Abstract

Human herpesvirus 8 (HHV-8) is the etiological agent of all forms of Kaposi&rsquo, s sarcoma (KS). K1 gene studies have identified five major molecular genotypes with geographical clustering. This study described the epidemiology of HHV-8 and its molecular diversity in Gabon among Bantu and Pygmy adult rural populations and KS patients. Plasma antibodies against latency-associated nuclear antigens (LANA) were searched by indirect immunofluorescence. Buffy coat DNA samples were subjected to polymerase chain reaction (PCR) to obtain a K1 gene fragment. We studied 1020 persons, 91% were Bantus and 9% Pygmies. HHV-8 seroprevalence was 48.3% and 36.5% at the 1:40 and 1:160 dilution thresholds, respectively, although the seroprevalence of HHV-8 is probably higher in Gabon. These seroprevalences did not differ by sex, age, ethnicity or province. The detection rate of HHV-8 K1 sequence was 2.6% by PCR. Most of the 31 HHV-8 strains belonged to the B genotype (24), while the remaining clustered within the A5 subgroup (6) and one belonged to the F genotype. Additionally, we reviewed the K1 molecular diversity of published HHV-8 strains in Africa. This study demonstrated a high seroprevalence of HHV-8 in rural adult populations in Gabon and the presence of genetically diverse strains with B, A and also F genotypes.

Abstract

International audience; Background - Although antiretroviral therapy (ART) has reduced the risk of Kaposi sarcoma (KS), KS cases still occur in HIV-infected people. Objective - To describe all KS cases observed between 2010 and 2015 in a country with high ART coverage. Methods - Retrospective study using longitudinal data from 44 642 patients in the French Dat'AIDS multicenter cohort. Patients' characteristics were described at KS diagnosis according to ART exposure and to HIV-plasma viral load (HIV-pVL) (≤50 or >50) copies/mL. Results - Among the 209 KS cases diagnosed during the study period, 33.2% occurred in ART naïve patients, 17.3% in ART-experienced patients and 49.5% in patients on ART, of whom 23% for more than 6 months. Among these patients, 24 (11.5%) had HIV-pVL ≤50 cp/mL, and 16 (66%) were treated with a boosted-PI-based regimen. The distribution of KS localization did not differ by ART status nor by year of diagnosis. Limitations - Data on human herpesvirus 8, treatment modalities for KS and response rate were not collected. Conclusion - Half of KS cases observed in the study period occurred in patients not on ART, reflecting the persistence of late HIV diagnosis. Factors associated with KS in patients on ART with HIV-pVL ≤50 cp/mL remain to be explored.

Abstract

International audience; The DNA-mediated innate immune response underpins anti-microbial defenses and certain autoimmune diseases. Here we used immunoprecipitation, mass spectrometry, and RNA sequencing to identify a ribonuclear complex built around HEXIM1 and the long non-coding RNA NEAT1 that we dubbed the HEXIM1-DNA-PK-paraspeckle components-ribonucleoprotein complex (HDP-RNP). The HDP-RNP contains DNA-PK subunits (DNAPKc, Ku70, and Ku80) and paraspeckle proteins (SFPQ, NONO, PSPC1, RBM14, and MATRIN3). We show that binding of HEXIM1 to NEAT1 is required for its assembly. We further demonstrate that the HDP-RNP is required for the innate immune response to foreign DNA, through the cGAS-STING-IRF3 pathway. The HDP-RNP interacts with cGAS and its partner PQBP1, and their interaction is remodeled by foreign DNA. Remodeling leads to the release of paraspeckle proteins, recruitment of STING, and activation of DNAPKc and IRF3. Our study establishes the HDP-RNP as a key nuclear regulator of DNA-mediated activation of innate immune response through the cGAS-STING pathway.

Abstract

International audience; BACKGROUND: Kaposi sarcoma (KS) is a complex tumor of uncertain clonality. Studying the viral clonality of the human herpesvirus 8 (HHV-8) in KS to determine clonality of the tumors, a strategy that has been used previously with Epstein-Barr virus and its associated tumors, may elucidate whether multicentric (disseminated) KS lesions correspond to metastatic lesions or to expansions of independent clones. METHODS: A series of 139 KS biopsies (from skin, lymph node, or tonsil) was obtained from 98 patients, with 59 biopsies from 18 patients with disseminated multicentric KS skin lesions. The degree of spindle cell infiltration in biopsies was established by direct observation of hematoxylin-eosin-stained sections, and HHV-8 viral load was quantified by real-time polymerase chain reaction. To determine cellular clonality, the size heterogeneity of the HHV-8-fused terminal repeat (TR) region was determined by probing of electrophoresed restricted genomic DNA from KS biopsies for the HHV-8 TR sequence. RESULTS: HHV-8 clonality analysis was performed on the 62 samples for which sufficient DNA was obtained. Most samples corresponded to histologically nodular lesions with high spindle cell infiltration and high viral load. A clonal HHV-8 pattern was determined for 59 samples; 11 were found to be monoclonal and 48 to be oligoclonal. The informative samples that were from disseminated KS skin lesions (n = 26, from six patients) were either monoclonal or oligoclonal, and the size of HHV-8 episomes varied between these samples. CONCLUSION: Although some tumor KS lesions were monoclonal expansions of HHV-8-infected spindle cells, most advanced lesions were oligoclonal proliferations. Furthermore, individual KS disseminated tumor skin lesions were found to represent distinct expansions of HHV-8-infected spindle cells. Thus, our results suggest that KS lesions, especially in patients with advanced skin tumors, are reactive proliferations rather than true malignancies with metastatic dissemination.

