Neonatal calf diarrhea is a frequent disease of calves and may result in dehydration and metabolic acidosis. The disease causes mortality and reduces growth and future productivity. Early identification of disease improves calf outcomes and thus there is increasing interest in technological methods for detecting disease. Dehydration leads to the blood becoming more concentrated and this can be measured using serum osmolality. Research in humans has shown that saliva conductivity is correlated with serum osmolality. Saliva conductivity may therefore offer a non-invasive opportunity to assess hydration status in calves. Furthermore, as blood pH is a prognostic indicator and there is ion exchange in the salivary ducts, saliva pH may act as an indicator of metabolic acidosis. This observational study aimed to assess the relationship of saliva conductivity and pH with the clinical and biochemical parameters of calves suffering from neonatal calf diarrhea. One hundred and forty-one dairy-bred calves were recruited onto the study at approximately 1 week of age. The health of the calves was assessed daily. Calves had blood and saliva samples taken weekly until 25 days of age or the development of neonatal calf diarrhea. When calves developed diarrhea, they were sampled for three consecutive days. Hematocrit, plasma total protein, saliva pH and saliva conductivity were measured at each sampling. Saliva pH and saliva conductivity were measured using portable meters ( LAQUAtwin-pH-33 and LAQUAtwin-EC22 ). In a subset of 30 matched samples, serum proteins and electrolytes were also measured. Saliva conductivity was not associated with diarrhea or dehydration. Saliva pH was lower in calves with diarrhea, regardless of hydration status. The Lin's concordance correlation coefficients between saliva variables and hematocrit and strong ion difference were negligible. Dehydrated calves with diarrhea had a higher hematocrit and albumin and the lowest sodium and SID. Calves with diarrhea and no dehydration had a lower plasma total protein. While saliva conductivity has been associated with measures of dehydration in humans, this does not appear to be the case in calves. Previous research has indicated that preweaned dairy calves reared in pairs compared with individually have improved performance and indicators of animal welfare. One hundred and thirty Holstein female calves completed the trial, with eighty-five being allocated to paired housing and forty-five calves being allocated to individual housing. Daily live weight gain (DLWG), treatments and mortality were recorded throughout the preweaning period. Salivary cortisol, latency to feed and latency to approach a novel object were assessed at batching. There were no significant differences in DLWG, mortality and disease treatments between the average of the pair and the individually housed calves, although the pair-reared calves were quicker to approach the milk feed after batching and interacted more quickly with a novel object. The heaviest born calves within the pair had the highest DLWG from birth to weaning, with a higher percentage of calves approaching the novel object, compared with the lightest born calf within the pair. This study shows that calves within a pair may have significantly different performance and welfare during the preweaning period, with the heavier calf outperforming and displaying less fear and more exploratory behaviour than the lighter calf within a pair.

Neosporosis and toxoplasmosis are major causes of abortion in livestock worldwide, leading to substantial economic losses. Detection tools are fundamental to the diagnosis and management of those diseases. Current immunohistochemistry (IHC) tests, using sera raised against whole parasite lysates, have not been able to distinguish between Toxoplasma gondii and Neospora caninum. We used T. gondii and N. caninum recombinant proteins, expressed in Escherichia coli and purified using insoluble conditions, to produce specific polyclonal rabbit antisera. We aimed to develop species-specific sera that could be used in IHC on formalin-fixed, paraffin-embedded (FFPE) tissue sections to improve the diagnosis of ruminant abortions caused by protozoa. Two polyclonal rabbit sera, raised against recombinant proteins, anti- Neospora -rNcSRS2 and anti- Toxoplasma -rTgSRS2, had specificity for the parasite they were raised against. We tested the specificity for each polyclonal serum using FFPE tissue sections known to be infected with T. gondii and N. caninum . The anti- Neospora -rNcSRS2 serum labeled specifically only N. caninum- infected tissue blocks, and the anti- Toxoplasma -rTgSRS2 serum was specific to only T. gondii- infected tissues. Moreover, tissues from 52 cattle and 19 sheep previously diagnosed by lesion profiles were tested using IHC with our polyclonal sera and PCR. The overall agreement between IHC and PCR was 90.1% for both polyclonal anti-rNcSRS2 and anti-rTgSRS2 sera. Background: Good stewardship of antibiotics can reduce the development and impact of antimicrobial resistance (AMR); therefore, understanding farmers' antibiotic use is of interest to stakeholders. To date, few qualitative studies have looked at farmers' antibiotic use on dairy farms in the UK., Methods: Semi-structured interviews were used to explore 15 Scottish dairy farmers' antibiotic use behaviours and the factors influencing their antibiotic use on farms., Results: Using an inductive process, the results from the interviews were analysed, and four key themes were extracted: use of antibiotics, awareness of AMR, determinants of antibiotic use and future aspirations., Limitations: Some of the farmers interviewed were wary about discussing their antibiotic use, which could mean that some contentious issues were not discussed. The farmers also all belonged to a single milk-buying group, which may limit the generalisability of the findings., Conclusions: The quantities of antibiotics used were felt to be driven by the disease prevalence on farms, cows being indoors more, increasing herd sizes resulting in increased stocking densities, retention of poorer cows and substandard housing. Farmer knowledge of antibiotics varied, and not all farmers interviewed were aware of AMR. Calf housing is naturally thermodynamic, with interactions between various elements such as wind speed, air temperature, and humidity. This study investigated the effect of the proportion of time for which calves were exposed to effective environmental temperatures below their lower critical temperature (LCT) on their daily liveweight gain (DLWG) within their first month of life. This study used the naturally occurring climatic environment, whereas other such studies have been conducted under climatically controlled conditions. Air temperature (°C), relative humidity (%), and wind speed (m/s) were recorded within the calf housing from birth until approximately 28 days of age, with calves being health-scored and weighed at regular intervals. Calves were housed from birth until 6-14 days old in individual hutches, and then moved into group housing igloo pens. Whilst individually housed, calves that spent less than 32% of their time below their LCT had a DLWG of 0.06 ± 0.34 kg/d (mean ± SE) compared to calves that spent more than 97% of their time below their LCT, which had a DLWG of -0.19 ± 0.045 kg/d. When group housed, calves that spent less than 1% of their time below their LCT had a DLWG of 0.59 ± 0.18 kg/d, whereas calves that spent more than 28% of their time below their LCT had a DLWG of 0.53 ± 0.23 kg/d. The proportion of time for which calves were exposed to effective environmental temperatures below their LCT had a significant effect on DLWG when calves were individually housed. Therefore, exposure to effective environmental temperatures below the LCT can be detrimental to the growth of the calf in the early stages of its life.

