Hancornia speciosahas bioactive molecules such as phenolic compounds which exhibit antioxidant and anti-microbial properties. Nevertheless, the separation of these substances from a medicinal plant can be differentaccording to the extraction process and solvent used. Thus, the present study aimed was to separate phenoliccompounds fromHancornia speciosaleaves in three steps by using a sequential pressurized liquid extraction andevaluated their antioxidant and antibacterial activity. Results showed that the temperature increase from 25 to60° C provides a significative enhancement in the extraction yield. The raw ethanol extract (39.0 ± 0.2%)showed the highest overall extraction yield. However, fractionated ethanol extract (FEE) at 60 °C exhibited thehighest amount of total phenolic compounds (347.0 ± 4.12 mg/g extract). Fractionated ethyl acetate extract(FAE) at 60 °C presented improved quantity of totalflavonoids (230.4 ± 3.5 mg/g extract) and rutin content(16.6 ± 0.3 mg/mL extract). The antioxidant activity results for IC50of DPPH indicated that FEE(3.8 ± 0.1 ?g/mL) was the best free radical scavenger. FAE and FEE were able to inhibit the growth of mul-tidrug-resistantE. coli. Basil is an aromatic herb that stands out fo its economic importance. It is consumed in natura and sed to obtain essential oil. The cultivation of this species in several regions of the world has allowed variations by natural crosses and euploidy, leading to the wide genetic variability found nowadays. Considering the importance of this species, we aimed to analyze the genetic diversity of 27 basil genotypes using ISSR molecular markers. Fourteen primers were employed for DNA amplification, resulting in 86% polymorphism. Based on the Jaccard?s dissimilarity index, the highest index (0.80) was observed between the individuals BAS001 and BAS012, while the lowest index (0.18) was detected between the genotypes BAS014 and BAS015. The genetic similarity among individuals was calculated, forming four distinct clusters. Most individuals (40.7%) were allocated in cluster I. The polymorphic information content (PIC) (0.89) indicated considerable levels of genetic diversity among genotypes. In this sense, the ISSR markers were efficient in the detection of polymorphisms between the accessions, suggesting the genetic variability of the collection. This result demonstrates the importance of the use of molecular markers and the advantages that this information provides to the breeding of the species. O manjericão é uma erva aromática que se destaca por possuir importância econômica. É consumido in natura e também utilizado na obtenção de óleo essencial. O cultivo desta espécie em diversas regiões do mundo permitiu que surgissem variações mediante cru-zamentos naturais e euploidia, ocasionando a ampla variabilidade genética existente. Considerando a importância desta espécie, este trabalho teve como objetivo analisar a diversidade genética de 27 genótipos de manjericão usando marcadores moleculares ISSR. Qua-torze primers foram utilizados para amplificação do DNA, resultando em 86% de polimorfismo. Com base no índice de dissimilaridade de Jaccard, observou-se o maior índice (0,80) entre os indivíduos BAS001 e BAS012, enquanto que o menor índice de dissimilari-dade (0,18) foi detectado entre os genótipos BAS014 e BAS015. A semelhança genética entre indivíduos foi calculada, formando quatro grupos distintos. A maioria dos indivíduos (40,7%) foi agrupada no grupo I. O conteúdo de informação polimórfica (PIC) (0,89) indicou níveis consideráveis de diversidade genética entre os genótipos. Neste sentido, os marcadores ISSR foram eficientes na detecção de polimorfismos entre os acessos e confirmaram que é possível inferir a variabilidade genética na coleção. A mandioca é a única espécie do gênero Manihot comercialmente produzida. A produção de mudas micropropagadas possui limitações, e entre estas está o alto custo com energia utilizadas nas salas de crescimento, e na taxa de sobrevivência das plantas na fase de aclimatização, porém a cultura de tecidos é uma ferramenta de multiplicação rápida e obtenção de plantas sadias (cultura de meristema), pois o método de propagação vegetativa acarreta problemas fitossanitários como disseminação de patógenos. Uma alternativa para diminuir gastos dessa técnica seria a redução de sacarose ao meio e indução de enraizamento sob luz natural em casa de vegetação. Sendo assim, o objetivo desse trabalho foi avaliar a influência da luz e sacarose na sobrevivência e desenvolvimento de mudas de mandioca micropropagada. As variedades BRS Formosa, Lagoão e BRS Verdinha, cultivadas em meio MS acrescido de diferentes concentrações de sacarose (10; 20 e 30 g.L-1), e após 30 dias em sala de crescimento, os frascos foram incubados em sala de