Abstract

International audience; Infection with human herpesvirus 8 (HHV-8), the etiological agent of Kaposi's sarcoma, has been shown to display strong familial aggregation, in countries in which HHV-8 infection is endemic. We investigated 40 large families (608 subjects aged one to 88 years) living in an isolated area of Cameroon in which HHV-8 is highly endemic. We performed a two-step genetic analysis for HHV-8 infection status (HHV-8+/HHV-8- determined by immunofluorescence) consisting of an initial segregation analysis followed by a model-based genome-wide linkage analysis. Overall HHV-8 seroprevalence was 60%, increasing with age. Segregation analysis provided strong evidence for a recessive major gene conferring predisposition to HHV-8 infection. This gene is predicted to have a major effect during childhood, with almost all homozygous predisposed subjects (∼7% of the population) becoming infected by the age of 10. Linkage analysis was carried out on the 15 most informative families, corresponding to 205 genotyped subjects. A single region on chromosome 3p22 was significantly linked to HHV-8 infection (LOD score=3.83, P=2.0 × 10(-5)). This study provides the first evidence that HHV-8 infection in children in endemic areas has a strong genetic basis involving at least one recessive major locus on chromosome 3p22.

Abstract

Background For an efficient immune response against viral infection, dendritic cells (DCs) must express a coordinate repertoire of receptors that allow their recruitment to the sites of inflammation and subsequently to the secondary lymphoid organs in response to chemokine gradients. Several pathogens are able to subvert the chemokine receptor expression and alter the migration properties of DCs as strategy to escape from the immune control. Findings Here we report the inhibitory effect of Human Herpesvirus 8 (HHV-8) on the migratory behavior of immature and mature DCs. We found that the virus altered the DC chemokine receptor expression and chemokine induced migration. Moreover HHV-8 was also able to interfere with basal motility of DCs by inducing cytoskeleton modifications. Conclusion Based on our findings, we suggest that HHV-8 is able to subvert the DC migration capacity and this represents an additional mechanism which interferes with their immune-functions.

Abstract

International audience; Childhood multicentric Castleman disease (MCD) is a rare and unexplained lymphoproliferative disorder. We report a human herpesvirus-8 (HHV-8)-infected child, born to consanguineous Comorian parents, who displayed isolated MCD in the absence of any known immunodeficiency. We also systematically review the clinical features of the 32 children previously reported with isolated and unexplained MCD. The characteristics of this patient and the geographic areas of origin of most previous cases suggest that pediatric MCD is associated with HHV-8 infection. Moreover, as previously suggested for Kaposi sarcoma, MCD in childhood may result from inborn errors of immunity to HHV-8 infection.