The aim of this study was to use an automated behavior-monitoring system to objectively assess the association between lying and activity behavior in the precalving, calving, and postcalving periods between multiparous and primiparous cows with (1) normocalcemia, (2) subclinical hypocalcemia, or (3) clinical hypocalcemia at calving. Behavioral data and blood serum samples were collected from 51 multiparous and 21 primiparous Holstein dairy cattle. Blood samples from the coccygeal vein were taken within 24 h of calving, and serum was analyzed to measure total calcium concentration. Cows were classified into one of 3 categories: normocalcemia (serum calcium concentration ≥ 2.0 mmol/L), subclinical hypocalcemia (serum calcium concentration < 2.0 mmol/L, absence of clinical signs), and clinical hypocalcemia (clinical signs and successful treatment). An activity sensor was fitted to the right hind leg of cows 3 wk before their expected calving date. Data for lying time, standing time, number of steps, and the total number of standing and lying bouts (postural transitions) were automatically collected and summed into 15-min blocks. Behavioral variables were summarized into 2-h and 24-h periods before analyses. Mixed effect models were used to analyze cow behavior in the entire 14 d before calving (d -14 to -1), on the day of calving, and the entire 21 d postcalving (d 1 to 21). In the precalving period, multiparous cows with normocalcemia had fewer postural transitions (18.5 ± 6.9 no./d) compared with cows with subclinical hypocalcemia (23.5 ± 8.0 no./d) and clinical hypocalcemia (23.5 ± 8.6 no./d). However, there was no association between blood calcium status on lying time (min/d) or step count (no./d) for multiparous cows. For primiparous cows, the step count of cows with subclinical hypocalcemia remained constant across the period, and the step count of cows with normocalcemia decreased from 842.8 steps/d on d -14 to 427.5 steps/d on d -1. Postpartum cows with clinical hypocalcemia were less active (fewer steps) and spent 88 min/d (1.5 h) and 125 min/d (2.1 h) more time lying down compared with cows with subclinical hypocalcemia and normocalcemia, respectively. Dairy cow farming plays an important role in the UK and worldwide economies. Significant challenges are currently being faced regarding sustainability of the dairy industry. Dairy cow subfertility remains an important issue limiting herd productivity, resulting in annual losses of hundreds of millions of pounds in the UK alone. To address this, accurate monitoring of reproductive status and early detection of fertility issues in individual cows is essential. The aim of this study was to gather farmer and veterinarian opinions on current practices and perceived gaps related to diagnosis of fertility issues and pregnancy testing in UK dairy farms. Using online questionnaires, data were collected and analyzed from a total of 40 farmers and 59 veterinarians. The results showed that non-seen bulling checks and ultrasound were the most frequent tools to detect fertility issues, and that most farmers tested post-calving, and often again before or during mating. Most farmers believed that current tests did not meet their expectations, with half of those being willing to pay more than they were currently paying for fertility testing. In regard to pregnancy testing, ultrasound was most commonly used, at 30-50 days post-insemination either in individual or groups of cows. Again, most farmers believed that current tests did not meet their expectations, and a majority would consider paying a higher cost for a test that was better than those currently available. In addition, a majority of farmers would consider using a test that could detect pregnancy within 2 weeks post-insemination, if such test existed, because they believed it would help improve their herds' reproductive performance. Overall, the opinions of farmers and veterinarians indicate that there is significant scope for improving dairy herd fertility monitoring practices in the UK, through development of improved assays that can diagnose pregnancy and infertility earlier, are less disruptive to farm operations and are more cost effective than available tools. Measuring core body temperature is used as part of the diagnostic process in assessing the health of animals. Typically in calves, this is carried out using a rectal thermometer which can be time consuming, stressful to the calf and is invasive by nature. A non-invasive technique that is gaining recognition is thermal imaging. This study investigated the use of thermal imaging as a technique to assess core body temperature in pre-weaned artificially reared calves. A total of 125 male and female calves had rectal temperatures measured daily from day 7 until day 40 of life, and at the same time had a thermal image taken of the area around the medial canthus of the eye. A weak correlation (r = 0.28) was found between calf rectal temperature and thermal image temperature. A multivariable predictive model for core body temperature increased the correlation (r = 0.32) when including the environmental parameters of air temperature (p < .001) and wind speed (p < .001) as well as reconstituted milk replacer consumption (p < .01). The effectiveness of a predictive model including these parameters for the detection of calves with a core body temperature ≥ 39.5 °C was examined and found to have a sensitivity of 0% and a specificity of 100%. The results of this study demonstrate the need to take thermal environmental parameters into consideration when using thermal imaging to assess body temperature. However, the results suggest that accurate measures of core body temperature using thermal imaging cannot be achieved under commercial farm conditions. This study aimed to investigate the effect of maternal stocking density during late pregnancy (approximately 60 ± 4 days before calving) on offspring performance during the pre-weaning period. Forty-five dairy calves were born to cows that went through either industry minimum standards (H: n = 24, high stocking density) or more extensive space allowances (L: n = 21, low stocking density) during the dry period. Body weight and average daily gain during the pre-weaning period (day 1-49) were measured. Observations were made of: (i) activity levels (day 2-6); ii) the level of training required to use an automatic feeder, and behavioural reactions to the group environment (d7); (iii) feeding and social behaviour in the group pen (day 7-21); and (iv) responses to weaning (day 40-49) and disbudding (day 28+). Compared to L calves, H calves made more frequent social contacts with pen mates in the group pen ( p = 0.003) and decreased their lying time around weaning ( p = 0.045). Among the healthy calves, L calves displayed more severe behavioural reactions to the disbudding procedure ( p < 0.001), a significant increase in salivary cortisol concentrations ( p = 0.013), and more frequent pain-related behaviour ( p = 0.036). This study indicated associations between maternal stocking density during late pregnancy and some welfare-relevant offspring outcomes during the pre-weaning period; these effects were found to be modulated by offspring health status., Competing Interests: There were no conflicts of interest.