crescimento e casa de vegetação por 15 dias. Em cultivos envolvendo diferentes tipos de luz, BRS Formosa e Lagoão, alcançaram melhores médias sob luz natural e altas concentrações de sacarose, enquanto que BRS Verdinha obteve melhor desenvolvimento em luz natural e baixa concentração de sacarose. Drought is the most limiting environmental factor to crop productivity and presents a great variability in the degree of tolerance among and within species, among varieties. The aim of this study was to characterize sugarcane accessions regarding tolerance to water stress during in vitro cultivation based on changes in biometric, physiological and biochemical characteristics, within species and among species, to support future breeding programs. Adventitious shoots of five sugarcane accessions: Saccharum robustum, Saccharum spontaneum and Saccharum officinarum species, cultivated in Murashige and Skoog medium supplemented with 2% sucrose and 4 g/l Phytagel were used in five water potentials, 0, -0.3, -0.6, -0.9, -1.2 MPa, induced by mannitol. Survival, length of shoots and roots, number of shoots and roots, biomass, proline content in leaves and activity of antioxidant enzymes were analyzed. There is difference among species, and also, within the same sugarcane species when submitted to in vitro drought stress, and S. officinarum was shown to be the most tolerant. Proline can be used as a biochemical indicator of response to drought in sugarcane accessions and its accumulation was intensified in S. robustum and S. spontaneum accessions. A abóbora (Cucurbita moschata Duch) é uma olerícola de grande aceitação em todo o Brasil, especialmente na região Nordeste. É bastante utilizada na alimentação diária do nordestino, onde é conhecida por Jerimum, para elaboração de diversos pratos e doces, que podem ser caseiros ou industrializados. Durante o processo de cocção, há perda de nutrientes e alteração na qualidade do alimento a ser consumido. O trabalho foi desenvolvido com o objetivo de avaliar as alterações ocorridas em abóboras após a cocção. Os 115 frutos avaliados foram oriundos do programa de melhoramento de abóbora da Embrapa Tabuleiros Costeiros (CPATC), colhidos em outubro de 2013, no Campo Experimental Pedro Arle, locali zado no município de Frei Paulo, Sergipe. As análises foram realizadas na Sala de Pós-colheita do CPATC, em Aracaju. Os pedaços, pesando cerca de 200g foram fervidos em 1L de água, onde permaneceram por cinco minutos. Antes e após a cocção, todas as amostras foram avaliadas quanto ao peso da massa fresca, firmeza e teor de sólidos solúveis (SS). Foi realizado o teste de Tukey para comparação de médias, ao nível de 5% de significância, utilizando-se o programa estatístico SAS. Não ocorreu perda de massa fresca significativa, que variou de 142,28g (antes) a 138,27 g (após a cocção). Entretanto, o teor de SS diminuiu significativamente de 11,68 para 11,08ºBrix, e a firmeza, de 12,75 a 5,69N. As características avaliadas neste trabalho, associadas a outras de importância nutricional e comercial, são importantes informações ao programa de melhoramento da espécie.

Myrcia lundiana Kiaersk. is a tree of the family Myrtaceae found in tropical and subtropical areas of the southern hemisphere that produces essential oil. The aim of this study was to characterize the genetic diversity of M. lundiana plants from a native population of Parque Nacional de Itabaiana, using inter-simple sequence repeat molecular markers. Thirty-five primers were tested, 20 of which were polymorphic, resulting in 135 polymorphic and informative bands. Results of the cluster analysis, obtained using the unweighted pair group method with arithmetic mean, grouped plants into three clusters: Cluster I - MLU001, MLU002, MLU003, MLU004, MLU005, MLU006, MLU018, MLU019, LU020, MLU021, LU022; MLU008, MLU011, MLU012, MLU014, MLU015, MLU017, MLU026, and MLU028; Cluster II - MLU007, MLU009, MLU010, MLU013, and MLU016; and Cluster III - MLU023, MLU024, MLU025, and MLU027. Jaccard similarity coefficients for pair-wise comparisons of plants ranged between 0.15 and 0.87. MLU014 and MLU015 presented low genetic diversity, with a similarity index of 0.87. Conversely, MLU007 and MLU019 presented high diversity, with a similarity index of 0.15. According to the structure analysis, three distinct clusters were formed. Genetic diversity of M. lundiana plants was intermediate, and expansion of its genetic diversity is necessary. MLU026 and MLU028 are the most suitable for selection in breeding programs, since they clearly represent all of the diversity present in these plants. Moreover, these results provide important information on the existing genetic variability, highlighting the importance of Parque Nacional de Itabaiana for the conservation of this species.

