Abstract

The synergy of judiciously engineered nanostructures and complex topology of light creates unprecedented opportunities for tailoring light-matter interactions on the nanoscale. Electromagnetic waves can carry multiple units of angular momentum per photon, stemming from both spin and orbital angular momentum contributions, offering a potential route for modifying the optical transition selection rules. However, the size difference between a vortex beam and quantum objects limits the interaction strength and the angular momentum exchange. Here, we demonstrate the sub-diffraction-limited focusing of a vortex beam using the high in-plane wave number modes present in hyperbolic metamaterials. The spin-orbit interaction within the hyperbolic structure gives rise to the formation of an optical skyrmion with a deep subwavelength structure, which may enable the exploration of new light-matter interaction phenomena., Comment: 11 pages, 3 figures

Abstract

BACKGROUND: Recurrent atypical teratoid/rhabdoid tumor (AT/RT) is, most often, a fatal pediatric malignancy with limited curative options. METHODS: We conducted a phase II study of Aurora kinase A inhibitor alisertib in patients aged 12 months. PFS did not differ by AT/RT molecular groups. Neutropenia was the most common adverse effect (n = 23/30, 77%). The 22 patients who received liquid formulation had a higher mean maximum concentration (Cmax) of 10.1 ± 3.0 µM and faster time to Cmax (Tmax = 1.2 ± 0.7 h) than those who received tablets (Cmax = 5.7 ± 2.4 µM, Tmax = 3.4 ± 1.4 h). CONCLUSIONS: Although the study did not meet predetermined efficacy end point, single-agent alisertib was well tolerated by children with recurrent AT/RT, and SD or PR was observed in approximately a third of the patients.

Abstract

The genetics of relapsed pediatric acute myeloid leukemia (AML) has yet to be comprehensively defined. Here, we present the spectrum of genomic alterations in 136 relapsed pediatric AMLs. We identified recurrent exon 13 tandem duplications (TD) in upstream binding transcription factor (UBTF) in 9% of relapsed AML cases. UBTF-TD AMLs commonly have normal karyotype or trisomy 8 with cooccurring WT1 mutations or FLT3-ITD but not other known oncogenic fusions. These UBTF-TD events are stable during disease progression and are present in the founding clone. In addition, we observed that UBTF-TD AMLs account for approximately 4% of all de novo pediatric AMLs, are less common in adults, and are associated with poor outcomes and MRD positivity. Expression of UBTF-TD in primary hematopoietic cells is sufficient to enhance serial clonogenic activity and to drive a similar transcriptional program to UBTF-TD AMLs. Collectively, these clinical, genomic, and functional data establish UBTF-TD as a new recurrent mutation in AML.SignificanceWe defined the spectrum of mutations in relapsed pediatric AML and identified UBTF-TDs as a new recurrent genetic alteration. These duplications are more common in children and define a group of AMLs with intermediate-risk cytogenetic abnormalities, FLT3-ITD and WT1 alterations, and are associated with poor outcomes. Abstract

We demonstrate a voltage-controlled exchange bias effect in CoFeB/MgO/CoFeB magnetic tunnel junctions that is related to the interfacial Fe(Co)Ox formed between the CoFeB electrodes and the MgO barrier. The unique combination of interfacial antiferromagnetism, giant tunneling magnetoresistance, and sharp switching of the perpendicularly-magnetized CoFeB allows sensitive detection of the exchange bias. It is found that the exchange bias field can be isothermally controlled by magnetic fields at low temperatures. More importantly, the exchange bias can also be effectively manipulated by the electric field applied to the MgO barrier due to the voltage-controlled antiferromagnetic anisotropy in this system.