Production diseases are highly prevalent in modern dairy herds, resulting in lost productivity and reduced animal welfare. Two important production diseases are mastitis and metabolic disorders. The availability of robust diagnostic tools that can detect animals at early stages of disease is crucial to prevent the high costs associated with lost productivity and the treatment of clinically and/or chronically diseased animals. Despite a variety of diagnostic methods being available to farmers and veterinarians, the incidence of these diseases in UK dairy herds has not changed over the last decade, underscoring the need for improved approaches for early disease detection. To this end, we administered a questionnaire to farmers and veterinarians to understand current diagnostic practices in the UK dairy cow sector, and to gather opinions on the suitability of currently available diagnostic tests in order to identify specific areas where improvement in diagnostic technologies and/or practices are needed. Data from a total of 34 farmers and 42 veterinarians were analyzed. Results indicated that most farmers surveyed used a combination of methods to diagnose mastitis and metabolic disorders, the most popular of which were visual inspection and milk recording somatic cell count data for mastitis, and body condition score and milk ketone testing for metabolic disorders. These preferences were not always in line with veterinarian recommendations of different diagnostic tools. Moreover, veterinarians indicated they were not satisfied with currently available diagnostic tools or how these were implemented by farmers. Both farmers and veterinarians recognized there was substantial room for improvement of current diagnostic tools, particularly in regard to the need to detect disease early. A majority of respondents preferred new diagnostic tests to be suitable for use with milk rather than blood or urine samples, and to yield results within 24 h. Finally, both groups surveyed identified economic cost as the most important barrier for the future uptake of new diagnostic technologies. The aim of this study was to objectively assess, using an automated behavioral monitoring system, any behavioral differences between primiparous and multiparous cows before calving, and to quantify any behavioral differences between assisted (dystocic) and unassisted (eutocic) calvings. Data were collected from 32 multiparous and 12 primiparous Holstein dairy cattle to describe normal calving behavior and parity differences. To quantify behavior related to calving difficulty, the data from 14 animals that had dystocia at calving were matched to cows that had an eutocic calving based on parity, locomotion score, calf breed, calf sex, month, and year of calving. An IceQube (IceRobotics Ltd., South Queensferry, United Kingdom) was fitted to the right hind leg of cows 4 wk before their expected calving date. Data for lying time, standing time, number of steps, motion index (total motion), and the total number of standing and lying bouts (postural transitions) were automatically collected and summed into 15-min blocks. Behavioral variables were summarized into 2-h periods and 24-h periods before analyses. Mixed-effect models were used to analyze cow behavior in the last 4 d before calving (d -4 to -1), and on the day of calving. In the 4 d before calving, compared with multiparous cows, primiparous cows lay down an average 2.8 h/d less, had 9.1 more postural transitions/d (37.7 ± 1.2 vs. 27.6 ± 0.7), walked 172 more steps/d, and had a higher motion index (2,673.2 vs. 1,981.5 units/d). There was an effect of 2-h period on all behavioral variables on the day of calving. No indicator of calving difficulty was found on the day of calving, nor the days leading up to calving. This study assessed risk factors associated with excessive negative energy balance (eNEB) in UK dairy cows between April 2006 and March 2015. Blood samples were analysed for β-hydroxybutyrate (BHB), non-esterified fatty acids (NEFA) and glucose. Following removal of all potential duplicate cows, a final dataset of 69,161 unique individual cows was obtained including biochemical results, individual cow and feed data. Generalised linear mixed-effect models and multivariable classification tree-based models showed that individual cow risk factors for eNEB included: (1) days relative to predicted calving date (dry cows); (2) days in milk (lactating cows); (3) body condition score (BCS; lactating cows ≥ BCS 4; OR 2.1); (4) milk yield (around 40 L per day); (5) parity (first lactation heifers; odds ratio [OR] 0.46 compared to older cows during lactation); and (6) chronic inflammatory conditions as assessed by globulin concentrations ≥ 50 g/L (OR 0.79 for cows with evidence of chronic inflammation). There was a higher prevalence during April to October (OR 1.19), and the lowest prevalence was in November. Feeding grass silage and wholecrop (silage made from cereal crops) to dry cows was associated with a reduced prevalence of eNEB, whereas access to grazed grass was associated with a higher prevalence in both the dry period (OR 1.32) and lactation (OR 1.33). Knowledge of the risk