Marine bivalves provide an important seafood resource, but conflicts between bivalve fisheries and conservation goals may arise.A scientific debate about cockle dredging in the Dutch Wadden Sea resulted in an apparent consensus among scientists of a severe and long‐lasting impact of the mechanical fisheries, and the fisheries were banned.Specifically, two highly cited papers concluded that the fisheries caused a strong reduction in bivalve recruitment for a period of 8 years and that the fisheries led to the loss of red knots from the Dutch Wadden Sea and a decline of the European wintering population.Here, we show that these papers had severe shortcomings in terms of data selection and data analysis and we challenge their conclusions. This article discusses the assessment of biological effects of contaminants in the Gulf of Finland, specifically focusing on sediment biotests with amphipods (Monoporeia affinis) and biomarker responses in clams (Macoma balthica). The Gulf of Finland is experiencing adverse effects due to human activities, leading to a decline in the marine environment's quality. The study collected sediments from contaminated offshore and coastal areas and conducted bioassays to determine the effect of contaminants on amphipods and clams. The results showed that the amphipods exposed to sediments from Narva Bay did not exhibit significant changes in biomarker activities, except for oxidative stress. In clams, enzymatic activities reflected in situ exposure to harmful compounds and conditions. The study concludes that the Sillamäe harbor had the most stressful conditions within the studied area. Taking advantage of the unique system of doubly uniparental inheritance (DUI) of mitochondria, we developed a reliable molecular method to sex individuals of the marine bivalve Macoma balthica rubra. In species with DUI (~100 known bivalves), both sexes transmit their mitochondria: males bear both a male‐ and female‐type mitogenome, while females bear only the female type. Male and female mitotypes are sufficiently divergent to reliably PCR‐amplify them specifically. Loop‐mediated isothermal amplification (LAMP) is a precise, economical and portable alternative to PCR for molecular sexing and we demonstrate its application in this context. We used 154 individuals sampled along the Atlantic coast of France and sexed microscopically by gonad examination to test for the congruence among gamete type, PCR sexing and LAMP sexing. We show an exact match among the sexing results from these three methods using the male and female mt‐cox1 genes. DUI can be disrupted in inter‐specific hybrids, causing unexpected distribution of mitogenomes, such as homoplasmic males or heteroplasmic females. To our knowledge, DUI disruption at the intra‐specific scale has never been tested. We applied our sexing protocol to control for unexpected heteroplasmy caused by hybridization between divergent genetic lineages and found no evidence of disruption in the mode of mitochondrial inheritance in M. balthica rubra. We propose LAMP as a useful tool to accelerate eco‐evolutionary studies of DUI. It offers the opportunity to investigate the potential role of, previously unaccounted‐for, sex‐specific patterns such as sexual selection or sex‐specific dispersal bias in the evolution of free‐spawning benthic species. Background/Aims: Occurring in marine invertebrates infectious haemic neoplasia (bivalves transmissible neoplasia, BTN) arises from genome instabilities leading to multilevel malfunctions and unregulated cell division of presumably haemocyte precursors. As its biochemical characterisation remains unknown, we here present the first data describing selected aspects of the physiology and biochemistry of the disease a in model clam Macoma balthica. We chose free amino acids (FAA) composition, mitochondrial respiration and enzymatic activity, oxidative stress enzymes activities and corticosteroids profile as markers of this contagious cancer. Methods: Selected markers were measured in neoplastic and healthy clams and two tissue types, haemolymph and solid tissue. FAA composition was assessed in the haemolymph samples using high performance liquid chromatography-mass spectrometry (LC/MS). Mitochondrial respiration analysis was performed on haemocytes using oxygen electrodes integrated system Seahorse XFp. Mitochondrial enzymes activities were measured using spectrophotometry (cytochrome oxidase, COX) and commercial kit (succinate dehydrogenase, SDH). Total Antioxidant Capacity (TAC), Acetylocholinesterase (AChE), Protein Carbonyl Content (CBO) and Malondialdehyde (MDA) levels were measured in the solid tissue using analytical kits, and glutathione (GSH) was measured spectrophotometrically. Corticosteroids profile, measured in the solid tissue, was obtained with Liquid Chromatography-Electrospray Ionization-Mass Spectrometry (HPLC-ESI-MS/MS) technique. Results: In both clam groups nine FAAs were detected with Asp, Glu, Pro, Ser constituting over 90% of total FAA content. Significantly higher Gln level was detected in BTN positive clams. In neoplastic clams, an impairment of mitochondrial metabolism was observed as a decrease in mitochondrial oxygen consumption and lower cytochrome c oxidase activity. In the neoplastic clams significantly higher concentration of low molecular weight antioxidants was found. Finally, we report high level of corticosterone and lower levels of dehydrocorticosterone, cortisol and cortisone in healthy clams and elevated cortisol level in BTN individuals. Conclusion: Neoplastic clams are characterized by altered mitochondrial metabolism, with a potential key role of glutamine (Gln) in cancer cells energy production. Despite low aerobic respiration, BTN cells have efficient antioxidative response to elevated concentration of ROS. Elevated cortisol level in BTN-positive clams may indicate an important role of this corticosteroid in cancer biochemistry. Thus, we here provide the first results of selected physiological and biochemical aspects of BTN, making an important step in studying cancer epidemiology in wildlife. [ABSTRACT FROM AUTHOR]