Abstract

The goal of this thesis is to obtain the statistics of transported quantities in non-equilibrium steady states of both classical and quantum systems. This transport is considered over a long time period where the large deviation principle is satisfied. Thus, the statistics of transported quantities will be described via the Scaled Cumulant Generating Functional (SCGF). Initially I introduce a simple approach for obtaining the SCGF dubbed ‘boot-strapping’. This is limited to systems that display a chiral separation and satisfy the Gallavotti-Cohen fluctuation relation which is expressed as a symmetry on the SCGF. I then present the primary result of this work, the Ballistic Fluctuation Formalism(BFF). This approach allows for the derivation of the SCGF for ballistically transported quantities in a maximal entropy state that has arisen in the long-time limit of a system governed by Euler hydrodynamics. The BFF represents an important new organisational principle for non-equilibrium states in both classical and quantum systems. The remainder of this thesis is the application of the BFF to three different systems. The first is a generalised Totally Asymmetric Exclusion Process, a classical system,where the SCGF for current transport is obtained. The second is a d-dimensional Con-formal Field Theory, a quantum system, where the first known expression for the SCGF of energy transport is obtained. Finally, the BFF is applied to integrable models that can be described by generalized hydrodynamics via the Thermodynamic Bethe Ansatz, where these can be either classical or quantum systems. The BFF is used to obtain a general expression for the SCGF applicable to any transported conserved quantity of any such system. These results are applied to the classical hard rod gas and the quantum Lieb-Liniger gas.

12. Fluctuations in ballistic transport from Euler hydrodynamics

Abstract

We propose a general formalism, within large deviation theory, giving access to the exact statistics of fluctuations of ballistically transported conserved quantities in homogeneous, stationary states. The formalism is expected to apply to any system with an Euler hydrodynamic description, classical or quantum, integrable or not, in or out of equilibrium. We express the exact scaled cumulant generating function (or full counting statistics) for any (quasi-)local conserved quantity in terms of the flux Jacobian. We show that the "extended fluctuation relations" of Bernard and Doyon follow from the linearity of the hydrodynamic equations, forming a marker of "freeness" much like the absence of hydrodynamic diffusion does. We show how an extension of the formalism gives exact exponential behaviours of spatio-temporal two-point functions of twist fields, with applications to order-parameter dynamical correlations in arbitrary homogeneous, stationary state. We explain in what situations the large deviation principle at the basis of the results fail, and discuss how this connects with nonlinear fluctuating hydrodynamics. Applying the formalism to conformal hydrodynamics, we evaluate the exact cumulants of energy transport in quantum critical systems of arbitrary dimension at low but nonzero temperatures, observing a phase transition for Lorentz boosts at the sound velocity., Comment: 27+22 pages, one figure

Abstract

We propose exact results for the full counting statistics, or the scaled cumulant generating function, pertaining to the transfer of arbitrary conserved quantities across an interface in homogeneous integrable models out of equilibrium. We do this by combining insights from generalised hydrodynamics with a theory of large deviations in ballistic transport. The results are applicable to a wide variety of physical systems, including the Lieb-Liniger gas and the Heisenberg chain. We confirm the predictions in non-equilibrium steady states obtained by the partitioning protocol, by comparing with Monte Carlo simulations of this protocol in the classical hard rod gas. Abstract

Ultra-compact, low-loss, fast, and reconfigurable optical components, enabling manipulation of light by light, could open numerous opportunities for controlling light on the nanoscale. Nanostructured all-dielectric metasurfaces have been shown to enable extensive control of amplitude and phase of light in the linear optical regime. Among other functionalities, they offer unique opportunities for shaping the wave front of light to introduce the orbital angular momentum (OAM) to a beam. Such structured light beams bring a new degree of freedom for applications ranging from spectroscopy and micromanipulation to classical and quantum optical communications. To date, reconfigurability or tuning of the optical properties of all-dielectric metasurfaces have been achieved mechanically, thermally, electrically or optically, using phase-change or nonlinear optical materials. However, a majority of demonstrated tuning approaches are either slow or require high optical powers. Arsenic trisulfide (As$_2$S$_3$) chalcogenide glass offering ultra-fast and large $\chi^{(3)}$ nonlinearity as well as a low two-photon absorption coefficient in the near and mid-wave infrared spectral range, could provide a new platform for the realization of fast and relatively low-intensity reconfigurable metasurfaces. Here, we design and experimentally demonstrate an As$_2$S$_3$ chalcogenide glass based metasurface that enables reshaping of a conventional Hermite-Gaussian beam with no OAM into an OAM beam at low-intensity levels, while preserves the original beam's amplitude and phase characteristics at high-intensity levels. The proposed metasurface could find applications for a new generation of optical communication systems and optical signal processing.

