Pediatric brain and spinal cancers are collectively the leading disease-related cause of death in children; thus, we urgently need curative therapeutic strategies for these tumors. To accelerate such discoveries, the Childrens Brain Tumor Network (CBTN) and Pacific Pediatric Neuro-Oncology Consortium (PNOC) created a systematic process for tumor biobanking, model generation, and sequencing with immediate access to harmonized data. We leverage these data to establish OpenPBTA, an open collaborative project with over 40 scalable analysis modules that genomically characterize 1,074 pediatric brain tumors. Transcriptomic classification reveals universal TP53 dysregulation in mismatch repair-deficient hypermutant high-grade gliomas and TP53 loss as a significant marker for poor overall survival in ependymomas and H3 K28-mutant diffuse midline gliomas. Already being actively applied to other pediatric cancers and PNOC molecular tumor board decision-making, OpenPBTA is an invaluable resource to the pediatric oncology community.

The giant rorqual whales are believed to have a massive food turnover driven by a high-intake lunge feeding style aptly described as the worlds largest biomechanical action. This high-drag feeding behavior is thought to limit dive times and constrain rorquals to target only the densest prey patches, making them vulnerable to disturbance and habitat change. Using biologging tags to estimate energy expenditure as a function of feeding rates on 23 humpback whales, we show that lunge feeding is energetically cheap. Such inexpensive foraging means that rorquals are flexible in the quality of prey patches they exploit and therefore more resilient to environmental fluctuations and disturbance. As a consequence, the food turnover and hence the ecological role of these marine giants have likely been overestimated.

Although failures and other experiences can capture attention and motivate organizations to learn and improve, this knowledge is not always retained over time, leaving some organizations dangerously prone to repeat the same mistakes repeatedly. We adapt theory on the attention-based view—and specifically on attentional engagement and vigilance—to shed new light on this process. While prior research has examined how competing events may draw attention away, our theory leads us to consider the circumstances that will motivate employees to maintain attention on learning from failure that has already occurred. Our framework examines the conditions that preserve attention to past failures by increasing the perception that related issues exist elsewhere, serving as continuing reminders of or cues about the failure when attention begins to drift away. We find support for related hypotheses involving a failure's complexity, the firm's culpability, and the ongoing use of routines related to those involved in the failure. Our findings contribute to the attention-based view by developing theory about attentional engagement and vigilance and by emphasizing the conditions that can keep attention focused on, rather drawn away from, past failure. The considerable power needed for large whales to leap out of the water may represent the single most expensive burst maneuver found in nature. However, the mechanics and energetic costs associated with the breaching behaviors of large whales remain poorly understood. In this study we deployed whale-borne tags to measure the kinematics of breaching to test the hypothesis that these spectacular aerial displays are metabolically expensive. We found that breaching whales use variable underwater trajectories, and that high-emergence breaches are faster and require more energy than predatory lunges. The most expensive breaches approach the upper limits of vertebrate muscle performance, and the energetic cost of breaching is high enough that repeated breaching events may serve as honest signaling of body condition. Furthermore, the confluence of muscle contractile properties, hydrodynamics, and the high speeds required likely impose an upper limit to the body size and effectiveness of breaching whales.

In recent years, longer and heavier trains have become more common, primarily driven by efficiency and cost‐saving measures in the railroad industry. Regulation of train length is currently under consideration in the United States at both the federal and state levels, because of concerns that longer trains may have a higher risk of derailment, but the relationship between train length and risk of derailment is not yet well understood. In this study, we use data on freight train accidents during the 2013–2022 period from the Federal Railroad Administration (FRA) Rail Equipment Accident and Highway‐Rail Grade Crossing Accident databases to estimate the relationship between freight train length and the risk of derailment. We determine that longer trains do have a greater risk of derailment. Based on our analysis, running 100‐car trains is associated with 1.11 (95% confidence interval: 1.10–1.12) times the derailment odds of running 50‐car trains (or a 11% increase), even accounting for the fact that only half as many 100‐car trains would need to run. For 200‐car trains, the odds increase by 24% (odds ratio 1.24, 95% confidence interval: 1.20–1.28), again accounting for the need for fewer trains. Understanding derailment risk is an important component for evaluating the overall safety of the rail system and for the future development and regulation of freight rail transportation. Originally devised for cancer control, mRNA vaccines have risen to the forefront of medicine as effective instruments for control of infectious disease, notably their pivotal role in combating the COVID-19 pandemic. This review focuses on fundamental aspects of the development of mRNA vaccines, e.g., tumor antigens, vector design, and precise delivery methodologies, - highlighting key technological advances. The recent, promising success of personalized mRNA vaccines against pancreatic cancer and melanoma illustrates the potential value for other intractable, immunologically resistant, solid tumors, such as glioblastoma, as well as the potential for synergies with a combinatorial, immunotherapeutic approach. The impact and progress in human cancer, including pancreatic cancer, head and neck cancer, bladder cancer are reviewed, as are lessons learned from first-in-human CAR-T cell, DNA and dendritic cell vaccines targeting glioblastoma. Going forward, a roadmap is provided for the transformative potential of mRNA vaccines to advance cancer immunotherapy, with a particular focus on the opportunities and challenges of glioblastoma. The current landscape of glioblastoma immunotherapy and gene therapy is reviewed with an eye to combinatorial approaches harnessing RNA science. Preliminary preclinical and clinical data supports the concept that mRNA vaccines could be a viable, novel approach to prolong survival in patients with glioblastoma. [ABSTRACT FROM AUTHOR]

Renowned marine mammal scientist Roger Payne, who passed away in 2023 at the age of 88, is best known for his work on humpback whale songs, which helped change public perception of whales. His research on live whales at sea and his recordings of humpback songs played a significant role in the environmental conservation movement of the early 1970s. Payne's contributions extended beyond whales, as he conducted groundbreaking research on other species and was known for his ability to merge science, art, and music.
