The in vitro maturation efficiency of porcine oocytes is relatively low, and this limits the production of in vitro porcine embryos. Since melatonin is involved in mammalian reproductive physiology, in this study, we have explored whether endogenously produced melatonin can help in porcine oocyte in vitro maturation. We have found, for the first time in the literature, that mitochondria are the major sites for melatonin biosynthesis in porcine oocytes. This mitochondrially originated melatonin reduces ROS production and increases the activity of the mitochondrial respiratory electron transport chain, mitochondrial biogenesis, mitochondrial membrane potential, and ATP production. Therefore, melatonin improves the quality of oocytes and their in vitro maturation. In contrast, the reduced melatonin level caused by siRNA to knockdown AANAT (siAANAT) is associated with the abnormal distribution of mitochondria, decreasing the ATP level of porcine oocytes and inhibiting their in vitro maturation. These abnormalities can be rescued by melatonin supplementation. In addition, we found that siAANAT switches the mitochondrial oxidative phosphorylation to glycolysis, a Warburg effect. This metabolic alteration can also be corrected by melatonin supplementation. All these activities of melatonin appear to be mediated by its membrane receptors since the non-selective melatonin receptor antagonist Luzindole can blunt the effects of melatonin. Taken together, the mitochondria of porcine oocytes can synthesize melatonin and improve the quality of oocyte maturation. The development of space exploration technologies puts higher demand on space power systems. The space reactor gas turbine cycle (SRGTC) has the advantages of compact configuration and small mass and volume and is one of the optimal options for future high-power space power systems. The SRGTC operates in an isolated island state and the radiator is fragile if it is hit by asteroids and space debris. The transient characteristics of the SRGTC are fundamental for safe operation in radiator loss accidents. In this paper, a dynamic model for the SRGTC is established, and the performance of the SRGTC with dual power conversion unit (PCU) modules (SRGTC-DPCU) after radiator loss was investigated. The results indicated that the waste heat of the system was accumulated in the radiator after radiator loss, which increased the consumed power of the compressor and further led to speed fluctuations. The bypass valve control and the temperature negative feedback effect can ensure the safe operation of the shaft and the reactor. More radiator loss decreased the speed far below the rated speed and exceeded the safety margin of the bypass valve control, which further led to system shutdown. There is a safety boundary of radiator loss accidents. Furthermore, the coupling effects of multiple PCUs after radiator loss were analyzed. The working fluid inventory was redistributed among the PCU modules. The reduction of working fluid inventory in the accident PCU module promoted the safety boundary. Space exploration technology is an important indicator of society's technological level. A space reactor coupled with a Brayton cycle is preferable for megawatt-scale space power systems. Noble gas binary mixtures have high chemical stability, heat transfer performance, and compressibility, making them the principal choice of working fluid for the space reactor Brayton cycle, which is also the key factor affecting the thermodynamic performance and mass of the system. This study developed thermodynamic performance and mass evaluation models for the space nuclear Brayton cycle and discovered the inherent relationship between system thermodynamic performance and mass. The effects of noble gas binary mixtures on system performance and mass were investigated. The results indicated that the elevated molar mass of noble gas binary mixtures reduced the aerodynamic load and mass of the turbomachines and increased the mass of the recuperator. There are optimal values of the total mass, specific mass of the system, and working fluid composition. Helium-xenon mixture is the optimal working fluid because it can achieve the highest thermodynamic efficiency and lowest mass. Furthermore, the optimal scheme of the helium-xenon Brayton cycle for a space nuclear power system was obtained by multi-objective optimization. Its power generation efficiency, specific mass, and helium molar fraction in the helium-xenon working fluid are 29.04%, 5.65 t ∙ MW − 1 and 77.5%, respectively.
