Simple Summary: Hepatocellular cancer (HCC) is the most common and lethal subtype of liver cancer without effective therapeutics. Understanding and targeting cancer stem cells (CSCs), a stem-cell-like subpopulation, which are emerging as effective ways to decipher tumor biology and develop therapies, may help to revolutionize cancer management. Cancer/testis antigen Maelstrom (MAEL) has been implicated in the regulation of CSC phenotypes, while the role of CSCs remains unclear. We demonstrated that MAEL positively regulates cancer stem-cell-like properties in HCC, and MAEL silencing provokes tumor cells' sensitivity to sorafenib. We further discovered that the MAEL-dependent stemness was operated via PGST2/IL8/AKT/STAT3 signaling. Collectively, our study suggests the MAEL/PGST2 axis as a potential therapeutic target against CSC and sorafenib resistance in HCC. Here, we found the upregulation of MAEL in HCC, with its expression negatively correlated with survival outcome. Functionally, MAEL promoted tumor cell aggressiveness, tumor stem-like potentials, and resistance to sorafenib in HCC cell lines. Transcriptional profiling indicated the dysregulation of stemness in MAEL knockout cells and identified PTGS2 as a critical downstream target transactivated by MAEL. The suppression effect of MAEL knockout in tumor aggressiveness was rescued in PTGS2 overexpression HCC cells. A molecular mechanism study revealed that the upregulation of PTGS2 by MAEL subsequently resulted in IL-8 secretion and the activation of AKT/NF-κB/STAT3 signaling. Collectively, our work identifies MAEL as an important stemness regulation gene in HCC. Targeting MAEL or its downstream molecules may provide a novel possibility for the elimination of CSC to enhance therapeutic efficacy for HCC patients in the future. [ABSTRACT FROM AUTHOR]

Background: Increasing evidence has indicated that Maelstrom (MAEL) plays an oncogenic role in various human carcinomas. However, the exact function and mechanisms by which MAEL acts in epithelial ovarian cancer (EOC) remain unclear. Results: This study demonstrated that MAEL was frequently overexpressed in EOC tissues and cell lines. Overexpression of MAEL was positively correlated with the histological grade of tumors, FIGO stage, and pT/pN/pM status (p < 0.05), and it also acted as an independent predictor of poor patient survival (p < 0.001). Ectopic overexpression of MAEL substantially promoted invasiveness/metastasis and induced epithelial-mesenchymal transition (EMT), whereas silencing MAEL by short hairpin RNA effectively inhibited its oncogenic function and attenuated EMT. Further study demonstrated that fibroblast growth factor receptor 4 (FGFR4) was a critical downstream target of MAEL in EOC, and the expression levels of FGFR4 were significantly associated with MAEL. Conclusion: Our findings suggest that overexpression of MAEL plays a crucial oncogenic role in the development and progression of EOC through the upregulation of FGFR4 and subsequent induction of EMT, and also provide new insights on its potential as a therapeutic target for EOC. Pachytene piRNAs are a class of Piwi-interacting small RNAs abundant in spermatids of the adult mouse testis. They are processed from piRNA primary transcripts by a poorly understood mechanism and, unlike fetal transposon-derived piRNAs, lack complementary targets in the spermatid transcriptome. We report that immunopurified complexes of a conserved piRNA pathway protein Maelstrom (MAEL) are enriched in MIWI (Piwi partner of pachytene piRNAs), Tudor-domain proteins and processing intermediates of pachytene piRNA primary transcripts. We provide evidence of functional significance of these complexes in Mael129 knockout mice that exhibit spermiogenic arrest with acrosome and flagellum malformation. Mael129-null mutant testes possess low levels of piRNAs derived from MAEL-associated piRNA precursors and exhibit reduced translation of numerous spermiogenic mRNAs including those encoding acrosome and flagellum proteins. These translation defects in haploid round spermatids are likely indirect, as neither MAEL nor piRNA precursors associate with polyribosomes, and they may arise from an imbalance between pachytene piRNAs and MIWI.