The study of seasonal dynamics of size-mass structure of Macoma balthica settlements has been investigated on the littoral zone of the southern part of the Kola Bay. Age structure has been calculated; data on the density and biomass of settlements have been obtained; the distribution of the mollusk on the littoral has been shown. On the littoral zone of the Kola Bay makoms are found everywhere, mostly inhabiting the lower and middle horizons of the littoral; this distribution depends on the content of food particles, which is an indirect indicator of the granulometric composition of soil and total organic content. The highest indicators of biomass and the number of settlements have been marked in the intertidal zone near the village of Abram-mys. This is due to the presence of silty-sandy soil – the most suitable habitat for shellfish, as the abundance of settlement figures increases as the silting of sand. The increase in the density of makom settlements in autumn occurs due to the active settling of juveniles. Shellfish from the right bank settlements surpass others on the morphometric parameters. Large shell sizes are explained by the presence of streams ensuring the inflow of food particles. Researches of the size-distribution of Macoma balthica mass parameters have shown that their values are increased from the upper to the lower horizon littoral as in the lower horizon the enhanced water exchange contributes to the self-cleaning processes and provides shellfish by food. The dominance of mollusks of younger age groups (0–3 years) is characteristic for all settlements. Clams with the maximum age of 8 years have been found in the New Bridge area. The presence in the samples of large number of mollusks of younger age groups indicates the normal development of Macoma balthica settlements.

Benthic macrofauna is an important component linking pelagic and benthic ecosystems, especially in productive coastal areas. Through their metabolism and behaviour, benthic animals affect biogeochemical fluxes between the sediment and water column. Mechanistic models that quantify these benthic-pelagic links are imperative to understand the functioning of coastal ecosystems. In this study, we develop a dynamic model of benthic macrofauna to quantify the relationship between organic matter input and benthic macrofaunal biomass in the coastal zone. The model simulates the carbon dynamics of three functional groups of benthic macrofauna and their sediment food sources and is forced by a hydrodynamic-biogeochemical model simulating pelagic physical and biological dynamics. The model reproduces measured time-series of macrofaunal biomass from two coastal sites with contrasting sedimentation in the Baltic Sea in 1993–2005 with comparatively high accuracy, including a major increase at one of the sites dominated by the bivalve Limecola (Macoma) balthica. This shift in community composition suggests altered pathways of organic matter degradation: 39% of simulated sedimentation was mineralised by macrofauna in 2005 compared to 10% in 1995. From the early 2000s onward macrofaunal biomass seems to be food-limited, as ca 80% of organic carbon sedimentation was processed by the deposit-feeding macrofauna at both sites. Abstract The frequency of seasonal and short-term hypoxia is increasing in coastal seas. How such repeated disturbances affect key species that have important roles for ecosystem processes and functions remains, however, unknown. By performing a field experiment we explored if the bivalve Macoma balthica can cope with short-term, recurring hypoxic stress, and investigated how hypoxia affects the condition of surviving bivalves. By combining data on different levels of biological organization, i.e., on physiology (biomarker response), behaviour and demography, we identified stress responses before the population declined. One pulse of hypoxic disturbance (3 days) resulted in behavioural alterations, as adult M. balthica extended their siphons, emerged towards the sediment surface and expressed decreased reburial rates. However, the demographic structure of the population remained unaltered. Several pulses of recurring hypoxic stress resulted in physiological response with changes in glutathione reductase and acetylcholinesterase enzyme activities. The recurring hypoxic disturbance was observed to affect juvenile bivalves before adults, while pro-longed hypoxia reduced the entire bivalve population. Our results clearly show that hypoxic stress changes the behaviour and physiology of M. balthica before demographic changes occur, which is likely to have severe implications for the contribution of this key species to ecosystem functioning. That a combination of measures at different levels of organization can detect disturbances at an early stage suggests that such an approach would be useful for assessing the effects of disturbances on marine ecosystems that are increasingly affected by anthropogenic change. Abstract The Baltic clam Limecola balthica L. (Tellinidae) is broadly used in ecophysiological, toxicological, evolutionary and environmental monitoring studies. However, it is poorly studied in respect of genome and gene functions. We obtained a transcriptome of Limecola b. balthica from Kamchatka (Western Pacific) generated with the use of Illumina high-throughput sequencing. We annotated 11,374 proteins, including 53 from the oxidative phosphorylation pathway and a number of pollution-stress biomarkers, recovered 254,540 single nucleotide variants within two annotated transcriptomes including 25,330 scorable in the previously published European data. Our results confirmed the available allozyme data indicating that nuclear genomes of the clams from the Baltic Sea were intermediate in their genetic composition between the Pacific (L. b. balthica) and the Atlantic (L. b. rubra) subspecies. At the same time, the mitochondrial genomes of Limecola from Kamchatka were nearly identical to the single published genome from the Baltic. The genomic diversity in Limecola was found to be high and comparable with that of other marine mollusks (0.0138 and 0.0142 heterozygous positions in the two studied transcriptomes). The data obtained in our study are a valuable resource for further development of genomic markers for evolutionary genetic and ecophysiological studies of L. balthica complex. [ABSTRACT FROM AUTHOR]

