WRKY protein is an important transcription factor in response to abiotic stress in plants. However, a systematic identification and characterization of WRKY genes has not been carried out for the medicinal plant Polygonatum sibiricum that has a strong ability to resist various abiotic stresses. In this study, we isolated a novel WRKY gene from P. sibiricum and compared its sequence structure with other plants. PsWRKY1 possesses two typical WRKY domains and two C2H2 zinc-finger motifs. Evolutionary analysis indicated that PsWRKY1 is most closely related to the WRKY protein from Eucalyptus grandis. Expression analysis showed that expression levels of PsWRKY1 were induced by cold and drought stresses but not salt stress. Overexpression of PsWRKY1 in Arabidopsis improved the seed germination and growth conditions of transgenic plants under drought and cold stresses. Furthermore, SOD activity and proline content in transgenic plants were higher than those in WT under cold and drought stresses, whereas MDA levels and relative electrolyte leakage in transgenic plants were lower than those in WT under same stresses. These results indicated that PsWRKY1 improved the tolerance to cold and drought stresses. OBJECTIVE: In order to investigate the role of the zinc finger E-box-binding homeobox 1 (ZEB1) expression in the incidence of non-small cell lung cancer (NSCLC), the effects of the ZEB1 expression on the pathogenesis and prognosis of NSCLC were investigated. PATIENTS AND METHODS: Correlations of the expression of ZEB1 in 88 clinical patients with NSCLC with clinicopathological features were determined. The patients were followed up for 60 months to study the correlation between the ZEB1 expression and the prognosis of NSCLC patients. To further explore the role of the ZEB1 expression in the incidence of NSCLC, the expressions of ZEB1 and E-cadherin in three NSCLC cell lines (A549, NCI-H1299 and NCI-H1975) and human normal lung epithelial BESA-2B cell line were measured, and the cell invasion ability was detected. The role of ZEB1 in NSCLC cell invasion was verified through the knockdown of ZEB1 by the short hairpin ribonucleic acids (shRNAs). In addition, its effects on the proliferation and apoptosis of NSCLC cells were confirmed by the overexpression of ZEB1. RESULTS: The expression of ZEB1 in NSCLC tissues was remarkably higher than that in normal tissues, and the expression level of ZEB1 was significantly related to the cancer stage and tumor size. The lower the expression of ZEB1 was, the higher the overall survival rate and the longer the survival time would be. The expression of ZEB1 was negatively correlated with that of E-cadherin in cell lines, and ZEB1-shRNAs markedly reduced the invasion ability of NSCLC cells. The overexpression of ZEB1 resulted in an increase in the proliferation activity and a significant decrease in the apoptosis of A549 cells. CONCLUSIONS: The expression of ZEB1 is closely related to the incidence and prognosis of NSCLC. Increased expression of ZEB1 is helpful for promoting the invasion of NSCLC and enhancing the proliferation activity of NSCLC. [ABSTRACT FROM AUTHOR]

The Lawn-Evans-Marshall model for Vickers-induced crack driving force is widely applied for determining fracture toughness of ceramic materials, yet its simplification that the volumes of Vickers impression and equivalent expanding cavity merely depend on the half-diagonal length of residual impression will bring significant error. In this work, the model is modified by considering the influence of elastic recovery in depth after completely unloading on the volumes. As a result, a new expression for crack driving force is obtained and correspondingly a new fracture toughness formula is presented based on the modified model, which is expressed as the well-known Anstis formula with a different constant multiplied by the ratio of residual impression depth to the maximum indentation depth. Both finite element analysis and experimental results demonstrate the effectiveness of the proposed formula and, especially, that it works much better than Anstis formula on materials with large ratio of hardness to elastic modulus. To study lead (Pb) distribution in organs and blood in the case of Pb poisoning, mice were firstly exposed to Pb as 0.1 mL or 0.2 mL of lead nitrate solution (0.1 mg/mL) by vein injection every other day. Then, after metabolic absorption, the Pb level in the blood and organs of the mice was measured using flame atomic absorption spectrometry. The resulting data showed that 93% of Pb in blood wass accumulated in red cells, but this percentage slightly decreased