The spermatogenesis of animal is essential for the reproduction and a very large number of genes participate in this procession. The Maelstrom (Mael) is identified essential for spermatogenesis in both Drosophila and mouse, though the mechanisms appear to differ. It was initially found that Mael gene is necessary for axis specification of oocytes in Drosophila , and recent studies suggested that Mael participates in the piRNA pathway. In this study, we obtained Bombyx mori Mael mutants by using a binary transgenic CRISPR/Cas9 system and analyzed the function of Mael in B. mori , a model lepidopteran insect. The results showed that BmMael is not necessary for piRNA pathway in the ovary of silkworm, whereas it might be essential for transposon elements (TEs) repression in testis. The BmMael mutation resulted in male sterility, and further analysis established that BmMael was essential for spermatogenesis. The spermatogenesis defects occurred in the elongation stage and resulted in nuclei concentration arrest. RNA-seq and qRT-PCR analyses demonstrated that spermatogenesis defects were associated with tight junctions and apoptosis. We also found that BmMael was not involved in the silkworm sex determination pathway. Our data provide insights into the biological function of BmMael in male spermatogenesis and might be useful for developing novel methods to control lepidopteron pests. Terminal regions of the Drosophila embryo are patterned by the localized activation of the Torso-RTK pathway, which promotes the downregulation of Capicua. In the short-germ beetle Tribolium, the function of the terminal system appears to be rather different, as the pathway promotes axis elongation and, in addition, is required for patterning the extra-embryonic serosa at the anterior. Here, we show that Torso signalling also induces gene expression by relieving Capicua-mediated repression in Tribolium. Given that the majority of Torso target genes remain to be identified, we established a differential gene-expression screen. A subset of 50 putative terminal target genes was screened for functions in early embryonic patterning. Of those, 13 genes show early terminal expression domains and also phenotypes were related to terminal patterning. Among others, we found the PIWI-interacting RNA factor Maelstrom to be crucial for early embryonic polarization. Tc-mael is required for proper serosal size regulation and head morphogenesis. Moreover, Tc-mael promotes growth-zone formation and axis elongation. Our results suggest that posterior patterning by Torso may be realized through Maelstrom-dependent activation of posterior Wnt domains. [ABSTRACT FROM AUTHOR]

Povzetek V članku so obravnavane značilnosti družinskega življenja v kontekstu poznomodernih družbenih sprememb in predstavljeni različni izzivi, s katerimi se družine srečujejo v vsakdanjem življenju (delitev družinskega dela, usklajevanje družine in dela idr.). Predstavljena so vprašanja s posebnim poudarkom na slovenskem kontekstu in posebnostih, kot so dobro razvite sorodstvene podporne mreže, tradicija zaposlitve žensk za polni delovni čas ipd. Avtorica obravnava nekatere značilnosti vojaških družin in izpostavlja, da bi morale raziskave o vojaških družinah upoštevati poseben odnos med vojaško organizacijo in družino ter tudi družbene okoliščine, ki so v zadnjih desetletjih bistveno spremenile družinsko življenje, ter predlaga vpeljavo koncepta družinske prakse. Abstract This article deals with the characteristics of family life in the context of late-modern social changes, and presents various challenges that families are consequently facing in everyday life (among others, the division of family labour and the reconciliation of family and work). The author addresses these issues with a special emphasis on Slovenia, taking into account the specificities of the country. The main characteristics of military families are also presented. The author argues that research into military families should take into account the specific relationship between the military organization and the family, as well as the social circumstances that have shaped family life significantly in recent decades. L'œuvre d'Edgar Allan Poe contient une série d'éléments qui peuvent être décrits comme des espaces clos littéraires : ces espaces textuels ne s'ouvrent pas au mouvement libre, à la communication, à l'échange, mais ils restent fermés sur eux et sont marqués d'un mouvement en spirale. La présente contribution ambitionne de mettre en relief cette caractéristique de l'œuvre littéraire de Poe en se fondant en particulier sur le conte The Purloined Letter et prenant aussi en compte la traduction française du texte par Charles Baudelaire. Celle-ci sert en même temps de base pour une réflexion sur des aspects de traductibilité. L'article part d'une microanalyse du premier paragraphe du récit de Poe pour montrer à quel point ce passage est composé selon une logique de la circularité et de la fermeture. Dans une deuxième étape, l'examen des espaces clos s'étend au niveau du récit global ainsi qu'à d'autres textes de Poe. L'analyse se focalise en particulier sur la structure de la mise en abyme et sur les formes de la spirale, du tourbillon, du maelström comme géométries immanentes, autoréflexives de l'œuvre littéraire de Poe. The Lofoten Maelstrom has been known for centuries as one of the strongest open-ocean tidal currents in the world, estimated to reach 3 m s−1, and by some estimates as much as 5 m s−1. The strong current gives rise to choppy seas when waves enter the Moskenes Sound, making the area extremely difficult to navigate. Despite its reputation, few studies of its strength exist and no stationary in situ measurements for longer time periods have been made due to the challenging conditions. By deploying for the first time in situ wave and current instruments, we confirm some previous estimates of the strength of the current. We also show that its strength is strongly connected with wave breaking. From a consideration of specific forcing terms in the dynamical energy balance equation for waves on a variable current, we assess the impact of the underlying current using a convenient metric formulated as a function of the horizontal current gradients. We find that the horizontal gradients are a likely explanation for the observed enhanced wave breaking during strong currents at a rising tide.