factors associated with eNEB in commercial dairy herds assists in both the implementation of herd monitoring programs and reduction of eNEB in dairy herds, with associated reductions in the risk of periparturient diseases and improved dairy cow performance., (Crown Copyright © 2019. The objectives of this study were to determine the individual animal and herd level prevalence of excessive negative energy balance (eNEB) in commercial UK dairy herds. Between April 2006 and March 2015, blood samples from 84,369 individual cows from 1748 different UK farms were received by a commercial laboratory service specializing in dairy cow nutritional monitoring. Following removal of all potential duplicate cows, the final dataset comprised 69,161 unique individual cows. The prevalence of eNEB was determined using plasma thresholds of β-hydroxybutyrate (BHB), non-esterified fatty acids (NEFA) and glucose. Overall prevalence of subclinical ketosis (SCK) in the first 20 days of lactation was 28.5%, 17.3% and 11.7% using BHB thresholds of 1.0, 1.2 and 1.4 mmol/L respectively. Prevalence of NEFA values ≥0.5 mmol/L in the last 10 days prior to calving was 26.0%, and 40.3% of cows had NEFA values ≥0.7 mmol/L in the first 20 days in milk (DIM). Combining BHB, NEFA and glucose showed that 52.0% of cows had one or more of the three biochemical measures of energy balance outside the respective threshold value in the last 10 days pre-calving, and 75.2% of cows showed a similar biochemical pattern in the first 20 DIM. The median herd prevalence of elevated BHB and/or NEFA was 37.5% in late pregnancy and 59.8% in the first 20 DIM, with substantial herd variation. In cattle, the serum protein haptoglobin (Hp) is a major acute phase protein (APP) that rises in concentration over a thousand fold following stimulation by pro-inflammatory cytokines. As such, this APP is a valuable biomarker for infection, inflammation and trauma in cattle. The assay for bovine Hp is becoming more commonplace in clinical pathology and in experimental studies when a biomarker of innate immunity is required. The most widely used assay for Hp utilises its binding to haemoglobin (Hp-Hb binding assay), which at low pH enables the preservation of the native peroxidase activity in the haemoglobin. This assay is used for all species, including species such as dog, cat and pig where the level of Hp is higher in healthy animals of these species than in healthy cattle, and therefore a bovine-specific immunoassay that can be automated would be desirable. Thus, a novel-automated species-specific immunoturbidimetric (IT) assay has been developed. Validation studies showed intra- and inter-assay CVs of below 5% and 9% respectively and a recovery of 99% from samples spiked with bovine Hp and a limit of quantification of 0.033 g/L. The assay is not affected by icterus or lipaemia but had moderate interference from haemoglobin and showed a significant correlation with the Hp-Hb binding assay. This novel IT assay for bovine Hp will allow automated analysis of this important bovine APP to identify changes in the Hp concentration not detectable by current Hp-Hb binding assays. Dry period management of the dairy cow focuses on maximising milk production during the subsequent lactation but may include procedures that negatively affect dry cow health and welfare. A survey of dairy farmers in the UK was conducted to investigate dry cow management procedures. The questionnaire was completed by 148 farmers. Most farms (84 per cent) kept dry cows in dynamic social groups. The median length of the dry period was 56 days, and 83 per cent of farms stopped milking abruptly, regardless of milk production level at dry-off. Twenty-seven per cent of cows from respondent farms produced more than 20 kg of milk per day at dry-off. The majority of farms (78 per cent) used antibiotic dry cow intramammary tubes at dry-off, in combination with internal or external teat sealants. Neck samples from 54 badgers and 32 tongue samples of the same badgers (Meles meles), collected in the Lothians and Borders regions of Scotland, were tested using polymerase chain reactions (PCRs) directed against the 18S ribosomal DNA and the internal transcribed spacer (ITS1) region of protozoan parasites of the family Sarcocystidae. Positive results were obtained from 36/54 (67%) neck and 24/32 (75%) tongue samples using an 18S rDNA PCR. A 468 base pair consensus sequence that was generated from the 18S rDNA PCR amplicons (KX229728) showed 100% identity to Sarcocystis lutrae. The ITS1 PCR results revealed that 12/20 (60%) neck and 10/20 (50%) tongue samples were positive for Sarcocystidae DNA. A 1074 bp consensus sequence was generated from the ITS1 PCR amplicons (KX431307) and showed 100% identity to S. lutrae. Multiple sequence alignments and phylogenetic analysis support the finding that the rDNA found in badgers is identical to that of S. lutrae. This parasite has not been previously reported in badgers or in the UK. Sarcocystis lutrae has previously only been detected in tongue, skeletal muscle and diaphragm samples of the Eurasian otter (Lutra lutra) in Norway and potentially in the Arctic fox (Vulpes lagopus).

