Abstract

Intestinal mucosal integrity is dependent on epithelial function and a regulated immune response to injury. Fucosyltransferase VII (Fuc-TVII) is an essential enzyme required for the expression of the functional ligand for E- and P-selectin. Trefoil factor 3 (TFF3) is involved in both protecting the intestinal epithelium against injury as well as aiding in wound repair following injury. The aim of the present study was to assess the interplay between barrier function and leukocyte recruitment in intestinal inflammation. More specifically, we aimed to examine how targeted disruption of Fuc-TVII either in wild-type or [TFF3.sup.-/-] mice would alter their susceptibility to colonic injury. TFF3 and Fuc-TVII double-knockout mice (TFF3/Fuc-[TVII.sup.-/-] mice) were generated by mating [TFF3.sup.-/-] and Fuc-[TVII.sup.-/-] mice. Colitis was induced by administration of dextran sodium sulfate (DSS) (2.5% wt/vol) in the drinking water. Changes in baseline body weight, diarrhea, and fecal blood were assessed daily. Upon euthanasia, extents of colonic inflammation were assessed macroscopically, microscopically, and through quantification of myeloperoxidase (MPO) activity. Colonic lymphocyte subpopulations were assessed at 6 days after administration of DSS by flow cytometry and immunohistochemistry. No baseline intestinal inflammation was found in TFF3/Fuc-[TVII.sup.-/-] [TFF3.sup.-/-], Fuc-[TVII.sup.-/-], or wild-type mice. Loss of Fuc-TVII resulted in a reduction in disease severity whereas [TFF3.sup.-/-] mice were markedly more susceptible to DSS-induced colitis. Remarkably, the loss of Fuc-TVII in [TFF3.sup.-/-] mice markedly decreased the severity of DSS-induced colitis as evidenced by reduced weight loss, diarrhea, decreased colonic MPO levels and improved survival. Furthermore, the loss of TFF3 resulted in increased severity of spontaneous colitis in IL-2/[beta-microglobulin-deficient mice. These studies highlight the importance of the interplay between factors involved in the innate immune response, mucosal barrier function, and genes involved in regulating leukocyte recruitment and other aspects of the immune response. trefoil factor family 3 doi: 10.1152/ajpgi.00228.2009.

Abstract

A study has been made on the properties of lead fluoride as a Cherenkov material for use in electromagnetic calorimetry. A prototype calorimeter module consisting of a 5 x 5 array of 2.1 x 2.1 x 18.5 |cm.sup.3~ crystals has been built and tested in a test beam at the Brookhaven AGS. Results are given on energy resolution, shower size and e/|Pi~ separation for electrons and pions in the range from 1-4 GeV. The light output has been measured to give |greater than or similar to~ 1000 photoelectrons per MeV in good quality crystals, and to provide useful signals down to as low as 32 MeV. Measurements were also made on radiation damage in lead fluoride using 60Co gamma rays and high energy ionizing particles, as well as on thermoluminescence after irradiation. It was found that only modest damage occurs up to a level of |similar to~ 30 Krad in large, calorimeter size crystals, and that the damage can be easily removed by optical bleaching.

Abstract

Radiographic contrast agents cause acute kidney injury (AKI), yet the underlying pathogenesis is poorly understood. Nod-like receptor pyrin containing 3-deficient (Nlrp3-deficient) mice displayed reduced epithelial cell injury and inflammation in the kidney in a model of contrast-induced AKI (CI-AKI). Unexpectedly, contrast agents directly induced tubular epithelial cell death in vitro that was not dependent on Nlrp3. Rather, contrast agents activated the canonical Nlrp3 inflammasome in macrophages. Intravital microscopy revealed diatrizoate (DTA) uptake within minutes in perivascular CX3CR1+ resident phagocytes in the kidney. Following rapid filtration into the tubular luminal space, DTA was reabsorbed and concentrated in tubular epithelial cells via the brush border enzyme dipeptidase-1 in volume-depleted but not euvolemic mice. LysM-GFP+ macrophages recruited to the kidney interstitial space ingested contrast material transported from the urine via direct interactions with tubules. CI-AKI was dependent on resident renal phagocytes, IL-1, leukocyte recruitment, and dipeptidase-1. Levels of the inflammasome-related urinary biomarkers IL-18 and caspase-1 were increased immediately following contrast administration in patients undergoing coronary angiography, consistent with the acute renal effects observed in mice. Taken together, these data show that CI-AKI is a multistep process that involves immune surveillance by resident and infiltrating renal phagocytes, Nlrp3-dependent inflammation, and the tubular reabsorption of contrast via dipeptidase-1.Copyright © 2018 American Society for Clinical Investigation. 26. Recovery of renal function in end-stage renal failure - Comparison between peritoneal dialysis and haemodialysis.