Through the video is created a carnival, and the audience has become a carnival maelstrom of followers. We need to analyze what kind of operation and management model shapes short video app development behind the audience. It is not hard to argue that the terror attacks on September 11, 2001, and the subsequent horrific violent acts that have been carried out in the name of Islam in cities like, for example, Madrid, London, Paris, Brussels, Berlin, Nice or Stockholm have all had a serious impact on public perceptions of Islam and Muslims in the West. One way of understanding the outcome of these processes is to argue that they have contributed to and produced and strengthened what Riem Spielhaus and I call narratives of inclusion and exclusion (Larsson and Spielhaus 2013, 2017). In this article I will use narratives of inclusion and exclusion as a backdrop and heuristic tool for analysing and discussing the impacts of these two grand ideal-type narratives on museums planning to display so-called Islamic artefacts.

This paper explores spectatorial encounters with criminal trials. Particularly focusing on the 2018 work of Australian contemporary visual artist Julie Fragar that followed her watching murder trials in the Supreme Court of Queensland, it is argued that the artist as a legal outsider grapples with the inhumanity of the trial. This grappling can go in two directions. For some there is a need to bring the human back, to see the person beneath the mask of the role that they are performing, to connect the gothic horror of the trial back to a redeeming humanism. For others, and this is evident in Fragar, the horror of the inhumanity is visceral and overpowering, and the grotesque masque of judgment needs to be witnessed. Both perform a corrective and critique to the business as usual of processing, judging and stamping onto human lives that is legal insider’s performance in the criminal trial.

In 1887, amateur theatrical performer Cora Urquhart Brown Potter turned professional amid a maelstrom of international newspaper coverage. Newspapers picked up the story of her career, feeding a desire for salacious gossip at the expense of the elite celebrity cast as a fallen woman. Yet, Potter and the press developed a symbiotic relationship, as her non-traditional path to the stage required that she transform her personal celebrity into a professional one in order to attract audiences and bookings. The papers obliged and, as the story developed and her celebrity transformed, they shifted their coverage of Potter’s journey from society columns, to theatrical columns, to sensational front-page spreads. Potter’s early career paralleled and capitalised on such new developments in the newspaper industry and its messaging. While the press continued to sell her scandal, Potter used the papers to profit from her society past while forging her future as a theatre professional.