with increasing exposure time and injection volume. For other target organs, the highest Pb level was in the kidney, followed by the liver, spleen, heart and lung, and was lowest in the brain. Moreover, the Pb level in the heart and brain is in a growth trend at all times for 0.1 mL and 0.2 mL of Pb injection exposure in 15 days, while the growth trend of Pb in other target organs become slow for 0.2 mL of injection after exposure Pb 11 days. Objective: Dose de-escalation of adjuvant therapy (DART) in patients with HPV(+)OPSCC was investigated in two prospective Phase II and III clinical trials (MC1273 and MC1675). We report the 30-day morbidity and mortality associated with primary TORS resection in patients enrolled in these trials., Materials and Methods: Patients with HPV(+)OPSCC, who underwent TORS resection between 2013 and 2020 were considered in this analysis. The severity of postoperative transoral bleeding was graded using both the Hinni Grade (HG) transoral surgery bleeding scale and the Common Terminology for Adverse Events (CTCAE) v5.0. Post-surgical complications within 30 days of surgery, as well as rates of tracheostomy, PEG and nasogastric tube placement., Results: 219 patients were included. A total of 7 (3.2 %) patients had a tracheostomy placed at the time of surgery, and all were decannulated within 26 days (median: 5, range: 2-26). There were 33 (15.1 %) returns to the emergency department (ED) with 10 (4.6 %) patients requiring readmission. Using the HG scale, 10 (4.6 %) patients experienced ≥ Grade 3 bleeding with no Grade 5 or 6 bleeds. In contrast, using the CTCAE scale, 15 patients (6.8 %) experienced ≥ Grade 3 bleeding with no Grade 5 bleeds. Objective: To investigate the effect of silencing hepatocyte growth factor receptor (c-Met) expression on the biological characteristics of HCT116 colon cancer cells. Methods: Cellular model of c-Met transient transfection was established by using small interfering RNA (siRNA), the expression of c-Met in colon cancer cells was detected by real-time fluorescence quantitative polymerase chain reaction (RT-qPCR) and western blot. The apoptosis assay, cell invasion assay, cell migration and other experiments were conducted to observe the effects of silencing c-Met on the biological characteristics of colon cancer cells. Results: RT-qPCR results showed that the relative expression levels of c-Met mRNA in siRNA-Met group, blank control group and siRNA negative control (siRNA-NC) group were 0.32±0.26, 1.01±0.03 and 1.05±0.23, respectively, and the difference was statistically significant ( P <0.05). Western blot analysis showed that the expression level of c-Met protein in the siRNA-Met group was 0.24±0.03, significantly lower than 1.23±0.06 in the blank control group and 1.18±0.11 in the siRNA-NC group ( P <0.05). The cell counting kit-8 (CCK8) results showed that the 72-hour absorbance (A) values of the siRNA-Met group, blank control group and the siRNA-NC group were 1.13±0.05, 1.48±0.08 and 1.53±0.07, respectively, and the difference was statistically significant ( P <0.01). Cell cycle results showed that the proportion of cells in G(2)/M phase was (14.65±1.41)% in siRNA-Met group , (5.07±0.70)% in blank control group and (5.63±0.71)% in siRNA-NC group, and the difference was statistically significant ( P <0.05). The expression levels of cell cycle regulatory proteins Cdc25c and cyclin B1 in siRNA-Met group were significantly decreased. The apoptotic rate in siRNA-Met group was (5.85±0.35)%, significantly higher than (1.00±0.17)% in blank control group and (0.91±1.14)% in siRNA-NC group ( P <0.05). The expression level of apoptosis-related protein Bcl-2 in the siRNA-Met group was significantly decreased while Bcl-2 associated X protein (BAX) expression level was significantly increased. The cell scratching result showed that the cell migration abilities of the siRNA-Met group, blank control group and the siRNA-NC group were (51.33±8.62)%, (100.00±3.72)% and (102.33±6.43)%, respectively, and the difference was statistically significant ( P <0.05). The number of cell penetrating into the basement membrane of the siRNA-Met group, blank control group and the siRNA-NC group were 47.50±10.60, 100.00±5.33 and 102.50±10.61, respectively, and the difference was statistically significant ( P <0.05). The expressions of invasion related proteins including MMP-2 and MMP-9 in siRNA-Met group were decreased significantly. Conclusions: c-Met plays an important role in maintaining the biological characteristics of colon cancer cells. Inhibition of c-Met may have important values in the treatment of colon cancer.
