheat, the conclusion was drawn that irrigation would have potentially beneficial impacts on wheat in this region; more water would be available and higher levels of evapotranspiration warranted, which is desirable from the agricultural viewpoint.
