Abstract

Treatment for Chagas disease, a parasitosis caused by Trypanosoma cruzi, has always been based on two drugs, nifurtimox and benznidazole, despite the toxic side effects described after prolonged prescription. In this work, we study a new prospective antitrypanosomal drug based on vanadium, here named VIVO(5Brsal)(aminophen). We found a good IC50 value, (3.76 ± 0.08) μM, on CL Brener epimastigotes. The analysis of cell death mechanism allowed us to rule out the implication of a mechanism based on early apoptosis or necrosis. Recovery assays revealed a trypanostatic effect, accompanied by cell shape and motility alterations. An uptake mostly associated with the insoluble fraction of the parasites was deduced through vanadium determinations. Concordantly, no drastic changes of the parasite transcriptome were detected after 6 h of treatment. Instead, proteomic analysis uncovered the modulation of proteins involved in different processes such as energy and redox metabolism, transport systems, detoxifying pathways, ribosomal protein synthesis, and proteasome protein degradation. Overall, the results here presented lead us to propose that VIVO(5Brsal)(aminophen) exerts a trypanostatic effect on T. cruzi affecting parasite insoluble proteins.

36. Holistic food safety evaluation of herbs: methods for the determination of organic and inorganic trace contaminants in Moringa stenopetalaas a case study

Abstract

Moringa stenopetalais considered a superfood due to the many bioactive compounds that it provides to the diet. However, like all edible plants, it is mandatory to guarantee food safety. Thus it is necessary to develop analytical methods that can rapidly and accurately determine hazardous pollutants, to evaluate compliance with food regulations. In this regard, two multi-component procedures were developed trying to cover some of the main organic and inorganic potential contaminants. A microwave-assisted digestion followed by electrothermal atomic absorption spectrometry was used for arsenic, cadmium, and lead determination, while a modification of the QuEChERS protocol followed by gas chromatography-tandem mass spectrometry was employed for the determination of 55 pesticides from different families. Both analytical methods were thoroughly validated according to international guidelines. The analyzed samples obtained from the Uruguayan market showed compliance with both, national and international, food regulations. The holistic approach employed in this research is not commonly presented in the literature, thus constituting a novel way to face food safety.

37. Evaluation of essential and potentially toxic elements in popcorn: study of cooking effect and in vitro bioaccessibility

Abstract

The influence of the cooking method on the elemental composition of popcorn, prepared according to the way it is consumed, was evaluated. Arsenic, calcium, cadmium, copper, iron, lead, magnesium, manganese, potassium, selenium, sodium and zinc levels were determined using atomic absorption spectrometry. Results showed that the losses after cooking varied considerably (2.9%–48.3%) depending on the element. A bioaccessibility study of cooked samples was also performed, using an in vitro approach. The most bioaccessible elements were sodium (77.4%–88.5%) and potassium (63.7%–81.8%), while considerably low values were observed for calcium (5.5%–13.7%) probably due to strong interactions with the matrix. The contribution to the Recommended Dietary Allowance was evaluated, finding an important coverage for selenium (36.7%–50.0%). Positive Pearson correlation coefficients were found for the pairs Cu/Fe, Cu/Mn, Fe/Mn Cu/K, Fe/K, K/Mn which can be explained by natural physiological mechanisms of the plant. To the best of our knowledge, this is the first study of bioaccessibility of essential elements in Uruguayan popcorn.

Abstract

This study investigated the in vitro bioaccessibility of aluminum, copper, iron, manganese, lead, selenium, and zinc in three important species of farmed insects: the yellow mealworm (Tenebrio molitor) , the house cricket (Acheta domesticus) and the migratory locust (Locusta migratoria). Results show that all three insect species constitute excellent sources of essential elements (Fe, Cu and Zn) for the human diet, contributing to the recommended dietary allowance, i.e. , 10%, 50%, and 92%, respectively. A higher accumulation of Se (≥1.4 mg Se/kg) was observed with increasing exposure concentration in A. domesticus , showing the possibility of using insects as a supplements for this element. The presence of Al and Fe nanoparticles was confirmed in all three species using single particle-inductively coupled plasma-mass spectrometry and transmission electron microscopy. The results also indicate that Fe bioaccessibility declines with increasing Fe-nanoparticle concentration. These findings contribute to increase the nutritional and toxicological insights of farmed insects. • Studied insects provide excellent sources of Fe, Cu, and Zn for the human diet. • Selenium-enriched diets significantly boost selenium levels in A. domesticus. • A higher presence of Fe-nanoparticles reduced Fe bioaccessibility. • Whether as nanoparticles or in ionic form, Al bioaccessibility remains below 10%.