Cambui (Myrciaria tenella O. Berg) is a native species from Brazil, which belongs to the family Myrtaceae. Molecular characterization is one of the most used tools for the study of the biotechnological potential of species because the diversity level between individuals can be inferred. Analysis of enetic diversity is fundamental to the direction of the strategies necessary to form and maintain a germplasm. This study aimed to evaluate the genetic diversity in a natural population of cambui using inter-simple sequence repeat (ISSR) molecular markers. The natural population, which provided the plant material, is found at the Private Reserve of Natural Heritage of Caju, which belongs to the experimental field of Embrapa Tabuleiros Costeiros, in the municipality of Itaporanga d’Ajuda, SE, Brazil. Young leaves of each individual were collected for DNA extraction and analysis of PCR-ISSR. Thirty primers were tested and the top 10 were selected. The use of these primers resulted in 71 fragments with 98.3% polymorphism. Similarity of individuals ranged between 0.30 and 0.92. The most similar individuals were C13 and C17 and the most distant were C1 and C41. Through UPGMA, six distinct groups were identified. This information may be used for conservation of these genetic resources, germplasm exchange, creation of germplasm bank and in future studies with this species. O objetivo do presente trabalho foi estudar as respostas morfogenéticas de explantes de jenipapeiro em diferentes condições de cultura in vitro para subsidiar futuros protocolos de multiplicação e produção in vitro de metabólitos secundários. Após 90 dias de cultivo, as plântulas de jenipapeiro foram segmentadas (segmentos nodal e foliar) e transferidas para meio MS suplementado com 30 g L-1 de sacarose e diferentes concentrações de 2,4 - Diclorofenoxiacético - 2,4-D (0; 4 e 8 mg L-1) combinadas com quatro concentrações de benzilaminopurina-BAP (0; 1; 2 e 3 mg L-1). Aos 120 dias a porcentagem de explantes com resposta morfoge-nética quanto à formação de calos e/ou organogênese foi avaliada. O 2,4-D nas concentrações de 4 e 8 mg L-1 induz aumento na resposta morfogenética, principalmente na formação de calos, segmentos foliar e nodal de Genipa americana L. acesso SIR. O BAP a 1,77 mg L-1 induz maior formação de calos em segmentos foliares e maior regeneração de brotos nos segmentos foliar e nodal. The objective of this work was to determine the mannitol concentration for in vitro germination and minimum growth of coconut (Cocos nucifera). The following accessions were used: Brazilian Red Dwarf Gramame (BRDG), Malaysian Yellow Dwarf (MYD), Brazilian Tall (BRA), Polynesian Tall (PYT), and Rennell Island Tall (RIT). Embryos were inoculated in an Y3 culture medium at different mannitol concentrations (0, 0.1, 0.2, 0.3, and 0.4 mol L?1). Inhibition of shoot development was verified at 0.1, 0.2, and 0.3 mol L?1, for MYD and BRDG, and at 0.1 and 0.2 mol L?1, for PYT, BRA, and RIT. This research had as objective to characterize genetically individuals of physic nut cultivated in experimental areas in Sergipe, Brazil by means of RAPD molecular markers. Leaves of 40 individuals were collected and DNA was isolated using CTAB 2% method. Were used 30 primers RAPD for DNA amplification, and this data was used to estimate the genetic similarity among the pairs of individuals, using Jaccard coefficient, and group them out for the UPGMA method. Also, the genetic structure and diversity of the populations were assessed using AMOVA. Of the 100 fragments generated, 29 of were polymorphic. A similarity average of 0.54 among the individuals was found and the amplitude similarities varied from 0.18 to 1.00. One of them (U5) was unit clusters and formed by the most divergent individuals.