The aims of the present study were to evaluate the effects of live yeast supplementation (Vistacell MUCL 39855, AB Vista, Marlborough, UK) on performance, rumination time, and rumen pH on dairy cows in commercial farm environments. Three trials were carried out, the trials lasted 12 (trial 1), 15 (trial 2), and 19 wk (trial 3). In each trial, 14 multiparous Holstein dairy cows were allocated to 2 groups that received (trial 1) a standard diet plus yeast, (trial 2) an acidogenic diet plus yeast, and (trial 3) grazing pasture plus yeast. Milk production, milk chemical characteristics, body weight and body condition score, rumination time, and rumen pH were monitored for each group throughout the 3 trials. No statistically significant differences were observed in any of the 3 trials for any of the recorded variables. In contrast, an effect of time (period or days in milk) on rumen pH was observed in all 3 of the trials, as time spent under the acidotic thresholds increased across the experimental periods; however the differences were not associated with live yeast supplementation. No effect of live yeast supplementation was observed in any of the 3 trials reported. Further research should include studies on animals at different stages of lactation (with emphasis on transition period and early lactation), consuming more challenging diets (higher level of inclusion of concentrates or starch), or under different environments such as grazing of succulent forages. Such studies might be required to elucidate any possible effect of live yeast supplementation of dairy cows when the rumen environment is under challenge., (Copyright © 2017 American Dairy Science Association. Automated systems for monitoring the behavior of cows have become increasingly important for management routines and for monitoring health and welfare. In the past few decades, various devices that record rumination have been developed. The aim of the present study was to compare rumination activity measured with a commercially available rumination collar (RC) against that obtained by direct visual observations and analysis of video recordings in commercial dairy cows. Rumination time from video recordings was recorded by a trained observer. To assess observer reliability, data were recorded twice, and the duration of recorded behaviors was very similar and highly correlated between these 2 measurements (mean=39±4 and 38±4 min/2 h). Measurements of rumination time obtained with RC when compared with analysis of video recordings and direct observations were variable: RC output was significantly positively related to observed rumination activity when dealing with animals housed indoors (trial 1 video recordings: slope=1.02, 95% CI=0.92-1.12), and the limits of agreement method (LoA) showed differences (in min per 2-h block) to be within -26.92 lower and 24.27 upper limits. Trial 1 direct observations: slope=1.08, 95% CI=0.62-1.55, and the LoA showed differences to be within -28.54 lower and 21.98 upper limits. Trial 2: slope=0.93, 95% CI=0.64-1.23, and the LoA showed differences to be within -32.56 lower and 19.84 upper limits. However, the results were poor when cows were outside grazing grass (trial 3: slope=0.57, 95% CI=0.13-1.02, and the LoA showed differences to be within wider limits -51.16 lower and 53.02 upper). Our results suggest that RC can determine rumination activity and are an alternative to visual observations when animals are housed indoors. However, they are not an alternative to direct observations with grazing animals on pasture and its use is not advisable until further research and validation are carried out., (Copyright © 2015 American Dairy Science Association. The objectives of this study were to determine (1) if stillborn calves born following dystocia present with specific injuries/pathological changes compared to stillborns delivered without difficulty, and (2) whether such stillborns differ in conformation from dystocic calves that survive. Post-mortem examinations were carried out on 20 stillborns that were either unassisted (N) or were 'farm-staff'-assisted/normally presented (FN) at birth. Evidence of greater trauma and bruising was observed in the FN calves and parameters such as body length, birth-weight and thyroid:body weight were similar. In a second part of the study birth-weight, body length and height, girth length, body mass (BMI), and ponderal (PI) indices were assessed in 490 calves. Regardless of the severity of dystocia, stillborns had greater body lengths and lower BMIs and PIs than calves born alive (P<0.05), suggesting prenatal factors contribute to their post-natal survival. FN calves were heavier than N calves (P<0.05), and both FN and farm-staff-assisted/malpresented calves had lower PIs than N calves (P<0.05). The study found that criteria such as grossly visible carcass haemorrhage, bruising, and brain congestion were not reliable in terms of identifying calves that had experienced dystocia. Half of the stillborns had breathed indicating they were alive and possibly had experienced pain/distress at time of delivery. The objective was to identify predisposing factors for increased risk of involuntary culling in adult Holstein-Friesian dairy cows. Data were sourced from Scotland's Rural College (SRUC) Dairy Research Centre. Between September 2003 and August 2010 175 cows were culled, a herd culling rate of 33.7%. The major reasons for involuntary culling were fertility (27.4%) and udder problems (26.9%). In the analysis, the culled cows were matched with their cohorts that survived to a later lactation. To identify predisposing factors, a binary logistic model was applied. Cows with higher than average body condition score (BCS) at last service were five times [Odds Ratio (OR) = 4.8] more likely to be culled due to infertility. Cows with low protein yield on day 60 ± 5 in lactation were ten times less likely (OR = 0.1) to be culled. In first lactation heifers, only BCS at last service increased the risk of involuntary culling due to infertility (OR = 13.0). A high milk yield acceleration was a significant (P = 0.04) factor in increasing the risk, five times (OR = 5.2) more, of culling cows due to udder problems. In conclusion, a high BCS at last service, high milk protein yield at around day 60 in lactation and acceleration of milk yield after calving exposed cows to a risk of being culled involuntarily. In practice, monitoring of traits that indicate metabolic imbalance could assist identifying cows at high risk of being culled and contribute to reducing the associated risk through a more effective timely decision.