Abstract

Background. Recovery of dialysis-independent renal function in long-term dialysis patients has not been studied extensively. The aim of this study was to investigate the effect of dialysis modality on the likelihood, timing and durability of recovery of dialysis-independent renal function.Methods. The study reviewed all patients in Australia and New Zealand who commenced dialysis for treatment of end-stage renal disease (ESRD) between 1963 and 2006. Dialysis modality was assigned at 90 days. A supplementary analysis was also conducted using a contemporary cohort that included data on comorbidities, smoking and eGFR at dialysis onset.Results. During the study period, 15912 individuals received peritoneal dialysis (PD) and 23658 received haemodialysis (HD). Renal recovery occurred in 176 (1.1) PD and 244 (1.0) HD patients. Using multivariate Cox proportional hazards regression analyses, dialysis modality was not independently predictive of time to renal recovery (HR 0.92, 95 CI 0.76-1.13, P = 0.4). Recovery was significantly more likely in patients with higher baseline eGFR, with no hypertension or peripheral vascular disease, and with certain causes of kidney failure (autoimmune renal disease, haemolytic uraemic syndrome, interstitial nephritis, obstructive uropathy, paraproteinaemia and renovascular nephrosclerosis). Recovery was less likely in MaoriPacific Islanders and polycystic kidney disease. Among patients who recovered, 328 (78) subsequently experienced renal death, mostly within the first year. The duration of renal recovery was not associated with initial dialysis modality (OR 0.82, 95 CI 0.50- 1.32).Conclusions. Dialysis modality is not associated with the likelihood, timing or durability of spontaneous recovery of dialysis-independent renal function in patients thought to have ESRD.

Abstract

This paper reports the range and statistical distribution of oxidation rates of atmospheric CH4 in soils found in Northern Europe in an international study, and compares them with published data for various other ecosystems. It reassesses the size, and the uncertainty in, the global terrestrial CH4 sink, and examines the effect of land-use change and other factors on the oxidation rate. Only soils with a very high water table were sources of CH4, all others were sinks. Oxidation rates varied from 1 to nearly 200 µg CH4 m-2 h-1, annual rates for sites measured for =1 y were 0.1-9.1 kg CH4 ha-1 y-1, with a log-normal distribution (log-mean ~ 1.6 kg CH4 ha-1 y-1). Conversion of natural soils to agriculture reduced oxidation rates by two-thirds - closely similar to results reported for other regions. N inputs also decreased oxidation rates. Full recovery of rates after these disturbances takes > 100 y. Soil bulk density, water content and gas diffusivity had major impacts on oxidation rates. Trends were similar to those derived from other published work. Increasing acidity reduced oxidation, partially but not wholly explained by poor diffusion through litter layers which did not themselves contribute to the oxidation. The effect of temperature was small, attributed to substrate limitation and low atmospheric concentration. Analysis of all available data for CH4 oxidation rates in situ showed similar log-normal distributions to those obtained for our results, with generally little difference between different natural ecosystems, or between short-and longer-term studies. The overall global terrestrial sink was estimated at 29 Tg CH4 y-1, close to the current IPCC assessment, but with a much wider uncertainty range (7 to > 100 Tg CH4 y-1). Little or no information is available for many major ecosystems, these should receive high priority in future research.

Abstract

The exchange of CH4 and CO2 over extensive tracts of peatland has recently been measured using micrometeorological methods. Fluxes of CH4 measured in Caithness (UK) during May and June 1994 using eddy covariance yielded fluxes in the range –70 to +170 µmol CH4 m–2 h–1 with a mean flux of 39 µmol CH4 m–2 h–1 Landscape emissions of CH4 from peatland are shown to vary with water table and temperature, increasing from 25 µmol CH4 m–2 h–1 at 5 per cent area to 50 µmol CH4 m–2 h–1 with 30 per cent within the footprint occupied by pool. A positive temperature response of 4.9 µmol CH4 m–2 h–1 {ring} C– in the temperature range 6–12{ring}C was also observed The mean day time fluxes of CO2 during May and June 1994, of —3.6 mmol m–2 h–1 were two orders of magnitude larger than the emission of CH4 of 40 µmol m–2 h–1, while at night (when net radiation is negative) the respiration flux of CO2 averaged +2.2 mmol m–2 h–1approximately two orders of magnitude larger than night-time CH4 emissions of 30 µmol m–2 h–1.
