In this paper we present a framework in which one can rigorously study the effect of spatio-temporal noise on traveling waves, stationary patterns, and oscillations that are invariant under the action of a finite-dimensional set of continuous isometries (such as translation or rotation). This formalism can accommodate patterns, waves, and oscillations in reaction-diffusion systems and neural field equations. To do this, we define the phase by precisely projecting the infinite-dimensional system onto the manifold of isometries. We outline a precise stochastic differential equation for the phase. The phase is then used to show that the probability of the system leaving the attracting basin of the manifold after an exponentially long period of time (in ε -2, the magnitude of the noise) is exponentially unlikely. Disorders of mineral metabolism, including secondary hyperparathyroidism, are thought to contribute to extraskeletal (including vascular) calcification among patients with chronic kidney disease. It has been hypothesized that treatment with the calcimimetic agent cinacalcet might reduce the risk of death or nonfatal cardiovascular events in such patients.In this clinical trial, we randomly assigned 3883 patients with moderate-to-severe secondary hyperparathyroidism (median level of intact parathyroid hormone, 693 pg per milliliter [10th to 90th percentile, 363 to 1694]) who were undergoing hemodialysis to receive either cinacalcet or placebo. All patients were eligible to receive conventional therapy, including phosphate binders, vitamin D sterols, or both. The patients were followed for up to 64 months. The primary composite end point was the time until death, myocardial infarction, hospitalization for unstable angina, heart failure, or a peripheral vascular event. The primary analysis was performed on the basis of the intention-to-treat principle.The median duration of study-drug exposure was 21.2 months in the cinacalcet group, versus 17.5 months in the placebo group. The primary composite end point was reached in 938 of 1948 patients (48.2%) in the cinacalcet group and 952 of 1935 patients (49.2%) in the placebo group (relative hazard in the cinacalcet group vs. the placebo group, 0.93; 95% confidence interval, 0.85 to 1.02; P=0.11). Hypocalcemia and gastrointestinal adverse events were significantly more frequent in patients receiving cinacalcet.In an unadjusted intention-to-treat analysis, cinacalcet did not significantly reduce the risk of death or major cardiovascular events in patients with moderate-to-severe secondary hyperparathyroidism who were undergoing dialysis. Background— Patients with kidney disease have disordered bone and mineral metabolism, including elevated serum concentrations of fibroblast growth factor-23 (FGF23). These elevated concentrations are associated with cardiovascular and all-cause mortality. The objective was to determine the effects of the calcimimetic cinacalcet (versus placebo) on reducing serum FGF23 and whether changes in FGF23 are associated with death and cardiovascular events. Methods and Results— This was a secondary analysis of a randomized clinical trial comparing cinacalcet to placebo in addition to conventional therapy (phosphate binders/vitamin D) in patients receiving hemodialysis with secondary hyperparathyroidism (intact parathyroid hormone ≥300 pg/mL). The primary study end point was time to death or a first nonfatal cardiovascular event (myocardial infarction, hospitalization for angina, heart failure, or a peripheral vascular event). This analysis included 2985 patients (77% of randomized) with serum samples at baseline and 2602 patients (67%) with samples at both baseline and week 20. The results demonstrated that a significantly larger proportion of patients randomized to cinacalcet had ≥30% (68% versus 28%) reductions in FGF23. Among patients randomized to cinacalcet, a ≥30% reduction in FGF23 between baseline and week 20 was associated with a nominally significant reduction in the primary composite end point (relative hazard, 0.82; 95% confidence interval, 0.69–0.98), cardiovascular mortality (relative hazard, 0.66; 95% confidence interval, 0.50–0.87), sudden cardiac death (relative hazard, 0.57; 95% confidence interval, 0.37–0.86), and heart failure (relative hazard, 0.69; 95% confidence interval, 0.48–0.99). Conclusions— Treatment with cinacalcet significantly lowers serum FGF23. Treatment-induced reductions in serum FGF23 are associated with lower rates of cardiovascular death and major cardiovascular events. Clinical Trial Registration— URL: http://www.clinicaltrials.gov . A joint research project was undertaken by the National Research Council of Canada (NRCC) and Canada Mortgage and Housing Corporation (CMHC) to study evacuation drills in highrise apartment buildings with mixed abilities occupants. A similar project had been carried out in 1993-94 on midrise apartment buildings. This second study collected real data on time and movement during evacuation drills in three highrise buildings. The buildings were located in Montreal, Calgary and Gloucester, Ont. These different locations reflected provincial variations in fire safety procedures. Each fire drill was organized, supervised and carried out with the full participation of local fire departments or local fire protection services.