High winter mortality (28 per cent) in female Jersey calves (80 IU/l in healthy females aged 3-24 weeks, and correlated with serum aspartate transaminase (AST). Copper supplementation of total mixed rations during lactation was excessive (40-60 mg added Cu/kg DM) and reduced to 16-28 mg Cu/kg, but supplementation of milk replacer and creep feed (10 and 35 mg added Cu/kg DM, respectively) continued. The syndrome recurred two years later, and liver Cu remained high in casualties (13.6 ± 2.6) and culled cows (6.38 ± 2.38 mmol/kg DM) prompting withdrawal of all Cu supplements. Mortality remained low (6-9 per cent) thereafter. Three years after removal of all Cu supplements, six culled newborn were examined postmortem; five had normal liver Cu (4.5 ± 1.73), but a sixth had 11.65 mmol/kg DM. In live, healthy calves (1-6 months old) sampled at the same time, GLDH and AST increased with age to levels found five years earlier, indicating possible subclinical hepatopathy. Causative links between Cu supplementation, high calf mortality and hepatopathy are plausible, and reductions in Cu supplementation may prove beneficial in other dairy herds.

Up to one-third of dairy calves are born after dystocia and this is a major cause of calf mortality. This study investigated the neonatal physiology, survival, health and subsequent growth of dairy calves following dystocia and is the first longitudinal study to analyse multiple effects and to look beyond the perinatal period. A total of 455 live born Holstein calves (N: No assistance, n=360; FN: Farmer assistance but normally presented calf, n=82; FM: Farmer assistance of malpresented calf, n=13) were followed from birth to first service (heifers) or until leaving the farm (bulls). Compared to N calves, FN and FM animals had higher salivary cortisol concentrations at day 1 (P<0.001) and FN calves had lower passive immune transfer (P=0.03). Dystocia had no biologically significant impact on rectal temperature throughout the first 4 days (P>0.05). During the first 60 days, FM calves had a higher proportion of days with non-routine health treatments (P<0.05) and, by the time of weaning, mortality in FN and FM heifers was higher than in N calves (2.8×; P<0.01). However, in surviving calves, growth to first service was not affected by dystocia category (P>0.05). Calves which survive dystocia experience lower passive immunity transfer, higher mortality and higher indicators of physiological stress. Such calves have poorer welfare in the neonatal period and possibly beyond. The experience of a difficult birth (dystocia) is traumatic and has adverse effects on the newborn in various species. Despite affecting up to 1 in 3 births in dairy cattle, studies on calves have been mostly limited to the first day of life. The objective of this study was to investigate the effects of dystocia on the survival to calving, growth to weaning, and subsequent fertility as nulliparous animals. Historical data from live-born Holstein heifer calves born from cows with various birth difficulty scores (no assistance; moderate; high difficulty) were obtained from 2 herds (Edinburgh herd: n=1,237; Crichton Royal Farm herd: n=721). Each herd was analyzed separately for birth weights, weaning weights, growth rate to weaning, number of services to conception, and age at first calving using REML and generalized linear mixed model analyses. Survival analysis (Cox proportional hazards model) was used in the Edinburgh herd to analyze the subsequent survival of live-born heifers, whereas descriptive data are presented for the Crichton Royal Farm herd. A higher mortality risk to weaning and to first service was observed in the live-born heifers that experienced moderate difficulty at birth compared with heifers born naturally. Surviving dystocial heifers had similar growth-to-weaning and fertility performance as heifers born naturally in both herds. It could be that the performance of dystocial heifers that survived to weaning was not affected or that it was compensated for by farm management. This study highlights long-term effects of the early experience of a difficult birth and thereby stresses the importance of preventing dystocia not just from the point of view of the adult cow, but also from the perspective of the calf. This paper describes the nematode control strategy adopted by a Scottish hill sheep farmer. It provides an example of the limited use of anthelmintic drugs, targeted towards control of the periparturient rise in faecal nematode egg output in ewes and exploitation of grazing management, resulting in limited exposure of naive sheep to infective larvae on pasture. Resistance to benzimidazole, imidazothiazole and macrocyclic lactone anthelmintics, and to a combination of a macrocyclic lactone and imidazothiazole anthelmintic drugs was diagnosed. The targeted use of a persistent anthelmintic drug to control a periparturient rise in faecal nematode egg output in ewes may be unnecessary and selects strongly for resistance when the reservoir of anthelmintic-susceptible nematodes in refugia is small at the time of treatment. However, the use of a persistent anthelmintic drug in a selective proportion of ewes can be important and probably does not select strongly for resistance when the reservoir of anthelmintic-susceptible nematodes in refugia is large at the time of treatment. The former circumstances might arise on many Scottish hill sheep farms, whereas the latter may occur on upland and lowground farms, depending on previous grazing management, anthelmintic use and winter weather conditions. These factors must be taken into account when preparing sustainable health plans for nematode parasite control in individual sheep flocks.