International audience; HHV-8 belongs to the family Herpesviridae, the subfamily Gammaherpesvirinae and the genus Rhadinovirus. While several viral homologs exist in both old-world and new-world non-human primates, HHV-8 is the only known human rhadinovirus. HHV-8 is considered to be the etiological agent of the four clinical-epidemiological forms of Kaposi's sarcoma (classic, endemic, post-transplant and epidemic/HIV-associated). In several African regions, epidemic KS is the most frequently diagnosed malignancy. In 2002, the estimated annual incidence of KS worldwide was approximately 65,000 cases, representing 1% of all diagnosed cancers. HHV-8 is also associated with primary effusion lymphoma, some cases of multicentric Castleman disease, and other rare lymphomas. All these tumors mainly affect immunodeficient patients. Recent studies indicate that KS tumors are due to mono- or oligoclonal expansion of latently HHV-8-infected spindle cells, Finthermore, advanced multicentric KS lesions are of multiclonal origin. HHV-8 is not a ubiquitous virus. It is mainly endemic in areas where classical or endemic Kaposi's sarcoma is highly endemic, including the Mediterranean basin and East and Central Africa. In the latter areas, the HHV-8 seroprevalence can reach 80% in the adult population. In the homosexual population (mainly in the U.S.A. and Europe), HHV-8 is mainly transmitted during repeated sexual contacts, whereas in Africa it is mainly transmitted from mother to child and among siblings. Saliva seems to play a major role in HH V-8 transmission. Molecular epidemiology studies of the K1 gene (one of the most variable genomic regions) have revealed different molecular subtypes, at least some of which appear to be linked mainly to the geographic origin of the samples rather than to the underlying disease.; L’HHV-8 est un virus de la famille des herpesviridae, de la sous-famille des Gammaherpesvirinae et du genre Rhadinovirus. Alors qu’il existe de nombreux homologues viraux de l’HHV-8 dans plusieurs espèces de primates non-humains, l’HHV-8 est le seul rhadinovirus connu chez l’homme. L’HHV-8 est considéré comme l’agent étiologique de toutes les formes du sarcome de Kaposi (classique, endémique, post-transplant et épidémique ou associée au VIH). Alors que l’incidence de la forme épidémique a fortement diminué ces dernières années dans les pays occidentaux, cette tumeur représente actuellement la prolifération la plus fréquente dans de nombreux pays d’Afrique Centrale et Australe. Le nombre de sarcome de Kaposi annuel est estimé à près de 65 000 cas, soit près de 1 % de tous les cancers diagnostiqués chaque année dans le monde. Cet herpèsvirus oncogène est aussi associé au lymphome des cavités, à certains cas de la maladie de Castleman multicentrique ainsi qu’à d’autres lymphomes rares. Ces dernières tumeurs survenant surtout dans un contexte d’immunodéficience. Des travaux récents ont démontré que le sarcome de Kaposi, dans sa forme tumorale avancée, était une prolifération mono ou oligoclonale de cellules fusiformes infectées par l’HHV-8 et que les lésions multicentriques, très fréquentes dans cette tumeur, étaient d’origine multiclonales. L’HHV-8 n’est pas un virus ubiquitaire. Il est principalement endémique dans les zones où le sarcome de Kaposi est fréquent, c’est-à-dire le pourtour du bassin méditerranéen et surtout l’Afrique Centrale et de l’Est, régions où la séroprévalence virale atteint près de 80 % chez les adultes. Alors que dans la population homosexuelle masculine (principalement aux USA et en Europe), le virus se transmet surtout durant les contacts sexuels, la transmission en zone de forte endémie (Afrique) se fait avant tout de la mère à l’enfant, puis entre enfants. La salive semble jouer un rôle majeur dans la transmission virale. Les études d’épidémiologie moléculaire portant sur le gène K1 (l’une des régions génomiques les plus variables) ont mis en évidence l’existence de plusieurs sous-types viraux qui ne semblent pas liés au type de maladie associée mais dans une grande mesure à l’origine géographique des patients.

International audience; We show human herpesvirus 8 with diverse molecular subtype D variants to be highly endemic among the Ni-Vanuatu population. Most K1 genes were nearly identical to Polynesian strains, although a few clustered with Australian or Taiwanese strains. These results suggest diverse origins of the Ni-Vanuatu population and raise questions about the ancient human population movements in Melanesia.

Abstract

International audience; HHV-8-associated solid lymphomas which develop in extracavitary sites during the course of primary effusion lymphoma (PEL) could represent the relapse of original PEL tumors in different anatomical sites, or newly occurring distinct HHV-8-associated lymphomas, such as multicentric Castleman disease-related microlymphomas. HHV-8 episome clonality might help identify which event takes place.

Abstract

Human herpesvirus 8 (HHV-8), also known as Kaposi’s arcoma (KS)-associated herpesvirus (KSHV), is the etioogical agent of all forms of Kaposi’s sarcoma, a tumor of ixed cellularity occurring frequently during human immundeficiency virus type 1 (HIV-1) infection (AIDS/epidemicS) and in transplant recipients (iatrogenic KS) (Boshoff nd Weiss, 2001; Hengge et al., 2002). Such tumor also ccurs among non-HIV-infected individuals, predominantly ither in aged men of Mediterranean and Middle East oriins (classic KS) or in inhabitants from East and Central frica (endemic KS) (Boshoff and Weiss, 2001; Hengge et l., 2002). Abstract

International audience; We evaluated the presence of human herpesvirus 8 (HHV-8) infection among groups of Amerindians in French Guiana. The overall prevalence of antibodies against lytic HHV-8 antigens was 23.0% (180/781), increasing from 18.4% in children 45 years). Seroprevalence was higher in Amerindians living in remote localities than it was in those living in the coastal region. Analysis of a 725-base pair fragment of the K1 gene amplified from DNA from a Wayampi Amerindian showed that the virus belonged to molecular subtype E, which has hitherto been found in only a few Amerindians in Brazil and Ecuador.

Abstract

International audience; The mRNA targets of microRNAs (miRNAs) can be identified by immunoprecipitation of Argonaute (Ago) protein-containing RNA-induced silencing complexes (RISCs) followed by microarray analysis (RIP-Chip). Here we used Ago2-based RIP-Chip to identify transcripts targeted by Kaposi's sarcoma-associated herpesvirus (KSHV) miRNAs (n = 114), Epstein-Barr virus (EBV) miRNAs (n = 44), and cellular miRNAs (n = 2337) in six latently infected or stably transduced human B cell lines. Of the six KSHV miRNA targets chosen for validation, four showed regulation via their 3'UTR, while two showed regulation via binding sites within coding sequences. Two genes governing cellular transport processes (TOMM22 and IPO7) were confirmed to be targeted by EBV miRNAs. A significant number of viral miRNA targets were upregulated in infected cells, suggesting that viral miRNAs preferentially target cellular genes induced upon infection. Transcript half-life both of cellular and viral miRNA targets negatively correlated with recruitment to RISC complexes, indicating that RIP-Chip offers a quantitative estimate of miRNA function.