The inclusion 'ideal' is one which has been both celebrated and maligned. It paved the way for equality; but also, out of its diversity sprang many misconceptions and concerns for children with Special Educational Needs and Disabilities (SEND). It is within this maelstrom of difficulty that the Special Educational Needs Co‐ordinator (SENCo) role is placed, so that those undertaking the position of SENCo face numerous barriers in the pursuit of providing effectively for children with SEND. Semi‐structured interviews were conducted with fifteen SENCos employed at mainstream primary schools within the North West of England. Findings revealed conflict between how SENCos viewed their role and responsibilities, and the expectations that they perceived to be placed on them by parents of children with SEND and mainstream colleagues. Cet article s’attache à montrer l’usage d’un texte littéraire comme outil en situation d’enquête ethnographique et artistique. Une descente dans le maelström d’Edgar Allan Poe - qui met en scène une image possible des abîmes (le maelström) -, devient le moyen de penser des dangers futurs difficilement perceptibles dans le présent en proposant un modèle dynamique qui fluctue entre mondes confinés et « devenir-abîme » des mondes. L’emboitement des techniques narratives et des techniques d’entretiens offre la possibilité de dégager des techniques de perceptions et d’alerte, de même que les attachements nécessaires face à des devenirs mouvants et potentiellement catastrophiques. This article aims to show the use of a literary short story as a tool in ethnographic and artistic investigation situations. A descent into Edgar Allan Poe’s maelström - which stages a possible image of the abyss (the maelstrom) – allows to address future dangers that are difficult to perceive in the present by proposing a dynamic model that fluctuates between confined worlds and the « becoming-abyss » of worlds. The interweaving of narrative and interviewing techniques offers the possibility of identifying techniques of perception and alert, as well as the necessary attachments in the face of changing and potentially catastrophic developments.

Mael, which comprises a high mobility group box and a MAEL domain, is one of the key players in piRNA-mediated transposon silencing. However, the mechanism whereby Mael is involved in this pathway remains unknown. Recent biochemical and structural studies, along with bioinformatic analyses of Mael-associating RNAs in vivo, have shed light on the functional aspects of Mael in the piRNA pathway. Resumen La crisis económica mundial que empezó en 2008 ha generado una caída del poder adquisitivo, reducción del consumo y contracción de la economía. Sin embargo, en medio de la crisis, se desarrolla una vorágine tecnológica que va a la par de un nuevo capitalismo mediante el incremento del uso de dispositivos digitales. El objetivo de este trabajo es analizar las nuevas claves de un capitalismo tecnológico generado por la democratización del consumo de bienes y servicios digitales y una arqueología tecnológica con periodos de obsolescencia cada vez más cortos. Es relevante la comprensión de este tipo de cambios, ya que permiten observar la relación entre el hombre y la tecnología y los usos que hacen de ella, con un enfoque en mejorar su calidad de vida y cerrar brechas como lo es la digital. Este trabajo usa una metodología del tipo documental de datos secundarios a nivel mundial sobre consumo tecnológico y su uso a tra vés de datos provenientes de ITU, Cepal, Eurostat y otros. Además, se realizó un análisis temporal del Smartphone desde el 2008 para establecer el crecimiento de su uso, teniendo en cuenta que ésta una de las tecnologías más utilizadas. Entre los hallazgos más notorios se evidencia que más del 75 % de la población a nivel mundial cuenta con un teléfono móvil, concluyen-do que esta tecnología facilita la gestión de la vida cotidiana en todos los aspectos: laboral, familiar, educativo, social, etc., es así que el Smartphone se convierte en una “navaja suiza” que facilita el diario vivir del ser humano. Abstract The global economic crisis that started in 2008 has generated a decrease in purchasing power, reduced consumption and a contraction in the economy. However, in the midst of the crisis, a technological maelstrom develops, which goes hand in hand with a new capitalism through the increase in the use of digital devices. The objective of this work is to analyze the new keys to a technological capitalism generated by the democratization of the consumption of digital goods and services and a technological archeology with increasingly shorter periods of obsolescence. The understanding of these types of changes is relevant since they allow us to observe the relationship between man and technology and its usesthe uses made of it, with a focus on improving their quality of life and closing gaps, such as the digital one. This work uses a documentary type methodology of secondary worldwide data on technological consumption and its use through data from ITU, Cepal, Eurostat and others. In addition, a temporal analysis of the Smartphone was carried out since 2008 to establish its growth, since it is one of the most widely used technologies. Among the most notorious findings, it is evidenced that more than 75 % of the world population has a mobile phone, concluding that this technology facilitates the management of daily life in all aspects: work, family, educational, social, etc. This is how the Smartphone becomes a “Swiss Army knife” that facilitates the daily life of human beings.