This report presents the temperature measurements from (7) full-scale fire resistance tests conducted at the National Fire Laboratory on non-insulated, loaded and non-loaded, regular gypsum board wall protected assemblies. Assemblies were 1x1 (one layer of gypsum board on both the exposed and unexposed sides) using wood studs and 2x2 (two layers of board on both the exposed and unexposed sides) using lightweight steel and wood studs. Three regular gypsum boards with different masses per unit area wereevaluated: 7.82 kg/m2 with no glass fibre in the gypsum core, 7.35 kg/m2 with glass fibre in the gypsum core and 7.27 kg/m2 with no glass fibre in the gypsum core. Tests were conducted to determine the fire resistance rating of the assemblies. The temperatures measured on the gypsum board surfaces and on the studs are presented.

The two assembly arrangements studied were: asymmetrical installation 1x2 (one layer of gypsum board on the exposed side and two layers of gypsum board on the unexposed side) and symmetrical installation 2x2 (two layers of boards on each of the exposed and unexposed sides) on wood studs. Twogypsum board thicknesses were considered: 12.7 mm and 15.9 mm Type X. The insulations used were glass, mineral and cellulose (blown dry) fibres. Tests were conducted to determine the effects of resilient channel installation, insulation types, and gypsum board thickness in symmetrical and asymmetrical installations on the fire resistance performance of gypsum board protected wall assemblies. The tcmperatures measured on the gypsum board surfaces and on the studs are presented.

Udgivelsesdato: 2001-Oct-15, p53 is a transcriptional activator which has been implicated as a key regulator of neuronal cell death after acute injury. We have shown previously that p53-mediated neuronal cell death involves a Bax-dependent activation of caspase 3; however, the transcriptional targets involved in the regulation of this process have not been identified. In the present study, we demonstrate that p53 directly upregulates Apaf1 transcription as a critical step in the induction of neuronal cell death. Using DNA microarray analysis of total RNA isolated from neurons undergoing p53-induced apoptosis a 5-6-fold upregulation of Apaf1 mRNA was detected. Induction of neuronal cell death by camptothecin, a DNA-damaging agent that functions through a p53-dependent mechanism, resulted in increased Apaf1 mRNA in p53-positive, but not p53-deficient neurons. In both in vitro and in vivo neuronal cell death processes of p53-induced cell death, Apaf1 protein levels were increased. We addressed whether p53 directly regulates Apaf1 transcription via the two p53 consensus binding sites in the Apaf1 promoter. Electrophoretic mobility shift assays demonstrated p53-DNA binding activity at both p53 consensus binding sequences in extracts obtained from neurons undergoing p53-induced cell death, but not in healthy control cultures or when p53 or the p53 binding sites were inactivated by mutation. In transient transfections in a neuronal cell line with p53 and Apaf1 promoter-luciferase constructs, p53 directly activated the Apaf1 promoter via both p53 sites. The importance of Apaf1 as a p53 target gene in neuronal cell death was evaluated by examining p53-induced apoptotic pathways in primary cultures of Apaf1-deficient neurons. Neurons treated with camptothecin were significantly protected in the absence of Apaf1 relative to those derived from wild-type littermates. A comparison of the effects of various oestrogenic substances on coronary and aortic atheroma in cholesterol-fed birds has been made.In general, oestrogenic activity paralleled the ability to regress coronary atherosclerosis but there were exceptions.Doisynolic acid, in a dose which produced side-effects comparable with oestradiol, did not significantly alter the incidence of coronary atherosclerosis.Epi-oestradiol, in an experiment of longer duration, and given in a dose which made it a potent oestrogenic substance with typical effects on serum lipids, gonads and sex characters, did not significantly reduce the incidence of coronary atherosclerosis.

Using specimens similar to those described in the draft revision of ASTM E 119, tests were conducted in floor and wall fire test furnaces. The tests were designed to compare the practical consequences of following the ASTM E 119 and ISO 834 specifications for measuring and controlling the nominal furnace temperature. It has been found that in fire tests of shorter duration, perhaps up to 1.5h, the ASTM test is slightly more severe than the ISO test. In longer tests the difference in severities is negligible.