Litter size determined ultrasonographically at 45-90 days after mating in eight groups of commercial sheep (n=2609) treated with slow release melatonin implants immediately prior to the natural breeding season showed a 19% treatment effect averaged over the eight groups, from 148% to 167% (P<0.01) compared with controls (n=2499). Control groups had a litter size ranging from 124% to 194% thereby representing most commercial production systems in the UK sheep industry. This is the first large scale study of slow release melatonin implants in commercial UK flocks to demonstrate a significant positive effect on litter size when used before the natural breeding season. The data indicate that increased productivity and profitability could be achieved while maintaining desirable purebred characteristics.

Between April 1999 and March 2004, metabolic profile analyses were performed on individual blood samples from 35,506 dairy cattle in the UK. Assessment of the cows' energy status by the analysis of plasma samples for beta-hydroxybutyrate, glucose and non-esterified fatty acids showed that 70.4 per cent of the cows in early lactation (10 to 20 days calved), 57.1 per cent of the cows in mid-lactation (51 to 120 days calved) and 57.7 per cent of the dry cows within 10 days of their predicted calving date had one or more energy metabolites outside the optimum range; in addition, 16 per cent of the cows in early lactation, 5.6 per cent of those in mid-lactation and 20.5 per cent of the dry cows within 10 days of their predicted calving date had a low plasma urea nitrogen concentration, indicating poor intakes of effective rumen-degradable protein. Abnormalities in the concentrations of magnesium, inorganic phosphate, copper, selenium and iodine were relatively uncommon. The transitional period, particularly in late pregnancy, was commonly identified as a constraint on productivity. Nutritional problems were most commonly associated with poor feed intakes and poor feed management, rather than with the formulation of the rations.

Evolved functions of integrin-alpha(v)beta(6) include roles in epithelial cell-extracellular matrix protein interactions and in the binding and activation of latent TGF-beta(1). Integrin-alpha(v)beta(6) is also exploited as a receptor by foot-and-mouth disease virus (FMDV) and may play a significant role in its transmission and pathogenesis. The ovine beta(6) integrin subunit was cloned and sequenced (EMBL accession no. AJ439062). Screening of normal ovine tissues by RT-PCR and immunocytochemistry confirmed that integrin-alphavbeta6 is restricted to sheep epithelial cells. Integrin-alphavbeta6 expression was detected in epithelia of the airways, oral cavity, gastrointestinal tract, kidney, sweat glands, hair follicle sheaths, and the epidermis of pedal coronary band (PB) but not of normal skin. Consistent with FMDV tropism, integrin-alphavbeta6 was detected within the basal layers of the stratified squamous epithelium of the oral mucosa and PB. In addition, integrin-alphavbeta6 appears to be constitutively expressed in the normal airways of both cattle and sheep. The latter finding suggests that ruminant airway epithelium presents a highly accessible target for initiation of infection with FMDV by inhalation.

The high-affinity receptor for IgE (FcepsilonRI) plays a central role in IgE-mediated allergic reactions. Cross-linking of FcepsilonRI by IgE-antigen complexes results in the activation of mast cells and basophils and is thought to contribute to the immunopathology of Heaves, a chronic obstructive pulmonary disease of horses. Recombinant protein corresponding to the extra-cellular portion of the FcepsilonRI alpha subunit, cloned and sequenced previously, was expressed using both mammalian cells and insect cells. The yield of expressed protein was considerably greater using insect cells and the baculovirus expression system. The recombinant proteins differed in size between the two systems, presumably due to differences in the extent of glycosylation. However, recombinant protein from both cell systems bound equine IgE present in bronchoalveolar lavage fluid from horses with Heaves. These results suggest that the recombinant extra-cellular part of FcepsilonRI should be a useful tool with which to study equine IgE responses.

Xanthones with amino substituents were synthesized to diminish the photoreactivity of the xanthone chromophore with DNA, with the objective of using these molecules to study their binding dynamics with DNA. The aminoxanthones showed a strong solvatochromic effect on their singlet and triplet excited-state photophysics, where polar solvents led to a decrease of the energies for the excited states. Quenching of the triplet excited states by nitrite anions was used to determine the binding dynamics, and a residence time in the microsecond time domain was estimated for the bound 2-aminoxanthone with DNA. The quenching experiments performed showed that this methodology will not be applicable to study the binding dynamics of a wide variety of guests with DNA.

