We report an experimental study aiming to clarify the role of boundary conditions (BC) in high Rayleigh number $10^8 < {\rm{Ra}} < 3 \times 10^{12}$ turbulent thermal convection of cryogenic helium gas. We switch between BC closer to constant heat flux (CF) and constant temperature (CT) applied to the highly conducting bottom plate of the aspect ratio one cylindrical cell 30 cm in size, leading to dramatic changes in the temperature probability density function and in power spectral density of the temperature fluctuations measured at the bottom plate, while the dynamic thermal behaviour of the top plate and bulk convective flow remain unaffected. Within our experimental accuracy, we find no appreciable changes in Reynolds number Re(Ra) scaling, in the dimensionless heat transfer efficiency expressed via Nusselt number Nu(Ra) scaling, nor in the rate of direction reversals of large scale circulation., Comment: 7 pages, 7 figures

8. First-order quantum phase transitions: test ground for emergent chaoticity, regularity and persisting symmetries

We present a comprehensive analysis of the emerging order and chaos and enduring symmetries, accompanying a generic (high-barrier) first-order quantum phase transition (QPT). The interacting boson model Hamiltonian employed, describes a QPT between spherical and deformed shapes, associated with its U(5) and SU(3) dynamical symmetry limits. A classical analysis of the intrinsic dynamics reveals a rich but simply-divided phase space structure with a H\'enon-Heiles type of chaotic dynamics ascribed to the spherical minimum and a robustly regular dynamics ascribed to the deformed minimum. The simple pattern of mixed but well-separated dynamics persists in the coexistence region and traces the crossing of the two minima in the Landau potential. A quantum analysis discloses a number of regular low-energy U(5)-like multiplets in the spherical region, and regular SU(3)-like rotational bands extending to high energies and angular momenta, in the deformed region. These two kinds of regular subsets of states retain their identity amidst a complicated environment of other states and both occur in the coexistence region. A symmetry analysis of their wave functions shows that they are associated with partial U(5) dynamical symmetry (PDS) and SU(3) quasi-dynamical symmetry (QDS), respectively. The pattern of mixed but well-separated dynamics and the PDS or QDS characterization of the remaining regularity, appear to be robust throughout the QPT. The dynamics inside the phase coexistence region exhibits a very simple pattern. A classical analysis reveals a robustly regular dynamics confined to the deformed region and well separated from a chaotic dynamics ascribed to the spherical region. A quantum analysis discloses regular bands of states in the deformed region, which persist to energies well above the phase-separating barrier, in the face of a complicated environment. We present a method to measure distributions of such degeneracies on the Riemann sheet of a selected level and apply it in classification of quantum phase transitions. Purpose: Within the Solve-RD project (https://solve-rd.eu/), the European Reference Network for Intellectual disability, TeleHealth, Autism and Congenital Anomalies aimed to investigate whether a reanalysis of exomes from unsolved cases based on ClinVar annotations could establish additional diagnoses. We present the results of the "ClinVar low-hanging fruit" reanalysis, reasons for the failure of previous analyses, and lessons learned. Methods: Data from the first 3576 exomes (1522 probands and 2054 relatives) collected from European Reference Network for Intellectual disability, TeleHealth, Autism and Congenital Anomalies was reanalyzed by the Solve-RD consortium by evaluating for the presence of single-nucleotide variant, and small insertions and deletions already reported as (likely) pathogenic in ClinVar. Variants were filtered according to frequency, genotype, and mode of inheritance and reinterpreted. Results: We identified causal variants in 59 cases (3.9%), 50 of them also raised by other approaches and 9 leading to new diagnoses, highlighting interpretation challenges: variants in genes not known to be involved in human disease at the time of the first analysis, misleading genotypes, or variants undetected by local pipelines (variants in off-target regions, low quality filters, low allelic balance, or high frequency). Conclusion: The "ClinVar low-hanging fruit" analysis represents an effective, fast, and easy approach to recover causal variants from exome sequencing data, herewith contributing to the reduction of the diagnostic deadlock. The aim of this work was to study the effect of different methods of multi-walled carbon nanotubes (MWCNTs) dispersion, and their influence on the microstructure and properties of aluminium alloy matrix composites produced using the powder metallurgy techniques, such as powder milling/mixing and hot extrusion. The main problem in the manufacturing of nanocomposites is the homogeneous distribution of MWCNTs in the metal matrix. To achieve their proper distribution a high-energy and low-energy mechanical milling, using a planetary ball mill, and mixing, using a turbulent mixer, were applied. Studies have shown that composite materials prepared using milling and extrusion have a much better dispersion of the reinforcing phase, which leads to better mechanical properties of the obtained rods. The low-energy mechanical mixing and mixing using the turbulent mixer neither change the powder morphology nor lead to adequate dispersion of the carbon nanotubes, which directly affects the resulting properties.