Nevertheless, this is not Ducasse’s only borrowing from the work of the American short-story writer, whom he probably discovered by way of Baudelaire.

Chromatin of male and female gametes undergoes a number of reprogramming events during the transition from germ cell to embryonic developmental programs. Although the rearrangement of DNA methylation patterns occurring in the zygote has been extensively characterized, little is known about the dynamics of DNA modifications during spermatid maturation. Here, we demonstrate that the dynamics of 5-carboxylcytosine (5caC) correlate with active transcription of LINE-1 retroelements during murine spermiogenesis. We show that the open reading frames of active and evolutionary young LINE-1s are 5caC-enriched in round spermatids and 5caC is eliminated from LINE-1s and spermiogenesis-specific genes during spermatid maturation, being simultaneously retained at promoters and introns of developmental genes. Our results reveal an association of 5caC with activity of LINE-1 retrotransposons suggesting a potential direct role for this DNA modification in fine regulation of their transcription., Blythe et al investigate the patterns of oxidized forms of the 5-methylcytosine DNA modification during spermatogenesis. They find that open reading frames of evolutionarily young and transcriptionally active LINE-1 retrotransposons are enriched in 5-carboxylcytosine (5caC) during spermatid maturation, suggesting a link between 5cac and LINE-1 activity.

The medieval crusades were complicated; their memory even more so. Their reuse across popular culture is a maelstrom of images, tropes, and ideas, further exacerbated when added to the mushrooming world of historical gaming. Not only is it impossible to imagine any kinds of games which have no inaccuracies, it is no longer possible to imagine any which would be deemed as accurate; we are no longer in the realm of historical accuracy but authenticity, and the extent to which a given imagined past coincides with our individual memory of that past. By exploring a range of tropes and historical icons established over time and which contribute to the mythical capital of popular memory, this chapter will explore the ways in which historical games construct authenticity through a range of crusading icons which resonate with popular ideas and tropes about the crusades.

Pachytene pi RNAs are a class of Piwi-interacting small RNAs abundant in spermatids of the adult mouse testis. They are processed from pi RNA primary transcripts by a poorly understood mechanism and, unlike fetal transposon-derived pi RNAs, lack complementary targets in the spermatid transcriptome. We report that immunopurified complexes of a conserved pi RNA pathway protein Maelstrom ( MAEL) are enriched in MIWI (Piwi partner of pachytene piRNAs), Tudor-domain proteins and processing intermediates of pachytene pi RNA primary transcripts. We provide evidence of functional significance of these complexes in Mael 129 knockout mice that exhibit spermiogenic arrest with acrosome and flagellum malformation. Mael 129-null mutant testes possess low levels of pi RNAs derived from MAEL-associated piRNA precursors and exhibit reduced translation of numerous spermiogenic mRNAs including those encoding acrosome and flagellum proteins. These translation defects in haploid round spermatids are likely indirect, as neither MAEL nor pi RNA precursors associate with polyribosomes, and they may arise from an imbalance between pachytene pi RNAs and MIWI. This chapter discusses the concept of ”˜homeland” and the diagnostic category of ”˜homelessness” derived from it with which Heidegger describes the contemporary situation of man, the situation that is characterized essentially by the planetary rule of technology and the homelessness to which it has given rise. Heidegger claims to give thought to such homelessness and, at the same time, to recollect symbolic resources—such as the basic determination ”homeland”—that help man to keep his balance in the precarious situation of the present, in the “maelstrom of nihilism.” He does so in the framework of what he calls the thought of Being. Two figures appear in the horizon of Heideggerian thought and become determinative for his thematization of homeland and homelessness: Nietzsche and Holderlin. Heidegger responds to the “Pelagians” of the twentieth century, that is, those who believe that man is only a human problem and is in a position to save himself: “Only a god can save us.”.