Abstract

Leukemia stem cells (LSCs) are thought to drive the genesis of acute myeloid leukemia (AML) as well as relapse following chemotherapy. Because of their unique biology, developing effective methods to eradicate LSCs has been a significant challenge. In the present study, we demonstrate that intrinsic overexpression of the mitochondrial dynamics regulator FIS1 mediates mitophagy activity that is essential for primitive AML cells. Depletion of FIS1 attenuates mitophagy and leads to inactivation of GSK3, myeloid differentiation, cell cycle arrest, and a profound loss of LSC self-renewal potential. Further, we report that the central metabolic stress regulator AMPK is also intrinsically activated in LSC populations and is upstream of FIS1. Inhibition of AMPK signaling recapitulates the biological effect of FIS1 loss. These data suggest a model in which LSCs co-opt AMPK/FIS1-mediated mitophagy as a means to maintain stem cell properties that may be otherwise compromised by the stresses induced by oncogenic transformation.

24. Faculty Liaisons: An Embedded Approach for Enriching Teaching and Learning in Higher Education

Abstract

This paper explores the experiences of a group of academic developers who support educational development work as Faculty Liaisons at a large, research-intensive university. These academic developers inhabit complex 'third spaces', providing support through an embedded partnership relationship that requires lateral movement across functional and organizational boundaries to create new professional spaces, knowledge, and relationships. The authors utilize narrative inquiry and auto-ethnographic approaches to present an interpretive qualitative analysis of their experiences supporting Faculty and University projects across complex and evolving organizational boundaries. From this analysis, they highlight key roles and responsibilities associated with their blended context and identify challenges that academic developers who occupy third spaces within academic organizations face as they negotiate competing interests, identities, and requirements associated with the diverse range of their projects and the blended experience of working in scholarly and administrative, central- and Faculty-based roles. The lessons they have learned from these experiences will be of particular interest to academic developers who are experiencing the flux of change within higher education settings that are impacting teaching and learning practices both for faculty in the classroom and for those across the institution who support them.

Abstract

While animal model studies have extensively defined the mechanisms controlling cell diversity in the developing mammalian lung, there exists a significant knowledge gap with regards to late-stage human lung development. The NHLBI Molecular Atlas of Lung Development Program (LungMAP) seeks to fill this gap by creating a structural, cellular and molecular atlas of the human and mouse lung. Transcriptomic profiling at the single-cell level created a cellular atlas of newborn human lungs. Frozen single-cell isolates obtained from two newborn human lungs from the LungMAP Human Tissue Core Biorepository, were captured, and library preparation was completed on the Chromium 10X system. Data was analyzed in Seurat, and cellular annotation was performed using the ToppGene functional analysis tool. Transcriptional interrogation of 5500 newborn human lung cells identified distinct clusters representing multiple populations of epithelial, endothelial, fibroblasts, pericytes, smooth muscle, immune cells and their gene signatures. Computational integration of data from newborn human cells and with 32,000 cells from postnatal days 1 through 10 mouse lungs generated by the LungMAP Cincinnati Research Center facilitated the identification of distinct cellular lineages among all the major cell types. Integration of the newborn human and mouse cellular transcriptomes also demonstrated cell type-specific differences in maturation states of newborn human lung cells. Specifically, newborn human lung matrix fibroblasts could be separated into those representative of younger cells (n = 393), or older cells (n = 158). Cells with each molecular profile were spatially resolved within newborn human lung tissue.
