Wear is one of the main failure forms of metal parts， and the surface coating technology is one of the important methods to protect metals from wear. TiC-TiB2 dual-phase composite wear-resistant ceramic coating is widely used as a protective coating for material surface due to dual advantages of carbide and boride ceramics，such as higher hardness and better wear and corrosion resistance. In this paper，the research progress of in-situ TiC-TiB2 composite coating was reviewed from two aspects: preparation method and main metal binders of TiC-TiB2 coating.The advantages and disadvantages of different preparation methods for TiC-TiB2 coating were compared， and the problems and possible solutions in practical applications were referred. The synthesis mechanisms and the reasons for improving the performance of TiC-TiB2 coating after adding different metal binders were summarized. The advantages and disadvantages of various metal binders in actual applications were analyzed. Finally， the future optimization directions and urgent problems of the preparation process of TiC-TiB2coating were pointed out.

In this study, selective laser melting (SLM) additive manufacturing was used for the fabrication of Al alloy with the surface structure based on the construction of mechanical interlocking mechanism to complete the joining with polyetheretherketone (PEEK). The application of hot-pressing process provided a direct joining between Al alloy-PEEK at 460 °C for 40 min. This work characterized the forming of both cylindrical and trapezoidal structures, indicating that the structures were formed completely, and the gaps were clear. Although some dimples or micropores were generated in the forming solid, these defects in the forming were beneficial for the flowing and filling of PEEK during hot-pressing joint. The microstructure of Al plate fabricated by SLM showed typical parallel columnar grains perpendicular to the deposition direction and stacked and fish scale like feature in the deposition direction, respectively. The microstructure characterizations of Al alloy-PEEK joints showed that both materials were joined without gaps and formed a strong mechanical interlocking, guaranteeing a firm bond with maximal tensile shear strength up to 81 MPa for trapezoidal structure. By comparison, the joint with cylindrical structure possessed a low 65 MPa shear strength due to weak cuneiform anchor interlocking. Fracture surface was investigated in more detail. The result showed that the joint with cylindrical structure mainly showed interfacial failure and pulling-out, tilting deformation and fracture of the supporting, while the joint with trapezoidal structure exhibited a separation between the top surface of the supporting and the PEEK and brittle cohesive fracture of the PEEK between the gaps. MicroRNAs (miRNAs) are endogenously expressed small noncoding RNAs and play critical roles in the regulation of post-transcriptional gene expression. Our previous study uncovered that chi-miR-487b-3p is widespread in different goat tissues, which is significantly higher in muscle, especially in lamb. Here, we demonstrate the role of chi-miR-487b-3p as a myogenic miRNA that regulates skeletal muscle development. chi-miR-487b-3p overexpression was demonstrated to significantly inhibit goat myoblast proliferation and differentiation, whereas chi-miR-487b-3p inhibition resulted in the opposite effects. Next, chi-miR-487b-3p was predicted to target the 3′UTR of insulin receptor substrate 1 (IRS1) gene by Target-Scan and miRDB. The results of dual-luciferase assay, RT-qPCR, and western blot all confirmed that IRS1 might be a direct target of chi-miR-487b-3p as its expression was negatively regulated by chi-miR-487b-3p. siRNA silencing of IRS1 further demonstrated significant inhibition on goat myoblast proliferation and differentiation, confirming the effect of IRS1 downregulation by chi-miR-487b-3p in myogenesis. In addition, chi-miR-487b-3p knockout goat myoblast clones were generated using CRISPR/Cas9 technology, and we further illustrated that chi-miR-487b-3p regulates goat myoblast growth through the PI3K/Akt signaling pathway by targeting IRS1. Collectively, our work demonstrated that chi-miR-487b-3p is a potent inhibitor of skeletal myogenesis and provided new insights into the mechanisms of miRNA on the regulation of goat growth. Abstract: A novel ZrB2 based material named “hybrid composite” was prepared with the addition of SiC whiskers plus SiC particles by hot-pressing. Microstructure and mechanical properties of the composites were investigated. The introduction of fine SiC particles can increase the relative density and mechanical properties of ZrB2. Mechanical properties especially fracture toughness were better than that of ZrB2 composites reinforced with SiC particles or SiC whiskers alone. Based on the results presented in this work, it can be stated that the combination of SiC whiskers and SiC particles can be used as reinforcing materials for diborides based ultra-high temperature ceramics, due to their cooperative effect in densification and reinforcement. During the gene editing process mediated by CRISPR/Cas9, precise genome editing and gene knock-in can be achieved by the homologous recombination of double-stranded DNA (dsDNA) donor template. However, the low-efficiency of homologous recombination in eukaryotic cells hampers the development and application of this gene editing strategy. Here, we developed a novel CRISPR/Cas9-hLacI donor adapting system (DAS) to enhance the dsDNA-templated gene editing, taking the advantage of the specific binding of the LacI repressor protein and the LacO operator sequence derived for the Escherichia coli lactose operon. The codon-humanized LacI gene was fused as an adaptor to the Streptococcus pyogenes Cas9 ( SpCas9 ) and Staphylococcus lugdunensis Cas9 ( SlugCas9-HF ) genes, and the LacO operator sequence was used as the aptamer and linked to the dsDNA donor template by PCR. The Cas9 nuclease activity after the fusion and the homology-directed repair (HDR) efficiency of the LacO-linked dsDNA template were firstly examined using surrogate reporter assays with the corresponding reporter vectors. The CRISPR/Cas9-hLacI DASs mediated genome precise editing were further checked, and we achieved a high efficiency up to 30.5% of precise editing at the VEGFA locus in HEK293T cells by using the CRISPR/SlugCas9-hLacI DAS. In summary, we developed a novel CRISPR/Cas9-hLacI DAS for dsDNA-templated gene editing, which enriches the CRISPR/Cas9-derived gene editing techniques and provides a novel tool for animal molecular design breeding researches.

Recently, with the development and the continuous improvement of various CRISPR systems represented by CRISPR/Cas9, gene editing technology has been gradually improved, and widely applied to the preparation of animal models of human diseases. The gene edited animal models provide important materials for the study of pathogenesis, pathological process, prevention and treatment of human diseases. At present, the gene edited animal models used in human disease research include mainly the rodent models represented by mice and rats, and large animal models represented by pigs. Among them, rodents differ greatly from humans in all aspects of their bodies and have short life span as well, which cannot provide effective evaluation and long-term tracking for the research and treatment of human diseases. On the other hand, pig is closer to human in physiology, anatomy, nutrition and genetics, which provides an important animal model in the field of organ transplantation and human disease research. In this paper, the application of the gene edited animal models was summarized in the researches of 5 human diseases such as neurodegenerative diseases, familial hypertrophic cardiomyopathy, cancer, immunodeficiency diseases and metabolic diseases. We hope this paper will provide a reference for the research of human diseases and the preparation of relative animal models.

In order to study the modulatory effects of compound polysaccharide Erweikang on the immune function of the immunosuppressive mice, the positive immune modulation of Erweikang at different doses and different time points was compared when normal saline and cyclophosphamide served as controls. The results showed that different doses of Erweikang could effectively reverse the decreased induction levels of NK, IL-2, gamma-IFN and the contents of TNF and IgF of the immunosuppressive mice after administration of Erweikang for 7 to 14 days. It was suggested that compound polysaccharide Erweikang presents positive immune modulation in dose- and time-dependent manners.