Long-term population dynamics of marine invertebrates can be shaped by environmental conditions as well as biotic factors, including predation, diseases, interspecific or intraspecific competition. Towards the northern edge of species ranges the role of biotic interactions gradually decreases while the impact of climate oscillations becomes more important. This study examined the long-term changes in abundance, individual growth rates and shell shape characteristics of Macoma balthica, one of the dominant species in White Sea soft-bottom intertidal communities. To test the role of predators in changes in clam abundance, we examined the number of moonsnails Amauropsis islandica. Macoma balthica exhibited spatially synchronous population dynamics at six sites in Kandalaksha Bay, where densities of clams varied between 140 and 8500 ind. m−2 during the 21-year period of observations. Statistical modelling using generalized additive models (GAM) shows that a combination of mild winter and warm summer led to an increase in M. balthica density the following year. Predation by A. islandica had no impact on changes in M. balthica density. Growth rates of M. balthica were higher during a cool decade, but clams that lived in a warmer period were characterized by more globose shells. Our results suggest that the climate oscillations can be regarded as the key factor causing the shift in abundance of M. balthica in the White Sea during the last two decades via recruitment and survival. We used transcriptomic sequence data to describe patterns of divergence and selection across different populations of a marine bivalve (Limecola balthica). Our analyses focused on a nuclear gene (atp5c1) that was previously detected in an FST scan as highly structured among populations separated by the Finistère Peninsula in France. This gene encodes the gamma subunit of the FO/F1 ATP synthase, a multi-protein complex that is paramount to cellular respiration and energy production. Analysis of non-synonymous to synonymous mutation ratios revealed that 65% of the gene is highly conserved (dN/dS ≤ 0.1, min = 0), while 6% of the gene is likely under positive selection (dN/dS ≥ 1, max = 2.03). All replacement mutations are clustered on a 46 residues portion of the protein, within an inter-peptide interaction zone. Comparative genomics suggests that these mutations are evolutionarily stable, and we hypothesize that they are involved in inter-population genetic incompatibilities with other subunits of the ATP synthase complex. The protein stability of the gamma subunit conferred by southern variants was inferred to be higher under warmer temperatures, suggesting that environmental conditions may contribute to the strength of genetic barriers in L. balthica.

Microplastics (MPs) are observed to be present on the seafloor ranging from coastal areas to deep seas. Because bioturbation alters the distribution of natural particles on inhabited soft bottoms, a mesocosm experiment with common benthic invertebrates was conducted to study their effect on the distribution of secondary MPs (different-sized pieces of fishing line < 1 mm). During the study period of three weeks, the benthic community increased MP concentration in the depth of 1.7–5.1 cm in the sediment. The experiment revealed a clear vertical gradient in MP distribution with their abundance being highest in the uppermost parts of the sediment and decreasing with depth. The Baltic clam Macoma balthica was the only study animal that ingested MPs. This study highlights the need to further examine the vertical distribution of MPs in natural sediments to reliably assess their abundance on the seafloor as well as their potential impacts on benthic communities. The combined effects of ocean warming and contaminants on marine ecosystems are poorly understood. In this study, we exposed model ecosystems comprising typical shallow coastal Baltic Sea communities to elevated temperature (+5 °C) and the flame retardant hexabromocyclododecane (HBCDD), both singly and in combination, for 13 days. Higher temperatures caused the release of PO4 from the sediment, which in turn stimulated the growth of the cyanobacteria Dolichospermum sp. This in turn led to an increase in the copepod Acartia bifilosa and other indirect effects in the plankton, interpreted as being caused by changes in predation, grazing, and competition. Elevated temperatures also stimulated benthic primary production and increased production of benthic mollusk larvae. Although increased temperature was the dominant driver of effects in these systems, HBCDD also appeared to have some effects, mainly in the zooplankton (both direct and indirect effects) and benthic meiofauna (an interactive effect with temperature). Although the study used model ecosystems, which are an approximation of field conditions, it highlights that interactive ecosystem effects between two stressors are possible and demonstrates the ecological and temporal complexity of such responses.