The main purpose of this work is to determine the effect of milling time on microstructure as well as technological properties of aluminium matrix nanocomposites reinforced with multi-walled carbon nanotubes (MWCNTs) using powder metallurgy techniques, including mechanical alloying. The main problem of the study is the agglomeration and uneven distribution of carbon nanotubes in the matrix material and interface reactivity also. In order to reach uniform dispersion of carbon nanotubes in aluminium alloy matrix, 5÷20 h of mechanical milling in the planetary mill was used. It was found that the mechanical milling process has a strong influence on the characteristics of powders, by changing the globular morphology of as-received powder during mechanical milling process to flattened one, due to particle plastic deformation followed by cold welding and fracturing of deformed and hardened enough particles, which allows to obtain equiaxial particles again. The obtained composites are characterised by the structure of evenly distributed, disperse reinforcing particles in fine grain matrix of AA6061, facilitate the obtainment of higher values of mechanical properties, compared to the initial alloy. On the basis of micro-hardness, analysis has found that a small addition of carbon nanotubes increases nanocomposite hardness.

Contains fulltext : 282710.pdf (Publisher’s version ) (Open Access), In 2016, guidelines for diagnostic Next Generation Sequencing (NGS) have been published by EuroGentest in order to assist laboratories in the implementation and accreditation of NGS in a diagnostic setting. These guidelines mainly focused on Whole Exome Sequencing (WES) and targeted (gene panels) sequencing detecting small germline variants (Single Nucleotide Variants (SNVs) and insertions/deletions (indels)). Since then, Whole Genome Sequencing (WGS) has been increasingly introduced in the diagnosis of rare diseases as WGS allows the simultaneous detection of SNVs, Structural Variants (SVs) and other types of variants such as repeat expansions. The use of WGS in diagnostics warrants the re-evaluation and update of previously published guidelines. This work was jointly initiated by EuroGentest and the Horizon2020 project Solve-RD. Statements from the 2016 guidelines have been reviewed in the context of WGS and updated where necessary. The aim of these recommendations is primarily to list the points to consider for clinical (laboratory) geneticists, bioinformaticians, and (non-)geneticists, to provide technical advice, aid clinical decision-making and the reporting of the results.

- Abstract
Research in global change ecology relies heavily on global climatic grids derived from estimates of air temperature in open areas at around 2 m above the ground. These climatic grids do not reflect conditions below vegetation canopies and near the ground surface, where critical ecosystem functions occur and most terrestrial species reside. Here, we provide global maps of soil temperature and bioclimatic variables at a 1-km² resolution for 0‐5 and 5‐15 cm soil depth. These maps were created by calculating the difference (i.e. offset) between in situ soil temperature measurements, based on time series from over 1200 1‐km² pixels (summarized from 8519 unique temperature sensors) across all the world’s major terrestrial biomes, and coarse-grained air temperature estimates from ERA5-Land (an atmospheric reanalysis by the European Centre for Medium-Range Weather Forecasts). We show that mean annual soil temperature differs markedly from the corresponding gridded air temperature, by up to 10° degrees C (mean = 3.0 +/‐ 2.1° degrees C), with substantial variation across biomes and seasons. Over the year, soils in cold and/or dry biomes are substantially warmer (+3.6 +/‐2.3° degrees C) than gridded air temperature, whereas soils in warm and humid environments are on average slightly cooler (‐0.7 +/‐ 2.3° degrees C). The observed substantial and biome-specific offsets emphasize that the projected impacts of climate and climate change on near-surface biodiversity and ecosystem functioning are inaccurately assessed when air rather than soil temperature is used, especially in cold environments. The global soil-related bioclimatic variables provided here are an important step forward for any application in ecology and related disciplines. Nevertheless, we highlight the need to fill remaining geographic gaps by collecting more in situ measurements of microclimate conditions to further enhance the spatiotemporal resolution of global soil temperature products for ecological