Data derived over four years from 434 dairy herds in 1998/99 to 244 in 2001/02 revealed average disposal rates of 22.6 per cent per year, half of which were for poor fertility, mastitis and lameness. The quartile of herds with the lowest disposal rates sold an average of 11.5 per cent annually and the quartile with the highest rates sold 35.5 per cent. Average annual disease rates over the four years were as follows: for assisted calving 7.8 per cent, for digestive disease 1.2 per cent, for ketosis 0.5 per cent, for hypomagnesaemia 0.5 per cent, for hypocalcaemia 5.0 per cent and for injuries 0.8 per cent. The incidence of mastitis increased from 36.0 to 43.3 per cent of cows per year. The incidence of lameness decreased from 23.3 per cent in 1998/99 to 20.7 per cent in 2000/01 but increased to 21.9 per cent in 2001/02. Data received from the same 219 farms during the first three years showed no effective differences from the full set of data for each of the three years. The lowest annual incidences of mastitis and lameness on individual farms were below 7 per cent and 2.5 per cent, respectively. In general, housing cows in cubicles was associated with a greater risk of lameness, and housing them in straw yards with a greater risk of mastitis. However, some of the lowest rates of lameness were recorded in cubicle-housed cows and some of the lowest rates of mastitis were recorded in cows housed in yards. Larger herds were not associated, in general, with higher rates of mastitis.

The administration of intraruminal identification boluses to a group of 76 lambs resulted in 24 (32 per cent) showing signs of pharyngeal damage. In 16 of them the bolus was found to be in the retropharyngeal region by means of a hand-held microchip scanner, radiography and endoscopy. Purulent tracts were identified in the dorsal pharynx. In 13 of the 16 lambs the surgical removal of the bolus under general anaesthesia was followed by a normal clinical recovery, although the lambs did not grow as well as similar unaffected animals; three of the 24 lambs died. The administration of a larger bolus to 10 yearling rams did not result in any clinical signs of pharyngeal damage. The most important factors influencing the occurrence of this type of injury among this group of lambs were the size and age of the lambs, the positioning of the lambs during bolus administration, the relative size of the dosing gun and bolus, and the large number of animals in the group.

This work describes analyses of the function of the murid herpesvirus 4 strain 68 (MHV-68) M2 gene. A frameshift mutation was made in the M2 open reading frame that caused premature termination of translation of M2 after amino acid residue 90. The M2 mutant showed no defect in productive replication in vitro or in lungs after infection of mice. Likewise, the characteristic transient increase in spleen cell number, Vbeta4 T-cell-receptor-positive CD8(+) T-cell mononucleosis, and establishment of latency were unaffected. However, the M2 mutant virus was defective in its ability to cause the transient sharp rise in latently infected cells normally seen in the spleen after infection of mice. We also demonstrate that expression of M2 is restricted to B cells in the spleen and that M2 encodes a 30-kDa protein localizing predominantly in the cytoplasm and plasma membrane of B cells.

Infection of mice by murine gammaherpesvirus 68 (MHV-68) is an excellent small-animal model of gammaherpesvirus pathogenesis in a natural host. We have carried out comparative studies of another herpesvirus, murine herpesvirus 76 (MHV-76), which was isolated at the same time as MHV-68 but from a different murid host, the yellow-necked mouse (Apodemus flavicollis). Molecular analyses revealed that the MHV-76 genome is essentially identical to that of MHV-68, except for deletion of 9,538 bp at the left end of the unique region. MHV-76 is therefore a deletion mutant that lacks four genes unique to MHV-68 (M1, M2, M3, and M4) as well as the eight viral tRNA-like genes. Replication of MHV-76 in cell culture was identical to that of MHV-68. However, following infection of mice, MHV-76 was cleared more rapidly from the lungs. In line with this, there was an increased inflammatory response in lungs with MHV-76. Splenomegaly was also significantly reduced following MHV-76 infection, and much less latent MHV-76 was detected in the spleen. Nevertheless, MHV-76 maintained long-term latency in the lungs and spleen. We utilized a cosmid containing the left end of the MHV-68 genome to reinsert the deleted sequence into MHV-76 by recombination in infected cells, and we isolated a rescuant virus designated MHV-76(cA8+)4 which was ostensibly genetically identical to MHV-68. The growth properties of the rescuant in infected mice were identical to those of MHV-68. These results demonstrate that genetic elements at the left end of the unique region of the MHV-68 genome play vital roles in host evasion and are critical to the development of splenic pathology.

The ultrasonographic anatomy of the elbow, carpal, fetlock and stifle joints of six clinically normal sheep was determined using a 7.5 MHz linear transducer with stand-off pad. Examination of the joints was performed both prior to and after experimental distension of the joint capsule in four cadavers with up to 10 mL isotonic saline. This distension enabled the joint capsule to be imaged ultrasonographically as a 2 mm thick echogenic line. Isolation of the joint at necropsy enabled definitive identification of the structures seen ultrasonographically. Arthrosonography of five sheep with chronic arthritis/synovitis affecting one or more joints was performed, and the gross pathological changes examined at necropsy. The gross thickening of the joint capsule was visible ultrasonographically as a hyperechoic band up to 20 mm thick. Ultrasonography was a valuable diagnostic aid in the evaluation of chronic ovine joint disease, and has potential value in the prognosis for chronic ovine arthritis/synovitis., (Copyright 1999 Harcourt Publishers Ltd.)
